【目的】 观察伯氏嗜木螨Caloglyphus berlesei生活史各发育阶段外部形态和超微形态特征。【方法】 采集中华地鳖Eupolyphaga sinensis培养床上的培养料, 分离伯氏嗜木螨。将分离获得的伯氏嗜木螨双蒸水洗涤后, 一部分用于制备常规玻片标本, 光镜下直接观察; 另一部分用2.5%戊二醛溶液固定, 70%酒精再次洗涤, 临界点干燥, 置于扫描电镜下观察不同发育阶段(包括卵、 幼螨、 若螨及成螨)的超微形态特征。【结果】 电镜下伯氏嗜木螨各发育阶段的足、 刚毛和外生殖器及其附属结构的形态均清晰可辨。幼螨足3对, 足上无叶状刚毛, 基节干发达; 若螨足4对, 出现第4背毛, 生殖区发育不全; 休眠体足爪和前跗节发达, 出现叶状毛、 胫节毛及膝节毛等结构, 生殖板骨化明显, 其两侧有吸盘和刚毛各1对。吸盘板上共有4对成对的吸盘、 1个单吸盘和2对类圆形微凸。成螨生殖感觉器骨化且呈心形, 雄雌成螨生殖感觉器的刚毛数量上有明显差异。【结论】 对伯氏嗜木螨形态和超微形态特征的观察有助于对其进一步科学分类和进行生活史研究, 并可为控制伯氏嗜木螨及其引起的过敏性疾病提供参考。  相似文献

We here report the case of a storage mite, Sancassania berlesei, infestation in the external auditory canal of a 46-yr-old male. He complained of feeling a foreign body and itching in the left external auditory canal for 1 mo, with accompanying otalgia for 3 days. Considering the duration of the patient's complaint and the 8-9-day life cycle of the mite, the mites are believed to have lived in the patient's ear for more than 3 generations.  相似文献

Meloidogyne polycephannulata n. sp. is described from specimens collected from an area cultivated with carrot cv. Brasilia, in the city of Rio Paranaíba, in the region of Alto Paranaíba, Minas Gerais State, Brazil. The perineal pattern of the female is circular to ovoid with a high dorsal arch that has widely spaced, coarse annulations. The lateral field may have a deep furrow separating the dorsal and ventral arches. The medial lips are short and wide, whereas the lateral lips are large and triangular. The female stylet is 15-16 μm long with wide knobs, distinctly divided by an indentation in the center. Its tip is slightly curved dorsally. The excretory pore opens 34-65 μm from the anterior end. Females retain eggs and second-stage juveniles in their body cavity, similar to that of the cyst-forming nematodes. Males are 1.3-1.7 mm long and have a high head cap that is rounded and slopes posteriorly. The labial disc is fused to the medial lips. The head region has several irregular annulations that are similar in appearance to the first or second body annules that are likewise irregular, making the head region appear to be extremely large. The stylet of the male is 21-24 μm long; it is slender, and has small, rounded knobs, that are distinctly indented medially and appear heart-shaped. The shaft has several tiny projections throughout its length. Mean second-stage juvenile length is 411.7 μm. The juvenile head cap is elevated, the medial lips are small, and the lateral lips are elongate to triangular-shaped. The head region has several short, incomplete and irregular transverse annulations. The juvenile stylet is 14-23 μm long with small, rounded, and sloping knobs. The thin tail ends with a short hyaline portion that is variable in size (16-26 μm) and with a small, rounded tip. Isozyme profiles of esterases from Meloidogyne javanica show 3 strong bands (SB) at Rm 46, 59, and 66; profiles of M. polycephannulata n. sp. show a SB at Rm 47 and a weak band (WB) at Rm 52; M. petuniae has two SB at Rm 44 and 53; M. phaseoli has a SB at 53, 58, and 64 Rm; M. brasilensis has three SB at Rm 40, 58, and 66 and a WB at Rm 71; M. pisi has a SB at Rm 40, 60, and 64 and two WB at 46 and 50 Rm. Data from sequencing the 18S rDNA region of M. polycephannulata n. sp. confirms that it is different from M. arabicida, M. arenaria, M. ethiopica, M. incognita, M. javanica, M. paranaensis, and M. thailandica. Sequence identity among these eight species ranged between 85 to 93.4%. Meloidogyne polycephannulata n. sp. reproduces very well on carrot and tomato; poorly on pepper; and not at all on cotton, peanut, tobacco, watermelon, and sweet corn.  相似文献

The root-knot nematode Meloidogyne ulmi is synonymised with Meloidogyne mali based on morphological and morphometric similarities, common hosts, as well as biochemical similarities at both protein and DNA levels. M. mali was first described in Japan on Malus prunifolia Borkh.; and M. ulmi in Italy on Ulmus chenmoui W.C. Cheng. Morphological and morphometric studies of their holo- and paratypes revealed important similarities in the major characters as well as some general variability in a few others. Host test also showed that besides the two species being able to parasitize the type hosts of the other, they share some other common hosts. Our study of the esterase and malate dehydrogenase isozyme phenotypes of some M. ulmi populations gave a perfectly comparable result to that already known for M. mali. Finally, phylogenetic studies of their SSU and LSU rDNA sequence data revealed that the two are not distinguishable at DNA level. All these put together, leave strong evidences to support the fact that M. ulmi is not a valid species, but a junior synonym of M. mali. Brief discussion on the biology and life cycle of M. mali is given. An overview of all known hosts and the possible distribution of M. mali in Europe are also presented.  相似文献

The free-living mite species Sancassania rodionovi (Zachvatkin) (Acari: Acaridae), is a serious pest of mushrooms in Iran. Studies were conducted to examine the development of this mite in relation to temperature on two mushroom species: Agaricus bisporus Lange (button mushroom) and Pleurotus ostreatus Kummer (oyster mushroom). The developmental time of this acarid mite was studied at eight constant temperatures, ranging from 5 to 40 degrees C, and developmental rates were modeled as a function of temperature. Sancassania rodionovi completed immature development in 17.35 +/- 0.58 and 20.17 +/- 0.88 d at 25 degrees C on button and oyster mushrooms, respectively. When the mite fed on button mushroom, the rate of development increased gradually from 10 to 35 degrees C. Using a linear model, the developmental zero was estimated to be 3.50 degrees C with a thermal constant of 357.14 degree-days. The Logan 10, Briere 1, and Thermodynamic models adequately described the data for this mite and yielded R2 values >0.95; these models provided estimates of optimum temperature for development of 33.244, 32.145, and 32.148 degrees C, respectively. Understanding the influence of temperature on development of S. rodionovi is discussed with respect to pest management in mushroom production.  相似文献

Total protein variation as revealed by two-dimensional electrophoresis (2D-E) was studied in 18 isolates from populations of Meloidogyne arenaria (six isolates), Meloidogyne incognita (10 isolates), and Meloidogyne javanica (one isolate) plus an unclassified isolate. Gels (80 x 60 x 0.75 mm) were silverstained and digitized in order to compare their protein patterns. Optical density and position of protein patterns were measured using statistical cluster analysis and computer-assisted image analysis software. Only those protein stains or positions that were clearly defined (i.e., without background) were considered. The number of positions in gels ranged from 86 to 203. Each of these positions had 95 clearly expressed proteins that were present in at least two replicates for each isolate. Spot position was considered a taxonomical character with two different states: presence (1) and absence (0). Accordingly, genetic distance was estimated among isolates and species, and a phylogenetic tree was constructed following the cladistic approach based on maximum parsimony analysis. Isolates of M. arenaria--M. javanica--Meloidogyne sp. and of M. incognita formed two separate monophyletic groups. Both groups were clearly defined on the basis of two sets of protein positions that can be considered as diagnostic characters. An attempt to identify these proteins by mass spectrometry was made. Group diagnostic proteins for M. incognita and M. arenaria (and for other proteins common to all isolates) were distinguished by protonated mass signals in the MALDI fingerprinting spectrum.  相似文献

A study was made of the effects of concentrations of 2 to 32 ppm of oximecarbamate, organophosphate and benzimidazole nematicides on the hatch, larval viability and migration of Meloidogyne javanica, M. incognita and M. hapla and on development of M. javanica in roots. Aldicarb at less than 8 ppm had little effect on hatch and methomyl markedly affected only the hatch of M. hapla. As little as 2 ppm of fenamiphos or thionazin markedly reduced hatch of all three species but less than 8 ppm ethoprophos significantly reduced only the hatch of M. incognita and phorate had little effect on hatch. Benomyl and thiabendazole had no significant effects on hatch. When egg masses of M. incognita were transferred from nematicides which suppressed hatch to water, hatching occurred, but aldicarb, fenamiphos, ethoprophos and thionazin significantly reduced total hatch. None of the nematicides killed larvae of the three species immersed in 16 and 32 ppm solutions of them for 3 days. Aldicarb at 2 ppm reduced migrations of all three species; the effects of methomyl, fenamiphos or thionazin on migration varied according to species, while phorate, ethoprophos, benomyl or thiabendazole had little or no effect on migration. Aldicarb or thionazin at 2 ppm stopped development of M. javanica in roots of tomato seedlings while methomyl, ethoprophos or fenamiphos at 4 ppm reduced development by 60% and at 8 ppm of ethoprophos or fenamiphos or 16 ppm of methomyl, development was stopped. Phorate had little effect on development and benomyl or thiabendazole had no effect. Nematicide concentrations which reduced development prevented the normal orientation of larvae in the roots and reduced or prevented giant cell formation.  相似文献

Caloglyphus (= Sancasania) polyphyllae discharges from a pair of opisthonotal glands a characteristic set of volatiles, i.e. three monoterpenes and seven hydrocarbons. Among them, beta-acaridial, which is known as the female sex pheromone of the species and has antifungal activity, was newly identified as the aggregation pheromone for unfeeding and unmating mites. Feeding mites, however, exhibited sexually aroused behavior instead of the tendency to cluster when exposed to beta-acaridial. This is the first example of the compound demonstrating two pheromone functions depending upon the circumstances faced by the mites.  相似文献

Meloidogyne hispanica n. sp. is described and illustrated from specimens obtained from peach rootstock, Prunus persica silvestris Batsch, from the Seville district of Spain. The perineal pattern of the female is oval shaped to rectangular with low dorsal arch and often widely spaced lateral lines with fringe-like striae. The stylet, 14.1 μm long, has broad, distinctly set off knobs. Males have a high, rounded head cap that slopes posteriorly. Labial disc and medial lips are fused to form elongate lip structures. The robust styler, 23.5 μm long, has large, rounded knobs that are slightly set off from the shaft. Mean second-stage juveniles length is 392.6 μm. The truncate head region is generally not annulated. The distinctly rounded and raised labial disc and the crescent-shaped medial lips form dumbbell-shaped lip structures. The stylet, 11.1 μm long, has rounded, posteriorly sloping knobs. The slender tail, 46.4 μm long, has large irregular-sized annules in the posterior region and ends in a bluntly rounded tip. Tomato was a good host; tobacco, pepper, and watermelon were poor hosts; cotton and peanut were nonhosts. Meloidogyne hispanica n. sp. reproduces by mitotic parthenogenesis and has a somatic chromosome number of 2n = 33-36. The esterase pattern is unique among Meloidogyne species.  相似文献

Specimens of Meloidogyne marylandi from Bermuda grass and a population from Zoysia grass were examined and compared morphologically by light and electron microscopy. The populations probably are conspecific and the differences noted in the Zoysia population, mainly those of second-stage juveniles (J2) with shorter tails, are considered normal variations rather than representing another form. Scanning electron microscope observations provided additional details of the perineal pattern and head of females and head and lateral fields of second-stage juveniles. Relationship of M. marylandi to closely related species is given. This species is currently known to occur only in Maryland, and populations previously reported from this state as M. graminis are now considered to be M. marylandi. Other reports of M. graminis in the United States now need to be reconfirmed by examination of voucher or recollected specimens.  相似文献

Sancassania polyphyllae (Acari: Acaridae) is associated with larvae of the white grub, Polyphylla fullo (Coleoptera: Scarabaeidae), and will feed on the infective juveniles of entomopathogenic nematodes in the families Steinernematidae and Heterorhabditidae which are important biological control agents of soil insect pests. We conducted laboratory studies to determine the potential negative effects this mite species might have on biological control of soil insect pests. Our objectives in this study were to (1) determine the response of S. polyphyllae adult mites to a nematode-killed insects on agar, (2) evaluate the predation by mites on Steinernema feltiae infective juveniles from nematode-killed insects on agar and in soil, and (3) assess predation efficiency of the mite on the infective juveniles in the soil. On agar, we found (1) significantly more adult female mites near or on a nematode-killed Ceratitis capitata (Diptera: Tephritidae) larva than near or on the freeze-killed larva or a bamboo mimic suggesting that a chemical or an odor from the nematode-killed larva attracted the mites, and (2) 10 mites consumed 96% of infective juveniles that emerged from an insect cadaver. In soil with a nematode-killed insect, the average number of infective juveniles recovered was <30 when mites were present, whereas the average number of infective juveniles recovered was >375 when the mites were absent. When the infective juveniles alone were placed in different depths in relation to the mites in the soil column for 4 and 10 days, S. polyphyllae was not as efficient at finding the infective juveniles when they were separated from each other in the soil lending support to the idea that the mites were cueing in on the cadaver as a food resource. Our data suggest that emerging infective juveniles from an insect cadaver in the soil in the presence of S. polyphyllae can adversely affect biological control because of nematode consumption by the mites.  相似文献

Summary Free-living nematodes may be attacked and eaten by soil mites which are normally considered fungivores or saprophages. Three species in the genus Tyrophagus, common inhabitants of grassland soils and also common pests in stored products, museums and laboratories, are predators of nematodes. All active stages of the mites will voraciously consume nematodes. When offered nematodes and a choice of other food (baker's yeast and algae), 11% of the Tyrophagus putrescentiae, 23% of the T. zachvatkini, and 56% of the T. similis tested fed on nematodes. Tyrophagus zechvatkini and similis were reared on a diet consisting entirely of nematodes, and developed at rates similar to a fungal diet and produced viable offspring. Agar cultures of Aphelenchus avenae which were inoculated with five mating pairs of Tyrophagus zachvatkini had populations that were one-third less than mite-free controls. Observations indicate that nematodes may be attacked by tyrophagid mites when in a dry, anhydrobiotic state.  相似文献

Total protein variation (up to ninety-five different positions) was revealed by two-dimensional electrophoresis (2-DE) in 18 isolates from populations of M. arenaria (6 isolates), M. incognita (10), M. javanica (1) plus an unclassified isolate in a previously reported study. Isolates of M. arenaria, M. javanica, Meloidogyne sp., and M. incognita formed two separate groups defined on the basis of two sets of protein positions that could be considered as diagnostic characters, but we could not identify these proteins by MALDI-TOF. To identify these marker positions, nano-liquid chromatography as peptides separation method was coupled to an ion-trap mass spectrometer for induced real-time fragmentation of eluted peptides. Group diagnostic proteins for M. incognita and M. arenaria were in-gel digested and on line analyzed by tandem mass spectrometry (LC-MS/MS). Six proteins out of seven selected spots were unambiguously identified by the analysis of the corresponding MS/MS (MS2) spectrum from parent ions fragmentation: Actin, Enolase, CG3752-PA protein similar to Aldehyde Dehydrogenase, HSP-60 and Translation initiation factor elF-4A. In M. incognita sample, de novo sequencing experiment of doubly charged ion at m/z=936.9 Da in spot 29 identified as enolase, reveals three residue substitutions (K to T, N to T, and D to E) when tentative sequence was compared with that of Anisakis simplex and Onchocerca volvulus enolase, thus three SNPs (single nucleotide polymorphisms) were also possibly identified.  相似文献

A new root-knot nematode, Meloidogyne aquatilis n. sp., attacking the roots of Spartina pectinata Link growing in the Ottawa River is described and illustrated. Meloidogyne aquatilis is distinguished from M. graminis by the light brown body color and by the absence of perineal lateral fields in the female. The male differs by the shorter stylet and by the hemizonid being separated by 7-9 annules from the excretory pore. The second-stage juveniles are also recognized by the 7-9-annule gap between the hemizonid and excretory pore and by the shorter tail with a disc-like subterminal tail structure, lower b ratio value, and inflated rectum. A key to the root-knot nematode species of Canada based on females, males, and juveniles is provided. The type host of Dolichodera fluvialis Mulvey and Ebsary, 1980, Spartina pectinata Link, is reported for the first time.  相似文献

A simple synthesis of beta-acaridial [(E)-1], the active principle of the sex, alarm and aggregation pheromone among astigmatid mites, was achieved in 5 steps from 1,2,4-butanetriol 2 in a 19% overall yield. Its analog, beta-acariolal 8, was also prepared in a 63% yield by oxidation of the intermediate, beta-acaridiol [(E)-7], with pyridinium dichromate (PDC). This synthetic route also gave beta-(Z)-acaridiol [(Z)-7] by using a Z-selective base in the Wittig reaction. (Z)-7 was oxidized to give a new monoterpene, beta-(Z)-acaridial [(Z)-1], which was detected as a trace component in the secretion of Caloglyphus polyphyllae, together with 8.  相似文献

Fecundity, longevity and survival to adulthood ofAcarus siro (L.) at constant and alternating temperatures were compared. Both fecundity and longevity were affected by alternating temperatures but the effect of the frequency of alternations was not significant. Significantly higher fecundity and longer life spans were recorded at constant temperatures of 14 and 21°C than at 28°C or at alternating temperature regimes. Alternating temperature regimes in comparison to constant regimes did not significantly change the number of males and females surviving to adulthood or the sex ratio. However, the lowest number of adults surviving was recorded when temperatures were changed every 12 h and the highest at a constant temperature of 14°C.  相似文献

Meloidogyne christiei n. sp. is described and illustrated from turkey oak (Quercus laevis) in Sanlando Park, Altamonte Springs, Florida. This new nematode species has a distinctive perineal pattern commonly with a high, squarish arch and coarse broken striae which tend to diverge at various angles, especially in and above the anal area. Female labial disc is indented, forming four points or prongs, unlike other species. Eggs are deposited inside the gall in a tubular, coiled manner. Vaginal muscles are exceptionally prominent and dense. SEM observations provided further detail of the perineal pattern and details of the head of females, males, and second-stage juveniles. Galls on the root commonly occur singly, but sometimes in small clusters, and appear as discrete nodules on the side of the root and without adjacent swelling. In general, only one female is found in each gall but occasionally two are present. In greenhouse tests, citrus, tobacco, cotton, pepper, watermelon, peanut, and tomato were not hosts. This nematode occurs throughout central Florida commonly on Q. laevis, the only known host.  相似文献

The present study was conducted to evaluate sublethal effects of B-azolemiteacrylic on the two-spotted spider mite, Tetranychus urticae Koch (Acari: Tetranychidae). Female adults of T. urticae were exposed to LC10 and LC30 of the acaricide, and the effects on treated females and their offspring were evaluated. The results showed that the fecundity of F0 female adults treated with LC10 and LC30 of B-azolemiteacrylic was reduced by 30.9 and 39.2%, respectively. Longevity and oviposition period of the females were significantly reduced as well. The developmental duration of egg and deutonymph stage of the F1 generation were not significantly different from that of the control. The protonymph stage after LC30 treatment lasted significantly longer, whereas the larva, deutonymph and female stage were significantly shorter than the control. The oviposition period of the F1 generation was significantly shortened, the fecundity of each female decreased significantly, and the ratio of female-to-male was reduced too. Moreover, the average generation period of T. urticae after LC10 and LC30 treatments was shorter than that of the control, and the net production rate (R0), intrinsic rate of increase (rm) and finite rate of increase (λ) were all reduced by 33.3, 7.5 and 1.9% (LC10 treatment) and by 51.3, 14.8 and 3.6% (LC30 treatment), respectively. The population doubling time was prolonged by 7.5 and 14.8% after LC10 and LC30 treatments, respectively, compared with the control. These results indicate that B-azolemiteacrylic may effectively inhibit the development rate of the F0 and F1 populations of T. urticae, which will help design integrated strategies for the comprehensive control of T. urticae and rational use of pesticides in the field.
