A rapid batch method was developed for the analysis of Si, Ca, Mg, and K in a large number of plant tissue samples by fusion with lithium metaborate (LiBO3) in graphite crucibles with the use of a molybdenum blue spectrophotometric analysis of silicon and FAAS for Ca, Mg, and K. Our method was tested for whole plant analysis of mature wild rice (Zizania palustris L.). Analysis of Si in plant tissue with LiBO3 in graphite crucibles is reliable and fast. Thirty-six samples can be ashed overnight, fused in one hour the next day, and analyzed for Si within approximately two hours.  相似文献

Diffusible IAA and dominance phenomena in fruits of apple and tomato   总被引:3,自引:0,他引:3  
The relationships between indole-3-acetic acid (IAA) diffusing out of the fruit and competition among fruits, and between fruits and shoot tips were investigated using apple ( Malus domestica Borkh. cv. Jonagold) and tomato ( Lycopersicon esculentum Mill.) plants. Dominant fruits always had more diffusible IAA than subordinated, inhibited fruits. Alterations in dominance – by fruit- or shoot tip removal – led to significant changes in diffusible IAA by the remaining fruits. This change could be detected one day after dominance modification.
It is suggested that diffusible IAA is involved in the correlative signal regulating dominance relationship between fruits, and between fruits and shoots in apple and tomato.  相似文献

To elucidate compositional changes of the arteries with aging, the authors investigated the relationships among average contents of calcium, phosphorus, sulfur, and magnesium in the arteries by inductively coupled plasma-atomic emission spectrometry. The arteries used were the thoracic and abdominal aortas, coronary, common carotid, anterior, middle and posterior cerebral, vertebral, basilar, internal thoracic, axillary, radial, truncus celiacus, common, internal and external iliac, femoral, popliteal, and umbilical arteries. It was found that high correlations were found between the average contents of calcium and phosphorus, between the average contents of calcium and magnesium, and between the average contents of phosphorus and magnesium in the arteries, but not between the average contents of sulfur and the other elements. These correlations revealed that as the content of calcium and phosphorus increased in the arteries, the magnesium content increased simultaneously in the arteries, but the sulfur content did not. It is likely that magnesium forms compounds with phosphorus in the arteries.  相似文献

To overcome difficulty in phytopathogenic fungi control during storage of apple fruits, the effect of different storage conditions on the occurrence and development of Fusarium avenaceum and Alternaria alternata infections on apple cultivar “Cripps Pink” was investigated during and after storage. Inhibitory effects of wild oregano essential oil on apple fruit rots caused by F. avenaceum and A. alternata were also tested as possible rot control measure. Artificially inoculated apple fruits were kept in cold storage with normal (NA) and controlled (CA) atmosphere for 95 days and at room temperature only. The obtained results indicated that different storage conditions significantly affect necrosis development on apple fruits caused by F. avenaceum and Aalternata after storage, as well as during shelf life.  相似文献

We estimated the fluxes, inputs and outputs of Ca, K,and Mg in a Mexican tropical dry forest. The studywas conducted in five contiguous small watersheds(12–28 ha) gauged for long-term ecosystem research. A total of five 80 × 30 m plots were used for thestudy. We quantified inputs from the atmosphere,dissolved and particulate-bound losses, throughfalland litterfall fluxes, and standing crop litter pools. Mean cation inputs for a six-year period were 3.03 kg/ha for Ca, 1.31 kg/ha for K, and 0.80 kg/ha for Mg. Mean outputs in runoff were 5.24, 2.83, and 1.79 kg/ha, respectively. Calcium, K, and Mgconcentrations increased as rainfall moved through thecanopy. Annual Ca return in the litterfall (11.4 g/m2) was much higher than K (2.3 g/m2)and Mg (1.6 g/m2). Litterfall represented 99%of the Ca, 84% of the Mg, and 53% of the K, totalaboveground return to the soil. Calcium concentrationin standing litter (3.87%) was much higher than K(0.38%) and Mg (0.37%). These concentrations werehigher (Ca), lower (K), or similar (Mg) to those inlitterfall. Residence times on the forest floor were0.86, 1.17, and 1.77 yr for K, Mg, and Carespectively. Compared to the residence time fororganic matter at the site (1.31 yr), these resultssuggest slow mineralization for Ca in this ecosystem. Budget estimates were calculated for a wet and a dryyear. Results indicated that nutrients accumulated inthe dry but that nutrients were lost during the wetyear. Comparison of Ca, K, and Mg losses in streamwater with the input rates from the atmosphere for thesix-year period show that inputs are lower thanoutputs in the Chamela tropical dry forestecosystem.  相似文献

Two clones of Populus trichocarpa varying in growth rate were grown in nutrient solutions with a range of K supply. In the leaves an antagonism between the levels of K and Ca+Mg was found, this was most extreme in the older leaves. Analysis of the contents of these elements in leaf vacuoles and cytoplasm suggests that in the vacuoles the total contents of K+Ca+Mg are controlled to maintain ionic and osmotic balance. At low K supply, the K deficit in the cytoplasm is not balance by accumulation of Ca and Mg, and it is suggested that other organic osmotica may accumulate. The regulation of K+Mg+Ca supplied to the leaves seems to be regulated by the roots and is not simple competition of ions for uptake sites at the plasmalemma.  相似文献

Changes in ribosome population, RNA species and DNA composition in flower buds of apples ( Malus pumila Mill. cvs Ralls and White winter pearmain) were investigated during breaking of dormancy and development. After bursting of flower buds, total ribosomes increased approximately 4-fold, and the polyribosomal fraction increased from 66% to 94% of total ribosomes. The newly synthesized ribosomes were identified by incorporation of radioactive precursor. The observed decrease in specific radioactivity of the monoribosomes is caused by the recruitment of monoribo-somes into polyribosomes after breaking of dormancy.
In both cultivars, the 25S and 18S rRNA peaks increased to a high level on April 8. The peaks of low molecular weight RNA were apparently increased after initial swelling of the flower buds. The DNA of flower buds was separated into three bands by electrophoresis. The median band is the main band of nuclear DNAs. The ahead band and the slow-moving band are satellite components of nuclear DNAs, and they obviously rose after initial swelling of the flower buds. On April 8, when the flower buds had opened, two other small DNA bands could be detected. These results suggest that the changes in level of different ribosome populations, RNA species and DNA composition are related to dormancy breaking development of apple flower buds.  相似文献

Changes in texture are an integral part of ripening in most fleshy fruits and these changes are thought to be determined, primarily, by alterations in cell wall structure. Electron energy loss spectroscopy (EELS) imaging was used to obtain quantitative information on the levels of calcium and nitrogen in the cell walls of apple (Malus domestica Borkh. cv. Cox's Orange Pippin) fruits. Samples of fruit cortex were prepared for EELS by high-pressure freezing and molecular distillation drying to minimize loss and redistribution of soluble cell wall components such as calcium. The EELS imaging successfully resolved calcium and nitrogen levels in the middle lamella and primary cell wall. When the elemental compositions of the cell walls of Cox's apples from two sites in the UK were compared at harvest or after 6 months storage, the orchard which always produced consistently firmer fruit had significantly lower levels of cell wall calcium and higher levels of cell wall nitrogen. This result was unexpected since firm texture in apples and other fruits has been commonly associated with elevated levels of fruit calcium. The nitrogen-rich material in the sections used for EELS was insoluble in acidified methanol, indicating that it represented a high-molecular-weight component in the cell wall. Furthermore, total tissue hydroxyproline levels were greatest in material with elevated cell wall nitrogen, suggesting enhanced levels of wall structural proteins in the tissue. These data indicate a correlation between increased amounts of cell wall nitrogen and firm fruit texture. The ability of ethylene to stimulate respiration and advance the onset of rapid ethylene production was investigated at different times during storage of 'Gloster 69' apples in 2 kPa O2 at 1.5–3.5°C. Ethylene stimulated respiration in apples at 15°C immediately after harvest; maximal rates were recorded at 10–1000 μl I−1 but attainment of these rates was delayed after low O2 storage until day 3 of treatment at 15°C. The onset of rapid ethylene production at 15°C occurred later in non-ethylene-treated apples after storage than after harvest. Ethylene production was induced in some apples during ethylene treatment for 3 or 6 days; in others it was induced about 20 days after treatment, but a proportion of the fruit showed no induction in the 45-day duration of experiments. An ethylene treatment at 10 μl I−1 led to a near maximal increase in the frequency of induction of ethylene production at all times. After storage apples were mainly induced during treatment or not induced, whereas after harvest induction after treatment was more frequent. The presence of 2000 μl l−1 norbornadiene during ethylene treatment inhibited the stimulation of respiration and the induction of ethylene production; this inhibition was only partly reversed by ethylene at 1000 μl l−1 the experiments suggest that receptors for ethylene were present at all stages but that response capacity changed during storage.  相似文献

To examine whether there were differences between races in regard to the relationships among element contents in the arteries, the authors investigated the relationships among the average contents of calcium, phosphorus, sulfur, and magnesium in the 18 kinds of the Thai artery. After ordinary dissection by medical students at Chiang Mai University was finished, the thoracic and abdominal aortas, ramifying site of the abdominal aorta into the common iliac arteries, coronary, common carotid, internal thoracic, subclavian, axillary, brachial, radial, superior and inferior mesenteric, renal, common iliac, internal iliac, and external iliac arteries were resected from the subjects who consisted of 12 men and 3 women, ranging in age from 39 to 84 yr. The femoral and posterior tibial arteries were resected from the subjects, consisting of 15 men and 5 women, ranging in age from 25 to 88 yr. The element content of the arteries was analyzed by inductively coupled plasma-atomic emission spectrometry. It was found that there were extremely significant direct correlations among the average contents of calcium, phosphorus, and magnesium in the 18 kinds of the Thai artery, but no significant correlations were found between the average contents of sulfur and elements, such as calcium, phosphorus, and magnesium. These results were in agreement with those of the Japanese arteries. Therefore, it was suggested that there was no significant difference between the arteries of the Thai and the Japanese in the relationships among calcium, phosphorus, sulfur, and magnesium.  相似文献

The influence of silicon treatment on the levels of calcium and magnesium in blood serum and tissues was studied in rats. The concentrations of both elements were estimated in samples of sera and tissues of rats receiving per os a soluble, inorganic silicon compound—sodium metasilicate nonahydrate (Na2SiO3·9H2O (REACHIM, USSR)), dissolved in the animals' drinking water. A decrease of magnesium concentration in serum was observed with accompanying elevation of registered calcemia. Moreover, a reduction of tissue calcium levels was found with a simultaneous increase of magnesium tissue pool. The results provide evidence for silicon involvement in mineral metabolism. It could result in a modification of pathological processes concerning bone tissue.  相似文献

Reduced glutathione (GSH) was applied to prevent browning in shoot tip explants of apple (Malus pumila Mill.). Development was compared between shoot tips treated either by (a) dipping into 0.1 mm GSH solution prior to culture (dip treatment), (b) dipping into the GSH solution and transferring to a medium containing 0.1 mm GSH (dip-and-add treatment), or (c) without dipping or culturing with GSH (control). In the dip treatment, 100% of shoot tips developed into normal shoots after 120 days, while the results with the dip-and-add treatment and control were 50 and 40%, respectively. The results show that application of antioxidant GSH in the initial phase of culture promoted the normal development of shoot tips. To examine whether an accumulation of elements in the arteries with aging differs between human and animal, the authors investigated the relationships among element contents in the arteries of the Japanese monkeys. The Japanese monkeys consisted of five males and four females, ranging in age from 2 to 29 yr. The aorta, common and external iliac, femoral, common carotid, subclavian, and axillary arteries were resected from the monkeys and element contents were determined by inductively coupled plasma-atomic emission spectrometry. It was found that there were very high correlations between calcium and phosphorus contents, between calcium and magnesium contents, and between phosphorus and magnesium contents in all of the monkey arteries. In addition, significant correlations were found among the other element contents in some, but not all of the arteries. These results were consistent with the foregoing findings of the human arteries. It is likely that magnesium forms compounds with phosphorus or calcium in the monkey arteries.  相似文献

An experiment was performed in which Ti4+-ascorbate was sprayed onto plum trees in several combinations with other commercial compounds containing Ca2+ and Mg2+ to study the effects on the commercial quality of fruits, with special focus on improving their resistance against postharvest handling damage.

All the treatments containing titanium increased the tree performance (branch elongation, flowering and fruit setting intensities) and fruit size. At harvest fruits from the Ti-treated trees showed improved resistance to compression and penetration, as well as a decrease in weight-loss during postharvest storage. A similar response was obtained for the external colour, though all the treatments seemed to delay somewhat the apparent ripening status. Nevertheless, the fruits from Ti-treated trees showed a better behaviour in the evolution of the colour parameters during storage than did the control fruits.

Titanium application significantly increased the calcium, iron, copper and zinc concentrations in peel and flesh. This improvement in the calcium absorption is explained as a consequence of the beneficial effect of titanium on the absorption, translocation and assimilation processes.  相似文献

To elucidate the accumulation of elements in the arteries with aging, the authors investigated age-related changes of elements in human arteries, such as the thoracic aorta, femoral, basilar, coronary, radial, and common iliac arteries by inductively coupled plasma-atomic emission spectrometry. The subjects consisted of 17 men and 9 women, ranging in age from 55 to 92 yr in the cases of the five arteries, except for the common iliac arteries, in which the subjects consisted of 16 men and 8 women, ranging in age from 65 to 93 yr. It was found that there were significantly direct correlations between calcium and phosphorus contents and between calcium and magnesium contents in all of the six arteries: thoracic aorta, femoral, basilar, coronary, radial, and common iliac arteries. Significantly direct correlations were also found between phosphorus and magnesium contents in the five arteries, except for the basilar artery. In contrast, significantly inverse correlations were found between calcium and sulfur contents and between phosphorus and sulfur contents in the four arteries, except for the coronary and radial arteries. These revealed that the accumulation of calcium and phosphorus in the arteries was accompanied by an increase of magnesium in the arteries and by a decrease of sulfur in the arteries.  相似文献

南方稻田土壤大面积酸化是水稻生产的主要限制因子.尽管石灰作为酸化土壤调理剂已广泛应用,但大量或长期施用石灰不仅会引起土壤板结,而且会导致土壤钙、钾、镁等元素的平衡失调.硅钙钾镁肥由于溶解度更低、养分全面是良好的替代材料.为了明确硅钙钾镁肥阻控土壤酸化的效果和作用,本研究采用连续4年的硅钙钾镁肥田间定位试验,以农民习惯施肥为对照,分析在农民习惯施肥基础上增施750、1125、1500和1875 kg·hm-2硅钙钾镁肥下稻田土壤pH、交换性酸、交换性盐基离子和有效硅的动态变化.结果表明: 农民习惯施肥导致土壤pH、土壤交换性盐基和盐基饱和度逐年下降,土壤交换性酸逐年增加.与之相反,硅钙钾镁肥处理显著提高了土壤pH值,提高幅度随硅钙钾镁肥施用次数或用量的增加而增大.连续多次施用硅钙钾镁肥有效促进了盐基离子在土壤中的累积和土壤交换酸的消耗,特别是土壤交换性Ca2+、Mg2+的累积和土壤交换性Al3+的消耗,硅钙钾镁肥用量越大,积累或消耗的量越多,但速率相对越慢.土壤交换性酸消耗量中,硅钙钾镁肥释放的交换性盐基离子和相应碱贡献了108.8%,是交换性酸减少的主要途径.硅钙钾镁肥在改良稻田土壤酸性的同时,土壤有效硅含量逐年增加,增幅随硅钙钾镁肥施用量的增加而显著增大.总之,农民习惯施肥导致土壤持续酸化,酸化率为2.86 kmol H+·hm-2·a-1,硅钙钾镁肥能有效阻控酸化过程,产生了大量碱(9.69~18.44 kmol OH-·hm-2·a-1),释放的Ca2+、Mg2+盐基离子和相应碱是土壤酸化阻控的主要作用因子.  相似文献

Headspace volatiles from apple-bearing twigs were collected in the field with a Radiello sampler during three different diurnal periods over the complete fruit growing season. Analyses by thermal desorption-GC-MS identified a total of 62 compounds in changing quantities, including the terpenoids alpha-pinene, camphene, beta-pinene, limonene, beta-caryophyllene and (E,E)-alpha-farnesene, the aldehydes (E)-2-hexenal, benzaldehyde and nonanal, and the alcohol (Z)-3-hexen-1-ol. The variations in emission of these plant odours were statistically related to temperature, humidity and rainfall in the field. Remarkably, rainfall had a significant positive influence on changes in volatile release during all three diurnal periods, and further factors of significance were temperature and relative humidity around noon, relative humidity in the late afternoon, and temperature and relative humidity during the night. Rainfall was associated consistently with an increase in the late afternoon in terpene and aldehyde volatiles with a known repellent effect on the codling moth, one of the key pests of apple fruit. During the summer of 2003, a season characterized by below-average rainfall, some postulated effects of drought on trees were tested by establishing correlations with rainfall. Emissions of the wood terpenes alpha-pinene, beta-pinene and limonene were negatively correlated with rainfall. Another monoterpene, camphene, was only detected in this summer but not in the previous years, and its emissions were negatively correlated with rainfall, further supporting the theory that drought can result in higher formation of secondary metabolites. Finally, the two green leaf volatiles (E)-2-hexenal and (Z)-3-hexen-1-ol were negatively correlated with rainfall, coinciding well with the expectation that water deficit stress increases activity of lipoxygenase. To our knowledge, this work represents the first empirical study concerning the influence of abiotic factors on volatile emissions from apple trees in situ.  相似文献

ABSTRACT

In Experiment 1, the effects of calcium maltobionate (MBCa) on calcium and magnesium absorption were examined using male rats. Four diets were designed in which 25%, 50%, and 100% of calcium carbonate (CaCO3, Control) were substituted with MBCa and were designated as MBCa-25, MBCa-50, and MBCa-100, respectively. The cecal concentration of short-chain fatty acids was significantly higher in groups MBCa-50 and MBCa-100; however, pH of cecal contents did not significantly differ among the groups. Retention rates of calcium and magnesium were significantly higher in all MBCa groups as compared to the Control. In Experiment 2, the efficiency of calcium absorption was compared using everted sacs of jejunum and ileum with CaCO3 and MBCa as calcium sources. More calcium from MBCa was absorbed as the concentration of calcium increased in comparison to CaCO3. It was concluded that MBCa is a better calcium source than CaCO3 in terms of both calcium retention and absorption.

The effects of simultaneous changes of calcium, magnesium, iron, copper, and zinc concentrations were evaluated in normal human T and B lymphocytes, cultured in cation-depleted media. Optimal concentrations for thymidine incorporation (TI) in both cell populations were Fe and Zn 15 μM and Cu 5 μM; for t cells Ca 2 mM and Mg 4 mM; for B cells Ca 4 mM and Mg 6 mM. TI decreased with increasing molarity of cations and the decrease was particularly apparent with Cu. Minimal amounts of Ca and Mg (0.5 mM) were necessary for growth, even in presence of optimal concentrations of Fe, Cu, and Zn. Fe and Cu showed synergistic stimulatory effects at low concentrations and synergistic inhibitory effects at high concentrations. Antagonism between Fe and Zn, Cu and Zn, and Ca and Zn was also demonstrated. CD4/CD8 increased with PHA stimulation in presence of Zn, and decreased with ConA stimulation in presence of Zn or Fe. The results demonstrate: (1) the relationship and interdependence of Fe, Cu, and Zn concentrations in modulating the growth of normal lymphocytes; (2) the stimulatory effects of Fe on B cells and Zn on CD8 positive cells; (3) the inhibitory effect of Cu at concentrations lower than those of Fe and Zn; (4) the requirement of Ca and Mg in certain concentration and ratio for the action of the other cations; and (5) the Ca and Mg requirement for the growth of B cells higher than T cells.  相似文献
