黄连(Coptis chinensis)是毛茛科著名药材,该文研究了黄连体内黄连素在组织器官中的分布规律和根尖屏障结构特征。在白光和荧光显微镜下,组织器官中黄连素在蓝色激发光下自发黄色荧光,黄连素-苯胺兰对染研究细胞壁凯氏带和木质化,苏丹7B染色栓质层,间苯三酚-盐酸染色木质化。结果表明:黄连不定根初生结构为维管柱、内皮层、皮层、外皮层和表皮组成;次生结构以次生木质部为主、次生韧皮部和木栓层组成。黄连根茎初生结构由角质层,皮层和维管柱组成;次生结构由木栓层、皮层和维管柱组成,以皮层和维管柱为主。叶柄结构为髓、含维管束的厚壁组织层、皮层和角质层。黄连不定根的屏障结构初生结构时期由栓质化和木质化的内皮层、外皮层;次生结构时期为木栓层组成;根状茎的为角质层和木栓层。黄连素主要沉积分布在不定根和茎的木质部,叶柄的厚壁组织层,木质部和厚壁组织是鉴别黄连品质的重要部位。黄连根尖外皮层及早发育,同时初生木质部有黄连素沉积结合,可能造成水和矿质吸收和运输的阻碍,也是黄连适应阴生环境的重要原因。  相似文献

Background and Aims

Casparian bands are characteristic of the endodermis and exodermis of roots, but also occur infrequently in other plant organs, for example stems and leaves. To date, these structures have not been detected in phellem cells of a periderm. The aim of this study was to determine whether Casparian bands occur in phellem cells using tests that are known to detect Casparian bands in cells that also contain suberin lamellae. Both natural periderm and wound-induced structures were examined in shoots and roots.

Methods

Using Pelargonium hortorum as a candidate species, the following tests were conducted: (1) staining with berberine and counterstaining with aniline blue, (2) mounting sections in concentrated sulphuric acid and (3) investigating the permeability of the walls with berberine as an apoplastic, fluorescent tracer.

Key Results

(1) Berberine–aniline blue staining revealed a modification in the radial and transverse walls of mature phellem cells in both stems and roots. Three days after wounding through to the cortex of stems, the boundary zone cells (pre-existing, living cells nearest the wound) had developed vividly stained primary walls. By 17 d, staining of mature phellem cells of wound-induced periderm was similar to that of natural periderm. (2) Mature native phellem cells of stems resisted acid digestion. (3) Berberine was excluded from the anticlinal (radial and transverse) walls of mature phellem cells in stems and roots, and from the wound-induced boundary zone.

Conclusions

Casparian bands are present in mature phellem cells in both stems and roots of P. hortorum. It is proposed that Casparian bands act to retard water loss and pathogen entry through the primary cell walls of the phellem cells, thus contributing to the main functions of the periderm.  相似文献

Certain anatomical characters of Jaumea carnosa, a salt marsh plant, which are diagnostic, adaptive, or otherwise important for an ecological anatomy study are described. The characters which follow are noteworthy. The plant has a sympodial construction forming an extensively branched rhizome system, and erect lateral shoots which eventually become prostrate. The leaves have very few, and only moderately developed succulent characters, such as: poorly defined palisade tissue, central mesophyll cells which are slightly enlarged anticlinally, and a complete lack of sclerification in the mesophyll. A continuous schizogenous canal system lined with epithelial cells lies in the ground tissues of all stem forms and leaves. The phellem is composed of thin-walled, only lightly suberized cells. Very broad parenchymatous areas develop in the secondary tissues of the rhizome, associated with a slowing of vascular cambial activity which creates depressions in the axis. Adventitious roots arise from ground tissues close to the primary xylem and from interfasicular regions close to the primary phloem in the rhizomes.  相似文献

菰(Zizania latifolia)是一种多年生挺水植物,为了探讨该植物根、茎和叶的解剖结构、组织化学及其质外体屏障的通透性生理。该文利用光学显微镜和荧光显微镜,对菰的根、茎、叶进行了解剖学和组织化学研究。结果表明:(1)菰不定根解剖结构由外而内分别为表皮、外皮层、单层细胞的厚壁机械组织层、皮层、内皮层和维管柱;茎结构由外而内分别为角质层、表皮、周缘厚壁机械组织层、皮层、具维管束的厚壁组织层和髓腔。叶鞘具有表皮和具维管束皮层,叶片具有表皮,叶肉和维管束。(2)不定根具有位于内侧的内皮层及其邻近栓质化细胞和外侧的外皮层组成的屏障结构;茎具内侧厚壁机械组织层,外侧的角质层和周缘厚壁机械组织层组成的屏障结构,屏障结构的细胞壁具凯氏带、木栓质和木质素沉积的组织化学特点,叶表面具有角质层。(3)菰通气组织包括根中通气组织,茎、叶皮层的通气组织和髓腔。(4)菰的屏障结构和解剖结构是其适应湿地环境的重要特征,但其茎周缘厚壁层和厚壁组织层较薄。由此推测,菰适应湿地环境,但在旱生环境中分布有一定的局限性。  相似文献

Seedlings of Taxus chinensis var. mairei were used as experimental materials to study the adaptation of Piriformospora indica to this plant under water stress. The materials were divided into two groups, namely, with or without inoculation with P. indica. Each group was subjected to four different levels of water stress. Vitality and physiological and biochemical indexes of the roots of T. chinensis var. mairei were regularly measured. Under water stress, T. chinensis var. mairei had significantly decreased root vitality; root vitality was higher in inoculated roots than in uninoculated roots. Under intense water stress, the inoculated roots had a higher soluble sugar content than the uninoculated roots. Under water stress, T. chinensis var. mairei experienced decreased activity of aerobic respiratory metabolic enzymes. The activity of anaerobic respiratory metabolic enzymes and alcohol dehydrogenase initially increased and then decreased, whereas that of lactate dehydrogenase increased. The inoculated roots had a higher activity of respiratory metabolic enzymes than the uninoculated roots. As water stress was further intensified, the roots had significantly decreased activity of aerobic respiratory metabolic enzymes and significantly increased activity of anaerobic respiratory metabolic enzymes. The activity of respiratory metabolic enzymes decreased faster in the uninoculated roots than in the inoculated roots. This study demonstrated that Piriformospora indica plays a positive role in enhancing the antihypoxic ability of T. chinensis var. mairei, thereby alleviating plant damage due to water stress.  相似文献

该研究采用石蜡切片和光学显微技术,对海南风吹楠营养器官的解剖结构及其对环境的适应性进行了探讨。结果表明:海南风吹楠为典型异面叶,叶片中脉发达,中部分化出髓,上表皮外侧具角质层,内侧具1层内皮层,下表皮外侧无角质层,有气孔器分布,气孔器为双环型,略下陷;栅栏组织3~4层细胞,海绵组织4~6层细胞。茎的初生结构中表皮轻微角质化,维管束为外韧型,8~10个初生维管束围绕髓排列为1轮;茎的次生结构中,表皮外部角质层加厚,维管柱紧密排列连成环状,次生韧皮部和次生木质部发达,形成层细胞3~5层。根的初生结构中表皮细胞外壁加厚,外皮层细胞体积大,形状不规则,内侧具1层形成层,内皮层具凯氏带,初生木质部为多原型,呈辐射状排列。根的次生结构中木栓层细胞5~6层,木栓层内侧具1层木栓形成层,栓内层细胞3层。海南风吹楠营养器官具有一定耐阴和耐旱结构特征,同时与其生活的热带雨林沟谷中高温荫湿的环境相适应。  相似文献

Four different ratios of river sand, ceramic pellets, vermiculite and perlite (1:1), and field soil were selected as the substrates in this experiment, and four gradient levels of root waterlogging, half waterlogging, full waterlogging and normal were set to investigate the effects of different gradients of waterlogging stress on the root morphology of Taxus chinensis var. mairei seedlings under different substrates. In this study, the root anatomical structure of Taxus chinensis var. mairei under different waterlogging stress was observed by the paraffin section method. The roots of T. chinensis var. mairei were diarch, with no pith and resin canals. There was a large number of tannins in the pericycle of the aerial adventitious roots of seedlings adapted to waterlogging. Also, the endodermis has obvious casparian strip thickening, and there were 4-5 layers of large parenchymatous cells in the close to the inner side of the pericycle in the vascular cylinder, which could increase the storage capacity, and transport capacity of the root. Under the treatment of root waterlogging stress, the development of plant roots in the mixed substrate of vermiculite and, perlite was the earliest. Under half waterlogging stress, T. chinensis var. mairei seedlings treated with various substrates all could better adapt to the environment of waterlogging stress. Under the stress of fully waterlogging, the roots of seedlings planted in river sand substrate developed secondary growth.  相似文献

Certain anatomical features, especially of roots, leaves, and fruits, provide useful criteria for delimiting a genus and defining its component species. Most species of Perideridia have 1-5 multistelic tuberous roots; however, P. Howellii and P. Kelloggii possess fascicles of up to 20 thickened fibrous roots, each of which is monostelic and composed of four or five primary xylem strands arranged alternately with isolated strands of secondary vascular tissues. The curious multistelic condition of the tuber apparently has not been reported for any other group of dicotyledons. An accessory vascular bundle located internally to the median collateral bundle of the leaf rachis is found in only three species of Perideridia. Depending on the species, the leaf mesophyll may be either dorsiventral or isolateral. In about two-thirds of the species, the oil ducts of the fruit are solitary in the intercostal intervals and paired on the commissure; in the remaining one-third they are more numerous in both situations, rising to 3-5 in the intervals and 12-13 on the commissure in P. Pringlei. Variation in number of oil ducts in the intervals assumes taxonomic importance because it has been used as a generic criterion in this group, but the present study shows no correlation between the number of oil ducts and other characters.  相似文献

蜈蚣草(Pteris vittata)是多年生的超积累砷植物,并用于修复受重金属污染的土壤。利用光学显微镜和荧光显微镜来研究蜈蚣草的解剖结构及组织化学特征,以此明确该物种适应干旱岩生环境,以及具有离子超富集作用的特点。结果表明(:1)蜈蚣草孢子体的根状茎、不定根和叶的结构均为初生结构,不定根的结构由内而外包括维管柱、内皮层、皮层、木质化厚壁组织层和表皮。(2)根状茎结构由内而外包括网状中柱、内皮层、皮层、表皮外覆盖的角质层。(3)羽状复叶的总叶柄的结构由内而外包括维管束、内皮层、皮层、厚壁组织层、表皮外覆盖的角质层。叶片为异面叶,表皮内方具厚壁层,叶表皮具角质层,仅下表皮有气孔。(4)蜈蚣草根表皮、皮层与根毛的表面富含果胶,皮层木质化;黄连素离子通透性试验结果显示,根毛、根表皮和皮层滞留大量黄连素离子。综上,植物体的内皮层、木质化厚壁组织层、异面叶和厚的角质层结构说明蜈蚣草适应岩生环境,根具木质化皮层和富含果胶的组织化学特点,以及离子通透性试验表明其与离子超积累功能有关。  相似文献

The lignin contents and anatomical structure of roots of wild cherry (Prunus avium L.) and pedunculate oak (Quercus robur L.) plantlets were compared to explain differences in response during transfer from in vitro to ex vitro conditions. Lignification of cell walls increased significantly in both oak and cherry roots during the period of acclimation and finally lignin content of root tissues of in vitro propagated plantlets reached the levels not significantly different from seedlings grown in soil. Later on when secondary tissues appeared, lignified secondary xylem constituted most of the tissues of both species. The most conspicuous interspecific difference in root structure was the presence of phi-thickenings in cortical layers just outer to endodermis in cherry roots cultivated ex vitro. Formation of phi-thickenings was avoided in vitro and their presence thus seems to be under environmental control. Suberised well established exodermis was present in roots of oak but not detected in those of cherry. Very early development of exodermis in oak roots, preceding suberisation of endodermis, was recorded in vitro but not in well aerated soil. While multilayered and well-developed cork occurred in oak, only thin walled and less suberised secondary dermal tissues were found in cherry.  相似文献

以桔梗(Platycodon grandiflorum A.DC)根为材料,运用石蜡切片和半薄切片法对其根的发育过程及结构进行解剖学观察,并对不同年限根的结构进行了比较。结果表明:桔梗根的结构发育过程包括原生分生组织、初生分生组织、初生生长和次生生长4个阶段。其原生分生组织由3群原始细胞组成,表现出典型分生组织的细胞学特征;初生分生组织包括根冠原、表皮原、皮层原和中柱原;初生结构由表皮、皮层和中柱组成,其中皮层薄壁细胞占主要地位,初生木质部为二原型;次生生长主要依靠维管形成层和木栓形成层的活动来完成,其次生结构从外到内由周皮和次生维管组织组成,次生维管组织占主导地位,其中以薄壁细胞为主,维管分子少量,分散在薄壁组织中。不同年限的根的结构基本相同,但它们在主根长度和直径、周皮厚度、木质部与韧皮部面积之比等方面存在差异。  相似文献

Rhodiola quadrifida is a rare mountain medicinal plant whose root extracts are used in traditional Chinese medicine as a hemostatic, antitussive, and tonic in the treatment of gynecological diseases. The aim of the study was to obtain R. quadrifida cultures at different degrees of differentiation in vitro and compare their growth characteristics and the content of salidroside and rosavin. Hairy roots were obtained by incubating cotyledons and hypocotyls in a suspension of Agrobacterium rhizogenes strain A4. The presence of the rolB and rolC genes was proven by polymerase chain reaction. The obtained roots were cultivated in Murashige-Skoog medium (MS). Calluses were obtained from the hairy roots in MS medium with the addition of hormones: 3 mg/L 2,4 D and 0.5 mg/L BAP. The presence of the main secondary metabolites of R. quadrifida, salidroside and rosavin, in calluses and salidroside in hairy roots by HPLC/MS was confirmed. The content of salidroside in callus culture was significantly higher than in hairy roots, 0.158 and 0.047%, respectively. The content of rosavin in callus culture was 0.07%. The content of rosavin and salidroside in callus culture was close to the level of these substances in the rhizomes of R. quadrifida plants growing in vivo, making this culture promising for its possible biotechnological use.  相似文献

采用石蜡切片和光学显微技术对江南油杉(Keteleeria fortunei(Murr.)Carr.var.cyclolepis(Flous)Silba)根、茎、叶的解剖结构进行观测,研究其形态结构对环境的适应性。结果显示:江南油杉叶片为异面叶,上表皮厚11.5μm,外侧覆盖厚4.5μm的角质层,下表皮厚8.6μm,外侧覆盖厚2.4μm的角质层,有气孔器分布,栅栏组织由1~2层细胞组成,海绵组织由2~3层细胞组成,主脉为单脉,厚474.1μm。茎的初生结构中表皮细胞1~2层,外皮层细胞4~6层,内皮层细胞6~8层,其内分布有树脂道;次生结构中木栓层细胞2~3层,栓内层细胞1~2层,皮层内有树脂道和分泌腔分布,维管束紧密排列连成环状。根的初生结构中外皮层细胞3层,内皮层细胞1~2层,具凯氏带,初生木质部为四原型;次生结构中木栓层细胞3~4层,栓内层细胞2~3层。江南油杉营养器官的解剖结构表现出较大的可塑性,使之既能较好地适应阳生环境又对阴生环境具备一定的适应性,还可耐受一定的干旱和寒冷。  相似文献

Soil contamination by toxic trace metal elements, like barium (Ba), may stimulate various undesirable changes in the metabolic activity of plants. The plant responses are fast and with, direct or indirect, generation of reactive oxygen species (ROS). To cope with the stress imposed by the ROS production, plants developed a dual cellular system composed of enzymatic and non-enzymatic players that convert ROS, and their by-products, into stable nontoxic molecules. To assess the Ba stress response of two Brassicaceae species (Brassica juncea, a glycophyte, and Cakile maritime, a halophyte), plants were exposure to different Ba concentrations (0, 100, 200, 300 and 500 μM). The plants response was evaluated through their morphology and development, the determination of plant leaves antioxidant enzymatic activities and by the production of plants secondary metabolites. Results indicated that the two Brassicaceae species have the ability to survive in an environment containing Ba (even at 500 μM). The biomass production of C. maritima was slightly affected whereas an increase in biomass B. juncea was noticed. The stress imposed by Ba activated the antioxidant defense system in the two species, noticed by the changes in the leaves activity of catalase (CAT), ascorbate peroxidase (APX) and guaicol peroxidase (GPX), and of the secondary metabolites, through the production of total phenols and flavonoids. The enzymatic response was not similar within the two plant species: CAT and APX seem to have a more important role against the oxidative stress in C. maritima while in B. juncea is GPX. Overall, total phenols and flavonoids production was more significant in the plants aerial part than in the roots, of the both species. Although the two Brassicaceae species response was different, in both plants catalytic and non-catalytic transformation of ROS occurs, and both were able to overcome the Ba toxicity and prevent the cell damage.  相似文献

为了明确地枫皮的结构特征及其在石灰岩山顶和山腰疏林间两种环境下生长的叶片解剖结构的差异,本研究采用石蜡切片和半薄切片对地枫皮营养器官进行解剖观察并评价了叶片结构对不同生态环境的响应。结果表明:地枫皮根中次生维管组织发达,木射线和韧皮射线明显。老茎的次生构造中,皮层贮藏物质丰富,内有大的石细胞群,韧皮射线和木射线明显;而髓细胞内含有大量晶簇和少量单晶。在叶片横切面观上,叶为异面叶,表皮细胞一层,上表皮无气孔分布,主脉中薄壁细胞中分散有石细胞。叶片解剖结构显示,随海拔高度上升,地枫皮趋向于旱生植物的特点,其主要表现在叶片表皮细胞外壁角质层加厚,栅海比增加、海绵组织排列由紧密变疏松。另外,根皮、茎皮和叶肉中都分布有大量油细胞。  相似文献

Panax notoginseng is a traditional Chinese medicinal plant. Root rot of P. notoginseng is one of the most serious diseases affecting P. notoginseng growth and causes wilted leaves, fewer lateral roots and rotten roots. Root rot is a soil-borne disease, and mainly occurs from June to August in Yunnan Province when the temperatures are high and the air is humid. In this study, the endophytic fungal genus Fusarium isolate E-2018.1.22-#3.2 was obtained from a P. notoginseng embryo. Fusarium isolate E-2018.1.22-#3.2 was identified as Fusarium striatum based on morphological characteristics and molecular analysis. The fungus was found to have conidiophores and macroconidia, and its ITS, LSU and TEF-1α genes shared 100%, 99.2% and 99% identities with the homologous genes of Fusarium striatum, respectively. Isolate F. striatum E-2018.1.22-#3.2 can cause root rot symptoms, including black, soft roots, fewer lateral roots and leaf wilt, in 93% of the experimental P. notoginseng plants, and could be re-isolated, fulfilling Koch’s postulates. When the P. notoginseng plants were treated with the fungicide pyraclostrobin, isolate F. striatum E-2018.1.22-#3.2 was unable to cause root rot. We have therefore demonstrated that F. striatum E-2018.1.22-#3.2 is able to cause root rot disease in P. notoginseng. This is the first report of root rot disease caused by F. striatum on P. notoginseng in China.  相似文献

This present study includes twelve species that represent the Ficus genus, namely; aspera, carica, tinctoria subsp. gibbosa, hirta, hispida, neriifolia, palmata, pumila, racemosa, septica, sur, and sycomorus, belonging to the Moraceae family. The species samples were collected from various locations in Egypt. The study focused on the anatomical and molecular characteristics of mature foliage leaves. Since the identification and classification of taxa are highly dependent on the anatomical features of leaves, the anatomical characteristics were recorded in the form of a comparison between the examined plants in the data matrix. This study aims to contribute to the identification of the studied species based on the anatomical details of the matured leaves. Anatomical characterization includes the variations in upper and lower epidermal layers that are covered by a thin or thick cuticle; the number of palisade and spongy layers; crystals; secretory elements; lithocysts; the midrib zone has parenchyma associated with mechanical tissue, vascular system, and investigation of trichomes; on the other hand, in the current study, the phylogenetic analysis was conducted by using the ITS and 5.8 S sequences. From the analysis of all the available data, it could be stated that there is an overall agreement with the anatomical character dendrogram.  相似文献
