We combined measurements of short-term (during gas exchange) and long-term (from plant dry matter) carbon isotope discrimination to estimate CO2 leakiness from bundle sheath cells in six C4 species (three grasses and three dicots) as a function of leaf insertion level, growth temperature and short-term irradiance. The two methods for determining leakiness yielded similar results (P > 0.05) for all species except Setaria macrostachya, which may be explained by the leaf of this species not being accommodating to gas exchange. Leaf insertion level had no effect on leakiness. At the highest growth temperature (36°C) leakiness was lower than at the two lower growth temperatures (16°C and 26°C), between which no differences in leakiness were apparent. Higher irradiance decreased leakiness in three species, while it had no significant effect on the others (there was an opposite trend in two species). The inverse response to increasing irradiance was most marked in the two NAD-ME dicots (both Amaranthus species), which both showed almost 50% leakiness at low light (300 μmol quanta m−2 s−1) compared to about 30% at high light (1,600 μmol quanta m−2 s−1). NADP-ME subtype grasses had lower leakiness than NAD-ME dicots. Although there were exceptions, particularly in the effect of irradiance on leakiness in Sorghum and Boerhavia, we conclude that conditions favourable to C4 photosynthesis (high temperature and high light) lead to a reduction in leakiness.  相似文献

Sensitivity to cold and freezing differs between populations within two species of live oaks (Quercus section Virentes Nixon) corresponding to the climates from which they originate. Two populations of Quercus virginiana (originating from North Carolina and north central Florida) and two populations of the sister species, Q. oleoides, (originating from Belize and Costa Rica) were grown under controlled climate regimes simulating tropical and temperate conditions. Three experiments were conducted in order to test for differentiation in cold and freezing tolerance between the two species and between the two populations within each species. In the first experiment, divergences in response to cold were tested for by examining photosystem II (PS II) photosynthetic yield (ΔF/F m′) and non-photochemical quenching (NPQ) of plants in both growing conditions after short-term exposure to three temperatures (6, 15 and 30°C) under moderate light (400 μmol m−2 s−1). Without cold acclimation (tropical treatment), the North Carolina population showed the highest photosynthetic yield in response to chilling temperatures (6°C). Both ecotypes of both species showed maximum ΔF/F m′ and minimum NPQ at their daytime growth temperatures (30°C and 15°C for the tropical and temperate treatments, respectively). Under the temperate treatment where plants were allowed to acclimate to cold, the Q. virginiana populations showed greater NPQ under chilling temperatures than Q. oleoides populations, suggesting enhanced mechanisms of photoprotective energy dissipation in the more temperate species. In the second and third experiments, inter- and intra-specific differentiation in response to freezing was tested for by examining dark-adapted F v/F m before and after overnight freezing cycles. Without cold acclimation, the extent of post-freezing declines in F v/F m were dependent on the minimum freezing temperature (0, −2, −5 or −10°C) for both populations in both species. The most marked declines in F v/F m occurred after freezing at −10°C, measured 24 h after freezing. These declines were continuous and irreversible over the time period. The North Carolina population, however, which represents the northern range limit of Q. virginiana, showed significantly less decline in F v/F m than the north central Florida population, which in turn showed a lower decline in Fv/F m than the two Q. oleoides populations from Belize and Costa Rica. In contrast, after exposure to three months of chilling temperatures (temperate treatment), the two Q. virginiana populations showed no decline in F v/F m after freezing at −10°C, while the two Q. oleoides populations showed declines in F v/F m reaching 0.2 and 0.1 for Costa Rica and Belize, respectively. Under warm growth conditions, the two species showed different F 0 dynamics directly after freezing. The two Q. oleoides populations showed an initial rise in F 0 30 min after freezing, followed by a subsequent decrease, while the Q. virginiana populations showed a continuous decrease in F 0 after freezing. The North Carolina population of Q. virginiana showed a tendency toward deciduousness in response to winter temperatures, dropping 58% of its leaves over the three month winter period compared to only 6% in the tropical treatment. In contrast, the Florida population dropped 38% of its leaves during winter. The two populations of the tropical Q. oleoides showed no change in leaf drop during the 3-months winter (10% and 12%) relative to their leaf drop over the same timecourse in the tropical treatment. These results indicate important ecotypic differences in sensitivity to freezing and cold stress between the two populations of Q. virginiana as well as between the two species, corresponding to their climates of origin.  相似文献

Isotope screening is a simple test for determining the photosynthetic pathway used by plants. The scope of this work was to classify the photosynthetic type of some herbs and medicinal plants through studies of the carbon isotope composition (δ13C). Also, we propose the use of carbon isotope composition as a tool to control the quality of herbs and medicinal plants. For studies of δ13C, δ13C‰ = [R (sample)/R (standard) − 1] × 10−3, dry leaves powdered in cryogenic mill were analyzed in a mass spectrometer coupled with an elemental analyzer for determining the ratio R = 13CO2/12CO2. In investigation of δ13C of 55 species, 23 botanical families, and 44 species possessed a C3 photosynthetic type. Six species found among the botanical families Euphorbiaceae and Poaceae were C4 plants, and 5 species found among the botanical families Agavaceae, Euphorbiaceae, and Liliaceae possessed CAM-type photosynthesis. Carbon isotope composition of plants can be used as quality control of herbs and medicinal plants, allowing the identification of frauds or contaminations. Also, the information about the photosynthetic type found for these plants can help in introducing and cultivating exotic and wild herbs and medicinal plants.  相似文献

One-year-old oak (Quercus mongolica Fisch.) seedlings were grown in growth chambers for 30 days to investigate the effects of the combination of elevated CO2 concentration ([CO2], 700 μmol/mol) and temperature (ambient T + 4°C) and only elevated temperature (ambient T +4°C) on leaf gas exchange, chlorophyll a fluorescence, and chlorophyll content. In the growth chambers, natural conditions of the Maoershan mountain regions of Heilongjiang Province (45–46°N, 127–128°E) of China for the average growth season were simulated. The results showed that the maximum net photosynthetic rate (P Nmax) was ≈ 1.64 times greater at elevated temperature than at ambient temperature. The irradiance saturation point (I s), apparent quantum yield (AQY), maximum photosystem II efficiency (F v/F m), and chlorophyll content significantly increased, while irradiance compensation point (I c) was not affected by elevated temperature. The combination of elevated [CO2] and temperature also significantly increased P Nmax by approximately 34% but much lower than that under elevated temperature only. In the case of factor combination, dark respiration (R d), I c, F v/F m, and total chlorophyll content increased significantly, while I s and AQY were not affected. Moreover, under elevated [CO2] and temperature, R d and I c, F v/F m were significantly higher than under elevated temperature only. The results indicated that the combination of elevated [CO2] and temperature expected in connection with the further global climate change may affect carbon storage of the coenotype of Q. mongolica in this region of China. When aspirating human red blood cells (RBCs) into 1.3 μm pipettes (ΔP = −2.3 kPa), a transition from blocking the pipette below a critical temperature T c = 36.3 ± 0.3°C to passing it above the T c occurred (micropipette passage transition). With a 1.1 μm pipette no passage was seen which enabled RBC volume measurements also above T c. With increasing temperature RBCs lost volume significantly faster below than above a T c = 36.4 ± 0.7 (volume transition). Colloid osmotic pressure (COP) measurements of RBCs in autologous plasma (25°C ≤ T ≤ 39.5°C) showed a T c at 37.1 ± 0.2°C above which the COP rapidly decreased (COP transition). In NMR T1-relaxation time measurements, the T1 of RBCs in autologous plasma changed from a linear (r = 0.99) increment below T c = 37 ± 1°C at a rate of 0.023 s/K into zero slope above T c (RBC T1 transition). In conclusion: An amorphous hemoglobin–water gel formed in the spherical trail, the residual partial sphere of the aspirated RBC. At T c, a sudden fluidization of the gel occurs. All changes mentioned above happen at a distinct T c close to body temperature. The T c is moved +0.8°C to higher temperatures when a D2O buffer is used. We suggest a mechanism similar to a “glass transition” or a “colloidal phase transition”. At T c, the stabilizing Hb bound water molecules reach a threshold number enabling a partial Hb unfolding. Thus, Hb senses body temperature which must be inscribed in the primary structure of hemoglobin and possibly other proteins. Species in the Laxa and Grandia groups of the genus Panicum are adapted to low, wet areas of tropical and subtropical America. Panicum milioides is a species with C3 photosynthesis and low apparent photorespiration and has been classified as a C3/C4 intermediate. Other species in the Laxa group are C3 with normal photorespiration. Panicum prionitis is a C4 species in the Grandia group. Since P. milioides has some leaf characteristics intermediate to C3 and C4 species, its photosynthetic response to irradiance and temperature was compared to the closely related C3 species, P. laxum and P. boliviense and to P. prionitis. The response of apparent photosynthesis to irradiance and temperature was similar to that of P. laxum and P. boliviense, with saturation at a photosynthetic photo flux density of about 1 mmol m-2 s-1 at 30°C and temperature optimum near 30°C. In contrast, P. prionitis showed no light saturation up to 2 mmol m-2 s-1 and an optimum temperature near 40°C. P. milioides exhibited low CO2 loss into CO2-free air in the light and this loss was nearly insensitive to temperature. Loss of CO2 in the light in the C3 species, P. laxum and P. boliviense, was several-fold higher than in P. milioides and increased 2- to 5-fold with increases in temperature from 10 to 40°C. The level of dark respiration and its response to temperature were similar in all four Panicum species examined. It is concluded that the low apparent photorespiration in P. milioides does not influence its response of apparent photosynthesis to irradiance and temperature in comparison to closely related C3 Panicum species.Abbreviations AP apparent photosynthesis - I CO2 compensation point - gl leaf conductance; gm, mesophyll conductance - PPFD photosynthetic photon flux density - PR apparent photorespiration rate - RuBPC sibulose bisphosphate carboxylase  相似文献

The chlorophyll fluorescence (F) temperature curves in a linear time-temperature heating/cooling regime were used to study heat-induced irreversible F changes in primary green leaves of spring barley (Hordeum vulgare L. cv. Akcent). The leaf segments were heated in a stirred water bath at heating rates of 0.0083, 0.0166, 0.0333, and 0.0500 °C s−1 from room temperature up to maximal temperature T m and then linearly cooled to 35 °C at the same rate. The F intensity was measured by a pulse-modulated technique. The results support the existence of the two critical temperatures of irreversible F changes postulated earlier, at 45–48 and 53–55 °C. The critical temperatures are slightly dependent on the heating rate. Two types of parameters were used to characterize the irreversibility of the F changes: the coefficient of irreversibility μ defined as the ratio of F intensity at 35 °C at the starting/ending parts of the cycle and the slopes of tangents of linear parts of the F temperature curve. The dependence of μ on T m revealed a maximum, which moved from 54 to 61 °C with the increasing heating/cooling rate v from 0.0083 to 0.0500 °C s−1, showing two basic phases of the irreversible changes. The Arrhenius and Eyring approaches were applied to calculate the activation energies of the initial increase in μ. The values varied between 30 and 50 kJ mol−1 and decreased slightly with the increasing heating rate.  相似文献

The anatomical features of leaves in 11 species of plants grown in a temperature gradient and a temperature + CO2 gradient were studied. The palisade parenchyma thickness, the spongy parenchyma thickness and the total leaf thickness were measured and analyzed to investigate the effects of elevated temperature and CO2 on the anatomical characteristics of the leaves. Our results show that with the increase of temperature, the leaf thickness of C4 species increased while the leaf thickness of C3 species showed no constant changes. With increased CO2, seven out of nine C3 species exhibited increased total leaf thickness. In C4 species, leaf thickness decreased. As for the trend on the multi-grades, the plants exhibited linear or non-linear changes. With the increase of temperature or both temperature and CO2 for the 11 species investigated, leaf thickness varied greatly in different plants (species) and even in different branches on the same plant. These results demonstrated that the effect of increasing CO2 and temperature on the anatomical features of the leaves were species-specific. Since plant structures are correlated with plant functions, the changes in leaf anatomical characteristics in elevated temperature and CO2 may lead to functional differences. A novel Gram-negative, slightly halophilic, catalase- and oxidase-positive, obligately aerobic bacterium, strain YIM-C248T, was isolated from a sediment sample collected from a salt-lake in the Qaidam Basin in Qinghai, north-west China. Cells were non-sporulating short rods, occurring singly or as doublets, motile with peritrichous flagella. Growth occurred with 1–15% (w/v) NaCl [optimum 2–4% (w/v) NaCl], at pH 6.0–10.0 (optimum pH 7.5) and at 4–35°C (optimum 25–30°C). The major cellular fatty acids were C18:1 ω7c, C12:0 3-OH, cyclo C19:0 ω8c, C16:0 and C16:1. The predominant respiratory quinone was Q-9 and the genomic DNA G + C content was 58.6 mol%. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain YIM-C248T should be assigned to the genus Halomonas. The sequence similarities between the isolate and the type strains of members of the genus Halomonas were in the range of 92.5–97.5%. The combination of phylogenetic analysis, DNA–DNA hybridization data, phenotypic characteristics and chemotaxonomic differences supported the view that strain YIM-C248T represents a new species of the genus Halomonas, for which the name Halomonas sediminis sp. nov. is proposed, with YIM-C248T (=CCTCC AA 207031 = KCTC 22167) as the type strain. The GenBank/EMBL/DBBJ accession number for the 16S rRNA gene sequence of strain YIM-C248T is EU135707.  相似文献

A novel Gram-negative, catalase- and oxidase-positive, non-sporulating, rod-shaped, aerobic bacterium, designated strain JSM 078120T, was isolated from sea water collected from a tidal flat of Naozhou Island, South China Sea. Growth occurred with 1–15% (w/v) total salts (optimum, 2–4%), at pH 6.0–10.0 (optimum, pH 7.5) and at 4–35°C (optimum, 25–30°C). The major cellular fatty acids were C18:1 ω9c, C16:0, C12:0 3-OH and C16:1 ω7c. The predominant respiratory quinone was ubiquinone Q-9, and the genomic DNA G + C content was 60.6 mol%. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain JSM 078120T should be assigned to the genus Marinobacter, being related most closely to the type strains of Marinobacter segnicrescens (sequence similarity 98.2%), Marinobacter bryozoorum (97.9%) and Marinobacter gudaonensis (97.6%). The sequence similarities between the novel isolate and the type strains of other recognized Marinobacter species ranged from 96.7 (with Marinobacter salsuginis) to 93.3% (with Marinobacter litoralis). The levels of DNA–DNA relatedness between strain JSM 078120T and the type strains of M. segnicrescens, M. bryozoorum and M. gudaonensis were 25.3, 20.6 and 18.8%, respectively. The combination of phylogenetic analysis, DNA–DNA relatedness, phenotypic characteristics and chemotaxonomic data supported the view that strain JSM 078120T represents a novel species of the genus Marinobacter, for which the name Marinobacter zhanjiangensis sp. nov. is proposed. The type strain is JSM 078120T (= CCTCC AB 208029T = DSM 21077T = KCTC 22280T). The GenBank/EMBL/DDBJ accession number for the 16S rRNA gene sequence of strain JSM 078120T is FJ425903.  相似文献

Floristic compositions, life forms, reproductive types for forage species, and their responses to desertification in Hunshandake desert were studied. 164 species, in 30 families and 94 genera, were identified with C3 (137 species), C4 (25 species), and CAM (2 species) photosynthesis. Of the 25 C4 species, 76 % were grasses and Chenopodiaceae species (hereafter chenopods). This suggests that the C4 species mainly occurred in a few families in the desert region. The reduction of C3 species and the increase of C4 species with desertification indicated that C4 species might have higher tolerance to environmental stresses (e.g. dry and poor soil). Relatively more hemicrytophyte and therophyte forms in the desert are related to the local temperate climate and vegetation dynamics. Relatively greater proportions of C4/C3 and clonal species/sexual species at mobile dune showed that the C4 species and clonal species could make greater contribution to sand land restoration in the Hunshandake desert.  相似文献

Summary Diurnal changes in chlorophylla fluorescence were determined in four species, differing in life form, in Portugal during the summer of 1989. These includedOpuntia ficus-indica, a CAM plant, andHelianthus annuus, Ficus carica andArbutus unedo, three C3 species. Steady state fluorescence yield,F S, and maximum fluorescence yield,F M′, were determined at different times of the day. Using the model of Genty et al. (1989), the photon use efficiency of photosystem II electron transport,φ e, was calculated from (F M′−F S)/F M′. Diurnal changes in relative rate of non-cyclic electron transport through photosystem II,J e, were derived by multiplyingφ e by the incident photon flux density (PFD). WhenJ e, determined for each species for various points in time throughout the day, was plotted against corresponding values of PFD, the light response curves obtained showed thatJ e was linearly dependent on PFD in low light and approached saturation in high light. The highest values ofJ e were observed inHelianthus annuus, followed byOpuntia ficus-indica, Ficus carica andArbutus unedo. The proportion of the xanthophyll zeaxanthin to total carotenoids, determined around noon, was inversely related to maximum rates ofJ e.  相似文献

A pale yellow-colored, moderately halophilic, Gram-negative, catalase- and oxidase-positive, non-sporulating, rod-shaped, motile, aerobic bacterium, designated strain JSM 073008T, was isolated from a sea anemone (Anthopleura xanthogrammica) collected from Naozhou Island, Leizhou Bay, South China Sea. The organism was able to grow with 1–20% (w/v) total salts (optimum, 5–10%), at pH 6.0–10.0 (optimum, pH 7.5) and 10–40°C (optimum, 25–30°C). The major cellular fatty acids were C16:0, C16:1 ω7c/iso-C15:0 2-OH and C18:1 ω7c. The polar lipids consisted of phosphatidylethanolamine, phosphatidylglycerol and an unidentified phospholipid. The predominant respiratory quinone was Q-8 and the genomic DNA G + C content was 47.5 mol%. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain JSM 073008T should be assigned to the genus Alteromonas, being most closely related to Alteromonas hispanica F-32T (sequence similarity 96.9%), followed by Alteromonas genovensis LMG 24078T (96.6%) and Alteromonas litorea TF-22T (96.4%). The sequence similarities between the novel isolate and the type strains of other recognized Alteromonas species ranged from 95.9% (with Alteromonas stellipolaris ANT 69aT) to 94.5% (with Alteromonas simiduii BCRC 17572T). The combination of phylogenetic analysis, phenotypic characteristics and chemotaxonomic data supported the view that strain JSM 073008T represents a new species of the genus Alteromonas, for which the name Alteromonas halophila sp. nov. is proposed. The type strain is JSM 073008T (=CCTCC AA 207035T = KCTC 22164T). The authors Yi-Guang Chen and Huai-Dong Xiao have contributed equally to this work.  相似文献

Photosynthetic pathway Types (C3, C4, and CAM) and life forms of native species from Hulunbeier rangelands, north China were studied. Of the total 258 species, 216 species in 132 genera and 42 families had C3 photosynthetic pathway, including dominant herbs, e.g. Stipa baicalensis Roshev. and Leymus chinensis (Trin.) Tzvel., Filifolium sibiricum Kitam. and Arudinella hirta (Thunb.) Koidz. 38 species in 28 genera and 10 families were found with C4 photosynthesis, and 4 species in 2 genera and 1 family had CAM photosynthetic pathway. The occurrence of C4 species was common in Gramineae and Chenopodiaceae, and the two families were leading ones within C4 plants. More than 52 % of the total 258 species were in H form, 21 % in Th form, 19 % in G form; the other life form Types, e.g. Ch, M, N, and HH, formed less than 3 %. 68 % of C4 species were in Th form and 24 % in H form, indicating that these Types were the dominant life forms for C4 species in the rangeland region. The occurrence of C4 species was closely related with plant habitats, disturbed lands had the highest C4 abundance (55 % of the total C4 species), followed by grasslands and sandy soil, and forests had the lowest C4 abundance (8 %). Hence the occurrence of C4 species could be efficient indicator for rangeland dynamics in Hulunbeier rangelands.  相似文献

The quantum yield for CO2 uptake was measured in C3 and C4 monocot species from several different grassland habitats. When the quantum yield was measured in the presence of 21% O2 and 340 cm3 m-3 CO2, values were very similar in C3 monocots, C3 dicots, and C4 monocots (0.045–0.056 mole CO2 · mole-1 quanta absorbed). In the presence of 2% O2 and 800 cm3 m-3 CO2, enhancements of the quantum yield values occurred for the C3 plants (both monocots and dicots), but not for C4 monocots. A dependence of the quantum yield on leaf temperature was observed in the C3 grass, Agropyron smithii, but not in the C4 grass, Bouteloua gracilis, in 21% O2 and 340 cm3 m-3 CO2. At leaf temperatures between 22–25°C the quantum yield values were approximately equal in the two species.  相似文献

Photosynthetic pathways (C3, C4, and CAM) and morphological functional types (e.g. shrubs, high perennial grasses, short perennial graminaceous plants, annual grasses, annual forbs, perennial forbs, halophytes, and hydrophytes) were identified for the species from salinity grasslands in Northeastern China, using the data from both stable carbon isotope ratios (13C) and from the references published between 1993 and 2002. 150 species, in 99 genera and 37 families, were found with C3 photosynthesis, and most of these species are dominants [e.g. Leymus chinensis (Trin.) Tzvel., Calamagrostis epigeios (L.), Suaeda corniculata (C.A. Mey.) Bunge]. 40 species in 25 genera and 8 families were identified with C4 photosynthesis [e.g. Chloris virgata Sw., Aeluropus littoralis (Gouan) Parlat] and 1 species with CAM photosynthesis. Gramineae is the leading family with C4 photosynthesis (27 species), Chenopodiaceae ranks the second (5 species). The significant increase of C4 proportions with intense salinity suggested this type plant is remarkable response to the grassland salinization in the region. 191 species were classified into eight morphological functional types and the changes of most of these types (e.g. PEF, HAL, and HPG) were consistent with habitats and vegetation dynamics in the saline grassland. My findings suggest that the photosynthetic pathways, combined with morphological functional types, are efficient means for studying the linkage between species and ecosystems in this type of saline grassland in Northeastern China. This revised version was published online in March 2005 with corrections to the page numbers.  相似文献

Seeds of Suaeda salsa were cultured in dark for 3 d and betacyanin accumulation in seedlings was promoted significantly. Then the seedlings with accumulated betacyanin (C+B) were transferred to 14/10 h light/dark and used for chilling treatment 15 d later. Photosystem 2 (PS2) photochemistry, D1 protein content, and xanthophyll cycle during the chilling-induced photoinhibition (exposed to 5 °C at a moderate photon flux density of 500 μmol m−2 s−1 for 3 h) and the subsequent restoration were compared between the C+B seedlings and the control (C) ones. The maximal efficiency of PS2 photochemistry (Fv/Fm), the efficiency of excitation energy capture by open PS2 centres (Fv′/Fm′), and the yield of PS2 electron transport (ΦPS2) of the C+B and C leaves both decreased during photoinhibition. However, smaller decreases in Fv/Fm, Fv′/Fm′, and ΦPS2 were observed in the C+B leaves than in C ones. At the same time, the deepoxidation state of xanthophyll cycle, indicated by (A+Z)/(V+A+Z) ratio, increased rapidly but the D1 protein content decreased considerably during the photoinhibition. The increase in rate of (A+Z)/(V+A+Z) was higher but the D1 protein turnover was slower in C+B than C leaves. After photoinhibition treatment, the plants were transferred to a dim irradiation (10 μmol m−2 s−1) at 25 °C for restoration. During restoration, the chlorophyll (Chl) fluorescence parameters, D1 protein content, and xanthophyll cycle components relaxed gradually, but the rate and level of restoration in the C+B leaves was greater than those in the C leaves. The addition of betacyanins to the thylakoid solution in vitro resulted in similar changes of Fv/Fm, D1 protein content, and (A+Z)/(V+A+Z) ratio during the chilling process. Therefore, betacyanin accumulation in S. salsa seedlings may result in higher resistance to photoinhibition, larger slowing down of D1 protein turnover, and enhancement of non-radiative energy dissipation associated with xanthophyll cycle, as well as in greater restoration after photoinhibition than in the control when subjected to chilling at moderate irradiance.  相似文献

Two novel strains of budding bacteria, Z-0071T and Z-0072, were isolated from dystrophic humified waters formed by xylotrophic fungi in the course of spruce wood degradation. The cells of both strains are coccoid (0.95–1.80 μm), nonmotile, single or arranged in pairs. The cells have a complex system of intracellular membranes and are covered with fimbriae and surrounded by a mucous capsule up to 0.3 μm thick. Both strains are aerobic organoheterotrophic, mesophilic, and acid-tolerant microorganisms that are able to grow under microaerobic conditions. They utilize N-acetyl-glucosamine, carbohydrates, and lactate as growth substrates. The strains grow in a pH range of 4.0–7.5 with an optimum at 6.0–6.5. The temperature range for growth is 4–30°C, with an optimum at 25–28°C. Strains Z-0071T and Z-0072, inhabitants of dystrophic low-mineral waters, are NaCl-sensitive: the NaCl content in the media above 0.5 g/l inhibited growth. The main fatty acids of strains Z-0071T and Z-0072 are C16:0, C18:1ω9c, and C18:2ω9c, 12c. The DNA G + C base content is 51.2–51.7 mol %. The sequences of the 16S rRNA gene fragments (1310 bp) of strains Z-0071T and Z-0072 were found to be identical. The obtained sequences showed a 94.3% similarity with the sequences of the type strain of the most closely related species Singulisphaera acidiphila MOB10≅T. The phenotypic and phylogenetic properties of strains Z-0071T and Z-0072 support classification of these strains within the genus Singulisphaera as a new species Singulisphaera mucilagenosa sp. nov., with the type strain Z-0071T (VKM B-2626).  相似文献

A moderate halophilic, Gram-negative, non-motile, rod-shape, and aerobe designated as strain HwaT was isolated from traditional fermented Korean seafood, which presented as a single cell or paired cells. Optimal growth occurred at 25°C in 10% (w/v) salts at pH 7.0–8.0; however, growth occurred in a temperature range of 10–32°C, a salts concentration of 5–25% (w/v) and pH 5.0–10.0. Tests for oxidase and catalase were positive. The cells produced poly-β-hydroxybutyric acid, but not exopolysaccharide. Based on the 16S rRNA gene sequence, not only was there low similarity between strain HwaT and all other species (94.1% similarity with H. subglaciescola DSM 4683T, 94.0% similarity with H. sulfidaeris Esulfide1T, 93.6% similarity with H. cerina SP4T and 93.0% similarity with H. halodurans DSM 5160T), but the phylogenetic analysis revealed that the isolate may be classified as a novel species belonging to the genus Halomonas in the class Gammaproteobacteria. The predominant fatty acids of strain HwaT were C18:1 ω7c, C16:0, C12:0 3-OH and C16:1 ω7c/C15:0 iso 2-OH. The DNA G+C content was calculated as 61.7 mol%. Based on phenotypic, genotypic, and phylogenetic characteristics, it is proposed that the strain designated as HwaT be assigned to the genus Halomonas as Halomonas jeotgali sp. nov. (=KCTC 22487T =JCM 15645T).  相似文献

The maximum quantum yields (a,c) for CO2 uptake in low-oxygen atmospheres were determined for 11 species of C3 vascular plants of diverse taxa, habitat and life form using an Ulbricht-sphere leaf chamber. Comparisons were also made between tissues of varied age within species. The species examined were Psilotum nudum (L.) P. Beauv., Davallia bullata Wall. ex Hook., Cycas revoluta Thunb., Araucaria heterophylla (Salisb.) Franco, Picea abies (L.) Karst., Nerium oleander L., Ruellia humilis Nutt., Pilea microphylla (L.) Karst., Beaucarnea stricta Lem., Oplismenus hirtellus (L.) P. Beauv. and Poa annua L. Quantum yields were calculated from the initial slopes of the response of CO2 uptake to the quantity of photons absorbed in conditions of diffuse lighting. Regression analysis of variance of the initial slopes of the response of CO2 uptake to photon absorption failed to show any statistically significant differences between age classes within species or between the mature photosynthetic organs of different species. The constancy of a,c was apparent despite marked variation in the light-saturated rates of CO2 uptake within and between species. The mean a,c was 0.093±0.003 for 11 species. By contrast, surface absorptance varied markedly between species from 0.90 to 0.60, producing proportional variation in the quantum yield calculated on an incidentlight basis. The ratio of variable to maximum fluorescence emission at 695 nm for the same tissues also failed to show any statistically significant variation between species, with a mean of 0.838±0.008. Mean values of a,c reported here for C3 species, in the absence of photorespiration, are higher than reported in previous surveys of vascular plants, but consistent with recent estimates of the quantum yields of O2 evolution.Abbreviations and Symbols A rate of CO2 uptake per unit projected area (mol · m–2 · s–1) - Fm the maximum fluorescence emission at 695 nm in saturating excitation light when closure of PSII reaction centres is maximal (relative units) - Fo the ground fluorescence at 695 nm when all PSII reaction centres are assumed open (relative units) - Fv the difference between Fm and Fo - JQ rate of CO2 uptake by the sample (nmol · s–1) - JQ rate of photon absorption by the sample (nmol · s–1) - Q absorbed photon flux per unit of projected area (nmol · m–2 · s–1) - 1 the light absorptance of photosynthetic organs (dimensionless) - s1 and s'1 the total and projected surface areas of the photosynthetic organs examined (m2) - a,c and i,c the quantum yields for CO2 uptake on an absorbed- and incident-light basis, respectively (dimensionless) - a,o the quantum yield for O2 evolution on an absorbed-light basis (dimensionless) This work was supported by grant PI7179-BIO, FWF, Austria to H.B-N. and by a British Council travel award to S.P.L.
