Uptake kinetics of arsenic species in rice plants   总被引:35,自引:0,他引:35  
Arsenic (As) finds its way into soils used for rice (Oryza sativa) cultivation through polluted irrigation water, and through historic contamination with As-based pesticides. As is known to be present as a number of chemical species in such soils, so we wished to investigate how these species were accumulated by rice. As species found in soil solution from a greenhouse experiment where rice was irrigated with arsenate contaminated water were arsenite, arsenate, dimethylarsinic acid, and monomethylarsonic acid. The short-term uptake kinetics for these four As species were determined in 7-d-old excised rice roots. High-affinity uptake (0-0.0532 mM) for arsenite and arsenate with eight rice varieties, covering two growing seasons, rice var. Boro (dry season) and rice var. Aman (wet season), showed that uptake of both arsenite and arsenate by Boro varieties was less than that of Aman varieties. Arsenite uptake was active, and was taken up at approximately the same rate as arsenate. Greater uptake of arsenite, compared with arsenate, was found at higher substrate concentration (low-affinity uptake system). Competitive inhibition of uptake with phosphate showed that arsenite and arsenate were taken up by different uptake systems because arsenate uptake was strongly suppressed in the presence of phosphate, whereas arsenite transport was not affected by phosphate. At a slow rate, there was a hyperbolic uptake of monomethylarsonic acid, and limited uptake of dimethylarsinic acid.  相似文献

Summary A study was made of the effects of the herbicides 2,4-D (2,4-dichlorophenoxyacetic acid) and MCPA (4-chloro-2-methyl-phenoxyacetic acid) on ion uptake, leakage and growth of rice seedlings. Using isotopically-labelled solutions containing different concentrations of 2,4-D or MCPA, it was established that 10–4 M 2,4-D or MCPA effectively inhibited potassium ion uptake, while K-ion leakage from the roots occurred only at 10–3 M. The growth of the rice seedlings was markedly retarded even at low (10–6 M) concentrations, and the roots and shoots showed different tolerances to the herbicide. At 10–8 M herbicide, the effects were not injurious, but rather favourable. Reduction in root length by herbicides was not in accordance with dry-matter production.  相似文献

InAnacystis nidulans uptake and efflux kinetics of fluoresceine were studied. It could be concluded that the distribution of fluoresceine depends on the pHc of the cells. The apparent photosynthetic Km (CO2) of air-grown Dunaliella salina is 2 M as measured both by the filtering centrifugation technique and by O2 electrode. These cells are capable of accumulating inorganic carbon (Cinorg) up to 20 times its concentration in the medium. It is suggested that air-grown Dunaliella cells are able to concentrate CO2 within the cell. Analysis of the efflux of Cinorg from cells previously loaded with H14CO 3 - demonstrated the existence of an internal pool which has an half-time of depletion of 2.5–7 min depending on the conditions of the experiment. This finding indicates that the internal Cinorg pool is not readily exchangeable with the external medium. Furthermore, the influence of the presence or absence of unlabelled Cinorg in the medium during the efflux experiment on the half-time observed indicate that efflux of Cinorg is not a simple diffusion process but is rather carrier-mediated.Abbreviation Cinorg inorganic carbon  相似文献

Sulfate efflux from an intracellular pool was observed with both wild-type and cys-11 cells of Neurospora and apparently occurs by way of the sulfate transport system. Efflux requires the presence of external sulfate or the related ions, chromate, selenate, or thiosulfate, and probably occurs by an exchange reaction. The sulfur amino acids, cysteine or methionine, do not promote sulfate efflux. The Km for efflux is much greater than the Km for sulfate uptake, which permits the accumulation of a considerable intracellular pool before efflux becomes significant. Substantial transmembrane movement of sulfate both influx and exit, was found to occur in azidetreated cells, although the net uptake of sulfate was abolished by this inhibitor. Both sulfate uptake and efflux are inhibited by p-chloromercuribenzoate which suggests that the sulfate permease possesses an essential sulfhydryl group.  相似文献

Summary Two15N-labelled slow-release nitrogen (N) sources, oxamide and isobutylidene diurea (IBDU), each at two particle sizes, and15N-labelled urea were compared at two rates as sources of N for rice (Oryza sativa) under two watering regimes which simulated a transplant (continuous flood, CF) and a direct-seeded (A/F) system of paddy rice culture. Highest grain yields were obtained from −8+10-mesh oxamide particles applied at the rate of 2,000 mg of N/5 kg of soil, CF series; this yield was slightly higher than that obtained from −3+4-mesh oxamide, A/F series. Incubating the N fertilizers in moist (22% water) soil for 21 days immediately before flooding and transplanting rice greatly reduced N supply because of nitrification during the preflood period, followed by denitrification after flooding. This resulted in less plant uptake of N and less grain yield from urea, fine oxamide and IBDU, A/F series. For coarse oxamide, N release during the preflood period resulted in higher N uptake and grain yield in the A/F rather than in the corresponding CF series. The pattern of fertilizer N uptake by rice plants was affected by kind of fertilizer, particle size of oxamide and IBDU, and watering regime. Uptake of fertilizer N generally paralleled uptake of soil N throughout the growth period. Plant tops continued to accumulate some N during the period of grain filling, but much of the N in plant tops was translocated to the grain after heading. There was a large decrease in dry weight, N content, and15N content of tops after heading. Root weight and N content increased rapidly at first, and then at a diminishing rate until maturity. Unexplained N deficits occurred in the CF series (14–23% of the N applied, depending on N rate and source), and in the A/F series for IBDU (37–43% of the N applied).  相似文献

Summary The uptake and distribution of potassium was examined in tomato plants, cv. Amberley Cross and Moneymaker, grown in peat/loam and given a nutrient feed either adequate or deficient in potassium.Detailed studies were also made of the distribution of potassium in various parts of tomato plants, cv. Amberley Cross, grown in sand and supplied with nutrient feeds containing four and seven concentrations of potassium. In these plants the concentration of potassium in stem, petiole and laminar tissue increased from the base to the apex of plants, irrespective of the potassium concentration in the nutrient feed. There was also a gradient of decreasing potassium concentration along leaves, from proximal to distal laminae. The concentration of potassium in all plant parts increased with increasing nutrient supply of potassium, petioles showing a greater response to nutrient potassium than laminar tissue.Marginal chlorosis and/or necrosis were observed when the potassium concentration in fully expanded leaves fell below 1.2 per cent of the dry weight for cv. Amberley Cross or 1.5 per cent for cv. Moneymaker. A 50 per cent incidence of chlorosis and/or necrosis occurred in leaf laminae containing 0.74 per cent potassium.Leaves rarely showed deficiency symptoms when the concentration of potassium in the petioles was higher than that in adjacent laminar tissue. However, chlorosis and/or necrosis occurred when the potassium concentration in petioles fell below that in laminae. In the diagnosis of the potassium status of tomato plants, the most suitable tissue for sampling for potassium analysis is considered to be the petioles of young fully expanded leaves.  相似文献

Summary In young oat plants, the mainstream of movement of Mn from nutrient solution to shoot may be regarded as passing through a labile fraction of Mn in the roots. It largely bypasses a relatively nonlabile fraction.Varietal differences in shoot-Mn can be attributed to variations in size and turnover rate of the labile fraction. Variations in pH and temperature of the substrate affect these quantities. But the effects are similar in both varieties, Sun II and Algerian, and, consequently, the varietal difference in shoot-Mn remains unmodified.Variations in total root-Mn can be largely attributed to variations in the size of the nonlabile fraction. This fraction, unlike the labile Mn, increases much more in plants of Algerian than in plants of Sun II, when the pH is raised or the temperature lowered. Consequently, the varietal difference in total root-Mn is easily disturbed by these substrate variations, and root-Mn can vary without relation to shoot-Mn.In several other varieties of oats, kinetic compartmentalisation of root-Mn could also be demonstrated. Summary The uptake and distribution of phosphorus was examined in tomato plants, cv. Kirdford Cross, grown in peat to which phosphate was added (P2) or omitted (P1). The plants received a liquid feed containing either a high (N2) or low (N1) concentration of ammonium nitrate. Initially, all plants were grown in peat containing an intermediate level of phosphate.There was a rapid net export of P from the leaves of plants transferred to the P1 medium resulting in deficiency symptoms before the fruit on the first truss had ripened. Most of the P absorbed by 11-week-old plants in the N1P2 and N2P2 treatments was located in the developing fruit, in the laminae of the mature leaves and in the lower parts of the stem. In the P1 treatments, the lowest fruit truss was the dominant sink for the limited supply of P, but there was also a significant concentration of P in the shoot apex and in the laminae. Increasing the supply of N to plants in the P2 treatment promoted the transport of P to the shoot and to the fruit trusses and also increased the total P uptake. However, plants in the N2 treatment required a significantly higher level of tissue P to prevent the symptoms of P deficiency occurring in the laminae. Generally, symptoms occurred in laminae of mature leaves containing less than 0.13 per cent P. Increases in concentration of tissue P in response to raising the level of applied P were greatest in the petioles of the mature leaves, and it is suggested that these petioles are the most suitable tissues for the assessment of the P status of tomato plants.  相似文献

Conidia of wild-type Neurospora crassa, preincubated for 3 1/2 h in growth medium, showed a typical triphasic pattern of adenine uptake. The three phases consisted of a quick initial uptake, followed by a plateau phase, and then by a resume lowered uptake. A study of the relative influx and efflux of [14C] adenine showed that the plateau phase in fact is a period of transmembrane movement of adenine and adenine metabolites. The efflux during the plateau phase essentially cancelled out all the influx during the same period. The uptake curve derived after taking into account the effluxed portion of radioactivity indicated that the second phase represents a period of lowered uptake activity. The beginning of the lowered uptake activity during the second phase is correlated with the presence of a high intracellular level of ATP derived from exogenous [14C]adenine. At the end of the secod phase, the intracellular level of ATP is much smaller and the rate of adenine uptake increases again. Analysis of the acid-soluble pool after feeding [14C]adenine indicated the presence of other 14C-nucleotides, but no detectable levels of bases and nucleosides were present. However, chromatographic analysis of the medium indicated that efflux results essentially in the accumulation of bases. The significance of this finding in relation to efflux is discussed.  相似文献

The presence and release of nanoparticles (NPs) into the environment have important implications for human health and the environment. A critical aspect of the risk assessment of nanoparticles is to understand the interactions of manufactured nanoparticles with plants. In this study, the uptake and distribution characteristics of two types of ceria nanoparticles with sizes of ca. 7 nm and 25 nm in cucumber plants were investigated using a radiotracer method and other techniques. With increasing concentration of the nanoparticles, concentration dependent absorption by the plant roots was noticed, but the majority of the particles only loosely adhered to the root surface. The seedlings treated with 7 nm ceria particles showed significantly higher ceria contents in both roots and shoots than those exposed to 25 nm ceria particles at all test concentrations (2, 20, and 200 mg L(-1)). Only very limited amounts of ceria nanoparticles could be transferred from the roots to shoots because the entry of nanoparticles into the roots was difficult. However, the results of tissue distributions of ceria nanoparticles in the plants and two dimensional distributions of the particles in the leaves imply that once they have entered into the vascular cylinder, ceria nanoparticles could move smoothly to the end of the vascular bundle along with water flow. To the best of our knowledge, this is the first detailed study of uptake and distribution of metal oxide nanoparticles in plants.  相似文献

Wide variation in CH4 flux was noticed among the ten rice cultivars grown under uniform field conditions. Cumulative CH4 flux ranged from 4.61 g m-2 to 20.25 g m-2. The rice cultivars could be ranked into three groups based on their CH4 flux potential. Rice cultivars could also be arranged based on their peak CH4 emission occurring either at vegetative, reproductive or at both growth stages. Of the several variables studied (root region redox potential, above- and underground biomass, grain and straw yield, duration of the crop, percent area occupied by the air space and root oxidase activity), only oxidase activity of the root tip exhibited a significant (negative) correlation with CH4 flux indicating an indirect effect of root oxidation potential on CH4 flux. Data presented in this study, demonstrate inherent variation in CH4 flux among different rice cultivars that can be used for developing future mitigation options.  相似文献

Glyphosate (N-phosphonomethylglycine) was readily transported in sugar beet plants (Beta vulgaris L., Klein E type, monogerm). Concentrations in sink leaves reached 2.5 to 13.7 micromolar in 10 hours from a 15 millimolar solution supplied to one mature leaf. Distribution of glyphosate followed that of [3H]sucrose used as a marker for materials transported by phloem, indicating that this is the primary means for distribution of glyphosate. Possible mechanisms of entry into the sieve tubes were evaluated using isolated leaf discs. Concentration dependence of uptake and kinetics of exodiffusion from tissue indicate a passive, nonfacilitated mechanism. Uptake was not affected by pH, eliminating the passive, weak acid mechanism. Permeability of the plasmalemma to glyphosate was calculated as 1.7 × 10−10 meters per second. This characteristic would allow slow entry and exit from the phloem, and together with other physiological parameters of the plant, is postulated to allow accumulation and transport in the phloem.  相似文献

An efflux of potassium ions was demonstrated in mutants of yeast cells lacking a functional high affinity carrier system for monovalent cations. This efflux showed the following characteristics: (a) It was stimulated by the presence of a substrate, either glucose or ethanol. (b) It was stimulated by several cationic organic molecules, such as ethidium bromide, dihydrostreptomycin, diethylaminoethyldextran, and also by trivalent cations, such as Al3+ and lanthanides; this stimulation also depended on the presence of a substrate. (c) K+ efflux was decreased in yeast mutants with decreased ATPase activity, which generated a lower membrane potential. (d) Although the efflux appeared to be of an electrogenic nature, producing hyperpolarization of cells, it was accompanied by the efflux of phosphate, probably as an anion partially compensating for the large amount of cations leaving the cell. (e) K+ efflux was also accompanied by an uptake of protons. (f) The efflux appeared more clearly in cells grown in YPD medium, and not in more complex media nor in the same YPD medium if supplemented with Ca2+ or Mg2+. Efflux of monovalent cations produced by Tb3+ and organic cationic agents was also demonstrated in wild type strains. This efflux system appears to be, at least partially, electrogenic, but seems to be also an exchange system for protons and to function as a symport with phosphate; it may be involved in the regulation of the internal pH of the cell, and appears to be regulated by its link to the energetic status of the cell, probably through the membrane potential.  相似文献
