Divergent mate preferences and subsequent genetic differentiation between populations has been demonstrated, but its effects on interspecific interactions are unknown. Associated species exploiting these mate preferences, for example, may diverge to match local preferences. We explore this idea in the sexually deceptive, fly‐mimicking daisy, Gorteria diffusa, by testing for association between genetic structure in the fly pollinator (a proxy for mate preference divergence) and geographic divergence in floral form. If genetic structure in flies influences interactions with G. diffusa, we expect phylogeographically distinct flies to be associated with different floral forms. Flies associated with forms exploiting only feeding behavior often belonged to several phylogeographic clades, whereas flies associated with forms exploiting male‐mating behavior always belonged to distinct clades, indicating the possibility of pollinator‐mediated floral divergence through phylogeographic variation in mating preferences of male flies. We tested this hypothesis with reciprocal presentations using male flies from distinct clades associated with separate floral forms. Results show that males from all clades exhibit similar preferences, making pollinator driven divergence through geographic variation in mate preference unlikely. Males, however, showed evidence of learned resistance to deceptive traits, suggesting antagonistic interactions between plants and pollinators may drive deceptive floral trait evolution in G. diffusa.  相似文献

Honeybee colonies offer an excellent environment for microbial pathogen development. The highest virulent, colony killing, bacterial agents are Paenibacillus larvae causing American foulbrood (AFB), and European foulbrood (EFB) associated bacteria. Besides the innate immune defense, honeybees evolved behavioral defenses to combat infections. Foraging of antimicrobial plant compounds plays a key role for this “social immunity” behavior. Secondary plant metabolites in floral nectar are known for their antimicrobial effects. Yet, these compounds are highly plant specific, and the effects on bee health will depend on the floral origin of the honey produced. As worker bees not only feed themselves, but also the larvae and other colony members, honey is a prime candidate acting as self‐medication agent in honeybee colonies to prevent or decrease infections. Here, we test eight AFB and EFB bacterial strains and the growth inhibitory activity of three honey types. Using a high‐throughput cell growth assay, we show that all honeys have high growth inhibitory activity and the two monofloral honeys appeared to be strain specific. The specificity of the monofloral honeys and the strong antimicrobial potential of the polyfloral honey suggest that the diversity of honeys in the honey stores of a colony may be highly adaptive for its “social immunity” against the highly diverse suite of pathogens encountered in nature. This ecological diversity may therefore operate similar to the well‐known effects of host genetic variance in the arms race between host and parasite.  相似文献

Most of our knowledge on learning and memory formation results from extensive studies on a small number of animal species. Although features and cellular pathways of learning and memory are highly similar in this diverse group of species, there are also subtle differences. Closely related species of parasitic wasps display substantial variation in memory dynamics and can be instrumental to understanding both the adaptive benefit of and mechanisms underlying this variation. Parasitic wasps of the genus Nasonia offer excellent opportunities for multidisciplinary research on this topic. Genetic and genomic resources available for Nasonia are unrivaled among parasitic wasps, providing tools for genetic dissection of mechanisms that cause differences in learning. This study presents a robust, high‐throughput method for olfactory conditioning of Nasonia using a host encounter as reward. A T‐maze olfactometer facilitates high‐throughput memory retention testing and employs standardized odors of equal detectability, as quantified by electroantennogram recordings. Using this setup, differences in memory retention between Nasonia species were shown. In both Nasonia vitripennis and Nasonia longicornis, memory was observed up to at least 5 days after a single conditioning trial, whereas Nasonia giraulti lost its memory after 2 days. This difference in learning may be an adaptation to species‐specific differences in ecological factors, for example, host preference. The high‐throughput methods for conditioning and memory retention testing are essential tools to study both ultimate and proximate factors that cause variation in learning and memory formation in Nasonia and other parasitic wasp species.  相似文献

Some parasitoid flies exploit odors derived from plants as olfactory cues for locating the food plants of host insects, but the role of visual cues associated with plants remains largely unknown. The generalist tachinid Exorista japonica Townsend (Diptera: Tachinidae) is attracted to odors derived from maize plants [Zea mays L. (Poaceae)] infested by the larvae of Mythimna separata (Walker) (Lepidoptera: Noctuidae). In this study, we examined the effects of visual parameters on the olfactory attraction of female flies to host‐infested plants. A paper plant model of one of four colors (blue, green, yellow, or red) was placed in front of a host‐infested plant, which was hidden behind a mesh screen in a wind tunnel. The landing rate of females was significantly higher on the green plant model than on the other three models. When an achromatic plant model of one of four gray scales (white, light gray, dark gray, or black) was tested, the response rate of females was significantly higher towards the white model and decreased as the brightness of models decreased. Few female flies responded to the green plant model without odors of the host‐infested plants. When the four color plant models were placed together in a cage filled with odors of host‐infested plants, females remained significantly longer on the green model than on the other three models. These results showed that E. japonica females preferred the color green when odors of the host‐infested plants were present and suggest that E. japonica uses visual as well as olfactory cues to locate the host habitat.  相似文献

1 Chemical cues involved in host location by Dastarcus helophoroides (Fairmaire) (Coleoptera: Bothrideridae), a parasitoid of Anoplophora glabripennis Motschulsky, Monochamus alternatus Hope, Massicus raddei (Blessig), Apriona germari (Hope), Apriona swainsoni (Hope) and Batocera horsfielde (Hope) in the family Cerambycidae, were investigated in a Y‐tube olfactometer. Responses of D. helophoroides adults toward odour sources from their host (M. raddei) and host tree (Quercus mongolicus Fisch. ex. Turcz.) were tested in either no‐choice or two‐way choice tests. Wood with larval tunnels and larvae frass were attractive to D. helophoroides. Both sexes showed attraction to wood with larval tunnels. 2 Volatile chemicals emitted from wood with larval tunnels and larval frass were collected on Porapak Q and analyzed by gas chromatography‐mass spectrometry. Several compounds identified from these sources, including α‐pinene, β‐pinene, Δ‐3‐carene, limonene and β‐myrcene, were tested against D. helophoroides in the same Y‐tube olfactometer. 3 The result showed that D. helophoroides adults were attracted to (R)‐(+)‐limonene, with or without the background odour from uninfested fresh wood. This may suggest that (R)‐(+)‐limonene is an important kairomone for this parasitic beetle. 4 Dose‐dependent response of D. helophoroides adults to (R)‐(+)‐limonene indicated that D. helophoroides adults showed significant attraction to (R)‐(+)‐limonene when release rates exceeded 2.08 μg/h in Y‐tube olfactometer tests. Moreover, significantly more A. glabripennis larvae were parasitized by D. helophoroides in the presence of R‐(+)‐limonene, relative to the control in cage experiments.  相似文献

Invasive species can be detrimental to a nation's ecology, economy and human health. Rapid and accurate diagnostics are critical to limit the establishment and spread of exotic organisms. The increasing rate of biological invasions relative to the taxonomic expertise available generates a demand for high‐throughput, DNA‐based diagnostics methods for identification. We designed species‐specific qPCR primer and probe combinations for 27 economically important tephritidae species in six genera (Anastrepha, Bactrocera, Carpomya, Ceratitis, Dacus and Rhagoletis) based on 935 COI DNA barcode haplotypes from 181 fruit fly species publically available in BOLD, and then tested the specificity for each primer pair and probe through qPCR of 35 of those species. We then developed a standardization reaction system for detecting the 27 target species based on a microfluidic dynamic array and also applied the method to identify unknown immature samples from port interceptions and field monitoring. This method led to a specific and simultaneous detection for all 27 species in 7.5 h, using only 0.2 μL of reaction system in each reaction chamber. The approach successfully discriminated among species within complexes that had genetic similarities of up to 98.48%, while it also identified all immature samples consistent with the subsequent results of morphological examination of adults which were reared from larvae of cohorts from the same samples. We present an accurate, rapid and high‐throughput innovative approach for detecting fruit flies of quarantine concern. This is a new method which has broad potential to be one of international standards for plant quarantine and invasive species detection.  相似文献

Queensland fruit fly [Bactrocera tryoni (Froggatt), Diptera, Tephritidae] is the most devastating insect pest impacting Australian horticulture. The Sterile Insect Technique (SIT) is an important component of tephritid pest management programs. However, mass‐rearing and irradiation (to render insects sterile) may reduce the fitness and performance of the insect, including the ability of sterile males to successfully compete for wild females. Manipulation of the gut microbiome, including the supplementation with bacterial probiotics shows promise for enhancing the quality of mass‐reared sterile flies, however there are fewer published studies targeting the larval stage. In this study, we supplemented the larval stage of mass‐reared B. tryoni with bacterial probiotics. We tested several individual bacteria that had been previously isolated and characterized from the gut of wild B. tryoni larvae including Asaia sp., Enterobacter sp., Lactobacillus sp., Leuconostoc sp. We also tested a consortium of all four of these bacterial isolates. The fitness parameters tested included adult survival in field cages, laboratory mate selection of bacteria supplemented males by bacteria nonsupplemented females, and laboratory locomotor activity of adult flies. None of the bacterial probiotic treatments in the current study was significantly different to the control for field survival, mate selection or locomotor activity of adult B. tryoni, which agree with some of the other studies regarding bacterial probiotics fed to the larval stage of tephritids. Future work is needed to determine if feeding the same, and/or other probiotics to adults, as opposed to larvae can positively impact survival, mating performance, mating competitiveness and locomotor activity of B. tryoni. The bacterial group(s) and function of bacterial species that increase fitness and competitiveness is also of interest to tephritid mass‐rearing programs.  相似文献

Phlebotomine sand flies are haematophagous dipterans of primary medical importance. They represent the only proven vectors of leishmaniasis worldwide and are involved in the transmission of various other pathogens. Studying the ecology of sand flies is crucial to understand the epidemiology of leishmaniasis and further control this disease. A major limitation in this regard is that traditional morphological‐based methods for sand fly species identifications are time‐consuming and require taxonomic expertise. DNA metabarcoding holds great promise in overcoming this issue by allowing the identification of multiple species from a single bulk sample. Here, we assessed the reliability of a short insect metabarcode located in the mitochondrial 16S rRNA for the identification of Neotropical sand flies, and constructed a reference database for 40 species found in French Guiana. Then, we conducted a metabarcoding experiment on sand flies mixtures of known content and showed that the method allows an accurate identification of specimens in pools. Finally, we applied metabarcoding to field samples caught in a 1‐ha forest plot in French Guiana. Besides providing reliable molecular data for species‐level assignations of phlebotomine sand flies, our study proves the efficiency of metabarcoding based on the mitochondrial 16S rRNA for studying sand fly diversity from bulk samples. The application of this high‐throughput identification procedure to field samples can provide great opportunities for vector monitoring and eco‐epidemiological studies.  相似文献

Stable flies (Stomoxys calcitrans [Diptera: Muscidae] L.) are blood‐feeding synanthropic pests, which cause significant economic losses in livestock. Stable fly antennae contain olfactory sensilla responsive to host and host environment‐associated odours. Field observation indicated that the abundance of stable flies increased significantly in grasslands or crop fields when cattle manure slurry was applied. Major volatile compounds emanating from manure slurry were collected and identified. Behavioural responses of stable flies to those compounds were investigated in laboratory bioassays and field‐trapping studies. Results from olfactometer assays revealed that phenol, p‐cresol and m‐cresol were attractive to adult stable flies. When tested individually, attraction was higher with lower dosages. Stable flies were most attracted to blends of phenol and m‐cresol or p‐cresol. Traps with binary blend lures caught more stable flies in field trials as well.  相似文献

Abstract 1. Enemy‐free space has been shown to mediate host shifts in herbivores, but this has not previously been documented in parasitoids. Also, natural enemies shown to maintain host shifts have always been from higher trophic levels, rather than competitors. 2. In Hawaii, an Australian parasitoid (Diachasmimorpha tryoni) of medflies that loses competition contests to a subsequently introduced Asian parasitoid (Fopius arisanus) has shifted its realised host range to attack non‐target gall flies on lantana. 3. The present study demonstrates experimentally that D. tryoni reproduction is: (i) lower on medflies in coffee when F. arisanus is present than when it is absent; (ii) higher in gall flies on lantana than on medflies in coffee, when F. arisanus is present; and (iii) higher in medflies on coffee than in gall flies on lantana, when F. arisanus is absent. This meets Berdegue et al.’s (Ecological Entomology, 21 , 203–217, 1996) three conditions to confirm the importance of enemy‐free space. 4. In the field, F. arisanus is abundant on medflies, whereas D. tryoni is rare on medflies, but is the dominant parasitoid of lantana gall flies. 5. Competitor‐free space is thus shown to be a key mechanism maintaining an apparent host shift by an introduced biocontrol agent onto a non‐target species.  相似文献

Abstract The olfactory responses of Aphidius gifuensis to odors from two host plants (Nicotiana tabacum and Brassica napus ssp.) and their complexes with different infestation levels of two host aphids (Myzus persicae and Lipaphis erysimi) were respectively examined in an olfactometer. The results showed that female A. gifuensis did not respond to odors of undamaged or mechanically damaged host plants, but significantly responded to odors of aphid/plant complexes. Moreover, A. gifuensis responded significantly to odors of both M. persicae and L. erysimi/plant complexes when host plants were infested by high levels of aphids, suggesting that quantity of aphid‐induced volatiles could be important for attracting A. gifuensis. When tested between aphid/plant complexes, A. gifuensis did not show its preference for either complex. The efficiency of A. gifuensis against aphids in open fields potentially could be improved by using its olfactory response to aphid/plant complexes.  相似文献

Melon fly, Zeugodacus cucurbitae (Coquillett) (Diptera: Tephritidae), is an important quarantine tephritid fruit fly with resident populations established in Hawai'i, USA. In the male‐annihilation approach, male flies are targeted using dispensers with cue‐lure (C‐L) and insecticides, typically organophosphates. The efficacy of the male annihilation approach is thought to be limited to individual male flies, contacting the lure and the pesticide, after which they die. Alternative classes of insecticides, such as fipronil, have been investigated for use in male‐annihilation. We hypothesized that ingestion of fipronil by male flies could lead to horizontal transfer and mortality in female flies. Horizontal insecticide transfer extends pesticide control beyond the individual contacting the toxicant through indirect contact via food sharing or other mechanisms. We tested the possibility for horizontal transfer of fipronil from male to female Z. cucurbitae through field and laboratory studies. Two repeated field trials were conducted to compare the numbers of female flies collected in fields treated with Amulet C‐L (0.34% fipronil active ingredient) bait stations, sanitation, and spot treatments of GF‐120 Fruit Fly Bait to numbers collected in fields where sanitation and spot‐treatments were used without Amulet C‐L. In fields with Amulet C‐L bait stations in conjunction with sanitation and weekly protein bait spot treatments of GF‐120 Fruit Fly Bait, female captures were significantly lower than those in field plots treated with weekly protein bait spot treatments and sanitation. In subsequent laboratory studies, all females died within 6 h after direct exposure to male flies that had access to Amulet C‐L for 1–4 min. The possibility that male regurgitant could be a mechanism for horizontal transfer and subsequent female mortality was determined by collecting regurgitated droplets from fipronil‐fed male flies and feeding them to males and females. Both male and female flies exposed to regurgitant from fipronil‐fed male flies or droplets containing fipronil had higher mortality than the male and female flies that were exposed to regurgitant or droplets with only the C‐L compound or sugar solution. Thus, female flies do experience mortality from exposure to regurgitant from males that have fed on fipronil laced solutions. This provides evidence of at least one mechanism of horizontal transfer of insecticide in tephritid fruit flies. These findings are discussed in the context of Zcucurbitae integrated pest management programs in Hawai'i.  相似文献

To evaluate the attractiveness of several mushroom‐growing substrates to the female mushroom fly Lycoriella ingenua (Dufour) (Diptera: Sciaridae), a pest of the cultivated white button mushroom, Agaricus bisporus (JE Lange) Emil J Imbach (Agaricales), we developed a two‐choice, static‐flow olfactometer. Behavioral assays using this olfactometer indicated that mushroom compost with A. bisporus mycelia growing in it was not more attractive than compost lacking growing mycelia. We also found that female flies were more attracted to compost lacking A. bisporus mycelia than to the actual commodity, the white button mushroom fruiting bodies. Flies were not, however, attracted to sterilized compost, suggesting the attraction is due to volatiles produced by microbial metabolism in the compost. We also found that female L. ingenua flies were attracted to the mycoparasitic green mold Trichoderma aggressivum Samuels & W Gams (Hypocreales). Flies preferred mushroom compost that had T. aggressivum growing in it over compost lacking T. aggressivum, providing an experimental outcome consistent with the anecdotal belief that L. ingenua flies are vectors of T. aggressivum spores that can infest mushroom‐growing houses.  相似文献

Analysis of genetic diversity and population structure among Quercus fabri populations is essential for the conservation and utilization of Q. fabri resources. Here, the genetic diversity and structure of 158 individuals from 13 natural populations of Quercus fabri in China were analyzed using genotyping‐by‐sequencing (GBS). A total of 459,564 high‐quality single nucleotide polymorphisms (SNPs) were obtained after filtration for subsequent analysis. Genetic structure analysis revealed that these individuals can be clustered into two groups and the structure can be explained mainly by the geographic barrier, showed gene introgression from coastal to inland areas and high mountains could significantly hinder the mutual introgression of genes. Genetic diversity analysis indicated that the individual differences within groups are greater than the differences between the two groups. These results will help us better understand the genetic backgrounds of Q. fabri.  相似文献

Many arctic‐alpine plant genera have undergone speciation during the Quaternary. The bases for these radiations have been ascribed to geographic isolation, abiotic and biotic differences between populations, and/or hybridization and polyploidization. The Cordilleran Campanula L. (Campanulaceae Juss.), a monophyletic clade of mostly endemic arctic‐alpine taxa from western North America, experienced a recent and rapid radiation. We set out to unravel the factors that likely influenced speciation in this group. To do so, we integrated environmental, genetic, and morphological datasets, tested biogeographic hypotheses, and analyzed the potential consequences of the various factors on the evolutionary history of the clade. We created paleodistribution models to identify potential Pleistocene refugia for the clade and estimated niche space for individual taxa using geographic and climatic data. Using 11 nuclear loci, we reconstructed a species tree and tested biogeographic hypotheses derived from the paleodistribution models. Finally, we tested 28 morphological characters, including floral, vegetative, and seed characteristics, for their capacity to differentiate taxa. Our results show that the combined effect of Quaternary climatic variation, isolation among differing environments in the mountains in western North America, and biotic factors influencing floral morphology contributed to speciation in this group during the mid‐Pleistocene. Furthermore, our biogeographic analyses uncovered asynchronous consequences of interglacial and glacial periods for the timing of refugial isolation within the southern and northwestern mountains, respectively. These findings have broad implications for understanding the processes promoting speciation in arctic‐alpine plants and the rise of numerous endemic taxa across the region.  相似文献

When an herbivorous insect enters a new geographic area, it will select host plants based on short and long distance cues. A conifer-feeding bark beetle that has been recently introduced to North America, the Mediterranean pine engraver, Orthotomicus erosus (Wollaston), has a potentially wide host range, especially among members of the Pinaceae. The long-distance response of the beetles to tree odors may be a key feature of the mechanism of host recognition and selection. We used a laboratory olfactometer to study the walking response of 1,440 O. erosus to odor cues from the bark and phloem of six North American tree species. The beetle moved toward the angiosperm non-host Betula papyrifera more than would be expected by chance, but had a neutral response to odors of two tree species that support reproduction and three species that do not. These results suggest that tree odors alone may not be adequate for O. erosus to recognize novel hosts.  相似文献

Floral variation among closely related species is thought to often reflect differences in pollination systems. Flowers of the large genus Impatiens are characterized by extensive variation in colour, shape and size and in anther and stigma positioning, but studies of their pollination ecology are scarce and most lack a comparative context. Consequently, the function of floral diversity in Impatiens remains enigmatic. This study documents floral variation and pollination of seven co‐occurring Impatiens spp. in the Southeast Asian diversity hotspot. To assess whether floral trait variation reflects specialization for different pollination systems, we tested whether species depend on pollinators for reproduction, identified animals that visit flowers, determined whether these visitors play a role in pollination and quantified and compared key floral traits, including floral dimensions and nectar characteristics. Experimental exclusion of insects decreased fruit and seed set significantly for all species except I. muscicola, which also received almost no visits from animals. Most species received visits from several animals, including bees, birds, butterflies and hawkmoths, only a subset of which were effective pollinators. Impatiens psittacina, I. kerriae, I. racemosa and I. daraneenae were pollinated by bees, primarily Bombus haemorrhoidalis. Impatiens chiangdaoensis and I. santisukii had bimodal pollination systems which combined bee and lepidopteran pollination. Floral traits differed significantly among species with different pollination systems. Autogamous flowers were small and spurless, and did not produce nectar; bee‐pollinated flowers had short spurs and large floral chambers with a wide entrance; and bimodally bee‐ and lepidopteran‐pollinated species had long spurs and a small floral chamber with a narrow entrance. Nectar‐producing species with different pollination systems did not differ in nectar volume and sugar concentration. Despite the high frequency of bee pollination in co‐occurring species, individuals with a morphology suggestive of hybrid origin were rare. Variation in floral architecture, including various forms of corolla asymmetry, facilitates distinct, species‐specific pollen‐placement on visiting bees. Our results show that floral morphological diversity among Impatiens spp. is associated with both differences in functional pollinator groups and divergent use of the same pollinator. Non‐homologous mechanisms of floral asymmetry are consistent with repeated independent evolution, suggesting that competitive interactions among species with the same pollination system have been an important driver of floral variation among Impatiens spp.  相似文献

Nemorilla maculosa Meigen (Diptera: Tachinidae) is a solitary endoparasitoid of the legume pod borer, Maruca vitrata Fabricius (Lepidoptera: Crambidae), a key pest of cowpea, Vigna unguiculata (L.) Walp. (Fabaceae) in Africa. A colony of N. maculosa, introduced for experimental purposes from Taiwan to the laboratories of the International Institute of Tropical Agriculture (IITA) in Benin, was used for our studies. Olfactory reponses of N. maculosa to leaves of infested or uninfested cowpea and yellow peabush, Sesbania cannabina (Retz.) Pers. (Fabaceae), and to M. vitrata larvae were evaluated in a four‐arm olfactometer. For all combinations of odor sources, responses between naïve and oviposition‐experienced female flies did not differ. Nemorilla maculosa females were attracted by odors from uninfested leaves of yellow peabush and flowers of cowpea when compared with clean air, and they were attracted to plants damaged by M. vitrata with larvae removed. However, the female fly did not discriminate between odors from infested and uninfested plants. The parasitic fly N. maculosa proved well able to use volatile compounds from various host plants (peabush and cowpea) to locate its host, with a more pronounced attraction by the combination of host larvae and infested host plant parts. These findings are discussed in light of the prospective use of N. maculosa as a biological control agent against the legume pod borer.  相似文献

A one‐step multiplex RT‐PCR method has been developed for the simultaneous detection of four viruses frequently occurring in tobacco (Cucumber mosaic virus, Tobacco mosaic virus, Tobacco etch virus and Potato virus Y). Four sets of specific primers were designed to work with the same reaction reagents and cycling conditions, resulting in four distinguishable amplicons representative of the four viruses independently. This one‐step multiplex RT‐PCR is consistently specific using different combinations of virus RNA as templates, and no non‐specific band was observed. It has high sensitivity compared to single RT‐PCR. Moreover, field samples in China can be tested by this method for virus detection. Our results show that one‐step multiplex RT‐PCR is a high‐throughput, specific, sensitive method for tobacco virus detection.  相似文献
