1H, 15N, and 13C NMR assignments for 116 amino acids (Gly893-Lys1008) of a bacterial collagen-binding domain (CBD) derived from Clostridium histolyticum class I collagenase were accomplished. Clostridial collagenases hydrolyze insoluble collagen. One to three copies of collagen-binding domains (CBDs) are present at their C-termini, each of which is the minimal segment required for the binding to the insoluble substrate. CBD has been shown to be able to anchor fused growth factors for up to 10 days in vivo. Structural analysis of the small domain with the unique function provides insights into designing a novel drug delivery vehicle by the rational drug design.  相似文献

Norovirus protease cleaves the virus-encoded polyprotein into six mature nonstructural proteins, presenting itself as an essential enzyme for the viral replication as well as an attractive target for the antiviral drug development. A deeper understanding of the structural mechanism of the protease-substrates/inhibitors interactions by means of solution NMR methods would facilitate a rational design of the virus protease inhibitor. We here report the backbone and side-chain resonance assignment of the protease from Norwalk virus, which is the prototype strain of norovirus. Angiogenin is an unusual member of the pancreatic ribonuclease superfamily that induces formation of new blood vessels and is a promising anti-cancer target. Here we report backbone and side chain 1H, 13C, and 15N resonance assignments for rat angiogenin (residues 24–145), excluding the N-terminal signal peptide. These data allow nuclear magnetic resonance structure and inhibitor-binding studies with the aim of providing angiogenin antagonists as potential therapeutics.  相似文献

S100 proteins belong to the EF-hand family of calcium binding proteins. Upon calcium binding, these proteins undergo a conformational change to expose a hydrophobic region necessary for target protein interaction. One member of the S100 protein family is S100A11, first isolated from chicken gizzard and termed calgizzarin. It was later isolated from other organisms and tissues including human placenta, pig heart and rabbit lung. The physiological target of S100A11 is thought to be annexin I, a phospholipid-binding protein involved in EGF receptor sorting. This work reports the 1H, 15N and 13C resonance assignments of rabbit apo-S100A11 determined using 15N, 13C-labelled protein and multidimensional NMR spectroscopy.  相似文献
