嗅觉在昆虫的多种行为中发挥关键作用。气味分子与嗅觉神经元树突上气味受体的结合,参与了昆虫嗅觉识别的初始过程。昆虫的嗅觉神经元表达两类气味受体: 一是传统气味受体,该类受体同源性较低,在少部分嗅觉神经元中表达; 二是Or83b家族受体,该类受体不感受气味,在不同昆虫间较为保守且在大多数嗅觉神经元中表达。目前,对于单个传统气味受体的气味分子配体特异性所知甚少; 对于Or83b家族受体,一般认为其可能具有将传统气味受体运送至嗅觉神经元树突膜上的功能。此外,有一些实验证据不支持昆虫气味受体为G蛋白偶联受体的观点。  相似文献

昆虫的嗅觉对昆虫的栖息地选择、觅食、群集、趋避、繁殖以及信息传递等行为具有重要的影响。对昆虫嗅觉机理的深入研究和嗅觉信号传导途径的完整阐述,是探索农业害虫的专一性防治的基础。嗅觉受体(olfactory receptors,Ors)是G蛋白偶联受体(G protein-coupled receptor)的一种,是嗅觉系统的关键成分。近年来嗅觉受体的研究日益受到关注。本文对昆虫嗅觉的基本过程、基因结构和表达调控特征、蛋白分子结构、生理功能、分布部位和相关配体的研究等进行了综述。  相似文献

【目的】克隆小菜蛾Plutella xyostella气味受体Or83b基因, 并进行原核表达, 为研究小菜蛾寄主选择行为的分子机理, 开发昆虫行为调节剂提供基础。【方法】提取小菜蛾的总RNA, 反转录获得总cDNA, 采用RT-PCR方法扩增目的基因, 将其克隆至T载体并测序, 然后将目的基因克隆到大肠杆菌Escherichia coli表达载体pET-32a (+)中表达。经酶切、 PCR及测序鉴定正确后转化BL21 (DE3)菌株, 用IPTG诱导表达, 通过SDS-PAGE, Western印迹鉴定表达蛋白。【结果】获得了编码小菜蛾Or83b的cDNA序列, 该基因阅读框长1 413 bp, 编码471个氨基酸, 预测的等电点为7.19, 命名为PlxyOr83b(GenBank登录号为GQ923610); 成功构建了pET-PlxyOr83b原核表达重组质粒, 目的基因获得高效表达, 其融合蛋白分子量为32.0 kD, Western blot 检测结果进一步表明PlxyOr83b在大肠杆菌DE3中得到正确表达。【结论】成功克隆和表达了小菜蛾气味受体基因PlxyOr83b, 该基因与其他昆虫Or83b基因基本一致。  相似文献

通过对丝状真菌G蛋白偶联受体(GPCR)的结构、分类以及功能方面进行综述,以期明确丝状真菌与其他模式生物GPCR之间的关系。基于已报道的模式生物及丝状真菌等不同生物中的GPCR,通过SMART保守结构域分析,以及利用Clustal X、MEGA等软件对上述GPCR进行遗传关系分析。明确丝状真菌典型GPCR具有七跨膜结构域,新型GPCR则含有PIPK、RGS等保守结构域,明确不同学者对于GPCR的分类情况,以及新型GPCR所具有的特殊功能,明确模式生物GPCR、丝状真菌GPCR分别各自聚类。丝状真菌中GPCR的数量较模式生物少,不同分类单元中真菌之间GPCR的数量也不尽相同,同时,丝状真菌GPCR除具有典型的七跨膜结构域外,还含有一些其他保守的结构域,上述研究为进一步开展其功能研究提供重要的理论基础。  相似文献

目前已经克隆了3种甘丙肽受体(GalR1, GalR2, GalR3),它们都是与G蛋白相偶联的受体.3种甘丙肽受体的氨基酸序列、药理学特性以及第二信使系统各不相同.GalR1/3受体可以抑制腺苷酸环化酶并可以激活钾通道,GalR2受体可以激活磷脂酶C并增加胞内钙离子浓度.用RNA印迹、反转录PCR以及原位杂交等技术对上述3种甘丙肽受体在人、大鼠和小鼠中的分布进行了研究,发现它们具有不同的分布特征,提示不同的甘丙肽受体可能参与不同的生理过程.  相似文献

In Drosophila, odorant receptors are encoded by an old and moderatelysized multigene family. Or22a and Or22b are two tandemly arrangedgenes of this family that have proved to be the result of arather young duplication. Nucleotide variation in the regionspanning both duplicates was surveyed in four natural populations(two African and two non-African) of Drosophila melanogasterand also analyzed in species of the melanogaster subgroup. Theintraspecific survey revealed a particular copy-number polymorphismin some of the studied populations, with the two genes (Or22aand Or22b) present in the long variant and a single chimericgene (Or22ab) present in the short variant. Estimated nucleotidediversity was higher in the short than in the long variant,despite the ancestral character of the latter variant in D.melanogaster. The general skew toward low-frequency variantsdetected in the non-African long variant and its reduced levelof silent polymorphism relative to divergence is consistentwith the recent fixation of an advantageous mutation at, ornearby, the Or22 long variant region. The nonnegligible frequencyof the short variant and the presence of a highly divergenthaplotype in the East African sample would point to direct orindirect selection for its maintenance in the species. Therewas evidence for a generally more rapid evolution of the Or22bcopy at both synonymous and nonsynonymous sites. In insects, many critical olfactory behaviours are mediated by the large odorant receptor (Or) gene family, which determines the response properties of different classes of olfactory receptor neurons (ORNs). While ORN responses are generally conserved within and between Drosophila species, variant alleles of the D. melanogaster Or22 locus have previously been shown to alter the response profile of an ORN class called ab3A. These alleles show potential clinal variation, suggesting that selection is acting at this locus. Here, we investigated if the changes seen in ab3A responses lead to changes in olfactory-related behaviours. We show that variation at the Or22 locus and in the ab3A neurons are not fully compensated for by other ORNs and lead to overall changes in antennal odorant detection. We further show that this correlates with differences in odorant preference behaviour and with differences in oviposition site preference, with flies that have the chimaeric short allele strongly preferring to oviposit on banana. These findings indicate that variation at the Or22 locus leads to changes in olfactory-driven behaviours, and add support to the idea that the ab3A neurons are of especial importance to the ecology of Drosophila flies.  相似文献

In mice, male-typical preference for female olfactory cues results largely from sexually differentiated testosterone production. It is currently unclear on which cells and tissues testosterone acts to produce male-typical preference for female olfactory cues. To further address the site of androgen action on olfactory preference, we have developed a loxP-based transgenic mouse that overexpresses androgen receptors (AR) only when activated by Cre. We used this transgene to overexpress AR globally in all tissues using a CMV-Cre driver and a Nestin-Cre driver to overexpress AR selectively in neural tissue. We then examined olfactory preference in transgenic and wildtype (Wt) littermates by simultaneously exposing animals to female-soiled, male-soiled and clean bedding. Ubiquitous overexpression of AR in CMV-AR mice increased preference for male bedding, whereas neural-specific AR overexpression in Nestin-AR transgenic mice did not differ from wildtype siblings in olfactory preference. Neural activation of olfactory brain areas in response to female-soiled bedding was also evaluated in these mice by measuring FOS immunoreactivity. This revealed a decrease in neural activity along the accessory olfactory pathway that accompanied the decrease in preference for female odors in CMV-AR males, compared to both Nestin-AR and Wt male siblings. Together, results indicate that androgens act via non-neural AR to mediate olfactory preference and neural responses to olfactory stimuli, and further suggest that AR in non-neural tissues can promote androphilic odor preferences in male mice.  相似文献

Animals use odors as signals for mate, kin, and food recognition, a strategy which appears ubiquitous and successful despite the high intrinsic variability of naturally-occurring odor quantities. Stimulus generalization, or the ability to decide that two objects, though readily distinguishable, are similar enough to afford the same consequence, could help animals adjust to variation in odor signals without losing sensitivity to key inter-stimulus differences. The present study was designed to investigate whether an animal's ability to generalize learned associations to novel odors can be influenced by the nature of the associated outcome. We use a classical conditioning paradigm for studying olfactory learning in honeybees to show that honeybees conditioned on either a fixed- or variable-proportion binary odor mixture generalize learned responses to novel proportions of the same mixture even when inter-odor differences are substantial. We also show that the resulting olfactory generalization gradients depend critically on both the nature of the stimulus-reward paradigm and the intrinsic variability of the conditioned stimulus. The reward dependency we observe must be cognitive rather than perceptual in nature, and we argue that outcome-dependent generalization is necessary for maintaining sensitivity to inter-odor differences in complex olfactory scenes.  相似文献
