Agricultural research of plant-derived endophytic fungi has grown in recent decades. We isolated 26 endophytic fungi from the leaves, stems and fruits of "the tree of life", Camptotheca acuminata, and tested them for antimicrobial activities based on growth inhibition measurements in a modified agar diffusion method. Fermentation broths from most of the isolates exhibited antifungal activity and 50% exhibited antibacterial activity; some of them also exhibited strong broad-spectrum antimicrobial activity. The strongest antimicrobial activity was exhibited by strains XSY10 and XSY15 against Rhizoctonia solani and Fusarium oxysporum f. sp. vasinfectum, with 75% and 67% inhibition, respectively. Strain XSJ01 gave strong activity against pathogenic bacteria, with inhibition zones more than 20 mm in diameter. The isolates were identified by molecular methods as belonging to nine taxa: Nigrospora, Diaporthe, Alternaria, Colletotrichum, Pestalotiopsis, Sordariomycete, Guignardiai, Penicillium, and Zythia. Based on these results, we conclude that the endophytic fungi of C. acuminata are promising sources of novel bioactive compounds.  相似文献

Abstract The antagonistic effect of Trichoderma reesei and Trichoderma harzianum was studied towards a range of phytopathogenic fungi; Alternia solani, Botrytis fabae, Cladosporium cucumerinum, Fusarium oxysporum, Fusarium tricintum . Coculture of the phytopathogens and Trichodema under laboratory conditions clearly showed dominance of the Trichoderma species. In all cases, Trichoderma overgrew the phytopathogens and subsequently developed a conidial lawn over the surface. In these studies the lytic action of the pathogen was clearly apparent and the inhibition of growth appears directly related to its ability to hydrolyze the cell walls of the tested microorganisms rather than through the inhibitory action of antibiotics or toxins. Proteinase, mannanase, laminarinase and chitinase activities were determined in the extracellular fluid of Trichoderma . Glucose and laminaribiose were detected after the in vitro hydrolysis of the cell walls of the phytopathogens. The data imply that the nature of T. reesei and T. harzianum antagonism is based on mycoparasitism (lysis) and appears to optimalize with contact between the mycelia.  相似文献

The aim of the present study was to evaluate the antifungal activity of fatty acids against phytopathogenic fungi. Two pot experiments were conducted by mixing palmitic and oleic acids in the soil in which poor plant growth was observed. In addition, the antifungal activities of nine fatty acids (butyric acid, caproic acid, caprylic acid, capric acid, lauric acid, myristic acid, palmitic acid, oleic acid, and linoleic acid) against four phytopathogenic fungi: Alternaria solani, Colletotrichum lagenarium, Fusarium oxysporum f. sp. Cucumerinum, and Fusarium oxysporum f. sp. lycopersici, were assessed by measuring mycelial growth and spore germination via Petri dish assay. The results of the pot experiments showed that the mixture of palmitic and oleic acids enhanced the growth of the seedlings of continuous-tomato and continuous-cucumber. Except for oleic acid, in the Petri dish assay, the fatty acids tested were observed to inhibit the mycelial growth of one or more tested fungi. In addition to the suppression of mycelial growth, butyric acid, caproic acid, caprylic acid, capric acid, lauric acid, and palmitic acid showed an inhibitory effect against spore germination and the extent of inhibition varied with both the type of fatty acids, and the fungi. In particular, capric acid displayed strong inhibitory effect against C. lagenarium on the mycelial growth and spore germination. The saturated fatty acids, i.e. palmitic acids, showed stronger antifungal activity than the unsaturated fatty acids, i.e. oleic acid. It suggests that fatty acids might be applicable to exploring for alternative approaches to integrated control of phytopathogens.  相似文献

从云南会泽铅锌矿废弃矿渣堆上的常见植物硬毛南芥(Arabis hirsuta)、毛萼香茶菜(Rabbosia eriocalyx)和倒挂刺(Rosa longicuspis)等6种植物的690个组织块中共分离得到内生真菌495株,内生真菌的分离频率在0.42—0.93之间,平均为0.72,所有植物茎内生真菌的分离频率都明显高于叶(P<0.05)。经形态学鉴定,内生真菌分属于茎点霉属(Phoma)、交链孢属(Alternaria)和派伦霉属(Peyronellaea)等20个分类单元,其中茎点霉属和派伦霉属为该废弃矿渣堆上常见植物的优势内生真菌属。6种植物内生真菌的多样性指数在1.05—2.29之间,与其它非重金属污染环境植物内生真菌的多样性指数相似,说明在重金属污染地区仍然存在多种重金属耐受的内生真菌种类。6种植物内生真菌的相似性系数(0.455—0.833)表明,会泽铅锌矿区植物内生真菌的宿主专一性较小。  相似文献

通过体外培养法,研究了药用植物温郁金内生真菌Chaetomium globosum L18对几种常见的植物病原菌的抑菌谱及其拮抗机理。结果表明,Chaetomium globosum L18对多种植物病原真菌和细菌均有不同程度的抑制作用,具有较广的抑菌谱,但对不同植物病原菌的抑制作用具有显著性差异(P<0.05),抑制率最高可达到92.9%;抑菌机制结果显示,竞争作用和重寄生作用是其主要的拮抗机制之一;发酵产物抑制作用测定发现,内生真菌Chaetomium globosum L18能够分泌产生抗菌物质抑制病原菌菌丝的生长和孢子萌发,可引起病原菌菌丝菌丝膨大成串珠状,分枝增多,分枝顶端膨胀后细胞壁破裂,原生质外溢,产生溶菌作用;使分生孢子萌发畸形,萌发率降低。  相似文献

Four polyketides, leptosphaerone C (1), penicillenone (2), arugosin I (3) and 9-demethyl FR-901235 (4), as well as five known compounds, bacillosporin A (5), bacillosporin C (6), sequoiamonascin D (7), sequoiatone A (8), and sequoiatone B (9) were isolated from the Penicillium sp. JP-1, an endophytic fungus isolated from Aegiceras corniculatum. Their structures were determined by spectroscopic methods, mainly by 2D NMR spectroscopic analyses. Compound 1 showed cytotoxicity against A-549 cells with an IC50 value of 1.45 microM, while compound 2 showed cytotoxicity against P388 cells with an IC50 value of 1.38 microM.  相似文献

从白花泡桐根、茎、叶中分离出19株内生真菌,对分离出的真菌分别利用琼脂块法和纸片扩散法进行初选和复选,通过形态学特征观察和rRNA基因ITS序列系统发育分析对抗菌活性菌株进行鉴定。抑菌试验结果显示JSD-8、JM-1和JM-10等3株内生真菌对枯草芽胞杆菌(Bacillus subtilis)CGMCC1.769、大肠杆菌(Escherichia coli)CGMCC1.1103和白色念珠菌(Candida albi-cans)ATCC10123均有一定的抗菌活性。鉴定结果表明,JSD-8、JM-1、JM-10分别属于串珠赤霉菌(Gibberella moniliformis)、长柄链格孢菌(Alternaria longipes)和托姆青霉菌(Penicilium thomii)。  相似文献

茅苍术内生真菌的分离鉴定及在组培苗中的回接   总被引:2,自引:0,他引:2  
从茅苍术的叶、茎和根中分离鉴定内生真菌,并观测内生菌回接对茅苍术组培苗生长的影响。共获得茅苍术内生菌16株,其中叶片中14株,根茎中各获得一株。分别为交链孢霉、镰刀霉、小克银汉霉、青霉、茎点霉、小菌核菌、孔球孢霉和不产孢类。经内生菌和茅苍术快繁苗实验初筛,得到两株对植物无害的内生菌:小菌核菌和孔球孢霉,将它们回接到茅苍术组培苗中,可以从苗叶片中染色观察到菌丝。其中小菌核菌能提高茅苍术炼苗的存活率,接入内生菌的茅苍术叶片超氧化物歧化酶和过氧化物酶活性均高于未接菌的对照,脂肪酸不饱和指数基本不变,说明分离自茅苍术的内生菌回接后可以与宿主建立共生关系。  相似文献

为充分开发黄花倒水莲(Polygala fallax)的内生真菌资源,获得具有抗植物病原真菌、抗氧化活性的内生真菌,该文以黄花倒水莲内生真菌为研究对象,使用平板对峙法检测内生真菌对6种植物病原真菌的抑菌活性,测定内生真菌发酵液的DPPH清除自由基能力和总还原能力,评价内生真菌的抗氧化活性,并对具有强抑菌活性和抗氧化活性的菌株进行形态和ITS鉴定。结果表明:(1)黄花倒水莲内生真菌中有2株内生真菌对香蕉专化尖孢镰刀菌、柑橘树脂病菌、叶点霉菌、香蕉具条叶斑病菌、茄病镰刀菌、三七根腐病菌具有明显的抑菌活性,抑菌率在50.3%~91.4%之间,其中HNLF-5对柑橘树脂病菌的抑菌率为73.2%,HNLF-44对香蕉专化尖孢镰刀菌抑菌率为91.4%。(2)内生真菌发酵液具有良好的抗氧化活性,DPPH清除率均在80%以上,总还原能力吸光值范围为0.279 2~0.748 8。(3)HNLF-44菌株为链格孢属真菌。该研究表明,药用植物黄花倒水莲内生真菌具有较好的生物活性,为后续从黄花倒水莲内生真菌中挖掘潜在新型抑菌活性和抗氧化活性物质奠定了基础。  相似文献

对喜树植株中的内生真菌进行分离纯化,共得到126株内生菌株。对126株内生菌株进行液体培养,并对其发酵产物进行抗菌活性测定。发现在供试质量浓度为5mg/mL时,126株菌株中有24株喜树内生真菌的发酵液对水稻纹枯病菌、稻瘟病菌以及辣椒疫霉具有不同程度的抑制菌丝生长的活性作用;其中,菌株XSY09的发酵液对此三种植物病原菌有明显的抑制作用,抑制率分别为74.97%、39.63%和58.49%。在0.1mg/mL的测试浓度下,菌株XSY09发酵液的乙酸乙酯相对三种植物病原菌均有较好的抑制作用。对该菌株的ITS序列进行测序分析,初步鉴定XSY09为炭疽菌属(Colletotrichum gloeosporioides)真菌。  相似文献

Recent research has shown the bioprospecting of endophytic fungi from Cupressaceae. Here, we further uncover that the healthy cypress plants such as Cupressus arizonica, Cupressus sempervirens var. cereiformis, and Thuja orientalis host highly bioactive endophytic Alternaria fungal species. Indeed, endophytic Alternaria alternata, Alternaria pellucida, and Alternaria tangelonis were recovered from healthy Cupressaceous trees. Biodiversity and bioactivity of recovered endophytic Alternaria species were a matter of biogeography and host identity. We further extracted such Alternaria’s metabolites and highlighted their significant antiproliferative, growth inhibitory, and antibacterial activities against the model target fungus Pyricularia oryzae and the model pathogenic bacteria Bacillus sp., Erwinia amylovora, and Pseudomonas syringae. In vitro assays also indicated that endophytic Alternaria species significantly inhibited the growth of cypress fungal phytopathogens Diplodia seriata, Phaeobotryon cupressi, and Spencermartinsia viticola. In conclusion, since the recovered Alternaria species were originally reported as pathogenic and allergenic fungi, our findings suggest a possible ecological niche for them inside the foliar tissues of Cupressaceous trees. Moreover, in this study, the significant bioactivities of endophytic Alternaria species in association with Cupressaceae plant family are reported.  相似文献

喜树内生真菌抗水稻纹枯病菌活性的研究   总被引:2,自引:0,他引:2  
从喜树健康组织叶、茎以及果实中经过分离、纯化共得到26株内生菌株。利用生长速率法测定这26株菌株对植物病原菌——水稻纹枯病病原菌的抑制作用。抑菌试验结果显示:26株菌株中有22株喜树内生真菌的发酵液对植物病原菌菌丝生长均有不同程度的抑制作用;其中,菌株XSY10的发酵液抑菌效果最强,达到了74.97%。水稻盆栽试验结果表明:喷施菌株XSY10次生代谢产物7d后,对水稻纹枯病的防治效果为34.77%。以上试验结果说明喜树内生真菌XSY10对植物病害的防治具有一定的功效。  相似文献

Chemical investigation of the endophytic fungus Penicillium sp. isolated from Limonium tubiflorum growing in Egypt afforded four new compounds of polyketide origin, including two macrolides, penilactone (1) and 10,11-epoxycurvularin (2), a dianthrone, neobulgarone G (7), and a sulfinylcoumarin, sulfimarin (14), along with twelve known metabolites (3-6, 8-13, 15 and 16). The structures of all compounds were assigned by comprehensive spectral analysis (1D and 2D NMR) and mass spectrometry. Compounds 3, 4, 13 and 16 showed pronounced antitrypanosomal activity with mean MIC values ranging from 4.96 to 9.75 ??M. Moreover, when tested against a panel of three human tumor cell lines compounds 3, 4, 6 and 12 showed selective growth inhibition against Jurkat and U937 cell lines with IC50 values ranging from 1.8 to 13.3 ??M. The latter compounds also inhibited TNF??-induced NF-??B activity in K562 cells with IC50 values ranging from 1.6 to 10.1 ??M, respectively.  相似文献

Three metabolites named phomopsin A (1), B (2) and C (3), together with two known compounds cytosporone B (4) and C (5), were isolated from the mangrove endophytic fungus, Phomopsis sp. ZSU-H76 obtained from the South China Sea. Their structures were elucidated by spectroscopic methods, mainly by 1D and 2D NMR spectroscopic techniques. The medium-sized cyclic phenol ether based on 1 or 2 is rare in natural products. In bioassays, compounds 1, 2, and 3 had no significant antibiotic activities, but compounds 4 and 5 inhibited two fungi Candida albicans and Fusarium oxysporum with an MIC ranging from 32 to 64 microg/ml.  相似文献

生活在宿主植物里的内生真菌是很重要的药用资源。喜树是中国的传统药用植物。从喜树植物中分离得到了大约50种菌株,其中一株产喜树碱的菌株通过形态学鉴定为青霉属,这是首次在喜树植物中发现产喜树碱的青霉属菌株。为研究简单序列重复相关序列扩增多态性(SRAP)方法在喜树内生真菌中应用的可行性,选择了十株喜树内生真菌进行SRAP多态性分析。SRAP引物共扩增出1 295条带,而这些菌株也被分为三大类。这些结果表明,SRAP研究喜树内生真菌具有高效性,是讨论喜树内生真菌的遗传多样性的有效方法。  相似文献

【目的】探讨回接内生真菌对葡萄叶内生真菌群落结构的影响。【方法】将葡萄叶中分离到的8株内生真菌回接到健康生长的玫瑰蜜葡萄叶片上,同时设置管理上喷施农药与未喷施农药两组处理,3个月后分析不同真菌菌剂处理后葡萄叶片内生真菌的分离率、优势度和多样性指数。【结果】5种菌剂的处理(Xylaria sp.、Nigrospora sp.、Alternaria sp.1、Alternaria sp.2和Colletotrichum sp.)在处理后的叶片中分离到了较多数量的接种菌。不同菌剂处理后葡萄叶片菌群结构差异显著。【结论】与对照处理的葡萄相比,菌剂的处理增加了叶内生真菌的分离率,但显著降低了内生真菌的多样性指数;发现喷施农药组叶片内生真菌的分离率较低,但多样性相对较高。  相似文献

Glucoside derivatives, xylarosides A (1) and B (2), were isolated from the broth extract of the endophytic fungus Xylaria sp. PSU-D14 along with two known compounds, sordaricin (3) and 2,3-dihydro-5-hydroxy-2-methyl-4H-1-benzopyran-4-one (4). The structures were assigned by spectroscopic methods. Sordaricin (3), one of the known metabolites, exhibited moderate antifungal activity against Candida albicans ATCC90028 with a MIC value of 32 microg/ml.  相似文献

采用组织分离法从古银杏健康组织中分离得到55株内生真菌, 其中28个分离菌株在PDA培养基上不产孢子, 占总分离菌株的50.9%, 其它菌株根据其在PDA培养基上的培养特征, 10株被鉴定为青霉、6株为曲霉、4株为交链孢霉、3株为简梗孢霉, 另外酵母、毛霉、小单头孢霉、镰孢霉各1株。考察内生真菌培养上清对7种受试指示菌的抑制作用, 共筛选得到23株至少对一种指示菌的生长有抑制作用的菌株, 其中11株为不产孢真菌, 占活性菌株的47.83%。对活性最强的一株菌进行形态学和分子生物学鉴定, 将其确定为Xyla  相似文献

从黄海海岸低盐药用植物芦竹(Arundo donax L.)中分离得到木霉属的内生真菌F0238,对其进行拮抗植物病原菌活性及作用机制的试验研究。结果表明F0238发酵液对Botrytis cinerea,Sclerotium rolfsii等8种植物病原菌有强的抑菌活性及抑制孢子萌发作用,且这种作用在发酵原液被稀释了160倍后仍然存在;对峙培养试验表明F0238菌对植物病原菌有很强的拮抗作用,拮抗机制主要为重寄生作用、营养竞争作用及抗生作用等。  相似文献

Microorganisms, especially endophytic fungi that reside in the tissue of living mangrove plants, seem to play a major role in meeting the general demand for new biologically active substances. During the course of screening for biologically active secondary metabolites from marine microorganisms, an antibiotic compound containing an indole and a diketopiperazine moiety was isolated from the culture medium of Penicilliumchrysogenum, (MTCC 5108), an endophytic fungus on the mangrove plant Porteresiacoarctata (Roxb.). The cell free culture medium of P. chrysogenum showed significant activity against Vibriocholerae, (MCM B-322), a pathogen causing cholera in humans. Bioassay guided chemical characterization of the crude extract led to the isolation of a secondary metabolite possessing a molecular formula C19H21O2N3. Its antibacterial activity was comparable with standard antibiotic, streptomycin. This compound (1) was found to be (3,1′-didehydro-3[2″(3′″,3′″-dimethyl-prop-2-enyl)-3″-indolylmethylene]-6-methyl pipera-zine-2,5-dione) on the basis of mass spectrometry, infrared spectroscopy and one and two-dimensional nuclear magnetic resonance analysis.  相似文献
