Genes for (methyl)phenol degradation in Pseudomonas putida strain H (phl genes) are located on the plasmid pPGH1. Adjacent to the phl catabolic operon we identified a cryptic transposon, Tn5501, of the Tn3 family (class II transposons). The genes encoding the resolvase and the transposase are transcribed in the same direction,
as is common for the Tn501 subfamily. The enzymes encoded by Tn5501, however, show only the overall homology characteristic for resolvases/integrases and transposases of Tn3-type transposons. Therefore it is likely that Tn5501 is not a member of one of the previously defined subfamilies. Inactivation of the conditional lethal sacB gene was used to detect transposition of Tn5501. While screening for transposition events we found another transposon integrated into sacB in one of the sucrose-resistant survivors. This element, Tn5502, is a composite transposon consisting of Tn5501 and an additional DNA fragment. It is flanked by inverted repeats identical to those of Tn5501 and the additional fragment is separated from the Tn5501 portion by an internal repeat (identical to the left terminal repeat). Transposition of phenol degradation genes could not
be detected. Analysis of sequence data revealed that the phl genes are not located on a Tn5501-like transposon.

The effect on nisin production of increasing nisin immunity/resistance genes in Lactococcus lactis subsp. lactis MG1363 was investigated. The 60-kb nisin immunity/resistance plasmid pND300, which was isolated from a non-nisin-producing
strain, encodes five genes involved in nisin immunity/resistance, which are very similar to those of the immunity/resistance
system encoded by the nisin-production transposon. The introduction of pND300 into MG1363(TnNip) resulted in the construct being able to produce significantly more nisin than the parent MG1363(TnNip). The introduction of pND314, which contains the nisin immunity/resistance genes subcloned into pSA3, into MG1363(TnNip) allowed the strain to grow more rapidly than the parent MG1363(TnNip) with a concomitant increase in the rate of nisin production. This work illustrates that introduction of pND300 and a derivative
containing the nisin immunity/resistance system of pND300 into MG1363 (TnNip) can result in significant alterations to the kinetics of nisin production. These observations indicate approaches that may
be used successfully to improve the economics of nisin production.

The structure and function of transposable elements that code for catabolic pathways involved in the biodegradation of organic compounds are reviewed. Seven of these catabolic transposons have structural features that place them in the Class I (composite) or Class II (Tn3-family) bacterial elements. One is a conjugative transposon. Another three have been found to have properties of transposable elements but have not been characterized sufficiently to assign to a known class. Structural features of the toluene (Tn4651/Tn4653) and naphthalene (Tn4655) elements that illustrate the enormous potential for acquisition, deletion and rearrangement of DNA within catabolic transposons are discussed. The recently characterized chlorobenzoate (Tn5271) and chlorobenzene (Tn5280) catabolic transposons encode different aromatic ring dioxygenases, however they both illustrate the constraints that must be overcome when recipients of catabolic transposons assemble and regulate complete metabolic pathways for environmental pollutants. The structures of the chlorobenzoate catabolic transposon Tn5271 and the related haloacetate dehalogenase catabolic element of plasmid pUO1 are compared and a hypothesis for their formation is discussed. The structures and activities of catabolic transposons of unknown class coding for the catabolism of halogenated alkanoic acids (DEH) and chlorobiphenyl (Tn4371) are also reviewed. 相似文献

From a plasmid carrying the tnpA gene and one inverted repeat sequence (IR) of transposon Tn3, plasmids containing a structure characteristic of transposons, i.e., two IRs flanking a tnpA gene, were generated spontaneously in vivo. They appear to have arisen either through the formation of a “staggered” head-to-head
dimer or by so-called one-ended transposition. These putative transposons could indeed transpose to, or form cointegrates
with, a recipient plasmid. Based on these findings it is proposed that a primeval transposase gene and its target site evolved
first, and subsequently gave rise to a “fully-fledged” transposon by head-to-head dimerization or one-ended transposition.

Tn4371 is a 55 kb transposon which encodes enzymes for the degradation of biphenyl and 4-chlorobiphenyl compounds into benzoate
and 4-chlorobenzo-ate derivatives. We constructed a cosmid library of Tn4371 DNA. The bph genes involved in biphenyl/4-chlorobiphenyl degradation were found to be clustered in the middle of the transposon. Sequencing
revealed an organisation of the bph genes similar to that previously found in Pseudomonas sp. KKS102, i.e. the bphEGF genes are located upstream of bphA1A2A3 and bphA4 is separated from bphA1A2A3 by bphBCD. Consensus sequences for σ54-associated RNA polymerase were found upstream of bphA1 and bphEGF. Plasmid RP4::Tn4371 was transferred into a mutant of Alcaligenes eutrophus H16 lacking σ54. In contrast to wild-type H16 exconjugants, the σ54 mutant exconjugants could not grow on biphenyl, indicating
the dependence of Tn4371bph gene expression on σ54. The Tn4371-encoded bph pathway was activated when biphenyl and various biphenyl-like compounds were present in the growth medium. Preliminary observations
indicate the presence of a region outside the catabolic genes downstream of bphA4 which is involved in mediating at least the basal expression of BphC.

When grown on vegetable oils and their derivatives, the smut fungus Ustilago maydis (DSM 4500 and ATCC 14826) produces several glycolipids under nitrogen-limiting conditions. With 45 g l−1 sunflower oil fatty acids (technical grade) a yield of 30 g l−1 glycolipid was achieved. The resulting mixture contained predominantly mannosylerythritol lipids together with smaller amounts
of cellobiose lipids. The production of the more polar cellobiose lipids was enhanced when glucose was used as carbon source.
The molecular structure of the main components of the glycolipid mixture were elucidated by a combination of NMR spectroscopic
and mass-spectrometric techniques.

Manipulation of the DNA coding for the desulphurizing activity in a new isolate of Arthrobacter sp. 总被引:3,自引:0,他引:3
A new bacterial strain able to cleave CS bonds from organosulphur heterocyclic compounds through the 4-S pathway and tentatively
classified as Arthrobacter sp. was recently isolated. In the present short article we describe the cloning and the characterization of the DNA encoding
the enzymes responsible for desulphurization in this microorganism, referred to as Arthrobacter sp. DS7. The desulphurization operon was found to be located in a large plasmid that also bears the genes conferring cadmium
and arsenic resistance. By shortening this plasmid, a new cloning vector was prepared and used to obtain a recombinant derivative
strain that desulphurizes dibenzothiophene despite of the presence of inorganic sulphur in the growth medium.

The effects of the substrate conditions on the volumetric productivity of Lactobacillus helveticus at different cell densities up to 60 g l−1 in a continuous stirred-tank reactor with microfiltration to retain the biomass were investigated. At low dilution rates,

p, gradually increased to a maximum at D = 1.2–1.5 h−1, because of reduced product inhibition. At higher D values, r
p unexpectedly decreased, although the substrate conditions further improved. The maxima of r
p at different cell densities coincided with a critical specific substrate utilization rate beyond which the cell metabolism
seems to be controlled through a catabolic modulator factor, and r
p decreases.

The gene dak1 encoding a dihydroxyacetone kinase (DHAK) isoenzyme I, one of two isoenzymes in the Schizosaccharomyces pombe IFO 0354 strain, was cloned and sequenced. The dak1 gene comprises 1743 bp and encodes a protein of 62 245 Da. The deduced amino acid sequence showed a similarity to a putative
DHAK of Saccharomyces cerevisiae and DHAK of Citrobacter freundii. The dak1 gene was expressed at a high level in Escherichia coli, and the recombinant enzyme was purified to homogeneity and characterized. The acetone powder of recombinant E. coli cells was used to produce dihydroxyacetone phosphate.

The Tn5-based transposon Tn5-KIL3 (Miksch et al. 1997c) bearing the kil gene of the ColE1 plasmid of Escherichia coli, which mediates controlled export of periplasmic proteins into the culture medium, was stably integrated into the chromosome
of Klebsiella planticola with high transposition frequency. A Bacillus hybrid β-glucanase located on an RSF1010-derived plasmid was mobilized from E.coli to K. planticola and used as a reporter protein to select strains with high expression and secretion competence. During fermentation experiments
it was shown that the production of β-glucanase in K. planticola was improved to an unexpectedly high level when the enzyme was secreted into the medium. Due to the stationary-phase promoter
used for the expression of the kil gene the secretion of β-glucanase into the medium started at the transition from the exponential to the stationary phase,
as in E. coli, and the fraction of secreted protein reached 90%. The results showed that K. planticola may represent an interesting organism for the production of heterologous proteins.

The biochemical mechanisms for growth tolerance to a 100% CO headspace in cultures, and butanol plus ethanol production from
CO by Butyribacterium methylotrophicum were assessed in the wild-type and CO-adapted strains. The CO-adapted strain grew on glucose or CO under a 100% CO headspace,
whereas, the growth of the wild-type strain was severely inhibited by 100% CO. The CO-adapted strain, unlike the wild-type,
also produced butyrate, from either pyruvate or CO. The CO-adapted strain was a metabolic mutant having higher levels of ferredoxin–NAD
oxidoreductase activity, which was not inhibited by NADH. Consequently, only the CO-adapted strain can grow on CO because
CO oxidation generates reduced ferredoxin which, via the mutated ferredoxin–NAD reductase activity, forms reduced NADH required
for catabolism. When the CO-adapted strain was grown at pH 6.0 it produced butanol (0.33 g/l) and ethanol (0.5 g/l) from CO
and the cells contained the following NAD-linked enzyme activities (μmol min−1 mg protein−1): butyraldehyde dehydrogenase (227), butanol dehydrogenase (686), acetaldehyde dehydrogenase (82) and ethanol dehydrogenase
(129).

The majority of lignin-degrading basidiomycetes are able to depolymerize humic acids. In this presentation the relationship
and possible similarities between enzymes involved in lignin degradation and humic acid depolymerization were examined on
the genetic level. We have cloned fragments of the gene encoding the extracellular ligninolytic enzyme laccase from Clitocybula dusenii, Nematoloma frowardii and a fungal strain designated i63-2, and compared the three sequences with those of several other published laccase genes.
The sequenced fragments displayed a high homology both on the DNA (97%–77%) and amino acid (100%–85%) level. Furthermore,
the expression of this gene in the above-mentioned fungi was demonstrated by a nested polymerase chain reaction with cDNA
as template.

Shuttle mutagenesis using signature-tagged transposons was employed to generate a library of individually tagged mutants
of the Neisseria meningitidis strain B1940, which belongs to serogroup B. The use of tagged transposons allowed us to monitor for enrichment for single
mutants during the process of shuttle mutagenesis, by amplification of the tags and subsequent sequence determination. Enrichment
of a single clone occurred during the transformation of the meningococci with transposon-containing plasmid DNA. Sequence
determination around the site of transposon insertion revealed that the transposon had mutagenized a previously unknown locus,
which was designated hrtA (high rate of transformation). hrtA-mediated transformation was independent of TnMax5 and tag sequences, and it most probably involved recombination events. The hrtA locus is restricted to meningococci and gonococci and is present in few apathogenic neisserial species. Chromosomal mapping
of hrtA and six further hrt sites revealed a random distribution of highly transforming DNA fragments on the meningococcal chromosome. In conclusion,
our data demonstrate that shuttle mutagenesis of naturally competent bacteria using signature-tagged transposons allows the
isolation of chromosomal DNA fragments, which exhibit a high transformation efficiency, and which, therefore, are likely to
be involved in horizontal gene transfer.

Previous investigations have reported that bacterial suspension cultures grow to higher stationary concentrations in space
flight than on Earth; however, none of these investigations included extensive ground controls under varied inertial conditions.
This study includes extensive controls and cell-growth data taken at several times during lag phase, log phase, and stationary
phase of Escherichia coli and Bacillus subtilis. The Marquardt-Levenberg, least-squares fitting algorithm was used to calculate kinetic growth parameters from the logistic
bacterial growth equations for space-flight and control growth curves. Space-flight cultures grew to higher stationary-phase
concentrations and had shorter lag-phase durations. Also, evidence was found for increased exponential growth rate in space.

A Tn5-based transposon bearing the kil gene (killing protein), mediating controlled export of periplasmic proteins into the culture medium, was constructed (Tn5-KIL3). This transposon contained the kil gene of the ColE1 plasmid under the growth-phase-dependent promoter of the fic gene (filamentation induced by cAMP) of Escherichia coli, an interposon located upstream of kil, a kanamycin/neomycin-resistance gene, a multiple cloning site and the mob site. The transposition of Tn5-KIL3 to Acetobacter methanolicus showed a moderate transposition frequency (10−5–10−6). By insertion of a Bacillus hybrid β-glucanase (bgl ) as a model protein into the transposon (Tn5-LF3) it was shown that the secretion function as well as the gene of the target protein had been transferred to and stably
integrated into the chromosome of A. methanolicus, and that the transposition of Tn5-LF3 was non-specific. β-Glucanase was highly overexpressed and secreted into the medium during stationary phase. Total and
extracellular production of β-glucanase varied depending on the integration site of the transposon. The viability of the bacterial
cells was not affected, and cell lysis did not occur.

The kinetics of nisin production was studied in batch cultures using a construct of Lactococcuslactis subsp. lactis C2SmPrt−, containing a transposon (TnNip) that encodes nisin production. The introduction of TnNip into C2SmPrt−significantly lowered the specific growth rate and the maximum A
620 reached was reduced from 15.2 to 11.0. The effect of nisin concentration and nutrient depletion on nisin production of the
construct, C2SmPrt−(TnNip), was examined. Nisin production was found to be inhibited by high concentrations of nisin, when grown in excess nutrient,
even though growth of the culture continued because nutrient limitation was not operating. However, in low nutrient concentrations
nisin production was limited by nutrient depletion. The specific growth rate of C2SmPrt−(TnNip) was altered, by using different nutrient concentrations and different sugars, in order to examine the relationship between
nisin production and growth. Nisin production was shown to be growth-associated for most of growth, but near the end of growth,
when the specific growth rate was 0.05 h−1 or less, the production ceased.

A 5.7-kbp region of the Clostridium thermocellum F1 DNA was sequenced and found to contain two contiguous and highly homologous xylanase genes, xynA and xynB. The xynA gene encoding the xylanase XynA consists of 2049 bp and encodes a protein of 683 amino acids with a molecular mass of 74 511 Da,
and the xynB gene encoding the xylanase XynB consists of 1371 bp and encodes a protein of 457 amino acids with a molecular mass of 49 883 Da.
XynA is a modular enzyme composed of a typical N-terminal signal peptide and four domains in the following order: a family-11
xylanase domain, a family-VI cellulose-binding domain, a dockerin domain, and a NodB domain. XynB exhibited extremely high
overall sequence homology with XynA (identity 96.9%), while lacking the NodB domain present in the latter. These facts suggested
that the xynA and xynB genes originated from a common ancestral gene through gene duplication. XynA was purified from a recombinant Escherichia coli strain and characterized. The purified enzyme was highly active toward xylan; the specific activity on oat-spelt xylan was
689 units/mg protein. Immunological and zymogram analyses suggested that XynA and XynB are components of the C. thermocellum F1 cellulosome.

A xylanase gene, xynF1, was cloned and characterized from a shoyu koji mould Aspergillus oryzae KBN616. The xynF1 gene was found to be comprised of 1484 bp with ten introns. The deduced amino acid sequence encodes a protein consisting
of 327 amino acids (35,402 Da) which is very similar to the fungal family F xylanases such as Aspergillus nidulans XlnC, Aspergillus kawachii XynA and Penicillium chrysogenum XylP. The intron/exon organization of xynF1 is very similar to that of the fungal family F xylanase genes. Plasmid pXPR64, which contains 64 copies of the xynF1 promoter region (PxynF1) in the same direction, was constructed and introduced into A. oryzae. This led to reduced expression of both xylanase and β-xylosidase genes in the transformants.

Twenty strains of Streptococcus bovis grew more slowly on lactose (1.21 ± 0.12 h−1) than on glucose (1.67 ± 0.12 h−1), and repeated transfers or prolonged growth in continuous culture (more than 200 generations each) did not enhance the growth
rate on lactose. Lactose transport activity was poorly correlated with growth rate, and slow growth could not be explained
by the ATP production rate (catabolic rate). Batch cultures growing on lactose always had less␣intracellular fructose 1,6-bisphosphate
(Fru1,6P
2) than cells growing on glucose (6.6 mM compared to 16.7 mM), and this difference could be explained by the pathway of carbon
metabolism. Glucose and the glucose moiety of lactose were metabolized by the Embden-Meyerhoff-Parnas (EMP) pathway, but the
galactose moiety of lactose was catabolized by the tagatose pathway, a scheme that by-passed Fru1,6P
2. A mutant capable of co-metabolizing lactose and glucose grew more rapidly when glucose was added, even though the total
rate of hexose fermentation did not change. Wild-type S. bovis grew rapidly with galactose and melibiose, but these galactose-containing sugars were activated by galactokinase and catabolized
via EMP. On the basis of these results, rapid glycolytic flux through the EMP pathway is needed for the rapid growth (more
than 1.2 h−1) of S.␣bovis.
