The phase behaviour of cuticular waxes from leaves of Hedera helix L. and Juglans regia L. was studied by Fourier transform infrared spectroscopy. For this purpose reconstituted waxes, isolated cuticular membranes, dewaxed polymer matrix membranes and whole leaves were studied in the horizontal attenuated total reflection and transmission modes. Melting curves of cuticular waxes were derived from temperature-dependent changes in the absorption maximum of the symmetric stretching mode of CH2 groups (νs, at approx. 2856–2848 cm−1). With increasing temperature absorption band doublets due to CH2 scissoring (δsciss) and rocking (δrock) movements (at approx. 1473–1471 and 730–720 cm−1, respectively) indicative of an orthorhombic arrangement of alkyl chains merged into a single peak. The area ratio of the peaks at approx. 720 and 730 cm−1 was used as a measure for aliphatic crystallinity of plant cuticular waxes at a given temperature. The investigations of reconstituted cuticular waxes and those still embedded in isolated cuticles or in situ on the leaf produced comparable results. Diffusion coefficients for thirteen lipophilic organic compounds in reconstituted waxes of Fagus sylvatica L. and Picea abies (L.) Karst. were measured to characterise the transport properties of the cuticular waxes that form the transport-limiting barrier of plant cuticles. Desorption kinetics (relative amounts desorbed versus time) were asymptotic, but could be linearized up to 50% desorption by plotting relative amounts desorbed versus the square root of time. Diffusion coefficients calculated from the slopes of the linear regressions ranged from 10–22 to 10–17 m2·s–1 and decreased with increasing molecular size. This size dependency of diffusion coefficients was analysed, assuming an exponential dependence of the diffusion coefficients on molar volumes, which allowed cuticular transport properties to be related to the physical structure of the wax. Barley (Hordeum vulgare), corn (Zea mays), bean (Phaseolus vulgaris), and radish (Raphanus sativus) seedlings were continuously irradiated under a lighting device for 5–10 d at an increased ultraviolet (UV)-B fluence rate. In their growth parameters, composition, and leaf surface, these four species responded differently to the increased UV-B exposure. Bean seedlings suffered the most serious effects, radish and barley less, and corn was hardly influenced at all. In all plant species, the fresh weight, the leaf area, the amounts of chlorophylls, carotenoids and the galactolipids of the chloroplasts were reduced. The lipid content of the corn and bean seedlings also diminished. But all the irradiated plants showed a rise in their protein content compared to the control plants. The content of flavonoids increased in barley and radish seedlings by about 50%. The effects on growth parameters and composition were more extensive with increasing UV-B fluence rates, at least as shown in the case of barley seedlings. The fresh weights fell proportionally with the chlorophylls and carotenoids. In contrast, the flavonoid content of barley leaves rose parallel to the increasing UV-B fluence rates and reached 180% of the value in the control plants with the highest UV-B fluence rate. Scorching appeared regularly in the form of bronze leaf discoloration at the highest UV-B fluence rates. The response of Antarctic, tropical and temperate microalgae of similar taxonomic grouping to ultraviolet radiation (UVR) stress was compared based on their growth and fatty acid profiles. Microalgae of similar taxa from the Antarctic (Chlamydomonas UMACC 229, Chlorella UMACC 237 and Navicula UMACC 231), tropical (Chlamydomonas augustae UMACC 246, Chlorella vulgaris UMACC 001 and Amphiprora UMACC 259) and temperate (Chlamydomonas augustae UMACC 247, Chlorella vulgaris UMACC 248 and Navicula incerta UMACC 249) regions were exposed to different UVR conditions. The cultures were exposed to the following conditions: PAR (42 μmol photons m−2 s−1), PAR + UVA (854 μW cm−2) and PAR + UVA + UVB (117 μW cm−2). The cultures were subjected to UVA doses of 46.1, 92.2 and 184.4 J cm−2 and UVB doses of 6.3, 12.6 and 25.2 J cm−2 by varying the duration of their exposure (1.5, 3 and 6 h) to UVR during the light period (12:12 h light-dark cycle). UVA did not affect the growth of the microalgae, even at the highest dose. In contrast, growth was adversely affected by UVB, especially at the highest dose. The dose that caused 50% inhibition (ID50) in growth was used to assess the sensitivity of the microalgae to UVB. Sensitivity of the microalgae to UVB was species-dependent and also dependent on their biogeographic origin. Of the nine microalgae, the Antarctic Chlorella was most tolerant to UVB stress (ID50 = 21.0 J cm−2). Except for this Chlorella, the percentage of polyunsaturated fatty acids of the microalgae decreased in response to high doses of UVB. Sesame (Sesamum indicum L.) is one of the most important oilseed crops, having seeds and oil that are highly valued as a traditional health food. The objective of this study was to evaluate leaf cuticular wax constituents across a diverse selection of sesame cultivars, and the responses of these waxes to drought-induced wilting. Water-deficit was imposed on 18 sesame cultivars by withholding irrigation for 15d during the post-flowering stage, and the effect on seed yield and leaf waxes compared with a well-watered control. Leaf cuticular waxes were dominated by alkanes (59% of total wax), with aldehydes being the next-most abundant class. Compared to well-irrigated plants, drought treatment caused an increase in wax amount on most cultivars, with only three cultivars having a notable reduction. When expressed as an average across all cultivars, drought treatment caused a 30% increase in total wax amount, with a 34% increase in total alkanes, a 13% increase in aldehydes, and a 28% increase in the total of unknowns. In all cultivars, the major alkane constituents were the C27, C29, C31, C33, and C35 homologs, whereas the major aldehydes were the C30, C32, and C34 homologs, and drought exposure had only minor effects on the chain length distribution within these and other wax classes. Drought treatments caused a large decrease in seed yield per plant, but did not affect the mean weight of individual seeds, showing that sesame responds to post-flowering drought by reducing seed numbers, but not seed size. Seed yield was inversely correlated with the total wax amount (-0.466*), indicating that drought induction of leaf wax deposition does not contribute directly to seed set. Further studies are needed to elucidate the ecological role for induction of the alkane metabolic pathway by drought in regulating sesame plant survival and seed development in water-limiting environments.  相似文献

Exclusion of UV (280–380 nm) radiation from the solar spectrum can be an important tool to assess the impact of ambient UV radiation on plant growth and performance of crop plants. The effect of exclusion of UV-B and UV-A from solar radiation on the growth and photosynthetic components in soybean (Glycine max) leaves were investigated. Exclusion of solar UV-B and UV-B/A radiation, enhanced the fresh weight, dry weight, leaf area as well as induced a dramatic increase in plant height, which reflected a net increase in biomass. Dry weight increase per unit leaf area was quite significant upon both UV-B and UV-B/A exclusion from the solar spectrum. However, no changes in chlorophyll a and b contents were observed by exclusion of solar UV radiation but the content of carotenoids was significantly (34–46%) lowered. Analysis of chlorophyll (Chl) fluorescence transient parameters of leaf segments suggested no change in the F v/F m value due to UV-B or UV-B/A exclusion. Only a small reduction in photo-oxidized signal I (P700+)/unit Chl was noted. Interestingly the total soluble protein content per unit leaf area increased by 18% in UV-B/A and 40% in UV-B excluded samples, suggesting a unique upregulation of biosynthesis and accumulation of biomass. Solar UV radiation thus seems to primarily affect the photomorphogenic regulatory system that leads to an enhanced growth of leaves and an enhanced rate of net photosynthesis in soybean, a crop plant of economic importance. Pea plants (Pisum sativum L. cv. Greenfeast) were grown and exposed to supplementary UV-B radiation from day 17 after planting under growth cabinet conditions. The effects of this exposure on the ultrastructure of chloroplasts and the total soluble sugar and starch concentrations were estimated. Supplementary UV-B radiation was shown to damage the structure of chloroplasts, as manifested by dilation of thylakoid membranes, a progressive disruption of the thylakoid structure and disintegration of the double membrane envelope surrounding the chloroplast, accompanied by the accumulation of large starch grains. Diurnal changes observed in starch concentration suggest that the higher concentration of starch in supplementary UV-B-treated leaves is due to its immobilization, rather than to any increase in starch synthesis: soluble sugars accumulated and remained at a higher level and then later declined.  相似文献

Water permeability and composition of soluble cuticular lipids of isolated cuticular membranes from leaves of Citrus aurantium L. were investigated for 3 successive years. The average water permeability coefficient determined using 169 cuticular membranes was 1.09·10–7 cm s–1 with a standard deviation of 0.78·10–7 cm s–1. There were no significant differences in water permeability between years. Cuticular membranes are characterized by a great variability in water permeability both within and between years. Both water permeability of individual membranes and variability between membranes are shown to be determined by soluble cuticular lipids contained within the cuticular membranes. The soluble cuticular lipids of Citrus leaves are composed of fatty acids, primary alcohols, esters, and hydrocarbons. They occur in amounts of 9.84 g cm–2, which represents approx. 3% of the total mass of isolated cuticular membranes. The specific weight of cuticular membranes (365.4 g cm–1) and total amount of soluble cuticular lipids did not vary significantly between years. Significant differences were observed for the amounts and composition of the constituent classes of lipids. Six homologues comprise 86% of the fatty acids (C16; C18; C19; C21; C24; C26), 83% of the primary alcohols (C24; C26; C28; C30; C32; C34) and 88% of the esters (C36; C38; C40; C41; C42; C44). Eleven major homologues amount only to 62% of the total hydrocarbons (C16; C17; C18; C20; C26; C27; C29; C30; C31; C32; C33). Variability in the composition of soluble cuticular lipids between years was much smaller than variability of water permeability and, therefore, no relation between composition of soluble cuticular lipids and water permeability could be found. The composition of free fatty acids (FFA) in relation to Hill reaction activity and photoperoxidation of lipids was studied in chloroplasts isolated from fresh, cold and dark-stored as well as illuminated leaves of Lycopersicon esculentum Mill., Phaseolus vulgaris L. and Cucumis sativus L. Following the cold and dark-storage of leaves the loss of Hill reaction activity is accompanied by approximately a 5-fold increase in the amount of FFA and by an increase in the percentage of unsaturated FFA, particularly that of linolenic acid. Illumination of the cold- and dark-stored leaves restores both Hill reaction activity and the content and composition of chloroplast FFA. Following the second and third cycles of cold storage and illumination of leaves the percentage of unsaturated fatty acids in chloroplasts increases while that of saturated ones decreases despite of the significant restoration of Hill reaction activity. Since the illumination of cold-stored leaves results in peroxidation of inhibitory fatty acids it seems likely that this phenomenon could, at least partially, be responsible for the restoration of Hill reaction activity. Inhibition of Hill reaction activity by exogenous linolenic acid in chloroplasts of fresh, cold-stored as well as cold-stored and illuminated leaves could be reversed following the incubation of chloroplast suspension with BSA, however only to a value measured in the absence of unsaturated fatty acid. The possible ameliorative effects of quercetin on soybean [Glycine max (L.) Merr.] leaves exposed to UV-B radiation were conducted in greenhouse. The symmetrical leaves supplied with quercetin solution (0.2%, 1%) were exposed to UV-B radiation (0, 3.5, 6.5 kJ m−2 d−1). 0.2% quercetin ameliorated leaf photosynthesis, improved leaf water content (LWC), and decreased lipid oxidation. The unfavorable effect on photosynthetic parameter was displayed in 1% quercetin treatment. The effect of quercetin on phenylalanine ammonia lyase (PAL) activity varied with the quercetin concentration, UV-B radiation intensity and leaf development. In the later development polyphenol oxidase (PPO) activity was increased significantly by quercetin treatments. We suggested that quercetin with suitable concentration could serve as UV-B protective agent partly due to its antioxidant capacity.  相似文献

Irradiation ofArabidopsis with ultraviolet (UV) light resulted in intensity- and wavelength-dependent increases in the levels of a small family of UV-absorbing flavonoids, which accumulate in the aerial parts of the plants. A gradient of sensitivity to UV-B radiation is described in the different leaves of developingArabidopsis plants whereby the earliest formed leaves become damaged by UV-B faster and more extensively than later formed leaves. This UV-sensitivity gradient tightly parallels differences in constitutive as well as UV-induced levels of flavonoid accumulation among the various leaves, suggesting a direct role of flavonoids in protection against damage by UV radiation. The level of accumulated flavonoids, both constitutive and UV-induced, in each leaf appear to be dependent on the specific developmental state of each leaf as well as the overall developmental state of the plant. The UV-mediated flavonoid response, along with the observed UV-induced damage, appear not to be systemic inArabidopsis but restricted very closely to the irradiated areas of leaves.I am deeply indebted to Robert Fischer and Bob Buchanan for providing access to their laboratories and for their invaluable help without which this work could not have been realized. The giant sporangiophores of Phycomyces blakesleeanus turn towards blue and away from ultraviolet C sources (wavelength under 310 nm). We have isolated fifteen mutants with normal blue tropism but defective ultraviolet tropism. Wild-type sporangiophores described a double turn when exposed successively to blue and ultraviolet beams coming from the same side; under certain conditions, the mutants turned only to the blue. The new uvi mutations modified the behaviour in heterokaryosis and were lethal in homokaryosis, i.e., they affected essential cellular components. The responses of the wild type and one of the mutants were registered and evaluated with a computer-aided device. The mutant behaved normally under blue light, but took longer than the wild type to turn away from the ultraviolet source. With very weak ultraviolet stimuli (10(-8) and l0(-9) W m-2), the wild type turned towards the source, but the mutant did not respond. Calculations of absorbed-energy distributions in the sporangiophore showed that Phycomyces responds differently to similar spatial distributions of blue and ultraviolet radiations. Wild-type and mutant sporangiophores had the same high ultraviolet absorption due to gallic acid. We conclude that ultraviolet tropism is not just a modification of blue phototropism due to the high ultraviolet absorption of the sporangiophores. Phycomyces has a separate sensory system responsive to ultraviolet radiation, but not to blue light.  相似文献

A mutantArabidopsis thaliana L., which displays a dramatic increase in sensitivity to ultraviolet-B (UV-B) radiation compared with wild-type plants, has been isolated by chemical mutagenesis. This mutation appears to affect UV-tolerance specifically, since mutant plants are indistinguishable from wild type with respect to their degree of resistance to other forms of stress. The UV-sensitive mutation proved to be recessive and to segregate as a single Mendelian locus. This single gene defect was shown to lead to a block in the synthesis of a group of flavonoids which normally accumulate in developing wild-typeArabidopsis and which increase in concentration when plants are exposed to UV radiation. One of these compounds has been identified as a rhamnosylated derivative of the flavonol, kaempferol. The results suggest that one or more of the flavonoids whose production has been blocked in the UV-sensitive mutant is essential for the protection ofArabidopsis against UV-radiation damage. Cuticular wax composition of healthy and and declining mature Norway spruce trees [Picea abies (L.) Karst.] was investigated in five European forest areas. The amount of extracted wax and the content of alkanes and secondary alcohols were analysed as a function of the factors sample area (five areas, detailed below), needle age (current year to 2 years) and decline class (Class O to Class 2). Using a GC-MS, alkanes from C20 to C31 and the following alcohols were quantified: 10-nonacosanol, 5,10-nonacosandiol, 4,10-nonacosandiol and the triterpenol 24-methylenecycloartanol. According to our results, the total wax content as well as the alkane and alcohol content of waxes show a large variation corresponding to sample area and needle age. Ageing caused a highly significant increase in alkane content and a highly significant decrease in total wax and alcohol content. The decline class significantly influenced only the content of the long chain alkane C31 (increase), the secondary alcohol 10-nonacosanol (decrease), and the triterpenic alcohol (decrease). Total wax weight was not influenced by tree damage. Thus, according to our results, needle ageing and progressive tree damage are correlated to different changes in the examined parameters.  相似文献

The impact of different doses of artificial ultraviolet (UV) radiation on the growth stages of a marine zooplankton was investigated using laboratory microcosms. Mortality percentages of naupliar and adult samples of Artemia franciscana were recorded in relation to different UV doses (single exposure: 75, 150, 300, 600, 1,200, 2,400, 3,900, 7,800 J m–2) at specific observation times after exposure (24, 48, 72, 96 and 120 h). The relationship between mortality percentage and UV dose showed significant differences in relation to the zooplankton growth stage. The elevated susceptibility of the naupliar samples to UV radiation is described through a mortality model based on a logistic equation. The data analysis shows that the slope of mortality versus dose remains the same for the two growth stages while the lethal dose in the naupliar stage was 3.3 smaller than that determined for the adult stage. The slope of the UV mortality rate versus post-incubation time was found to be significantly different (P<0.05) at low UV doses for the two life stages examined, i.e. naupliar and adult. The lower value of LD50 in naupliar stages compared to that for adults confirms that in the early growth stage this marine zooplankton is more susceptible to UV radiation.  相似文献

选用2个抗旱性不同的紫花苜蓿品种,敖汉(强抗旱)和三得利(弱抗旱),设置空气湿度(45%-55%和75%-85%)和土壤水分胁迫(75%和35%田间持水量)处理,分析紫花苜蓿叶表皮蜡质含量、组分及晶体结构、气体交换参数、水势及脯氨酸含量的变化规律。结果表明,单独土壤水分胁迫时,紫花苜蓿叶表皮蜡质晶体结构及蜡质总量无显著变化;敖汉蜡质组分中烷类、酯类含量增加,醇类含量下降;三得利醇类含量下降,烷类、酯类含量变化不显著。低空气湿度胁迫时,两品种蜡质总量无显著变化,烷类和酯类含量显著增加,醇类含量显著下降,叶表皮片状蜡质晶体结构熔融呈弥漫性,扩大了对叶表面积的覆盖,其蒸腾速率显著低于正常湿度。复合胁迫处理时,叶表皮片状蜡质晶体结构继续呈弥漫性,烷类、酯类、未知蜡质组分含量均高于单独胁迫处理,醇类含量最低,而蜡质总量除三得利显著高于对照外,其余均无显著差异。紫花苜蓿叶表皮蜡质各组分含量(除醇类)及蜡质总量与光合速率呈显著负相关,与蒸腾速率无显著相关关系。蜡质总量与叶水势呈显著正相关。总体上,敖汉蜡质总量显著高于三得利,蜡质组分中烷类物质的增加有助于提高植株的抗旱性。在复合胁迫下,强抗旱品种主要通过气孔因素控制水分散失,而弱抗旱品种通过气孔和非气孔因素共同控制植物水分散失。  相似文献

Sexual dimorphism is presumed to reflect adaptive divergence in response to selection favouring different optimal character states in the two sexes. Here, we analyse patterns of sexual dimorphism in the cuticular hydrocarbons of the Australian field cricket Teleogryllus oceanicus using gas chromatography. Ten of the 25 peaks found in our chromatographs, differed in their relative abundance between the sexes. The presence of sexual dimorphism in T. oceanicus is discussed in reference to a review of sexual dimorphism in cuticular hydrocarbons of other insects. We found that this trait has been examined in 103 species across seven different orders. Seventy-six of these species (73%) displayed sex specificity of cuticular hydrocarbons, the presence/absence of which does not appear to be directly linked to phylogeny. The occurrence of sexual dimorphism in cuticular hydrocarbons of some but not other species, and the extent of variation within genera, suggest that this divergence has been driven primarily by sexual selection.  相似文献

The increasing levels of ultraviolet-B (UV-B) radiation reaching the earth's surface caused by ozone destruction have prompted many studies of UV-B effects on plants. Most of these studies have focused on physiological and growth responses of plants to increased UV-B, but these measures may not be closely related to future survival of plant populations. We examined the effects of two different levels of increased UV-B on total female fitness, including seed number and quality, in rapid-cycling strains of Brassica nigra and B. rapa (Brassicaceae). We also measured the effects of UV-B on fitness components, particularly those related to pollination success. Two separate experiments, examining two different levels of UV-B, were performed. Sixty plants of each species were grown under control and enhanced levels of UV-B for a total of 480 plants (60 plantsx2 speciesx2 UV-B levelsx2 experiments). Increased UV-B was generally detrimental to growth and flowering in both species; however, total seed production was actually greater at higher UV-B doses in three of four dose/plant species combinations examined. UV-B had little effect on pollination success or offspring quality in either species. Therefore, in spite of the detrimental effects of UV-B on growth and flowering that we found, there is little evidence that fitness of these plant species would suffer with increasing UV-B, and we caution against using solely physiological or growth measurements to infer effects of UV-B on plant population fitness.  相似文献

Experimental evidence for a membranebound microsomal ester synthetase from Bonus barley primary leaves is reported. The results are consistent with at least two mechanisms for the synthesis of barley wax esters: an acyl-CoA-fattyalcohol-transacylase-type reaction and an apparent direct esterification of alcohols with fatty acids. Biosynthesis of wax esters was not specific with regard to the chain length of the tested alcohols. The microsomal preparation readily catalyzed the esterification of C16-, C18-, C22- or C24-labelled alcohols with fatty acids of endogenous origin. Exogenous long-chain alcohols were exclusively incorporated into the alkyl moieties of the esters. Addition of ATP, CoA and-or free fatty acids was not effective in stimulating or depressing the esterifying activity of the microsomal fraction.
