The conservation status of small breeding areas of the Goosander (Mergus merganser merganser) in Central Europe is unclear. Geographic isolation of these areas suggests restricted gene flow to and from large North-European populations. On the other hand, migrating Goosanders from northern Europe join the Central European breeding population for wintering. To evaluate the conservation status of the small breeding areas we assessed the genetic structure of M. merganser populations in Europe by examining two nuclear marker systems (microsatellites and Single Nucleotide Polymorphisms, SNP) and mitochondrial (mtDNA) control region sequence variation for Goosanders in 11 sampling areas representing three of five distinct breeding areas and two subspecies (M. m. merganser and M. m. americanus). Overall population differentiation estimates including both subspecies were high, both based on mtDNA () and nuclear markers (θ ST = 0.219; 95% CI 0.088–0.398, SNP and microsatellites combined). Within Europe, mtDNA revealed a strong overall () and significant pairwise population differentiation between almost all comparisons. In contrast, both nuclear marker systems combined revealed only a small overall genetic differentiation (θ ST = 0.022; 95% CI 0.003–0.041). The strong genetic differentiation based on female-inherited mtDNA but not on biparentally inherited nuclear markers can be explained by sex-biased dispersal and strong female philopatry. Therefore, small breeding areas in Europe are endangered despite large male-mediated gene-flow, because when these populations decline, only males—but due to strong philopatry not females—can be efficiently supplemented by migration from the large North European populations. We therefore propose to manage the small breeding areas independently and to strengthen conservation efforts for this species in Central Europe.  相似文献

The scaly-sided merganser, Mergus squamatus, is considered one of the most threatened sea duck species in the Palearctic with limited breeding and wintering distribution in China and Russia. To provide information for future conservation efforts, we sequenced a portion of the mitochondrial (mt) DNA control region in four species of mergansers and three additional sea duck taxa to characterize the evolutionary history of the scaly-sided merganser, infer population trends that may have led to its limited geographic distribution, and to compare indices of genetic diversity among species of mergansers. Scaly-sided mergansers exhibit substantially lower levels of mtDNA genetic diversity (h = 0.292, π = 0.0007) than other closely related sea ducks and many other avian taxa. The four haplotypes observed differed by a single base pair suggesting that the species has not experienced a recent population decline but has instead been at a low population level for some time. A phylogenetic analysis placed the scaly-sided merganser basal to North American and European forms of the common merganser, M. merganser. Our inclusion of a small number of male samples doubled the number of mtDNA haplotypes observed, suggesting that additional genetic variation likely exists within the global population if there is immigration of males from unsampled breeding areas.  相似文献

Between 1984 and 1990 a total 221 Carlin tags used to mark salmon Salmo salar smolts in the River North Esk, NE Scotland, were recovered from the stomachs of goosanders Mergus merganser and red-breasted mergansers M. serrator . Both Carlin-tagging and adipose-clipping affected the predation of salmon smolts by sawbill ducks. The mean (± S.D.) sizes of tagged smolts taken by both species were similar (117 ± 3 mm) and significantly smaller than the mean sizes of smolts in the river, possibly due to a reduction in the swimming performance of small smolts bearing tags. Large adipose-clipped smolts (±mean smolt size) were predated significantly more than unclipped smolts, but no such difference was observed for small smolts (相似文献

To control swimmer’s itch in northern Michigan inland lakes, USA, one species of bird, the common merganser (Mergus merganser), has been relocated from several lakes since 2015. Relocation efforts are driven by a desire to reduce the prevalence of the swimmer’s itch-causing parasite Trichobilharzia stagnicolae. The intention of this state-sponsored control effort was to interrupt the life cycle of T. stagnicolae and reduce parasite egg contribution into the environment from summer resident mergansers such that infections of the intermediate snail host Stagnicola emarginata declined. Reduced snail infection prevalence was expected to substantially reduce the abundance of the swimmer’s itch-causing cercarial stage of the parasite in water. With no official programme in place to assess the success of this relocation effort, we sought to study the effectiveness and impact of the removal of a single definitive host from a location with high definitive host and parasite diversity. This was assessed through a comprehensive, lake-wide monitoring study measuring longitudinal changes in the abundance of three species of avian schistosome cercariae in four inland Michigan lakes. Environmental measurements were also taken at these lakes to understand how they can affect swimmer’s itch incidence. This study demonstrates that the diversity of avian schistosomes at the study lakes would likely make targeting a single species of swimmer’s itch-causing parasite meaningless from a swimmer’s itch control perspective. Our data also suggest that removing the common merganser is not an effective control strategy for the T. stagnicolae parasite, likely due to contributions of the parasite made by non-resident birds, possibly migrants, in the autumn and spring. It appears likely that only minimal contact time between the definitive host and the lake ecosystem is required to contribute sufficient parasite numbers to maintain a thriving population of parasite species with high host specificity.  相似文献

Genetic studies of waterfowl (Anatidae) have observed the full spectrum of mitochondrial (mt) DNA population divergence, from apparent panmixia to deep, reciprocally monophyletic lineages. Yet, these studies often found weak or no nuclear (nu) DNA structure, which was often attributed to male‐biased gene flow, a common behaviour within this family. An alternative explanation for this ‘conflict’ is that the smaller effective population size and faster sorting rate of mtDNA relative to nuDNA lead to different signals of population structure. We tested these alternatives by sequencing 12 nuDNA introns for a Holarctic pair of waterfowl subspecies, the European goosander (Mergus merganser merganser) and the North American common merganser (M. m. americanus), which exhibit strong population structure in mtDNA. We inferred effective population sizes, gene flow and divergence times from published mtDNA sequences and simulated expected differentiation for nuDNA based on those histories. Between Europe and North America, nuDNA ФST was 3.4‐fold lower than mtDNA ФST, a result consistent with differences in sorting rates. However, despite geographically structured and monophyletic mtDNA lineages within continents, nuDNA ФST values were generally zero and significantly lower than predicted. This between‐ and within‐continent contrast held when comparing mtDNA and nuDNA among published studies of ducks. Thus, male‐mediated gene flow is a better explanation than slower sorting rates for limited nuDNA differentiation within continents, which is also supported by nonmolecular data. This study illustrates the value of quantitatively testing discrepancies between mtDNA and nuDNA to reject the null hypothesis that conflict simply reflects different sorting rates.  相似文献

The field metabolic rates (FMRs) of nine captive goosanders, Mergus merganser , released on two Scottish rivers were estimated using the doubly-labelled water (DLW) technique. Mean (± S.E.) FMR was 2.322±0.239ml CO2 g−1 h−1 and daily energy expenditure (DEE) 1939 ± 184 kJ per day. This was significantly greater (x 1.5) than previous estimates which assumed DEE to be three times the basal metabolic rate (BMR) based on regression equations predicting BMR from body mass. FMR of captives and dietary data from previous studies were used to estimate daily consumption of salmon, Salmo salar , smolts and parr by natural populations of these ducks on the river North Esk, north-east Scotland. Goosanders are likely to consume 480-522 g fish per day of which two-thirds are juvenile salmon; equivalent to a daily intake of 10-11 smolts and 48-52 parr. Annual predation of smolts by goosanders was estimated to be between 8000 and 15 000 or 3 and 16% of annual production.  相似文献

Summary Diel and seasonal foraging activity patterns of goosanders (Mergus merganser L.) and red-breasted mergansers (M. serrator L.) were studied during the breeding season on the Rickleå River in northern Sweden (64°05N). In addition, the locomotor activity patterns of their most important prey species, the river lamprey (Lampetra fluviatilis (L.)) and the three-spined stickleback (Gasterosteus aculeatus L.), were recorded.During spring, as mergansers arrived in the breeding area, both species exhibited a bimodal, diurnal pattern of activity with morning and evening peaks. By June (the spawning period for the river lamprey and the migratory period for the three-spined stickleback), goosander activity had shifted to late in the evening. This new activity peak correlated well with the main activity period in the river lamprey. Red-breasted mergansers remained predominantly diurnal throughout the observation period. Their diurnal activity correlated with the activity of their major prey, the three-spined stickleback.Both merganser species utilize a specialized foraging technique, i.e. probing the river-bottom with the bill, to catch hiding fish more or less at random. This behaviour probably helps them to forage during the relatively bright summer nights and thus prey upon the abundant nocturnal river lamprey. The goosander preys upon the river lamprey to a greater extent than does the red-breasted merganser, thus leading to a temporal segregation in food resource utilization between the two duck species.  相似文献

We investigated the effect of temperature on development and demographic parameters such as the intrinsic rate of natural increase (r m) of the two spider mite species Tetranychus merganser Boudreaux and T. kanzawai Kishida at eleven constant temperatures ranging from 15 to 40°C at intervals of 2.5°C. Both male and female T. merganser and T. kanzawai completed development from egg to adult at temperatures ranging from 15 to 37.5°C. The longest developmental duration of immature stages was found at 15°C and the shortest developmental duration was found at 35°C for both species. Using linear and non-linear developmental rate models, the lower thermal thresholds for egg-to-adult (female and male) and egg-to-egg development were estimated as 12.2–12.3°C for T. merganser and as 10.8°C for T. kanzawai. The highest developmental rates were observed at around 35°C, whereas the upper developmental thresholds were around 40°C for both species. In fact, at 40°C, a few eggs of either species hatched, but no larvae reached the next stage. The r m-values of T. merganser ranged from 0.072 (15°C) to 0.411 day−1 (35°C), whereas those of T. kanzawai ranged from 0.104 (15°C) to 0.399 (30°C). The r m-values were higher for T. kanzawai than for T. merganser at temperatures from 15 to 30°C, but not at 35°C (0.348 day−1). Total fecundity of T. merganser was also higher than that of T. kanzawai at 35°C. These results indicate that higher temperatures favor T. merganser more than T. kanzawai.  相似文献

Several subspecies are defined within Codium fragile, including the invasive C. fragile ssp. fragile, first reported in New Zealand in 1973. An endemic subspecies, C. fragile ssp. novae‐zelandiae, is also found throughout New Zealand. The two subspecies exhibit morphological and molecular variation, although these have never been evaluated together. We compared variation between subspecies at locations in Auckland, identifying subspecies using rps3‐rpl16 DNA sequence data, and assessing gross morphological differences, anatomical utricle characters and morphometrics. The taxonomic utility of the morphometric data sets was assessed by linear discriminant analysis. Utricle characters and measurements varied within individual thalli and between different preservation methods. The phenotypes of both subspecies were highly variable and influenced by environment. Accurate subspecies delimitation using morphological data was not possible; the discriminant analyses performed no better than chance for all combinations of the morphological data. Specimens from New Zealand, Canada, Australia and Ireland were sequenced using both the rps3‐rpl16 and tufA plastid markers. The tufA elongation factor was shown to be a good candidate for differentiating subspecies of C. fragile. This marker is twice the length of the rps3‐rpl16 spacer, shows greater variation between ssp. fragile and novae‐zelandiae, and is less prone to sequencing error. A simple restriction enzyme digest of the tufA amplicon can distinguish ssp. fragile and ssp. novae‐zelandiae. Our study expands the known range of the ssp. fragile in New Zealand, including the first record of this subspecies from the west coast of Auckland, and points to a need to re‐evaluate morphological and molecular criteria for subspecies currently defined within C. fragile.  相似文献

Cosmodela Rivalier, 1961, represents a genus of tiger beetles that currently contains 13 described species widely distributed across Southeast Asia. A phylogenetic analysis based on the mitochondrial DNA fragments 16S and COI and the nuclear marker wingless and a phylogeographic analysis using a COI fragment were carried out on two subspecies of Cosmodela aurulenta (C. a. juxtata and C. a. aurulenta). The results support the hypothesis that these two subspecies are significantly different to be considered as separate species that diverged during the Pleistocene. TAXONDNA analysis was used to investigate the capability of the COI region as a marker for discriminating both entities and to quantify intra‐ and intertaxa genetic variation. The minimum distance between C. aurulenta and C. juxtata was 2.7837%, and no overlap of intra‐ and intertaxa genetic divergence was observed. Both taxa, here considered as valid species, occur in sympatry in the Malay Peninsula, with C. aurulenta most probably originating from the area and C. juxtata a secondary colonizer that expanded southwards from the Asian mainland. Our data infer a continental origin of the Indonesian samples of C. aurulenta, and they most likely dispersed across the land bridges that emerged during glacial maxima to form Sundaland.  相似文献

The Northern Goshawk Accipiter gentilis is a medium‐sized bird of prey inhabiting boreal and temperate forests. It has a Holarctic distribution with 10 recognized subspecies. Traditionally, it has been placed within the Accipiter [gentilis] superspecies, together with Henst's Goshawk A. henstii, the Black Sparrowhawk A. melanoleucus, and Meyer's Goshawk A. meyerianus. While those four taxa are geographically separated from each other, hence referred to as allospecies, their phylogenetic relationships are still unresolved. In the present study, we performed phylogenetic analyses on the Accipiter [gentilis] superspecies, including all recognized subspecies of all four allospecies, using partial sequences of two marker loci of the mitochondrial genome, the control region and the cytochrome b gene. We found a deep split within A. gentilis into two monophyletic groups, a Nearctic clade (three subspecies) and a Palearctic clade (seven subspecies). The Palearctic clade is closely related to A. meyerianus, and together these two were more closely related to the other Old World taxa A. henstii and A. melanoleucus, which in turn were reciprocally monophyletic sister species. As a consequence, A. gentilis as usually conceived (including all Holarctic subspecies) was non‐monophyletic. We found a strong genetic homogeneity within Palearctic A. gentilis despite the fact that it comprises seven subspecies distributed from the Atlantic coast in Western Europe to Eastern Siberia. Relationships between the four clades could not be resolved unambiguously. Our results, if confirmed by more integrative data, would imply a taxonomic revision of Nearctic A. gentilis into a separate allospecies, Accipiter [gentilis] atricapillus.  相似文献

Fusobacterium nucleatum, which has four subspecies (nucleatum, animalis, vincentii and polymorphum), plays an important role in promoting colorectal cancer (CRC). However, as there is no efficient method of differentiating these subspecies in the context of a rich gut microbiota, the compositions in CRC remain largely unknown. In this study, a PCR-based differentiation method enabling profiling of Fnucleatum infection in CRC at the subspecies level was developed. Based on the analysis of 53 Fnucleatum genomes, we identified genetic markers specific to each subspecies and designed primers for the conserved sequences of those markers. The PCR performance of the primers was tested with Fnucleatum and non-nucleatum Fusobacterium strains, and complete consistence with taxonomy was achieved. Additionally, no non-specific amplification occurred when using human DNA. The method was then applied to faecal (n = 58) and fresh-frozen tumour tissue (n = 100) samples from CRC patients, and wide heterogeneity in Fnucleatum subspecies compositions in the gut microbiota among CRC patients was observed. Single-subspecies colonization was common, whereas coexistence of four subspecies was rare. Subspecies animalis was most prevalent, while nucleatum was not frequently detected. The results of this study contribute to our understanding of the pathogenicity of Fnucleatum at the subspecies level and the method developed has potential for clinical and epidemiological use.  相似文献

The common midwife toad (Alytes obstetricans), widely distributed in the northern half of the Iberian Peninsula and part of Western Europe, is currently subdivided into four subspecies: A. o. obstetricans, A. o. boscai, A. o. pertinax and A. o. almogavarii. However, the delimitation of these subspecies and their ranges are still under discussion because strong discordances have been found between morphological and molecular data, and especially among different genetic markers. Here, we screen a set of novel microsatellite loci and mtDNA sequences of A. obstetricans populations representative of all currently recognized subspecies to investigate the correspondence between genetic groupings inferred from clustering analysis of microsatellite genotypes and the described subspecies and test whether patterns of mtDNA variation are concordant with those genetic clusters. Our results confirm previous expectations of extremely high intraspecific diversity in A. obstetricans in Iberia. Analyses of microsatellite and mtDNA data were concordant in recovering five well‐defined groups, of which three correspond to previously defined subspecies, while the two additional clusters correspond to populations of subspecies A. o. boscai separated by the Douro River. Our results suggest the occurrence of two distinct genetic units within A. o. boscai that likely result from a long independent evolutionary history, thus deserving special attention from a conservation point of view.  相似文献

Summary Seasonal and diel activity patterns of mergansers, gulls, and terns along a river in northern Sweden were documented, as were those of their fish prey. The seasonal and diel activity patterns of goosandersMergus merganser and gulls (Larus canus, L. argentatus, andL. fuscus) were closely related to that of the river lampreyLampetra fluviatilis. During the peak spawning of the river lamprey, birds showed a nocturnal peak in fishing activity. During the summer solstice, birds were active for 24 h. The activity patterns of red-breasted merganserMergus serrator, ternsSterna spp., and three-spined sticklebacksGasterosteus aculeatus were also similar. Activity pattern of the prey apparently influenced breeding time, diel activity and foraging area of the twoMergus species. Social relations between gulls probably corrdinated their peak in fishing, which coincided with the time lampreys were most efficiently exploited.  相似文献

The majority of 170 rice accessions used in this study were diverse landraces or varieties from a putative mini-core collection of Chinese germplasm along with some widely used parental lines in genetic analysis or breeding (a few from abroad). The population was genotyped using 84 SSR or InDel markers on chromosome 7 and 48 markers on other chromosomes. The phenotyping of heading date, plant height and panicle length were carried out in different locations for 2 years. Based on morphological characterization, distance-based clustering and model-based estimation of marker data, the population showed a predominant structure with two subpopulations in correspondence with indica and japonica subspecies. The estimation of linkage disequilibrium in 2 Mb windows varied along chromosome 7 and showed parallel changes with inter-subspecies differentiation of marker loci (Fst). Based on the mixed linear model considering population structure and family relatedness [i.e. the (Q + K) model], one to three associated markers (P ≤ 0.0001) per trait per experiment were scanned out on rice chromosome 7. Most significant loci were repeated for the data from both field experiments while two loci were associated with two or three traits. Marker-based allelic effects were shown in a couple of associated markers as examples. The application of association results in breeding program was also discussed. Defense behavior of three, free living giant (Megapis) honey bee subspecies, Apis laboriosa, A. dorsata dorsata and A. dorsata breviligula, was compared. Disturbed worker bees responded with characteristic dorso-ventral defense body twisting (DBT). Workers of A. laboriosa twisted the thorax by 55°, and the two other A. dorsata subspecies by about 10° more. A. laboriosa workers raised the tip of the abdomen by 90° and workers of the two other bee subspecies by about 20° higher. Differences in those traits were highly significant between A. laboriosa and both A. dorsata subspecies, but were not significant between those two subspecies. The whole cycle of DBT was the most vigorous in A. d. breviligula (0.11 s), and it was twice as vigorous as in A. d. dorsata (0.26 s) and trice as in A. laboriosa (0.32 s). A. laboriosa twisted the body together with wings folded over the abdomen, while the two A. dorsata subspecies raised the abdomen between spread wings. This supports the opinion to treat A. laboriosa as a separate species. Anthropogenic activity, especially modern apiculture, has considerable impact on the natural distribution of the Western honeybee, Apis mellifera, leading to the spread, replacement and fragmentation of many subspecies. This creates demand for the conservation of some subspecies, in particular, Apis mellifera mellifera, which once was widely distributed in Western Europe and nowadays is endangered through habitat loss and fragmentation. Moreover, A. m. mellifera may be further endangered by hybridisation in populations that now occur in artificial sympatry with other subspecies. Here, we quantify and compare individual hybridisation between sympatric and allopatric honeybee populations of A. m. mellifera and A. m. carnica using microsatellite markers and a Bayesian model-based approach. We had a special focus on pure breeding populations, which are a major tool in honeybee conservation. Our results demonstrate that subspecies are still highly differentiated, but gene flow is not prevented by the current management strategies, creating urgent demand for an improved conservation management of A. m. mellifera. However, the occurrence of a high number of pure individuals might suggest that some sort of hybrid barrier acts against the complete admixture of the two subspecies.  相似文献

The Lahontan cutthroat trout (Oncorhynchus clarkii henshawi) is threatened by habitat destruction, over‐harvest and hybridization with nonnative trout. Currently, three Geographic Management Units (GMUs) are recognized within the taxon. Here, we describe a suite of 68 single‐nucleotide polymorphism (SNP) genetic markers for use in the study and management of Lahontan cutthroat trout and a closely related subspecies, the Paiute cutthroat trout (O. c. seleneris). These include markers variable within the two subspecies (n = 35), diagnostic for the two subspecies (n = 23) and diagnostic for Yellowstone cutthroat trout (O. c. bouvieri) and other closely related subspecies (n = 10). Sixty‐three markers were discovered by Sanger sequencing of 171 EST loci in an ascertainment panel including Lahontan cutthroat trout from four populations representing all GMUs. Five markers were identified in a secondary sequencing effort with a single population of Lahontan cutthroat trout. TaqMan assays were validated on six Lahontan cutthroat trout populations and a diverse panel of other trout. Over 90% of the markers variable in Lahontan cutthroat trout were polymorphic in at least two populations, and 66% were variable within all three GMUs. All Lahontan diagnostic markers were also fixed for the Lahontan allele in Paiute cutthroat trout. Most of the Yellowstone diagnostic markers can also be used for this purpose in other cutthroat trout subspecies. This is the first set of SNP markers to be developed for Lahontan cutthroat trout, and will be an important tool for conservation and management.  相似文献

Understanding the spatiotemporal distribution of genetic variation and the ways in which this distribution is connected to the ecological context of natural populations is fundamental for understanding the nature and mode of intraspecific and, ultimately, interspecific differentiation. The Petunia axillaris complex is endemic to the grasslands of southern South America and includes three subspecies: P. a. axillaris, P. aparodii and P. asubandina. These subspecies are traditionally delimited based on both geography and floral morphology, although the latter is highly variable. Here, we determined the patterns of genetic (nuclear and cpDNA), morphological and ecological (bioclimatic) variation of a large number of P. axillaris populations and found that they are mostly coincident with subspecies delimitation. The nuclear data suggest that the subspecies are likely independent evolutionary units, and their morphological differences may be associated with local adaptations to diverse climatic and/or edaphic conditions and population isolation. The demographic dynamics over time estimated by skyline plot analyses showed different patterns for each subspecies in the last 100 000 years, which is compatible with a divergence time between 35 000 and 107 000 years ago between P. a. axillaris and P. a. parodii, as estimated with the IMa program. Coalescent simulation tests using Approximate Bayesian Computation do not support previous suggestions of extensive gene flow between P. a. axillaris and P. a. parodii in their contact zone.  相似文献
