In the present study, Bacillus subtilis Ch9 was evaluated as a probiotic in grass carp, Ctenopharyngodon idella (Valenciennes, 1844). For 56 days the grass carp (50 ± 2.5 g) were given a feed containing B. subtilis Ch9 in three concentrations: 1.0 × 109 (T1), 3.0 × 109 (T2) and 5.0 × 109 (T3) CFU kg?1 feed in triplicate treatments. The control group (T0) was given feed without B. subtilis Ch9 for the same period. Determined were the specific growth rate (SGR), feed conversion ratio (FCR), and digestive enzyme activities in the intestine and hepatopancreas as well as the intestinal microflora. After 56 days, fish receiving the diets supplemented with B. subtilis Ch9 showed significantly higher SGR and lower FCR (P < 0.05) than those fed the control diet. There was no significant different in SGR and FCR among T1, T2 and T3 nor was the survival rate affected (P > 0.05) by the dietary treatments. From days 14 to 56 of the experiment, higher protease, amylase and lipase activities in the foregut, midgut hindgut and hepatopancreas were observed in T1, T2 and T3 (P < 0.05) compared with the control over a short‐term (14–28 days). Enzyme activity did not increase after long‐term feeding with B. subtilis Ch9 (56 days), but was still higher than that of control fish (P < 0.05). Fish fed the probiotic had an increase in trend of total aerobic and facultative anaerobic bacterial quantity (P > 0.05), but the ratio of Bacillus was significantly higher (P < 0.05) than in control fish. The total anaerobic bacterial quantity, Bifidobacterium and Lactobacillus were significantly higher (P < 0.05) in fish fed B. subtilis Ch9 compared with fish fed control feed. In conclusion, an optimum dose of B. subtilis Ch9 could modulate intestinal microflora, induce digestive enzyme activity and potentially promote the digestion and absorption of nutrients, as well as improve the growth performance of grass carp significantly.  相似文献

Eight feeding trials using 708 cross-bred pigs were conducted to evaluate the addition of virginiamycin and a commercially-available lactobacillus fermention product (probiotic) to starter, grower and finisher diets based on maize and soya bean meal. Virginiamycin was added at a level of 11 mg/kg of diet, and lactobacillus probiotic at 1 g/kg in starter diets and 500 mg/kg in grower and finisher diets. In the combined analysis of four starter trials, pigs fed on diets containing virginiamycin tended to eat more and grow faster than pigs fed on the control diet, but feed efficiency was not different. Furthermore, daily gain, daily feed intake and feed efficiency were not improved by the addition of lactobacillus probiotic to starter diets. In the combined analysis of a starter—grower—finisher and a grower—finisher trial, virginiamycin supplementation did not alter overall pig performance compared with control. Pigs fed on a positive control diet containing chlortetracycline, penicillin and sulfamethazine had a nonsignificantly greater daily gain and feed intake.Daily gain for the lactobacillus probiotic-fed pigs was lower (P < 0.05) than that of the negative controls, but daily feed intake and feed efficiency were not significantly different. In three growing—finishing trials in which virginiamycin and lactobacillus probiotic were compared in a 2 × 2 factorial arrangement of treatments, daily gain and feed intakes were greater (P < 0.01) for pigs given virginiamycin. The lactobacillus probiotic did not improve daily gain, feed intake or feed efficiency. Overall, no interaction between virginiamycin and lactobacillus probiotic occurred.  相似文献

The wild-type Bacillus subtilis strain 8130 secreted metabolites that stimulated two to three times the growth of the test cultures of lactic acid bacteria. It exhibited endoglucanase activity that depended on the composition of nutrient medium. The addition of the product of two-stage culturing of B. subtilis 8130 to the diet of pigs (0.2% of fodder weight) made it possible to increase the daily weight gain by 19% and decrease the consumption of mixed fodder by 10%. Digestion of protein, fat, and other organic compounds increased by 3–4% and cellulose by 12%. It was shown that B. subtilis 8130 is a probiotic with targeted action stimulating digestion (primarily the digestion of cellulose). The enrichment of a dry-beer pellet with the product of solid-phase fermentation by bacillus (1 × 108 cells per gram dry pellet) allowed the pellet to entered into the diet of a calf (6% of the weight of fodder with probiotic), causing additional weight gain by 12% and a 10% economy of fodder consumption.  相似文献

The objective of this study was to evaluate the effects of Lactobacillus plantarum MA2, an isolate from Chinese traditional Tibet kefir, on cholesterol-lowering and microflora of rat in vivo. Rats were fed on cholesterol-enriched experimental diet, supplemented with lyophilized L. plantarum MA2 powder, with a dose of 1011 cells/day per mice. The results showed that L. plantarum MA2 feeding significantly lowered serum total cholesterol, low-density lipoprotein cholesterol, and triglycerides level, while there was no change in high-density lipoprotein cholesterol. In addition, liver total cholesterol and triglycerides was also decreased. However, fecal cholesterol and triglycerides was increased significantly (P < 0.05) in comparison with the control. Also, L. plantarum MA2 increased the population of lactic acid bacteria and bifidobacteria in the fecal, but it did not change the number of Escherichia coli as compared to control. Moreover, pH, moisture, and organic acids in the fecal were also measured. The present results indicate the probiotic potential of the L. plantarum MA2 strain in hypocholesterolemic effect and also increasing the probiotic count in the intestine.  相似文献

Summary A DNA segment covering the signal sequence coding region, the ribosome binding site, and the promoter of the staphylokinase (sak) 42D gene (Behnke and Gerlach 1987) was cloned into pUC19 to form a portable expression-secretion unit (ESU). Fusion of human interferon α1 (hIFNα1) and hybrid hIFNα1/2 genes to thissak ESU resulted in secretory expression of the two gene products in bothEscherichia coli andBacillus subtilis. While most of the IFNα was exported to the periplasmic space ofE. coli, about 99% was secreted to the culture medium by recombinantB. subtilis strains. The total yield inE. coli was 1.2×105 IU/ml. This level of expression and export led to instability of the recombinant strains that was spontaneously relieved in vivo by inactivation of thesak ESU through insertion of an IS1 element. No such instability was observed withB. subtilis although expression and secretion levels reached even 3×106 IU/ml. Proteolytic degradation of IFNα by extracellular proteases was avoided by a combination of constitutive expression and secretion during the logarithmic growth phase and the use of exoprotease-reduced host strains. The IFNα1 protein purified fromB. subtilis culture supernatant was correctly processed, carried the expected 11 amino acid N-terminal elongation that resulted from DNA manipulations and proved to be homogenous in Western blotting experiments. The same recombinant plasmid that directed efficient secretion of hIFNα1 inB. subtilis gave poor yields when introduced intoStreptococcus sanguis.  相似文献

In order to develop a multi-microbe probiotic preparation of Lactobacillus reuteri G8-5 and Bacillus subtilis MA139 in solid-state fermentation, a series of parameters were optimized sequentially in shake flask culture. The effect of supplementation of B. subtilis MA139 as starters on the viability of L. reuteri G8-5 was also explored. The results showed that the optimized process was as follows: water content, 50 %; initial pH of diluted molasses, 6.5; inocula volume, 2 %; flask dry contents, 30~35 g/250 g without sterilization; and fermentation time, 2 days. The multi-microbial preparations finally provided the maximum concentration of Lactobacillus of about 9.01?±?0.15 log CFU/g and spores of Bacillus of about 10.30?±?0.08 log CFU/g. Compared with pure fermentation of L. reuteri G8-5, significantly high viable cells, low value of pH, and reducing sugar in solid substrates were achieved in mixed fermentation in the presence of B. subtilis MA139 (P?<?0.05). Meanwhile, the mixed fermentation showed the significantly higher antimicrobial activity against E. coli K88 (P?<?0.05). Based on the overall results, the optimized process enhanced the production of multi-microbe probiotics in solid-state fermentation with low cost. Moreover, the viability of L. reuteri G8-5 could be significantly enhanced in the presence of B. subtilis MA139 in solid-state fermentation, which favored the production of probiotics for animal use.  相似文献

This trial was conducted to study the effects of different Bacillus subtilis concentrations on immune response of broiler chickens. There were 5 treatment groups: control, with no added B. subtilis supplementation, and 4 treatment groups receiving feed supplemented with different concentrations of B. subtilis. The trial was conducted with 225 broilers. The weight of broiler chickens in all groups receiving feed supplemented with B. subtilis was significantly higher and the feed conversion was better independently of the concentration than that of the control chickens. The degree of diffuse lymphohistiocytic infiltration and the number of solitary lymphoid follicles in the mucosa increased in accordance with the B. subtilis concentration of the feed. The birds from the groups fed B. subtilis-supplemented diets had significantly increased antibody responses to vaccination against Newcastle disease virus. The appearance of increased diffuse lymphohistiocytic infiltration and solitary lymphoid follicles in the mucosa and a stronger response to NDV indicate increased immunological response in chickens fed with a B. subtilis-supplemented diet.  相似文献

A urinary tract infection (UTI) is a multi-factorial disease including cystitis, pyelonephritis, and pyelitis. After Escherichia coli, Proteus mirabilis is the most common UTI-associated opportunistic pathogen. Antibiotic resistance of bacteria and infection recurrence can be connected to biofilm formation by P. mirabilis. In this study, human and sheep isolates of P. mirabilis were investigated for antibiotic sensitivity using an antibiotic disk test. Co-aggregation of the tested potential probiotic bacilli, Bacillus amyloliquefaciens B-1895 and Bacillus subtilis KATMIRA1933, with the isolated pathogen was also evaluated. Then, the anti-biofilm activity of naturally derived metabolites, such as subtilin and subtilosin, in the bacilli-free supernatants was assessed against biofilms of P. mirabilis isolates. The isolated pathogens were sensitive to 30 μg of amikacin and 5 μg of ciprofloxacin but resistant to other tested antibiotics. After 24 h, auto-aggregation of B. amyloliquefaciens B-1895 was at 89.5% and higher than auto-aggregation of B. subtilis KATMIRA1933 (59.5%). B. amyloliquefaciens B-1895 strongly co-aggregated with P. mirabilis isolates from human UTIs. Cell-free supernatants of B. amyloliquefaciens B-1895 and B. subtilis KATMIRA1933 showed higher antimicrobial activity against biofilms of P. mirabilis isolated from humans as compared with biofilms of sheep isolates. According to our knowledge, this is the first report evaluating the anti-biofilm activity of probiotic spore-forming bacilli against clinical and animal UTI isolates of P. mirabilis. Further studies are recommended to investigate the anti-biofilm activity and the mode of action for the antimicrobial substances produced by these bacilli, subtilosin and subtilin.

The present study investigated the effect of enriched Artemia with Bacillus subtilis on growth performance, reproductive factors, proximate composition, intestinal microflora, and resistance to Aeromonas hydrophila of ornamental fish, Poecilia latipinna. Using a completely randomized design, the experiment included three groups. The first group was fed with commercial food without any probiotic. The second group was fed with unenriched Artemia, and the last group consumed long-time enriched Artemia with Bacillus subtilis. The bacteria B. subtilis with a density of 1 × 105 CFU mL−1 was added daily to Artemia culture medium. The total microflora and Bacillus subtilis counts were significantly increased in enriched Artemia compared to the unenriched group (P < 0.05). In fish fed groups, growth factors did not show any significant difference (P > 0.05). The maximum relative fecundity (28.65 ± 2.52 egg number g−1), fry production (62.93 ± 4.6 individual per female), and fry survival (70.97 ± 1.56%) obtained in the third group were found to be significantly more than those in the first and the second groups. Moreover, intestinal bacterial count for Bacillus revealed that the higher concentration of bacteria was significantly related to the third group (6.24 ± 0.11 log CFU g−1) (P < 0.05). Maximum protein and fat contents were observed in fish fed with Bacillus-enriched Artemia; however, no significant difference was found between control and unenriched Artemia groups (P > 0.05). The highest amount of ash was observed in fish fed with commercial food without any probiotic (P < 0.05). At the end of the feeding period, each of the three groups along with positive group (oxytetracycline 100 mg kg−1 of commercial food) was exposed to A. hydrophila (BCCM5/LMG3770) bacteria intraperitoneally. Based on the results, the lowest cumulative mortality was significantly found in group three (68.75 ± 3.6%) and positive group (62.5 ± 7.0%) compared to control and unenriched Artemia groups (P < 0.05). Hence, B. subtilis with a concentration of 1 × 105 CFU mL−1 during the period of Artemia culturing can improve the reproductive parameters, intestinal microflora, and resistance to pathogenic bacteria of Poecilia latipinna.

The purpose of our work was to observe the influence of probiotic preparation BioPlus 2B on average daily gains of weaned pigs, feed efficiency and to evaluate some metabolic indices. The weaned pigs, at the age of 42 days, were included into the trial and divided into four groups. Pigs in groups A (n = 5) and B (n = 4) received BioPlus 2B also before weaning. Only group A received BioPlus 2B, at the concentration of 3.2 × 109 CFU per kg of feed, after weaning continually. Groups C (n = 5) and D (n = 4) did not receive BioPlus 2B until the start of the trial, but group C was given BioPlus 2B at the same concentration as group A during the experiment. Blood samples for determination of metabolic indices were collected at the start of experiment, i.e. on 42nd day of pigs life, and then on 56th, 70th, 84th and 91st days of pigs life. The following biochemical indices were evaluated within the trial: serum levels of total proteins, albumin, urea, total cholesterol and total lipids. Total serum protein level (p < 0.0001) and serum albumin level (p = 0.0024) in groups A and B were significantly higher in comparison with groups C and D on day 56 of pigs life. Serum urea level in group D was significantly (p = 0.049) higher than in group A on 70th day of pigs life. Serum level of total cholesterol in group B on day 56 and 84 of pigs life was significantly (p = 0.0004) higher than in groups C and D. Total serum lipid level was significantly (p < 0.05) higher in B group compared to other groups on 56th, 70th, 84th days of pigs life. Average daily gains (ADG) in A group, even if non-significantly, were about 10% better than in groups B, C, D between 57th and 77th days of pigs life. ADG in groups A and B were 14% better in comparison with that in groups C and D between 78th and 91st days of pigs life (p = 0.036). The best feed efficiency in the trial was in group A, approximately 13%, 16% and 21% better than that in the groups B, C and D, repectively. Sixty-week-old Hy-Line brown laying hens were randomly divided into five groups and fed different diets over a period of 84 days. Experimental treatments included a basal diet (control); the basal diet supplemented with 1.0 × 106B. licheniformis yb-214245; the basal diet supplemented with 1.0 × 106B. subtilis yb-114246; a combination of both strains in a 2:1 ratio (6.6 × 105:3.3 × 105B. licheniformis yb-214245:B. subtilis yb-114246); and the latter, added with 5 mg/kg flavomycin. Basal diet supplementation with the combined Bacillus species improved egg-laying performance in aging hens significantly (P < 0.05). Eggshell strength improved significantly with this treatment, compared to the control or the antibiotic-supplemented groups (P < 0.05). The levels of total cholesterol, triglycerides, and very low-density lipoprotein cholesterol in egg yolk declined significantly more in the Bacillus-treated group than in the control or the antibiotic-supplemented groups (P < 0.01). Small intestinal morphology was better in the hens treated with the Bacillus combination than in the hens in the control group (P < 0.05). The total number of aerobic bacteria (Bacillus, Lactobacillus, and Bifidobacterium) in the cecum was significantly higher in all the Bacillus-supplemented hens than either in the control or the antibiotic-supplemented hens (P < 0.01); additionally, the number of E. coli and Salmonella was significantly lower than in the control group (P < 0.01). In conclusion, diet supplementation with the combination of Bacillus species used here for aging laying hens improved their growth performance, cecal bacterial composition, egg quality, and small intestine morphology.

Cloning and expression of the L-phenylalanine dehydrogenase (PheDH) gene from Bacillus sphaericus in B. subtilis was performed. It was ligated into the pHY300PLK shuttle vector and the resulting plasmid, pHYDH encoding polypeptide with molecular weight of 340 kDa, then transformed in B. subtilis ISW1214 and Escherichia coli JM109 competent cells for expression. Bacillus subtilis ISW1214/pHYDH only produced PheDH enzyme (4700 U/l). The recombinant PheDH was purified to near homogeneity as judged by SDS–polyacrylamide gel electrophoresis (M r 41000 Da) and the result was 40-fold with a yield of about 54%. Apparent K m values for L-phenylalanine (Phe), L-tyrosine and NAD+ were 0.24, 0.48 and 0.19 mM respectively. The optimum pH of the recombinant enzyme was 11 for the oxidative deamination, 10.2 for the reductive amination. The features of recombinant PheDH enzyme were comparable with the wild type PheDH protein.  相似文献

【背景】碱性蛋白酶是工业用酶中占比最大的酶类,广泛应用于清洁、食品、医疗等行业。近期研究发现碱性蛋白酶在生产生物活性肽方面有巨大潜力,这将进一步拓宽其在保健食品领域中的应用。【目的】利用枯草芽孢杆菌异源表达地衣芽孢杆菌来源的碱性蛋白酶SubC。【方法】通过筛选3种枯草芽孢杆菌宿主菌株(Bacillus subtilis 1A751、MA07、MA08)和6种信号肽(AmyE、AprE、NprE、Pel、YddT、YoqM),同时优化诱导剂浓度、发酵培养基和发酵时长,最终得到最优重组菌株MA08-AmyE-subCopt。【结果】重组菌株MA08-AmyE-subCopt的胞外酶活力为3.33×103 AU/mL,胞外蛋白分泌量为胞内可溶蛋白表达量的4倍,与携带野生型信号肽的对照组菌株WT相比,酶活提高了73.4%。【结论】异源碱性蛋白酶SubC在枯草芽孢杆菌中成功表达,为碱性蛋白酶SubC的表达和在保健食品领域的工业化应用提供了理论基础。  相似文献

Summary The illegitimate recombination between Staphylococcus aureus plasmids pE194 (or pGG20, the hybrid between pE194 and Escherichia coli plasmid pBR322) and pBD17 (plasmid pUB110 without HpaII C-fragment) was studied in Bacillus subtilis. Cointegrates were generated with the frequency of 1–3x10-8. Among 22 hybrids analysed 9 types of recombinants were found. Nucleotide sequences of all three parental plasmids were involved in intermolecular recombination. Nucleotide sequencing of recombinant DNA junctions revealed that in 8 cases recombination occurred between short homologous regions (9–15 bp). One recombinant was formed using nonhomologous sites. The similarity was demonstrated between nucleotide sequences of the recombination sites of two types of cointegrates and those used for pE194 integration into the B. subtilis chromosome. Possible mechanisms of illegitimate recombination are discussed.  相似文献

Probiotics have growth promoting effects even under periods of heat stress challenge. More information is needed to understand the mechanisms by which probiotics maintain the growth performance. The aim of this study was to evaluate the effect of a probiotic based on Bacillus subtilis bacteria on growth related mechanisms of broilers under heat stress conditions. Specifically, growth performance, skeletal bone characteristics, skeletal muscles size, intestinal villus-crypt structure, intestinal bacteria, growth hormone (GH), insulin-like growth factor-1 (IGF-1), cholesterol, and glucose. A total of 1200 one day old Ross 308 male broilers were randomly distributed into 4 treatments, with 12 replicates per treatment and 25 birds per replicate. A 2 × 2 factorial arrangement was used; the main factors were environmental temperature (thermoneutral or heat stress) and diet (control or control + B. subtilis; 3 × 107 cfu/kg of feed). From d 22 to 35 of age, birds were either exposed to thermoneutral conditions (21 °C) or chronic heat stress (30 °C). During the same period, each group was divided into 2 subgroups and fed either the control diet or the B. subtilis supplemented diet. The results demonstrated that B. subtilis had positive effects (P < 0.05) on the body weight gain, feed conversion ratio, villus height, crypt depth, villus surface area, absorptive epithelial cell area and viable counts of intestinal beneficial bacteria. B. subtilis increased (P < 0.05) serum GH, IGF-1 and maintain normal levels of cholesterol and glucose under heat stress conditions. In addition, broilers fed B. subtilis under heat stress conditions exhibited higher (P < 0.05) skeletal muscles size and improved (P < 0.05) tibia traits and lower (P < 0.05) abdominal fat pads deposition compared with the controls. B. subtilis had no effect on rectal temperature under thermoneutral or heat stress conditions. It is concluded that B. subtilis can be used as growth promoters in broilers, particularly during the periods of heat stress conditions.  相似文献

Fermented feed (FF) is widely applied to improve swine performance. However, the understandings of the effects of FF on the immune status and gut microbiota of lactating sows and whether probiotics are the effective composition of FF are still limited. The present study aimed to investigate the performance, immune status and gut microbiota of lactating sows fed with a basal diet supplemented with Bacillus subtilis and Enterococcus faecium co-fermented feed (FF), with the probiotic combination (PRO) of B. subtilis and E. faecium and control diet (CON) as controls. Compared with the CON group, FF group remarkably improved the average daily feed intake of sows and the weight gain of piglets, while significantly decreased the backfat loss, constipation rate of sows and diarrhoea incidence of piglets. The yield and quality of milk of sows in FF group were improved. Besides, faecal acetate and butyrate were promoted in FF group. Additionally, FF increased the level of IgG, IgM and IL-10 and decreased the concentration of TNF-α in serum. Furthermore, FF reduced the abundance of Enterobacteriaceae and increased the level of Lactobacillus and Succiniclasticum, which were remarkably associated with growth performance and serum immune parameters. Accordingly, microbial metabolic functions including DNA repair and recombination proteins, glycolysis and gluconeogenesis, mismatch repair and d -alanine metabolism were significantly upregulated, while amino acid metabolism was downregulated in FF group. Overall, the beneficial effects of FF were superior to PRO treatment. Altogether, administration of FF during lactation improved the performance and immune status, and modulated gut microbiota of sows. Probiotics are not the only one effective compound of FF.  相似文献

We have previously shown that galacto-rhamnogalacturonan fibers can be enzymatically extracted from potato pulp and that these fibers have potential for exerting a prebiotic effect in piglets. The spore-forming Bacillus species are widely used as probiotics in feed supplements for pigs. In this study, we evaluated the option for further functionalizing Bacillus feed supplements by selecting strains possessing the enzymes required for extraction of the potentially prebiotic fibers. We established that it would require production and secretion of pectin lyase and/or polygalacturonase but no or limited secretion of galactanase and β-galactosidase. By screening a library of 158 Bacillus species isolated from feces and soil, we demonstrated that especially strains of Bacillus amyloliquefaciens, Bacillus subtilis, and Bacillus mojavensis have the necessary enzyme profile and thus the capability to degrade polygalacturonan. Using an in vitro porcine gastrointestinal model system, we revealed that specifically strains of B. mojavensis were able to efficiently release galacto-rhamnogalacturonan from potato pulp under simulated gastrointestinal conditions. The work thus demonstrated the feasibility of producing prebiotic fibers via a feed containing Bacillus spores and potato pulp and identified candidates for future in vivo evaluation in piglets.

Effects of Bacillus subtilis E20 isolated from fermented soybean on immune parameters and the disease resistance of the white shrimp (Litopenaeus vannamei) after 98 days of B. subtilis E20 feeding were evaluated in this study. Shrimp fed B. subtilis E20-containing diets at concentrations of 106 (E206), 107 (E207), and 108 (E208) cfu kg?1, respectively, had significantly increased survival rates of 13.3%, 16.7%, and 20%, compared to the control (fed no probiotic) after being challenged with Vibrio alginolyticus. There were no significant differences in the total hemocyte count, respiratory burst, or superoxide dismutase glutathione peroxidase among all treatments. Shrimp fed a higher concentration of the probiotic (E208) exhibited significant increases in phenoloxidase activity, phagocytic activity, and clearance efficiency compared to control shrimp. In addition, B. subtilis E20 showed a weaker inhibitory effect against the growth of Aeromona hydrophila with around a 0.3-cm inhibitory zone, but showed no inhibitory effects against other selected pathogens, such as white shrimp pathogens: V. alginolyticus and Vibrio vulnificus. These results suggest that the increased resistance of shrimp after B. subtilis E20 consumption occurs through immune modifications, such as increases in phenoloxidase activity, phagocytic activity, and clearance efficiency against V. alginolyticus.  相似文献

Summary The effects of the restriction system of Bacillus subtilis strain M on plasmid transformation were studied. Plasmid pHV1401 DNA prepared from B. subtilis transformed the restriction-proficient M strain 100 times more efficiently than the DNA prepared from Escherichia coli, while the two DNA preparations transformed restriction-deficient derivatives of that strain with similar efficiencies. This indicates that transformation with pHV1401 is sensitive to the M restriction system. pHV1401 contains three CTCGAG (XhoI sites). Successive removal of these abolished the effect of restriction. The study aimed to evaluate the effects of early antibiotic intervention (EAI) on bacterial fermentation patterns and mucosal immune markers in the colon of pigs with different protein level diets. Eighteen litters of piglets at day (d) 7 were fed creep feed without or with growth promoting antibiotics until d 42. At d 42, pigs within each group were further randomly assigned to a normal- or low-crude protein (CP) diet. At d 77 and d 120, five pigs per group were slaughtered for analyzing colonic bacteria, metabolites, and mucosal gene expressions. Results showed that low-CP diet increased propionate and butyrate concentrations at d 77 but reduced ammonia and phenol concentrations (P < 0.05). EAI increased p-cresol and indole concentrations under normal-CP diet at d 77 (P < 0.05). Low-CP diet significantly affected (P < 0.05) some bacteria groups (Firmicutes, Clostridium cluster IV, Clostridium cluster XIVa, Escherichia coli, and Lactobacillus), but EAI showed limited effects. Low-CP diet down-regulated gene expressions of pro-inflammatory cytokines, toll-like receptor (TLR4), myeloid differentiating factor 88 (MyD88), and nuclear factor-κB p65 (NF-κB p65) (P < 0.05). EAI up-regulated mRNA expressions of interleukin-8 (IL-8) and interferon-γ (IFN-γ) under normal-CP diet at d 77 (P < 0.05). Furthermore, reductions of E. coli and ammonia under low-CP diet were positively correlated with down-regulated gene expressions of pro-inflammatory cytokines, which were positively correlated with the down-regulated TLR4-MyD88-NF-κB signaling pathway. In conclusion, EAI had short-term effects under normal-CP diet with increased aromatic amino acid fermentation and gene expressions of pro-inflammatory cytokines. Low-CP diet markedly reduced protein fermentation, modified microbial communities, and down-regulated gene expressions of pro-inflammatory cytokines possibly via down-regulating TLR4-MyD88-NF-κB signaling pathway.
