Effects of endogenous carbon monoxide on collagen synthesis in pulmonary artery in rats under hypoxia 总被引:3,自引:0,他引:3
To study the role of endogenous carbon monoxide (CO) in collagen metabolism during hypoxic pulmonary vascular remodeling, a total of 18 Wistar rats were used in the study and they were randomly divided into three groups: hypoxia group (n = 6), hypoxia with zinc protoporphyrin-IX (ZnPP-IX) group (n = 6) and control group (n = 6). The measurement of mean pulmonary artery pressure (mPAP) and carboxyhemoglobin (HbCO) formation in lung tissue homogenates was measured. A morphometric analysis of pulmonary vessels was performed, in which the percentage of muscularized arteries (MA); partially muscularized arteries (PMA) and nonmuscularized arteries (NMV) in small and median pulmonary vessels, relative medial thickness (RMT) and relative medial area (RMA) of pulmonary arteries were analyzed. Collagen type I and III and transforming growth factor-beta3 (TGF-beta3) expressions were detected by immunohistochemical assay. The expressions of procollagen type I and III and TGF-beta3 mRNA were detected by in situ hybridization. The results showed that ZnPP-IX significantly increased mPAP and markedly decreased HbCO formation in lung tissue homogenates in rats under hypoxia (P < 0.01). In the hypoxia rats treated with ZnPP-IX, the percentage of muscularized arteries of small and median pulmonary vessels was obviously increased, and RMT and RMA of intra-acinar muscularized pulmonary arteries were markedly increased compared with hypoxic rats. Ultrastructural changes, such as hyperplasia and hypertrophy of endothelial cells (ECs) and smooth muscle cells (SMCs) and the increased number of SMCs in synthetic phenotype were found in intra-acinar pulmonary muscularized arteries of hypoxic rats treated with ZnPP-IX. Meanwhile, ZnPP-IX promoted the expression of collagen type I and III and TGF-beta3 protein in pulmonary arteries of rats under hypoxia (P < 0.01). Furthermore, ZnPP-IX elevated obviously the expressions of procollagen type I and III mRNA, and TGF-beta3 mRNA in pulmonary arteries of rats under hypoxia (P < 0.01). The results of this study suggested that ZnPP-IX played an important role in promoting collagen synthesis in pulmonary arteries of rats with hypoxic pulmonary structural remodeling by increasing the expression of TGF-beta3. The above findings also suggested a possible role of endogenous CO in the pathogenesis of chronic hypoxic pulmonary hypertension. 相似文献

Eight Ni proteins are known and three of these, CO dehydrogenase (CODH), acetyl-CoA synthase (ACS), and hydrogenase, are Ni-Fe-S proteins. In the last three years, the long-awaited structures of CODH and ACS have been solved. The bioinorganic community was shocked, as the structures of the active sites of CODH and ACS, the C- and A-cluster, respectively, which each had been predicted to consist of a [Fe4S4] cluster bridged to a single Ni, revealed unexpected compositions and arrangements. Crystal structures of ACS revealed major differences in protein conformation and in A-cluster composition; for example, a [Fe4S4] cluster bridged to a binuclear center in which one of the metal binding sites was occupied by Ni, Cu, or Zn. Carbon monoxide is produced in the endothelial cells and has possible vasodilator activity through three different pathways. The aim of this study was to demonstrate circulatory effects after administration of saturated carbon monoxide blood and to describe the pharmacokinetics of carbon monoxide. Six pigs were anesthetized and 150 ml blood was removed. This blood was bubbled with carbon monoxide until the carboxyhemoglobin (COHb) levels were 90-99%. A specific amount of this blood was then injected back to the animal. At predetermined times; arterial and mixed venous blood was drawn and analyzed for carbon monoxide. Systemic and pulmonary vascular resistance index (SVRi and PVRi) were measured and exhaled air was sampled and measured for carbon monoxide. Blood samples were gathered over 300 minutes along with measurements of invasive pressures, heart rate, cardiac output, oxygen saturation (SpO2), Hb, temperature and blood gases. We conclude that this type of exposure to carbon monoxide appears to have little or no effect on general vasomotor tone and, after correcting for basal levels of carbon monoxide, elimination occurs through the lungs as predicted by a single compartment model. The half-life of carbon monoxide was determined to be 60.5 minutes (SEM 4.7). 相似文献

This study examined plasma volume changes (deltaPV) in humans during periods with or without changes in body hydration: exercise-induced dehydration, heat-induced dehydration and glycerol hyperhydration. Repeated measurements of plasma volume were made after two injections of Evans blue. Results were compared to deltaPV calculated from haematocrit (Hct) and blood haemoglobin concentration ([Hb]). Eight well-trained men completed four trials in randomized order: euhydration (control test C), 2.8% dehydration of body mass by passive controlled hyperthermia (D) and by treadmill exercise (60% of their maximal oxygen uptake, VO2max) (E), and hyperhydration (H) by glycerol ingestion. The Hct, [Hb], plasma protein concentrations and plasma osmolality were measured before, during and after the changes in body hydration. Different Hct and [Hb] reference values were obtained to allow for posture-induced variations between and during trials. The deltaPV values calculated after two Evans blue injections were in good agreement with deltaPV calculated from Hct and [Hb]. Compared to the control test, mean plasma volume declined markedly during heat-induced dehydration [-11.4 (SEM 1.7)%] and slightly during exercise-induced dehydration [-4.2 (SEM 0.9)%] (P < 0.001 compared to D), although hyperosmolality was similar in these two trials. Conversely, glycerol hyperhydration induced an increase in plasma volume [+7.5 (SEM 1.0)%]. These results would indicate that, for a given level of dehydration, plasma volume is dramatically decreased during and after heat exposure, while it is better maintained during and after exercise. 相似文献

AbstractThe underlying mechanism of the central nervous system (CNS) injury after acute carbon monoxide (CO) poisoning is interlaced with multiple factors including apoptosis, abnormal inflammatory responses, hypoxia, and ischemia/reperfusion-like problems. One of the current hypotheses with regard to the molecular mechanism of CO poisoning is the oxidative injury induced by reactive oxygen species, free radicals, and neuronal nitric oxide. Up to now, the relevant mechanism of this injury remains poorly understood. The weakening of antioxidant systems and the increase of lipid peroxidation in the CNS have been implicated, however. Accordingly, in this review, we will highlight the relationship between oxidative stress and CO poisoning from the perspective of forensic toxicology and molecular toxicology. 相似文献

The present study was conducted to determine how 10 weeks of root hypoxia and a low P supply altered relative growth, and carbon acquisition and partitioning in a moderately flood tolerant pine. Pond pine (Pinus serotina Michx.) seedlings were grown in continuously flowing solution culture at 5 or 100 μM P, under aerobic or hypoxic solution conditions. Staggered harvests were used to ascertain changes in biomass allocation and relative growth over time. Carbon dioxide exchange rates (CER) were determined by infrared gas analysis, and needles were analyzed for inorganic P (Pi), sucrose, reducing sugars and starch. Although aeration treatment had no significant effect on shoot dry weight or shoot ontogeny, root dry weight of hypoxic seedlings was significantly lower than that of aerobic seedliings after 8 weeks, regardless of the P treatment. Mean relative growth rates (RGR) of roots in the high P treatment initially decreased under hypoxia, but recovered by the sixth week with the production of adventitious roots. Two weeks of hypoxic growth conditions decreased CER and stomatal conductances of seedlings in the high P treatment by more than 30% relative to their aerobic counterparts. Stomatal closure was not accompanied by a decrease in intracellular CO2, but was accompanied by an increase in starch accumulation. Recoveries of CER, stomatal conductance and carbohydrate metabolism coincided with the recovery of root growth. Low P growth conditions did not significantly affect shoot or root dry weight until the sixth week of treatment. However, differences in seedling RGR, particularly needle RGR, were discernable during the first 2 weeks. Low P treatment effects on CER paralleled changes in needle RGR, with needle RGR more affected than CER. After 6 weeks, CER of aerobically grown seedlings in the low P treatment were only 15% lower than CER of seedlings in the high P treatment, despite a 31% and 75% reduction in needle RGR and Pi concentrations, respectively. Increased starch concentrations of recently expanded needles at this time were probably a result of diminished growth. The inhibitory effect of a low P supply on shoot growth, more specifically on needle expansion and emergence of new fascicular needles, probably limited net carbon fixed per plant more than any direct effect of low P on CER per se. 相似文献

1 Populations of the spruce bark beetle, Ips typographus (L.), are known to grow rapidly in storm‐disturbed stands as a result of relaxation from intraspecific competition. In the present study, it was tested whether a second mechanism, escape in space from natural enemies, also contributes to the rapid population increase. 2 The experiment was conducted during the initiation phase of five local outbreaks of I. typographus triggered by a storm‐disturbance in November 1995 in southern Sweden. 3 The impact of natural enemies on the ratio of increase (number of daughters per mother) of I. typographus was compared pairwise between disturbed stands with high numbers of storm‐felled trees and undisturbed stands without wind‐felled trees. 4 Enemy impact was assessed by comparing the ratio of increase in uncaged (exposed to enemies) and caged (protected from enemies) bolts colonized by I. typographus prior to being placed in the stands. The experiment was conducted in the second and third summers after the storm‐felling. 5 Enemy impact was about twice as high in stands without wind‐felled trees compared with in stands with wind‐felled trees in the second summer whereas there was no significant difference between the stand types in the third summer. 6 The result demonstrates that spatial escape from enemies contributes to the rapid population growth of I. typographus after storm‐disturbances. 相似文献

Interaction energy with sulphur compounds and free volume in imidazolium-based ionic liquid were calculated by molecular dynamics (MD) simulations to examine their effects on desulphurisation. From microstructure analysis and energy contribution calculation, it is found that an increasing fractional free volume in ionic liquid and an enhancement of interaction with solute by tuning the structure of ionic liquid or oxidising sulphur compounds are favourable for desulphurisation, which allows more efficient packing of sulphur compounds in ionic liquids and more easily extraction of sulphur compounds from fuel. The eastern Asian (EAS)-eastern North American (ENA) floristic disjunction is one of the best-known biogeographic patterns in the Northern Hemisphere. Recent paleontological and molecular analyses have illuminated the origins of the biogeographic pattern, but subsequent diversification and evolution of the disjunct floras in each of the two continents after isolation remains poorly understood. Although similar in climate and floristic composition, EAS has twice as many species as ENA in genera occurring in both regions. Explaining such differences in species diversity between regions with similar environmental conditions (diversity anomalies) is an important goal of the study of the global patterns of biodiversity. We used a phylogenetic approach to compare rates of net speciation and molecular evolution between the two regions. We first identified EAS-ENA disjunct sister clades from ten genera (Asarum, Buckleya, Carpinus, Carya, Cornus, Hamamelis, Illicium, Panax, Stewartia, and Styrax) that represent diverse angiosperm lineages using phylogenetic analyses of ITS (internal transcribed spacer of nuclear ribosomal DNA) sequence data. Species richness and substitution rate of ITS between sister clades were compared. The results revealed a pattern of greater species diversity in the EAS counterparts. A positive relationship between species diversity and ITS substitution rate was also documented. These results suggest greater net speciation and accelerated molecular evolution in EAS. The data support the idea that a regional difference in net speciation rate related to topographic heterogeneity contributes to the diversity anomaly between EAS and ENA. The close relationship between rates of ITS evolution and species richness further suggests that species production may be directly linked to rate of nucleotide substitution. 相似文献

The kinetic parameters of in vivo ethylene metabolism by seedlings of Pisum sativum L. cv. Alaska have been determined. The oxidation of ethylene to CO2, (Ox) and the incorporation of ethylene into the tissue (TI) were both shown to display Michaelis-Menten kinetics (Km Ox = 0.9 × 10–6 M liquid phase, Vmax Ox = 2.4 × 10–10 moles g– dry mass h–1 Km TI = 1.6 × 10–6 M liquid phase, Vmax TI = 4.5 × 10–10 moles g–1 dry mass h–1). Propylene competitively inhibited both Ox (Ki = 7.0 × 10–6 M) and TI (Ki = 3.7 × 10–7 M). A system comparable to Ox was absent from imbibed cotyledons of Vicia faba L. cv. Aquadulce even at saturating concentrations of ethylene similar to those used in kinetic analysis on Pisum. Silver ions were shown to inhibit TI but promoted Ox, while carbon dioxide inhibited Ox but promoted TI. Kinetic data on both these effects are presented. Mark–recapture and fixed-station passive integrated transponder (PIT) telemetry were used to compare movements, distribution and survival of stocked juvenile chub Leuciscus cephalus and roach Rutilus rutilus with those of wild conspecifics. Daily activity of wild fish activity was affected by a combination of river flow and temperature, whereas stocked fishes were not influenced by environmental factors. PIT telemetry recorded exploratory movements of stocked L. cephalus immediately after stocking, a substantial number of stocked fish moved both downstream and upstream during periods of elevated flow, and proportionally more stocked fish moved during the first 6 weeks after release than later on. Proportionally more stocked fish than wild fish moved through PIT antennae, stocked L. cephalus moved greater distances than wild L. cephalus and were more widely distributed than wild fish. Minimum estimates of survival after 5 months were 50·5% for stocked R. rutilus and 28·0% for stocked L. cephalus . Ultimately, stocked cyprinids appeared to be able to cope with elevated flows and most remained in the river section local to the stocking location. 相似文献

Methane flux (FCH4) measurements using the eddy covariance technique have increased over the past decade. FCH4 measurements commonly include data gaps, as is the case with CO2 and energy fluxes. However, gap‐filling FCH4 data are more challenging than other fluxes due to its unique characteristics including multidriver dependency, variabilities across multiple timescales, nonstationarity, spatial heterogeneity of flux footprints, and lagged influence of biophysical drivers. Some researchers have applied a marginal distribution sampling (MDS) algorithm, a standard gap‐filling method for other fluxes, to FCH4 datasets, and others have applied artificial neural networks (ANN) to resolve the challenging characteristics of FCH4. However, there is still no consensus regarding FCH4 gap‐filling methods due to limited comparative research. We are not aware of the applications of machine learning (ML) algorithms beyond ANN to FCH4 datasets. Here, we compare the performance of MDS and three ML algorithms (ANN, random forest [RF], and support vector machine [SVM]) using multiple combinations of ancillary variables. In addition, we applied principal component analysis (PCA) as an input to the algorithms to address multidriver dependency of FCH4 and reduce the internal complexity of the algorithmic structures. We applied this approach to five benchmark FCH4 datasets from both natural and managed systems located in temperate and tropical wetlands and rice paddies. Results indicate that PCA improved the performance of MDS compared to traditional inputs. ML algorithms performed better when using all available biophysical variables compared to using PCA‐derived inputs. Overall, RF was found to outperform other techniques for all sites. We found gap‐filling uncertainty is much larger than measurement uncertainty in accumulated CH4 budget. Therefore, the approach used for FCH4 gap filling can have important implications for characterizing annual ecosystem‐scale methane budgets, the accuracy of which is important for evaluating natural and managed systems and their interactions with global change processes. 相似文献

The present study was designed to determine whether becoming a mother during the adolescent period changes maternal responsiveness or maternal motivation, assessed through hormonal, autonomic, and hedonic responses to recorded infant cries and interactions with their babies. Fifty-six teen mothers were compared to 58 teen non-mothers and 49 adult mothers. Teen mothers reported more sympathy and alertness in response to recorded infant cries compared to non -mother teens; however, among the teen women there were no differences between mothers and non-mothers in heart rate and cortisol responses to infant cries. In contrast, in comparison to adult mothers, teen mothers reported the same levels of sympathy and alertness to infant cries; however, adult mothers showed an 'alerted' pattern of heart rate and cortisol response to infant cries not seen in the teen mother group. Inclusion of the covariate, fathers' employment classification as an index of SES or time of testing and cortisol sampling did not affect this pattern of results. Taken together, these results show that where self-report is used as a measure of maternal responsiveness, teen mothers are no different in responsiveness than adult mothers; however, where physiological and interactional measures of responsiveness are considered, teen mothers are less likely to show heightened or selective responses to infant cries or respond 'attentively' to the infant. 相似文献

Comments are presented on an article published in October 2020 in Ecology and Evolution (“Predictive ability of a process‐based versus a correlative species distribution model”) by Higgins et al. This analyzed natural distributions of Australian eucalypt and acacia species and assessed the adventive range of selected species outside Australia. Unfortunately, inappropriate variables were used with the MaxEnt species distribution model outside Australia, so that large climatically suitable areas in the Northern Hemisphere were not identified. Examples from a previous analysis and from the use of the freely available spatial portal of the Atlas of Living Australia are provided to illustrate how the problem can be overcome. The comparison of methods described in the Higgins et al. paper is worthwhile, and it is hoped that the authors will be able to repeat their analyses using appropriate variables with the correlative model. 相似文献

Direct counts of fish obtained by night spotlighting were compared with species composition and population estimates obtained from three-pass electrofishing obtained across 29 sites along small clear streams in the Otago region of New Zealand. The influence of habitat variables on the relative efficiency of each method was also examined. The same seven species of fishes were identified by both methods. Juvenile brown trout Salmo trutta were the only species present in sufficient numbers to allow comparison of abundance estimates using the two methods. A total of 777 brown trout were counted by spotlight and 803 brown trout were caught using electrofishing. Estimates of abundance obtained by spotlighting reflected population estimates obtained by three-pass electrofishing across most habitats. Electrofishing produced higher population estimates relative to spotlighting in fast-flowing turbulent riffle habitats, whereas counts obtained by spotlighting tended to be higher relative to electrofishing in slow-flowing pool habitats. The results suggest that spotlighting is an effective method for assessing fish composition and brown trout abundance in small clear water streams, although the extremes of water velocity may influence efficiency of both spotlighting and electrofishing. 相似文献

Objective: To establish a comparison between treatment with conventional complete dentures and implant‐retained overdentures in elderly patients by conducting a literature review. Materials and methods: A search of English language peer‐review literature was completed using Medline up to 2008 focusing on evidence‐based research. Randomised clinical trials (RCTs) and longitudinal prospective studies were favoured in the review, using a general hierarchical classification. Articles that did not focus exclusively on the comparison of patient satisfaction between complete dentures and overdentures were excluded from further evaluation. The last search was conducted in February 2008. Key terms included quality of life, patient satisfaction, edentulism, complete denture and overdenture. Results: Among the 90 articles found in the initial search, 27 met the inclusion criteria. This included 18 RCTs and eight prospective and one cohort study. Most of the articles stated superiority of the mandibular implant‐retained overdenture therapy over the conventional complete denture regarding patient satisfaction and quality of life. Conclusion: Even with implant treatment presenting higher patient satisfaction and improvement of quality of life, it was not possible to establish a direct comparison between the studies due to differences in adopted methodologies. 相似文献

Suillus and Boletinus were studied using Ohta medium. In media with glucose or trehalose, all tested strains grew well. With mannose and cellobiose, strains generally grew well, except for one strain of Suillus. Utilization of dextrin and soluble starch differed with each strain, and that of sucrose and glycerol was low for all strains. Utilization of four amino acids, arginine, glutamic acid, aspartic acid, and alanine, was similar to that of ammonium tartrate for Suillus strains, but mycelial growth with amino acids was clearly lower than with ammonium tartrate for the Boletinus strain. The effect of glucose and ammonium tartrate concentrations for nine strains of the genera Suillus and Boletinus was studied with ranges for glucose of 1–100 and 200g/l, respectively, and for ammonium tartrate of 0.2–5 and 20g/l, respectively. Six strains showed maximal growth at a glucose concentration greater than 25g/l, and one strain showed maximal growth at 70g/l. The results indicate that these fungi are adapted to relatively high concentrations of carbon sources. In general, glucose concentration at mycelial growth maximum decreased as ammonium tartrate concentration increased, and at higher concentrations of glucose, mycelial growth decreased more rapidly in higher concentrations of ammonium tartrate. 相似文献

The quantitative relationship between carcinogenicity in rodents and mutagenicity in Salmonella typhimurium was examined, by using 10 monofunctional alkylating agents, including N-nitrosamides, alkyl methanesulfonates, epoxides, β-propiolactone and 1,3-propane sultone. The compounds were assayed for mutagenicity in two S. typhimurium strains (TA1535 and TA100) and in plate and liquid assays. The mutagenic activity of the agents was compared with their alkylating activity towards 4-(4′-nitrobenzyl)pyridine and with their half-lives (solvolysis constants) in an aqueous medium. No correlations between these variables were found, nor was mutagenic activity correlated with estimates of carcinogenicity in rodents.
There was a positive relationship between carcinogenicity and the initial ratios of 7-: O6-alkylguanine formed or expected after their reaction with double-stranded DNA in vitro. The results suggest that alkylation of guanine at position O6 (or at other O atoms of DNA bases) may be a critical DNA-base modification that determines the overall carcinogenicity of these alkylating agents in rodents. 相似文献
