Anthropogenic stratospheric ozone depletion causes an increase in UVB radiation impinging on the earth's surface, which is a threat to plants not adapted to higher UVB irradiances. Investigations were carried out among tropical marine macrophytes, Turbinaria turbinata, Sargassum polyceratium var. ovatum, Padina sanctae-crucis, Lobophora variegate, Dictyota spec., Halimeda discoidea, Udotea flabellum, Thalassia testudinum and Syringodium filiforme collected from 0.3 to 26 m depths at the Belizean barrier reef, where ultraviolet radiation (UV)-irradiances are naturally high. Photoinhibition was induced under full solar, UV (UVA + UVB), and UVA only-depleted radiation conditions. Photosynthetic activity during high radiation stress and during recovery in reduced solar radiation was determined in vivo by measuring fluorescence changes using a PAM fluorometer device. Generally, UV caused an additional decrease of photosynthetic performance during high light stress which varies according to species, depth of growth and UV penetration at the site of collection; an observation in concordance with the conventional harmful UV-radiation effects on phototrophs. When solar radiation was reduced by 50%, significant photosynthetic recovery was observed. However, some shallow water species which are adapted to high UV were observed to recover less under treatment with depleted solar UVB radiation. Our result supports earlier reports that UVB causes not only negative effects on photosynthesis, but may also facilitate or induce recovery processes in aquatic macrophytes acclimated to high solar radiation which grow at the upper shoreline. Among the eulittoral macroalgae, e.g. Dictyota spec., P. sanctae-crucis, and H. discoidea and the seagrass T. testudinum, initiation of photosynthetic recovery processes in the presence of low irradiance of short UV-wavelengths may present an ecophysiological advantage compared to macrophytes which initiate photosynthetic recovery process during low light or in the absence of UV.  相似文献

Several unicellular algae were exposed to artificial UV-B (280–320 nm) radiation after adaptation to high (43 W m−2) and low (19 W m−2) visible light. UV-B radiation had different effects on rates of photosynthesis, motility and absorption spectra for these species. Photosynthesis of Euglena gracilis and the diatom Phaeodactylum tricomution was more sensitive to UV-B inhibition than that of the dinoflagellates Heterocapsa triquetra and Prorocentrum minimum . Not only UV-B radiation but also high visible light had a photoinhibitory effect on photosynthesis in all four organisms. The effect on photosynthesis was observed both on the quantum yield and on the light saturation rate of photosynthesis. The dinoflagellates, in contrast to E. gracilis and P. tricorntum , absorbed strongly in the UV region (334 nm) and their absorption peaks increased after growth under high visible light or with or without UV-B radiation for one week. The swimming speed of H. triquetra decreased more after low visible light and UV-B radiation compared to high visible light and UV-B radiation. The negative effects of UV-B radiation on P. minimum and E. gracilis were most pronounced after high visible light.  相似文献

The major UV-B screening pigments of the epidermal layer of Scots pine (Pinus sylvestris) needles are flavonol 3-o-glycosides (F3Gs) esterified with hydroxycinnamic acids at positions 3" and 6". Acylation is the last step in biosynthesis and is catalysed by position-specific hydroxycinnamoyl transferases (3" and 6"HCT). The UV-B dependence of these enzyme activities was studied in primary needles of Scots pine seedlings grown under different UV-B conditions in environmentally controlled sun simulators. 6"HCT activity was induced upon UV-B irradiation while 3"HCT activity was not induced but showed high constitutive values. To investigate the biosynthesis of diacylated F3Gs during needle development under natural conditions, the HCT activities and metabolite contents were analysed in needles of field-grown mature pine trees. Accumulation of diacylated compounds as well as of 6"HCT activity occurred transiently in the first year of needle development only. In contrast, 3"HCT activity exhibited broad maxima in two consecutive years during needle growth. The data suggest that acylated F3Gs are first formed as soluble compounds which are then translocated into the cell wall to be bound by their hydroxycinnamoyl residues.  相似文献

A convenient and sensitive method for the quantitative determination of poly(ethylene glycol) 400 in plasma and urine with capillary gas chromatography-mass spectrometry has been developed. The sample preparation involves solid-phase extraction with subsequent derivatization with heptafluorobutyric anhydride, which proved to give the most stable derivative. The derivatization procedure was optimized using experimental design, and different solid-phase extraction columns were evaluated. The limit of quantitation was 1 μmol/l (0.4 μg/ml) for both plasma and urine.  相似文献

Response of adenine nucleotides (ATP, ADP, AMP) and adenylate energy charge (EC) to atrazine, a triazine herbicide, was evaluated as an indicator of metabolic state in Zostera marina L. (eelgrass), a submerged marine angiosperm. Short-term (6 h) atrazine stress reduced ATP and total adenylates (AT) at both 10 and 100 ppb, but EC remained constant. Net productivity decreased at 100, but not at 10 ppb atrazine over 6 h. Long-term (21 day) atrazine stress was evidenced by growth inhibition and 50% mortality near 100 ppb. EC was reduced at 0.1, 1.0 and 10 ppb atrazine, but ATP and EC increased with physiological response to severe stress (100 ppb) after 21 days. Apparently, ATP and AT decrease over the short-term but rebound over the long-term with severe atrazine stress, increasing beyond control levels before plant death results. Supplementing adenine nucleotide and EC results with more conventional quantitative analyses should afford greater knowledge of physiological response to environmental variation.  相似文献

BACKGROUND: Depletion of the stratospheric ozone layer leads to an increase in ultraviolet-B (UVB: 280-320 nm) radiation reaching the earth's surface, and the enhanced solar UVB radiation predicted by atmospheric models will result in reduction of growth and yield of crops in the future. Over the last two decades, extensive studies of the physiological, biochemical and morphological effects of UVB in plants, as well as the mechanisms of UVB resistance, have been carried out. SCOPE: In this review, we describe recent research into the mechanisms of UVB resistance in higher plants, with an emphasis on rice (Oryza sativa), one of the world's most important staple food crops. Recent studies have brought to light the following remarkable findings. UV-absorbing compounds accumulating in the epidermal cell layers have traditionally been considered to function as UV filters, and to play an important role in countering the damaging effects of UVB radiation. Although these compounds are effective in reducing cyclobutane pyrimidine dimer (CPD) induction in plants exposed to a challenge exposure to UVB, certain levels of CPD are maintained constitutively in light conditions containing UVB, regardless of the quantity or presence of visible light. These findings imply that the systems for repairing DNA damage and scavenging reactive oxygen species (ROS) are essential for plants to grow in light conditions containing UVB. CONCLUSION: CPD photolyase activity is a crucial factor determining the differences in UVB sensitivity between rice cultivars. The substitution of one or two bases in the CPD photolyase gene can alter the activity of the enzyme, and the associated resistance of the plant to UVB radiation. These findings open up the possibility, in the near future, of increasing the resistance of rice to UVB radiation, by selective breeding or bioengineering of the genes encoding CPD photolyase.  相似文献

The green flagellate Euglena gracilis has been used as a model organism to elucidate the possible large-scale and short-term effects of waste substances from the pulp and paper industry on photosynthetic efficiency (PE). Different concentrations of waste substances before and after treatment in a cleaning system were studied. The uncleaned sample at concentrations up to 1:10 and the cleaned sample at concentrations up to 1:5 showed stimulating effects on the PE after 7 days of incubation compared to the control. The effects of waste substances on the PE of E. gracilis were also studied in combination with short-term studies (20 and 40 min) of ultraviolet-B radiation (UV-B, 280–320 nm). It was shown that increasing concentrations of the uncleaned sample had continuously stimulating effects on the PE and worked protectively against UV-B radiation. The cleaned sample exhibited no effects, or negative effects, on the PE of E. gracilis together with UV-B radiation compared to the experiments with only UV-B radiation. At the concentration 1:1 of the cleaned sample an increase in the PE was detected due to the high concentration of the coloured substances and a decrease in the UV-B penetration. PE revealed itself to be highly sensitive for detecting toxic effects on E. gracilis and is thus very promising for use in regular toxicity tests of waste water from pulp and paper industry. Abstract A β-(1–5)-galactofuran was isolated and characterized from fraction F1S (alkali- and water-soluble) of the cell wall of most of the species of Eupenicillium . In E. cryptum, E. euglaucum and E. nepalense the galactan contained galactofuranose with different linkages in addition to β-(1–5). Fraction F1I (alkali-soluble, water-insoluble) was an α-glucan in certain species while in other it was a =gb-glucan. Xylose was detected in some species in F1I or in F3 (alkali-soluble at 70°C). The most abundant fraction (F4), resistant to the alkali treatment, was a β-glucan-chitin complex. Excepting this component, the β-(1–5)-galactofuran was the polysaccharide which appeared more frequently in the cell wall of species of Eupencillium and it may have chemotaxonomic relevance.  相似文献

We investigate lateral organization of lipid domains in vesicles versus supported membranes and monolayers. The lipid mixtures used are predominantly DOPC/DPPC/Chol and DOPC/BSM/Chol, which have been previously shown to produce coexisting liquid phases in vesicles and monolayers. In a monolayer at an air-water interface, these lipids have miscibility transition pressures of approximately 12-15 mN/m, which can rise to 32 mN/m if the monolayer is exposed to air. Lipid monolayers can be transferred by Langmuir-Sch?fer deposition onto either silanized glass or existing Langmuir-Blodgett supported monolayers. Micron-scale domains are present in the transferred lipids only if they were present in the original monolayer before deposition. This result is valid for transfers at 32 mN/m and also at lower pressures. Domains transferred to glass supports differ from liquid domains in vesicles because they are static, do not align in registration across leaflets, and do not reappear after temperature is cycled. Similar static domains are found for vesicles ruptured onto glass surfaces. Although supported membranes on glass capture some aspects of vesicles in equilibrium (e.g., gel-liquid transition temperatures and diffusion rates of individual lipids), the collective behavior of lipids in large liquid domains is poorly reproduced.  相似文献

Cholesterol incorporation into lipid bilayers, in the form of multilamellar vesicles or extruded large unilamellar vesicles, has been quantitated. To this aim, the cholesterol contents of bilayers prepared from phospholipid:cholesterol mixtures 33-75 mol% cholesterol have been measured and compared with the original mixture before lipid hydration. There is a great diversity of cases, but under most conditions the actual cholesterol proportion present in the extruded bilayers is much lower than predicted. A quantitative analysis of the vesicles is thus required before any experimental study is undertaken.  相似文献

温室条件下,用0(Control)、8.65kJm-2d-1(TI)及11.2KJm-2d-1(t2)不同剂量的UV-B辐射处理蚕豆幼苗。Ca2 .ATPase及Mg2 -ATPase的活性在辐射处理期间下降。在处理21d,T1和T2微粒体膜的MDA含量明显高于对照,同时IUFA急剧下降,且呈明显的剂量效应。14及21d时,膜磷脂的含量也明显下降。脂氧合酶(Lox)活性在第7及14天与对照相比都显著升高,而21d后迅速下降。结果表明,增强UV-B对微粒体膜的伤害可能是一方面导致正常酶合成与分解之间的平衡失调,另一方面导致了膜脂过氧化作用。  相似文献

Seventeen different, chemically defined phosphatidylcholines, dispersed in aqueous medium in the form of large unilamellar vesicles, have been tested for solubilization by the non-ionic detergent Triton X-100. The temperatures (either 20 °C or 45 °C) were such that the bilayers were always in the liquid-disordered state. For each case, the solubilization parameters, Don (total detergent: lipid mole ratio producing the onset of solubilization) and D50 (total detergent: lipid mole ratio producing 50% solubilization), were determined under equilibrium conditions. Both parameters varied generally in parallel. When double bonds were introduced to the acyl chains, other factors remaining constant, solubilization became more difficult, i.e., more detergent was required. Cis-unsaturated phospholipids required more detergent than the corresponding trans-isomers. Increasing chain length in saturated phospholipids between C12 and C16 decreased moderately the detergent/lipid ratios causing solubilization. Acyl and alkyl phospholipids were equally susceptible to Triton X-100 solubilization. Lipid chain order, as measured by DPH fluorescence polarization, seemed to facilitate solubilization, perhaps because more ordered bilayers have a smaller capacity to accommodate detergent monomers without breaking down into lipid-detergent mixed micelles.  相似文献

The lethal balanced strain Va / Ba is the only tool available for analysing the lethal genes on the O chromosome of Drosophila subobscura . This strain presents the X-ray-induced inversions OVIII+210 and the naturally occuring O3+4. Recombination is possible if wild chromosomes carry the O3+4 arrangement without any other overlapping inversion. Loukas et al . (1980) developed a mathematical method to standardize all of the types of wild lethal chromosomes. Although this method has proved useful, results are sometimes anomalous. Here the method is analysed and possible solutions to these anomalies are suggested.  相似文献

We report here on a series of studies aimed at characterization of the structural and dynamical properties of the synthetic lipid diphytanoyl phosphatidylcholine, in multilamellar dispersions and vesicle suspensions.This lipid exhibits no detectable gel to liquid crystalline phase transition over a large temperature range (?120°C to +120°C).Examination of proton nuclear magnetic resonance (NMR) free induction decays obtained from multilayer dispersions of diphytanoyl phosphatidylcholine provided an estimate of the methylene proton order parameter. The estimated magnitude of 0.21 is comparable to those determined for other phospholipids.Sonication of aqueous dispersions of diphytanoyl phosphatidylcholine led to formation of bilayer vesicles as determined by the measurement of the outer/inner choline methyl proton resonances, vesicle sizes in electron micrographs, and comparison of proton NMR linewidths between multilayer and sonicated dispersions. Ultracentrifugation studies of diphytanoyl phosphatidylcholine vesicles in H2O and 2H2O media yielded a value of 1.013 ± 0.026 ml/g for the partial specific volume of this lipid.We have measured spin lattice relaxation rates for the methyl and methylenemethyne protons of the hydrocarbon chains of diphytanoyl phosphatidylcholine in bilayer vesicles over a range of temperatures and at two NMR frequencies (100 and 220 MHz). The observed relaxation rates for the methylene protons in this system were approximately twice those previously reported for dipalmitoyl phosphatidylcholine at comparable temperatures and resonance frequencies, whereas the relaxation rates measured for the methyl protons were greater than those of the straight chain lipid by an order of magnitude.Measurement of the spin lattice relaxation rates of the hydrocarbon protons of the diphytanoyl phosphatidylcholine in a 10 mol% mixture of the branched-chain lipid in a deuterated host lipid, diperdeuteropalmitoyl phosphatidylcholine, showed a discontinuity in the temperature dependence of the proton NMR longitudinal relaxation rates of the branched-chain lipid in the region of the gel to liquid crystalline phase transition temperature of the deuterated dipalmitoyl phosphatidylcholine host lipid. This result may be taken as evidence of lateral phase separation of a liquid cyrstalline phase enriched in diphytanoyl phosphatidylcholine from a gel phase enriched in diperdeuteropalmitoyl phosphatidylcholine at temperatures below the phase transition temperature of deuterated host lipid. This conclusion is supported by the observation of an abrupt change in the hydrocarbon methylene linewidth (at 100 MHz) of 10 mol% diphytanoyl phosphatidylcholine in diperdeuteropalmitoyl phosphatidylcholine over the temperature range where lateral phase separation is taking place according to differential thermograms.  相似文献

Based on previous studies, Candida utilis pyruvate decarboxylase (PDC) proved to be a stable and high productivity enzyme for the production (R)-phenylacetylcarbinol (PAC), a pharmaceutical precursor. However, a portion of the substrate pyruvate was lost to by-product formation. To identify a source of PDC which might overcome this problem, strains of four yeasts -- C. utilis, Candida tropicalis, Saccharomyces cerevisiae and Kluyveromyces marxianus -- were investigated for their PDC biocatalytic properties. Biotransformations were conducted with benzaldehyde and pyruvate as substrates and three experimental systems were employed (in the order of increasing benzaldehyde concentrations): (I) aqueous (soluble benzaldehyde), (II) aqueous/benzaldehyde emulsion, and (III) aqueous/octanol-benzaldehyde emulsion. Although C. utilis PDC resulted in the highest concentrations of PAC and was the most stable enzyme, C. tropicalis PDC was associated with the lowest acetoin formation. For example, in system (III) the ratio of PAC over acetoin was 35 g g(-1) for C. tropicalis PDC and 9.2 g g(-1) for C. utilis PDC. The study thereby opens up the potential to design a PDC with both high productivity and high yield characteristics.  相似文献

Ectotherm animals including insects are known to undergo seasonal restructuring of the cell membranes in order to keep their functionality and/or protect their structural integrity at low body temperatures. Studies on insects so far focused either on fatty acids or on composition of molecular species in major phospholipid classes. Here we extend the scope of analysis and bring results on seasonal changes in minor phospholipid classes, lysophospholipids (LPLs), free fatty acids, phytosterols and tocopherols in heteropteran insect, Pyrrhocoris apterus. We found that muscle tissue contains unusually high amounts of LPLs. Muscle and fat body tissues also contain high amounts of β-sitosterol and campesterol, two phytosterols derived from plant food, while only small amounts of cholesterol are present. In addition, two isomers (γ and δ) of tocopherol (vitamin E) are present in quantities comparable to, or even higher than phytosterols in both tissues. Distinct seasonal patterns of sterol and tocopherol concentrations were observed showing a minimum in reproductively active bugs in summer and a maximum in diapausing, cold-acclimated bugs in winter. Possible adaptive meanings of such changes are discussed including: preventing the unregulated transition of membrane lipids from functional liquid crystalline phase to non-functional gel phase; decreasing the rates of ion/solute leakage; silencing the activities of membrane bound enzymes and receptors; and counteracting the higher risk of oxidative damage to PUFA in winter membranes.  相似文献

The effect of ethanol (ETOH) on the incorporation of [14C]oleic acid (18:1) into lipid in human monocyte-like U937 cells was investigated. With increasing time of exposure to ETOH, the percentage of the label distributed into neutral lipid (NL) declined from 35 per cent (3 h) to 10 per cent (24 h) accompanied by increased incorporation into phospholipid (PL). [14C] 18 : 1 was preferentially incorporated into triglyceride (TG) and phosphatidylcholine (PC), comprising over 65 per cent and 50 per cent of the label associated with NL and PL, respectively. Low concentrations of ETOH (⩽ 1·0 per cent; v/v) had no effect. At concentrations greater than 1·5 per cent, there was enhanced incorporation into TG and diacylglycerol (DAG) in a 24-h incubation period, while at 16 h the label in phosphatidylethanolamine (PE) was decreased. The effect of ETOH on the CDP-choline or ethanolamine pathway was examined by monitoring the incorporation of [3H]choline or [14C]ethanolamine into PC or PE, respectively. At low concentrations ETOH had no effect on either choline uptake or the incorporation into PC. Higher concentrations (≥ 1·5 per cent) for 3 and 6 h resulted in a slightly decreased choline uptake, and the reduction (40–50 per cent) of incorporation into PC suggests that the CDP-choline pathway was inhibited. There was a similar inhibition of the incorporation of [14C]ethanolamine into PE. When the cells were incubated for 3 h in the presence of 2 per cent ETOH and with labelled 18 : 1 and PL-base, the ratios of incorporation (base/18 : 1) into PC and PE fractions decreased, indicating that the major inhibition lay in blockage of the availability of the base moiety for PL formation. Analysis of the distribution of the label into metabolites revealed that ETOH inhibited the conversion of [14C] ethanolamine into [14C]phosphorylethanolamine. The reduction in incorporation was not due to the enhanced breakdown of base-labelled PL. Our results indicate that ETOH has an inhibitory effect on the CDP-choline or ethanolamine pathway.  相似文献
