Abstract: Two populations of the psocid, Liposcelis bostrychophila Badonnel, were exposed to two CO2-enriched atmospheres (35% CO2 + 21% O2, and 55% CO2 + 21% O2, balance N2) for 30 generations. Controls were reared in normal atmospheres. The reserves of triacylglycerol and polysaccharides were evaluated in adults of the two experimental and the control populations in generations F15 and F30. The utilization rate of triacylglycerol and polysaccharides in the CO2-enriched atmospheres were also determined in generation F30. The results indicated that the reserves of triacylglycerol and polysaccharides increased significantly during selection for CO2 resistance; the higher the resistance level, the greater the reserves. Exposure of these populations to controlled atmosphere was associated with a steady utilization of the reserves. By contrast, the unselected population responded to controlled atmospheres by accelerated utilization of triacylglycerol and polysaccharides. Comparison of the utilization rates during CO2 exposure showed that triacylglycerol is the main energy source, and polysaccharides contribute to metabolic energy supply only to a small extent.  相似文献

Abstract. The presence or absence of epistasis, or gene interaction, is explicitly assumed in many evolutionary models. Although many empirical studies have documented a role of epistasis in population divergence under laboratory conditions, there have been very few attempts at quantifying epistasis in the native environment where natural selection is expected to act. In addition, we have little understanding of the frequency with which epistasis contributes to the evolution of natural populations. In this study we used a quantitative genetic design to quantify the contribution of epistasis to population divergence for fitness components of a native annual legume, Chamaecrista fasciculata . The design incorporated the contrast of performance of F2 and F3 segregating progeny of 18 interpopulation crosses with the F1 and their parents. Crosses were conducted between populations from 100 m to 2000 km apart. All generations were grown for two seasons in the natural environment of one of the parents. The F1 often outperformed the parents. This F1 heterosis reveals population structure and suggests that drift is a major contributor to population differentiation. The F2 generation demonstrated that combining genes from different populations can sometimes have unexpected positive effects. However, the F3 performance indicated that combining genes from different populations decreased vigor beyond that due to the expected loss of heterozygosity. Combined with previous data, our results suggest that both selection and drift contribute to population differentiation that is based on epistatic genetic divergence. Because only the F3 consistently expressed hybrid breakdown, we conclude that the epistasis documented in our study reflects interactions among linked loci.  相似文献

Glycinebetaine (GB) is a compatible solute synthesized by species in a number of plant families, including some grasses such as sorghum ( Sorghum bicolor L. Moench). A recombinant-inbred (RI) population was developed from a cross between GB-deficient (IS2319) and GB-accumulating (P932296) sorghum genotypes. The proportions of GB-accumulating lines in both the F5 and F7:8 generations of this RI population were higher than expected, possibly due to some beneficial effect of GB accumulation on plant growth, survival or seed set. GB levels varied widely in lines of the RI population that were grown under controlled conditions, suggesting genetic control not only for the presence or absence of GB, but also for the level of GB. This hypothesis was tested by analysing individual plants from lines identified as accumulating low, medium or high concentrations of GB based on the F5 and F7:8 screens. The level of GB was conserved within lines, supporting the hypothesis of genetic control of relative GB levels within accumulating lines. Two pairs of near-isogenic lines (NILs) with contrasting GB levels within pairs were developed from the RI population. The stable inheritance of the GB phenotype and isogenicity of these NILs were confirmed with progeny tests and molecular marker analysis, respectively. Labelling studies demonstrated that the deficiency in GB accumulation was at the choline oxidation step.  相似文献

In order to test whether there is genetic coadaptation in geographic populations of Drosophila bipectinata with respect to body size, reciprocal crosses were made among five strains derived from ecogeographically different localities in India. Wing length was used as an index of body size, and was measured in all the five strains, and their crosses in F and F2 generations. The statistical analysis of the data show that there is significant interpopulation variation in body size and in all the crosses, there is an increase in body size in F1 generation when compared with mid-parent value. Further, there is a decrease in body size in F2 generation as compared to F1 in most of the crosses with increased variability. These results provide evidence for genetic coadaptation in geographic populations of D. bipectinata .  相似文献

Abstract. Twenty cultivars of spring wheat were examined for variation in abscisic acid (ABA) accumulation following partial dehydration of excised leaves. A 3-fold range of ABA concentration was obtained.
A cross between two cultivars which differed in drought-induced ABA accumulation was used to study the heritability of ABA accumulation and to develop lines differing in their capacity to accumulate ABA. Broad sense heritability was 0–32 between the F2 and F3 generations and 0–70 between the F3 and F4 generations. Apparent homozygosity for ABA accumulation was achieved in several selections at the F4. The possible significance for drought resistance of differences in capacity to accumulate ABA is discussed.  相似文献

Abstract. The metabolic rates, as expressed by oxygen (O2) consumption, carbon dioxide (CO2) production, and losses in wet and dry weights, were examined for adults of three strains of the red flour beetle Tribolium castaneum (Herbst), during exposure to two modified atmospheres (MAs). Exposure of a strain selected for resistance over twenty-one generations to an atmosphere of 65% CO2, 20% O2 and the balance nitrogen (N2), termed a high carbon dioxide concentration atmosphere (HCC) and exposure of an unselected strain to HCC, showed considerable levels of aerobic metabolism during exposure. For the unselected strain water loss and mobilization of energy reserves were rapid and mortality was followed by rapid desiccation. For the HCC-resistant strain water balance was maintained and energy reserves were utilized more slowly over a prolonged period. Exposure of a strain selected for resistance over twenty-one generations to a low oxygen concentration atmosphere (LOC) of 0.5% O2 in N2, and an unselected strain to LOC, revealed that even at 0.5% O2, metabolism was largely aerobic in both strains. Maintenance of water balance was not a major factor causing mortality of either strain during exposure to LOC. In air, metabolic rates of both the resistant strains were lower than that of the unselected strain.  相似文献

An F3 resource population originating from a cross between two divergently selected lines for high (D+ line) or low (D− line) body weight at 8-weeks of age (BW55) was generated and used for Quantitative Trait Locus (QTL) mapping. From an initial cross of two founder F0 animals from D(+) and D(−) lines, progeny were randomly intercrossed over two generations following a full sib intercross line (FSIL) design. One hundred and seventy-five genome-wide polymorphic markers were employed in the DNA pooling and selective genotyping of F3 to identify markers with significant effects on BW55. Fifty-three markers on GGA2, 5 and 11 were then genotyped in the whole F3 population of 503 birds, where interval mapping with GridQTL software was employed. Eighteen QTL for body weight, carcass traits and some internal organ weights were identified. On GGA2, a comparison between 2-QTL vs. 1-QTL analysis revealed two separate QTL regions for body, feet, breast muscle and carcass weight. Given co-localization of QTL for some highly correlated traits, we concluded that there were 11 distinct QTL mapped. Four QTL localized to already mapped QTL from other studies, but seven QTL have not been previously reported and are hence novel and unique to our selection line. This study provides a low resolution QTL map for various traits and establishes a genetic resource for future fine-mapping and positional cloning in the advanced FSIL generations.  相似文献

Convict cichlids, Cichlasoma nigrofasciatum , inbred for four and five generations exhibited moderate and severe morphological deformities. Deformed fishes were characterized by a more vertically sloping forehead, irregular jaw, hyoid, opercula and fin structure, reduced growth and irregular swimming behaviour. Inbred fish (F4, F5) did not differ from F1s in their age of maturation or the average size of their first broods. Brood survival was 18.7% in F5 inbred fish, compared to 75.5% in F1s.  相似文献

Spanish and Swedish populations of Helophorus lapponicus are shown on fossil evidence to have been isolated from one another for about 10 000 years, since the end of the Pleistocene. When cross-bred they show some evidence of heterosis in the F1, with reduced viability in the F2 and back-cross generations. The level of compatibility of these populations is similar to that found when different populations of Lepidoptera species are crossed, and quite different from the incompatibility revealed by studies of interspecific hybridization. The degree of evolutionary stability revealed is consistent with the apparent absence of evolution revealed by studies of Pleistocene fossil Coleoptera.  相似文献

Abstract.  1. Changes in the frequency of Cry1Ac resistance genes and shifts in tolerance of cotton bollworm, Helicoverpa armigera , to the Cry1Ac toxin were assessed using bioassays of F1 and F2 offspring of isofemale lines from Anci County of Hebei Province (a multiple-crop system including corn, soybean, peanut, and Bt cotton) and Xiajin County of Shandong Province (an intensive Bt cotton planting area) in Northern China during 2002–2005.
2. A conservative analysis of the overall results indicated that there was a small increase in the frequency of major, non-recessive resistance genes over time.
3. The relative average development ratings [RADR – growth rate of a line on a Bt diet in proportion to the growth rate on a non-Bt (NBT) diet] of the bollworm larvae in F1 tests increased significantly from year to year, indicating a gradual trend towards higher tolerance to Cry1Ac in the field populations.
4. There were also significant positive correlations between RADR of the lines in the F1 generation and the RADR of their F2 offspring, indicating that the tolerance was genetically based.
5. Quantitative genetic simulation analysis showed that resistance of H . armigera to Bt cotton in Xiajin could evolve to a high level in 11–15 years if no effective resistance management measures are carried out.  相似文献

Hierarchy is the main criterion for informativeness in a data set, even if no explicit reference to evolution as a causal process is provided. Sequence data (nuclear ribosomal DNA ITS) from Armeria (Plumbaginaceae) contains a certain amount of hierarchical structure as suggested by data decisiveness and distribution of tree lengths. However, ancillary evidence suggests that extensive gene flow and biased concerted evolution in these multicopy regions have significantly shaped the ITS data set. This argument is discussed using parsimony analysis of four data sets, constructed by combining wild sequences with those from different generations of artificial hybrids (wild + F1, F2, and backcrosses; wild + backcrosses; wild + F1; wild + F2). Compared to the F1 hybrids, F2 show a certain degree of homogenization in polymorphic sites. This effect reduces topological disruption caused by F1 and is considered to be illustrative of how extensive gene flow and biased concerted evolution may have modeled the wild ITS data. The possibility that hierarchy has arisen as a result of—or despite a significant contribution from—those two such potentially perturbing forces raises the question of what kind of signal are we recovering from this molecular data set.  相似文献

Supernatant malate dehydrogenase (MDH) isozymes (as visualized by starch gel electrophoresis) are encoded by two distinct gene loci in both the largemouth and smallmouth bass. When an interspecific F1 hybrid is formed between these two fish, a unique MDH isozyme is generated. The results of freeze-thaw molecular hybridization (which is the first application of this technique to MDH) indicate that this unique isozyme in the F1 hybrid is a heterodimer composed of one subunit of each parental type. The F1 hybrids produced F2 hybrids which in turn formed the F3 hybrid population. The inheritance of alleles at the MDH-B locus is consistent with a single Mendelian autosomal locus. Furthermore, there is no evidence of linkage between the lactate dehydrogenase-E locus and the MDH-B locus.  相似文献

Abstract:  Against the background of conflicting reports of variable sex ratio distribution in different populations of the African rice gall midge (AfRGM), Orseolia oryzivora Harris and Gagne, the number and sex ratios of F1 adult progeny produced per female from wild and screen house populations were investigated in the screen house at 27 ± 3°C, 60–70% relative humidity and 12 : 12 h (light : dark) photoperiod. Results indicated the occurrence of a single sex-biased ratio in all the F1 adult progeny produced per female of this pest. Regardless of the source, whether from wild or screen house population, each mated female AfRGM produced a full complement of either sex only but never both. The difference between the number of eggs laid and F1 adult progeny that emerged per female from wild and screen house populations was not significant (P > 0.05), indicating consistency in the occurrence of this phenomenon among AfRGM populations. The reason for this could not be ascertained in this study but AfRGM eggs must be fertilized for development and eclosion to occur. No F2 adults emerged when a cohort of emergents from the same female were allowed to re-infest susceptible seedlings, indicating absence of parthenogenesis.  相似文献

High salt stresses were exerted upon seedlings of the common variety of Rhodes grass ( Chloris gayana Kunth.) in order to select a salt resistant strain and to ascertain the characteristics which differentiate between the original parent population and the currently selected one. Five generations of seedlings were grown on sand culture and irrigated with NaCl solutions up to 0.7 M . The most successful survivors of each generation were selected and grown to maturation and seed production. Plants of the F5-selected population showed a marked improvement in their survival and regeneration capabilities after harvesting. Some effects of selection were found in seed germination and in water use. No apparent differences between the parent and the F5 populations were found in growth of unclipped plants, ion content or protein synthesis. The data presented indicate that the capability to survive through salt stresses can be raised in Rhodes grass by mass selection. Fundamentally this is achieved by improving the regeneration capability of the plants after multiple clippings.  相似文献

To characterize the genetic basis of voluntary calcium consumption, we tested C57BL/6J mice (B6; with low avidity for calcium), PWK/PhJ mice (PWK; with high avidity for calcium) and their F1 and F2 hybrids. All mice received a series of 96-h two-bottle preference tests with a choice between water and the following: 50 m m CaCl2, 50 m m calcium lactate, 50 m m MgCl2, 100 m m KCl, 100 m m NH4Cl, 100 m m NaCl, 5 m m citric acid, 30 μ m quinine hydrochloride and 2 m m saccharin. Most frequency distributions of the parental and F1 but not F2 groups were normally distributed, and there were few sex differences. Reciprocal cross analysis showed that B6 × PWK F1 mice had a non-specific elevation of fluid intake relative to PWK × B6 F1 mice. In the F2 mice, trait correlations were clustered among the divalent salts and the monovalent chlorides. A genome screen involving 116 markers showed 30 quantitative trait loci (QTLs), of which six involved consumption of calcium chloride or lactate. The results show pleiotropic controls of calcium and magnesium consumption that are distinct from those controlling consumption of monovalent chlorides or exemplars of the primary taste qualities.  相似文献

Marek's disease (MD) is a lymphoproliferative disease caused by a member of the herpesvirus family, and the best understood genetic resistance to MD involves the chicken major histocompatibility complex (MHC) B -complex. Preliminary observations have suggested that MHC-like Rfp-Y genes might also influence the incidence of MD. This study describes the differentiation and definition of unique Rfp-Y genes in inbred lines 63 and 72, lines that possess identical B -complex genes, but that are resistant or susceptible to MD, respectively. To assess if Rfp-Y genes affect susceptibility to MD, 265 63× 72 F2 chickens were challenged with the JM strain of MD virus at 1 week of age and were evaluated for MD lesions at up to 10 weeks of age. Genotyping of the F2 chickens for Rfp-Y haplotypes was performed by restriction fragment length polymorphism analysis of genomic DNA using Taq I and a B-FIV probe. Analysis of variance and interval mapping procedures were used to determine association between the Rfp-Y haplotypes and the phenotypic MD values of the F2 chickens. The cosegregation analysis of 265 F2 chickens indicated that there was no association between Rfp-Y haplotypes and MD susceptibility. Furthermore, the fact that the Rfp-Y haplotypes fit the 1:2:1 segregation ratio and the Rfp-Y allele frequencies did not differ significantly from 0·5 in the full population or in selected subpopulations (of either 40 MD-resistant or 39 MD-susceptible chickens) also indicated that Rfp-Y haplotypes do not significantly influence MD susceptibility. We conclude that Rfp-Y haplotypes do not play a major role in determining the genetic susceptibility to MD in 63× 72 F2 White Leghorn chickens.  相似文献

Genetic analysis of resistance to the inhibitory effects of UV radiation on growth of rice ( Oryza sativa L.) cultivars was carried out. Some experimental plants were grown in visible radiation supplemented with UV radiation containing a large amount of UV-B and a small amount of UV-C in a phytotron, while others were grown without UV radiation. The degree of resistance to UV radiation was estimated in terms of the degree of reduction caused by supplemental UV radiation in the fresh weight of the aboveground plant parts and the chlorophyll content per unit fresh weight. Fresh weight and chlorophyll content in F2 plants generated by reciprocally crossing cv. Sasanishiki, a cultivar more resistant to UV radiation, and Norin 1, a cultivar less resistant to such radiation exhibited a normal frequency distribution. The heritabilities of these two properties in F2 plants were low under conditions of non-supplemental UV radiation. Under elevated UV radiation, the F2 population shifted to the lower range of fresh weight and chlorophyll content, and the means were close to those of Norin 1. The heritabilities of these two properties were the same in the reciprocal crosses, indicating that maternal inheritance was not involved. Inheritance of chlorophyll content per unit fresh weight was further determined in F3 lines generated by self-fertilizing F2 plants of Sasanishiki and Norin 1. The results showed that the F3 population was segregated into three genotypes, namely, resistant homozygotes, segregated heterozygotes and sensitive homozygotes, with a ratio of 1:65:16.
It was thus evident that the resistance to the inhibitory effect of elevated UV radiation in these rice plants was controlled by recessive polygenes.  相似文献

Abstract:  Populations of Plutella xylostella L. with high (laboratory re-selected) and low levels (unselected) of resistance to the Bacillus thuringiensis ( B.t. ) toxin Cry1Ac were maintained under controlled environment conditions in the absence of any further selection pressure. Larvae were maintained either in discrete generations on freshly excised leaves replaced ad lib (standard culture system) or in microcosm cages holding up to 12 potted Chinese cabbage plants, allowing progressively overlapping generations of insects. After 12 generations in the microcosm cages, the LC50 values of both selected and unselected populations were not significantly different from those of a standard susceptible population. Under standard culture conditions, the LC50 value for unselected insects after 12 generations was almost identical to that of unselected insects in the microcosm cages, whereas the LC50 value for selected insects was 1250-fold greater. The fecundity of the selected population was significantly lower compared with that of the same population in the microcosm cages after 12 generations. There was no significant change in the fecundity of the unselected population after 12 generations in the microcosm cages. The results show, that differences in the culture system (microcosm vs. standard) and in the resistance level (re-selected vs. unselected) can have a marked influence on the stability of resistance to a B.t. toxin in an insect population. Possible factors involved in determining the rate at which resistance declines are discussed.  相似文献
