The aim of study is to use erythrocyte ABO genotyping to assist serological typing in the identification of suspected ABO subtypes. Nine samples with discrepancies in ABO serological forward and reverse typing were subjected to ABO 6–7 exon sequencing, and their respective results were cisAB01O1, Bw12O2, Ael05B, Bel03O1, Ael05O1, B(A)04O1, B(A)02O1, and two cases of O1O2 weak antibody for the reverse typing test. The ABO genotyping and serological ABO forward and reverse typing were combined for subtype identification. In the results of ABO genotyping, cisAB and B(A)04 are only found B gene in genotyping, thus the presence of A subtype B was excluded. The samples with a weak antibody for serological O type in reverse typing were tested ABO genotyping to exclude subtype A or B. We summarized a combination of ABO genotyping and serological typing can be used for identification of suspected ABO blood groups.  相似文献

黄曲毒素B1(aflatoxinB1,简称AFB1)抗原的制备是AFB1免疫检测研究的第一步。研究了回流温度和时间对黄曲霉毒素B1肟(aflatoxinB1oxime,简称AFB1O)产生的影响,通过统计分析得到85℃,回流2hAFB1O的产率最高,为89%。在此基础上进一步研究了AFB1O与载体蛋白─-牛血清蛋白(BSA)反应的起始摩尔比对产物摩尔比的影响,随着反应起始摩尔比的增加,产物的摩尔比也稍有增加,但是增加幅度不显著,而AFB1O的利用率则随着起始摩尔比的增加而减少。选择20∶1为AFB1O与牛血清蛋白BSA的反应起始摩尔比,得到摩尔比为6.3∶1的AFB1O与BSA的连接物。  相似文献

目的:比较SHA.LIN评分和S.T.O.N.E评分对经皮肾镜取石术结石清除率的预测价值。方法:选择我院于2016年1月-2017年12月期间行经皮肾镜取石术患者67例为研究对象,在术前对所有患者进行SHA.LIN评分和S.T.O.N.E评分。根据手术结果将患者分为结石清除组(n=49)和结石残留组(n=18),对两组患者的一般资料、SHA.LIN评分、S.T.O.N.E评分进行统计对比。采用多因素Logistic回归分析方法分析患者术后结石残留的影响因素。采用绘制ROC曲线的方法分析SHA.LIN评分和S.T.O.N.E评分对结石清除率的预测结果的敏感性和特异性。结果:67例患者术后结石清除者49例、结石残留者18例,结石清除率为73.13%。结石残留组患者手术时间、术中失血量、住院时间、结石最大截面积、最大累积截面积、结石解剖分布肾盂的发生率、受累肾盏数均高于结石清除组,穿刺通道长度低于结石清除组(P0.05)。结石清除组患者的SHA.LIN评分、S.T.O.N.E评分均低于结石残留组(P0.05)。经多因素Logistic回归分析显示,手术时间、术中失血量、SHA.LIN评分、S.T.O.N.E评分、受累肾盏数是结石残留的影响因素(P0.05)。通过绘制ROC曲线可知,SHA.LIN评分的敏感性为91.25%、特异性为89.12%、曲线下面积(AUC)为0.912(95%CI 0.869~0.948);S.T.O.N.E评分的敏感性为78.75%、特异性为84.24%、AUC为0.782(95%CI 0.690~0.871)。结论:经皮肾镜取石术患者结石清除率与SHA.LIN评分、S.T.O.N.E评分明显相关,两种评分系统均能预测患者的结石清除率,但SHA.LIN评分的敏感性、特异性高于S.T.O.N.E评分。  相似文献

利用聚丙烯酰胺凝胶电泳技术,对采自泡沙参复合体整个分布区的15个天然居群进行了等位酶 分析。7种酶系统共10个等位酶基因位点的检测表明,本复合体植物具有高水平的遗传变异性,居群 水平上的遗传多样性指标分别为:多态位点百分率P=0.40~0.70,等位基因平均数A=1.5~2.5,平 均期望杂合度He=0.102~0.291。3个类群在居群水平上的平均多样性水平由高至低依次为;A. wawreana(A=2.10,P=0.63, He=0.237),A.lobophylla(A=1.85,P=0.60,He=0.204)和A. potaninii(A=1.83,P=O.60,He=0.188)。居群遗传结构和交配系统的分析表明,本复合体植物的大 部分遗传变异存在于居群之内,Fst=0.176(A.wawreana)和0.115(A.potaninii),并以异交为主,异 交率估计值分别为t=0.86(A.wawreana),0.74(A.potaninii)和0.63(A.lobophylla),这是本复合 体植物具有丰富遗传变异的主要原因之一。根据15个居群的遗传一致度(I)值进行聚类分析发现, 15 个居群可分为明显的2支,由A.lobophylla 2个居群构成的一支与由A. wawreana 和A.potaninii 13 个居群构成的一支在遗传上的分化十分明显,而6个A.wawreana 居群和7个A.potaninii 居群间的 遗传差异不明显,是两个分化不充分的类群。因此,本文的等位酶证据说明,A.lobophylla是遗传上独立的好种,同时不支持将A.wawreana 和A. potaninii 分别作为独立种的处理。  相似文献

目的:探究冰片提取物对沙眼衣原体感染后的He La细胞模型CT703和CT259表达的影响。方法:将成功建立的40例感染L2血清型沙眼衣原体的人宫颈癌上皮(He La)细胞模型随机分成A、B两组,分别添加冰片提取物和等剂量生理盐水。观察感染后He La细胞模型的包涵体数目及大小、RNA抽提结果完整性以及CT70与CT259的表达变化。结果:染色后的40例受沙眼衣原体感染的He La细胞模型体内均发现包涵体,在同一时间点B组细胞内包涵体比A组大,且数目比A组多(P0.05);两组提取的总RNA的OD值均在1.8~2.0之间,通过RNA凝胶电泳结果可清楚发现28S、18S及5S条带;同一时间点CT259和CT703扩增产物的平均灰度值比较,A组感染后He La细胞模型样本基因表达量低于B组,差异具有统计学意义(P0.05)。结论:冰片提取物能够有效降低经沙眼衣原体感染的He La细胞模型中CT703与CT259基因表达量。  相似文献
