Due to the unpredictable climate change, drought stress is being considered as one of the major threats to crop production. Wheat (Triticum aestivum L. cv. BARI Gom-26) being a dry season crop frequently faces scarcity of water and results in a lower yield. Therefore, this experiment aims to explore the role of different organic amendments (OAs) in mitigating drought stress-induced damage. The pot experiment consisted of different organic amendments viz. compost, vermicompost and poultry manure @0.09 kg m−2 soil, biochar @2.5% w/w soil and chitosan @1% w/w soil which was imposed on the plants grown under both well-watered and drought conditions. Results showed that drought stress reduced plant height (15%), SPAD value (16%), relative water content (13%), number of spikelet spike−1 (17%), number of grains spike−1 (12%), and 100-grain weight (18%). Organic amendments act as a protectant and reduce drought stress-induced damages by enhancing the morpho-physiological and yield attributes. Vermicompost enhanced SPAD value by 18%, number of spikelets spike−1 by 20%, number of grains spike−1 by 17%, whereas poultry manure increased plant height by 16% under drought condition compared to control plant. Unlike other OAs applied, vermicompost was proved to be capable of reducing the higher lipid peroxidation and proline content raised by drought condition. Drought stress-induced increment of catalase, ascorbate peroxidase and glutathione reductase activities were also efficiently modulated by the organic amendment application. The present study concluded that OAs play significant roles in alleviating drought stressinduced damages by improving the morpho-physiological attributes and among the different types of OAs used vermicompost performed better which in addition ceased the production of reactive oxygen species.  相似文献

The effects of intercycle cover crops on Rotylenchulus reniformis population densities in pineapple were evaluated in one greenhouse and two field experiments. In the greenhouse, Crotalaria juncea, Brassica napus, and Tagetes erecta were planted for 3 months and then incorporated. These treatments were compared to weedy fallow with or without 1,3-dichloropropene (1,3-D) in three soils (Makawao fallow, Wahiawa fallow, and Wahiawa pineapple) naturally infested with R. reniformis. All cover crop incorporation suppressed R. reniformis numbers in cowpea more than did the weedy treatment in the Makawao (P < 0.05) but not in the Wahiawa soils. Crotalaria juncea treatment increased bacterivorous nematodes and nematode-trapping fungal population densities more than the other treatments in Makawao fallow and Wahiawa pineapple-planted soils. The field trials included the same plants as well as Sinapis alba. Treatments with Crotalaria juncea and 1,3-D maintained lower R. reniformis population densities on pineapple longer than other cover crops or weedy fallow treatments. Crotalaria juncea could have suppressed R. reniformis because it is a poor host and because it enhances nematode-trapping fungi when incorporated into soil. Treatment with 1,3-D reduced microbial activities but produced the greatest pineapple yield.  相似文献

Two greenhouse experiments were conducted to examine the effect of Crotalaria juncea amendment on Meloidogyne incognita population levels and growth of yellow squash (Cucurbita pepo). In the first experiment, four soils with a long history of receiving yard waste compost (YWC+), no-yard-waste compost (YWC-), conventional tillage, or no-tillage treatments were used; in the second experiment, only one recently cultivated soil was used. Half of the amount of each soil received air-dried residues of C. juncea as amendment before planting squash, whereas the other half did not. Crotalaria juncea amendment increased squash shoot and root weights in all soils tested, except in YWC+ soil where the organic matter content was high without the amendment. The amendment suppressed the numbers of M. incognita if the inoculum level was low, and when the soil contained relatively abundant nematode-antagonistic fungi. Microwaved soil resulted in greater numbers of M. incognita and free-living nematodes than frozen or untreated soil, indicating nematode-antagonistic microorganisms played a role in nematode suppression. The effects of C. juncea amendment on nutrient cycling were complex. Amendment with C. juncea increased the abundance of free-living nematodes and Harposporium anguillulae, a fungus antagonistic to them in the second experiment but not in the first experiment. Soil histories, especially long-term yard waste compost treatments that increased soil organic matter, can affect the performance of C. juncea amendment.  相似文献

The objective of this work was to evaluate the effect of Beauveria bassiana (Bb 1205) on controlling Fusarium oxysporum f. sp. lycopersici (Fol 17108) in tomato plants in greenhouse conditions. Inoculation of Bb 1205 was the most promising among the agronomic variables and expression of the activity of the enzymes β-1,3-glucanases and chitinases. Inoculation of Bb 1205 occurred at a concentration of 1 × 108 conidia·mL−1, which was administered onto the leaves, directly into the soil and via injection. Infection with Fol 17108 occurred with 1 × 106 spores·mL−1, which were added directly to the soil. Spectrophotometry was used for measuring agronomic parameters, namely activity of chitinases and β-1,3-glucanases in foliage and roots. When Bb 1205 was added to the soil, the chlorophyll index and aerial part length showed significant differences. In addition, it was determined that root length, fresh weight of foliage, flower, and fruit count increased 82 days after inoculation (dai). Chitinase activity induced by Bb 1205 in leaves and roots of tomato plants infected with Fol 17108 was observed when injected into the stem at 32 dai (41.8 and 11.6-fold, respectively). Inoculation on the foliage showed a 10-fold increase of β-1,3-glucanases in the roots after 82 dpi. As for leaves, a 3.8-fold increase was found when the stem was inoculated. In the different in vivo applications, Bb 1205 activated its defenses by expressing the chitinase enzymes and β-1,3-glucanase, thus reducing the damage caused by Fol 17108, demonstrating increase plant growth thereafter.  相似文献

Chinese Torreya grandis (Torreya grandis cv. Merrillii) is a unique economic tree species in China. Intensive management related to application of chemical fertilizer and herbicides caused serious soil quality degradation of Chinese Torreya grandis plantations. Totally, 120 soil samples were collected from the main disbtributed areas of Chinese Torreya grandis in Southeastern China. In this area, soil pH values varied from 3.68 to 6.78, with a median value of 4.91, implying a trend of acidification. The average concentrations of organic matter, available nitrogen, available phosphorus and available potassium were 27.52 g kg−1, 135.77 mg kg−1, 15.12 mg kg−1, and 153.43 mg kg−1, respectively. The results of spatial analysis revealed that target variables had clear spatial patterns. The soil fertility was relevantly high in most of the study area, with soil nutrient imbalances existing. The stand age had a large effect on soil properties, which were also mainly influenced by human activities. Therefore, it is necessary to change the fertilization method for a sustainable management of Torreya grandis plantation.  相似文献

Water insufficiency is the hampering feature of crop sustainability, especially in arid and semi-arid regions. So, the effectual usage of all water resources especially underground brackish water represents the core priority in Saudi Arabia. The present study aimed to recognize the influence of different types of water irrigation (tap water as a control, salinized well water, and magnetized salinized well water) with or without soil amendments (soil without any amendment as a control, peat-moss, ferrous sulfate, and peat-moss plus ferrous sulfate) on petunia plant growth and flowering as well as ion content. Irrigating Petunia plants with saline well water adversely affected growth and flowering as compared to tap water and magnetized saline well water. Additionally, plants irrigated with magnetized water showed a significant enhancement in all the studied vegetative and flowering growth parameters as compared to those irrigated with salinized well water. Furthermore, mineral contents and survival of Petunia plants irrigated with magnetized well water were higher than those irrigated with tap water. Irrigation with magnetized well water significantly reduced levels of Na+ and Cl ions in leaves of Petunia plants indicating the role of magnetization in alleviating harmful effects of salinity. In conclusion, we recommend the utilization of magnetized saline well water for irrigating Petunia plants either alone or in combination with soil amendments (peat moss plus ferrous sulfate).  相似文献

The influence of Chloris gayana, Crotalaria juncea, Digitaria decumbens, Tagetes patula, and a chitin-based soil amendment on Hawaiian populations of Rotylenchulus reniformis was examined. Chloris gayana was a nonhost for R. reniformis. The nematode did not penetrate the roots, and in greenhouse and field experiments, C. gayana reduced reniform nematode numbers at least as well as fallow. Tagetes patula was a poor host for reniform nematode and reduced reniform nematode numbers in soil better than did fallow. Crotalaria juncea was a poor host for R. reniformis, and only a small fraction of the nematode population penetrated the roots. Crotalaria juncea and D. decumbens reduced reniform nematode populations at least as well as fallow. A chitin-based soil amendment, applied at 2.24 t/ha to fallow soil, did not affect the population decline of reniform nematode.  相似文献

Aconitum vilmorinianum, a well-known traditional Chinese herb, is recently being threatened by overexploitation and environment disturbance. This study was conducted to provide propagation methods through in vitro germination and explant cultivation. Germination was stimulated up to 66.00% on Murashige and Skoog (MS) medium containing 2.0 mg L−1 6-benzylaminopurine (BAP), 0.1 mg L−1 1-napthaleneacetic acid (NAA), and 30 g L−1 sucrose. Three bacteria (Pantoea agglomerans, Erwinia persicina, and Pseudomonas tolaasii) would be responsible for consistent contamination during germination. The latter two were effectively eradicated after disinfected. The influence of explant types and hormone combinations on direct and indirect organogenesis was evaluated in the present work. The frequency of shoot induction from axillary bud explants was 100% on the MS fortified with 2.0 mg L−1 BAP and 0.3 mg L−1 NAA. Shoots multiplication was optimized on MS medium supplemented with 0.1 mg L−1 thidiazuron (TDZ) and 0.1 mg L−1 NAA. High callus induction percentage (96.67%) was obtained from stem segments on MS medium with 2.0 mg L−1 2,4-D, then successfully regenerated into shoots on MS medium in the presence of 0.1 mg L−1 TDZ and 0.2 mg L−1 NAA. The present work could be useful for the utilization and conservation of this valuable species.  相似文献

Sugarcane is one of the major important sugar yielding crops in Bangladesh. As an exhaustive crop, sugarcane removes a huge amount of plant nutrients from the soil. However, the combined use of organic and inorganic fertilizers can be a good approach to deal with nutrient depletion and promote sustainable crop production as well as improve soil health. Therefore, an attempt was made to identify the most fruitful and profitable integrated nutrient management on the aspects of growth, yield and quality of sugarcane in two consecutive growing seasons. Seven treatments: T1 =Control, T2 =165:55:120:30:10:2.5:4 kg N:P:K:S:Mg:Zn:B ha−1, T3 =Poultry Litter (PL) at 5 t ha−1 +95:51:87:9:10:2.5:4 kg N:P:K:S:Mg:Zn:B ha−1, T4 =Cow Dung (CD) at 15 t ha−1 + 36:52:60:17:10:2.5:4 kg N:P:K:S:Mg:Zn:B ha−1, T5 =Press Mud (PM) at 15 t ha−1 +10:50:43:0:10:2.5:4 kg N:P:K:S:Mg:Zn:B ha−1, T6 =Mustard Oil Cake (MOC) at 0.5 t ha−1 +140:54:115:25:10:2.5:4 kg N:P:K:S:Mg:Zn:B ha−1 and T7 =GM (Green Manure) at 5 t ha−1 +140:53:100:28:10:2.5:4 kg N:P:K:S:Mg:Zn:B ha−1 were used in this experiment. Two years data showed that treatment T3 produced the maximum amount of tillers, total dry matter yield, millable sugarcane, cane yield and sugar yield, followed by the T4 treatment. The highest stalk heights were recorded in the T3 treatment, which was statistically similar to all other treatments except T1 and T2. The juice quality parameters viz., brix and pol in cane were found significant in treatment T3 while the highest purity was obtained in the T7 treatment. All the data of Jaggery (goor) quality parameters, the highest sucrose content, color transmittance, Jaggery (goor) recovery and the lowest ash content of Jaggery (goor) were observed in the T3 treatment, which was statistically similar to the T4 treatment in both seasons. The highest cost of production was obtained from the T6 treatment while the highest gross return, net return and BCR were recorded in the T3 treatment. No significant changes were found in one cycle of sugarcane in initial and post-harvest soil characteristics viz., pH, organic carbon, total N, and available P, K and S contents due to integrated use of different fertilizer packages. From the experimental findings, it was concluded that treatment T3 followed by T4 treatment would be the better productive and profitable integrated nutrient management technology for ensuring higher yields and quality of sugarcane without soil fertility degradation in the High Ganges River Floodplain soils.  相似文献

Effect of sunn hemp (Crotalaria juncea) hay amendment on nematode community structure in the soil surrounding roots of yellow squash (Cucurbita pepo) infected with root-knot nematodes was examined in two greenhouse experiments. Soils were from field plots treated long-term (LT) with yard-waste compost or no yard-waste compost in LT experiment, and from a short-term (ST) agricultural site in ST experiment. Soils collected were either amended or not amended with C. juncea hay. Nematode communities were examined 2 months after squash was inoculated with Meloidogyne incognita. Amendment increased (P < 0.05) omnivorous nematodes in both experiments but increased only bacterivorous nematodes in ST experiment (P < 0.05), where the soil had relatively low organic matter (<2%). This effect of C. juncea amendment did not occur in LT experiment, in which bacterivores were already abundant. Fungivorous nematodes were not increased by C. juncea amendment in either experiment, but predatory nematodes were increased when present. Although most nematode faunal indices, including enrichment index, structure index, and channel index, were not affected by C. juncea amendment, structure index values were affected by previous soil organic matter content. Results illustrate the importance of considering soil history (organic matter, nutrient level, free-living nematode number) in anticipating changes following amendment with C. juncea hay.  相似文献

Chickpea yield is decreasing day by day due to drought stress, which could be an immense risk for future food security in developing countries. Management practices could be the most excellent approach to diminish loss due to this abiotic factor. The current research work was designed to explore the tolerance reaction of chickpea genotypes against management practices, through morphological and biochemical parameters and evaluate yield performance across drought prone location of Bangladesh. Four genotypes BD-6048, BD-6045, BD-6090, BD-6092 and eight management practices, e.g., severe water stress (SWS), i.e., without irrigation, 10 cm thick mulching with rice straw (MRS), 10 cm thick mulching with water hyacinth (MWH), organic amendment through compost (OAC) @ 3 t ha−1, organic amendment through cow dung @ 5 t ha−1 (OACD), organic amendment through poultry manure @ 2 t ha−1 (OAPM), inorganic amendment through proline application (IAPA) as foliar spray and 16 h hydro-priming (HP). The study revealed that the genotypes BD-6048 showed excellent performance because of the highest chlorophyll, carotenoids, phosphorus, potassium, proline and protein content. The highest pod number plant−1 also increased seed yield in BD-6048. Considering management practices, IAPA increased relative water content, carotenoids, leaf phosphorus and potassium compared to other management practices and severe water stress. Finally, BD-6084 was selected as best genotype because of a significant increase in chlorophyll a and b, carotenoids, and relative water content with IAPA. Identified top performing genotypes can be used for releasing variety and cultivated for sustainable production in drought prone area of Bangladesh.  相似文献

The effect of water deficit was determined on both in vitro and soil seedling as well as in cells in suspension of Agave americana L. In order to do the establishment of cells, the formation of callus was induced; for it two auxins were evaluated: 2,4-dichlorophenoxyacetic acid (2,4-D) and 4-mino-3,5,6-trichloropicolinic acid (picloram) at three concentrations (0.25, 0.5 and 0.75 mg L−1) in three explants (leaf, root and meristems) cultured in MS semisolid medium. The callogenesis response was related to the type and section of the explant, as well as the regulator used, and a cell suspension was established using 0.5 mg L−1 naphthaleneacetic acid (NAA) + 0.5 mg L−1 Benzylaminopurine (BAP). Seedlings were exposed to polyethyleneglycol (15% and 30% w/v) with a water potential of −0.87 and −2.67 MPa, respectively, under soil conditions. Water stress was applied through restricted irrigation. Fresh weight, root system growth, and chlorophyll concentration were some of the parameters that were affected by the effect of water deficit on A. americana L. Chlorophyll concentration values were significantly decreased by 15 at 30% PEG (19.6 SPAD units) compared to the control treatment. In in vitro plants, the highest concentration of proline was found in the roots, being the treatment with 30% polyethylene glycol where the highest concentration of this osmoregulator was obtained (62.5 mg g−1 DW). Under restricted irrigation conditions, an increase in proline concentration was observed both in the aerial part (2.2 µg 100 g−1 DW) and in the root system (1.8 µg 100 g−1 DW). However, the concentrations found were approximately ten times greater, less than those found under in vitro conditions. Therefore, the accumulation of proline can be considered an indicator of stress in Agave Americana L. growth in vitro.  相似文献

Miscanthus, is a promising bioenergy crop, considered superior to other bioenergy crops because of its higher water and nutrient use efficiency, cold tolerance, and higher production of biomass. Broadleaf weeds and grass weeds, cause major problems in the Miscanthus field. A field experiment was conducted in 2018 and 2019, to assess the effects of pre-emergence (alachlor and napropamide) and post-emergence herbicides (nicosulfuron, dicamba, bentazon, and glufosinate ammonium) on broadleaf and grass weeds in M. sinensis and M. sacchariflorus fields. The weed control efficiency and phytotoxicity of pre- and post-emergence herbicides were evaluated at 30 days after treatment (DAT) and compared to those of the control plots. The results showed wide variations in the susceptibility of the weed species to the treated herbicides. Treatment with nicosulfuron 40 g.a.i.ha−1 provided the most effective overall weed control (with 10% visual injury), without affecting the height and biomass of neither Miscanthus species in the field. Post-emergence herbicides such as glufosinate ammonium 400 g.a.i.ha−1 and dicamba 482 g.a.i.ha−1 were effective and inhibited the growth and density of the majority of weeds to a 100%; however, they showed significant phytotoxicity (toxicity scale of 1–10) to both species of Miscanthus. The application of glufosinate ammonium caused severe injuries to the foliar region (90% visual injury) of both Miscanthus sps. Comparatively, M. sinensis showed a slightly higher tolerance to the herbicides nicosulfuron, bentazon and napropamide with 10% visual injury at the recommended dose than M. sacchariflorus. The present study clearly showed that infestation of broadleaf and grass weeds in Miscanthus fields can cause significant damage to the growth and biomass of Miscanthus and applying pre-emergence and post-emergence herbicides effectively controls the high infestation of these weeds.  相似文献

An efficient regeneration system of Rhynchostylis retusa was established to provide technical reference for the application of tissue culture tube seedlings in production. The mixtures of callus and protocorm from aseptic germination were used as explants. The optimal media of each stage was selected for callus proliferation, protocorm occurrence and growth, rejuvenation and rooting via a single, complete combination and orthogonal experiment. The results showed that the optimal medium for callus proliferation, protocorms occurrence and growth was 1/2 Murashige and Skoog (MS) medium adding 50 g·L−1 banana puree, 0.1 mg·L−1 α-naphthaleneacetic acid (NAA), 1.5 mg·L−1 6-benzylaminopurine (6-BA) and 1.0 mg·L−1 kinetin (KT) with 17.33 proliferation coefficient of callus and 19.63 occurrence coefficient of buds after 90 days. Then the buds occurred from protocorm were cultured on 1/2 MS medium including 100 g·L−1 banana puree, 1.0 mg·L−1 NAA, 2.0 mg·L−1 6-BA and 0.05 mg·L−1 KT, in which the proliferation coefficient of callus was 10.32 and occurrence coefficient of buds reached 17.87. In the further subculture, the same medium was simultaneously used for callus proliferation, protocorm occurrence and bud growth. The plantlets developed roots in 1/2 MS medium containing 70 mL·L−1 coconut water and 1.5 mg·L−1 NAA with 100% rooting rates after 90 days. The survival rate was more than 90% after domestication and transplantation. This regeneration protocol will provide technique foundation for protecting wild resource and developing artificial cultivation.  相似文献

Effect of cover crops intercropped with pineapple (Ananas comosus) on Rotylenchulus reniformis population densities and activity of nematode-trapping fungi (NTF) were evaluated in two cycles of cover crop and pineapple. Sunn hemp (Crotalaria juncea), rapeseed (Brassica napus), African marigold (Tagetes erecta), or weeds were intercropped with pineapples. Beds planted with sunn hemp or rapeseed had lower population densities of R. reniformis than African marigold, weeds, or pineapple plots during cover crop growth, and the subsequent pineapple-growing periods. Rapeseed was a good host to Meloidogyne javanica and resulted in high population densities of M. javanica in the subsequent pineapple crop. Fireweed (Erigeron canadensis) occurred commonly and was a good host to R. reniformis. Bacterivorous nematode population densities increased (P ≤ 0.05) most in sunn hemp, especially early after planting. Nematode-trapping fungi required a long period to develop measurable population densities. Population densities of NTF were higher in cover crops than weeds or pineapples during the first crop cycle (P < 0.05). Although pineapple produced heavier fruits following sunn hemp than in the other treatments (P < 0.05), commercial yields were not different among rapeseed, weed, and sunn hemp treatments.  相似文献

Hydrangea bretschneideri Dipp is a highly popular ornamental plant for garden decoration. Genetic engineering technology has been successfully used in many plant species, but it is limited in Hydrangea. Here we established an efficient regeneration system by using stem segments as explants for the first time. In our study, the plant growth regulators (PGRs) were evaluated at the different regeneration processes, including axillary shoots regeneration and root induction. We found that the optimal concentration for axillary buds’ induction was 2.0 mgL−1 6-BA and 0.5 mgL−1 1 IAA, its highest induction rate was 70%. Moreover, the highest axillary shoots proliferation coefficient was 10.7 on the Murashige and Skoog (MS) medium with 2.0 mgL−1 6-benzyladenine (BA), 0.2 mgL−1 indole-3-butyric acid (IBA), and 1.0 mgL−1 gibberellin A3 (GA3). The highest frequency of root induction was 80.0 ± 0.06% by culturing the elongated shoots in 1/2 MS medium containing 0.1 mgL−1 IBA. In summary, our study will provide an effective technology for large-scale propagation and important pathway for promoting the popularization and application of Hydrangea bretschneideri Dipp.  相似文献

The present study aimed to develop a protocol for somatic embryogenesis and encapsulation of coffee embryos (Coffea arabica L.), for the conservation of genotypes with characteristics of commercial interest. Somatic embryos were induced from leaf explants in Murashige and Skoog medium (MS) supplemented with 1 mg · L−1 of 2,4-dichlorophenoxiacetic acid (2,4-D) combined with 2 mg · L−1 of benzyladenine (BA). Somatic embryos (SE) at the globular stage were encapsulated in a sodium alginate matrix; two treatments were tested: MS + 5 mg · L−1 BA + 1 mg · L−1 NAA + 3% (w/v) alginate, and MS + 7 mg · L−1 BA + 5.7 mg · L−1 indoleacetic acid (IAA) + 3% (w/v) alginate. Alginate was complexed with 100 mM calcium chloride (CaCl2). Viability of the encapsulated SE was determined by staining with 0.01% fluorescein diacetate (FDA) after 0, 15, 30, and 45 days of storage at 4°C. Embryo viability was 100% in both treatments.  相似文献

Sustainable crop production is dependent on improvement of soil health using different management strategies. A study was conducted in the 2008/09 cropping season to investigate soil macrofauna order diversity and abundance under organic matter transfer system and improved fallows in a high rainfall (>800 mm year?1) area of Zimbabwe. Macrofauna were sampled using monoliths to a depth of 25 cm and Shannon‐Wiener diversity was used for diversity testing. Order diversity varied significantly with treatment under both systems. Under improved fallows, Leucaena trichandra had the highest fauna orders (6) followed by Calliandra colorthysus (5), then Acacia anguistissima (4) while Leucaena pallida and miombo forest had the least orders. Crotalaria juncea had the highest faunal orders (5), among organic material transfer system while other treatments had ≥3 orders. Unfertilized maize had one order. Calliandra colorthysus had the highest order diversity followed by maize stover > cattle manure > Cr. juncea > fully fertilized maize > unfertilized maize. Major groups identified were termites, earthworms, beetles, millipedes and ants. All taxa combined, cattle manure and miombo forest had the highest macrofauna abundance. This study showed that application of organic nutrient resources and use of improved fallows significantly influenced soil macrofauna order diversity and abundance.  相似文献

Immature embryos of inbred maize (Zea mays) lines (H8183, H8184, and H8185) were used for Agrobacterium infection. We used the β-glucuronidase gene (GUS) as the target gene and the glufosinate resistance gene (bar) as the selection marker. We conducted research on several aspects, such as different genotypes, coculture conditions, screening agent concentrations, and concentrations of indole-3-butytric acid (IBA), 6-benzylaminopurine (6-BA), and ascorbic acid (Vc) in the differentiation medium. We optimized the genetic transformation system, and the obtained results indicated that among the three lines studied, the induction rate of H8185 was the highest at 93.2%, followed by H8184, with H8183 having the lowest induction rate (80.1%). The best coculture method was that using the N6 coculture medium layered with a sterile filter paper. Using orthogonal analysis, we found that the optimal combination of the three factors in the differentiation medium was A3 (1 mg mL−1 IBA), B3C1 (1.6 mg mL−1 6-BA), and D3 (1.5 mg mL−1 Vc). Through GUS staining analysis, Bar test-strip analysis, and polymerase chain reaction, five transgenic plants were finally obtained. This study established the optimal conditions for genetic transformation in maize.  相似文献

Remediation of soil acidity is crucial for increasing crop production and improving environmental quality of acid infertile soils. Soil incubation and greenhouse pot experiments were carried out to examine the interactions between phosphate rock (PR), coal combustion by-product (BP), dolomitic lime (L), and cellulose (C) in an acidic soil and their effects on ryegrass (Lolium perenne L. cv Linn) growth. BP and PR application increased plant P content and dry matter yield (DMY) of shoots and roots by improving soil Ca availability and reducing Al toxicity. Application of BP at low rates (5 to 10 g BP kg-1) with PR appeared to decrease both plant P content and DMY compared to PR application alone. The reduced DMY is due to an increased Al concentration in soil solution as a result of displacement of sorbed Al by Ca of BP. Increases in DMY were obtained by addition of lime along with PR and BP at low rates or by increasing BP application rates above 15 g kg-1. This improved plant response was likely related to alleviation of Al toxicity by CaCO3 contained in the BP. In addition to raising the pH to an acceptable level for plant growth, the dolomitic lime supplied needed Mg for plants, thereby maintaining a good balance between available Ca and Mg for plants in the BP- and PR-amended soils. The addition of cellulose to the BP- and PR-amended soils reduced water-soluble Al and increased DMY. Plant growth increased PR dissolution by 2.4 to 243% in a soil with low available P. Use of BP at moderate rates with PR and dolomitic lime appears to be the best combination in increasing crop yields on infertile acidic soils.  相似文献
