韩国赫坎按蚊复合体3成员种核糖体DNA内转录间隔2区的比较(英文) 总被引:5,自引:0,他引:5
本文对韩国中华按蚊、雷氏按蚊和八代按蚊核糖体DNA (rDNA)内转录间隔 2区 (ITS2 )序列进行了比较研究。用PCR扩增的rDNA ITS2片段直接测序 ,每种蚊测定 3个个体 ,结果显示 :韩国中华按蚊、雷氏按蚊和八代按蚊的rDNA ITS2序列长度分别为 4 6 8bp、 4 51bp和 4 53bp ,GC含量分别为 4 4 .87%、 4 6 .2 %和 4 5.7% ,3种按蚊序列差异范围为 12 .16 %— 30 .74 %。研究表明 ,rDNA ITS2序列差异可用于韩国中华按蚊、雷氏按蚊和八代按蚊的分子鉴别。 相似文献

我国中华按蚊群体分子遗传多态研究 总被引:10,自引:0,他引:10
利用RAPD-PCR技术研究了采自我国9省10个代表点的中华按蚊Anopheles sinensis自然群体的遗传多态现象,根据23个RAPD等位基因位点进行了分析。结果显示:①多态位点比例为68.2%~86.4%,期望平均杂合度为0.249~0.348,说明中华按蚊群体具广泛的遗传多态现象;②利用3种方法计算Fst和θ,平均值为0.069~0.111,相应的迁移率Nm为2.0~3.4,表明基因流水平较低;③中华按蚊各自然群体间的遗传一致性达0.8795~0.9973,平均遗传距离为0.041±0.033,属种内变异范围。聚类分析显示,群体间遗传距离与地理位置无对应关系。 相似文献

ABSTRACT To figure out how far spread the malaria parasite infected mosquitoes from Demilitarized Zone (DMZ). We investigated the sporozoite-infected mosquitoes in Chosan-ri and Hajiseok-ri of Paju-shi, Tonae-dong of Koyang-shi, Mudeung-ri of Yonchon-gun, Soyo-dong of Tongduchon-shi, and Tukjeong-ri of Yangju-gun, Kyonggi-do province in 1998. They were known as highly epidemic areas in the Republic of Korea. Anopheline mosquitoes were collected by human biting collection method from the 2nd week of June to the 5th week of August. A total of 3,256 anopheles mosquitoes were captured in these areas. A sandwich enzyme-linked immununosorbent assay (ELISA) for identifying Plasmodium vivax sporozoites in mosquitoes was used in this survey. Only one mosquito which was captured in Chosan-ri, on 2nd week of July showed positive reaction in this test. This mosquito was identified as Anopheles sinensis. This showed a 0.068% (1/1,465) positive rate in Chosan-ri on 2nd week of July. We could not detect positive mosquitoes the other areas. So we concluded the parasite infected mosquitoes may be highly limited near the DMZ. 相似文献

Abstract Differences in sequences of ribosomal DNA second internal transcribed spacer (ITS) among Anopheles sinensis, An. lesteri and An. yatsushiroensis from Korea were compared. The PCR amplified rDNA-ITS2 fragments were sequenced directly. Three samples of each species were individually determined.
Lengths of the ITS2 regions were 468bp in An sinensis , 451bp in An lesteri and 453bp in An yatsushiroenesis and GC contents were 44.87 % 46.2 % and 45.7 % respectively. Variations of the sequences ranged from 12.16 % to 30.7 % among the three species. The differences of the rDNA-ITS2 sequences would be useful for molecular identification of the three members of Anopheles hycanus complex from Korea. 相似文献

The susceptibility of members of the Anopheles sinensis Group in Korea to insecticides was evaluated under laboratory conditions using 15 insecticides currently used by local public health centers in Korea. The insecticides included eight pyrethroids, six organophosphates and a pyrazol analogue. Based on their LC50 values, the order of susceptibility of An. sinensis larvae to the insecticides was bifenthrin, chlorfenapyr, α-cypermethrin and λ-cyhalothrin, with values of 0.009, 0.04, 0.06 and 0.08 p.p.m., respectively. The least susceptibility was obtained with fenitrothion, with an LC50 of 7.7 p.p.m. In the comparative resistance test, the resistance ratios (RR) of 14 insecticides were compared to each other using two strains of members of the An. sinensis Group collected in the locality in 2001 and 2008. Anopheles spp. demonstrated higher RR to organophosphates such as fenthion, and low RR for the pyrethroids. Among the organophosphates, fenthion had the highest RR of 33.3 and 270.0 fold differences for LC50 and LC90 values, respectively. Among the pyrethroids, permethrin was observed to have the highest RR of 3.8 and 1.8 fold differences for LC50 and LC90 values, respectively. However, there were no significant differences in susceptibility to chlorfenapyr, chlorpyrifos, deltamethrin and fenitrothion. An. sinensis s. l. was more susceptible to the six insecticides bifenthrin, λ-cyhalothrin, α-cypermethrin, cypermethrin, cyfluthrin and pyridafenthion, showing 0.03, 0.06, 0.3, 0.3, 0.4 and 0.4 fold differences in resistance rates (RR LC50 ), respectively. 相似文献

ABSTRACT The Ultrastructure of rectum epithelial cells in the mosquito larvae, Anopheles sinensis Wiedemann, was studied using electron microscope. The rectal epithelium forms rectal papillae composed of the absorptive cells and the surrounding basal cells. Moreover, rectal epithelium was covered with thin cuticular intima. Apical plasma membrane of the epithelial cells had infoldings and in between them, mitochondria developed into elongated shape were attached. In addition, the membrane infoldings reach down into the cell cytoplasm to form several layers of leaflet-like prolongations. On both sides of these prolongations were also large, well-developed mitochondria. Their formation was that mitochondria were attached to 3 μm length and 4–13 layers of membrane wrinkle lump. Many spherites, which are lamelated crystals that form an illusory structure in concentric circles inside of the cytoplasm of epithelial cell were observed. Basal plasma membrane in the epithelial cells was also wrinkled to promulgate into the cytoplasm to become basal infoldings producing canaliculi in basal labyrinth formation. There were many mitochondria scattered in these formations as well. On the bottom of the epithelial cell, basal lamina was attached and between basal lamina and muscle bundle was subepithelial space, which is connective tissue. The granular formulation of 0.5% pyriproxyfen is a mosquito insect growth regulator larvicide. It was evaluated for residual activity for killing and/or emergence inhibition against 3rd stage larvae of Anopheles sinensis in the laboratory. Mortality rates ranged from 60.9% to 79.7% at 0.01, 0.05 and 0.1 mg/L of water during the first seven days. Except for the lowest concentration (0.01 mg/L) mortality rates were greater than 80% from day 8 through 28. Mortality rates for 0.01 mg/L concentrations were 70.4% from days 8‐14, and thereafter exceeded 80% through day 28. The residual activity gradually increased post‐treatment since the granular formulation of pyriproxyfen has a gradual solubility and increases concentration of active ingredient over time. The mortality rates of An. sinensis larvae and pupae eventually reached 100% for the three (0.01, 0.05, 0.1 mg/L) concentrations. 相似文献

Zika virus(ZIKV) has been isolated from mosquitoes such as Aedes, Mansonia uniformis, and Culex perfuscus; However,the isolation of ZIKV from Anopheles sinensis and Culex tritaeniorhynchus has not yet been reported. In June and July2018, 22,985 mosquitoes and 57,500 midges were collected in Jiangxi Province in southeastern China. Among them, six strains of ZIKV were isolated from mosquitoes: four from An. sinensis and two from Cx. tritaeniorhynchus. Molecular genetic analysis showed that the ZIKV isolated from An. sinensis and Cx. tritaeniorhynchus belonged to genotype 2 in the Asian evolutionary branch of ZIKV. In addition, the ZIKV strains isolated from An. sinensis and Cx. tritaeniorhynchus had amino acid substitutions identical to ZIKV strains prevalent in South America since 2015. This study is the first to isolate ZIKV from mosquito specimens collected in the wild of Jiangxi Province, China; This is also the first time that ZIKV has been isolated from An. sinensis and Cx. tritaeniorhynchus. Given that An. sinensis and Cx. tritaeniorhynchus have a very wide geographical distribution in China and even in eastern and southern Asia, the isolation of several strains of ZIKV from these two mosquitoes poses new challenges for the prevention and control of ZIKV infection in the mainland of China and countries and regions with the same distribution of mosquitoes. 相似文献

Abstract Phenol oxidase (PO activity was investigated in cell-free hemolymph collected from Anopheles sinensis and Aedes albopictlcs female adults intrathoracically inoculated with Brugia dayi microfilariae (mff), inoculated with saline alone, or from uninoculated mosquitoes. The activity of PO from uninoculated 1-day-old mosquitoes was significantly greater than those of mosquitoes on 3, 12 and 16 day. PO activity in mff-inoculated A. sinensis at 24 h postinoculation (PI) was 2–3 times higher as compared with uninoculated or saline-inoculated mosquitoes. Inoculation of B. malayi mff into A. albopictus elicited ?-fold increase in PO activity at 24 h PI as compared with uninoculated mosquitoes. Immune-activated levels of PO activity in A. alboplctus was significantly higher as compared with those seen in A. sinensis. The relationship of observed differences in PO activity to differences in immunological capability between A. sinensis and A. albopictus is discussed. 相似文献
