Summary Cytological studies in desynaptic plants, isolated at the F6 generation of an intervarietal cross of Corchorus olitorius L., have shown variable numbers of bivalents and univalents in the PMC's at metaphase I, resulting in irregular distribution of chromosomes at anaphase I. The progenies of the desynaptic plants consisted of 9.24 percent of all possible primary trisomies except trisomie 6. The desynaptic condition is controlled by a pair of simple recessive genes.  相似文献

Among the pesticides evaluated against the jute pests under field conditions, endosulfan 35 EC at 350 g a.i./ha was found to be the most effective insecticide for controlling semilooper, Bihar hairy caterpillar and myllocerus weevil. Endosulfan and fenpropathrin provided good control of yellow mite as well. Even, endosulfan-treated plots gave maximum fibre and stick yield with greater benefit–cost ratio (BCR) over others. Next to endosulfan and chlorpyriphos, neemazal (azadirachtin 50,000 ppm) are useful and safer options against semilooper, Bihar having caterpillar, myllocerus and yellow mite. Methoxyfenocide 10 EC at 200g a.i./ha provided good control (70.60%) of S. obliqua even after 14 days of spraying, but it was not effective against other pests of jute. Further, almost similar efficacy of neemazal and chlorpyriphos was found against the pest complex of jute and based on BCR, neemazal could be an alternative choice over chlorpyriphos.  相似文献

Rice necrosis mosaic virus-inoculated jute plants looked more juvenile with enhanced leaf size than control ones. Such enlarged leaves and those from control ones were collected separately and extracted in chilled methanol. The crude tissue extract in case of inoculated ones showed three times higher activity in bioassay for cytokinin over control. Biological assay with fractions of crude extract separated by thin layer chromatography (TLC) and high performance liquid chromatography (HPLC) was ascertained, and one fraction from inoculated sample showing higher activity in terms of growth over respective control was selected. Ultraviolet/visible absorbance spectra of TLC-separated fractions revealed the presence of different quantities of similar kind of compounds in both control and inoculated samples. A major compound with same m/z value of 217.1 was found in both cases as revealed through electrospray ionisation mass spectrophotometry analysis along with some other minor compounds. Different proton–proton coupling and a molecular structure of the highly substituted aliphatic groups were found in the material from inoculated sample having higher cytokinin-like activity than the control one, which was noticed through 1H one-dimensional nuclear magnetic resonance analysis.  相似文献

Germination was increasingly inhibited at concentrations above 5 and 20 mM and stopped altogether at 80 and 50 mM NaF in rice and jute respectively, whereas the inhibition of seedling growth began at much lower concentrations. Of the enzymes, RNase activity was increased by fluoride, while α-amylase, protease, phytase and ATPase activities exhibited distinct inhibition from the control.  相似文献

Summary Nearly 400 individuals at two locations and over a number of years were crossed and subsequently scored for selfing versus outcrossing in eight monohybrid populations of opium poppy (Papaver somniferum). Two different marker loci, petal colour (R/r) and capsule size (B/b) were used to determine the male gametes that had effected fertilizations in F2 recessives (rr and bb). The estimates of the outcrossing parameter were found to vary with year, location and for the marker locus used ( range: 0.0988–0.3704). Study of two dihybrid crosses involving the two loci simultaneously, further confirmed that outcrossing at the R/r locus was significantly greater than that at the B/b locus. The nature of the outcrossing was, in general, nonrandom. Selfmg predominated in this species; however, there was a high frequency of natural outcrossing for generating variations in P. somniferum.CIMAP publication No. 1086  相似文献

The anatomical characteristics were studied of one X-ray induced macromutant, Undulating stem. This true-breeding mutant produces only primary phloic fibres. The secondary phloic fibres, which are the derivatives of vascular cambium, are found lacking, though there is formation of secondary phloem pyramids from the cambial tissue. Inheritance of fibre (bast) in jute (C. capsularis) is monogenic and recessive.  相似文献

Activities of photosynthetic and photorespiratory enzymes viz.,ribulose bisphosphate carboxylase, phosphoenol pyruvate carboxylaseand glycolate oxidase from jute (Corchorus olitorius L.; cv.JRO 632) leaves were compared with those from maize (C4) andsunflower (C3) leaves. The photosynthetic CO2 fixation products,the release of 14CO2 in light and dark following photosynthesisin 14CO2, chlorophyll a: b ratio, gross leaf photosyntheticrate and dry matter production rate were also studied. We report the first complete microsatellite genetic map of jute (Corchorus olitorius L.; 2n = 2 × = 14) using an F6 recombinant inbred population. Of the 403 microsatellite markers screened, 82 were mapped on the seven linkage groups (LGs) that covered a total genetic distance of 799.9 cM, with an average marker interval of 10.7 cM. LG5 had the longest and LG7 the shortest genetic lengths, whereas LG1 had the maximum and LG7 the minimum number of markers. Segregation distortion of microsatellite loci was high (61%), with the majority of them (76%) skewed towards the female parent. Genomewide non-parametric single-marker analysis in combination with multiple quantitative trait loci (QTL)-models (MQM) mapping detected 26 definitive QTLs for bast fibre quality, yield and yield-related traits. These were unevenly distributed on six LGs, as co-localized clusters, at genomic sectors marked by 15 microsatellite loci. LG1 was the QTL-richest map sector, with the densest co-localized clusters of QTLs governing fibre yield, yield-related traits and tensile strength. Expectedly, favorable QTLs were derived from the desirable parents, except for nearly all of those for fibre fineness, which might be due to the creation of new gene combinations. Our results will be a good starting point for further genome analyses in jute.  相似文献

A perfectly monofactorially segregating male sterility mutant in jute, associated with a morphological marker (ribbon leaf, rl) was found to form a pleiotropic complex with several undesirable characters, which make it unsuitable for hybrid breeding, in spite of unimpaired viability. It is improbable that the unfavorable characters can be made harmless by selection for compensating genes.  相似文献

Summary Morphologically similar leaf characters of two X-ray induced and independently isolated jute mutants from a common mother cultivar, JRO-632, were controlled by the same locus. However, they differed significantly in such quantitative characters as plant height, middle diameter, days to flower, node number and fibre yield/plant. Combining ability analysis from a 9 X 9 diallel set of crossing including these two mutants revealed that the mutants significantly differed in general combining ability (gca) effects for most of the traits in either direction or magnitude. Specific combining ability (sca) effects of the inter-mutant cross, as well as crosses with the common mother cultivar, JRO-632, also differed for most of the traits studied. It was suggested that X-irradiation induced random mutations effecting changes in the common background genotype, independent of the mutated locus.  相似文献

Controlled environments were used to establish the base-linegrowth and to evaluate the photoperiodic response of two Tossajute (Corchorus olitorius) cultivars. Fresh and dry weightsof shoots and roots, leaf areas and stem lengths were much greaterat long day (LD) 12·5, 11·5 than LD 11·5,12·5 photoperiods for both cultivars. Root lengths weregreater at LD 12·5, 11·5 initially but were longestat LD 11·5, 12·5 at 35 days from emergence. Meanrelative growth rates (MRGR) were similar irrespective of photoperiodor cultivar but declined with plant age. The superior growthof cultivar ‘Yaya’ at LD 12·5, 11·5was due to more growth in the first 7 days, while, for cultivar‘Angbadu’ it was a combination of growth for 7 days,and higher MRGR up to 21 days. Summary Reproductive effort (RE) of a day-length neutral mutant TCJ-5, its parent, and two other cultivars of jute (Corchorus capsularis) was estimated as reproductive biomass/aerial biomass. Plant height at flowering and aerial biomass were significantly higher in the mutant, while the reproductive biomass at 55 days after flowering was statistically equal. Therefore, the estimated RE was significantly lower in the mutant compared to the parent and other cultivars. The lower RE of the mutant was due to delayed initiation of flowering and additional vegetative growth in this period. The results show alteration of RE by a recessive mutation.  相似文献

A multiplex nested PCR assay was developed by optimizing reaction components and reaction cycling parameters for simultaneous detection of Corchorus golden mosaic virus (CoGMV) and a phytoplasma (Group 16Sr V‐C) causing little leaf and bunchy top in white jute (Corchorus capsularis). Three sets of specific primers viz. a CoGMV specific (DNA‐A region) primer, a 16S rDNA universal primer pair P1/P7 and nested primer pair R16F2n/R2 for phytoplasmas were used. The concentrations of the PCR components such as primers, MgCl2, Taq DNA polymerase, dNTPs and PCR conditions including annealing temperature and amplification cycles were examined and optimized. Expected fragments of 1 kb (CoGMV), 674 bp (phytoplasma) and 370 bp (nested R16F2n/R2) were successfully amplified by this multiplex nested PCR system ensuring simultaneous, sensitive and specific detection of the phytoplasma and the virus. The multiplex nested PCR provides a sensitive, rapid and low‐cost method for simultaneous detection of jute little leaf phytoplasma and CoGMV. Based on BLASTn analyses, the phytoplasma was found to belong to the Group 16Sr V‐C.

Significance and Impact of the Study

Incidence of phytoplasma diseases is increasing worldwide and particularly in the tropical and subtropical world. Co‐infection of phytoplasma and virus(s) is also common. Therefore, use of single primer PCR in detecting these pathogens would require more time and effort, whereas multiplex PCR involving several pairs of primers saves time and reduces cost. In this study, we have developed a multiplex nested PCR assay that provides more sensitive and specific detection of Corchorus golden mosaic virus (CoGMV) and a phytoplasma in white jute simultaneously. It is the first report of simultaneous detection of CoGMV and a phytoplasma in Corchorus capsularis by multiplex nested PCR.  相似文献

A mutant of Corchorus olitorius L. (cultigen, JRO-632) isolatedfrom R2 generation of 90 kR X-rayed progeny is characterizedby excessive leaf-fall. Although the initial rate of leaf-fallis comparable with that of the mother strain, after 65 daysof growth, leaf-fall is much more rapid as revealed from comparativecounts of the number of leaves and nodes. Formation of a separation layer has been observed at the abscissionzone of the sixth leaf above the last abscissed leaf of themutant, whereas, in the mother strain it is initiated just abovethe last abscissed leaf under the same climatic conditions inthe field. A crude extract from senescent leaves of the mutant acceleratesseparation layer formation in the abscission zone of the motherstrain. Transgenic hairy root system is important in several recalcitrant plants, where Agrobacterium tumefaciens-mediated plant transformation and generation of transgenic plants are problematic. Jute (Corchorus spp.), the major fibre crop in Indian subcontinent, is one of those recalcitrant plants where in vitro tissue culture has provided a little success, and hence, Agrobacterium-mediated genetic transformation remains to be a challenging proposition in this crop. In the present work, a system of transgenic hairy roots in Corchorus capsularis L. has been developed through genetic transformation by Agrobacterium rhizogenes harbouring two plasmids, i.e. the natural Ri plasmid and a recombinant binary vector derived from the disarmed Ti plasmid of A. tumefaciens. Our findings indicate that the system is relatively easy to establish and reproducible. Molecular analysis of the independent lines of transgenic hairy roots revealed the transfer of relevant transgenes from both the T-DNA parts into the plant genome, indicating the co-transformation nature of the event. High level expression and activity of the gusA reporter gene advocate that the transgenic hairy root system, thus developed, could be applicable as gene expression system in general and for root functional genomics in particular. Furthermore, these transgenic hairy roots can be used in future as explants for plantlet regeneration to obtain stable transgenic jute plants.  相似文献

The effect of protein, isolated from Jute (Corchorus olitorius) seed was studied upon albino rats with respect to some of their serum, liver and intestinal enzymes and liver lipids. An increase in the body weight (including the weight of the liver) was noted in test animals after feeding with a Jute seed protein enriched diet. It was also observed that AST, ALT and total lipid of liver increased significantly whereas AST and ALT of serum were decreased. An increase in the concentration of lipids in the liver was found and this may be due to excess of the seed protein in the diet. An overall observation reveals that there is slight fatty infiltration in the liver of test animals.  相似文献
