We identified 14 novel polymorphic microsatellite loci in the black‐capped vireo (Vireo atricapillus). We also attempted to amplify and genotype these loci in other Vireo species, including the white‐eyed vireo (Vireo griseus), red‐eyed vireo (Vireo olivaceus), and blue‐headed vireo (Vireo solitarius). In 33 genotyped black‐capped vireos from two locations, total alleles ranged from six to 20, with observed heterozygosity ranging from 0.58 to 0.91 and expected heterozygosity from 0.65 to 0.93. Two loci had detectable levels of null alleles. Many of the loci were able to be amplified in the related Vireo species.  相似文献

Abstract Theory predicts that compared with active searchers, ambush foragers should have lower rates of energy intake, slower growth, and higher survival rates. We tested these predictions with data on two species of sympatric, saurophagous, small‐bodied, viviparous elapid snakes: the broad‐headed snake, Hoplocephalus bungaroides, and the small‐eyed snake, Rhinoplocephalus nigrescens. Demographic parameters and growth curves for both species were estimated from a long‐term (9 years) mark‐recapture study in Morton National Park, south‐eastern Australia. The ambush predator (H. bungaroides) displayed slower juvenile growth and later maturation (5 years for males, 6 years for females) than did the active forager (R. nigrescens, 3 years). Litter sizes were similar in both species, but reproductive frequency was higher in R. nigrescens (90–100%) than in H. bungaroides (50%). Juvenile survival was lower in the active searcher (31%) than in the ambush forager (55%), but adult survivorship was similar (74% vs 82%). Our results support the hypothesis that ambush foragers display ‘slow’ life history traits, but additional phylogenetically independent comparisons are needed to evaluate the generality of this pattern.  相似文献

The ant Cataglyphis cursor is exceptional in that unmated workers are potentially able to lay both male and female eggs. We characterized eight pairs of primers for microsatellite loci, developed from genomic DNA for this species. Variability was tested with DNA from 19 workers and all eight loci were highly polymorphic, displaying 5–10 alleles and a high level of heterozygosity. Cross‐species amplifications indicate that these microsatellites might be useful in genetic studies of other species belonging to the genus Cataglyphis.  相似文献

Five tetranucleotide microsatellite loci were isolated from the striped dolphin (Stenella coeruleoalba). Characterization of these loci revealed between three and 11 alleles at each locus and observed heterozygosity ranged from 0.192 to 0.775. All sets of primers amplified homologous loci when tested on four related species and showed polymorphisms in most locus–species combinations. A sixth microsatellite, which appeared to be monomorphic in the striped dolphin, was also shown to be polymorphic in three related species.  相似文献

A microsatellite library was developed using genomic DNA of the endangered dusky gopher frog, Rana sevosa. Polymerase chain reaction (PCR) primers and conditions are presented for R. sevosa (eight loci) and two sister taxa — other gopher frogs, Rana capito (seven loci) and crawfish frogs, Rana areolata (three loci). Polymorphism of each microsatellite locus was evaluated for each species. All loci have moderate to high genetic variation in terms of allelic richness (four to 10 alleles per locus), observed heterozygosity (0.595–0.946), and expected heterozygosity (0.531–0.856).  相似文献

This paper reports the development of microsatellite primers for a widespread birch species Betula pendula. By screening genomic libraries with (TC)10 and (TG)10 probes, 38 microsatellite sequences were obtained, of which 23 proved to be polymorphic. Thirty genotyped individuals had two to 20 alleles per loci and a heterozygosity range of 0.17–0.92. Cross‐species amplification tests of four primer pairs on diverse birch species showed positive results in all cases. Also, one of two loci tested on related genus Alnus gave positive result.  相似文献

Eight microsatellite loci were characterized from the Kaka (Nestor meridionalis), a New Zealand parrot, using a polymerase chain reaction‐based isolation technique. Locus‐specific primers were used to genotype nine Kaka populations and tested on 25 other species of parrot. The number of alleles observed within Kaka ranged from one to 16 in the 12–126 individuals screened and two loci exhibited greater than 60% heterozygosity. Furthermore, these primers are likely to be useful in population‐level studies of two other New Zealand parrots.  相似文献

We describe the isolation and characterization of 11 microsatellite loci in the sociobiologically enigmatic ant Lasius austriacus. The polymerase chain reaction primers were tested on a population in East Austria. The number of alleles ranged from four to 19 and the observed heterozygosity from 0.200 to 0.900. Cross‐species amplification tests were performed, with some loci polymorphic in all species tested, for the closely related invasive species Lasius neglectus, three further Lasius species, another formicine, Formica polyctena, and the invasive myrmicine species Tetramorium tsushimae.  相似文献

Seventeen microsatellite DNA loci from the Australian short‐finned eel (Anguilla australis Richardson) were isolated and their amplification characteristics were described. The polymerase chain reaction primers were tested on 40 eel individuals. The primers amplified loci with relatively high numbers of alleles, ranging from five to 14 with an average of nine per locus. Mean observed heterozygosity (HO) and expected heterozygosity (HE) were 0.6779 and 0.7374, respectively, indicating that these markers would be useful for population studies. No loci deviated significantly from Hardy–Weinberg equilibrium (P = 0.05) and no evidence was found for genotypic disequilibrium among loci at a 5% significance level.  相似文献

We isolated five polymorphic microsatellite loci from a library of two thousand recombinant clones of two fungus‐growing ant species, Cyphomyrmex longiscapus and Trachymyrmex cf. zeteki. Amplification and heterozygosity were tested in five species of higher attine ants using both the newly developed primers and earlier published primers that were developed for fungus‐growing ants. A total of 20 variable microsatellite loci, developed for six different species of fungus‐growing ants, are now available for studying the population genetics and colony kin‐structure of these ants.  相似文献

Six new microsatellite loci were isolated and characterized in 32 individuals from a farm population of gilthead seabream (Sparus aurata). Expected heterozygosity at all loci was high, ranging from 0.835 to 0.958 with between 10 and 27 alleles per locus. A multiplex polymerase chain reaction protocol was developed using four of the loci. Cross‐species amplification of the loci was tested in six species of the Sparidae family and four loci were successfully amplified in two or more related species.  相似文献

Ruditapes philippinarum and R. variegate are commercially important shellfish in Korea. In order to understand the processes and organization of genetic diversity and genetic resources for the sustainable management of fisheries resources we developed 13 primer pairs for microsatellite loci in R. philippinarum and tested their cross-amplification in R. variegate. Twelve primers amplified in both species. Nine loci were polymorphic in R. philippinarum and eight in R. variegate, each with nine to 26 alleles per locus. Unbiased expected heterozygosity levels varied from 0.73 to 0.94 in R. philippinarum. All polymorphic loci possessed species-specific alleles. No linkage disequilibrium was found. Results indicated that these microsatellite were highly polymorphic and will be useful for conservation genetics of both species.  相似文献

Microsatellite markers were isolated from red‐flanked bushrobin, Tarsiger cyanurus, for parentage analysis. From an enriched genomic library using the fast isolation by AFLP of sequences containing repeats (FIASCO) method, six polymorphic loci were obtained. Seven to 21 alleles were identified in an analysis of 19 red‐flanked bushrobin, with observed heterozygosity ranging from 0.842 to 1.00. All loci showed Hardy–Weinberg equilibrium and linkage equilibrium. Total exclusion probability using six loci was 0.998 for neither parent known, and 0.999 for one parent known. We also tested the utility of these loci on four species of Turdidae. Although one of the loci failed to amplify in any species, the other primers successfully amplified in at least two of the species we tested.  相似文献

The small red‐eyed damselfly, Erythromma viridulum (Charpentier), is the first recorded example of a migrant damselfly establishing colonies in the British Isles. To examine the population genetic structure of E. viridulum, a partial genomic library enriched for CA microsatellite loci was constructed. Of the 42 loci tested, 19 amplified spurious bands and 13 were monomorphic, leaving 10 polymorphic loci that resolved distinct alleles within the expected size range. The number of alleles ranged between two (LIST14‐021, LIST14‐40) and eight (LIST14‐002). Observed and expected heterozygosities varied between 0.000–0.698 and 0.045–0.688, respectively.  相似文献

We describe the isolation and characterization of seven dinucleotide microsatellite loci developed from the red‐billed gull (Larus novaehollandiae scopulinus). Locus‐specific primers were used to genotype individuals from 13 populations of this subspecies as well as individuals from closely related subspecies from Australia and New Caledonia. The primers were tested successfully on other species of gulls and shorebirds. The number of alleles observed within the red‐billed gull ranged from three to 17, and observed heterozygosity varied from 0.359 to 0.787. These microsatellites are likely to be useful for studies of mating systems and population genetics in a wide range of gull species.  相似文献

Twenty‐six microsatellite loci were isolated and characterized from the mangrove species Rhizophora mangle using (GT)n and (CT)n repeats. Eighty‐four per cent of the clones contained microsatellite sequences; the most common dinucleotides were the (GA/CT) and (CA/GT) repeats. Ten primers were selected to investigate the polymorphism among individuals of R. mangle from two natural populations of the Colombian Pacific Coast. The observed heterozygosity per locus varied from 0.20 to 0.80, the power of discrimination was 0.32–0.84 and the power of exclusion was 0.03–0.75. This set of microsatellites offers an efficient tool for population genetics studies on this species.  相似文献

Fourteen polymorphic microsatellite loci were isolated and characterized for the Neotropical cichlid Geophagus brasiliensis and tested on 30 individuals belonging to a single population. Among the 14 loci described, four showed potential presence of null alleles, inferred from the excess of homozygous genotypes, and three of these loci showed significant deviations from Hardy–Weinberg equilibrium. Fifty‐nine different alleles were detected (ranging from two to eight alleles per locus), with estimates of observed and expected heterozygosity ranging from 0·167 to 0·700 and from 0·269 to 0·825. We developed primers for the amplification of 24 polymorphic nuclear microsatellites in apricot (Prunus armeniaca L.). Thirteen loci originated from three genomic libraries enriched for TC, TG and AAG motifs. Eight loci were developed from three fruit EST (Expressed‐Sequence‐Tag) libraries and three from a leaf cDNA microsatellite‐enriched library. There were up to nine alleles per polymorphic locus in 12 different cultivars. No difference in allele numbers were shown between cDNA and genomic‐source loci. Mean expected heterozygosity was 0.65 (range: 0.15–0.87). Mendelian segregation was confirmed for all loci. These markers should be helpful for diversity studies, genome mapping and cultivar identification in apricot and related species.  相似文献

Ten polymorphic microsatellites were isolated from the red‐bellied yellow tail fusilier Caesio cuning, a reef‐associated fish which occurs widely in the Indo‐Pacific region. The species is exploited by both small‐scale and commercial fisheries. Fifty individuals from six populations were genotyped using primers that reliably amplified 10 polymorphic loci. The number of alleles per locus ranged from three to 11. Observed heterozygosity (HO) and expected heterozygosity (HE) ranged between 0.400 to 0.880 and 0.337 to 0.843, respectively. These microsatellite loci may be used to study population structure, genetic diversity and connectivity of C. cuning in the range of its distribution.  相似文献

We characterized 11 microsatellite primer pairs for the village indigobird Vidua chalybeata. The loci were highly polymorphic, with 7–13 alleles per locus. Gene diversity, estimated as expected heterozygosity, ranged from 0.52 to 0.86, and was generally matched by levels of observed heterozygosity (0.49–0.91). Many of these primer pairs amplified polymorphic loci in cross‐species amplification trials with a variety of estrildid and ploceid finches and a sparrow, Passer griseus. These primers will be valuable for genetic analyses of the brood parasitic indigobirds and whydahs (genus Vidua) as well as other Old World finches.  相似文献
