Seeds of barley (Hordeum vulgare L.) and mung bean (Vigna radiata(L.) Wilczek), with orthodox seed storage behaviour, were imbibedfor between 8 h and 96 h at 15 °C and 25 °C, respectively,while barley seeds were also maintained in moist aerated storageat 15 °C for 14 d. These seeds and seedlings, together withcontrols, were then dried to various moisture contents between3% and 16% (wet basis) and hermetically stored for six monthsat —20°C, 0°C or 15°C. In both species, neitherdesiccation nor subsequent hermetic storage of the control lotsresulted in loss in viability. The results for barley seedsimbibed for 24 h were similar to the control, but desiccationsensitivity increased progressively with duration of imbibitionbeyond 24 h in barley or 8 h in mung bean; these treatmentsalso reduced the longevity of the surviving seeds in air-drystorage. Loss in viability in barley imbibed for 48 h was mostrapid at the two extreme seed storage moisture contents of 3·6%and 14·3%, and in both these cases was more rapid at15 °C than at cooler temperatures. Similarly, for mung beanimbibed for 8 h, loss in viability was most rapid at the lowest(4·3%) moisture content, but in this case it was morerapid at –20 °C than at warmer temperatures. Ricinodendron rautanenii (manketti) seeds respond to as littleas 10–2 µl I–1 ethylene. Both dry and imbibedseeds respond to ethylene treatments. The length of the imbibitionperiod prior to the ethylene treatment is not important as seedswhich were simultaneously gassed and imbibed germinated successfully.Seeds were sensitive to the gas at temperatures between 25 and35 °C. No temperature effect was observed in seeds whichwere transferred from temperatures below 20 °C, or above35 °C, to 30 °C. Immature Glycine max (L.) Merrill seeds, initially between 50and 450 mg f. wt, were grown and matured successfully in vitro.Excised seeds were floated in a liquid medium containing 5 percent sucrose, minerals and glutamine in flasks incubated at25 °C under 300 to 350 µE m–2 s–1 fluorescentlight. During 16 to 21 d in culture, seeds grew to a matured. wt of 100 to 600 mg per seed at an average rate of 5 to 25mg d. wt per seed d–1 depending on initial size. Growthrates were maximal during the first 8 to 10 d in vitro but declinedwith loss of green colour in the cotyledons. Seed coats rupturedwith rapid cotyledon expansion during the first 2 d in culture.Embryos were tolerant to desiccation and 80 to 90 per cent germinatedif removed from culture before complete loss of green colour.The growth of excised seeds in vitro exceeded the growth ofseeds in detached pods, but when windows were cut in pods topermit direct exposure of seeds to the medium, seed growth wascomparable. Peanut (Arachis hypogaea L. ) seed powder accumulated ATP fromAMP and phosphoenolpyruvate (PEP) at a rate of approx. 100 pmolmin–1mg powder at 35° C. When peanut seed powderwas incubated with various substrates, which may result in PEPor AMP (ADP) synthesis, then ATP accumulated. The best substratecombinations examined so far were AMP + succinate, NADH2, andAMP + malate + NAD, with activities of 33, 12 and 12 pmol minmg–1powder,respectively; AMP + malate showed very low activity. Some combinationsexhibited linear activities with time, while others had an exponential-typeprofile. The temperature dependence of the ATP accumulationdemonstrated by the Ahrrenius plot had a double phase with atransition point at 25° C. The Ea values between 15°C and 25° C were 25 000–50 000 cal/mol, while above25° C the Ea values fluctuated between 6000 and 8000 cal/mol(depending on the substrate). The AMP + PEP combination exhibiteda single-phase profile between 15° C and 40° C, withan Ea value of 22 000 cal/mol. In the presence of some substrates,ethephon (ethylene) had a stimulatory effect and caused an increasein the Ea values at the high temperature phase. A comparisonof seed powder from dormant seeds with that from non-dormantseeds revealed that some substrate combinations accumulate ATPfaster in non-dormant seeds and others do so in dormant seeds. Thermal analyses of freezing events in hydrated lettuce (LactucasativaL.) seeds show a correlation between low temperature exotherms(LTEs) (evidence of ice crystal formation) and seed death. Yet,weather patterns common to the Northern Great Plains of NorthAmerica regularly create conditions where non-dormant seedsof native plants hydrate with snow melt and are subsequentlyexposed to -30 °C or colder conditions. To determine ifsuch weather patterns decimate dispersed seeds, we measuredthe effects of freezing on fully hydrated winterfat (Eurotialanata(Pursh) Moq.) seeds harvested from the Northern Plainsat two USA and one Canadian location. Survival of hydrated seedsto -30 °C at a cooling rate of 2.5 °C h-1was similarto that of seeds not subjected to cooling, even though botha high temperature exotherm (HTE) and an LTE were observed.Although the LTE was not related to winterfat seed survival,freeze-stressed seeds had reduced germination rates and reducedseedling vigour, particularly for the collection with the lightestseeds. The temperature of LTEs was similar among seed collectionswith a mean of -17.6 °C, but was warmer when the seeds wereimbibed at 0 °C compared to 5, 10 or 20 °C. We founda significant correlation between the HTE and LTE temperatures.The difference and the correlation may be due to the highermoisture content of seeds imbibed at 0 °C. After pericarpremoval, only one exotherm in the range of the LTE was observed.This was also true for the naked embryo. Low temperature tolerance was investigated in the imbibed seedof 15 seed lots compnsmg seven cultivars of Lactuca sativa L.During rapid cooling (20 °C h–1) some seeds of allseed lots survived to –16 °C but none to –20°C. The majority of seed lots retained over 50 per centviability above –14 °C due to isolation of the embryofrom external ice by the endosperm, and subsequent embryo super-cooling.Certain seed lots, including all three seed lots of cv. TomThumb, showed high mortality at temperatures above –10°C. Correlation of mortality with the formation of externalice suggested that the endosperm is not an effective nucleationbarrier in these seed lots. Survival to –20 °C was increased at slower coolingrates (6 to 1 °C h–1) due to freeze desiccation ofthe embryo, but seed lots varied considerably in their toleranceof specific cooling rates. A model to explain this variationwas developed incorporatmg (1) seed lot super-cooling limittemperature, (2) the rate at which freeze dehydration of thesupercooled embryo took place, (3) the moisture content at whichnucleation (at –20 °C) was no longer certain and (4)the.initial equilibrium moisture content of the fully imbibedseed. Factors (1), (2) and (3) were found to be relatively constant,but low (or artificially reduced) seed moisture content wasclosely correlated with high survival at natural cooling rates.Seed size fractions of similar moisture content from a singlecultivar showed that more small seeds survive cooling at 3 °Ch–1 to –20 °C than larger seed. Seed conditioning and germination in witchweed (Striga asiatica(L.) Kuntze) were temperature-dependent. With higher conditioningtemperatures, shorter conditioning time was required for germinationwith terminal dl-strigol (strigol) treatment at 30 °C. Maximumgermination (80–100%) was obtained by conditioning inwater at 20, 25, 30 and 35 °C for 14, 7, 5 and 3 d, respectively,and terminally treating with 10–6 M strigol at 30 °C.Seeds conditioned in 10–8 M strigol instead of water germinatedmuch less with the same terminal strigol treatment. Generally,conditioning was slower when seeds were conditioned in strigolrather than water. The reduction in germination rate by pretreatmentin strigol or pretreatment at low temperatures could be overcomeby increasing the terminal strigol concentration in the germinationtest. Conditioned seeds did not germinate at 10 and 15 °Cwith a terminal 10–6 M strigol treatment but yielded closeto maximum germination at 25, 30 and 35 °C with the sameterminal strigol treatment. To obtain maximum germination, boththe minimum conditioning temperature and the minimum germinationtemperature for conditioned seeds were 20 °C. Intact seeds of P. sativum and P. elatius leaked electrolytes,sugars and proteins for as long as 48 h after the beginningof imbibition. Initial leakage was higher at 25 °C thanat 5 °C, but its extent was much greater at the lower temperature.Transfer of seed from 5 °C to 25 °C after 5 h imbibitionresulted in leakage for 48 h at the initial rate at 5 °C.The transfer from 25 °C to 5 °C gave leakage at a rateequivalent to the initial rate at 25 °C. The NADP$-specific isocitrate dehydrogenase was partially purifiedfrom photosynthetically-grown Rhodospirillum rubrum. The pHoptimum is between 7.5 and 9.0 in phosphate buffer. The apparentKm is 3.1x10–5 M for isocitrate, 5.1x10–5 M forNADP$, 1.7x10–5 M for manganese, 1.5x10–4 M formagnesium, and 3.5x10–3 M for inorganic orthophosphate.Arsenate exerts a slight inhibition. The Q10 between 17.5°Cand 40°C is 1.62, and the energy of activation at 25°Cis 9.74 Kcal/mole. Glyoxylate and oxalacetate cause concertedinhibition of the enzyme activity. Various nucleotides inhibitthe activity. The kinetics of inhibition by ATP was found tobe mixed type with respect to NADP$ and isocitrate, the Ki valuesbeing 1.17x10–3 M and 1.10x10–3 M respectively.The inhibition between ATP and orthophosphate is competitivewith a Ki of 10–4M. Intact wild-type tomato (Lycopersicon esculentum cv. Moneymaker)seeds do not complete germination to the same percentage orat the same speed as intact ABA-deficient sitiens (sitw) mutantseeds when seeds of both genotypes are imbibed on polyethyleneglycol (PEG) solutions of –0.3 to –1.5 MPa osmoticpotential. However, if the thicker testas of wild-type seedsare removed (stripped) from the micropyle without damaging theendosperm, both the percentage and speed of germination at lowexternal water potential are similar to that of sitw mutantseeds. Removing the micropylar end of the testa from sitw seedsdid not enhance either the speed or percentage of germinationon PEG solution. Despite similar germination percentage and speed between strippedwild-type seeds and either stripped or intact sitw seeds underosmotic stress, some differences in seed metabolism are evidentbetween genotypes. After removal of the embryo from developing seeds of Pisum sativum,the ‘empty’ ovules (seed coats without enclosedembryo) were filled with a solution (pH 5.5) containing mannitol(usually 400 mM) to which various salts were added. A solutioncontaining two isotopes ((a) [2H]-sucrose/[–14C]aminoisobutyricacid (AIB) or (b) [3H]valine/[14C]asparagine mixture) was administeredto the plant via the petiole subtending the fruiting node, and[2H]solute and [14C]solute unloading from the seed coat wasmeasured, in pulse-labelling experiments of about 5 h. The presenceof 25 or 50 mM K+ in the ‘empty’ ovule enhancedthe release of sucrose from the seed coat particularly duringthe first hours of the experiment, but the stimulating effectof K+ on the release of labelled solutes derived from aminoacids was much smaller. The presence of 25 mM CaCl2 did notaffect the release of sucrose or amino acids from the seed coat.The effect of K+ on sucrose and amino acid release is explainedas an inhibition of sucrose and amino acid resorption from theseed coat apoplast into seed coat cells, after unloading fromthe seed coat unloading sites. It is suggested that amino acidrelease is much less affected by K+ than sucrose release, becausefar less resorption of amino acids by seed coat parenchyma cellstakes place during amino acid transport into the seed coat cavity. Effects of dehydration, storage temperature and humidificationon germination of Salix alba andS. matsudana seeds were studied.Newly released seeds showed 100% germination before and afterdehydration to 11–12% moisture content. Germination ofthe high vigour lot (100% initial normal germination) was notaffected by dehydration to 6.7% moisture content but germinationdecreased with further dehydration to 4.3%. The lower vigourlot (75% initial normal germination) was more susceptible todehydration and germination decreased following dehydrationto 6.7% moisture content. Dry seeds of both species survivedimmersion in liquid nitrogen without loss of viability. Thegermination of seeds stored with 9% moisture content decreasedto 35–40% in 5 months at -20°C or in 2 months at 5°C.However, at 25°C seeds entirely lost viability within 2weeks. Seeds showed improved performance when stored at -70°C> - 20°C > 5°C > 25°C and tolerated dehydrationto a moisture content in equilibrium with 15% relative humidity.Results suggest that they are orthodox in storage behaviouralthough they are short-lived. Humidification treatment of lowvigour seed lots resulted in a remarkable increase in germinationpercentage. A stable freeze-dried powder was prepared of partly purifiedribulose bisphosphate carboxylase from wheat leaves. As withpreparations from other leaves it is necessary to incubate theenzyme with Mg2$ and CO2 to achieve maximum activity. At 25°C this activity was 0.75 IU mg–1 protein for a preparationactivated at 50 °C for 10 min; the Km for CO2 was 15 µM. The time for reactivation of enzyme that had been inactivatedthrough the absence of CO2 and Mg2$ was influenced by the lengthof the inactivating treatment. After a short inactivation periodthe enzyme was reactivated within a few minutes, whereas aftera longer period several hours were needed. Enzyme in the latterstate had some properties in common with enzyme inactivatedby lower temperatures but in the presence of CO2 and Mg2$. Theenzyme kinetic characteristics are similarly affected by bothkinds of inactivation; the maximum velocity is decreased butthe affinity for CO2 is not affected. Reactivation following a long inactivating treatment becomesmore dependent on Mg2$ concentration as the temperature is increasedfrom 0 to 20 °C.  相似文献

Immature Glycine max (L.) Merr. seeds initially at 50–70mg fresh weight were successfully grown and matured in vitroin detached pods. Surface sterilized pods were floated in aliquid medium containing 5 per cent sucrose, minerals, and glutaminein 125 ml Erlenmeyer flasks and incubated at 25 °C under350–400 µE m–1 s–1 white light. Seedswhich were matured in vitro increased tenfold in dry weight,were visually similar to commercial seeds of the same size,were tolerant to desiccation and germinated with normal seedlinggrowth. Excised pods transported dye from the pedicel to thegrowing seed within 120 min. Soya bean pod culture is a usefultechnique to study the influence of single or combinations ofchemical or environmental parameters on regulation of seed growth,seed maturation, and subsequent germination events without theconfounding interactions with the mother plant. Biochemical properties, i.e. endogenous abscisic acid, proline,sugars, respiration, adenosine phosphates and adenylate energycharge, and growth and moisture content were measured duringthe development of seeds of Machilus thunbergii. As dry matteraccumulated in the embryo during development, moisture content,ABA, proline, respiration and sugars all declined. At maturity,the dry mass of the seeds failed to attain a plateau beforethe period of natural seed shedding; the axis and cotyledonsreached moisture contents of 58 and 45%, respectively. Dryingof immature seeds at 73% relative humidity and 25 °C for30 d resulted in a complete loss of viability at all developmentalstages tested with the exception of mature seeds that were ableto tolerate a 5% decrease in moisture content before germinationdeclined. ABA was detected in all embryos tested, with a maximum value16.·16 µg g-1 d. wt about midway through development.Although the presence of ABA induced no tolerance to desiccationof mature seeds, it did coincide with decreased content of waterin the developing seeds and decreased respiration. Desiccationdamage of M. thunbergii seeds occurred when moisture contentwas still high (45%) and this damage was not related to theabsence of oligosaccharides in the mature seeds. We concludethat developing embryos and mature seeds of M. thunbergii haveproperties common to many recalcitrant seeds, with seeds beingsensitive to desiccation at all stages, having a prominent ABApeak, little proline, lacking oligosaccharides, and specifically,little dormancy and a moderate rate of respiration of matureseeds (0·9 µmol O2 min-1 g-1 f. wt). Adenosinetriphosphate content and energy charge decreased from stagefour to stage eight of seed development, then increased againto 103 nmol g-1 d. wt and 0·73, respectively, in matureseeds. The effect of alternating temperatures on the times taken byseeds of lentil (Lens culinaris Medikus) to germinate was investigatedusing a two-way temperature-gradient plate. Between 5 and 25°C,warmer temperatures increased the rate of germination. Variationamong the individual seeds in the times required for germinationat different constant temperatures within this range were describedwell by a log-normal distribution of thermal times, accumulatedabove a base temperature of 1·5°C. Even with amplitudesas great as 20°C, no effect of alternation per se on thethermal time required for germination was detected—whetherthe cool temperature was applied for 8 or 16 h d-1. Similarly,in alternating temperature regimes where the minimum temperatureof the diurnal cycle was between 0°C and the base temperature,the thermal times required for germination (where no thermaltime accrued during the periods when temperature was below Tb)were in close agreement with those values provided by the modeldetermined at warmer constant temperatures. However, where theminimum temperature applied was < 0°C the germinationof all but the earliest germinators was delayed beyond modelpredictions, and more so where the sub-zero minimum temperaturewas applied for 16 rather than 8 h d-1. The results, therefore,contradict the view that alternation in temperature per se reducesthe thermal time required for seed germination. Seeds stored under various conditions showed deteriorative changesin extremely dry (1% r.h. at 10 °C) or humid (93% r.h. at25 °C) conditions after 6 weeks storage, when little orno loss of viability had taken place; no changes were detectedin intermediate conditions (45% r.h. at 10 °C). The lossof electrolytes from seeds into water increased after 3 weeksof humid storage, and subsequently dead areas developed on thecotyledons of seeds held in either humid or dry conditions.With time in storage some of the seeds in dry conditions showeda reduction in the rate of imbibition, and consequently a lowlevel of electrolyte leakage. Other seeds showed an increasein leakage following dry storage. No change in solute content(sugars, potassium, and electrolytes) was detected in seedsstored in humid conditions, suggesting that the increased electrolyteleakage was caused by an impaired ability to retain solutes.Thus increased leakage was recorded in seeds whose cotyledonscontained no dead areas as revealed by vital staining, and wastherefore attributable to changes in living cells, possiblydeterioration in cell membranes. Viability began to declineafter 6 weeks in humid storage at 25 °C and after 2 d in94% r.h. at 45 °C, but was maintained in both dry and intermediateconditions. Exposure of Solanum nigrum L. seeds to high temperatures duringimbibition affected their leakage pattern: (1) The rate of leakageof total electrolytes was markedly increased with elevationof temperature. The increase was highest during the first 3h of imbibition but with a reduced rate thereafter. (2) Leakageof Na+ was almost complete after 6 h of imbibition at both temperatures,but much more Na+ leaked out at 50?C than at 25?C. (3) A markedincrease in leakage of K+ occurred after 24 h of exposure to50?C so that after 96 h three times more K+ leaked out at 50?Cthan at 25?C. (4) After 6 h of imbibition Ca11 and Mg++ continuedto leak out at 25?C and at 50?C at a similar rate. (5) Imbibitionat an elevated temperature induced a marked increase in theleakage of both nucleic acids and proteins. (6) Malate dehydrogenasewas not detected in the leachate at 25?C, but was found after48 h at 50?C. It is assumed that this enzyme was of cytoplasmicorigin, indicating heat damage to membranes. Seed germination in Talinum triangulare as affected by photoperiod,with or without previous incubation in the dark in water at25 or 4 °C, was studied. The time course and quantity ofseed germination in photoperiods of 1 h and above were similarwith or without dark pretreatment, but the time to half maximumgermination was reduced from 12 days in non-dark pretreatedseeds to 4 days in seeds given 20 days in the dark at 25°C.A photoperiod of 0·25 h gave a lower rate and total germinationthan photoperiods of 1 h and above. Un-pretreated seeds required17 cycles of 24 h photoperiod for maximum germination as comparedwith 7 or less cycles if the seeds received more than 10 daysdark pretreatment at 25 °C. Both the rate and total germinationin light increased as the length of dark pretreatment at 25°C was increased from zero to 30 days. Incubation of theseeds in water in the dark at 4 °C for 5 to 30 days priorto illumination at 21 °C, reduced both the rate and quantityof seed germination in light as compared with those similarlyincubated in the dark at 25 °C. However, previous incubationin the dark for 30 days at 4 °C partially substituted forthe light requirement. Two simultaneous measurements, extracellular potential V andK$(86Rb) transport, and the intracellular potential of corticalcell E and potential V, were used to study the effects of externalKCl on two-day-old bean roots. High, external KCl concentrations(>10 mM) markedly enhanced K$ loss from tissues in the elongatingregion to the external solution and induced depolarization ofthe membrane potential difference (PD=V–E). When Phaseolus roots were returned to a solution with a lowerconcentration of K$, the K$ loss and the potential difference,PD, were restored to their previous values. K$ transport fromother parts of the root to the elongating region, however, didnot recover, and the potential, E, increased. These resultsclearly demonstrate that treatment of Phaseolus roots with ahigh external K$ concentration inhibits K$ translocation throughthe stele to the elongating cortical cells and is dependenton depolarization of the intracellular potential.
