Transport of cargo to, through and from the Golgi complex is mediated by vesicular carriers and transient tubular connections. In this review, we describe vesicle tethering events with the understanding that similar events occur during transport via larger structures. Tethering factors can be generally divided into a group of coiled-coil proteins and a group of multi-subunit complexes. Current evidence suggests that these factors function in a variety of membrane-membrane tethering events at the Golgi complex, interact with SNARE molecules, and are regulated by small GTPases of the Rab and Arl families.  相似文献

To date, three different structural gene mutations have been identified in patients with carbonic anhydrase II deficiency (osteopetrosis with renal tubular acidosis and cerebral calcification). These include a missense mutation (H107Y) in two families, a splice junction mutation in intron 5 in one of these families, and a splice junction mutation in intron 2 for which many Arabic patients are homozygous. We report here a novel mutation for which carbonic anhydrase II-deficient patients from seven unrelated Hispanic families were found to be homozygous. The proband was a 2 1/2-year-old Hispanic girl of Puerto Rican ancestry who was unique clinically, in that she had no evidence of renal tubular acidosis, even though she did have osteopetrosis, developmental delay, and cerebral calcification. She proved to be homozygous for a single-base deletion in the coding region of exon 7 that produces a frameshift that changes the next 12 amino acids before leading to chain termination and that also introduces a new MaeIII restriction site. The 27-kD truncated enzyme produced when the mutant cDNA was expressed in COS cells was enzymatically inactive, present mainly in insoluble aggregates, and detectable immunologically at only 5% the level of the 29-kD normal carbonic anhydrase II expressed from the wild-type cDNA. Metabolic labeling revealed that this 27-kD mutant protein has an accelerated rate of degradation. Six subsequent Hispanic patients of Caribbean ancestry, all of whom had osteopetrosis and renal tubular acidosis but who varied widely in clinical severity, were found to be homozygous for the same mutation.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Structure of nonmodified scales and lateral-line scales (Sll) is compared in 179 species of 105 genera of 13 families of the order Pleuronectiformes. Morphological diversity of Sll depends on the level of development and of interaction of two component parts—the scale plate and the tubular part. Of the considered characters, the most individually variable character is whether the tubular part is complete or incomplete. The interspecies differences may manifest themselves in structure of the scale plate, the level of unification of opening of the tubular part and of the cutaneous canaliculus, or in completeness or incompleteness of the tubular part. As a rule, Sll in species of the same genus is similar in structure. In some cases, special traits of this structure may be diagnostic characters of genera and contribute to the solution of controversial issues of generic position of species. The families of Pleuronectiformes are characterized by a different ratio of taxa with certain involved characters of nonmodified scales and Sll. However, their distribution in this order does not give a simple idea on their relationships and on the direction of evolutionary transformations. This follows from the fact that the structure of Sll depends on a wide complex of factors—morphogenesis of nonmodified scales, level of ossification of trunk canal, time of anlage, and mutual influence of these structures at early stages of ontogenesis.  相似文献

Summary Four families of chelonians, the Carettochelyidae, Trionchidae, Pelomedusidae and Chelidae, possess nasal erectile tissue and masses of smooth muscle within the snout. These specialized tissues appear to control the internal diameter of the narial passage. Two of these families, the carettochelids and trionchilds, can change the angle of the tubular snout relative to the head. The trionchids have numerous pharyngeal papillae that function as respiratory surfaces. The angle of the snout and its internal diameter are important to these animals in utilizing the snout to draw clear water into the buccal cavity while buried in the substrate.  相似文献

Epithelial–mesenchymal transition (EMT) of tubular cells is one of the mechanisms which contribute to renal fibrosis and transforming growth factor-β (TGF-β) is one of the main triggers. Heparanase (HPSE) is an endo-β-D-glucuronidase that cleaves heparan-sulfate thus regulating the bioavailability of growth factors (FGF-2, TGF-β). HPSE controls FGF-2-induced EMT in tubular cells and is necessary for the development of diabetic nephropathy in mice.The aim of this study was to investigate whether HPSE can modulate the expression and the effects of TGF-β in tubular cells.First we proved that the lack of HPSE or its inhibition prevents the increased synthesis of TGF-β by tubular cells in response to pro-fibrotic stimuli such as FGF-2, advanced glycosylation end products (AGE) and albumin overload.Second, since TGF-β may derive from sources different from tubular cells, we investigated whether HPSE modulates tubular cell response to exogenous TGF-β. HPSE does not prevent EMT induced by TGF-β although it slows its onset; indeed in HPSE-silenced cells the acquisition of a mesenchymal phenotype does not develop as quickly as in wt cells. Additionally, TGF-β induces an autocrine loop to sustain its signal, whereas the lack of HPSE partially interferes with this autocrine loop.Overall these data confirm that HPSE is a key player in renal fibrosis since it interacts with the regulation and the effects of TGF-β. HPSE is needed for pathological TGF-β overexpression in response to pro-fibrotic factors. Furthermore, HPSE modulates TGF-β-induced EMT: the lack of HPSE delays tubular cell transdifferentiation, and impairs the TGF-β autocrine loop.  相似文献

Although the superficial similarity between Polygalaceae and Fabaceae flowers is well known, a comparison between their recently more precisely defined 'keel flowers' reveals a wealth of functional congruences with regard to visual attraction, flower mechanics (interplay of fixed and mobile floral parts, presence of special contrivances such as tongue guide, foot handles, pollen cache, etc.), pollen presentation and nectar storage. Although keel flowers are principally addressed to bees, the increasingly pronounced fusion of floral parts gave rise to tubular flowers and has widened the spectrum of pollinators in the Polygalaceae. The intimate functional correspondence does not affect the assessment that the floral architecture (in terms of homologies) is quite different between the families. This is discussed with reference to the sistergroup relationship of the two families emanating from molecular systematics.  相似文献

The abdominal lateral line system in Trichoptera, consisting of filaments and lamellae with associated primary and secondary setae, was studied in larvae and pupae of 70 genera representing 26 families. Filaments are slender, generally bifid, tubular extensions of the cuticle that together form a fringe on the lateral abdominal surface of larvae and pupae of many but not all families of case-making Trichoptera (Integripalpia). Larvae and pupae of the Annulipalpia and Spicipalpia lack a lateral fringe of bifid filaments, but single filaments occur in larvae of a few unrelated genera. A lateral fringe may help in regulating the flow of water which is maintained through the case by abdominal undulation. Lamellae occur only in larvae of the Integripalpia, where they are variously distributed among families and genera, and probably have a sensory function. Three types of lamellae are recognized: forked, serrate and single. Filaments and lamellae have significant taxonomic value, and their patterns of occurrence raise questions about the phylogeny of Trichoptera.  相似文献

In angiogenesis associated with tissue repair and disease, fibrin and inflammatory mediators are often involved. We have used three- dimensional fibrin matrices to investigate the humoral requirements of human microvascular endothelial cells (hMVEC) to form capillary-like tubular structures. bFGF and VEGF165 were unable to induce tubular structures by themselves. Simultaneous addition of one or both of these factors with TNFalpha induced outgrowth of tubules, the effect being the strongest when bFGF, VEGF165, and TNFalpha were added simultaneously. Exogenously added u-PA, but not its nonproteolytic amino-terminal fragment, could replace TNFalpha, suggesting that TNFalpha-induced u-PA synthesis was involved. Soluble u-PA receptor (u- PAR) or antibodies that inhibited u-PA activity prevented the formation of tubular structures by 59-99%. epsilon-ACA and trasylol which inhibit the formation and activity of plasmin reduced the extent of tube formation by 71-95%. TNFalpha or u-PA did not induce tubular structures without additional growth factors. bFGF and VEGF165 enhanced of the u- PAR by 72 and 46%, but TNFalpha itself also increased u-PAR in hMVEC by 30%. Induction of mitogenesis was not the major contribution of bFGF and VEGF165 because the cell number did not change significantly in the presence of TNFalpha, and tyrphostin A47, which inhibited mitosis completely, reduced the formation of tubular structures only by 28-36%. These data show that induction of cell-bound u-PA activity by the cytokine TNFalpha is required in addition to the angiogenic factors VEGF165 and/or bFGF to induce in vitro formation of capillary-like structures by hMVEC in fibrin matrices. These data may provide insight in the mechanism of angiogenesis as occurs in pathological conditions.  相似文献

BAR and ENTH domains are families of alpha-helical lipid bilayer binding modules found in proteins that function in endocytosis, actin regulation and signaling. Several members of these families not only bind the bilayer, but also participate in the regulation of its curvature. These properties are thought to play physiological roles at sites of membrane budding and at other sites where narrow tubular membranes occur in vivo. Studies of BAR and ENTH domains and of their flanking regions have provided new insights into mechanisms of membrane deformation and curvature sensing, and have emphasized the importance of amphipathic helices, thought to intercalate in one of the leaflets of the lipid bilayer, in the generation of membrane curvature. Structural studies and database searches are rapidly expanding the BAR and ENTH domains families, with the identification of new related domains and subfamilies, such as F-BAR (also called EFC) domains and ANTH domains, respectively. Here we present a short overview of the properties of these domains based on evidence obtained from genetics, cell biology, biochemistry and structural biology.  相似文献

In distal renal tubular acidosis (dRTA) the tubular secretion of hydrogen ion in the distal nephron is impaired, leading to the development of metabolic acidosis, frequently accompanied by hypokalemia, nephrocalcinosis, and metabolic bone disease. The condition can be familial, when it is usually inherited as an autosomal dominant, though there is a rarer autosomal recessive form associated with nerve deafness. It has been shown that the autosomal dominant form of dRTA is associated with a defect in the anion exchanger (AE1) of the renal collecting duct intercalated cell. This transporter is a product of the same gene (AE1) as the erythrocyte anion exchanger, band 3. In this review we will look at the evidence for this association. Studies of genomic DNA from families with this disorder have shown, both by genetic linkage studies and by DNA sequencing, that affected individuals are heterozygous for mutations in the AE1 gene whilst unaffected family members have a normal band 3 sequence. Mutations have been found in the region of proposed helices 6 and 7 of the membrane domain of band 3 and involve amino acids Arg-589 and Ser-613, and in the COOH-terminal domain of band 3. Studies of red cell band 3 from these families have provided information on the effect these mutations have on the structure and function of erythrocyte band 3. Expression studies of the erythroid and kidney isoforms of the mutant AE1 proteins, in Xenopus laevis oocytes, have shown that they retained chloride transport activity, suggesting that the disease in the dRTA families is not related simply to the anion transport activity of the mutated proteins. A possible explanation for the dominant effect of these mutant AE1 proteins in the kidney cell is that these mutations affect the targeting of AE1 from the basolateral to the apical membrane of the alpha-intercalated cell.  相似文献

Sixty-eight patients with osteogenesis imperfecta (OI) classified according to Sillence were evaluated for dentinogenesis imperfecta (DI). Orthopantomograms of 51 of the 68 were examined. Type I DI was recognized in 22 patients from 16 families. DI was observed in 4/45 patients with type I OI, in one of two patients with type III, and in 13/16 patients with type IV OI. Four of the five patients with an unidentified type of OI had DI. The expression of type I DI was variable. Discoloration and pulpal obliteration were the major manifestations. Teeth from 14 patients from 12 families were studied histologically. Eight of the 14 patients were from six families who had clinical and/or radiographic evidence of DI. Irregularity of the dentin matrix and tubular pattern in the circumpulpal dentin and normal mantle dentin were observed. Interfamilial variability was greater than intrafamilial variability. The expression of DI was mild in one family with type I OI. There was no further relation between the type of OI and the severity of DI.  相似文献

Terminal flower-like structures (TFLS) occur in many angiosperms that possess indeterminate inflorescences such as spikes, racemes, or spadices. We describe and review TFLS in early-divergent angiosperms, especially the magnoliid order Piperales and the monocot order Alismatales, in which floral interpretation is controversial. Essentially similar TFLS occur in a wide range of taxa. Among magnoliids, they occur in some Piperales (Saururaceae and a few Piperaceae), but are absent from Chloranthaceae. Among monocots, they occur in some early-divergent families such as Acoraceae, Aponogetonaceae, Juncaginaceae, Potamogetonaceae, and Ruppiaceae. Similar TFLS with obscure organ identity are recorded in mutants of Arabidopsis. TFLS can often be interpreted as pseudanthia (close aggregations of reduced flowers), but in some cases the entire terminal pseudanthium is very similar to a true flower. In some cases, elaborated TFLS could therefore have given rise to what are normally termed 'true' (i.e. euanthial) flowers. Data presented here on terminal pseudanthia in Potamogeton and Ruppia support a pseudanthial evolutionary origin of reproductive units in the alismatid families Zannichelliaceae and Cymodoceaceae. Furthermore, in some alismatid species, either the entire inflorescence apex or an individual primordium at or near the inflorescence tip can be transformed into a filamentous or tubular (or intermediate) structure. A tubular structure enclosing stamens and carpels is described in Piper. This indicates that pseudanthium formation can provoke morphological novelties, perhaps due to new patterns of overlap between expression zones of regulatory genes and/or new spatial constraints.  相似文献

High Performance Liquid Chromatography (HPLC) was used to determine specific sugar ratios (fructose, glucose and sucrose) in nectars of nine families of flowering plants. All nectars contained all three sugars with the exception of that of Asclepias. Asclepias nectar was nearly a pure sucrose solution. Sucrose/hexose ratio was correlated with flower morphology, with tubular flowers having more sucrose and open flowers having more hexose. Open flowers contained nectar whose concentration was more affected by relative humidity than tubular flowers. Available nectar in unbagged flowers was found in volumes of 0.1 to 5 μl (17.5 to 68.2% sugar). Total sugar available per blossom amounted to 0.07 to 3.54 mg. We note that care must be taken in converting % concentration to mg sugar. Energetic yield was not as variable as the other measures and ranged from 0.72 to 3.58 cal/μl. Total daily nectar production was measured in five families and 24-hr sugar production varied from 0.64 to 5.52 mg per flower. Insect nectar feeders frequently searched many blossoms with little or no reward, but were rewarded sufficiently at rare “lucky hit” blossoms which contained relatively large nectar rewards. Insect pollinators did not seek nectars of specific sucrose-hexose ratios, but instead took nectar where caloric reward and accessibility made it most profitable.  相似文献

Haba Value of computer‐assisted quantitative nuclear morphometry for differentiation of reactive renal tubular cells from low‐grade urothelial carcinoma Objective: To assess whether computer‐assisted quantitative morphological parameters can be an effective tool for objectively distinguishing reactive renal tubular cells from low‐grade urothelial carcinoma cells (LG‐UCs) in voided urine. Methods: Nuclear morphometry was performed by a computer‐assisted image analyser system on Papanicolaou‐stained cytological specimens. The circumference of reactive renal tubular cells (n = 40) or LG‐UC (n = 20) nuclei were manually traced, and the following nuclear morphometric parameters were analysed: (i) area, (ii) perimeter, (iii) roundness factor, (iv) maximum length, and (v) linear factor. For each nuclear measurement, we calculated the maximum, minimum, mean and standard deviation. Results: The mean nuclear area and nuclear perimeter were higher in reactive renal tubular cells compared to the LG‐UCs. The mean of roundness and linear factors (reflecting a tendency for the nuclear outline to be regular and oval, respectively) were higher in LG‐UCs compared with reactive renal tubular cells. Among nuclear areas, the nuclear perimeter, roundness factors and maximum length did not show any significant differences between reactive renal tubular cells and LG‐UCs. On the other hand, the linear factor showed a mean higher value among LG‐UCs than reactive renal tubular cells (P = 0.023). Conclusions: Of five quantitative nuclear morphological parameters, only linear factor was statistically significant in differentiating reactive renal tubular cells in renal disease from LG‐UCs.  相似文献

利用新一代高通量测序手段——转录组测序(RNA-Seq)技术研究在300 mmol.L-1NaCl胁迫下两个甜瓜(Cucumis melo L.)品种‘玉露’和‘冰雪脆’的转录因子基因表达变化。这两个甜瓜品种在叶绿素荧光参数上的差异表明其在盐胁迫下有不同的生理反应。转录组测序结果表明,盐胁迫与对照相比,‘玉露’共有属于19个转录因子家族的56个转录因子基因表达发生变化(在转录水平,属于7个转录因子家族的22个转录因子上调表达,属于14个转录因子家族的34个转录因子下调表达)。‘冰雪脆’有属于20个转录因子家族的47个转录因子基因表达发生变化(在转录水平上,属于5个转录因子家族的17个转录因子上调表达,属于17个转录因子家族的30个转录因子下调表达)。盐胁迫下,两个甜瓜品种差异表达的转录因子既表现特异性,也存在部分重叠。‘玉露’有29个转录因子特异响应,‘冰雪脆’有20个特异响应。盐胁迫响应重叠的转录因子有27个,其中9个上调表达,18个下调表达。采用实时荧光定量PCR对几个转录因子进行了盐胁迫下的表达检测,其趋势与转录组分析结果基本一致。  相似文献

Renal proximal tubular epithelium can regenerate after various insults. To examine whether the tubular repair process is regulated by surrounding peritubular capillaries, we established an in vitro human tubulogenesis model that mimics in vivo tubular regeneration after injury. In this model, HGF, a potent renotropic factor, dose dependently induced tubular structures in human renal proximal tubular epithelial cells cultured in gels. Consistent with regenerating tubular cells after injury, HGF-induced tubular structures expressed a developmental gene, Pax-2, and a mesenchymal marker, vimentin, and formed a lumen with aquaporin-1 expression. Electron microscopic analysis showed the presence of microvilli on the apical site of the lumen, suggesting that these structures are morphologically equivalent to renal tubules in vivo. When cocultured with human umbilical vein endothelial cells (HUVEC), HGF-induced tubular formation was significantly enhanced. This could not be reproduced by the addition of VEGF, basic FGF, or PDGF. Protein array revealed that HUVEC produced various matrix metalloproteinases (MMPs). The stimulatory effects of coculture with HUVEC or HUVEC-derived conditional medium were almost completely abolished by addition of the tissue inhibitor of metalloproteinase (TIMP)-1 or TIMP-2. These data suggest that endothelial cell-derived factors including MMPs play a critical role in tubulogenesis and imply a potential role of peritubular capillary endothelium as a source of factor(s) required for tubular recovery after injury.  相似文献

Two specimens of Thysanoptera with forked sensilla on third and fourth antennal segments were described from the Lebanese Neocomian and the Spanish Albian ambers, and attributed to the new genus Tethysthrips n. gen. in the family Thripidae Stevens 1829. One specimen with a tubular tenth abdominal segment was also discovered in the Lebanese Neocomian amber, and attributed to the new genus Rohrthrips n. gen. belonging to the family Phlaeothripidae Uzel 1895. Thripidae and Phlaeothripidae are nowadays the most species-rich families of Thysanoptera. The present discoveries of Early Cretaceous fossils show how diversified these families and thrips already were at that time. Moreover, this tubuliferan Rohrthrips specimen has plesiomorphies no longer present in the recent genera, in particular on the wings. Therefore it brings new insight in the evolution of Tubulifera.  相似文献

Summary The mechanism of induction of tubular outgrowths in vitro on floating collagen gels and the influence of extracellular factors on this process have been investigated using the clonal rat mammary epithelial cell line, Rama 25. Growth of Rama 25 on such floating gels causes their contraction. Contraction of the gel is accompanied by a 10-fold increase in the number of cells per unit area, a change in cell shape, and a convolution of the epithelial cell sheet. Gels folded over manually show an 11-times higher incidence of tubules along the folds than on the flat surface. Tubular formation begins when cords of cells develop from local proliferations of the cell sheet and become canalized. Tubules follow wrinkles in the gel and branch to yield monopodial, dichotomous, or lobular architecture. Hydrocortisone and insulin, in the presence of serum, stimulate both narrow and thick tubular structures on folded gels, whereas extra additions of 1 ng/ml cholera toxin or 100 ng/ml epidermal growth factor preferentially stimulate thick tubular structures. Floating glutaraldehyde-fixed gels, very thick collagen gels, and collagen gels prepared on the top of rigid steel grids fail to support the formation of tubules, suggesting that flexibility and access of the medium to basal surfaces are important to their genesis. Incorporation of hyaluronic acid into the gel matrix preferentially inhibits the thick tubular outgrowths. Thus, the branching tubular structures generated by Rama 25 can be influenced in different ways by various extracellular factors in the medium and in the gel. During the course of this work E. J.
