In this work the possibility of using microwaves for immunomodulation in the immunization of animals with thymus-independent antigen was studied. The projection zones of the thymus and adrenal glands of the test animals were subjected to the action of decimeter, or ultrahigh frequency (UHF), waves, while the corresponding zones of the control animals were subjected to imitation UHF irradiation. Vi-antigen was shown to be a thymus-independent antigen for rabbits (according to the results of the evaluation of the functional state of thymocytes, as manifested by the adsorption of acridine orange by the chromatin of thymocyte nuclei and by the content of RNA and DNA in the thymus). The action of UHF waves on the projection zone of the thymus was accompanied by a decrease in the glucocorticoid activity of the adrenal cortex, observed simultaneously with a pronounced immunostimulating effect. The UHF irradiation of the zone of the adrenal glands was accompanied by immunosuppression in combination with enhanced glucocorticoid activity of the adrenal cortex.  相似文献

Morphophysiological characteristics and peculiarities of adrenal gland of bank vole (Clethrionomys glareolus) were studied in the area of natural biogeochemical province with abnormally high content of nickel, cobalt and chromium. The control population inhabited area with usual content of these elements. We used 4-factor analysis of variance to estimate the influence of geochemical conditions, phase of population cycle, sex and reproductive state on the morphophysiological characteristics of animals and functional activity of adrenal gland. Animals from area with high concentration of Ni, Co and Cr show an increase in relative mass of adrenal glands, fascicular zone of adrenal cortex, size of cells and their nuclei. All these changes can be considered as an evidence of increased secretion of glucocorticoids. It is shown that phase of population cycle influences fatness of animals, size of nuclei, cells and adrenal cortex. Females in comparison with males are characterized with higher indexes of liver and adrenal gland, as well as morphometric indexes of adrenal cortex. The maturation of animals is accompanied with increase in body mass, fatness and relative mass of adrenal glands, the size of cortex zone, nuclei and cells themselves. It is supposed that the effect of "geochemical factor" results in intensification of glucocorticoid secretion of adrenal costex, thus increasing non-specific resistance of animals inhabiting area with high concentration of heavy metals. Such factors as "phase of population cycle", "sex" and "reproductive state", influence mineralocorticoid activity, glucocorticoid and androgenic functions of adrenal cortex. Studies were done to determine the effects of age on steroidogenesis in the inner (zona reticularis) and outer (zona fasciculta plus glomerulosa) zones of the guinea pig adrenal cortex. In 35-day-old animals, cortisol production by adrenal outer zone cells was approximately twice as great as that by inner zone cells. With aging, cortisol secretion by inner zone cells decreased to very low levels, but there was no detectable change in the capacity for cortisol production by the outer zone. However, the outer zone comprised a progressively decreasing fraction of the total adrenal mass in older animals. To determine the basis for the decline in cortisol production by inner zone cells with aging, the activities of several steroidogenic enzymes were determined. Microsomal 21-hydroxylase activity was greater in the inner than outer zone but was not significantly affected by age. By contrast, 17-hydroxylase activity was greater in the outer zone at all ages, and decreased with aging in the inner but not the outer zone. Mitochondrial cholesterol sidechain cleavage and 11β-hydroxylase activities were also higher in the outer than inner zone and declined in the zone only in older animals. The decrease in inner zone cholesterol sidechain cleavage activity with aging was proportionately greater than the age-dependent changes in other enzyme activities. The results indicate that the effects of aging on steroidogenesis are both zone- and enzyme-specific. The overall decline in cortisol secretion by the guinea pig adrenal cortex with aging is attributable to both a decrease in cortisol production by the cells of the zone reticularis and a disproportionate increase in the mass of the gland comprised by this zone. The decrease in cortisol secretion correlates closely with a decline in cholesterol sidechain cleavage activity in the zona reticularis, and may be causally related.  相似文献

It has been reported that cells isolated from the inner zone of the guinea pig adrenal cortex fail to have a steroidogenic response to ACTH. To further explore this, adenylate cyclase activity of membrane particles and cAMP production by cells prepared from the inner and outer adrenocortical zones were determined. The cAMP response to ACTH and forskolin was similar for cells from both zones. Basal adenylate cyclase activity was significantly higher in the inner zone; and while absolute responses to ACTH, GppNHp, GTP, NaF, and forskolin were greater for the inner zone, relative responses were similar for the two zones. These observations suggest that the inner zone of the guinea pig adrenal cortex may have a defect in ACTH action at a step(s) beyond cAMP formation.  相似文献

The action of ultrahigh frequency (UHF) waves on the projection zone of the adrenal glands has been found to suppress both primary and secondary immune response, which is probably due to an increase in the glucocorticoid function of the adrenal cortex. The synthesis of nonspecific immunoglobulins has proved to be more resistant to the suppressive action of UHF than the synthesis of specific immunoglobulins.  相似文献

Low-density lipoprotein (LDL) receptor activity and the concentration of cholesterol were measured in the outer (glomerulosa/fasciculata) and inner (reticularis) zones of the adrenal cortex of the guinea pig to examine the relation between cholesterol content and LDL receptor activity. While the concentration of cholesterol was 2-3-times higher in the outer cortical zone, the maximum high-affinity binding capacity for LDL was essentially the same for the two zones, or slightly higher for the inner zone. Adrenocorticotrophic hormone (ACTH) caused a significant increase in LDL receptor activity only in the outer zone, but led to a reduction in the cholesterol content in both adrenocortical zones. The treatment of animals with 17 alpha-ethinyl-estradiol also resulted in a reduction of cholesterol in both adrenocortical zones, but an increase in LDL receptor number only in the outer zone. The latter effect was partially reversed by the administration of dexamethasone. Aminoglutethimide, which was used in a dose that did not block steroidogenesis but did block the hydrolysis of cholesteryl esters in response to ACTH, did not prevent the ACTH-induced increase in LDL receptor number in the outer zone. Thus, the number of LDL receptors was increased in the zona fasciculata by ACTH in the absence of a reduction in cellular cholesterol content, while the number of LDL receptors in the zona reticularis was not increased by ACTH even in the face of a reduction in cellular cholesterol. Exclusive of the experiments employing aminoglutethimide, when the cellular cholesterol content was plotted against LDL binding activity, an excellent inverse correlation was revealed for the zona fasciculata, but essentially no correlation was noted for the zona reticularis. It is concluded that the outer and inner cortical zones of the guinea pig adrenal are quite distinct in the nature of their LDL receptor activity and regulation: the LDL receptor of the outer zone appears to function in a way similar to what has been reported for the whole adrenal cortex of other species in that receptor number correlates with tissue cholesterol content and is primarily regulated by ACTH; the LDL receptor number of the inner zone, however, does not correlate with tissue cholesterol content and is apparently not regulated by ACTH.  相似文献

The effect of irradiation of the central nervous system by microwaves (MW) at a frequency of 2450 MHz and power 5-40 W on the regulation of cerebral circulation and oxygen supply to the nervous tissue were studied in rabbits. Local irradiation of the exposed cerebral cortex resulted in hyperemia and hyperoxia in the zone of exposure induced by the hyperthermal effect of MW. When the region of the medulla oblongata was irradiated even with low MW power (not leading to hyperthermia), the local circulation and oxygen tension increased in the whole brain, apparently due to the impairment of the regulation of the cerebral blood flow and oxygen supply to the brain tissue.  相似文献

In experiments on 123 male rats have been found that the combination of immobilizing and cold (4 degrees C) stresses during 6 hours results in the formation of 4.5 gastric mucosa ulcers in every rat, marked activation of lipoperoxidation and depression of antioxidant activity of myocardium, significant increase of relative mass of adrenal glands, decrease of spleen mass and death of 80% animals accompanied by the reduction of concentration of thyroid hormones in blood serum and hypothermia (to 28 degrees C). The prestress injection of small doses of thyroidine decreased of mucosa defect rate and the number of ulcers to 2.8, made less marked the changes of relative mass of adrenal glands and spleen and the changes of concentration of thyroid hormones in blood serum, significantly limited the activation of lipoperoxidation, depression of power in antioxidant systems of myocardium, hypothermia (to 32.5 degrees C) and significantly increased the survival rate (to 70%).  相似文献

The model of chronic intermittent stress by immobilization during pregnancy may produce alterations in the mechanisms that maintain adrenal gland homeostasis. In earlier investigations using this model, significant variations in plasma prolactin and corticosterone levels, and adrenal gland weights were observed. We hypothesized that chronic stress causes changes in apoptosis in the adrenal glands of pregnant rats. We identified and quantified apoptotic cells in the adrenal cortex and examined their ultrastructural characteristics using transmission electron microscopy. Adrenal glands of pregnant rats at gestation days 12, 17 and 21 were studied for control and experimental (stressed) rats. Immunolabelling techniques, stereological analysis and image quantification of adrenal gland sections were combined to determine differences in apoptosis in the different cell populations of the adrenal cortex. The apoptotic index of the experimental rats showed a significant reduction at gestation day 17, while at days 12 and 21 there were no differences from controls. Moreover, the apoptotic index of the reticular zones in control and experimental animals showed a significant increase compared to the glomerular and fascicular zones at the three gestation times studied. Chronic stress by immobilization reduced the caspase-dependent apoptotic index at gestation day 17, which may be related to variations in plasma concentrations of estrogens and prolactin.  相似文献

The effect of alcoholic typhoid vaccine and of the cultural vaccine against the tickborne encephalitis on the adrenocortical secretion in guinea pigs was studied. Functional condition of the adrenal glands was assessed histochemically. Immunization was accompanied by increase in the activity of the adrenal cortex, the most pronounced the first 3 days. Comparative analysis showed that typhoid vaccine produced a more pronounced stree on the adrenal gland function.  相似文献

The guinea pig adrenal cortex is composed of two chromatically distinct concentric zones. The steroidogenic response to ACTH by the two zones is likewise distinct: ACTH stimulates cholesterol side-chain cleavage activity in the outermost zone, but fails to do so in the inner zone. This despite the fact that adenylate cyclase activation by ACTH and cAMP formation are similar for the two zones. To further examine this model, protein kinase activity and protein phosphorylation have been examined. It was found that the cAMP-dependent, Ca2+/phospholipid-dependent, and Ca2+/calmodulin-dependent protein kinase activities were significantly higher in the outer zone than in the inner zone by 70, 60 and 800%, respectively. Although the physiological meaning of a zonal difference in protein kinase activity is not as yet clear, the marked difference in Ca2+/calmodulin-dependent protein kinase activity between the inner and outer zones correlates well with the marked difference in steroidogenesis that exists between the two zones. Of the Ca2+/calmodulin-dependent protein kinases known to exist, there is preliminary evidence to suggest the presence of kinase III in the guinea pig adrenal cortex. Protein phosphorylation induced by the three kinase systems in the two adrenocortical zones revealed notable differences in phosphoprotein patterns. In addition, it was found that exogenous calmodulin was phosphorylated and that the kinase responsible for this was more active in the inner zone.  相似文献

This work was carried out to study dynamics of morphometric structure of thyroid and adrenal glands in bulls brought up in early postnatal ontogenesis at lowered (-4.4-(-)8.0 degree centigrade) and raised (8.7-20.5 degree centigrade) environment temperature with further grow and fattening using intensive technology with application of new biogenic matters. For the first time it has been experimentally proved the expediency of correction of morphometric structure of investigated endocrine glands in bulls under condition of joint application of Suvar with Polistim or with DAFS-25 during the various periods of postnatal ontogenesis. Special features of morphometric structures of the thyroid glands (diameter, the area of follicles, thickness of thyroid epithelium, follicles index) and the adrenal glands (weight, width of zones of cortical and brain substances, the areas of cells and nuclei, and nuclear cytoplasmic ratio) in animals maintained under condition of adaptive technology have been investigated. It has been established that the endocrine glands morphometric parameters in bulls brought up at both lowered and raised temperature are more relief under combined effect of Suvar and DAFS-25 than Suvar and Polistim.  相似文献

The regional distribution of steroid and drug metabolism was studied in intact cells and microsomal fractions obtained from the chromatically distinct inner (zona reticularis) and outer (zona fasciculata plus zona glomerulosa) zones of the guinea pig adrenal cortex. Cells isolated from the outer cortical zone produced far more cortisol than cells from the inner zone and cortisol production was stimulated by adrenocorticotropic hormone only in cells from the outer zone. Among the factors which may contribute to the greater cortisol production by the outer zone are a higher rate of 17 alpha-hydroxylation and ratio of 17 alpha- to 21-hydroxylase activities in that zone, both of which favor cortisol synthesis. In contrast, steroid 21-hydroxylase activity was far greater than 17 alpha-hydroxylase activity in microsomes obtained from the inner zone of the adrenal cortex. Microsomal metabolism of various xenobiotics such as benzo(a)pyrene and ethylmorphine proceeded far more rapidly in the inner than outer cortical zone. The zonal differences in metabolism appeared to result in part from differences in the ability of xenobiotics to interact with microsomal cytochromes P-450 in the two zones. The results indicate that the inner zone has a minor role in cortisol production by the adrenal cortex, but its involvement in the production of other steroids cannot be excluded. In contrast, the inner zone appears to have the major role in the metabolism of at least some xenobiotics which may account for its greater vulnerability to the toxic effects of chemicals requiring metabolic activation.  相似文献

Injury of individual nuclei of the anterior and the middle portions of the hypothalamus in rabbits by electrocoagulation through the preliminarily implanted electrodes was accompanied by the change in the morpho-functional condition of the hypothalamo-hypophysial neurosecretory system (HHNS). The nearer the focus of injury to the region of the supraoptic nuclei -- the greater the functional activity of the HHNS. In combining the injuries of individual hypothalamic nuclei with the atherogenic diet there occurred marked morpho-functional changes both in the HHNS and in the adrenal glands; as to animals with experimental atherosclerosis, it acquired under these conditions a more severe form than in the animals on the atherogenic diet alone. Results of investigations pointed to the presence of a close association of the genesis of the atherosclerotic process with the neurogenic factors.  相似文献

The structure and ultrastructure of the following regions of the hypothalamo-hypophysial neurosecretory system (HHNS) in the population cycle of lemming were studied: supraoptic (SON), paraventricular (PVN) and arcuate nuclei (AN), the median eminence (ME) and posterior pituitary (PP). Being compared to the state of endocrine glands and gonads, and the level of reproduction stress in the population cycle of the lemming described earlier, the obtained data revealed a certain regularity. First, at the stages of decrease, increase and in the first period of the stage of population peak the activity of the SON-PP complex and AN is gradually increased. This coincides with intensified reproduction, intensified functioning of the thyroid gland and adrenal cortex. Second, while the population is decreasing, all the above functions become depressed, except that of the thyroid gland which, on the contrary, becomes active. In the second period of the stage of population peak, when reproduction stops abruptly, this regularity, however, does not hold, the activity of SON-PP complex and AN is sharply lowered, whereas that of PVN becomes very high. The structure of the thyroid damaged as a result of previous hyperfunction, is restored, and the adrenal cortex shows signs of hypercorticism. The present work is devoted to specificity of the HHNS functioning at the stage of population peak, referred to as crucial, and the pathways and mechanisms by which neurohormones produced by this system can affect the endocrine glands. In addition, the causes likely to lead to the mass death of animals at this stage, and biological significance of this fact are also given attention to.  相似文献

Steroidogenesis is not stimulated by ACTH in the inner zone of the guinea pig adrenal cortex; adenylate cyclase is normally stimulated. To further explore the lack of a steroidogenic response to ACTH in the inner zone, cAMP-dependent protein kinase activity and protein phosphorylation were examined in the outer and inner adrenocortical zones. To summarize: total cAMP-dependent protein kinase activity was 40% higher in the outer zone than in the inner zone; of the total cAMP-dependent protein kinase activity, cytosol contained 80% for the outer and 70% for the inner zone. In both zones only the type II isozyme was present. Qualitative and quantitative differences in protein phosphorylation were noted for the two zones.  相似文献

Body growth and circulating levels of hormones were assessed in young rats and rabbits exposed to a 50-Hz electric field of 50 kV/m. Eight-week-old male rats were exposed 8 h/day for 4 weeks and rabbits were exposed 16 h/day from the last 2 weeks of gestation to 6 weeks after birth. The body and the organ growth of exposed rats were not statistically different from those of sham-exposed controls. No important differences from controls were observed in plasma levels of corticosterone, TSH, ACTH, and T4 or in adrenal levels of epinephrine, norepinephrine, and corticosterone although T3 was slightly, but significantly, decreased. No large histological changes in the thyroid or adrenals were noted. In rabbits, organ and body weights of exposed animals were comparable to those of controls. Plasma levels of various hormones (ACTH, GH, T3, T4, corticosterone, cortisol), serum glucose, triglycerides, and cholesterol were not significantly altered. Adrenal content of cortisol was lower, however, in exposed rabbits. No histological changes of the thyroid or adrenal glands were observed.  相似文献

The activity of 3beta-OH-steroid dehydrogenase in the adrenal cortex of 21-day fetuses in control and 3 days after decapitation in utero was estimated quantitatively by microspectrophotometry. The enzyme activity calculated as per conditioned cell in the fascicular zone exceeded that in the forming glomerular zone. After hypophysectomy the activity of the 3beta-OH-steroid dehydrogenase decreased in both the fascicular and glomerular zones.  相似文献

Previous studies showed that apolipoprotein-E (apoE) mRNA is regulated in rat adrenal gland by treatments that alter adrenal gland cholesterol content and steroidogenesis. In the present study cell types expressing apoE mRNA were determined by in situ hybridizations using an [alpha-35S]UTP-labeled RNA probe. Autoradiographic grains were counted to compare apoE expression in adrenal glands from control and experimentally treated animals. In control adrenal gland, zona (z.) fasciculata and z. reticularis exhibited the highest level of apoE mRNA expression, with lower levels in z. glomerulosa and medulla. Dexamethasone (DEX) treatment selectively increased apoE mRNA 3-fold in outer z. fasciculata, but not in other adrenal zones. ApoE mRNA expression appeared to be lower in adrenal glands from 4-aminopyrazolopyrimidine-treated rats, in that differences among adrenal gland zones were abolished. DEX treatment increased adrenal gland cholesteryl ester and oil red O staining in z. fasciculata cells in which the apoE mRNA concentration was increased as well as in other cortical cells in which apoE mRNA was unchanged. Aminoglutethimide administration led to a large increase in oil red O staining throughout the cortex, including z. fasciculata, without affecting apoE mRNA expression. These data suggest that adrenal gland apoE mRNA expression is not closely coupled to cellular cholesterol concentrations. Increased apoE mRNA expression in z. fasciculata of DEX-treated animals suggests an inverse relationship between apoE mRNA concentration and the level of steroidogenesis. This result is consistent with the proposal that apoE may play a role in regulating the utilization of cholesterol for steroid production.  相似文献
