对广州地区常见的21科41种(变种)植物叶片几丁质酶系研究的结果表明,所有受试植物都具有几丁质酶活性。几丁质酶不仅存在于被子植物的双子叶植物和单子叶植物中,而且也存在于裸子植物及蕨类植物中。几丁质酶活性及比活性均较高的有蕹菜、葱、蕨类植物、蒜、茄科植物、玉米、菜豆、番木瓜等。植物一般都具有两种几丁质酶:外切酶和内切酶。几丁质外切酶活性及比活性均较高的有蕨类植物、葱、茄科植物等。几丁质内切酶活性及比活性均较高的有蕹菜、裸子植物等。不同植物几丁质外切酶与内切酶的比例相差较大。有些植物的几丁质酶系以外切酶为主,如茄科植物、大部分豆科植物、番木瓜等;有些植物以内切酶为主,如裸子植物、伞形科植物、蕹菜等;有些植物的外切酶与内切酶含量相差不大。  相似文献

Genes from the biocontrol fungus Trichoderma atroviride encoding the antifungal proteins endochitinase or exochitinase (N-acetyl--D-hexosaminidase) were inserted into Marshall McIntosh apple singly and in combination. The genes were driven by a modified CaMV35S promoter. The resulting plants were screened for resistance to Venturia inaequalis, the causal agent of apple scab, and for effects of enzyme expression on growth. Disease resistance was correlated with the level of expression of either enzyme when expressed alone but exochitinase was less effective than endochitinase. The level of expression of endochitinase was negatively correlated with plant growth while exochitinase had no consistent effect on this character. Plants expressing both enzymes simultaneously were more resistant than plants expressing either single enzyme at the same level; analyses indicated that the two enzymes acted synergistically to reduce disease. Selected lines, especially one expressing low levels of endochitinase activity and moderate levels of exochitinase activity, were highly resistant in growth chamber trials and had negligible reduction in vigor relative to control plants. We believe that this is the first report of resistance in plants induced by expression of an N-acetylhexosaminidase and is the first report of in planta synergy between an exochitinase and an endochitinase.  相似文献

Chitinolytic enzymes produced by ovine rumen bacteria   总被引:1,自引:0,他引:1  
Two strains of clostridia, isolated from the rumen fluid of sheep as potential antagonists toward anaerobic fungi showed a complete array of chitinolytic enzymes. Enzyme tests in cultures demonstrated endochitinase, exochitinase,N-acetylglucosaminidase, chitosanase and chitin deacetylase activities mainly in the extracellular fractions. In all samples, the highest was the activity of exochitinase (600–1100 nmol mL−1 h−1); the activity of endochitinase (280–500 nmol mL−1 h−1) was also significant. Chitinases were stimulated in the presence of reducing compounds and no dependence on cations was observed. In both strains different isoforms of chitinases of molar mass 36–96 kDa were detected. The chitinases from our isolates lyzed cell walls of anaerobic fungiin vitro and inhibited the activity of fungal β-1,4-endoglucanases. Of the two bacteria examined, one was more effective in both antifungal effects.  相似文献

It has been reported that the susceptibility of insect herbivores to entomoviruses is affected by phytochemicals ingested during the acquisition of viral inoculum on the foliage of host plants. However, the relationship between this susceptibility and phytochemicals is poorly understood. To test this hypothesis of plant‐mediated effects on this susceptibility, we measured the effects of foliage from three plants, soybeans (Glycine max), collards (Brassica oleracea) and water convolvuluses (Ipomoea aquatica), on the susceptibility of larval beet armyworm (Spodoptera exigua) to nucleopolyhedrovirus (NPV), and analysed six foliar chemicals (total phenolics, peroxidase [POD], catalase [CAT], superoxide dismutase [SOD], endochitinase and exochitinase) in the three plants, respectively. The results of exponential modelling indicated that the LD50s (median lethal dose) of NPV to larvae increased with the increase in both phenolics and POD but declined with the increase in four other foliar chemicals, while the opposite trend was found between median lethal time (LT50) of NPV and the six foliar chemicals. This study reveals that phenolics and POD decrease host susceptibility to the entomoviruses and that CAT, SOD, endochitinase and exochitinase increase this susceptibility.  相似文献

Since chitin is degraded by a combination of both endo- and exochitinases, it is likely that both enzymes will be present in a crude extract. Currently used substrates for detecting endochitinase activity suffer from the fact that they could easily be digested by the contaminating exochitinase, thus giving either a false-positive or an inaccurate reading of the endochitinase activity. Using Photorhabdus luminescens, a bacterium symbiotically associated with insect-parasitic nematode Heterorhabditis bacteriophora as an exemplary system, we have identified these two chitinases by a simple "fluorimetric zymography" procedure. The exochitinase is a metalloenzyme and its activity is inhibited by 1,10-phenanthroline. Once the exochitinase activity is detected in a crude extract, its contribution must be eliminated before accurate determination of the endochitinase activity can be carried out. Specific properties of these enzymes including the pH activity profile, the requirement of metal ions for activity, and the molecular weight of the enzymes are among the factors to be considered in developing assaying procedures for endochitinase.  相似文献

Effects of Water Stress on Fruit Quality Attributes of Kiwifruit   总被引:5,自引:0,他引:5  
Four-year-old kiwifruit vines (Actinidia deliciosa(A. Chev.)C. F. Liang et A. R. Ferguson var.deliciosacv. Hayward) werestudied to determine response of the plant and effects on fruitquality when irrigation water was withheld either early or latein the growing season. The greatest effect on fruit growth occurredwhen water was withheld early in the season. Harvest weightof fruit from early-stressed vines was approx. 25% less thanthe weight of fruit on control vines. Early season water stressresulted in a transient increase in concentrations of solublecarbohydrates in both leaves and fruit. This was accompaniedby a reduction in stomatal conductance of the leaves. Starchlevels in leaves but not fruit were reduced by both stress treatments.Concentrations of sucrose at harvest in fruit from vines stressedlate in the season were markedly higher than in other fruit,and softness of the fruit was unaffected. Xyloglucan endotransglycosylase (XET) activity was measured in apple (Malus domestica Borkh. cv. Braeburn) pericarp and kiwifruit (Actinidia deliciosa [A. Chev.] C. F. Liang et A. R. Ferguson var. deliciosa cv. Hayward) outer pericarp and core tissues in order to establish whether a correlation exists between the activity of the enzyme and different stages of fruit development Whereas the growth rate of kiwifruit paralleled changes in XET activity throughout fruit growth, that of apple did not. Both fruits showed the highest XET activity, on a fresh weight basis, in the first two weeks after anthesis when cell division was at its highest. XET activity then decreased sharply, but as the fruit increased in size (4–8 weeks after anthesis) there was a concomitant increase in XET activity in both fruits. In the latter stage of fruit development (16–26 weeks after anthesis) XET activity increased to peak at harvest in apple fruit. During this time there was relatively little increase in fruit size and presumably therefore minimal cell expansion. XET activity then declined as fruit softened after harvest. In core tissue from kiwifruit, XET activity increased throughout the later stages of fruit growth to harvest maturity in a similar manner to apple, but continued to increase after harvest until fruit were ripe. In contrast, XET activity in the outer pericarp of kiwifruit did not increase until ripening after harvest. In apple tissue up to 30% of the XET activity was cell wall bound and could not be solubilised, even in buffer containing 2 M NaCl. The results implicate XET in cell wall assembly during cell division and expansion early in apple and kiwifruit growth. However, the disparity between apple and kiwifruit with respect to XET activity late in fruit development and ripening and the different affinities of the enzyme for the cell wall in each fruit, suggest that XET has several roles in plant development, not all of which are related to cell wall loosening during periods of accelerated growth.  相似文献

Chitinolytic activity of Serratia marcescens Nima (130 U ml−1) was up to 43 times higher than those produced by other S. marcescens strains. This strain synthesized an endochitinase (Chi-60), an exochitinase (Chi-50) and a novel N-acetylglucosaminidase. This latter showed two putative isoforms (Chi-180.5 and Chi-180.8) with isoelectric points of 5 and 8.1, respectively.  相似文献

 Localization of chitinolytic activities in Fagus sylvatica (beech) mycorrhizas was examined using a range of fluorogenic 4-methylumbelliferyl [4-MU-(GlcNAc)1–4] substrates in order to distinguish between exochitinase, endochitinase and β-N–acetylglucosaminidase activities. The validity of the technique was confirmed using onion epidermis cells. In the beech mycorrhiza, endochitinase activity was not detectable above background fluorescence. Exochitinase activity was detected in the fungal sheath and the Hartig net. β-N–Acetylglucosaminidase activity was also mainly associated with the fungal sheath and Hartig net. Individual fungal hyphae extending from these structures also showed substantial β-N–acetylglucosaminidase activity. The cortical cell walls of the host in the Hartig net region also fluoresced brightly. The localization of β-N–acetylglucosaminidase activity was confirmed using a chromogenic histochemical reagent, 5-bromo-4-chloro-3-indolyl-N–acetyl-β-d-glucosaminide (X-GlcNAc). Magnesium deficiency was associated with large yield reductions in a five-year-old commercial kiwifruit (Actinidia deliciosa) orchard. The effect on yield resulted primarily from a reduction in fruit numbers, there being no difference in mean fruit weight between fruit harvested from affected and unaffected vines. Magnesium deficiency had no deleterious effect on postharvest storage characteristics of fruit stored at 0.5–1°C for 18 weeks; fruit from deficient vines were firmer but had slightly lower soluble solids than fruit from control vines. Although deficiency symptoms were first observed on the basal leaves of the non-fruiting shoots mid season, indications of the impending deficiency could be established very early in the season using foliar analysis. Magnesium concentrations in youngest fully expanded leaves (YFEL) on the affected vines were less than 2.0 g kg−1 DM four weeks after budbreak and remained below this value for the rest of the season; concentrations in YFEL on unaffected vines did not decrease below this value and gradually increased after fruitset to 4.5 g kg−1 DM at harvest. To avert potential production losses, it is suggested that soluble magnesium fertilizers (containing at least 200 kg ha−1 Mg) should be broadcast early in the season if foliar magnesium concentrations less than 2.0 gkg−1 DM are measured four–six weeks after budbreak.  相似文献

Barley ( Hordeum vulgare L.) chitinases (EC 3.2.1.14) were found to be distributed and induced in highly tissue specific patterns. Out of 6 chitinases investigated 3 were present in leaves and only a class II chitinase (molecular mass 24 846 ± 5 Da, pI≥9.8) was markedly induced in leaves heavily infected with powdery mildew ( Erysiphe graminis f. sp. hordei ). The class II chitinase and a novel class III chitinase (molecular mass 30 kDa, pI≥9.8) were found in intercellular washing fluid of leaves, suggesting extracellular deposition. Neither of these two proteins were induced after infiltration of sodium salicylate (2 m M , pH 6.5) or nickel chloride (2 m M ). The class III chitinase showed exochitinase activity in addition to endochitinase activity. No grain specific chitinases were found in leaves after any of the stresses applied. In contrast, 3 grain specific chitinases and one of the leaf chitinases were found in in vitro cultures.  相似文献

Five cDNAs for genes differentially expressed during fruit development of kiwifruit (Actinidia deliciosa var.deliciosa cv. Hayward) were isolated from a library made from young fruit, 8–10 days after anthesis. One gene (pKIWI503) has low levels of expression in young fruit but is induced late in fruit development and during fruit ripening, and has some homology to plant metallothionein-like proteins. The other four genes are highly expressed in young fruit with reduced expression in the later stages of fruit development. pKIWI504 has strong homology to plant metallothionein-like proteins and pKIWI505 exhibits homology to the -subunit of the mitochondrial ATP synthase gene. The two other genes (pKIWI501 and 502) encode proteins with no significant homology to other known sequences.  相似文献

The novel chitinolytic bacterium Clostridium beijerinckii strain JM2 was isolated from the stool of healthy volunteers supplied daily per orally with 3 g of chitosan. The bacterium grown on colloidal chitin produced a complete array of chitinolytic enzymes. Significant activities of endochitinase, exochitinase and chitosanase were excreted into the medium (301, 282 and 268 nkat/μg protein, respectively). The high cellular activity of N-acetyl-β-glucosaminidase (NAGase) and chitosanase were detected (732.4 and 154 nkat/μg protein, respectively). NAGase activity represented the main activity associated with the cellular fraction. The activities of both enzymes tested increased from 20 to 50 °C; the optimum reaction temperature estimated being 50 °C. Endochitinase as well as NAGase showed an activity in the pH interval of 4.0–8.0; the optimum pH values were 6.5 and 6.0, respectively. The extracellular endochitinase complex consisted of six isoenzymes with molar mass of 32–76 kDa; in the cellular fraction five bands with molar mass of 45–86 kDa were detected. Exochitinase activity was demonstrated in the form of three bands (with molar mass of 30–57 kDa), NAGase activity displayed one band of 45 kDa.  相似文献

The role of the chitinolytic enzymes in plants is not necessarilyrestricted to plant defense. Tomato plants transformed with an endochitinaseand a chitobiosidase gene from Streptomyces albidoflavus andgrowth under greenhouse conditions showed a significant reduction in plantheight, and reduced time to flowering compared with the control(non-transformed) plants. The levels of chitobiosidase and endochitinaseactivity in the transgenic tomato plants were positively correlated with earlyflowering, and negatively correlated with plant height. We have not determinedwhether these effects are exclusively due to the expression of the transgenesof endochitinase and chitobiosidase from S. albidoflavus orthe additive effect of these 2 enzymes combined with the endogenouschitinolytic enzymes produced by the plants. However, when control plants were trimmed,early flowering was observed compared with the controls that were not trimmed, whichindicates that wound induced proteins such as chitinolytic enzymes affect thetime of flowering. In addition, the expression of the endochitinase andchitobiosidase genes significantly increased the number of flowers and fruit onthe plants, resulting in an increase in yield of fruit. One of the primarygoals of crop breeding programs is to increase the productivity of plants. These twogenes were directly associated with plant productivity, and should be studied further.  相似文献

Four methods of determining the substrate requirements for synthesisof a kiwifruit [Actinidia deliciosa (A. Chev.) C. F. Liang etA. R. Ferguson var. deliciosa cv. Hayward] berry were comparedusing data derived from common kiwifruit berry samples collectedfrom anthesis to fruit maturity. The four methods were basedon fruit proximal analysis, elemental analysis, heats of combustion,or tissue carbon content. All methods gave similar patternsof seasonal costs and values of final cost to the plant (mean1.21 g glucose g–1 season–1) but there was lessagreement for growth respiration (mean 0.147 g glucose g–1season–1). This is the first time that a continuous recordof growth cost over the course of development has been presented,and the trends in seasonal cost reflect the uptake into andsynthesis of the different biochemical constituents in the fruit.The differences between the results of each method reflect theunderlying assumptions used in their development. Transgenic broccoli plants expressing a Trichoderma harzianum endochitinase gene were obtained by Agrobacterium tumefaciens-mediated transformation. PCR and Southern blot analysis confirmed the presence of the gene in plants initially selected via resistance to kanamycin. Primary transformants (T0) and selfed progeny (T1) were examined for expression of the endochitinase gene using a fluorometric assay and for their resistance to the fungal pathogens Alternaria brassicicola and Sclerotinia sclerotiorum. All transgenic plants with elevated endochitinase activity had the expected 42 kDa endochitinase band in western blot analysis, whereas no such band was detected in the non-transgenic control. Leaves of most mature T0 plants had 14–37 times higher endochitinase activity than controls; mature T1 plants had higher endochitinase activity (100–200 times that in controls), in part because of lower control values. T0 plantlets in vitro or young plants in soil had higher absolute and relative endochitinase activity. When detached leaves of T0 plants were inoculated with A. brassicicola, lesion size showed a significant negative correlation with endochitinase levels. After inoculation of two-month old T0 plants with A. brassicicola, all 15 transgenic lines tested showed significantly less severe disease symptoms than controls. In contrast, lesion size on petioles of T0 and T1 plants inoculated with S. sclerotiorum was not statistically different from controls.  相似文献

Ice-nucleating capability of the kiwifruit vine, Actinidia deliciosa, was examined. Pseudomonas viridiflava, present as an epiphyte on the vine, was an effective icenucleating agent. The presence of other sources of ice nuclei on the surface of the vines, leaves and fruit, which could not be inhibited by anti-bacterial agents, was also demonstrated.  相似文献

Herbivorous insects may benefit from avoiding the smell produced by phytopathogens infecting plant host tissue if the infected tissue reduces insect fitness. However, in many cases the same species of phytopathogen can also infect host plant tissues that do not directly affect herbivore fitness. Thus, insects may benefit from differentiating between pathogen odors emanating from food and nonfood tissues. This is based on the hypothesis that unnecessarily staying attentive to pathogen odor from nonfood tissue may incur opportunity costs associated with not responding to other important survival functions. In this study adults of Drosophila suzukii Matsumura, an invasive larval frugivore, showed reduced attraction to the odor of raspberry fruit, a food tissue, when infected with Botrytis cinerea Pers., a ubiquitous phytopathogen, in favor of odors of uninfected raspberry fruit. Moreover, D. suzukii oviposited fewer eggs on infected raspberry fruit relative to uninfected raspberry fruit. Larval survival and adult size after eclosion were significantly reduced when reared on B. cinerea‐infected raspberry relative to uninfected fruit. Interestingly, when the behavioral choice experiment was repeated using Botrytis‐infected vs. ‐uninfected strawberry leaves, a nonfood tissue, in combination with fresh raspberry fruit, odor from B. cinerea‐infected leaves did not reduce D. suzukii attraction to raspberries relative to raspberries with uninfected leaves. These behavioral results illustrate the important role context can play in odor‐mediated interactions between insects, plants and microbes. We discuss implications of our findings for developing a repellent that can be useful for the management of D. suzukii.  相似文献
