In order to assess the potential genotoxic effects of occupational exposure to petrochemicals, the incidence of sister-chromatid exchanges (SCE) in cultured lymphocytes was studied. Blood samples were taken from 233 individuals (184 exposed and 49 worksite controls) in an oil refinery and from 47 community control persons. The data showed a non-significant elevation of SCE frequency in occupationally exposed workers when compared to non-exposed individuals. The mean SCE frequency per cell ranged from 7.55 ± 0.55 in blood of lube oil blending and canning (LOBC) workers to 9.13 ± 0.71 in catalytic cracking and water treatment (CCWT) workers. The control values were 6.2 ± 0.67 and 7.21 ± 0.45 in the community and worksite individuals, respectively. Furthermore, the SCE frequencies were influenced neither by age nor by smoking.  相似文献

Fifty river buffalo (Bubalus bubalis, 2n?=?50) cows reared in two different provinces of Campania (southern Italy) underwent cytogenetic investigations to ascertain possible differences in their chromosome stability. One group (Caserta province) was under legal sequestration due to the presence in the milk mass of higher mean values of dioxins [21.79 pg/g of fat as sum of polychloro-dibenzo-dioxins (PCDDs), polychloro-dibenzo-furans (PCDFs) and dioxin-like polychlorobiphenyls (DL-PCBs)] than both those permitted (6.0 pg/g of fat as WHO-TEQ) and those (1.3 pg/g of fat as WHO-TEQ) observed in the control group raised in Salerno province. A cytogenetic study of papaya workers exposed to ethylene dibromide   总被引:3,自引:0,他引:3  
Ethylene dibromide (EDB) has been shown to be carcinogenic in animal studies and mutagenic in vitro. One cytogenetic study of workers exposed to low levels of EDB for short durations was negative. To test whether exposure to low levels of EDB over long periods caused cytogenetic changes, we have assessed the frequencies of sister-chromatid exchanges (SCE) and chromosomal aberrations (CA) in the peripheral blood lymphocytes of 60 men occupationally exposed to EDB. These men worked in papaya-packing plants where EDB was used to fumigate the fruit after harvest to kill fruit-fly larvae. 42 other men who worked at a nearby sugar mill served as controls. The average duration of exposure of the papaya workers was 5 years. 82 full shift personal breathing-zone air samples indicated that the papaya workers were exposed to a geometric mean of 88 ppb of EDB, as an 8-h time weighted average (TWA). Peaks up to 262 ppb were measured. The proposed OSHA 8-h TWA for EDB is 100 ppb, while NIOSH recommends 45 ppb. No differences in SCE levels were found between exposed and nonexposed workers. No differences were found in the total CA frequency between exposed and nonexposed workers. SCE levels were significantly increased in men who smoked cigarettes (p = 0.0001) and in men who smoked marijuana (p = 0.01). ABSTRACT

The aim of this study was to compare metabolic aspects potentially associated with metabolic syndrome (MetS), in addition to serum leptin levels and sleep, according to exposure to night work among nursing staff. A cross-sectional study involving 60 nursing staff was conducted. Sociodemographic, sleep (Karolinska Sleep Questionnaire), physical activity (International Physical Activity Questionnaire), and nutrition data (16-Food Intake Questionnaire) were collected. Body measurements and blood pressure were taken and blood collected to determine glycemia, total cholesterol and portions of low-density lipoprotein and high-density lipoprotein and triglycerides, and leptin levels. The sample was divided into three groups according to exposure to night work (nonexposed, currently exposed, past exposure). Furthermore, to evaluate the relationship between exposure time and prevalence of MetS, the night-exposed groups (past and current) were divided according to time exposed to night shifts (<10 years and ≥10 years). Mean age of participants was 39.8 ± 10.5 years. The groups were homogenous with regard to sociodemographic characteristics, physical activity, dietary patterns, and health aspects. The prevalence of MetS in the population studied was 32%, above the rate for the general population. However, there were no significant differences among the groups. In addition, a higher proportion of participants with hypertriglyceridemia and diastolic arterial hypertension was observed in the currently exposed group. The currently exposed group also reported less sleep and higher sleep debt than the other groups on workdays. Although no differences were observed among the groups regarding MetS, the currently exposed group had more sleep disturbances than the others, and a higher prevalence of two out of three risk factors for the MetS diagnosis.  相似文献

Sister chromatid exchange (SCE) frequency and high-frequency cells (HFCs) were analyzed in 50 storage battery plant workers with mean blood lead level (BLL) of 40.14±9.99 μg/dL. The mean BLL in the control group (n=30) was 9.77±1.67 μg/dL. This difference in mean BLLs between control and exposed group was statistically significant (p<0.05) and reflects clearly the lead exposure in the workers. Urinary aminolevulinic acid (U-ALA) was also determined in both control (3.37±0.89 mg ALA/g creatinine) and exposed groups (12.39±6.18 mg ALA/g creatinine) and U-ALA excretion was statistically higher (p<0.05) in lead-exposed workers. The relationship between biomarkers of lead exposure/effect and HFC percentage was higher than the relationship between biomarkers of lead exposure/effect and SCE frequency. Accordingly, HFC analysis seemed to be more sensitive than the SCE analysis as a cytogenetic biomarker for lead exposure. Additionally, the statistically significant correlation (r 2=0.880, p<0.01) between U-ALA excretion and HFC percentage in lead-exposed workers supported the probability of ALA mediated indirect mechanism for lead genotoxicity.  相似文献

Abstract

There is apprehension about widespread use of electrical and electromagnetic gadgets which are supposed to emit electromagnetic radiations. Reports are controversy. These electromagnetic fields (EMFs) have considerable effect on endocrine system of exposed subjects. This study was focused to assess the possible bioeffects of extremely low-frequency (ELF)-EMFs on epinephrine level, DNA damage and oxidative stress in subjects occupationally exposed to 132?kV high-voltage substations. The blood sample of 142 exposed subjects and 151 non-exposed individuals was analyzed. Plasma epinephrine was measured by enzyme-linked immunosorbent assay, DNA damage was studied by alkaline comet assay along with oxidative stress. Epinephrine levels of sub-groups showed mean concentration of 75.22 ± 1.46, 64.43 ± 8.26 and 48.47 ± 4.97 for high, medium and low exposed groups, respectively. DNA damage ranged between 1.69?µm and 9.91?µm. The oxidative stress levels showed significant increase. The individuals employed in the live-line procedures were found to be vulnerable for EM stress with altered epinephrine concentrations, DNA damage and increased oxidative stress.  相似文献

The cytogenetic damage in floriculturists of Morelos State, Mexico, exposed to pesticides, was evaluated by mean of biological tests based on sister chromatid exchanges (SCE) in lymphocytes of peripheral blood and micronuclei (MN) in exfoliated cells of the buccal mucosa. Besides the cytogenetic analysis, the effects of pesticides exposure on the cell proliferation kinetics (CPK) by the replication index (RI) were also studied. The mitotic index (MI) to detect cytotoxic effects was also determined. Greenhouses of the towns of Santa Catarina, Jiutepec and Yecapixtla were selected for the study, because the application of chemicals to the flowers is uncontrolled. As non-exposed group, people of the town of Temisco were chosen; their activity was not related to pesticides. The SCE were analyzed in the peripheral blood of 30 persons, 22 women and 8 men, with 10 and 1.5 years of exposure to pesticides, respectively, and of 30 persons, 28 women and 2 men, that were considered as the non-exposed group. Samples of buccal mucosa were also taken from each person. Significant differences between exposed and non-exposed groups were found in SCE, CKP and MI. Besides, the MN frequencies in the exposed group were three times higher than in the non-exposed group.  相似文献

Abstract. Changes in the discontinuous gas exchange cycle of pupal beet armyworm, Spodoptera exigua (Hübner) (Lepidoptera: Noctuidae), exposed or not to Cry1C Bacillus thuringiensis toxin, are examined against developmental age (1–7 days) and at different temperatures (10–25 °C) using flow through respirometry. Both exposed and nonexposed pupae exhibit discontinuous gas exchange, but only at 10 °C; the frequency of cyclic release of CO2 increases with increasing temperatures. The three phases of the discontinuous gas exchange cycle are distinct for both treatment groups. However, the duration of each phase is significantly greater for pupae exposed previously to toxin. The closed phase is 40 ± 14% longer, the flutter phase 23 ± 19% longer, and the open phase is 28 ± 12% longer when pupae were exposed to toxin. Respiratory water loss is 4.5 ± 1.3% for toxin exposed pupae and 2.1 ± 2.4% for unexposed pupae. Furthermore, the exposed pupae have significantly greater cuticular permeability (26.01 ± 1.9 µg cm−2 h−1 mmHg−1) than the nonexposed pupae (9.64 ± 0.9 µg cm−2 h−1 mmHg−1). However, in both strains, cuticular transpiration (>93%) far exceeds respiratory transpiration. Overall, total water loss is significantly greater in pupae whose larvae are exposed to toxin compared with pupae from nontreated larvae. Toxin exposed pupae have a mean cycle duration of 60 ± 2.5 min whereas that of nonexposed pupae is 42 ± 1.8 min.(ml g−1 h−1) of the open phase is greater earlier in pupal life followed by a minimum at mid-pupal stage and an increase at late-pupal development in both treatment groups. Combining all 7 days, closed, flutter and open phase (ml g−1 h−1), pupae exposed to toxin produce significantly more CO2 during each phase. On average, toxin exposed pupae produce 52 ± 12, 43 ± 10 and 15 ± 37% more CO2 than the untreated pupae during the closed, flutter and open phases, respectively. Therefore, the present study reinforces the need to use insects of similar developmental age in studies of insect respiration patterns and energy metabolism.  相似文献

The neurotoxicity of manganese (Mn) is well known, however, the neurochemical effect caused by this metal is less well investigated. In this study, urinary homovanillic acid (HVA) and vanillymandelic acid (VMA), two end products of catecholamine metabolism, were measured in 39 workers chronically exposed to Mn in a manganese smelting plant. The average duration of Mn exposure was 17.4 yr. Nineteen nonexposed workers were also studied. Concentrations of Mn in serum (MnS) and in urine (MnU) were measured by Zeeman graphite furnace atomic absorption spectrophotometry (ZAAS), and HVA and VMA determined by high performance liquid chromatography (HPLC). For Mn-exposed workers, the concentration of MnS was nearly 2.8 times (1.61 ± 0.16 mg/L vs 0.56 ± 0.16 mg/L) and MnU about 4.5 times higher (7.62 ± 0.17 mg/L vs 1.69 ± 0.16 mg/L) than the nonexposed. Although the geometric mean concentration of HVA in exposed workers was similar to that of the nonexposed (3.09 ± 1.39 mg/g ere. vs 2.99 ± 1.40 mg/g cre.), the VMA concentration was significantly higher (3.02 ± 1.43 mg/g cre. vs 2.49 ± 1.58 mg/g cre.,p = 0.033). Multiple regression analysis showed that although there were no correlations between any of these parameters with the duration of exposure to Mn, both HVA and VMA showed significant correlations with increase in MnS and MnU. These data provide evidence that exposure to Mn was associated with measurable increase in catecholamine metabolites. This finding is compatible with recent observations in laboratory animals that Mn interferes with neurochemical metabolism.  相似文献

ABSTRACT

Recently we evaluated by actigraphy the rest-activity circadian rhythm (RAR) in breast cancer (BC) survivors at 5 years from primary diagnosis, as well as in a control group with similar age and body mass index (BMI). RAR, analyzed by Cosinor method, resulted significantly different in BC survivors compared to healthy subjects: BC survivors showed lower values of MESOR and Amplitude (A), while acrophase (φ) was similar in the two groups.

Now, using non-parametric methods we have detected Interdaily Stability (IS), Intradaily Variability (IV), nocturnal activity (L5), and daily activity (M10) on the same sample of previous study: 15 BC survivors at 5 years from the primary diagnosis (mean age = 56.7 ± 6.6 yrs; mean BMI = 24.5 ± 3.8 Kg/m2) and 13 healthy controls (mean age = 54.4 ± 7.2 yrs; mean BMI = 25.2 ± 2.8 Kg/m2).

The non-parametric indices showed that in BC-group IV was significantly higher than in Ctrl-group (0.86 vs. 0.65 a.u. in BC and Ctrl, respectively; p <.01), while L5 (11.27 vs. 34.41 a.c. in BC and Ctrl, respectively; p <.0001) and M10 (326.82 vs. 428.07 a.c. in BC and Ctrl, respectively; p <.01) were significantly lower compared to Ctrl-group.

The data suggest that BC patients need constant clinical assessment of RAR characteristics along the years following the primary diagnosis. The analysis of RAR in all its components, parametric and non-parametric, is important to detect alterations in the sleep-wake cycle and can be useful for developing new strategies for health protection, such as structured and tailored physical activity programs, to improve circadian activity level in order to raise the quality of life in BC survivors.  相似文献

Due to high mercury levels in many Mediterranean aquatic organisms, people who live in this area and consume large amounts of seafood are exposed to a toxicological hazard. A group of 51 fishermen exposed to mercury through eating contaminated seafood from the northern Tyrrhenian Sea underwent cytogenetic monitoring. This work is part of a research project consisting of the evaluation of micronuclei (MN), chromosomal aberrations (CA) and sister-chromatid exchanges (SCE) in peripheral blood lymphocytes. Here we present data on mercury levels in blood and on micronucleus frequencies in peripheral blood lymphocytes of fishermen. The range of mercury concentrations in blood was 10.08–304.11 ng/g fresh weight, the average was 88.97±54.09 ng/g. Micronucleus frequency was defined with at least 2000 binucleated cells scored for each person; the average was 8.74 ± 2.56 expressed on 1000 binucleated cells. A statistical correlation was found between MN frequency and total mercury concentration in blood (p = 0.00041, r = 0.674), as well as between MN frequency and age (p = 0.017). No other parameters taken into account correlated with MN frequency.  相似文献

Thyroid hormones are essential for body homeostasis. The scientific literature contains restricted proofs for effects of environmental chemical factors on thyroid function. The present study aimed at evaluating the relationship between toxicological parameters and concentration of thyrotropic hormone in persons occupationally exposed to lead, cadmium and arsenic. The studies were conducted on 102 consecutive workers occupationally exposed to lead, cadmium and arsenic (mean age 45.08 ± 9.87 years). The estimated parameters characterizing occupational exposure to metals included blood cadmium concentration (Cd-B), blood lead concentration (Pb-B), blood zinc protoporphyrin concentration (ZnPP) and urine arsenic concentration (As-U). Thyroid function was evaluated using the parameter employed in screening studies, the blood thyrotropic hormone concentration (TSH). No differences were disclosed in mean values of toxicological parameters between the subgroup of persons occupationally exposed to lead, cadmium and arsenic with TSH in and out of the accepted normal values. Logistic regression demonstrated that higher blood total bilirubin concentrations (ORu = 4.101; p = 0.025) and higher Cd-B (ORu = 1.532; p = 0.027) represented independent risk factors of abnormal values of TSH in this group. In conclusion, in the group of workers exposed to lead, cadmium and arsenic, higher blood cadmium concentration seems to augment the risk of abnormal hormonal thyroid function.  相似文献

This study was aimed to examine the influence of acclimatization on the change of concentration of stress hormones in men’s serum exposed to heat stress during physical training. The study included a total of 40 men, aged 19–21 years, divided randomly into four groups: CTRL group: control, exposed to the Exercise Tolerance Testing in comfortable conditions; O group: exposed to Exercise Tolerance Testing in a warm environment; P group: exposed to passive acclimation to heat for 10 days, followed by Exercise Tolerance Testing in a warm environment; A group: exposed to active acclimation to heat for 10 days, followed by Exercise Tolerance Testing in a warm environment. All participants were tested for thermoregulation and acclimatization, skin and tympanic temperature, heart rate (HR), hormonal status and sweating. The mean skin temperature was the lowest in the control group of subjects exposed to physical exertion under comfortable conditions, and at each point of measurement it was statistically significantly different from that of the other study groups (p?<?0.001). Sweating intensity was statistically significantly the lowest in the CTRL group (0.32?±?0.04 l/m2/h; p?<?0.001), compared to all other groups. Cortisol was significantly altered in O group (632.2?±?92.3; 467.2?±?89.7), testosterone levels were significantly altered in P (19.2?±?9.3; 16.4?±?7.3) and in A groups (22.1?±?12.4; 14.9?±?9.9), while prolactin was changed in O (392.1?±?51.3; 181.4?±?42.3), P (595.1?±?191.1; 191.2?±?52.5), and A group (407.4?±?189.3; 173.4?±?43.9) after the experimental period. The impact of acclimatization on hormonal indicators emphasizes its importance in the response of the endocrine system of soldiers to perform military activities in warm climates.

Objectives: The aim of this study was to evaluate the effect of oral functional training and nutrient supplements to improve the nutrition of malnourished elderly people in a nursing home. Background: Malnutrition is a frequent problem in the elderly requiring long‐tem care; however, it is not clear whether oral functional training can be effective to improve nutrition. Subjects and methods: Fourteen subjects of 82 residents (mean age 85.7 ± 6.2 years) in a nursing home, who had a serum albumin level of ≤3.8 g/dl and understood the purpose of this study, were randomly divided into two groups of seven: one group served as the supplement group (mean age 87.0 ± 4.9 years) to which a high‐calorie and high‐protein diet was provided, and the other as the oral training plus supplement group (mean age 84.6 ± 10.1 years) to which oral functional training was given by a dental hygienist once a week as well as the above diet. Nutritional status was evaluated using serum biochemical values as indices at 4 months after the start of the intervention. Results: In the supplement group, serum albumin was 3.44 ± 0.36 g/dl at the start of the study (before intervention) and 3.24 ± 0.45 g/dl at 4 months after intervention. In the oral training plus supplement group, it was 3.56 ± 0.22 g/dl before intervention and significantly increased to 3.70 ± 0.33 g/dl after intervention (p < 0.05: Wilcoxon signed‐rank test). Conclusion: Nutritional supplements alone were not adequate, but with oral functional training to maintain and improve feeding function, nutritional improvement in the elderly could be observed.  相似文献

The frequencies of sister-chromatid exchange (SCE) in peripheral blood lymphocytes of 40 workers at a phosphate fertilizer factory in North China were studied. HF and SiF4 are main air pollutants in the factory, there is also some dust containing fluoride, phosphate fog, NH3 and SO2. It was shown that the chemicals caused an increase in SCE, and also induced cell mitotic delays. The mean SCEs/cell of the workers and the non-exposed controls were 7.47 ± 0.31 and 4.94 ± 0.14 (p < 0.01) respectively. SCEs/cell in 75% of 40 workers were higher than 6 while 40 controls all had values lower than 6. SCE frequencies of the workers increased with length of the chemical exposure period up to 10 years. Smoking enhanced the SCE frequencies induced by the chemicals.  相似文献

In experiments to assess the effects of several biological, chemical, and physical variables on sister-chromatid exchange (SCE) induction in cultured lymphocytes exposed to mitomycin C (MMC) before PHA stimulation we observed: (1) high SCE frequencies in female cells, and normal SCE frequencies in Y-bearing metaphases in mixed cultures containing equal numbers of MMC-treated female lymphocytes and untreated male lymphocytes; (2) small, but statistically significant, decreases in SCEs with increasing pH after G0 exposure in the pH range 6.6–7.6; (3) pronounced reductions in MMC-induced SCEs in lymphocytes exposed at 4°C vs. 37°C. In other studies, SCE induction was evaluated in cultures exposed during G0 to MMC concentrations ranging from 0.25 to 2.5 μg/ml for varying time intervals ranging from 5 min to 24 h. For all concentrations tested SCE induction varied as a linear function of G0 exposure time. To compare SCE induction between cultures, we calculated the mean frequencies of SCEs induced per metaphase/unit dose MMC/unit G0 exposure time (SCE/μg/h). A mean frequency of 20.7 ± 4.8 SCE/μg/h was observed for 41 lymphocyte cultures suggesting that a single term adequately describes the rate of SCE induction following G0 exposure to a 10-fold range in concentration of MMC for time intervals of 30 min to 24 h.  相似文献

Objective : To evaluate the short‐term impact of portion‐controlled food provision in combination with an Internet behavioral weight loss program on weight, blood cholesterol, and blood glucose levels. Design and Methods : Fifty participants, mean age 46 ± 10.7 years and mean body mass index 35.1 ± 3.8 kg/m2, were randomized to one of two study groups, an Internet behavioral weight loss program (Internet‐alone; n = 25) or an Internet behavioral weight loss program plus a commercially available portion‐controlled diet (Internet + PCD; n = 25) for 12 weeks. Results : An intent‐to‐treat analysis found that the mean weight change in the Internet + PCD group was ?5.7 ± 5.6 kg and in the Internet‐alone group (n = 25) was ?4.1 ± 4.0 kg (P = 0.26). Participants in the Internet + PCD group achieved significantly greater improvements in blood glucose (?2.6 ± 5.7 vs. 1.4 ± 11.0 mg/dl; P = 0.05) and LDL cholesterol (?8.2 ± 18.0 vs. ?0.6 ± 21.0 mg/dl; P = 0.04), compared with Internet‐alone group. Conclusions : These data suggest that there may be short‐term clinical benefit in using a PCD in conjunction with a behavioral Internet‐based weight loss program to enhance weight loss and improve health indicators.  相似文献

This double-blind study tested the effects of pulsating electromagnetic field (PEMF) therapy sessions on the changes in peripheral cardiovascular function in a group of aging adults after 12 weeks of treatment. Each therapy session involved 15 min of exposure to low-frequency PEMF with asymmetrical waveforms emitted by the Impulser? Pro mattress. The treatment was provided 5 days per week for a total of 60 sessions. Resting blood pressure and arterial stiffness index were determined for peripheral cardiovascular function. Fifty-four older men and women (mean age 59.8 ± 3.5 yrs) completed the entire protocol involving either the PEMF or a sham treatment. The results include statistically significant reductions in systolic and pulse blood pressure, while no significant difference in diastolic pressure or the index of arterial stiffness was observed. These findings suggest that the PEMF treatment might be linked to improvements in peripheral resistance or circulation.  相似文献

The present detailed study aimed to establish for the first time the ovarian and uterine hemodynamic change in dairy cows after the end of the puerperal period to investigate a possible association between Doppler indices and volume of blood flow. Twenty cows weighing 500–600 kg (mean 400 ± 50 kg) and 4–5 years of age (mean 3.2 ± 0.5 years) categorized into two main groups (true anestrum, with corpus luteum, n = 10) and (normal cyclic, n = 10), The period of examination started from day 39 till day 70 after calving with day after day routinely examination by b-mode and Doppler performed on both ovaries with ovarian arteries (OA) and uterus with uterine arteries (MUA) in the ipsilateral (ipsi) and contralateral (contra) side to ovulation, in addition to a thickness in horns and body was measured. Estradiol and progesterone were also measured. Results showed that both Doppler indices in the OA and MUA ipsi and contra had a positive (P ≤ 0.001) correlation with contra Doppler indices, but revealed a negative (P ≤ 0.05) correlation with ipsi and contra Doppler velocities, blood flow rate and volume in anestrum cows. Both ovarian and uterine ipsi indices showed an increase (P ≤ 0.05) in the anestrum cows, and both PSV and EDV of both arteries ipsilateral showed (P ≤ 0.05) a decrease in the anestrum cows, the ipsi and contra ovarian and uterine colored % were lower in anestrum group than the normal group. Estradiol (P ≤ 0.05) decreased in anestrum cows than the normal, while progesterone increased in the anestrum group.
