The Role of Roots in Control of Bean Shoot Growth   总被引:10,自引:0,他引:10

Restriction of root growth by growing bean plants (Phaseolusvulgaris L.) in very small pots led to the development of dwarfplants. The leaves of those plants were smaller and their internodesshorter than those of control plants which were grown in largerpots and had developed a more extensive root system. A largequantity of starch—much more than in control plants —accumulated in the leaves and shoots of the dwarf plants. Increasingthe amount of minerals which was supplied to the roots, enhancedleaf growth of the control plants but failed to affect the dwarfones, in spite of the fact that in both cases the treatmentincreased the content of N, P and K in all the plant organs.The leaf water content was similar in both treatments, but theleaf water potential was higher in the dwarf plants. Exogenousapplication of gibberellic acid (GA3) to the dwarf plants overcamethe reduction of stem growth completely, and that of the leavespartially. Application of the cytokinin, benzyladenine (BA)did not affect stem growth, but increased that of the primaryleaves. A combined supply of GA2 + BA restored completely thegrowth of the stem and the primary leaves, and partially thatof the trifoliate leaves. It is concluded that a limited rootsystem restricts shoot growth through an hormonal system inwhich at least gibberellins and cytokinins are involved, andthat the dwarfing is not a consequence of mineral or assimilatedeficiency, or due to water stress. Effects of plant hormones were examined on the dark- and light-inducedmovements of Cassia fasciculata. Indole-3-acetic acid (IAA),gibberellic acid (GA3) and 6-benzylaminopurine (6-BAP) inhibitedthe scotonastic movement whereas abscisic acid (ABA) enhancedit. After brief treatments (5 to 30 min), the ABA effect wasinhibitory rather than promotional. Hormonal treatment in theacidic range gave the best physiological response for ABA, butthe greatest efficiency of IAA, GA3 and 6-BAP was obtained withpH values close to neutrality. Three to 5 h were needed beforeexpression of the physiological effect triggered by GA3 and6-BAP, while 5 min treatments were sufficient for IAA and ABA.Light-induced movements were largely enhanced by IAA and slightlyby GA3 but inhibited by 6-BAP and ABA. The results are discussedin relation to the ionic changes in the pulvinar motor cells,regulating leaflet movements. 1. A study was made of the influence of gibberellic acid (GA2)on nucleic acid, protein, and cell-wall synthesis in pea internodesin vivo. 2. GA3-treated fifth internodes finally contained more thantwice as much total RNA and protein as comparable untreatedones, and the contents of RNA and protein were closely relatedto the length of internode cortical cells. 3. Cell elongation, RNA, protein, and cell-wall synthesis werestimulated 24–48 h before there was any demonstrable GA3effect on DNA synthesis and cell division. 4. Treated fifth internodes finally contained twice as manycortical cells as control internodes, a response that was matchedby a proportionate increase in the amount of DNA. 5. Internodes treated with actinomycin D or cycloheximide failedto elongate in response to GA3 treatment, indicating that bothRNA and protein synthesis are essential for gibberellin-stimulatedcell elongation to occur in this tissue. 6. 5-fluorodeoxyuridine at concentrations which completely blockcell division did not prevent cells from elongating in the presenceof GA3. 7. With the possible exception of pectic substances there wasno change in the relative proportions of each of the major cell-wallconstituents in treated, as compared to control internodes.  相似文献

Seeds ofTaxus maireiare known for their deep dormancy whichcan only be broken by a procedure involving warm stratificationfollowed by cold stratification. Treatments with alternatingtemperatures of 25/15 or 23/11 °C (12 h light) for 6 monthsfollowed by 5 °C for 3 months were successful in overcomingseed dormancy. After 6 months of warm stratification, cytologicalchanges observed included: enlargement of the embryo; a decreasein the number of lipid bodies; appearance of ER; and increasesin mitochondria, plastids, dictyosomes, vacuoles and microbodiesin the shoot apical meristem. Cold stratification followingthe warm treatment induced cell division, and one or two distinctnucleoli in the shoot apical meristem cells were observed. Bothwarm and cold stratification reduced endogenous ABA concentrationsfrom the original 8888 pg per freshly harvested seed to 392and 536 pg, respectively. Treatment with exogenous gibberellinsafter seeds had been warm-stratified showed that GA4and GA7wereeffective at promoting seed germination, but GA3was not. Theseresults suggest that the strong seed dormancy ofT. maireicouldbe caused by a high ABA content and underdevelopment of theembryos in freshly shed seeds. We conclude that warm stratificationwith alternating temperatures increases the growth of embryosby cell expansion and enlargement and decreases ABA content,but seeds still remain ungerminated. The sprouting of partially stratified yam bulbils was promotedby exogenous benzyladenine, cycocel (CCC) and indole-3-aceticacid and suppressed by gibberellin A3(GA3. Light exposure causedslight suppression of sprouting. GA3 raised the content of batatasinsbut not that of abscisic acid. We suggest that the sprouting-inhibitingactions of GA3 and light are exerted by raising the batatasinlevel in the bulbils. A critical stage in flower bud growth in the spike of Gladioluswhich is initiated by gibberellic acid (GA3) and sustained bysucrose has been identified. This corresponds to the stage atwhich separation of the outer bract occurs. In buds at differentdevelopmental stages isolated and held in water, it is the samebud stage that first shows increased growth. Buds not inducedby light were shown to respond more significantly to GA3 andsucrose than those induced by light. Maize (Zea mays L., hybrid Cargill 147) seedlings, grown inaseptic conditions, were inoculated with three strains of Azospirillumlipoferum (Al op 33, Al iaa 320, and ATCC 29708) or culturedin different concentrations of indol-3-acetic acid (IAA) orgibberellin A3 (GA3). After 48 h, root length, root surfacearea, root dry weight, and shoot dry weight were measured inall treatments. Gibberellin content was evaluated in selectedroots of control and Azospirillum inoculated seedlings by gaschromatography-mass spectrometry-selected ion monitoring withthe use of deuterated gibberellins as internal standards. Thethree strains of A. lipoferum, IAA (2 ng ml–1), and GA3(40 to 400 pg ml–1) significantly enhanced root growth.Improvement of root hair growth and density was obtained mainlywith A. lipoferum strain Al op 33 and GA3 40 pg ml–1.GA3 was identified by gas chromatography-mass spectrometry (byboth, full scan and selected ion monitoring) in a free acidfraction from roots of the seedlings inoculated with A. lipoferum.In the roots of the non inoculated seedlings GA3 was found afterhydrolysis of a fraction expected to contain glucosyl conjugates. The effect of abscisic acid (ABA) and of gibberellic acid (GA2)on the metabolism of kinetin in detached leaves was investigated.Kinetin labelled in either the purine ring or the N-6 substitutedside chain was fed into leaves of Rumex pulcher. The leaveswere extracted 4 or 20 h later and the extract chromatographedon paper which was subsequently scanned for radioactivity. Fourmajor radioactive peaks were revealed. Two peaks were tentativelyidentified, one kinetin and its riboside, and the other adenine.ABA effected a significant decrease in the transformation ofkinetin to adenine while GA3 had no consistent effect. The germination performance of maize seeds (Zea mays L. cv.Partap-1) pre-treated individually with the substituted phthalimideAC 94,377 (1-(3-chlorophthalimido)-cyclohexanecarboxamide),GA4+7 and ABA was markedly improved under sub- and supra-optimaltemperature regimes. ABA was especially stimulatory at the sub-optimaltemperature. Metabolic alterations in the germinating embryosof treated seeds revealed an increased accumulation of solublesugars and proteins compared with the controls under stressingtemperatures. The activities of acid phosphatase, invertase,catalase and peroxidase were seemingly related to the alleviationmetabolism. It is suggested that gibberellins and abscisic acidmay have positive regulatory effects in triggering the systemfor stress alleviation. The interactive effect of NaCl salinity and gibberellic acidin the activities of cellulase and pectin lyase, and on thecontent of auxin and chlorophyll, has been determined duringleaf growth (fifth from base) in rice. The linear growth, chlorophyllcontent, activity of cellulase, and the auxin level of leaveswere markedly decreased when plants were exposed to salt stress(12 dS m–1). However pectin lyase activity did not registerany significant alteration in the leaves of salt-stressed plantscompared with the control. Treatment of plants with gibberellicacid (GA3) (10 ppm) increased the leaf growth and chlorophyllcontent with a concomitant rise in the activity of cellulaseunder stressed as well as non-stressed conditions. A markedincrease in the content of auxin was discernible in the leavesof salt-stressed plants treated with GA3 compared with non-treatedsalinized ones. An appreciable increment in the activity ofpectin lyase in response to GA3 administration was detectedonly in the leaves of non-stressed plants. These results indicatethat enhancement of cellulase activity and the augmentationof endogenous auxin content may be involved in the stimulationof rice leaf growth by GA3 under saline conditions. The growth response of Tapinanthus bangwensis (Engler and K.Krause) Danser to exogenous growth substances, indole-3yl-aceticacid (IAA), kinetin, and gibberellic acid (GA3), not only varieswith the concentrations of each of these substances in the mediumbut follows a similar trend in each case. Kinetin, IAA, andGA3 were found to enhance growth but not by very appreciableamounts, kinetin being the most effective growth promoter. Highconcentrations of these substances (5.0 and 10.0 ppm) exceptGA3 were toxic to the parasite. Gibberellic acid induced theproduction of accessory leaves at all the concentrations used.Kinetin was also effective in inducing accessory leaves butonly at a concentration of 1.0 ppm. Some of the lower concentrationsof the growth substances affected significantly some aspectsof the parasite's growth and development. Gibberellins GA1, GA4, GA8, GA9, GA19, GA20, GA29, GA44, GA81,indole-3-acetic acid (IAA) and abscisic acid (ABA) were identifiedin cambial region tissues of Eucalyptus globulus by comparingmass spectra and Kovats retention indices with those of authenticstandards. Using stable isotope labelled internal standardsGA19, GA20 and GA44 were quantified at levels of 2–7 ng(g fr wt)-1, other GAs were present at levels < 1 ng (g frwt)-1. Levels of IAA and ABA ranged from 417–1, 140 ng(g fr wt)-1 and 86–305 ng (g fr wt)-1 respectively. Experiments were carried out to explore the involvement of gibberellins(GAs) in the light-induced germination of Arabidopsis thaliana(L.) Heynh, using wild type (WT) and phytochrome-deficient mutants(phyA, phyB and phyAphyB deficient in phytochrome A, B and Aplus B, respectively). Seed germination of WT and phytochrome-deficientmutants was inhibited by uniconazole (an inhibitor of an earlystep in biosynthesis of GA, the oxidation of ent-kaurene) andprohexadione (an inhibitor of late steps, namely, 2rß-and 3rß-hydroxylation). This inhibition was overcomeby simultaneous application of 10-5 M GA4. The relative activityof GAs for promoting germination of uniconazole-treated seedswas GA4>GA1=GA9>GA20. The wild type and the phyA and phyBmutants had an increased response to a red light pulse in thepresence of GA1, GA4, GA9, GA20 and GA24 but there were no significantdifferences in activity of each GA between the mutants. Therefore,neither phytochrome A nor hytochrome B appears to regulate GAbiosynthesis from GA12 to GA4 during seed germination, sincethe conversion of GA12 to GA9 is regulated by one enzyme (GA20-oxidase). However, GA responsiveness appears to be regulatedby phytochromes other than phytochromes A and B, since the phyAphyBdouble mutant retains the photoreversible increased responseto GAs after a red light pulse. Effects of three phytohormones (IAA1, GA3 and kinetin) on grain-fillingand the pattern of 32P translocation from individual leavesto grains were studied at intervals of 7 days during the progressof reproductive development of rice (Oryza saliva L. cv. Jaya).The plants were sprayed with 100 µg ml–1 aqueoussolutions of the hormones at 100 days, when the plants wereentering the reproductive stage. Kinetin produced a pronouncedeffect on grain-filling as well as on 32P mobilization fromindividual leaf to grains and increased yield, possibly by increasingleaf longevity. GA3 and IAA also increased the grain-fillingand 32P mobilization significantly over control but the effectswere less marked than those of kinetin. Two cDNA probes (pRP60 and pRP80) were used to examine the hormonalregulation of expression of two cysteine endopeptidase genesin germinated whole seeds and de-embryonated seeds of rice.The pRP60 protein is a major rice cysteine endopeptidase namedREP-1 that digests rice glutelin, the main seed storage protein.The pRP80 protein has features of a cysteine endopeptidase buthas not yet been confirmed to be one. Neither mRNA was detectablein dry seeds, and the levels per seed increased sharply uponimbibition, reached peaks at d 6 to d 9, and then decreased.Both mRNAs were expressed in a seed-specific manner, but theaccumulation of pRP60 mRNA was about ten times greater thanthe other. When seeds were de-embryonated and incubated, theamounts of both mRNAs were reduced to very low levels, but inthe presence of 0.01 to 1 µM. gibberellic acid (GA3) bothagain reached high levels. Addition of abscisic acid (ABA) oruniconazole, an inhibitor of gibberellin biosynthesis, partlyeliminated the effect of GA3. Protein immunoblot analysis showedthat the accumulation of the REP-1 protein in seeds was regulatedby GA3 and ABA similarly to that of pRP60 mRNA, but the changein pRP60 mRNA with time after the onset of imbibition precededthat of the REP-1 protein. Dormancy was induced during storage of Liatris spicata cormsgrown in Israeli summer conditions, but plants left in soilcontinued vegetative growth. Corms of winter-grown plants sproutedfreely. Treatment with GA3 restored both sprouting and floweringin summer-grown corms, but in winter corms GA2 was effectiveonly after corms were stored at low temperature. All the plantsflowered after 4 weeks at 2 °C and GA3 treatment. The content of gibberellins in the main bud of freshly excavatedcorms decreased during the first 18 d of storage but increasedto the initial level after 4 months of cold storage. The numberof flowering stems increased to 2.5 per corm when corms werecold-stored up to 75 d, but decreased with a longer storage. Evidence that the synthesis of GA3 is involved in the growthof floral orga'ns of Pharbitis nil is presented. GAs in floralorgans at different developmental stages were surveyed usingTLC followed by the bioassay with two dwarf rice seedlings,‘Tanginbozu’ and ‘Waito-C’. The amountof GAs in the petal and stamen increased rapidly after the petalemerged from calyx, reached a maximum 12 hr before anthesis,then declined markedly thereafter. The GA content in the calyxremained unchanged before and after anthesis, and that in thepistil increased after anthesis. Pharbitis flowers containedat least two active GAs, one of which was probably GA3, theother appeared to be GA19. GA3 was detected in relatively largeamounts in both the petal and stamen during their rapid elongation.In the calyx, which showed little increase in fresh weight duringrapid flower growth, GA9 was the dominant GA. Exogenously suppliedGA3 promoted elongation of sections in excised young filaments.Sucrose was necessary for definite growth promotion by GA3.GA19 had little effect on filament elongation, and IAA was ratherinhibitive.
