Acyl-CoA: phospholipid acyl-transferase activity as well as phospholipase A activity were detected in inner and outer membrane preparations from rat liver mitochondria. Both enzyme systems have an optimum pH around 8 and act preferentially on phosphatidylethanolamine. While phospholipase A activity is much lower in the inner membrane than in the outer membrane of mitochondria the reverse is true for the incorporation of (14C)-oleic acid into endogenous phosphatidylethanolamine. These results bring an indirect evidence that the inner membrane per se possesses a phospholipase A activity.  相似文献

1. The mitochondria isolated from human or rat liver were fractionated into submitochondrial particles and purified inner and outer membrane. According to different marker enzymes the inner membranes were enriched about 5-6-fold and the outer membranes about 12-14-fold. The electron microscopical appearance of the membranes was that expected on the basis of enzymic characterization. 2. A comparison of the average amino acid composition of the membrane proteins from the two types of mitochondria has been made. In the case of submitochondrial particles there were statistically significant differences between the human and rat hydrolysates for only five amino acids. Analysing the purified mitochondrial membranes there were significant differences between the two species for nine amino acids in the case of outer membranes and for 12 amino acids in the case of inner membranes. 3. With one exception all amino acids that were increased or decreased in the outer membrane exhibited a similar trend in the inner membrane of human compared with rat liver mitochondria. It appears that liver mitochondrial membranes have a species-dependent pattern of amino acid composition of their proteins.  相似文献

Experiments on dissociation and reassociation of outer and inner membranes of cauliflower (Brassica oleracea L.) mitochondria have been carried out in order to investigate the possible occurrence of an intermembrane electron transfer. With NADH as electron donor, it has been shown that electron transfer can take place between an antimycin-insensitive NADH-cytochrome c reductase, on the outer membrane, and the cyanide-sensitive cytochrome oxidase, on the inner membrane, provided a mobile carrier such as cytochrome c is present in the intermembrane space. In intact plant mitochondria, part of the oxidation of exogenous NADH could be carried out by this pathway.  相似文献

The incorporation of radioactivity derived from [2-14C] riboflavin into the flavins of rat liver mitochondrial outer membranes was studied. These membranes were found to contain about 0.6 nmol of non-covalently bound flavins per mg protein; the majority is in the form of FAD (73%) and FMN (24%). The membranes also contain about 1.5 nmol per mg of covalently bound flavins.After labeling, radioactive flavins appeared in the non-covalently bound flavins for about 4 h. Most of this radioactivity was in FAD (77%). Neither the rate nor extent of this labelling was affected by cycloheximide (1 mg/kg) administered 30 min prior to the radioactive riboflavin. With the covalently bound flavins, radioactivity was incorporated into the coenzymes for at least 18 h, but the rate of incorporation was much slower. After cycloheximide, radioactive flavins continued to appear in covalently bound flavins for about 2 h, but then stopped. Labeling of both types of flavins after [14C] riboflavin was considerably slower than the incorporation of [3H] leucine into outer membrane proteins. These results suggest that with flavoproteins from the mitochondrial outer membranes, the incorporation of flavins occurs after synthesis of the various apoenyzmes is complete.  相似文献

The outer membranes of mitochondria prepared from the liver of rats kept 12 days on a choline-deficient diet were analyzed for changes in phospholipid and protein content. The total amount of phospholipid in the outer membranes was not affected by the deficiency. There was, however, a significant decrease in the amount of phosphatidylcholine and an increase in phosphatidylethanolamine. The alterations in the membrane phospholipids were reflected in a reduction in the fluorescence of the membrane probe, 8-anilino-1-naphthalene sulfonate. Choline deficiency also affected the protein composition of the outer membranes as judged by electrophoretic analysis; however, the activity of several enzymes which serve as markers for the outer membrane was not affected by the deficiency.  相似文献

1. The amount and types of phospholipid and the fatty acid composition of the various phospholipids were examined in intact rat liver mitochondria, in mitochondria devoid of their outer membrane (preparation A) and in very small pieces derived from the disruption of the inner-membrane complexes (preparation B). The latter two preparations were obtained by digitonin treatment and carry out oxidative phosphorylation. 2. The ratio mug.atoms of phospholipid P/mg. of protein was 0.163 for intact mitochondria, decreased to 0.118 on removal of the outer membrane and increased markedly to 0.292 on disruption of the inner-membrane complex. 3. Examination of the various types of phospholipid present showed that the molar proportions cardiolipin:phosphatidylcholine:phosphatidylethanolamine were approx. 1:6:6 for intact mitochondria and 1:3:3 for preparations A and B. 4. There was a correlation between the recovery of cardiolipin and adenosine triphosphatase activity in the conversion of intact mitochondria into preparations A and B. 5. The fatty acid contents of the various types of phospholipid purified by thin-layer chromatography were identical in all three preparations. Our results show a considerably higher content of arachidonic acid and lower content of oleic acid for phosphatidylcholine, phosphatidylethanolamine and phosphatidylinositol than have previously been reported for mitochondrial phospholipids.  相似文献

1. Rat liver mitochondria were examined in the electron microscope by using negative staining in the presence of 0·3m-sucrose. The intact outer membrane does not appear to be freely permeable to the stain. Where the stain penetrated through a tear it was seen that the inner membrane had randomly oriented grooves, many of which contained round structures varying between 200 and 900å in diameter. Laminar structures containing two to five layers of approx. 50å each were found at the periphery. 2. When the outer membrane was removed by treating the mitochondria with digitonin several types of inner-membrane complexes were formed and they showed a general correlation with those observed in sectioned samples of the same preparations. The main types were: (a) a condensed form looking very much like the intact mitochondrion without the outer membrane (this still showed the grooves, some of which contained the round structures, and the laminar whirls at the edges); (b) a more transparent form containing tubules of uniform width and various lengths (some of these appeared to terminate in a hole at the surface of the inner membrane); (c) a large torn sac, probably the inner membrane, containing some tubules and vesicles. 3. When the inner-membrane complex was further treated with digitonin it was disrupted and the resulting material consisted of pieces of membrane, doughnut-shaped units and lamellar structures. Most of these pieces varied in size between 500 and 1000å.  相似文献

The reaction of gamma-glutamyltranspeptidase with phenobarbital or with thiobarbituric acid resulted in a irreversible loss of its enzymatic activity. The inactivation followed pseudo-first-order kinetics. Half-maximal velocity of inactivation (Ki) at 37 degrees C in the presence of phenobarbital or thiobarbituric acid was calculated to be 43 mM and 20 mM, respectively. The inactivation of the enzyme activity by both these inhibitors was prevented by serine borate, a known competitive inhibitor, and by the substrate, reduced glutathione, suggesting an active-site-directed nature of the these inhibitors. Maleate provided slight protection against inactivation by thiobarbituric acid. Complete inactivation of the enzyme with tritium-labeled phenobarbital resulted in a stoichiometric incorporation of radioactivity into the enzyme protein. Upon sodium dodecyl sulfate polyacrylamide gel electrophoresis of tritium-labeled phenobarbital-enzyme complex, nearly all the radioactivity was found to be associated with the small subunit (Mr = 22 000) of the enzyme, indicating that the catalytic component of the enzyme is on the small subunits.  相似文献
