Summary The present paper describes for the first time the fine structure of ovarian granulosa cells as seen after administration of clomiphene to the domestic fowl for a 28 days period.The main cytoplasmic changes of the granulosa cells seemed to be an increase in the number of mitochondria, dense bodies and complex bodies. The Golgi apparatus became enlarged, and there was an increase in the endoplasmic reticulum, annular desmosomes and cytoplasmic processes.All the observations made are similar to those made after administration of steroids and gonadotropins. In conclusion, therefore, the present study has demonstrated that administration of clomiphene exerts a stimulating effect on the granulosa cells. The mechanism of this effect is discussed. 相似文献

Summary The fine structure of granulosa cells of the domestic fowl as seen after administration of steroids is described. Diaethylstilboestrol, estradiol and hydroxy-progesterone were given as intramuscular injections for a 28-days period. The main cytoplasmic changes of the granulosa cells were an increase in the number of mitochondria and dense bodies. The Golgi apparatus became enlarged, and occupied a large portion of the cell. The nucleus was found located adjacent to the oocyte, and there was an increase in number of the annular desmosomes. The investigation has demonstrated that even if steroids in high dosages induce atrophic alterations of the ovary, they stimulated proliferation of the granulosa cells of the small follicles. Apparently the small follicles with the granulosa cells retain the ability to regain development, which may be to some importance when steroids are used therapeutically (gynecologic disorders, contra-ception). 相似文献

Summary Pig oocytes and their surrounding granulosa cells obtained from mature Graafian follicles at a stipulated time near to ovulation were studied in some details electronmicroscopically. Particular emphasis is given to the corona radiata cell processes and to the heterogeneous population of mitochondria in the oocyte.The corona radiata cell processes contain various components such as filaments, mitochondria, multivesicular bodies and lipid droplets in their matrix. The contact relationship of the corona radiata cell processes to the oocytes is maintained by desmosomes. Usually, the two parallel surface membranes forming the desmosome are separated by a space of about 200 Å. Occasionally, the two membranes approximate each other to form a junction having a gap of about 70 Å. Apparently the membranes become fused in some regions.Of particular interest is the distribution and structural characteristics of the single-membrane-bounded structures, and their relationship to the cytomembranes and the mitochondria. On the basis of the present and earlier (Norberg, 1972) observations, the question arises whether the formation and development of mitochondria of pig oocytes depend, at least partly, on a metamorphosis of single-membrane-bounded structures derived from less complex membraneous elements. Final conclusions concerning this problem demand integrated morphological and biochemical investigation regarding the biosynthesis of mitochondria.This work was supported by the Agricultural Research Council of Norway. 相似文献

Direct production of gonadal steroids from sulfated adrenal androgens may be an important alternative or complementary pathway for ovarian steroidogenesis. The conversion of sulfated adrenal androgens, present in serum at micromolar concentrations in adult women, into unconjugated androgens or estrogens requires steroid sulfatase (STS) activity. STS activity has not been characterized in the rat ovary. Substantial STS activity was present in homogenates of rat ovaries, primary cultures of rat granulosa cells, and a granulosa cell line, as determined by conversion of radiolabeled estrone sulfate (E1S) to unconjugated estrone. The potent inhibitor estrone sulfamate eliminated the STS activity. Using E1S as a substrate with microsomes prepared from a granulosa cell line, the Km of STS activity was approximately 72 μM, a value in agreement with previously published data for rat STS. Therefore, ovarian cells possess STS and can remove the sulfate from adrenal androgens such as dehydroepiandrosterone sulfate (DHEA-S). Using DHEA-S as a steroidogenic substrate represents an alternative model for the production of ovarian steroids versus the “two cell, two gonadotropin” model of ovarian estrogen synthesis, whereby thecal cells produce androgens from substrate cholesterol and granulosa cells convert the androgens into estrogens. The relative contribution of STS activity to ovarian steroidogenesis remains unclear but may have important physiological and pathophysiological implications. 相似文献

Summary The structure of follicular layer of growing and atretic follicles in the ovary of the domestic goose, was studied by electron microscopy. In small follicles, the wall is lined with a narrow layer of tightly packed small, cuboidal cells separated from the thecal tissue by the basal lamina. During growth, they transform into tall, columnar cells arranged in a single row. The cells display several peculiar ultrastructural features. First, annulate lamellae are commonly observed. Second, cytoplasmic dense-cored granules accumulate in close association with fenestrated cisternae and networks of tubuli derived from the RER. They consist of spheres and strands of amorphous substance of unknown origin. Third, the cells contain many transosomes, a unique organelle of the avian follicle cell consisting of a dense plaque associated with ribosome-like particles. The mature forms of transosomes are located at the tips of lateral and apical cell projections, while bodies thought to be their precursors, are found in the apical cytoplasm. In follicles larger than 8 mm in diameter, most of the transosomes and their precursors have disappeared. Follicular atresia occurs in all of the size-classes of follicles investigated. A loss of transosomes (in follicles up to 8 mm in diameter) and an accumulation of lipid droplets are the first atretic events detectable by electron microscopy. Morphologic features, including deep nuclear indentations, accumulation of lipid droplets frequently encireled by membrane whorls, dilation and disintegration of RER cisterns, swelling of mitochondria and accumulation of dense irregular masses of unknown origin in the cytoplasm, are taken as evidence for advanced degradation. We conclude that necrosis is the dominant type of cell death of the follicular cells during atresia. However, a small fraction of cells, characterized by dark condensed cytoplasm, seems to die by apoptosis. 相似文献

In the present paper, we have studied the expression of the Phosphatase and TENsin homolog deleted on chromosome 10 (PTEN) and its putative biological role in the sheep ovary. We found by Northern-blot, immunohistochemistry and immunoblot that PTEN is highly expressed in granulosa cells from large differentiated follicles (LF) in comparison with small proliferating follicles (SF) (P < 0.001), with no clear effect of follicle quality. Moreover, the PTEN lipid phosphatase activity is also higher in LF than in SF (P < 0.01). In contrast, levels of the phosphorylated form of AKT (pAKT) are lower in LF than in SF (P < 0.0001). IGF-I and insulin but not FSH, LH or forskolin are able to stimulate the expression of PTEN mRNA (P < 0.001) and protein by ovine granulosa cells after 48 h of culture in vitro. An IGF-1 time course analysis showed that expression of PTEN protein appeared after 12h of culture, concomitant with the fall of the pAKT levels, which peaked after 6h of stimulation with IGF-I. Moreover, transfection experiments showed that overexpression of PTEN in ovine granulosa cells induced a decrease and an increase in E2F and p27 promoter activity, respectively (P < 0.05). Overall, our present data show for the first time that the expression of PTEN increases during terminal follicular growth. This increase, that might be induced by IGF-I but not FSH, would participate in the proliferation/differentiation transition of ovine granulosa cells in differentiating follicles. 相似文献

Follicular growth, development and ovulation are highly ordered processes that involve the expression of many genes under precise temporal and spatial regulation. However, information on stage-specific gene expression during the antral follicle phase in sheep is not well understood. In the present study, suppressive subtractive hybridization (SSH) was performed to screen genes that were differentially expressed in the granulosa cells between large follicles (LF, >5 mm) and small follicles (SF, 3–5 mm), and subtractive cDNA library was constructed. Furthermore, with dot-blot analysis, a total of 90 clones randomly selected from the library were proven to be differentially expressed in the granulosa cells. Among these, 38 exhibited high homology to known genes, 14 sequences were corresponding to novel expressed sequence tags (ESTs). Four ESTs, LAPTM4A, SERPINE2, GSTA1, and INHBA, were further examined the reproducibility of the SSH data by the real-time quantitative PCR. Results confirmed an increase expression of respective mRNA in granulosa cells of large follicles compared with that of small follicles. It is concluded that we have identified several genes (known or unknown) that may effect follicular growth, dominance or ovulation in ewes. 相似文献

Summary The purpose of this study is to investigate the ultrastructural features of perivascular cells as found in the neural lobe of the hypophysis. In particular, an inquiry was made into the nature of the relationship of such cells to neurosecretory fibers and endings. The latter, in fact, are often invaginated within the perivascular cells and enveloped by their processes; furthermore, they often reveal a certain number of empty granules as well as characteristics of degenerative nature. In the course of this study the localization of the perivascular cells has been investigated as well as that of their processes within the extensive interlobular network typical of the hypophysial neural lobe of rodents. Based on the data gathered, the hypothesis is put forward that the perivascular cells play an important role in the turnover of neurosecretory endings, both under physiological and experimental conditions, contributing thereby to the release of post-hypophysial hormones.With the technical assistance of Vincenzo Panetta. 相似文献

Summary The ultrastructure of the polymorphic vesicular component of the ultimobranchial gland of the domestic fowl (Gallus gallus domesticus) has been described in detail, together with the structure of the cell strands interconnecting the vesicles and the parathyroid nodules lying within the ultimobranchial stroma. The vesicles frequently appear to arise from the nodules by way of the cell strands. The strands show a structure of their component cells intermediate between that of the parathyroid and the vesicular cells, although the position at which the strand changes from an essentially parathyroid structure to an essentially vesicular structure is very variable. The degree and kind of secretory activity within different cell types has been described. A review of the structure of ultimobranchial glands throughout the vertebrates shows that similar tissue with a similar secretory potential has been observed in all vertebrate classes, suggesting a functional significance for this part of the gland. 相似文献

Summary The ultrastructure of the lutein cells in the mink throughout pregnancy and the regression periodpost partum is described. To correlate the fine structure with the changes in the peripheral plasma progesterone levels, the concentrations of progesterone were measured by a rapid competitive protein-binding assay.Even during the delay period (e.g. as long the plasma progesterone levels remain at the basal level, <8ng/ml), the lutein cells in the mink exhibit structural criteria of functional activity. However, the increase in progesterone secretion is accompanied by some morphological transformations, characterized by the presence of more and more small dense homogenous bodies in the cytoplasm, which become irregular and scalloped during the stage with maximum release of progesterone. At this stage the agranular endoplasmic reticulum is often cisternal or vesicular.During the decline of the progesterone levels, typical and moderate electron-dense lipid droplets are found increasingly more within the lutein cells. The expanded agranular ER is now more sparse, while the granular ER becomes more pronounced, often forming parallel arrays. During this phase the mitochondria become elongated, dumb-bell, or cup shaped. After parturition the corpora lutea consist of cells in various stages of degeneration. At day 14post partum only a few lutein cells are still identifiable.Evidently the observed morphological changes take place in the lutein cells during the life span of corpora lutea. This feature lends further support to the concept that the mink lutein cells are steroid-producing cells and furthermore, that the corpora lutea may be the main sites of gestagen production during pregnancy. 相似文献

Summary In an attempt to identify pancreatic islet cells emitting formaldehyde-induced fluorescence (FIF), the pancreatic islets of the domestic fowl were studied by combined fluorescence, ultrastructural, silver-impregnation and immunohistochemical methods in the same section or in consecutive semi-thin and ultra-thin sections. The results indicate that islet cells emitting intense FIF exhibit a strongly argyrophil reaction with the Grimelius' silver method and also immunohistochemical reaction with anti-glucagon serum, but not with anti-5-HT serum. Therefore, the fowl islet A cell, a peptide hormone-producing cell, stores simultaneously catecholamine as biogenic amine. The islet B and D cells did not display any FIF, any argyrophil reaction with the Grimelius' silver method, or any immunoreactivity with anti-glucagon or anti-5-HT sera. The fluorescent but non-argyrophil cells dispersed in the exocrine acinus may well be PP cells. 相似文献

Summary The fine structure of the thecal gland of the domestic fowl, is described for the first time. In the fowl, the glands are located as islets of lipid-laden cells in the theca interna and also in the interfollicular regions. They appear as well defined structures, organized like endocrine glands, quite different from the surrounding theca interna cells. Each gland is composed of two different cell types, the steroid-producing cell, and a cell type never described before, named the enclosing cell. Both cell types are surrounded by a common, distinct basal membrane. The steroid-producing cells are characterized by their content of organelles typical of steroid-producing cells in other organs. The enclosing cells are char cterized by their peripheral location within the gland and their membranous contact with the steroid-producing cells, long processes with desmosomes and their relation to the nerve fibers. They do not contain the organelles typically found in steroid-producing cells. So far, the real function of the enclosing cells is unknown. The following structures are demonstrated in ovarian steroid-producing cells of the fowl for the first time: cytoplasmic microtubules and filaments, intramatrical lipid-like droplets, attachment devices, the polarity of the steroidproducing cells of the thecal gland. 相似文献

Summary In the rostral hypothalamus of the domestic fowl, the magnocellular neurosecretory nuclei show a peculiar differentiation. Golgi studies of the supraoptic and paraventricular nuclei of the fowl reveal at least two major cell types: 1) large multipolar neurons, and 2) small interneurons. Golgi impregnations provide a detailed cytoarchitectural picture of the large-sized cells; the latter may well correspond to the neurosecretory cells demonstrated in the same regions by selective staining, and immunocytochemical and electron microscopical techniques.Electron microscopically, neuronal perikarya are observed to contain variable amounts of neurosecretory granules (100–200 nm in diameter; mean diameter of 160 nm) scattered throughout the cytoplasm. The diameters of these granules do not differ statistically in the two principal nuclear areas examined. The perikarya of these neurons display only a few axosomatic synapses containing electron-lucent and dense-cored vesicles (70–90 nm in diameter). Numerous nerve terminals of this type also end on the dendritic ramifications in the surrounding neuropil. 相似文献

Summary The fine structure of the interstitial gland of the rat ovary was studied at estrus and on Days 4, 6, 10, 14 and 18 of pregnancy. At estrus, ovarian interstitial cells have small nuclei with dense irregular clumps of heterochromatin. Mitochondria are small and rod-shaped and have predominantely lamellar cristae. Numerous osmiophilic lipid droplets are present. At Days 4 and 6, nuclear heterochromatin is reduced, and nucleoli are larger and complex. Mitochondria are enlarged and often bizarre-shaped and have tubular cristae. Golgi and smooth endoplasmic reticulum are more conspicuous. At Day 10, prominent ultrastructural features include nuclei with conspicuous heterochromatin, smaller mitochondria with both lamellar and tubular cristae, numerous ribosomes and lipid droplets with decreased osmiophilia. At Days 14 and 18, nuclei have increased heterochromatin, mitochondria are small and have lamellated cristae and an increase in the size and number of lipid droplets occurs. These observations suggest that steroidogenic activity of interstitial cells is highest during the first half of pregnancy and regresses during the last half. It is suggested that the interstitial gland is an important ovarian source of pregnancy hormone(s) during the first half of gestation and that LH may modulate steroidogenic activity in this ovarian component. 相似文献

We previously demonstrated the presence of 2-methoxyestradiol (2-ME) in swine follicular fluid. Present study was aimed first of all to investigate if swine granulosa cell produce 2-ME; in addition, we tried to assess a potential effect of hypoxia in modulating 2-ME output. Finally, we explored the effect of 2-ME in an angiogenesis bioassay set up in our lab. Our data show that cultured granulosa cells are able to produce 2-ME; interestingly, the secretion of the hormone appeared to be stimulated by hypoxia. Angiogenesis bioassay points out that 2-ME displays an inhibitory effect on neovascularisation. Therefore our data suggest that 2-ME could be a local effector in determining the fine tuning responsible for follicle angiogenesis. These data deserve special attention since the ovary is a valuable experimental model in angiogenesis research. 相似文献

Summary The fine structure of ovarian steroid-producing cells of the domestic fowl as seen after administration of steroids is described. Diaethylstilboestrol, estradiol and hydroxyprogesterone were given as intramuscular injections for a 28-days period. The main cytoplasmic changes of the steroid-producing cells were an increase in the number and the size of lipid droplets due to a possible expansion of the endoplasmic reticulum and a confluence of the smooth endoplasmic reticulum and the lipid droplets. The mitochondria became paler, markedly reduced in size and number, and contained less cristae. The Golgi apparatus became smaller and tended to disappear. The most prominent nuclear changes were irregular outline of the nuclear membrane, shrinkage of the nucleus, formation of pseudo-inclusions, decreased number of nuclear bodies with a tendency to vacuolization and small nucleoli. These alterations have never been demonstrated in ovarian steroid-producing cells before, and they are consistent with atrophic changes observed in steroid-producing cells in other organs. The present study substantiates the view that the thecal gland must be concerned with endocrine function. The possible functions of the different cell types of the thecal gland are discussed. 相似文献

Neutral endopeptidase (NEP) is a zinc metallopeptidase ubiquitously distributed in various tissues in mammals. This peptidase is involved in the post-secretory metabolism of various neuropeptides and peptide hormones in vivo, such as enkephalins, bradykinin, atrial natriuretic peptide, substance P and endothelins. In this paper we show that NEP is expressed in ovaries as a 110-kDa glycosylated integral membrane protein with enzymatic properties similar to those of the kidney protein. Using immunohistochemistry, we localize the peptidase in the granulosa cells of follicles at all stages of maturation, with the exception of atretic follicles. We also observe immunoreactive staining in the epithelia that lines the blood vessels in the medulla and the surface of the ovary. The co-localization of NEP and bioactive peptides known to be physiological substrates of NEP in other tissues suggests an important role for this protein in processes such as follicle maturation, ovulation, and/or regulation of ovarian blood flow, by modulating the physiological function of these peptides. 相似文献

Summary The ultrastructure of corpora lutea obtained during the preimplantation, implantation and early postimplantation periods has been studied in 20 western spotted skunks. Fine structure of granulosa lutein cells was correlated with progesterone levels. The corpus luteum of the prolonged (7 month) preimplantation period contained undifferentiated small granulosa cells and differentiated large granulosa lutein cells. The former ranged in size between 12 and 20 and the latter between 20 and 45 . The ratio of small and large cells was about equal in an animal 2 days prior to nidation whereas only few small cells and numerous large cells were observed in an animal estimated to be 8 to 12 hours from nidation. Occasionally small cells were observed amidst large ones during the 24 hour nidation period, i.e. adhesion of trophoblast with the luminal uterine epithelium, but small cells were absent in animals after this period. Small cells had some smooth and rough endoplasmic reticulum, rod-shaped mitochondria with platelike cristae, small Golgi complex, and relatively smooth plasma membranes. Large lutein cells had abundant smooth endoplasmic reticulum, membranous whorls of smooth endoplasmic reticulum, usually round mitochondria with tubular and lamellar cristae, a well developed Golgi complex, variable amounts of lipid droplets, and highly plicated and ruffled plasma membranes. Peripheral plasma progesterone levels during the prolonged preimplantation period ranged between 1.1 and 7.9 ng/ml, but during implantation it was between 8 and 16.6 ng/ml. It is suggested that plasma progesterone levels fluctuate during the time of implantation and should not be regarded as a basis to predict actual nidation in the western spotted skunk.This research was supported in part by Grant Number HD06556 from the National Institute of Child Health and Human Development. 相似文献
