Ten pony mares that had developed stallion-like sexual behavior as the result of anabolic steroid treatment (boldenone undecylenate, 0.55 mg/kg intramuscularly (i.m.), once weekly for 12 injections) were evaluated for ability to aid in detecting estrus in cycling mares. In across-the-fence estrus detection trials, androgenized mares failed to elicit signs of estrus or diestrus. In 10-min pasture trials, in which each androgenized mare was placed in a group of 10 cycling mares (six of which were estrous), seven of the 10 androgenized mares elicited estrous behavior from one or two mares. Observations of the 10 androgenized mares among a pasture group of 21 cycling mares indicated that approximately one third of the mares in estrus could be identified on the basis of their response to androgenized mares.  相似文献

I have used an exercise involving domestic cats in the General Genetics course at the University of Nebraska-Lincoln for the past 5 years. Using a coherent set of traits in an organism familiar to the students makes it easy to illustrate principles of transmission and population genetics. The one-semester course consists primarily of sophomores and juniors who have either taken a one-semester introductory biology course, a one-semester cell biology course, or have a strong high school biology background. The students are given a handout and asked to determine the genotype at seven unlinked loci of at least one cat. To fill out the form, the students have to grasp such concepts as dominance, incomplete dominance, temperature-sensitive mutations, epistatic interactions, sex linkage, and variable expressivity. Completing the form reinforces these concepts as they observe the cat's phenotype and fill in the genotype. I then analyze the collected data and use it in my lectures on population genetics to illustrate the Hardy-Weinberg equilibrium, calculate allele frequencies, and use statistics. This allows the students to look at population genetics in a very positive light and provides concrete examples of some often misunderstood principles.  相似文献

Computer-based molecular simulation techniques are increasingly used to interpret experimental data on biomolecular systems at an atomic level. Direct comparison between experiment and simulation is, however, seldom straightforward. The available experimental data are limited in scope and generally correspond to averages over both time and space. A critical analysis of the various factors that may influence the apparent degree of agreement between the results of simulations and experimentally measured quantities is presented and illustrated using examples from recent literature.  相似文献

The sternal epithelium of Porcellio scaber was used as a novel model to study the subcellular elemental distribution in control and Ca(2+)-transporting stages in situ. The anterior sternal epithelium (ASE) is specialized for transport of cuticular Ca to sternal CaCO(3) deposits during premolt, and from these deposits during intramolt. The less specialized posterior sternal epithelium transports Ca(2+) to and from the cuticle. In the ASE cells basal [Na], [Cl], and [Mg] are higher than in the apical side. The basal [Na] increases from 105 to 173 mmol/kg dry mass between control and Ca(2+)-transporting stages, accompanied by a decrease in [Cl] and [K]. The [Mg] increases, suggesting transepithelial Mg(2+)-transport. Cytosolic [Ca] varied insignificantly between 4.5 and 5.7 mmol/kg dry mass, however, the number of Ca hot-spots with concentrations between 15 and 50 mmol/kg dry mass increased during transport. Mitochondrial [Ca] decreased in the ASE from 3.3 in the control to 1.0 in the late premolt and to 2.0 mmol/kg dry mass in the intramolt stage. The results suggest Na(+)-dependent mechanisms for transcellular Ca(2+)-transport and the presence of Ca(2+)-binding proteins. Organelles, probably the smooth endoplasmic reticulum, sequester Ca(2+) during intracellular Ca(2+)-transport. A role of mitochondria as a storage site for cuticular Ca is excluded.  相似文献

We have been investigating a fluorescence dual binding biosensor to detect calpastatin. Calpastatin is a protein found in meat and it is a regulator of meat tenderness. The ability to accurately predict the calpastatin concentration of beef with a biological sensor at the time of grading would lead to a more accurate assessment of the overall palatability of beef when it reaches the consumer. Meat can then be labeled as tender or tough, which would greatly enhance meat processors' ability to grade meat, allowing them to recover lost revenue. The biosensor technique utilized the chemical transduction principle of fluorescence resonance energy transfer (FRET). FRET requires the use of two fluorophores, termed a donor and acceptor. In this study, the donor fluorophore was conjugated to the protein, mu-calpain, while the acceptor fluorophore was conjugated to a monoclonal antibody. The results showed that in the presence of calpastatin, the labeled mu-calpain and antibody would bind to calpastatin, reducing the distance between the two proteins and eliciting a measurable change in fluorescence. The FRET dual binding technique was tested in heated and unheated meat extract, and a limit of detection for calpastatin was 120 ng/ml in diluted heated meat extract with no significant response in the unheated meat extract. Stable response times were achieved within 5 min. The proof-of-principle of utilizing a FRET dual binding technique to detect calpastatin in heated meat extract has been established.  相似文献

Administration of a droperidol-fentanyl combination to laboratory rabbits at a dose of 2.5 mg/kg and 0.05 mg/kg body weight, respectively, produced vasodilatation which greatly facilitated blood withdrawal from the central auricular artery. The method eliminated the need for vacuum devices or topical irritants and posed little risk or discomfort to the animal.  相似文献

Thirteen serotypes of Salmonella isolated from imported ornamental aquarium frogs, snails, and their waters were shown to be multi-drug-resistant. Among the resistances exhibited were resistance to gentamicin, sulfamethoxazole-trimethoprim, cephalothin, and nalidixic acid. Frog isolates displayed eight different patterns and snails isolates had two different resistance patterns. The most common serotype, Salmonella typhimurium, was resistant to 18 antibacterials while other salmonellae were resistant to 9 to 16 antibacterials. Resistances in S. typhimurium and S. bovis-morbificans were conjugative and a number of R plasmids participated in the resistance. The plasmid-mediated resistance in S. typhimurium was stable and the levels of resistance conferred were markedly higher than in the other salmonellae tested. Resistance of other serotypes was non-conjugative and resistance to the beta-lactam antibiotics was unstable.  相似文献

Summary The conventional saccharification of cassava root by enzymatic hydrolysis is improved by using a little amount of cellulase and cellobiase in addition to conventional enzyme, glucoamylase. With new saccharification[glucoamylase 0.45SGU/g cassava, cellulase 4.5NCU/g cassava, cellobiase 0.09U/g cassava, pH 4.3, temperature 60°C, total volume 465ml : 100g of cassava/400ml of water], the reaction time was reduced by about 5 hours, the concentration of reducing sugar was increased by 40%, glucose production was enhanced by 10% , and the viscosity was reduced by 30%.  相似文献

Over the past 7 years, there have been a significant number of studies describing the structural and electronic properties, as well as the chemical reactivity, of synthetic peroxomanganese adducts. Many redox-active manganese enzymes, including manganese-containing superoxide dismutases, extradiol catechol dioxygenases, and ribonucleotide reductases, are proposed to feature peroxomanganese intermediates in their catalytic cycles. The recent efforts to model these intermediates using synthetic complexes have thus provided a strong complement to mechanistic studies of the enzymes. This review provides both a summary and a perspective of work in this area, with an emphasis on the relationship between geometric and electronic structure and chemical reactivity for η2-peroxomanganese(III) and η1-alkylperoxomanganese(III) adducts.  相似文献

Pollen walls can be viewed, like all other structural entities, as bodies subject to the laws of mathematics and mechanics. This view is compatible with an interpretation of pollen biology in terms of water relations. The past decade has seen some impressive successes in the metabolic engineering of biotechnologically important plant pathways. However, plant metabolic engineering currently proceeds more by trial and error than by intelligent system design. A change in philosophy away from studying pathways in isolation and towards studying metabolism as a network is necessary. To support this development, improvements in technologies for metabolic analysis, a wider adoption of metabolite-profiling approaches and significant innovations in data analysis methodologies are required.  相似文献

The amount of adenosine triphosphate (ATP) stored in the muscle available for immediate use is limited, and once used, must be resynthesized in the muscle. Ribose, a naturally occurring pentose sugar, helps resynthesize ATP for use in muscles. There have been claims that ribose supplements increase ATP levels and improve performance. Other studies have provided mixed results on the effectiveness of ribose as an ergogenic aid at high doses. None of these studies have compared the impact of the recommended dose of ribose on athletes and nonathletes under exercise conditions that are most conducive for effectiveness. The purpose of this study was to evaluate the effectiveness of ribose as an ergogenic aid at the dose recommended for supplements currently on the market during an exercise trial to maximize its efficacy. Male subjects (n = 11) performed 2 trials 1 week apart. Each trial consisted of three 30-second Wingate tests with a 2-minute recovery between each test. Trials were counterbalanced, with 1 trial being performed with 625 mg of ribose and the other with a placebo. Peak power, mean power, and percent decrease in power were recorded during each Wingate test. Repeated-measures analysis of variance (p > 0.05) found no significant differences between ribose and placebo. These results suggest that ribose had no effect on performance when taken orally, at the dose suggested by the distributor.  相似文献
