Soils previously treated with landfarming to reduce petroleum hydrocarbon concentrations are often left with a less biodegradable residual fraction that can present challenges for additional treatment. Four possible polishing technologies were tested on the bench scale for weathered hydrocarbons present in fine-grain soils obtained from a previously landfarmed area at an active oil refinery. The technologies included additional bioremediation (both biostimulation and bioaugmentation tested), soil washing, chemical oxidation, and low-temperature thermal desorption. Multiple parameters were tested separately for each technology to identify possible factors that were relevant across technologies. Extractable hydrocarbons comprised only approximately 35% of the organic carbon in the soils, and this component was considerably less affected by biological, surfactant, and oxidant treatment than organic materials that are not quantified by the TEH analysis. Treatment testing of thermal desorption indicated removal of large quantities of extractable hydrocarbons despite the presence of high organic matter. The additional demand to the system would likely result in considerably large timeframes (biological treatment), reagent quantities (soil washing and oxidation), or energy input (thermal desorption) for treatment of target hydrocarbons on a full scale.  相似文献

The need for microbial cell disruption has hindered the large scale production of commercial biotechnological products of intracellular derivation. The intracellular nature of many recombinant products and the potential use of the bacterial storage product, PHB as a commodity thermoplastic have renewed interest in the improvement of this unit operation. This paper provides a review of processes of a mechanical, physical, chemical or biological nature used for cell disruption on both the laboratory and large scale. Applicability of the techniques to large scale operation is discussed. Modification of existing processes is suggested for the reduction of energy requirements and improved process economics. The requirements for the liberation of granular intracellular products such as inclusion bodies and virus-like yeast particles are distinguished from those for the liberation of soluble products, mainly proteinaceous in nature. The integrated nature of the process with both upstream and downstream processes is addressed. Finally, the recent approach of selective liberation of soluble products of interest is reviewed.  相似文献

Hexachlorocyclohexane (HCH) contaminated soils were treated for a period of up to 64 days in situ (HCH dumpsite, Lucknow) and ex situ (University of Delhi) in line with three bioremediation approaches. The first approach, biostimulation, involved addition of ammonium phosphate and molasses, while the second approach, bioaugmentation, involved addition of a microbial consortium consisting of a group of HCH-degrading sphingomonads that were isolated from HCH contaminated sites. The third approach involved a combination of biostimulation and bioaugmentation. The efficiency of the consortium was investigated in laboratory scale experiments, in a pot scale study, and in a full-scale field trial. It turned out that the approach of combining biostimulation and bioaugmentation was most effective in achieving reduction in the levels of α- and β-HCH and that the application of a bacterial consortium as compared to the action of a single HCH-degrading bacterial strain was more successful. Although further degradation of β- and δ-tetrachlorocyclohexane-1,4-diol, the terminal metabolites of β- and δ-HCH, respectively, did not occur by the strains comprising the consortium, these metabolites turned out to be less toxic than the parental HCH isomers.  相似文献

The effectiveness of bioremediation is often a function of the microbial population and how they can be enriched and maintained in an environment. Strategies for inexpensive in situ bioremediation of soil contaminated with petroleum hydrocarbons include stimulation of the indigenous microorganisms by introduction of nutrients (biostimulation) and/or through inoculation of an enriched mixed microbial culture into soil (bioaugmentation). To demonstrate the potential use of bioremediation in soil contaminated with kerosene, a laboratory study with the objective of evaluating and comparing the effects of bioattenuation, biostimulation, bioaugmentation, and combined biostimulation and bioaugmentation was performed. The present study dealt with the biodegradation of kerosene in soil under different bioremediation treatment strategies: bioattenuation, biostimulation, bioaugmentation, and combined biostimulation and bioaugmentation, respectively. Each treatment strategy contained 10% (w/w) kerosene in soil as a sole source of carbon and energy. After 5 weeks of remediation, the results revealed that bioattenuation, bioaugmentation, biostimulation, and combined biostimulation and bioaugmentation exhibited 44.1%, 67.8%, 83.1%, and 87.3% kerosene degradation, respectively. Also, the total hydrocarbon-degrading bacteria (THDB) count in all the treatments increased with time up till the second week after which it decreased. The highest bacterial growth was observed for combined biostimulation and bioaugmentation treatment strategy. A first-order kinetic model equation was fitted to the biodegradation data to further evaluate the rate of biodegradation and the results showed that the specific degradation rate constant (k) value was comparatively higher for combined biostimulation and bioaugmentation treatment strategy than the values for other treatments. Therefore, value of the kinetic parameter showed that the degree of effectiveness of these bioremediation strategies in the clean up of soil contaminated with kerosene is in the following order: bioattenuation < bioaugmentation < biostimulation < combined biostimulation and bioaugmentation. Conclusively, the present work has defined combined biostimulation and bioaugmentation treatment strategy requirements for kerosene oil degradation and thus opened an avenue for its remediation from contaminated soil.  相似文献

The results obtained during the laboratory remediation modeling of oil-containing waste (652 g/kg) and natural radionuclides (Ra226, Th232, and K40) using the methods of landfarming, biostimulation, and bioaugmentation are given in the article. It is found that landfarming and biostimulation decrease the content of oil products and phytotoxicity of the waste. Landfarming is the most rapid process.  相似文献

Bioremediation, involving bioaugmentation and/or biostimulation, being an economical and eco-friendly approach, has emerged as the most advantageous soil and water clean-up technique for contaminated sites containing heavy metals and/or organic pollutants. Addition of pre-grown microbial cultures to enhance the degradation of unwanted compounds (bioaugmentation) and/or injection of nutrients and other supplementary components to the native microbial population to induce propagation at a hastened rate (biostimulation), are the most common approaches for in situ bioremediation of accidental spills and chronically contaminated sites worldwide. However, many factors like strain selection, microbial ecology, type of contaminant, environmental constraints, as well as procedures of culture introduction, may lead to their failure. These drawbacks, along with fragmented literature, have opened a gap between laboratory trials and on-field application. The present review discusses the effectiveness as well as the limitations of bioaugmentation and biostimulation processes. A summary of experimental studies both in confined systems under controlled conditions and of real case studies in the field is presented. A comparative account between the two techniques and also the current scenario worldwide for in situ biotreatment using bioaugmentation and biostimulation, are addressed.  相似文献

The aim of this study was to investigate the removal of ibuprofen in laboratory scale constructed wetlands. Four (planted and unplanted) laboratory‐scale horizontal subsurface flow constructed wetlands were supplemented with ibuprofen in order to elucidate (i) the role of plants on ibuprofen removal and (ii) to evaluate the removal performance of a bioaugmented lab scale wetland. The planted systems showed higher ibuprofen removal efficiency than an unplanted one. The system planted with Juncus effusus was found to have a higher removal rate than the system planted with Phalaris arundinacea. The highest removal rate of ibuprofen was found after inoculation of gravel previously loaded with a newly isolated ibuprofen‐degrading bacterium identified as Sphingobium yanoikuyae. This experiment showed that more than 80 days of CW community adaptation for ibuprofen treatment could be superseded by bioaugmentation with this bacterial isolate.  相似文献

This review paper addresses the role of green roofs in urban drainage considering both management of water quantity and quality. Results from investigation of full scale installations as well as from laboratory models are reviewed. The following factors affecting runoff dynamics from green roofs are discussed: type of green roof and its geometrical properties (slope); soil moisture characteristics; season, weather and rainfall characteristics; age of green roof; vegetation. Design parameters as suggested by different authors are also reviewed. Factors which affect influence of a green roof on runoff water quality are discussed in general terms followed by the review of data regarding concentrations of phosphorus, nitrogen, and heavy metals in green roof runoff, its’ pH, and first flush effect. Linking among fertilization, runoff pollution and vegetation development is given a particular focus. The review indicates clearly that there is a need for more research into a green roof performance in an urban environment. The differences measured by few existing studies between the early years performance of green roofs and the later years indicate a need for long term monitoring of green roofs.  相似文献

The conditions necessary for the establishment and maintenance of Enhanced Biological Phosphate Removal (EBPR) from wastewaters are discussed in the light of our inability to achieve levels of EBPR from artificial sewage in a laboratory‐scale system. Adequate levels of P removal and polyP accumulation by sludge biomass could only be restored by the imposition of stringent anaerobiosis (Eh < –120 mV) and by increasing the short chain fatty acid composition of the influent. Subsequent laboratory‐scale investigations into several possible alternative strategies to achieve enhanced levels of P removal and polyP accumulation from artificial sewage medium indicated that a reduction in the operational pH of the system to approximately 5.5 could achieve comparable levels of P removal under fully‐aerobic conditions. Acid stimulated P uptake and polyP formation might serve as the basis of novel alternative technologies for eutrophication control at wastewater treatment facilities.  相似文献

污泥厌氧发酵生产挥发性脂肪酸相较产甲烷,是更具应用价值的污泥稳定途径及资源化利用方式,得到国内外学者的普遍重视。考虑到产酸量低和产酸过程的不稳定性是限制污泥发酵产酸的主要问题,采用生物强化方法实现挥发性脂肪酸的大量积累,与物理和化学方法相比,具有成本低、无二次污染等优点。根据生物强化制剂的类型,归纳了微生物纯培养物、微生物混合培养物及生物酶强化对污泥厌氧发酵产酸的影响,并在此基础上对生物强化技术控制污泥定向产酸、调控奇偶数碳比率等方面的应用进行讨论。此外,分析了影响挥发性脂肪酸产量和组分的因素,如pH、温度、底物、水力停留时间和污泥龄等。最后对生物强化技术的发展方向进行了展望,以期为深入探究污泥资源化利用提供借鉴。  相似文献

The performance of a laboratory scale rotating biological contactor (RBC) towards shock loadings of 2-fluorophenol (2-FP) was investigated. During a period of ca. 2 months organic shock loadings of 25 mg L?1 of 2-FP were applied to the RBC. As no biodegradation of 2-FP was observed, bioaugmentation of the RBC with a 2-FP degrading strain was carried out and, along ca. 6 months, organic shock loadings within a range of 25-200 mg L?1 of 2-FP were applied. Complete biodegradation of 50 mg L?1 of 2-FP was observed during operation of the reactor. The RBC showed to be robust towards starvation periods, as after ca. 1month of non-supply of the target compound, the reactor resumed 2-FP degradation. The inoculated strain was retained within the biofilm in the disks, as the 2-FP degrading strain was recovered from the biofilm by the end of the experiment, thus bioaugmentation was successfully achieved.  相似文献

The use that 30 general practitioners in four group practices made of open access laboratory and radiological facilities was studied for one year. We were particularly interested in whether general practitioners hoped to exclude rather than confirm abnormality when requesting investigations. All but two of the general practitioners studied used investigations to exclude abnormality to a greater extent than to confirm it. The rate at which the practices investigated patients and the number of investigations requested were appreciably different and were different for individual general practitioners, part time general practitioners requesting more investigations than trainees and full time general practitioners. Haematological investigations accounted for over 30% of requests for investigations in all but one practice, biochemical investigations being requested as often as bacteriological investigations in two of the four practices. The ratio of expected to unexpected results varied among general practitioners; no general practitioner had more unexpected results and the range of ratios was similar for full time and part time general practitioners and for trainees. The proportion of patients with abnormalities uncovered by each practice increased disproportionately as the use of investigations increased, supporting a higher rather than lower rate of investigation among general practitioners. Compared with the results of other studies the use of the radiological facilities available was low. X ray examinations of the skeleton were requested more than chest and contrast media examinations by three of the four practices. Most x ray examinations were used to exclude rather than confirm abnormality by all the practices, with over 85% of results confirming the general practitioner''s initial diagnosis.  相似文献

In biotechnological processes, often cell disruption has been an inevitable step as current host cells express most of the desired products intracellularly. Thus, an appropriate cell disruption technique must be selected considering different factors including the target product, process scale, and cell wall structure. In the current study, as a novel method, the efficacy of cell disruption via laser was tested qualitatively and quantitatively in batch and continuous systems, respectively. Laser-induced cell lysis can be a clean, rapid and convenient alternative to the other conventional disruption techniques. Our investigations in the continuous system with a flow rate of 800 μL/sec proved efficient (~ 90%) Pichia pastoris cell disruption at the wavenumber 1,064 nm with the energy input of 284 mW after four complete rounds of circulation. The main mechanism of cell disruption is assumed to be thermolysis via instant heat increase in the laser-treated spot. The results of the current study showed that continuous laser system could be applied in laboratory and industry scale for cell disruption.  相似文献

Animal tissue is likely to continue to be an important source of enzymes and protein hormones well into the 21st century. Aqueous phase systems show considerable potential and specific advantages for extractive purification of proteins from animal tissue. Although no industrial process is yet in place for commercial production of a protein from animal tissue, the potential for the system has, however, been demonstrated at laboratory scale for a number of enzymes, and at pilot scale for a few, using simple phase systems and also affinity partitioning systems. In this paper we demonstrate how broad scale comparative physiology has an important role to play in informing a variety of assumptions made in macroecology. We do so by examining large-scale geographic variation in insect development, thermal tolerance and metabolic rate. From these studies, and those from the literature on insect water loss and thermoregulation, we show that there is often a bias to the geographic extent of available empirical data. Studies of cold hardiness are most usually undertaken at high latitudes, while investigations of upper thermal tolerances and water loss are most common in warm arid regions. Likewise, we demonstrate that much variation in insect physiological tolerances is partitioned at higher taxonomic levels, which has important implications for comparative physiology. Intriguingly, data on the full range of variables we review are available for only three species. We also show that, despite its importance, body size is regularly reported in only some kinds of investigations (metabolic rate, water loss rate), whereas in others (upper lethal temperature, cold hardiness, development) this variable is often ignored. In short, although large-scale comparative physiology can contribute considerable understanding to both physiology and ecology, there is much that remains to be done.  相似文献

Abstract

Humans have utilised plant derived natural products as medicines for millenia. Moreover, many contemporary pharmaceuticals are also natural products or derivatives thereof. However, the full potential of these compounds remains to be exploited because often they are: complex and difficult to synthesise; found in low quantities; produced by undomesticated and sometimes rare plants; and, their synthesis is routinely influenced by weather conditions. Potentially, the in vitro culture of cells from the corresponding plant species could circumvent some of these problems but the growth of plant cells on an industrial scale is also problematic. The recent isolation and culture of cambial meristematic cells (CMCs), stem cells which ordinarily generate the plant vasculature, may now provide a key platform technology to help realise the full potential of plant natural products.  相似文献

The numbers of vaginal swabs, faecal specimens, throat swabs, and urine specimens submitted by 104 city general practitioners to the bacteriology laboratory at Aberdeen differed widely. The doctors who made most use of the laboratory service rarely did so equally for all four investigations but usually for only one or two. Similarly, those who used the service least often made frequent use of one particular investigation. Further studies are needed to identify and evaluate the reasons for the apparently substantial disagreement among general practitioners concerning the value of these diffferent bacteriological investigations in general practice.  相似文献
