Upland rice cultivars were evaluated in the greenhouse for susceptibility to the rice blast disease caused by Pyricularia grisea Sacc., on two upland soils from the Philippines previously considered to be “blast conducive” and “blast non-conducive”. Under monocyclic inoculation tests plants grown in conducive soil showed significantly greater lesion development than plants of the same cultivar grown in non-conducive soil: cultivars considered to be susceptible to the isolates used showed increased number of susceptible-type lesions; resistant cultivars showed increased number of hypersensitive resistant-type lesions. A similar effect was observed under polycyclic tests where several generations of the pathogen were allowed to develop on the test plants. Dilution of conducive soil with non-conducive soil resulted in a corresponding reduction of disease severity, although this was most pronounced on resistant cultivars. Removal of leaf epicuticular waxes (LEW) using organic solvents increased the number of resistant-type lesions on resistant cultivars grown in both soils following inoculation. Susceptible plants were not suitable for quantifying the relative blast conduciveness of a soil because of the extreme environmental sensitivity of the bioassay and the tendency of lesions to coalesce. Comparing numbers of resistant-type lesions on leaves of plants stripped of LEW and inoculated with an incompatible P. grisea isolate among plants grown in different soils proved to be a satisfactory means of distinguishing the relative blast conduciveness of soils under controlled conditions. This method was field tested in eastern India and results corroborated farmer assessment of which soils were blast conducive. Using incompatible isolate-cultivar combinations and LEW-free leaves is proposed as a simple bioassay for assessing blast conduciveness of soils and should prove useful in regional characterization of rice blast risk.  相似文献

Samba mahsuri (BPT 5204) is a cultivar of the medium slender grain indica variety of Oryza sativa grown across India for its high yield and quality. However, this cultivar is susceptible to several diseases and pests including rice blast. The analysis of near isogenic lines indicated the presence of a resistance gene, Pi-1(t) in the donor cultivar C101LAC which is highly resistant to the rice blast fungus Magnaporthe grisea (M. grisea). C101LAC was crossed with susceptible indica rice cultivar (BPT 5204) to generate the mapping population. A mendelian segregation ratio of 3:1 for resistant to susceptible F2 plants using bulk segregation analysis confirmed the presence of a major gene pi-1(t) by simple sequence repeats marker RM224 to the highly virulent blast isolate DRR 001.  相似文献

Phytotoxicity of AAL-toxin and fumonisin B1 to six cultivars of tomato was compared with the pathogenicity of their fungal sources, Alternaria alternata and Fusarium moniliforme, respectively. These include two AAL-toxin susceptible cultivars with genotypes(asc/asc), three resistant cultivars (Asc/Asc), and a heterozygous cultivar (Asc/asc.) A. alternata spores were pathogenic to the susceptible but not to the resistant cultivars F. moniliforme was not pathogenic to any of the tomatoes. Filtrates of both fungi grown on rice containing their respective toxins caused necrosis within 48 h and eventually mortality on susceptible cultivars but not on the resistant lines. The heterozygous cultivar Asc/asc showed minimal damage and no mortality after 14 days exposure to both filtrates and both toxins. The spores of both fungi had no effect on heterozygous intact plants. Tomato leaf disc bioassays with AAL-toxin and fumonisin B1 at 1μM caused cellular leakage and reduced chlorophyll content in susceptible cultivars and minimal effects on the heterozygous and resistant varieties.  相似文献

Jasmonate is an important endogenous chemical signal that plays a role in modulation of plant defense responses. To understand its mechanisms in regulation of rice resistance against the fungal pathogen Magnaporthe oryzae, comparative phenotype and proteomic analyses were undertaken using two near‐isogenic cultivars with different levels of disease resistance. Methyl‐jasmonate (MeJA) treatment significantly enhanced the resistance against M. oryzae in both cultivars but the treated resistant cultivar maintained a higher level of resistance than the same treated susceptible cultivars. Proteomic analysis revealed 26 and 16 MeJA‐modulated proteins in resistant and susceptible cultivars, respectively, and both cultivars shared a common set of 13 proteins. Cumulatively, a total of 29 unique MeJA‐influenced proteins were identified with many of them known to be associated with plant defense response and ROS accumulation. Consistent with the findings of proteomic analysis, MeJA treatment increased ROS accumulation in both cultivars with the resistant cultivar showing higher levels of ROS production and cell membrane damage than the susceptible cultivar. Taken together, our data add a new insight into the mechanisms of overall MeJA‐induced rice defense response and provide a molecular basis of using MeJA to enhance fungal disease resistance in resistant and susceptible rice cultivars.  相似文献

Different cultivars of cow pea and garden pea seeds were surveyed for susceptibility or resistance towards the toxigenic and aflatoxin-producing mould (Aspergillus flavus IMI 102135). The results show that aflatoxin production varied among the different cultivars of both cow pea and garden pea. Morphological and histological characters of the different cultivars tested did not show any relation between colour, shape and size of seeds and the amount of aflatoxin produced. The chemical analysis of the different constituents obtained from both seed coats and seed kernels with susceptible, partially resistant and resistant cow pea and garden pea cultivars revealed that the resistant cultivars of cow pea (namely: Balady cultivar) and garden pea (namely: Melting Sugar cultivar) contained lower levels of sodium and higher levels of phosphate and potassium.  相似文献

The effect of N form and Si nutrition on rice (Oryza sativa L.) susceptibility to blast disease (caused by Pyricularia oryzae Cav.) was assessed in the greenhouse with nutrient solution culture. The N form supplied to the susceptible cultivar IR50 affected the relative infection efficiency (RIE) of P. oryzae measured as lesions/cm2 leaf. Plants given NO3 - were more susceptible than plants receiving NH4 +-N. This result may partially explain why plants grown in nonflooded soil, where NO3 - is the main source of inorganic N, are more susceptible to blast than plants grown in flooded soils, where NH4 + is the main inorganic N source. Nitrate-N and Mn concentration were higher in leaf blades of plants grown with NO3 -. Total-N, Si, and Fe concentration were not affected by N form. The addition of Si significantly increased IR50 resistance to blast. With 2.2 mol m-3 Si in solution, RIE values were lower by more than 90% than the control with no Si added in solution. The effect of Si accumulation in leaves at various positions was further studied in cultivars having differing levels of resistance (IR50, IR36, and IAC165). Silicon addition significantly reduced RIE in the three cultivars. Silicon concentration in the topmost leaves (the only leaves showing typical blast lesions) was not significantly different among the three cultivars when 2.2 mol m-3 Si was used. Silicon was an important component in the mechanism of resistance to blast and it was effective regardless of the original level of resistance of the cultivar used. DNA markers that allow for identification of resistance genes in rice germplasm have a great advantage in resistance breeding because they can assess the existence of the genes without laborious inoculation tests. Functional markers (FMs), which are designed from functional polymorphisms within the sequence of genes, are unaffected by nonfunctional allelic variation and make it possible to identify an individual gene. We previously showed that the resistance function of the rice blast resistance gene Pit in a resistant cultivar, K59, was mainly acquired by up-regulated promoter activity through the insertion of a long terminal repeat (LTR) retrotransposon upstream of Pit. Here, we developed PCR-based DNA markers derived from the LTR-retrotransposon sequence and used these markers to screen worldwide accessions of rice germplasm. We identified 5 cultivars with the LTR-retrotransposon insertion out of 68 rice accessions. The sequence and expression pattern of Pit in the five cultivars were the same as those in K59 and all showed Pit-mediated blast resistance. The results suggest that the functional Pit identified using the markers was derived from a common progenitor. Additionally, comparison of the Pit coding sequences between K59 and susceptible cultivars revealed that one nucleotide polymorphism, which caused an amino acid substitution, offered another target for a FM. These results indicate that our DNA markers should enhance prediction of Pit function and be applicable to a range of rice varieties/landraces cultivated in various regions worldwide and belonging to the temperate japonica, tropical japonica, and indica groups.  相似文献

稻瘟病是世界范围内影响水稻(Oryza sativa)生产的主要病害。抗稻瘟病基因的发掘和育种利用是控制稻瘟病经济、环保的有效措施。为了揭示云南地方水稻品种子预44广谱持久抗瘟机制, 利用江南香糯和子预44杂交构建的F7重组自交群体, 采用苗期稻瘟病菌自然诱发接种法, 通过调查田间抗瘟性表型数据, 结合基因型数据对子预44中的数量抗瘟性位点进行了分析。结果表明, 在连锁系数(logarithm of odds, LOD)大于2.0的域值上, 共检测出13个QTLs, 分别位于第1、2、6、8、12号染色体上。不同位点表型贡献值差异较大, 范围为5.8%-21.9%, 其中8号染色体上标记RM72-RM404之间的QTLs可解释约61.9%的表型变异, 很可能为一个主效抗瘟QTL位点。多个位点的主效和微效抗性相结合可能是子预44持久稻瘟病抗性的分子基础。  相似文献

Blast, caused by Magnaporthe grisea, is one of the most important diseases in rice production regions of the world including Iran. To determine progress of rice blast disease on the selective cultivars and lines also to assay some components of partial resistance, a set of Iranian rice cultivars (Local and breeding) along with near-isogenic lines (NILs) and breeding lines from International Rice Research Institute (IRRI) were tested with some field races of the fungus in blast nursery and five of selective races in greenhouse. These experiments were conducted in a Randomized complete Block Design (RCBD) with three replications (except greenhouse experiment on the leaves). Traits in this study consisted of Infection Neck Number (INN), Neck Lesion Size (NLS), Infection Type (IT), percent Diseased Leaf Area (DLA) and Area Under Disease Progress Curve (AUDPC); also IT, Sporulation Lesion Number (SLN), Sporulating Region Diameter (SRD) and percent DLA were measured in leaf blast in greenhouse (one replication). The Iranian local cultivars and NILs i.e. Co-39 and C104-PKT located as susceptible group for AUDPC, IT, INN and NLS. Iranian breeding cultivars, breeding lines from IRRI and NILs (except Co-39 and C104-PKT) were resistant or indicated hypersensivity reaction (HR). Some cultivars (Fujiminori, Onda, and Hassan Saraii) were semi susceptible to leaf blast in nursery. The main point is correlation in 1% (a = 0.0001) between the traits in greenhouse and blast nursery. Neck node infection of Haraz cultivar in greenhouse experiment to IA-89 race is very important, because Haraz is a resistant cultivar to blast disease in Iran.  相似文献

Protocols for selecting plant tissues of winter oilseed rape (Brassica napus subsp. oleifera) with resistance to Leptosphaeria maculans by either stem or leaf inoculation of both soil and in vitro grown plant material are described. The stem inoculation procedure gave good correlation (r = 0. 92) between the 50 day stem disease scores of eight out of nine cultivars of soil grown winter oilseed rape inoculated with isolate 41A4 of L. maculans and the N. A. B. esistance ratings or resistance data from field trials. The exception was the cultivar Liradonna. Inoculation of stems of five cultivars with isolates 41A4, 433 and 478 indicated a range of isolate virulence 478 > 41A4 > 433. This was the inverse of that observed in leaf inoculations. Application of the stem inoculation procedure to in vitro shoot cultures allowed differentiation of resistant and susceptible cultivars, including the cultivar Liradonna, after 20 days incubation at 20°C. The protocol was also applicable to plantlets regenerated from thin cell layer explants grown in vitro. Inoculations with isolate 433 allowed the differentiation of resistant, intermediately resistant and susceptible leaf material of soil grown plants, when leaf discs from young leaves were incubated on water agar supplemented with BAP (1 × 10?5 M) at 25°C for 10 days. Intermediately resistant leaves were resistant after 10 days and susceptible after 15 days of incubation. Leaves of shoot cultures grown in vitro were more susceptible than the corresponding soil grown material. However, inoculation of old leaves with isolate 41A4 (an isolate of less virulence on leaves than 433) distinguished the cultivars after 15 days of incubation. These protocols allow the accurate assessment of resistance to L. maculans at the stem or leaf level and are of use in traditional as well as in vitro selection programmes.  相似文献

It has been argued that resistant cultivars may enhance the susceptibility of insect pests to insecticides, and thus can reduce crop production costs by lowering the rate of insecticide application. Here we studied the susceptibility of the cowpea seed beetle, Callosobruchus maculatus (Coleoptera: Chrysomelidae), reared on either a susceptible or resistant cultivar of the mung bean (Vigna radiate ), to a herbal extract taken from the prickly parsnip, Echinophora platyloba (Umbelliferae). After selection of a susceptible (Parto) and a resistant (Sahar) cultivar, the median lethal concentration (LC50) of the herbal extract was determined as 24.41 and 20.28 μL/250 mL air against beetles reared on susceptible and resistant cultivars, respectively. Rearing on resistant cultivar had a significant effect on biological parameters of C. maculatus regardless of insecticide application. However, herbal extract showed a significant interaction with host resistance in terms of oviposition rate and pre‐adult development time. These results provide evidence that the resistant cultivar increases the susceptibility of C. maculatus to herbal extract. The use of plant resistance in combination with narrow‐spectrum insecticides seems to be of both economic and environmental importance, as it can reduce the costs of crop production and lower the doses of insecticides required for satisfactory pest control.  相似文献

The peroxidase activities in leaves from resistant and susceptible cultivars of wheat infected and non-infected by Erysiphe graminis DC were studied. In non-infected wheat, soluble and ionic bound peroxidase activity level was found to be higher in the resistant cultivar than that in the one susceptible to Erysiphe graminis DC. After infecting wheat leaves with Erysiphe graminis DC a remarkable increase in the activity of soluble and ionic bound peroxidases was detected 5 days after inoculation only in the resistant cultivar. In the susceptible cultivar a high increase in the activity of the soluble and ionic bound peroxidases occurred only 15 days after inoculation. Using ion exchange chromatography four peroxidase fractions were obtained from infected susceptible and resistant cultivars as from non-infected ones. The fraction II in non-inoculated resistant cultivars was much higher than that in the susceptible one. This fraction increased after inoculation in both cases reaching a higher level in resistant cultivars. Fraction I was higher in the susceptible cultivar. Electrofocusing profiles of peroxidase from the susceptible and resistant cultivar differed from one another. New peroxidase bands after inoculation appeared only in the resistant cultivar.  相似文献

A field experiment in which main‐crop potatoes were grown every other year was conducted on a sandy soil from 1994 to 1999. The aim of the experiment was to control soil‐borne pathogens of potato with ecologically sound methods. Potato grown as a trap crop from the end of April to the end of June (8 wk) was used to control potato cyst nematodes (PCN) (Globodera pallida), and its effects on other important soil pathogens and on the growth of a subsequent potato crop were also assessed. Additional experimental treatments were a potato crop from which the haulm was removed and a green manure crop. Three potato cultivars with different degrees of resistance to PCN were grown as the main crop. Duplicate sets of the experiment were run concurrently. The PCN were effectively controlled by the potato trap crop. When a highly resistant potato cultivar was grown as a main crop after the trap crop, the post‐harvest soil infestation was very low. When a moderately resistant cultivar was grown after the trap crop the soil infestation also remained low. When the trap crop was alternated with a susceptible potato cultivar as a main crop, soil infestation increased slightly, but the degree of control when compared with no trap crop averaged 96%. Soil infestation with root‐knot nematodes (mainly Meloidogyne hapla) increased when potato was grown as a trap crop, but soil infestation with the root‐lesion nematode Pratylenchus crenatus was not affected. Stem canker caused by Rhizoctonia solani was not affected by the trap crop but black scurf (sclerotia of R. solani) on tubers was reduced. Soil infestation with Verticillium dahliae declined in one of the duplicate sets of the experiment but not in the other. However, stem infections by V. dahliae were significantly decreased in both sets, although the effect depended on the PCN‐resistance level of the potato cultivar. When a highly resistant potato cultivar was grown Verticillium stem infections were not significantly affected, they were decreased with a moderately resistant cultivar but the decrease was most pronounced with a PCN‐susceptible cultivar. Senescence of a following potato crop was not influenced by the trap crop when a highly PCN‐resistant cultivar was grown, but it was delayed in the case of a moderately resistant or a susceptible cultivar, resulting in higher tuber yields for those cultivars. The experiment proved that a trap crop can be an alternative to chemical soil disinfection but, for several reasons, the potato itself is not an ideal crop for this purpose; a trap crop other than potato must be developed.  相似文献

Rice production is of great importance to food security in Nigeria. The African rice gall midge (AfRGM), Orseolia oryzivora Harris & Gagné, is a major pest of lowland rice in Nigeria and many other Africa countries. Current management efforts are directed at integrating host plant resistance with biological agents. The type and abundance of some phytochemicals are known to affect resistance in host plants, so in this study we measured rice culm hardness, rice culm stickiness and the type and quantity of phytosterols in rice cultivars, and investigated the effects of these factors on resistance to the AfRGM. The study was conducted at 24 ± 2°C, 60 ± 5% relative humidity, and a 12 h light : 12 h dark photoperiod. Three rice cultivars were studied: one Oryza glaberimma cultivar, Tog 7442 (resistant), and two O. sativa cultivars, Cisadane (tolerant) and ITA 306 (susceptible). We found no significant differences in culm hardness and stickiness among the cultivars tested. Using gas chromatography (flame ionization detection) analysis, we identified four major phytosterols: campesterol, 24‐methylenecholesterol, stigmasterol and β‐sitosterol. 24‐Methylenecholesterol was the most abundant sterol in the resistant O. glaberimma cultivar (79.20 μg), whereas β‐sitosterol was most abundant in the two susceptible O. sativa cultivars (Cisadane, 71.27 μg; ITA 306, 66.37 μg). The comparatively high concentration of 24‐methylenecholesterol in this cultivar of O. glaberimma may be an indicator of resistance to AfRGM.  相似文献

Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae) is the major pest of tomato plant. Since using relatively resistant cultivars may reduce the number of sprayings as well as postpone the development of the resistance to pesticides, the present study focused on evaluating the damage of T. absoluta on eleven 45-day-old tomato cultivars under greenhouse condition. Larval mines on the leaves as well as the terminal bud damage were considered. Damaged leaves, active mines and damaged terminal buds were significantly different among the cultivars. Cluster analysis using SPSS software resulted in grouping the cultivars into four categories as relatively resistant, partially resistant, partially susceptible and susceptible. The host plant’s growing characteristics (height and leaflet number) were assessed and likewise the weight of the resulted pupae. Differences in vulnerability of the cultivars showed that tomato cultivars possess resistant traits and the identification and utilisation of these traits can give rise to resistant varieties.  相似文献

The influence of resistant and susceptible potato cultivars on Globodera rostochiensis population density changes was studied at different nematode inoculum levels (Pi) in the greenhouse and field. Soil in which one susceptible and two resistant cultivars were grown and fallow soil in pots was infested with cysts to result in densities of 0.04-75 eggs/cm³ soil. A resistant cultivar was grown in an infested field with Pi of 0.7-16.7 eggs/cm³ soil. Pi was positively correlated with decline of soil population densities due to hatch where resistant potatoes were grown in the greenhouse and in the field but not in fallow soil. However, Pi was not correlated with in vitro hatch of G. rostochiensis cysts in water or potato root diffusate. Under continuous culture o f a resistant cultivar, viable eggs per cyst declined 60-90% per plant growth cycle (4 weeks) and the number of cysts containing viable eggs had decreased by 77% after five cycles. The rate of G. rostochiensis reproduction on both resistant and susceptible cultivars was negatively correlated with Pi. These data were used to predict the effect of resistant and susceptible potato cultivars on G. rostochiensis soil population dynamics.  相似文献

Because of the frequent breakdown of major resistance (R) genes, identification of new partial R genes against rice blast disease is an important goal of rice breeding. In this study, we used a core collection of the Rice Diversity Panel II (C‐RDP‐II), which contains 584 rice accessions and are genotyped with 700 000 single‐nucleotide polymorphism (SNP) markers. The C‐RDP‐II accessions were inoculated with three blast strains collected from different rice‐growing regions in China. Genome‐wide association study identified 27 loci associated with rice blast resistance (LABRs). Among them, 22 LABRs were not associated with any known blast R genes or QTLs. Interestingly, a nucleotide‐binding site leucine‐rich repeat (NLR) gene cluster exists in the LABR12 region on chromosome 4. One of the NLR genes is highly conserved in multiple partially resistant rice cultivars, and its expression is significantly up‐regulated at the early stages of rice blast infection. Knockout of this gene via CRISPR‐Cas9 in transgenic plants partially reduced blast resistance to four blast strains. The identification of this new non‐strain specific partial R gene, tentatively named rice blast Partial Resistance gene 1 (PiPR1), provides genetic material that will be useful for understanding the partial resistance mechanism and for breeding durably resistant cultivars against blast disease of rice.  相似文献

Five rice cultivars, one hybrid (WR96), three modern (BR16, BR26, and BRRI Dhan27) and one local (Pari) were screened for reaction to brown leaf spot disease caused by Cochliobolus miyabeanus and performance of yield-related characters. The severity of brown leaf spot varied with growth stages of rice plant as well as different cultivars tested under field condition. Low disease severity was observed at maximum tillering stage compared to moderate to high at dough stage, with hybrid cultivar WR96 showing highest disease, while local cultivar Pari had the lowest. Brown spot disease severity in different cultivars under induced epiphytic condition also followed the similar trend. The results also revealed that most of the yield-contributing characters examined showed wide variations among the cultivars. Modern cultivar BR16 produced the highest panicle length, number of grain per panicle and grain yield per hectare. At the same time as local cultivar Pari generated the lowest number of tiller per plant, panicle length, grain number per panicle and grain yield per hectare. Moreover, hybrid cultivar WR96 produced the highest percentage of spotted grain per panicle and seed yielding C. miyabeanus, and also the lower percentage of seed germination, while the reverse was observed in local cultivar Pari. These findings may allow producers and breeders to select rice cultivar, resistant or tolerant to brown leaf spot disease and to avoid significant reductions in grain yields.  相似文献

The content of phenolic compounds as well as the activity of polyphenoloxidase and peroxidase in the red raspberry callus tissue after D. applanata infection were investigated. Two red raspberry cultivars: Latham — relatively resistant and M. Promise — susceptible were used. In the M. Promise cultivar the amount of phenols, especially o-diphenols, decreased after infection and stayed more or less on the same level in Latham cultivar. The activity of polyphenoloxidase increased in both of the investigated cultivars, however in the tissue of Latham cultivar this rise was more pronounced than in the M. Promise one. The changes in peroxidase activity after infection were inconsiderable. The relation between the changes observed after infection in the phenolic metabolism and the differentiated susceptibility of callus tissue to D. applanata of the investigated cultivars was discussed. The obtained results explain partially the browning of infected callus tissue of one of the investigated cultivar i.e. M. Promise which had already been observed in the earlier investigations.  相似文献

Ten mungbean cultivars were evaluated for their resistance to iron deficiency in view of chlorosis symptoms, plant growth and seed yield under field conditions on a calcareous soil in Thailand. The KPS2 cultivar was highly susceptible; the KPS1, PSU1 and Pag-asa 1 cultivars were somewhat susceptible; the VC1163B cultivar was moderately tolerant; the CN36, CN60, UT1 and CNM-I cultivars were tolerant; and the CNM8509B cultivar was very tolerant to iron deficiency. Foliar application of a solution of 5 g L-1 ferrous sulphate was effective in correcting chlorosis that was induced by iron deficiency, and it enhanced both the growth and the yield of susceptible cultivars. Compared with the susceptible cultivar KPS2, the tolerant cultivar UT1 had a greater ability to lower the pH of the nutrient solution in response to iron deficiency. The root-associated Fe3+-reduction activity of UT1 that had been grown in -Fe medium was similar to that of the plants grown in +Fe medium when the acidification of the medium occurred. Acidification of the medium in response to iron deficiency might contribute to the efficient solubilization of iron from calcareous soils, and it related more closely to the resistance to iron deficiency than Fe3+ reduction by roots in mungbean cultivars.  相似文献
