Morphology has a crucial effect on productivity and the supply of substrate for cultures of filamentous fungi. However, cultivation parameters leading to the desired morphology are often chosen empirically as the mechanisms governing the processes involved are usually unknown. For coagulating microorganisms like Aspergillus niger the morphological development is considered to start with the aggregation of conidia right after inoculation. To elucidate the mechanism of this process, kinetic studies were carried out using an in-line particle size analyzer. Based on the data obtained from these experiments a model for conidial aggregation is proposed in this article. It consists of two separate aggregation steps. The first one takes place immediately after inoculation, but only leads to a small decrease of total particle concentration. Most suspended conidia aggregate after a second aggregation step triggered by germination and hyphal growth. Aggregation velocity of this second phase is linearly dependent on the particle growth rate.  相似文献

Aspergillus niger reproduces asexually by forming conidia. Here, the minimal nutrient requirements were studied that activate germination of A. niger conidia. To this end, germination was monitored in time using an oCelloScope imager. Data was used as input in an asymmetric model to describe the process of swelling and germ tube formation. The maximum number of spores (Pmax) that were activated to swell and to form germ tubes was 32.54% and 20.51%, respectively, in minimal medium with 50 mM glucose. In contrast, Pmax of swelling and germ tube formation was <1% in water or 50 mM glucose. Combining 50 mM glucose with either NaNO3, KH2PO4, or MgSO4 increased Pmax of swelling and germination up to 15.25% and 5.4%, respectively, while combining glucose with two of these inorganic components further increased these Pmax values up to 25.85% and 10.99%. Next, 10 mM amino acid was combined with a phosphate buffer and MgSO4. High (e.g. proline), intermediate and low (e.g. cysteine) inducing amino acids were distinguished. Together, a combination of an inducing carbon source with either inorganic phosphate, inorganic nitrogen or magnesium sulphate is the minimum requirement for A. niger conidia to germinate.  相似文献

Conidia of Aspergillus niger TMB 2022 were immobilized in calcium alginate for the production of citric acid. A 1-mL conidia suspension containing ca. 2.32 x 10(8) conidia were entrapped into sodium alginate solution in order to prepare 3% Ca-alginate (w/v) gel bead. Immobilized conidia were inoculated into productive medium containing 14% sucrose, 0.25% (NH(4))(2)CO(3), 0.25% KH(2)PO(4), and 0.025% MgSO(4).7H(2)O with addition of 0.06 mg/L CuSO(4).5H(2)O, 0.25 mg/L ZnCl(2), 1.3 mg/L FeCl(3).6H(2)O, pH 3.8, and incubated at 35 degrees C for 13 days by surface culture to produce 61.53 g/L anhydrous citric acid. Under the same conditions with a batchwise culture, it was found that immobilized conidia could maintain a longer period for citric acid production (31 days): over 70 g/L anhydrous citric acid from runs No. 2-4, with the maximum yield for anhydrous citric acid reaching 77.02 g/L for run No. 2. In contrast, free conidia maintained a shorter acid-producing phase, ca. 17 days; the maximum yield for anhydrous citric acid was 71.07 g/L for run No. 2 but dropped quickly as the run number increased.  相似文献

We have examined the effects of the enzyme inhibitors 2,4,6-trinitrobenzene sulfonic acid (TNBS) and 5,5'-dithiobis-2-nitrobenzoic acid (DTNB) on ethylene and CO2 production in apple and tomato fruit discs. In the past these inhibitors have been used to inhibit membrane bound enzyme systems in various animal tissues. The amino reactive inhibitor TNBS was shown to decrease ethylene production in tomato discs without affecting rates of respiration; similar results were obtained with apple. The effects of the sulfhydryl reactive inhibitor DTNB were not as clearcut as TNBS. There was little effect of DTNB on ethylene production in tomato discs, however, in apple discs ethylene production was significantly reduced. DTNB also reduced the respiration rate in apple discs, although not to the same extent as ethylene production. The inhibition of DTNB was reversed by a brief treatment with dithioerythritol. The results indicate that ethylene production takes place at the cell surface.  相似文献

Lipid profiles of conidia of Aspergillus niger and a fatty acid auxotroph   总被引:2,自引:0,他引:2  
Conidial lipids of the wild-type (V35) Aspergillus niger and its unsaturated fatty acid auxotroph (UFA2) were compared. The wild type contained lower levels (7.6%) of phospholipids and higher levels (28.4%) of glycolipids than the mutant (16.5 and 22.2%, respectively). Oleic (33.4%), linoleic (22.5%), palmitic (12.8%), stearic (7.4%), and linolenic (6.2%) were the main fatty acids of the wild type (V35). The mutant grew only in the presence of unsaturated fatty acid having at least one delta 9cis double bond, and its conidial fatty acid profile was influenced by the exogenous acid. Analyses of the fatty acids of UFA2 grown in the presence of different fatty acid supplements support the original view that the mutant is defective in delta 9-desaturase activity.  相似文献

Galactose in the furanoic conformation appears to be limited to bacteria and lower eukaryotes. Galactofuranoic (Galf)-containing glycoconjugates that occur in organisms pathogenic or allergenic to man are frequently antigenic and immunodominant. We have used an immunochemical approach, employing a monoclonal antibody that recognises Galf epitopes, to investigate the presence of Galf-containing glycoconjugates within conidia and conidiophores of Aspergillus niger. ELISA and immunofluorescence microscopy indicated that specific and saturable binding sites were found on both. Inhibition studies confirmed that this binding was to Galf-containing glycoconjugates. Interestingly, the conidiophore heads were particularly rich in these glycoconjugates. Western blotting identified a Galf glycoprotein of 150-200 kDa from disrupted conidia.  相似文献

Cholesterol and phospholipids remain tightly associated with the ferroxidase-II protein from human serum following extensive purification. Purified ferroxidase-II preparations show a consistent ratio of protein, phospholipid, and cholesterol. Thin-layer chromatographic analyses indicate that phosphatidyl choline accounts for 70% of the bound phospholipid. Treatment of purified ferroxidase-II with phospholipase C or A results in a loss of ferroxidase activity which parallels the hydrolysis of phospholipid. A lipid-depleted form of ferroxidase-II can be prepared by gel-filtration following treatment with phospholipase C. However, hydrolysis, not removal, of the lipid is sufficient for the loss of ferroxidase activity. These studies indicate that the bound lipid components are essential to the maintainence of the catalytic activity of ferroxidase-II.  相似文献

Data on the lipid composition and carbohydrate composition of Aspergillus niger conidia make it possible to characterize the individual germination stages and differentiate between the conidia capable of germination and those that lost the germination capacity. The following criteria are proposed: the ratio of phosphatidylcholine and phosphatidylethanolamine, the ratio of mannitol and arabitol, and the levels of sterols and free fatty acids. The role of these compounds in the biochemical background of cell transition from dormancy to active metabolism and their use as indices of the quality of inocula in biotechnological processes are discussed.  相似文献

Productivity of fungal cultures is closely linked with their morphologic development. Morphogenesis of coagulating filamentous fungi, like Aspergillus niger, starts with aggregation of conidia, also denominated as spores. Several parameters are presumed to control this event, but little is known about their mode of action. Rational process optimization requires models that mirror the underlying reaction mechanisms. An approach in this regard is suggested and supported by experimental data. Aggregation kinetics was examined for the first 15 h of cultivation under different cultivation conditions. Mechanical stress was considered as well as pH-dependent surface interaction. Deliberations were based on a two-step aggregation mechanism. The first aggregation step is only affected by the pH-value, not by the fluid dynamic conditions in the bioreactor. The second aggregation step, in contrast, depends on the pH-value as well as on agitation and aeration induced power input. For the given experimental set-up, agitation had much more influence than aeration. In addition, hyphal growth rate was determined to be the driving force for the second aggregation step.  相似文献

It is shown that the rate of sporogenesis, the appearance of low-molecular-weight thiols, and the activation of carbohydrate synthesis in spores adversely affected the viability of Aspergillus niger conidia during storage. Conversely, the prevalence of trehalose over mannitol and the absence of glycerol, erythritol, and glucose in carbohydrate composition facilitated viability of conidia. The data obtained are discussed in regard to the biochemical criteria that may be used to characterize quiescent state of fungi and retaining the viability of the inoculum.  相似文献

The enzymatic properties of intracellular myrosinase produced by Aspergillus niger AKU 3302 were investigated. Maximum activity occurred at pH 6.2, and the enzyme was stable in a pH range of 7.6 to 8.0 at 5°C for 24 hr. Optimum temperature was about 34°C. Enzyme activity was stimulated by copper (I), (II), manganese (II) and cobalt (II) and was inhibited by mercury (II) and stannous (II) ions. However, metal complexing agents and DFP had little effect, while PCMB was a strong inhibitor. In contrast to plant myrosinase, this enzyme was neither activated nor inhibited by L-ascorbic acid. Glucosides and δ-gluconolactone inhibited enzyme activity but sugars were ineffective. The Km value for sinigrin was 3.3 × 10?3 M and that for p-nitrophenyl β-glucoside was 1.5 × 10?3 M. The relation between fungous myrosinases and β-glucosidase is discussed in comparison to plant myrosinase.  相似文献

Aspergillus niger produces two extracellular glucoamylases (GAI of Mr 85 300 and GAII of Mr 77 600) separable on DEAE-cellulose. The enzymes differes in electrophoretic mobility, thermostability and substrate specificity. The GAI/GAII ratio depends on the concentration and form of nitrogen (nitrate or ammonium) in the culture medium. Proteinase VIII from Bacillus subtilis converts GAI to a form showing properties similar to those of GAII. Possible proteolytic degradation of GAI to GAII by Asp. niger endogenous proteinase(s) is suggested.  相似文献

Bacteriol. 91:1883-1887. 1966.-Trehalases (soluble trehalase and coat-bound trehalase) were found in the conidia of Aspergillus oryzae, and the total activity of the trehalases increased during the germination process. The soluble trehalase was purified by diethylaminoethyl-cellulose column chromatography; its optimal pH, Michaelis constant, and heat stability were studied. In vitro, the trehalases were competitively inhibited by d-mannitol, which was also contained in the conidia. Since the trehalose content in the conidia decreased at an early stage of germination, it was assumed that trehalase might begin to hydrolyze trehalose after the inhibitory effect of d-mannitol decreased.  相似文献

The effect of cations Fe2+ and Fe3+ on the decomposition of apple pectin by the enzymic preparations of Asp. niger and Asp. awamori has been examined. Fe ions have delayed the process of enzymic decomposition of the pectin molecule. In the presence of Fe3+ far less amounts of monogalacturonic acid are formed. The presence of Fe ions makes the pectin molecule more stable to the effect of pectolytic enzymes.  相似文献
