The ATP-dependent establishment of a positive membrane potential (measured as S14CN-accumulation) in membrane vesicles isolated from the roots of Atriplex nummularia Lindl. was not inhibited by NaMes and KMes at concentrations up to 140 millimolar. On the other hand, the formation of ΔpH (measured as 14C-methylamine accumulation or quenching of quinacrine fluorescence), was depressed by NaMes concentrations as low as 30 millimolar. Supply of NaMes after the ΔpH had been established brought about partial dissipation within 30 seconds. Extent of dissipation of ΔpH increased with NaMes concentration over the range tested (up to 180 millimolar). The H+/Na+ exchange indicated by these results was not due to the creation of a Na+ diffusion potential. Formation of ΔpH in these vesicles was stable to NO3 up to 100 millimolar; further, the dissipating effect of Na+ supply was apparent on a ΔpH formed in the presence of 30 millimolar NO3. Additional evidence that the origin of the membrane vesicles observed in this investigation was not the tonoplast and was probably the plasmalemma included the vanadate sensitivity of the establishment of the membrane potential.  相似文献

Summary Growth reduction or cessation is an initial response of Atriplex nummularia L. cells to NaCl. However, A. nummularia L. cells that are adapted to 342 and 428 mM NaCl are capable of sustained growth in the presence of salt. Cells that are adapted to NaCl exhibit a reduced rate of division compared to unadapted cells. Unlike salt adapted cells of the glycophyte Nicotiana tabacum L., A. nummularia L. cells do not exhibit reduced rate of cell expansion after adaptation. However, the cell expansion rate of unadapted A. nummularia L. cells is considerably slower than that of unadapted glycophyte cells and this normally low rate of cell expansion may contribute to the enhanced capacity of the halophyte to tolerate salt. Turgor of NaCl adapted cells was equivalent to unadapted cells indicating that the cells of the halophyte do not respond to salt by osmotic over adjustment as reported for the glycophyte tobacco (Binzel et al. 1985, Plant Physiol. 79:118–125).  相似文献

Membrane vesicles were isolated from the roots of the halophyte Atriplex nummularia Lindl. H+-translocating Mg2+-ATPase activity was manifested by the establishment of a positive membrane potential (measured as SCN accumulation); and also by the establishment of a transmembrane pH gradient (measured by quinacrine fluorescence quenching). H+-translocation was highly specific to ATP and was stable to oligomycin. Growing the plants in the presence of 400 millimolar NaCl doubled the proton-translocating activity per milligram of membrane protein and otherwise modulated it in the following ways. First, the flat pH profile observed in non-salt-grown plants was transformed to one showing a peak at about pH 6.2. Second, the lag effect observed at low ATP concentration in curves relating SCN accumulation to ATP concentration was abolished; the concave curvature shown in the double reciprocal plot was diminished. Third, sensitivity to K-2 (N-morpholino)ethanesulfonic acid stimulation was shown in salt-grown plants (about 40% stimulation) but was absent in non-salt-grown plants. Fourth, the KCl concentration bringing about 50% dissipation of ATP-dependent SCN accumulation was 20 millimolar for salt-grown plants and 50 millimolar for non-salt-grown plants. Vanadate sensitivity was shown in both cases. No clear NO3 inhibition was observed.  相似文献

The present study aimed to uncover the interconnected mechanisms underlying salt tolerance of Atriplex nummularia, a potential fodder plant for saline agriculture. The plants were grown in gravel/hydroponic quick check system in the greenhouse and irrigated with various seawater salinities (sws) (0, 25, 50, 100 and 150 %). Raising NaCl salinity stimulated the plant growth, which was highest at 50 % sws. Growth stimulation was mainly due to positive water balance, bestowed by plant ability to adjust osmotically and to minimize water loss via transpiration. Osmotic adjustment was mainly achieved by substantial accumulation of Na+ and Cl?. This was associated concurrently with sharp decrease in K+ and NO3 ? concentrations, resulting into ion imbalance. However, the plants were able to maintain adequate ion ratios in their roots and juvenile leaves, where the metabolic activities are expected to be highest. Salt-induced reduction in transpiration rates was coincided with progressive decrease in net photosynthesis (P N). This reduction was proportionally higher than those observed for photosynthesis, leading to improve photosynthetic water use efficiency (PWUE). Reduction in NO3 ? concentration may contribute to the overall reduction in total soluble protein (TSP), total N content and hence net photosynthetic rates. However, photosynthetic nitrogen use efficiency (PNUE) and nitrogen use efficiency (NUE) were transiently increased, peaked at moderate salinities, indicating that the plants could effectively regulate nitrogen utilization for C-assimilation machinery. Thus, plant’s ability to maintain carbon and nitrogen assimilation in equilibrium through well-coordinated regulatory mechanisms could be considered as a key determinant for salt tolerance in A. nummularia.  相似文献

Abstract Salt-stimulated ATPase activity in membrane preparations obtained from roots of Atriplex nummularia Lindl. at pH 5 was not suscep-tible to inhibition by KC1 or NaCl up to 450 mol m-3 but showed a broad peak of activity between 150 and 300 mol m?3. At pH 8 stimulation occurred at 50 mol m?3 but concentrations above 100 mol m?3 depressed activity below the level of the MgATPase activity. By contrast, preparations from roots of Pisum sativum L. at pH 5 showed maximal stimulation at 25 to 50 mol m?3 of NaCl or KC1; concentrations higher than 150 mol m?3 depressed activity below that of MgATPase activity. At pH 8 maximal stimulation was observed at 5 to 10 mol m?3 NaCl or KC1 while the threshold for inhibition was reduced to 15 mol m?3. With increasing salt concentrations the pH profiles for NaCl stimulation of Atriplex ATPase activity (expressed as the difference between treatment and control) showed a progressive displacement of the apparent optimum towards lower pH. The shift was not apparent when stimulation was expressed as a percentage of MgATPase activity. This shift may be accounted for if NaCl stimulated the monovalent salt-activated ATPase activity but simultaneously inhibited MgATPase activity.  相似文献

Arabis stelleri var.japonica evidenced stronger osmotic stress tolerance than Arabidopsis thaliana.Using an A.thaliana microarray chip,we determined changes in the expression of approximately 2 800genes between A.stelleri plants treated with 0.2 M mannitol versus mock-treated plants.The most significant changes in the gene expression patterns were in genes defining cellular components or in genes associated with the endomembrane system,stimulus response,stress response,chemical stimulus response,and defense response.The expression patterns of three de novo proline biosynthesis enzymes were evaluated in A.stelleri var.japonica seedlings treated with 0.2 M mannitol,0.2 M sorbitol,and 0.2 M NaCl.The expression of Δ1-pyrroline-5-carboxylate synthetase was not affected by NaCl stress but was similarly induced by mannitol and sorbitol.The proline dehydrogenase gene,which is known to be repressed by dehydration stress and induced by free L-proline,was induced at an early stage by mannitol treatment,but the level of proline dehydrogenase was increased later by treatment with both mannitol and NaCl.The level of free L-proline accumulation increased progressively in response to treatments with mannitol,sorbitol,and NaCl.Mannitol induced L-proline accumulation more rapidly than NaCl or sorbitol.These findings demonstrate that the osmotic tolerance of the novel halophyte,Arabis stelleri,is associated with the accumulation of L-proline.  相似文献

Atriplex nummularia is a halophyte widely employed to recover saline soils and was used as a model to evaluate the water potentials in the soil-plant system under drought and salt stresses. Potted plants grown under 70 and 37% of field capacity irrigated with solutions of NaCl and of a mixture of NaCl, KCl, MgCl2 and CaCl2 reproducing six electrical conductivity (EC): 0, 5, 10, 20, 30, and 40 dS m?1. After 100 days, total water (Ψw, plant) and osmotic (Ψo, plant) potentials at predawn and midday and Ψo, soil, matric potential (Ψm, soil) and Ψw, soil were determined. The type of ion in the irrigation water did not influence the soil potential, but was altered by EC. The soil Ψo component was the largest contributor to Ψw, soil. Atriplex is surviving ECs close to 40 dS m?1 due to the decrease in the Ψw. The plants reached a Ψw of approximately ?8 MPa. The water potentials determined for different moisture levels, EC levels and salt types showed huge importance for the management of this species in semiarid regions and can be used to recover salt affected soils.  相似文献

Atriplex nummularia plants are able to grow well in the absence of significant amounts of Na+. Medium levels of salinity (100 mM NaCl or KCl) did not cause substantial inhibition of growth but increasing concentrations of salt induced a progressive decline in length and weight of the plants. This inhibition was significantly higher in KCl grown plants than in NaCl grown plants. In addition, although it has been proposed that both K+ and Na+ are involved in the osmotic adjustment of plants in response to high soil salinity, we show that Na+ ions contribute more efficiently than K+ ions to perform this function. Our results also indicate that most of the osmotic adjustment of the plant was due to the accumulation of inorganic ions. The strong inhibition of Rb+ transport caused by internal sodium suggests that this cation could be efficiently used by the plant and, as a consequence, the transport of other monovalent cations is down-regulated.  相似文献

Chenopods are generally regarded as non-host plants for mycorrhizal fungi and are believed not to benefit from colonization by mycorrhizal fungi. Perennial Atriplex nummularia Lindl., growing under field conditions, showed a relatively high level of colonization by mycorrhizal fungi (10–30% of root length colonized) in spring and summer. Accordingly, two glasshouse experiments were designed to assess the effects of inoculation with mycorrhizal fungi (with a single species or a mixture of different species) on growth, nutrient uptake, and rhizosphere bacterial community composition of A. nummularia at high and low salinity levels (2.2 and 12 dSm–1). Only low and patchy colonization by mycorrhizal fungi (1–2 of root length colonized) was detected in inoculated plants under glasshouse conditions which was unaffected by salinity. Despite the low colonization, inoculation increased plant growth and affected nutrient uptake at both salinity levels. The effects were higher at an early stage of plant development (6weeks) than at a later stage (9–10 weeks). Salinity affected the bacterial community composition in the rhizosphere as examined by ribosomal intergenic spacer amplification (RISA) of 16S rDNA, digitization of the band patterns and multivariate analysis. The effects of inoculation with mycorrhizal fungi on growth of A. nummularia may be attributed to (i) direct effects of mycorrhizal fungi on plant nutrient uptake and/or (ii) indirect effects via mycorrhizal-induced changes in the bacterial community composition.  相似文献

The choice of the best species to cultivate in semi-arid and arid climates is of fundamental importance, and is determined by many factors, including temperature and rainfall, soil type, water availability for irrigation and crop purposes. Soil or water salinity represents one of the major causes of crop stress. Species of the genus Atriplex are characterized by high biomass productivity, high tolerance to drought and salinity, and high efficiency in use of solar radiation and water. Based on a search of the international literature, the authors outline an agro-climatic zoning model to determine potential production areas in Argentina for Atriplex halimus and Atriplex numularia. Using the agroclimatic limits presented in this work, this model may be applied to any part of the world. When superimposed on the saline areas map, the agroclimatic map shows the suitability of agro-ecological zoning for both species for energy purposes on land unsuitable for food production. This innovative study was based on the implementation of a geographic information system that can be updated by further incorporation of complementary information, with consequent improvement of the original database.  相似文献
