A selective agar plate medium (tannin-treated brain heart infusion agar supplemented with colistin-oxolinic acid) was developed to enumerate tannin-protein complex-degrading Streptococcus bovis in the feces of koalas. The role of litter tannins in controlling soil nitrogen (N) cycling may explain the competitive ability of Kalmia relative to black spruce (Picea mariana), although this has not been demonstrated experimentally. Here, the protein-precipitation capacities of purified tannins and leaf extracts from Kalmia and black spruce were compared. The resistance to degradation of tannin-protein precipitates from both species were compared by monitoring carbon (C) and N dynamics in humus amended with protein, purified tannins or protein-tannin precipitates. The purity of the precipitates was verified using solid-state (13)C nuclear magnetic resonance (NMR) spectra. The ability of mycorrhizal fungi associated with both species to grow on media amended with tannin-protein complexes as the principal N source was also compared. The protein precipitation capacity of Kalmia tannins was superior to those of black spruce. Humus amended with protein increased both mineral and microbial N, whereas humus amended with tannin-protein precipitates increased dissolved organic N. Mycorrhizal fungi associated with Kalmia showed better growth than those associated with black spruce when N was provided as tannin-protein precipitates. These data suggest that Kalmia litter increases the amount of soil N sequestered as tannin-protein complexes, which may improve the competitive ability of Kalmia relative to black spruce by favouring N uptake by mycorrhizas associated with the former.  相似文献

Summary The midgut pH of late instar gypsy moth (Lymantria dispar L.) larvae is strongly alkaline, and varies with diet, larval stadium, and time since feeding. Midgut pH rises with time since feeding, and does so more quickly, reaching greater maximum values, on some diets than others. Leaf tissues of 23 tree species resist increases in alkalinity differentially; this trait and differing initial leaf pH may explain the impact of diet on gut pH. Third instar larvae may have gut conditions favorable for tannin-protein binding shortly after ingesting certain foods, but with time midgut alkalinity becomes great enough to dissociate tannin-protein complexes. Older instars rarely exhibit gut pHs low enough to permit tannin activity. Alkaline gut conditions may explain the gypsy moth's ability to feed on many tanniniferous plant species, especially in later instars. Consequences for pathogen effectiveness are discussed.  相似文献

Tannase activity of bacteria capable of degrading tannin-protein complexes was determined by a newly developed visual reading method. The method is based on two phenomena: (i) the ability of tannase to hydrolyze methyl gallate to release free gallic acid and (ii) the green to brown coloration of gallic acid after prolonged exposure to oxygen in an alkaline condition. The method has been successfully used to detect the presence of tannase in the cultures of bacteria capable of degrading tannin-protein complexes.  相似文献

Underwater audiograms of a northern fur seal, a Caspic seal and a dolphin aphalina were measured under conditions of full or partial (the head above the water) submergence of animals using the method of instrumental conditioned reflexes with food reinforcement. The possibility and peculiarities of sound conduction through the body of marine mammals were investigated by isolating the auricle from the medium of sound spreading (under conditions of partial submergence). By the same technique, the hearing thresholds of Caspic seal were measured in the presence of broad- and narrow-band noises with different central frequencies depending on the medium (underwater or in air) the signal and the noise masker were presented and on the sound-conducting ways (under conditions of full or partial submergence of animals). It was found that aerial and underwater sound-conducting canals of the Caspic seal were functionally connected with each other. The level of hearing masking in the Caspic seal is determined by the tracts of signal and noise conduction, by the differences in sensitivity to the signal and masker, and by their spectral structure. Apparently, the tissues of the seal body considerably change the amplitude-frequency characteristics of the sound.  相似文献

An evaluation on the applicability of Mitis Salivarius agar (MS) medium, commonly used for the detection of oral streptococci in human and animals, to dog specimens and the development of a new selective medium for isolating streptococci from the canine oral cavity are described. Oral samples from dogs were cultured on MS medium under anaerobic conditions. The predominant facultative anaerobic bacteria on MS plates were gram-negative rods. Selectivity of streptococci on MS medium was 21.2%. A new selective medium, designated MS-CAN-AE, was developed for the isolation of streptococci from the canine oral cavity. The average growth recovery of laboratory and clinically isolated strains of streptococci on MS-CAN-AE medium was 84.1% of that on MS medium. Gram-positive rods and gram-negative rods and cocci rarely grew on the MS-CAN-AE. The selectivity of MS-CAN-AE was 95.0% for clinical samples. MS-CAN-AE medium will be helpful for investigations of streptococci in the canine oral cavity.  相似文献

Tannins are water-soluble polyphenolic compounds having wide prevalence in plants. Hydrolysable and condensed tannins are the two major classes of tannins. These compounds have a range of effects on various organisms – from toxic effects on animals to growth inhibition of microorganisms. Some microbes are, however, resistant to tannins, and have developed various mechanisms and pathways for tannin degradation in their natural milieu. The microbial degradation of condensed tannins is, however, less than hydrolysable tannins in both aerobic and anaerobic environments. A number of microbes have also been isolated from the gastrointestinal tract of animals, which have the ability to break tannin-protein complexes and degrade tannins, especially hydrolysable tannins. Tannase, a key enzyme in the degradation of hydrolysable tannins, is present in a diverse group of microorganisms, including rumen bacteria. This enzyme is being increasingly used in a number of processes. Presently, there is a need for increased understanding of the biodegradation of condensed tannins, particularly in ruminants.  相似文献

Cultivation conditions affecting feather degradation by Bacillus sp. FK 46 were investigated. The results showed that feather was almost completely degraded under the following conditions: 1% whole chicken feather as a substrate at the initial medium pH of 9 with 5% bacterial inoculum, at a temperature of 37 degrees C and a shaking speed of 250 rev/min. Glucose, methanol, Tween 80 and Triton X-100, however, had no effect on feather degradation. After feather was degraded, its residue and fermented broth would become a protein feed for animals.  相似文献

We have developed a computerized system for quantitative morphometric analysis of the number and position of secretory granules and organelles in pancreatic islet beta cells following tolbutamide treatment. Data from animals injected with tolbutamide for 1, 2, and 3 days were compared to tissues obtained from untreated control animals. Pancreatic islets removed by a collagenase technique were perfused with an appropriate medium to restore a basal state. After fixation and embedding, thick sections of beta cells were viewed by electron microscopy. Morphometric studies of randomly selected or serially cut cells were performed with computer programs for digitization, quantify, rotational, and perspective display. Tolbutamide treatment resulted in graded granule depletion which was maximal at 72 hr relative to control animals. Reduced granule density was associated with significant reduction in total cell area or cytoplasmic area, but was without effect on nuclear size. Since granule depletion improved visualization of subcellular structures, this will enable us to pursue studies of exocytosis under a variety of physiological conditions.  相似文献

Protective effect of the natural dipeptide carnosine on the antioxidant system of rats under conditions of oxidative stress caused by chronic cadmium administration was investigated. Oxidative status of experimental animals were evaluated based on a number of informative parameters of iron-induced chemiluminescence. It was shown that the introduction of cadmium for 7 days reduces the duration of the latent period of chemiluminescence in the brain, liver, and blood plasma suggesting the depletion of endogenous antioxidant defense. Coexposure to carnosine and cadmium led to significant increase in the level of antioxidant protection in plasma, liver, and brain of animals. Carnosine also prevented the increase of lipid hydroperoxides in the brain and prevented the development of lipid peroxidation content in liver and plasma of animals. Mechanism of the protective effect of carnosine under conditions of oxidative stress induced by cadmium administration was shown on human neuroblastoma SH-SY5Y cell culture. Addition of the cadmium to the incubation medium to a final concentration of 5 μM reduced cell viability of a culture, as was determined by MTT assay; simultaneous addition of carnosine (0.25 mM final concentration) with cadmium resulted in increased cell viability during 24 hours of incubation. Thus, carnosine in a final concentration of 1 mM effectively prevented the development of necrotic lesions of neuroblastoma cells, inhibiting the formation of reactive oxygen species as measured by flow cytometry. The results indicate the ability of carnosine to prevent the development of oxidative stress under the toxic action of cadmium.  相似文献

Light conditions during mycelial growth are known to influence fungi in many ways. The effect of visible-light exposure during mycelial growth was investigated on conidial tolerance to UVB irradiation and wet heat of Metarhizium robertsii, an insect-pathogenic fungus. Two nutrient media and two light regimens were compared. Conidia were produced on (A) potato dextrose agar plus yeast extract medium (PDAY) (A1) under dark conditions or (A2) under continuous visible light (provided by two fluorescent lamps with intensity 5.4 W m(-2)). For comparison, the fungus was also produced on (B) minimal medium (MM) under continuous-dark incubation, which is known to produce conidia with increased tolerance to heat and UVB radiation. The UVB tolerances of conidia produced on PDAY under continuous visible light were twofold higher than conidia produced on PDAY medium under dark conditions, and this elevated UVB tolerance was similar to that of conidia produced on MM in the dark. The heat tolerance of conidia produced under continuous light was, however, similar to that of conidia produced on MM or PDAY in the dark. Conidial yield on PDAY medium was equivalent when the fungus was grown either under continuous-dark or under continuous-light conditions.  相似文献

Escherichia coli K-12 Hfr and F- strains were successively implanted in axenic chicks. Conjugation with exchanges of chromosomal genes occurred with high frequencies in the gut of the chicks and could continue as long as fertile strains coexisted in this environment. Almost all of the expected recombinant types were recovered in the feces under these experimental conditions. Furthermore, these recombinants were analogous to those obtained after conjugations in vitro. Recombinants formed in the gut were more numerous when the F- strain was seeded before the Hfr strain. The recombinants showed no apparent selective advantage over the parental strains in the intestinal medium. They were maintained throughout the experimental period and represented more than 10% of the total intestinal flora. The chick gut is usually rapidly colonized by other bacterial types under natural conditions. The possible effects of other components of the bacterial flora on conjugation of E. coli in holoxenic animals will require subsequent work with more complex microbiological conditions.  相似文献

Lactobacilli constitute a significant part of the microbial population in the gastrointestinal tract of humans and animals. Sensitivity of intestinal lactobacilli to 11 chemotherapeutic drugs was tested with the method of serial dilutions in the solid medium MRS under aerobic and anaerobic conditions. Criteria for dividing the organisms into sensitive and resistant ones with respect to the drugs are proposed. Prevalence of lactobacilli polyresistant strains in the intestine contents of healthy persons at the age of 25 to 50 years being in prolonged constant contact with antibiotics was shown. On the whole 77 various combinations of the antibiotic resistance markers were detected in 141 tested strains. The most frequent were isolated simultaneously resistant to 6 or 7 antibiotics. Different levels of the antibiotic resistance in the strains tested under the aerobic and anaerobic conditions were observed.  相似文献

AIMS: A contribution towards the elucidation of the mechanisms of tannins on bacteria growth inhibition, with particular focus on the interaction between tannins and bacterial proteins. METHODS AND RESULTS: The interaction between tannic acid (TA) and Lactobacillus hilgardii, a wine spoilage bacterium, was investigated by a combination of physiologic and proteomic approaches. Growing tests were performed on medium supplemented with TA at concentrations ranging from 100 to 1000 mg l(-1) demonstrating the inhibitory effect of TA on the growth rate. Total proteins extracted from cells unexposed and exposed to TA were then analysed by 2D-electrophoresis and significant quantitative variations with a marked decrease of protein intensity upon TA exposure were observed. Most of the proteins, identified by ESI tandem Mass Spectrometry, were metabolic enzymes of different pathways, located in cytoplasm and membrane. CONCLUSIONS: The effects of TA on cells are deduced by the involvement of metabolic enzymes, and functional proteins on the tannin-protein interaction. These results might be related to the altered functions of the cell metabolism. SIGNIFICANCE AND IMPACT OF THE STUDY: The possible role of tannins in the inhibition of the bacterial survival and growth in a natural environment such as wine. A similar approach could be applied for evaluating the effects of tannins on food borne and pathogenic bacteria.  相似文献

The purpose of the present study was to examine the utilization of fatty acids (FA) and muscle substrates by skeletal muscle in young, middle-aged, and old adult rats under conditions of euglycemia with low insulin levels. Male Fischer 344 x Brown Norway rats aged 5, 15, or 24 mo underwent hindlimb perfusion with a medium of 8 mM glucose, 1 mM palmitate, 25 microU/ml insulin, [1-(14)C]palmitate, and [3-(3)H]glucose. Glucose and palmitate uptake were similar among age groups. The percent and total palmitate oxidized (nmol.min(-1).g(-1)) were 30-36 and 41-49% lower (P < 0.05) in 15-mo- and 24-mo-old than in 5-mo-old animals. Compared with 5-mo- and 15-mo-old animals, pre- and postperfusion muscle triglyceride (TG) levels were significantly (P < 0.05) elevated 91-305% in red and 118-219% in white muscles of 24-mo-old animals. Fatty acid-binding protein content was 40-64% higher (P < 0.05) in 24-mo- than in 5-mo- or 15-mo-old animals. In red muscle, hormone-sensitive lipase (HSL) content was 28% lower (P < 0.05) in 24-mo- than in 5-mo-old animals. These results indicate that, under euglycemic conditions in the presence of low insulin levels, the reduction in FA disposal to oxidation and the decrease in HSL content may contribute to the accumulation of TG in muscle of old animals.  相似文献

Velocities of bovine spermatozoa in a medium containing glucose were similar under true anaerobic and aerobic conditions. Spermatozoa were not able to sustain motility under anaerobic conditions when glycolysis was inhibited, but regained motility when re-aerated. This demonstrates that immobilisation was due to lack of oxygen and that conditions under which motility was analysed were truly anaerobic. Sperm motility parameters were not significantly different in the presence and absence of 4 microM antimycin A and 4 microM rotenone when glucose was present in the medium. After each incubation, functionality of sperm mitochondria was assayed by washing sperm into the medium which supported respiration but not glycolysis, and motility was visually assessed. All sperm samples were highly motile in this medium indicating that their mitochondria were functional. When glycolysis was inhibited, antimycin and rotenone abolished sperm motility immediately after addition. Bovine sperm can maintain similar levels of motility aerobically and anaerobically if a glycolysable substrate is available. Available data on bovine sperm energetics support this view.  相似文献

The effects of long-term starvation on the body composition of the isopod Porcellio scaber (Latreille) and the collembolan Orchesella cincta (L.) were studied, by determining the body composition in starved and fed animals. A period under summer conditions (19 degrees C, 75% RH and L/D 16/8 photoperiod), was followed by a period under winter conditions (5 degrees C, 75% RH and LD 6/18 photoperiod). O. cincta was held under summer conditions for 3weeks, during which its protein and lipid content decreased, while its water content increased. In P. scaber, the same occurred during the 6weeks they were kept under summer conditions. During subsequent weeks under winter conditions, changes in cold tolerance of the animals were investigated. Cold tolerance and haemolymph osmolality were measured once a week. Starved animals had lower cold tolerance than fed ones. For P. scaber a decreased haemolymph osmolality was found in starved animals compared to fed ones. This is assumed to be caused by a combination of the consumption of carbohydrates out of the haemolymph and of protein reserves and the accumulation of body water. O. cincta appeared to be capable of osmoregulation, as haemolymph osmolality did not differ between starved and fed animals, despite differences in body water content. Decreased cold tolerance in starved animals of both species may be caused by increased water content or, more probably, by the decrease in reserves needed to produce cryoprotective substances.  相似文献

Digestibility trials using common ringtail possums ( Pseudocheirus peregrinus ), small folivorous marsupials, were used to determine effects of tannins on a herbivore which has a specialized hind gut and which normally consumes tannin-rich eucalypt leaves. In one group, untreated leaves were fed to possums, and in a second group polyethylene glycol (PEG) was added to the eucalypt leaves to inactivate tannins. Animals maintained weight, and intakes were not different between the two dietary groups. Digestibility was determined chemically. Digesta were examined histologically. The presence of bacterial rafts in the stomachs of possums fed either diet indicated that PEG did not alter the coprophagic behaviour of possums. Cell wall digestion did not appear to be inhibited by tannins. Tanning of cell contents was predominant, rather than tanning of cell walls. PEG increased faecal excretion of tannins, suggesting that some tannins from normal leaves were digested during gut passage. Dry matter digestibility was higher when animals were fed normal tannin-rich leaves than when they were fed leaves in which tannins were inactivated. The difference could be explained in part by the high digestibility of tannins. The presence of tannin did not reduce nitrogen digestion. We suggest that dissociation of tannin-protein complexes may take place in the specialized caecum. We propose that some other folivorous marsupials may have a similar capacity to overcome tannins. This capacity may allow them to consume a tannin-rich diet.  相似文献
