Noradrenaline induced vasodilation in isolated perfused eel or trout gills was inhibited by propranolol, aβ-adrenergic blocking agent. The noradrenaline effect was much reduced in gills of fish which had been kept in 1 mg/l solutions of the detergent LAS (linear alkylate sulphonate; 12C chain length). LAS produced concentration-dependent vasodilation, which was inhibited by propranolol, in eel and trout gills. Noradrenaline produced additional vasodilation in eel and trout gills which had been dilated by 2.10−8 m LAS but had no effect on trout gills perfused with 2.10−7 m LAS. Gills from trout which had been kept in 0.1 mg l-1 LAS responded to noradrenaline normally but the propranolol inhibition appeared to be potentiated.  相似文献

Urotensin II (UII) was first discovered in the urophyses of goby fish and later identified in mammals, while urotensin II-related peptide (URP) was recently isolated from rat brain. We studied the effects of UII on isolated heart preparations of Chinook salmon and Sprague–Dawley rats. Native rat UII caused potent and sustained, dose-dependent dilation of the coronary arteries in the rat, whereas non-native UII (human and trout UII) showed attenuated vasodilation. Rat URP dilated rat coronary arteries, with 10-fold less potency compared with rUII. In salmon, native trout UII caused sustained dilation of the coronary arteries, while rat UII and URP caused significant constriction. Nω-nitro-l-arginine methyl (l-NAME) and indomethacin significantly attenuated the URP and rat UII-induced vasodilation in the rat heart. We conclude that UII is a coronary vasodilator, an action that is species form specific. We also provide the first evidence for cardiac actions of URP, possibly via mechanisms common with UII.  相似文献

Cannulated brown trout, Salmo trutta , were exposed for 36 h to synthetic water with a low calcium content of pH 5 and similar synthetic water dosed with aluminium to raise the filterable A1 from 5 to 290 μg 1−1 over the 36-h period. There were no significant disturbances of plasma concentrations of glucose, cortisol or catecholamine (adrenaline and noradrenaline) in fish held in water of pH 5. The addition of aluminium to this acidic synthetic water resulted in a generalized endocrine stress response with a four-fold increase in plasma glucose concentration after 18 h and a significant increase in plasma cortisol concentration from 24 h onwards when filterable A1 exceeded 200 μg 1−1. Plasma catecholamine concentration indicated an adrenergic stress response in aluminium-exposed brown trout. A transient doubling in noradrenaline after 6 h in A1 was followed by a larger increase in both plasma adrenaline and noradrenaline concentrations in fish surviving the 36-h exposure to A1.  相似文献

Rainbow trout, Oncorhynchus mykiss, were subjected to either physical stress (twice daily chasing to exhaustion for 5 days) or a period of 2 months of fasting. Following these treatments, the levels of catecholamines, adrenaline and noradrenaline, stored within the kidney and posterior cardinal vein (PCV) were determined. The ability of the catecholamine-storing chromaffin cells to release catecholamines in response to cholinergic stimulation was measured using an in situ saline-perfused PCV preparation. In the physically stressed fish, the concentration (μg catecholamine g?1 tissue) of noradrenaline within the anterior and middle thirds of the kidney increased; the concentration of adrenaline was unchanged in all tissues. The content (μg) of noradrenaline or adrenaline, within the various tissues, was similar in both groups of fish with the exception of a higher noradrenaline content in the middle third of the kidney in the physically stressed fish. The total catecholamine content (μg catecholamine) of these tissues (kidney+PCV) was unaffected by physical stress. With the exception of a lower concentration of adrenaline in the middle third of the kidney, the concentrations of catecholamines were unaffected by fasting. There was a trend towards a greater content (μg) of noradrenaline within all of the tissue regions of the fed fish, however, a significant difference was only observed in the anterior third of the kidney. The content of adrenaline in the fed fish was greater in all regions of the kidney as well as the middle third of the PCV. The total catecholamine content (kidney + PCV) was lower in the fasted fish owing to significantly lower PCV and kidney masses. Prolonged physical stress caused a decrease in the responsiveness of the chromaffin cells to the cholinoceptor agonist carbachol (10?8 to 10?4mol). The ED50 (the dose of carbachol required to elicit a half maximal response) for catecholamine release was 0·96 ± 10?6mol carbachol in the physically stressed fish and 0·84 ± 10?7 in the control fish. Fasting did not alter the pattern of catecholamine release. The ED50 values were 0·96 ± 10?7 and 1·24 ± 10?7 mol for fasted and fed fish, respectively. Thus, a physical stress affected both catecholamine storage and release whereas fasting affected only storage and not the release process.  相似文献

Sympathetic activation leading to increased levels of blood catecholamines, and stimulation of the hypothalamic-pituitary inter-renal axis leading to increased cortisol, are difficult to avoid when handling animals. Yet, in research on effects of acute stress, elicitation of such responses must be minimized in the control groups. The work examines means to achieve a minimally disturbed state in rainbow trout (Oncorhynchus mykiss). Level of arousal was determined by adrenaline and cortisol concentrations in plasma, and by the spleen:somatic index. Fish were prepared for bleeding by rapid capture and concussion, by infusion of anesthetic into the undisturbed home tank, by confinement in black boxes, or by being fed alpha- and beta-receptor antagonists. Even when done quickly, netting and concussion yielded fish with ca. 200-pmol adrenaline/ml plasma. Cortisol was elevated (to > 10 ng/ml) within 30 s of stress initiation. Surreptitious infusion of anesthetic (2-phenoxyethanol, PE) into tanks yielded fish with lower adrenaline levels (means 19.34 and 19.58 pmols/ml in home tank and black boxes, respectively). Among fish given phentolamine and propranolol, spleen:somatic indices and plasma adrenaline were higher than in diet controls, whether undisturbed or stressed, indicative of successful receptor blockade. Since careful infusion of 2-PE yielded the lowest adrenaline levels, and requires no special apparatus, it is the method of choice for obtaining minimally stressed fish.  相似文献

Linear alkyl benzene sulphonate (LAS), one of the main ingredients used in synthetic detergents to enhance their cleansing properties. Indiscriminate and untreated discharge of detergents and their residues in both lantic and lotic habitats pose a variety of ecological threats and also adversely affect aquatic fauna. In vivo, LAS metabolism and biotransformation occurs via monooxygenases in liver, leading to Reactive Oxygen Species, ROS, production and consequently oxidative stress by disturbing cellular antioxidant enzymatic equilibrium. Present study aims to evaluate the activities of two widely distributed antioxidant enzymes viz., catalase (CAT) and superoxide dismutase (SOD) and ROS induced histological impairments in liver of freshwater fish, Channa punctatus. For the estimation of oxidative stress and hepatic impairments, well acclimatized fishes were divided in three groups. Fish of group G1 serves as control whereas fish of the other two groups, G2 and G3 were exposed to two fractions, 1/20th and 1/10th of 96 h LC50 of LAS for 24, 48, 72 and 96 h of exposure periods. Our results showed a significant induction in CAT and SOD activities in liver tissue of C. punctatus in a dose and time dependent manner. ROS induced histopathological impairments in hepatic tissues are characterized by loosely arranged, irregularly distributed and degenerated hepatocytes with increased vacuolization and pyknotic nuclei. The results are quite suggestive that LAS intoxication generates oxidative stress by ROS production which brings about histopathological impairments in exposed fish.  相似文献

Movement rhythms in juvenile Atlantic salmon, Salmo salar L.   总被引:1,自引:0,他引:1  
Nocturnal downstrean migration of juvenile Atlantic salmon is usually interpreted as increased locomotor activity. The frequency of downstream passages of 0–1 + salmon in an endless stream channel was greater by night than by day in both smoking and non-smolting fish in autumn and spring. Movement increased at dusk, and decreased after dawn. Mature male 1 + fish moved slightly less than immatures in October, but significantly more in November. Total movement frequency was lower at full moon than at other moon phases, and movement was reduced when the moon was up. Under turbid conditions by day, the threshold water velocity inducing nett downstream movement was 8.2 cm s−1, and the relative velocity of fish swimming downstream was never more than one third that of fish holding station at the normal maximal flow of 25–30 cm s−1.
At the end of their first growing season in October, fish which had been offered food continuously through 24 h did not differ in size from those fed by day only, but the latter were significantly larger than those offered food only at night.
We conclude that: (1) the fish fed actively by day, and not by night; (2) station-holding represented activity, and downstream nocturnal movement represented relative inactivity (displacement) which occurred on loss of visual orientation, hence migration resulted from reduced activity; (3) lack of displacement in early autumn has adaptive value for maturing fish, but not for non-spawners.  相似文献

The production of Atlantic salmon gynogenomes by the combined use of a novel method for sperm irradiation and differently timed high hydrostatic pressure shocks is described. Sperm solutions were exposed to UV irradiation in a temperature-controlled flow-through device. Eggs fertilised with such sperm were exposed to shocks of 9500 psi at 30 min or approximately 7 h after fertilisation in order to produce meiotic and mitotic gynogenomes respectively. Yields of meiotic gynogenomes were generally high (up to 95%); those of mitotic gynogenomes were lower (range 2–20%). Analyses of the offspring by ploidy status and fingerprinting confirmed their gynogenetic origin. Small numbers of mitotic gynogenetic fish were grown on for 2 years in fresh and salt water. S1/S2 ratios were lower in gynogenetic fish and mean age at maturity was greater. Of the presumptive gynogenetic fish subjected to destructive sampling (n = 87) all were female.  相似文献

Summary Linear alkylbenzene sulfonate (LAS) is a widely used anionic surfactant. Although approximately 1 million metric tons of LAS are produced annually, relatively little is known about the bacteria or the genetic factors that control LAS degradation in the environment. The objectives of this research were to: i) compare bacterial populations in wastewater and pristine pond systems; ii) determine the frequency of plasmids in bacteria from these sites; and iii) compare the frequency of DNA sequences coding for aromatic catabolism in isolates from these two sites. Plate counts indicated that exposure to wastewater resulted in higher levels of both heterotrophic bacteria and bacteria capable of growing on LAS containing medium (LAS/YEPG). In addition to higher numbers, a higher proportion of heterotrophs from the wastewater system were capable of growth on LAS/YEPG medium. Thus, the high levels of LAS in the wastewater system apparently selected fro organisms that were able to tolerate and/or degrade, it. Mineralization of14C-ring labelled LAS in any habitat related to the presence of organisms that grew on LAS/YEPG. Although may of these isolates could carry out primary degradation, no isolate, could mineralize14C-ring LAS in pure culture. A higher incidence of plasmids was found in bacteria from the wastewater pond and among bacteria that grew on LAS containing medium. However, the presence of plasmid, DNA did not necessarily confer the ability to degrade LAS nor was the ability to degrade LAS dependent on the presence of a plasmid. The incidence of selected genotypes for aromatic catabolism was similar among isolates on LAS/YEPG at both sites, suggesting that LAS ring degradation may be present in other populations or encoded by alternative sequences. In conclusion, LAS mineralization is mediated by a consortium and the evidence that initial attack of LAS is plasmid mediated is inconclusive.  相似文献

The overwinter mortality rate of underyearling S2 Atlantic salmon parr (fish that will remain in fresh water for at least two years prior to smoltification) was almost ten times greater than that of S1 parr (fish that will smoltify after 1 year in fresh water) kept under similar conditions. The loss of condition ( K factor) of the S2 parr was proportionally greater than that of S1 parr during the early winter months and this coincided with a marked lymphocytopenia and thrombocytopenia in the S2 parr. Limited evidence indicates that S2 parr have chronically elevated blood cortisol levels during the winter months. It is suggested that chronic stress resulting (in part) from nutritional deficiency has a debilitating effect on the defence systems of these fish. The ultimate cause of mortality in most cases was bacterial fin-rot and/or fungal ( Saprolegnia ) infection. These findings are discussed in relation to the habitat requirements of overwintering salmonid fish.  相似文献

Hypo-osmoregulatory ability in juvenile Atlantic salmon, Sulrno salur L., was improved by cortisol treatment. Implantation of a vegetable shortening pellet containing cortisol (50 mg kg−1) resulted in elevated plasma cortisol titres. Maximum cortisol levels (160–170ngml−1) were observed at days 6 and 12 after the implantation and dropped significantly by day 55. Cortisol-implanted fish in fresh water developed a twofold increase in gill Na+/K+-ATPase activity at days 6 and 12, and a threefold increase by day 55. Intestinal mucosa Na+/K+-ATPase activity was not affected by cortisol. Cortisol-implanted fish exposed to 28 ppt sea water for 48 h tended to show an improved ability to regulate their plasma osmolarity and reduce their ionic load. The osmo-regulatory ability attained at days 12 and 55 was further evaluated by exposing fish to 37 ppt sea water for 96 h. While all the control fish died relatively early in these tests, cortisol-implanted fish showed a clear reduction in their mortality rate. These results indicate that cortisol can induce biochemical and organismal changes during winter months that typify preadaptive events normally occurring in the spring.  相似文献

Fall Chinook salmon Oncorhynchus tshawytscha were fed practical diets medicated with azithromycin (30 mg kg(-1) fish for 14 d) or erythromycin (100 mg kg(-1) fish for 28 d) either 1, 2, or 3 times beginning 14 d after initiation of exogenous feeding (February) and ending at smoltification (June). Average tissue concentrations of azithromycin increased from 19.0 microg g(-1) in fry to 44.9 microg g(-1) in smolts, and persisted in the tissues > 76 d after treatment ceased. Tissue concentrations of erythromycin were comparatively low, ranging from 0.2 microg g(-1) in fry to 10.4 microg g(-1) in smolts. Erythromycin was not detectable 21 d post-treatment. Neither antibiotic caused histopathologically significant lesions in the trunk kidney or other organ tissues. The high tissue concentrations and prolonged retention of azithromycin in Chinook may be factors that increase the efficacy of the antibiotic against Renibacterium salmoninarum, compared with erythromycin, particularly in early life history stages before covertly infected fish show clinical signs of disease.  相似文献

1. Coho salmon, exposed to sublethal levels of aqueous copper (1/4 and 1/2 LC50), lost appetite and ceased growing or showed decreased rates of growth. 2. Recovery of appetite and growth rate was faster in fish exposed to 1/4 of the LC50 than in those exposed to 1/2 of the LC50. 3. Copper levels were elevated in liver gill and kidney of exposed fish with the liver tissue accumulating a much larger amount of the metal than any other tissue. 4. The concentration of liver copper became constant at about the time that growth rate recovered. 5. The exposed fish exhibited much higher resistance to elevated aqueous copper levels than did the controls. 6. The results suggest that coho salmon may become acclimated to higher levels of copper and that acclimated fish are more tolerant to copper than control animals.  相似文献

The effects of three anaesthetics on induction and recovery were compared in Chinook salmon (Oncorhynchus tshawytscha). Heart rate (HR), cardiac output (Q), dorsal aortic pressure (DAP) and stroke volume (SV) were measured in minimally disturbed salmon during 5 min anaesthetic inductions with approximately equi-potent concentrations of MS222 (100 ppm), metomidate (6-10 ppm) and AQUI-S (60 ppm). MS222 induction caused a steady decline in DAP only, while metomidate induction did not affect any cardiovascular variable. AQUI-S caused a biphasic response, and within 2 min had depressed HR, Q, DAP and SV by between 20 and 50%. In the final 3 min HR returned to pre-anaesthesia levels, and Q and SV climbed to greater than pre-anaesthesia levels. Blood samples taken pre- and post-anaesthesia showed all inductions caused hypoxaemia (oxygen partial pressure of dorsal aortic blood (PaO2): MS222 47 mmHg, metomidate 35 mmHg, AQUI-S 21 mmHg). Haematocrit and plasma adrenaline and noradrenaline levels increased slightly in AQUI-S treated fish only. Recovery was monitored for 6 h post-anaesthesia, and was similar for each anaesthetic. All cardiovascular variables had returned to control levels within 5 min with the exception of DAP, which was initially slightly elevated (up to 20%) but returned to control values within 30 min. Anaesthesia is usually preceded by handling. Netting prior to anaesthesia caused significant increases in HR, Q and SV, which masked any anaesthetic dependent effects. Recovery from anaesthesia combined with surgery was also generally anaesthetic independent and recovery was prolonged, compared to anaesthesia alone. These data suggest limiting fish handling/manipulation is more important in minimising cardiovascular disturbance than the choice of anaesthetic.  相似文献

We isolated and characterized yeast mutants whose growth is sensitive to a local anestheticum tetracaine and, at the same time, temperature sensitive. These mutants were collectively called las mutants (local anestheticum sensitive). The las21 mutants were analyzed in this study. The wild type LAS21 gene was cloned by exploiting temperature sensitivity of the las21 mutants and we found that LAS21 encodes ORF YJL062w which has not been analyzed before. Las21p is putative membrane protein belonging to the major facilitator super family containing plural membrane spanning domains. Complete elimination of the LAS21 ORF did not kill the cells but made their growth temperature sensitive. Interestingly, the complete loss of the LAS21 gene canceled the sensitivity to tetracaine. The ability of the las21 mutants to grow at a higher temperature was recovered in the various media containing an osmotic stabilizer or salts. Furthermore, temperature sensitivity of the las21 mutants was partially suppressed by introduction of PKC1, encoding protein kinase C, on a high copy vector. We found some genetic interactions between LAS21 and Ras/cAMP cascade genes. These results suggest that LAS21 defines unknown pathway regulating the stress response of yeast.  相似文献

Amoebic gill disease (AGD) in cultured salmonids causes severe multifocal hyperplastic lesions in the gills with the potential to influence respiratory and acid–base physiology. Atlantic salmon Salmo salar affected with AGD were surgically implanted with dorsal aortic catheters and, following recovery, were confined for 5 min ( n  = 16) or left undisturbed ( n  = 8). Confinement caused an acute extracellular acidosis that was corrected in 6 h amongst surviving fish. There was a gradual increase in plasma lactate concentrations that peaked at 1 h post-confinement then declined by 9 h recovery. In a second experiment, AGD-affected fish were confined then recovered either in a tank of static water ( n  = 9) or while being forced to swim at 1·5 body lengths s−1 ( n  = 6). There was no significant difference between fish recovered by swimming and those in static water in terms of recovery of the acute extracellular acidosis and lactate accumulations coincident with exhaustive exercise. Confinement severely compromised the survival of AGD-affected Atlantic salmon, although survivors appeared to recover similarly to other studies. Forced swimming of AGD-affected Atlantic salmon following confinement did not facilitate recovery and is unlikely to be a useful strategy for mitigating the effects of stressful episodes such as crowding and fish movement and commercial handling.  相似文献

Twelve groups of 10 Atlantic salmon Salmo salar parr were screened for dominance in small tanks with a single localized food source using a serial removal method and the top, middle and bottom two fish in each group were classified as high, medium and low social status, respectively. These 72 ranked fish were weighed and formed into four groups or waves, each consisting of six fish of each status category. The fish were given dye marks according to status and wave and were then introduced into an artificial stream in four waves over a 12-day period. Patterns of settlement/emigration were recorded over a 2-month period, at the end of which the fish were removed from the stream, identified and reweighed, and daily specific growth rates calculated. Fifty-one fish established feeding stations in the flume, the remainder emigrating via a downstream trap. The average growth rate over the 2-month period was 0·87% day−1. Probability of settlement and growth rates following settlement were significantly higher in fish from the earlier waves. In contrast, salmon that were dominant in the initial trial were less likely than subordinate fish to settle in the experimental stream and showed lower postsettlement growth rates. The possibility that fish have individually consistent behavioural profiles that promote success in different competitive environments is discussed with reference to published literature for other groups. A small-scale trial at the end of the main experiment confirmed the prior residence effect and demonstrated that 1 day was sufficient for such an effect to be evident.  相似文献

This study was performed to determine the efficacy of orally administered oxolinic acid and Vetoquinol, an oxolinic acid ester, in the treatment of experimental induced furunculosis in Atlantic salmon Salmo salar held in seawater. Two strains of the causative bacterium Aeromonas salmonicida subsp. salmonicida, 1 sensitive (VI-88/09/03175) and 1 resistant (3475/90) to oxolinic acid, were used. In 2 trials, cohabitational challenges were performed by introducing 8 fish challenged in advance by an intraperitoneal injection of 2.2 x 10(4) colony forming units of strain 3475/90 (Trial 1) or strain VI-88/09/03175 (Trial 2) to 10 aquaria each containing 40 healthy fish. The treatment groups in both trials consisted of 4 groups receiving either oxolinic acid (2 groups) or Vetoquinol (2 groups) and 1 control group. An unchallenged, unmedicated group was used to determine the natural mortality in the population. The recommended therapeutic dose of 25 mg oxolinic acid kg-1 fish at Days 1, 2, 4, 6, 8 and 10 following initiation of treatment was used. Oral medication initiated at Day 10 (Trial 1) or Day 11 (Trial 2) following challenge significantly (p < 0.05) lowered the specific mortality in all drug-treated groups compared to the untreated control groups. Mortality in Vetoquinol-treated groups was significantly (p < 0.05) lower than in oxolinic acid-treated groups in Trial 1 whereas no significant (p < 0.05) difference in survival rate was found between the medicated groups in Trial 2.  相似文献

Endurance swimming of diploid and triploid Atlantic salmon   总被引:1,自引:0,他引:1  
When groups of diploid (mean ±  s . e . fork length, L F) 33·0 ± 1·4 cm and triploid (35·3 ± 0·5 cm) Atlantic salmon Salmo salar were forced to swim at controlled speeds in a carefully monitored 10 m diameter 'annular' tank no significant difference was found between the maximum sustained swimming speeds ( U ms, maintainable for 200 min) where the fish swam at the limit of their aerobic capability. Diploids achieved 2·99 body lengths per second (bl s−1)(0·96 m s−1) and triploids sustained 2·91 bl s−1(1·02 m s−1). The selection of fish for the trials was based on their ability to swim with a moving pattern projected from a gantry rotating at the radius of the tank and the selection procedure did not prove to be significant by ploidy. A significant difference was found between the anaerobic capabilities of the fish measured as endurance times at their prolonged swimming speeds. During the course of the experimentation the voluntary swimming speed selected by the fish increased and the schooling behaviour improved. The effect of the curvature of the tank on the fish speeds was calculated (removing the curved effect of the tank increased the speed in either ploidy by 5·5%). Implications of the endurance times and speeds are discussed with reference to the aquaculture of triploid Atlantic salmon.  相似文献
