Summary The Limulus ommatidium consists of 4 to 20 retinula cells surrounding the dendrite of the eccentric cell. Adjoining membranes are differentiated into the microvillous rhabdome in the central area of the ommatidium. Three types of pigment cells envelop the sensory cells. The distal pigment cells cover the periphery of the distal half of the ommatidium; proximal pigment cells (beneath the base of the ommatidium) and intraommatidial pigment cells provide glial wrapping for the sensory cells, the partitions between them, and the peripheral loose framework. Processes of the overlying cone cells penetrate into the ommatidium and lie at the edges of the rhabdomal fins. Numerous neurosecretory axons terminate at all levels of the ommatidium on pigment cells, conveyed there either by enveloping pigment cells or by separate neuroglial cells. Tight junctions in the ommatidium are confined to the contacts between rhabdomal miorovilli. The periphery of the rhabdome is surrounded by continuous adhering junctions except at the tip and exit of the eccentric cell dendrite. Summary Eight classes of organic sulfur compounds, comprising 42 substances, were tested in vitro for activity against the yeast phase ofHistoplasma capsulatum Darling. Activity was found in the classes of thiols, thio acids, disulfides, and thiolsulfonates. The classes of sulfinates, sulfenamides, penicillamine analogs, and reaction products of thiols with aldehydes or ketones were unpromising. A deoxyribonuclease was partially purified from the free-living nematodeCaenorhabditis elegans. The DNase functioned as an endonuclease and introduced both single-strand nicks and double-strand breaks into DNA. The enzyme hydrolyzed double-stranded DNA seven times more rapidly than single-stranded DNA. DNase activity was not affected by the addition of divalent cations below 1mm but was inhibited at higher ionic concentrations. In addition, the enzyme was not inhibited in the presence of 10mm EDTA. The enzyme was inhibited by salt concentrations greater than 20mm. Three independent mutations in thenuc-1 gene were shown to reduce nuclease activity to less than 1% of that seen in wild-type organisms. Zusammenfassung Die Doppelbrechung der Cornealinse und der Rhabdomere im Facettenauge von Calliphora erythrocephala (MEIG.) wurde untersucht.Der Gangunterschied wurde in Schnitten parallel zur Ommatidienachse gemessen. Die Differenz der Brechungsindices — die Doppelbrechung — zwischen dem außerordentlichen und dem ordentlichen Strahl ist (n en o) = 0,0012. Die Cornealinse ist ein einachsig, negativ doppelbrechender Kristall. Die optische Achse verläuft parallel zur Ommenachse.Die Kristallkegel und die Rhabdomerenkappen sind isotrop. Die Rhabdomere selbst sind anisotrop. Der Gangunterschied in den Sehstäben 1–6 (50 nm) scheint größer zu sein als im siebenten Rhabdomer (18 nm). Die Rhabdomere der siebenten und achten Sehzelle liegen jedoch genau hintereinander in einer Achse und dieTubuli sind zueinander senkrecht orientiert. Polarisationsoptisch gesehen liegen die beiden Sehstäbe in Subtraktionsstellung. Die Doppelbrechung der Rhabdomere ist (n en o) = – 0,0004.
Investigations with polarized light on the eye of Calliphora erythrocephala (Meig.)
Summary The birefringency of the corneal lens and of the rhabdomeres in the compound eye of Calliphora erythrocephala (MEIG.) was investigated.The phase difference was measured in sections parallel to the axis of the ommatidium. The difference of the refractive indices — the birefringency — between the extraordinary and the ordinary beam is (n e – n o) = –0,0012. The corneal lens is a negative birefringent crystal. Its optical axis runs parallel to the axis of the ommatidium.The crystalline cones and the extracellular distal processes of the rhabdomeres are isotropic. The rhabdomeres are anisotropic. The phase difference along the rhabdomeres No. 1–6 (50 nm) seems to be higher than in the seventh (18 nm). As rhabdomere No. 8 is situated beneath rhabdomere No. 7 and the tubules of these two rhabdomeres are perpendicularly orientated, the phase differences are partially cancelled. The birefringency of the rhabdomeres is (n en o) = –0,0004.

Summary Using patch-clamp techniques, we have studied Ca2+-activated K+ channels in the basolateral membrane of freshly isolated epithelial cells from rabbit distal colon. Epithelial cell clusters were obtained from distal colon by gentle mechanical disruption of isolated crypts. Gigaohm seals were obtained on the basolateral surface of the cell clusters. At the resting potential (approximately –45 mV), with NaCl Ringer's bathing the cell, the predominant channels had a conductance of 131±25 pS. Channel activity depended on voltage as depolarization of the membrane increased the open probability. In excised inside-out patches, channels were found to be selective for K+ over Na+. Channel activity correlated directly with bath Ca2+ concentration in the excised patches. Channel currents were blocked by 5mm TEA+ and 1mm Ba2+. In cell-attached patches, after addition of the Ca2+ ionophore A23187, which increases intracellular Ca2+, open probability was markedly increased. Channel activity was also regulated by cAMP as addition of 1mm dibutyryl-cAMP in the bath solution in cell-attached patches increased channel open probability over 20-fold. Channels that had been activated by cAMP were further activated by Ca2+. We conclude that the basolateral membrane of epithelial cells from descending colon contains a class of potassium channels, which are regulated by intracellular Ca2+ and cAMP.  相似文献

Patch clamp techniques were used to record whole cell and single channel Na+ currents from NB41A3 neuroblastoma cells grown in culture. Cells were grown for two weeks in control medium or medium supplemented with 30 mm d-glucose of 30 mm l-fucose.Cells exposed to glucose or l-fucose had smaller whole cell Na+ currents than cells grown in unsupplemented medium, consistent with earlier studies (Yorek, Stefani & Wachtel, 1994). Whole cell macroscopic currents showed no change in activation or inactivation kinetics. Single channel current properties and opening probability were also unchanged.The number of [3H]saxitoxin binding sites, and therefore the total number of Na+ channels, was not reduced in cells grown in glucose or l-fucose (Yorek et al., 1994). Therefore, we conclude that some of the channels must have been rendered nonfunctional by the conditioning media. The finding that single channel properties are not altered suggests that channels become nonfunctional in an all-or-none manner.This work was supported by Merit Review Awards to M.A. Yorek and R.E. Summary The structure of Euglena granulata and Chlamydomonas eugametos has been studied using polarization and electron microscopy, cinematography, and chemical extraction procedures, with the main focus on the structure of the eyespot.The 50–60 granules which form the extrachloroplastic eyespot of E. granulata are large bodies, up to 1200 m in diameter. They are found in the cytoplasm near the base of the reservoir and are associated with the parabasal body which contains a large crystal. The eyespot granules are contained within membranes having a unit membrane structure; 2 or 3 are usually present in a single eyespot packet; microtubules are also contained within the packet. The eyespot granules have the structure of a positive spherite and clearly exhibit birefringence; this structure is modified by fixation.The granules of the chloroplastic eyespot of C. eugametos are about 75 m in diameter and are contained within the chloroplast in an ordered array. Occasionally, the eyespot contains elongate or helical bodies mixed with the granules. Extraction with organic solvents caused the removal of materials which formed the eyespot granules as well as that of the osmophilic globules in the chloroplasts.Several hypotheses which concern the function of the eyespots in these and other species are discussed in the light of our results. the possible origin and demise of the eyespot granules are also discussed.Supported in part by NSF Grant GB-313. High activity of carboxypeptidases was detected in the hepatopancreas of the crab Paralithodes camtschatica, while aminopeptidase activity in this tissue was low. Two crab carboxypeptidases were purified by chromatography on DEAE-cellulose, phenyl-Sepharose, and Sephadex G-75 to homogeneity. The molecular weight values of carboxypeptidases I and II were 40,000 and 37,000, respectively. The isoelectric point value for both carboxypeptidases was 4.5. The crab carboxypeptidases were activated by NaCl, with maximal activity of carboxypeptidases I and II at 1.0 M and 0.6 M NaCl, respectively. Using 19 N-blocked dipeptides with the general structures Bz-Gly-X and Z-Gly-X, broad substrate specificity of the purified enzymes was demonstrated. Under optimal conditions the values of K m and k cat for the hydrolysis of Bz-Gly-l-Phe, Bz-Gly-l-Arg, and Bz-Gly-l-Lys by crab carboxypeptidases were determined. Inhibition data led to classification of the crab enzymes as metallopeptidases. Both carboxypeptidases were stable under neutral and mildly alkaline conditions. In addition, the presence of 1 M NaCl decreased the thermostability of the crab carboxypeptidases. Received August 13, 1999; accepted November 19, 1999.  相似文献

Interactions between the reactive disulfide fungal metabolite, gliotoxin (GTX), and rabbit skeletal ryanodine receptor (RyR) calcium release channels have been examined. RyRs in terminal cisternae vesicles formed a covalent complex with 100 μm 35S-GTX, which was reversed by 1 mm dithiothreitol (DTT) or 1 mm glutathione. GTX (80–240 μm), added to either cytoplasmic (cis) or luminal (trans) solutions, increased the rate of Ca2+ release from SR vesicles and the frequency of opening of single RyR channels in lipid bilayers. Channel activation was reversed upon addition of 2 mm DTT to the cis solution, showing that the activation was due to an oxidation reaction (2 mm DTT added to the cis solution in the absence of GTX did not affect RyR activity). Furthermore, RyRs were not activated by trans GTX if the cis chamber contained DTT, suggesting that GTX oxidized a site in or near the membrane. In contrast to cis DTT, 2 mm DTT in the trans solution increased RyR activity when added either alone or with 200 μm trans GTX. The results suggest that (i) GTX increases RyR channel activity by oxidizing cysteine residues that are close to the membrane and located on RyR, or associated proteins, and (ii) a disulfide bridge or nitrosothiol, accessible only from the luminal solution, normally suppresses RyR channel activity. Some of the actions of GTX in altering Ca2+ homeostatsis might depend on its modification of RyR calcium channels. Received: 12 November 1999/Revised: 14 March 2000  相似文献

In humans, the quantity of α-l-fucosidase in serum is determined by heredity. The mechanism controlling levels of the enzyme in serum is unknown. Lymphoid cell lines derived from individuals with either low, intermediate, or high α-l-fucosidase in serum were established. Steady-state levels of intracellular and extracellular α-l-fucosidase as well as rates of synthesis and secretion of enzyme overlapped among the cell lines. Thus,vivo} serum phenotypes were not expressed in this system. No appreciable differences in the qualitative processing of newly made α-l-fucosidase were observed among these lymphoid cell lines. Cells pulse-labeled with35S-methionine from 0.25 to 2 hr had an intracellular form of enzyme with aM r=58,000. Cells pulsed for 1.5 hr and chased for 21 hr with unlabeled methionine had an intracellular form ofM r=60,000 and an extracellular form ofM r=62,000. All three enzyme forms were glycoproteins with a common polypeptide chain ofM r=52,000 but with different carbohydrate moieties. No evidence for a high molecular mass precursor form of α-l-fucosidase was found. Fucosidosis is a rare, inherited disease in which α-l-fucosidase activity in tissues and body fluids is low or absent. The mutations for fucosidosis and the serum polymorphism map separately. Lymphoid cells from two siblings with fucosidosis had 8-fold to 341-fold less intracellular α-l-fucosidase protein with 11-fold to 56-fold lower specific activities than control cells. Residual mutant enzyme was a glycoprotein with a polypeptide chain virtually the same size (M r=52,000) as control enzyme. However, residual mutant enzyme was hypoglycosylated and hypersecreted as compared to control enzyme. This research was supported by National Institutes of Health Grant DK 32161.  相似文献

Summary If an insect is able to determine the direction of polarization in any point of the sky, this ability does not in itself guarantee that the insect can orientate unambiguously. Such would only be the case, if every point in the sky had its own exclusive direction of polarization. In thee-vector pattern of the sky, however, each direction of polarization is found at many different points. For compass orientation the insect has therefore to use some information on the geometry of thee-vector pattern in the sky. In general, eache-vector occurs twice at a given elevation (Fig. 1). The angular separation between the positions of identicale-vectors depends on the elevations of thee-vectors above the horizon and on the height of the sun. Except at sunrise and sunset, 180°.If a bee is trained to fly in a certain direction to a food source, the direction of its waggle dance on a horizontal comb points directly towards the goal (provided that the bee is able to view the sky). However, if the bee is only allowed to view a singlee-vector in the sky (or a single artificially adjustede-vector), it should perform ambiguous orientation. One expects the bee to prefer two dance directions separated by the proper angular distance . One of these two dance directions should point at the food source.The bees indeed dance in two directions. However, there are two unexpected results: (1) The angular distance between the two preferred directions invariably amounts to =180°. (2) One of the preferred directions points closely, but not exactly at the goal. What one can deduce from these single-e-vector tests is that the bee uses a rather generalized internal representation of thee-vector pattern in the sky. This paper describes the generale-vector characteristic applied by a dancing bee that only views a singlee-vector in the sky (diameter of the celestial patch or the artificially polarized light source 10°). This generale-vector characteristic of the bee (Fig. 9) more closely fits the meane-vector distribution near the zenith thane-vector distributions in other parts of the sky (Fig. 11).This article is dedicated to Prof. Dr. H. Summary The cytological influence of light and dark adaptation (LA and DA) on the retinular cells of the spider crab Libinia emarginata has been studied by light and electron microscopy in four adaptive states: 17 hours darkness, 5 hours darkness, 5 hours diffuse light and 17 hours diffuse light. The rhabdom's fine structure is typical of decapods but its dual overall form and position mingle certain features of both apposition and superposition compound eye types. Distal and proximal retinal pigments both showed adaptive migration, but the distal pigment cells moved over a restricted range, and DA separated the retinular cell pigment granules into two groups, perinuclear and basilar.In the rhabdom no changes in its position, dimensions or microvillus fine structure were observed with LA or DA. But at the base of the rhabdom microvilli the rate of pinocytosis was strongly affected by the eye's adaptive state, being lowest after 17 hours DA and greatest after 17 hours LA; the wall of the 0.1 microvesicles so formed, looked like the membrane of the rhabdom microvillus and they were the same size as the vesicles in multivesicular bodies and in vesicular lamellar bodies.Three categories of complex cytoplasmic particles about 1 in diameter (multivesicular bodies, vesicular lamellar bodies and purely lamellar bodies) were all increased in number by decreased DA and by increased LA; similar quantitative effects occurred in the endoplasmic reticulum and in the ribosomes.The pinocytotic vesicles and the complex cytoplasmic bodies may represent part of an intracellular system to dispose of rhabdom metabolites whose production was initiated or increased by light absorption.Cytoplasmic and perirhabdomal vacuoles mainly distal in location, were also affected by light, but inversely; their maximal extent occurred after 17 hours DA; less DA or any LA significantly decreased their presence and aggregation.The data reported are of interest not only because they correlate retinal fine structure with the metabolism of vision but also because they provide a new and specific tool for distinguishing active from inactive neurosensory cells in the optic pathway.This research was initiated with the aid of U.S. Public Health Service Grant NB-03076 and has been continued with the support of U.S. Air Force Grant AFOSR-1064. Summary Action potentials in directional statocyst interneurons are measured during sinusoidal oscillation of the crabCarcinus around horizontal axes. Peak response of the interneurons precede peak position of the animal by 120 ° (Fig. 3). Optimum response of an interneuron occurs when the plane of oscillation of the crab coincides with the circumferential plane of the vertical canal of the statocyst providing input to the interneuron in question (Fig. 6). Directionality is explained in terms of unidirectional sensory receptors inCarcinus statocyst. Drones (Apis mellifera ) were attracted to a lure scented with queen pheromone suspended at a height of 11–18m. An unscented test object, usually a black sphere, was lowered on a vertical string at a horizontal distance of 50 cm or 103 cm from the scented lure. Drones often made a rapid, direct, displacement from the scented lure to the test object. Drones responded to objects that subtended an angle as small as 0.41° which is less than the acceptance angle of a single ommatidium. We calculate that a drone can respond to a stimulus corresponding to a reduction in the light incident on one ommatidium at a time of 8%; the brevity of the stimulus on each ommatidium would seem to be a factor in the design of the eye.  相似文献

Summary Specially in stress conditoin, some hyaline droplets occur in the perikarya of the neurosecretory hypothalamic cell of Zoarces viviparus. They first appear as filamentous macromolecular material in the intracisternal space, then tend to accumulate in the form of irregular electron dense rods or reticulum. Summary The present studies examined some of the properties of Cl channels in renal outer medullary membrane vesicles incorporated into planar lipid bilayers. The predominant channel was anion selective having aP Cl/P K ratio of 10 and a unit conductance of 93 pS in symmetric 320mm KCl. In asymmetric KCl solutions, theI-V relations conformed to the Goldman-Hodgkin-Katz equation. Channel activity was voltage-dependent with a gating charge of unity. This voltage dependence of channel activity may account, at least in part, for the striking voltage dependence of the basolateral membrane Cl conductance of isolated medullary thick ascending limb segments. The Cl channels incorporated into the planar bilayers were asymmetrical: thetrans surface was sensitive to changes in ionized Ca2+ concentrations and insensitive to reducing KCl concentrations to 10mm, while thecis side was insensitive to changes in ionized Ca2+ concentrations, but was inactivated by reducing KCl concentrations to 50mm.  相似文献

Summary Repetitive stimulation of human peripheral nerves in situ produces an amplitude oscillation of the evoked action potentials. The purpose of this experiment was to describe the dynamics of the response as a function of the characteristics of the electrical stimuli. Two attempts to define precisely the origin of the response have proved ineffective.These studies were supported in part by Grant MH 8786-01 from the National Institute of Health and in part by Grant GR 2000 from the National Science Foundation. Ionic currents responsible for the action potential in scorpion muscle fibers were characterized using a three-intracellular microelectrode voltage clamp applied at the fiber ends (8–12°C). Large calcium currents (I Ca) trigger contractile activation in physiological saline (5 mm Ca) but can be studied in the absence of contractile activation in a low Ca saline (2.5 mm). Barium (Ba) ions (1.5–3 mm) support inward current but not contractile activation.Ca conductance kinetics are fast (time constant of 3 msec at 0 mV) and very voltage dependent, with steady-state conductance increasing e-fold in approximately 4 mV. Half-activation occurs at –25 mV. Neither I Ca nor I Ba show rapid inactivation, but a slow, voltage-dependent inactivation eliminates I Ca at voltages positive to –40 mV. Kinetically, scorpion channels are more similar to L-type Ca channels in vertebrate cardiac muscle than to those in skeletal muscle.Outward K currents turn on more slowly and with a longer delay than do Ca currents, and K conductance rises less steeply with voltage (e-fold change in 10 mV; half-maximal level at 0 mV). Summary Water bugs (Notonecta glauca) were set into flight in a room with a homogeneously illuminated ceiling and a light-emitting platform on the floor. In these conditions polarized UV light from the platform was more effective in causing the animals to fly down to the surface of the platform than was unpolarized UV light several times as intense. Experiments with an array of baffles that restricted the directions from which the polarization film on the platform could be seen showed that the polarized UV light is effective in eliciting descent only when the e-vector is perpendicular to the median sagittal plane of the animal (horizontal). It can be concluded that polarized UV light with horizontal e-vector is distinguished, as a special sensory quality, from unpolarized UV light.Notonecta thus provides an example of true polarization vision.The special orthogonal arrangement of the microvilli in the rhabdomeres of the UV visual cells in the ventral part of the eye (cf. Schwind 1983 b and Schwind et al., in press) is suggestive with regard to polarization vision. The microvilli of the two UV visual cells in the ommatidia looking forward and down are horizontal and vertical, respectively, and hence could serve as a two-channel analyzer system capable of distinguishing the polarized UV light reflected by a water surface from unpolarized UV light.  相似文献
