Optical rotatory dispersion of E. coli ribosomes and their constituents   总被引:3,自引:0,他引:3  
Investigations have been made on the optical rotatory dispersion properties of E. coli ribosomes and their constituent RNA and proteins. The results indicate that (1) no conformational changes are involved in the formation of a 70S particle from the 50S and 308 subunits, (2) the E. coli ribosomal proteins are similar to most globular proteins with little α-helix content, and (3) the conformation of RNA and proteins inside the ribosome is very similar to that in the free state.  相似文献

Optical rotatory dispersions between 200 and 600 mμ are presented for mononucleosides and 5′-mononucleotides in neutral, acid, and in some cases alkaline solutions. All display a single Cotton effect in the ultraviolet region (above 220–240 mμ); the purine ones are negative in sign and the pyrimidine ones positive, with the crossovers (zero rotations) in most cases close to the wavelengths of their respective absorption maxima. The visible rotatory dispersions obey the one-term Drude equation, except for TMP and UMP, which show anomalous dispersions. The rotational strengths of the dichroic bands were estimated from the Cotton effect profiles; cytosine mononucleotides and mononucleotides show the strongest rotational strengths among the compounds studied.  相似文献

Osmotic pressure studies were carried on tobacco mosaic virus (TMV) and its components, protein and RNA, as well as on bis(3,3′-aminopropyl)amine, reported to be present in TMV preparations. Solvents were phosphate and barbital buffers at different values of pH and ionic strength. Measurements were made at room temperature. The Donnan effect was exhibited by TMV protein in phosphate buffer of 0.01 ionic strength at pH values ranging between 5.8 and 7.5. The observed values of the Donnan effect at pH 5.8 and 5.97 were in reasonable agreement with theoretical values calculated from the charge obtained by hydrogen ion titration. TMV-RNA in phosphate buffer at pH 7.5 and ionic strength 0.01 did not exhibit more than 1% of the expected Donnan effect. This is explained tentatively as the result of firm binding of metal ions. Negative values of osmotic pressure were observed with bis(3,3′-aminopropyl)amine. Similar anomalous osmosis was sometimes observed with TMV protein and with TMV. In agreement with earlier observations, TMV did not exhibit the Donnan effect in phosphate buffer of 0.01 ionic strength at pH values ranging from 5.5 to 8.0. However, TMV dialysed extensively in the presence of EDTA at pH 8.5 and TMV produced by reconstitution of purified protein and RNA did exhibit the Donnan effect in both phosphate and barbital buffers. The magnitude was of the same order as that calculated from the net charge determined by hydrogen ion titration. When reconstituted TMV, which did exhibit Donnan effect, was treated with calcium ions, the effect was abolished.  相似文献

Peroxidases (EC 1.11.1.7) have been implicated in the responses of plants to physical stress and to pathogens, as well as in a variety of cellular processes including cell wall biosynthesis. Tissue samples from leaf, root, pith, and callus of Nicotiana tabacum were assayed for specific peroxidase isozymes by analytical isoelectric focusing. Each tissue type was found to exhibit a unique isozyme fingerprint. Root tissue expressed all of the detectable peroxidase isozymes in the tobacco plant, whereas each of the other tissues examined expressed a different subset of these isozymes. In an effort to determine which peroxidase isozymes from Nicotiana tabacum are involved in cell wall biosynthesis or other normal cellular functions and which respond to stress, plants were subjected to either wounding or infection with tobacco mosaic virus. Wounding the plant triggered the expression of several cationic isozymes in the leaf and both cationic and anionic isozymes in pith tissue. Maximum enzyme activity was detected at 72 hours after wounding, and cycloheximide treatment prevented this induction. Infection of tobacco with tobacco mosaic virus induced two moderately anionic isozymes in the leaves in which virus was applied and also systemically induced in leaves which were not inoculated with virus.  相似文献

On reaching the stem each virus usually first moved downward, but in some plants both upward and downward movement occurred simultaneously and in a few
upward movement occurred first.
All three viruses travelled through the stem at approximately the same rate. Each was capable of travelling more than 80 cm. during the first 12 hr. after entering the stem, giving a minimal average rate of about 8 cm. per hr.
Uninfected pieces of stem invariably occurred between infected pieces. Maximum lengths of stem through which virus particles had apparently passed without causing infection, were 44.5, 49 and 39 cm. for the three viruses.  相似文献

Measurements of optical rotatory dispersion (ORD) and circular dichroism (CD) have been made in the range of 600-210 mμ for the β-glycan carbanilates as for instance, 2,3,6-tricarbanilylcellulose (I), 2,3,6-tricarbanilylmannan (II), 2,3-dicarbanilylcellulose (III), and octacarbanilylcellobiose (IV) and also for the α-glycan carbanilates, such as 2,3,6-tricarbanilylamylose (V), tricarbanilylpullulan (VI), 2,3-dicarbanilylamylose (VII), and octacarbanilylmaltose (VIII). Furthermore, the 2,3,4,6-tetracarbanilyl-α-methyl-glucopyranoside (IX) and the 1,2,3,4,6-pentacarbanilylglucose (X) have been measured in dioxane at 20°C. For the β-glycans a small negative CD in the region of 238–240 mμ and nearly symmetrical ORD curve with a crossover point at 238–240 mμ are found; this indicates a simple negative Cotton effect. In the case of α-glycosides, a strong negative CD with a maximum at 240–242 mμ and a strong positive CD with a maximum at 223–225 mμ were found; the ORD curves are asymmetrical and cross the abscissa in two places, at 241–243 and 220–222 mμ. With 2,3,4,6-tetracarbanilyl-α-methylglucoside (IX) no CD and ORD in the ultraviolet region and with 1,2,3,4,6-pentacarbanilyl-glucopyranoside (X) the ORD, but not the CD, could be measured. The ORD curve is nearly symmetrical, like those of the β-glycans but is of opposite sign. It seems impossible to discuss the striking difference of the CD and ORD spectra between the α-and the β-glycans in terms of contributions of single independant chromophores influenced by their individual different steric arrangements and their spatial relation to the glycosidic bond in C1. The exciton theory of Moffitt, which is suitable for explaining the ORD and CD spectra of helical polymers, has been applied to α- and β-glycans. A structure with helical parts is proposed for the α-glycans while a nearly planar arrangement is assumed for the β-glycans.  相似文献
