Recent reduction in the ozone shield due to manufactured chlorofluorocarbons raised considerable interest in the ecological and physiological consequences of UV‐B radiation (λ=280–315 nm) in macroalgae. However, early life stages of macroalgae have received little attention in regard to their UV‐B sensitivity and UV‐B defensive mechanisms. Germination of UV‐B irradiated spores of the intertidal green alga Ulva pertusa Kjellman was significantly lower than in unexposed controls, and the degree of reduction correlated with the UV doses. After exposure to moderate levels of UV‐B irradiation, subsequent exposure to visible light caused differential germination in an irradiance‐ and wavelength‐dependent manner. Significantly higher germination was found at higher photon irradiances and in blue light compared with white and red light. The action spectrum for photoreactivation of germination in UV‐B irradiated U. pertusa spores shows a major peak at 435 nm with a smaller but significant peak at 385 nm. When exposed to December sunlight, the germination percentage of U. pertusa spores exposed to 1 h of solar radiation reached 100% regardless of the irradiation treatment conditions. After a 2‐h exposure to sunlight, however, there was complete inhibition of germination in PAR+UV‐A+UV‐B in contrast to 100% germination in PAR or PAR+UV‐A. In addition to mat‐forming characteristics that would act as a selective UV‐B filter for settled spores under the parental canopy, light‐driven repair of germination after UV‐B exposure could explain successful continuation of U. pertusa spore germination in intertidal settings possibly affected by intense solar UV‐B radiation.  相似文献

  • We studied the resistance of Parmotrema austrosinense to UV‐B stress. We focused on the effects of a high dose UV‐B radiation on the content of chlorophylls, carotenoids and UV‐B screening compounds.
  • Photosynthetic parameters were measured by chlorophyll fluorescence (potential and effective quantum yields, photochemical and non‐photochemical quenching) and evaluated in control and UV‐B‐treated lichens. Lichens from two different locations in Cordoba, Argentina, were selected: (i) high altitude and dry plots at (Los Gigantes) and (ii) lowland high salinity plots (Salinas Grandes).
  • UV‐B treatment led to a decrease in the content of photosynthetic pigments and UV‐B screens (absorbance decrease in 220–350 nm) in the samples from Salinas Grandes, while in Los Gigantes samples, an increase in UV‐B screen content was observed. Chlorophyll fluorescence parameters showed a UV‐B‐induced decline in FV/FM, ΦPSII and qP indicating limitation of primary photosynthetic processes in photosystem II (PSII) of symbiotic alga, more pronounced in Salinas Grandes samples. Protective mechanism of PSII were activated by the UV‐B treatment to a higher extent in samples from Salinas Grandes (NPQ 0.48) than in Los Gigantes samples (NPQ 0.26).
  • We concluded that site‐related characteristics, and in particular different UV‐B radiation regimen, had a strong effect on resistance of the photosynthetic apparatus of P. austrosinense to UV‐B radiation.

The filamentous cyanobacterium Fischerella muscicola TISTR8215 was tested for the presence of ultraviolet (UV)‐absorbing mycosporine‐like amino acids (MAAs) and their induction by UV radiation. Reverse‐phase high performance liquid chromatographic coupled with photodiode‐array detection studies revealed the presence of a MAA having an absorption maximum at 332 nm and a retention time of around 16.1 min. Based on absorption maximum, the compound was designated as M‐332. This is the first report for the occurrence of a MAA and its inducibility as influenced by UV radiation in Fischerella strains studied so far. Photosynthetically active radiation (PAR) had no significant impact on MAA induction. PAR + UV‐A radiation significantly induced the synthesis of M‐332; however, PAR + UV‐A + UV‐B radiation conferred highest impact on MAA synthesis. The cultures exposed to alternate light and dark conditions showed the induction of M‐332 synthesis mostly during the light period in contrast to the decreased levels of M‐322 during the dark period suggesting a circadian induction of its synthesis. Overall results indicate that F. muscicola may protect itself from deleterious short wavelength UV radiation by synthesizing the photoprotective compounds particularly during summer time in its natural brightly‐lit habitats.  相似文献

In nature, ultraviolet‐B (UV‐B) radiation is highly heterogeneous, both spatially and temporally. Plants exposed to UV‐B radiation produce UV‐B absorbing compounds that function as a protective filter. For clonal plants under heterogeneous UV‐B radiation conditions, integration among ramets can allow irradiated ramets to benefit un‐irradiated ramets by causing them to increase their UV‐B absorbing compounds content. In this study, we evaluated integration between pairs of clonal ramets of Glechoma longituba under heterogeneous or homogeneous UV‐B conditions. We determined the levels of UV‐B absorbing compounds, nitric oxide (NO) and hydrogen peroxide (H2O2) and measured the activity of phenylalanine ammonia‐lyase (PAL) in connected ramet pairs under homogeneous or heterogeneous UV‐B conditions. Under heterogeneous UV‐B conditions, the UV‐B absorbing compounds content increased in leaves of irradiated and un‐irradiated ramets, but not in the connecting stolons. The NO content increased in irradiated and un‐irradiated leaves and stolons, but the H2O2 content did not. Application of NO synthesis inhibitors and an NO blocker to irradiated ramets blocked the increase in UV‐B absorbing compounds and PAL activity in un‐irradiated ramets. These results suggested that NO is involved in the integration process for UV‐B absorbing compounds among ramets. Our findings suggested that a UV‐B‐induced increase in NO transmits a signal to un‐irradiated ramets via the stolon, leading to an increase in PAL activity and UV‐B absorbing compounds content. The internal translocation of signal enables members of clonal networks to function as a whole unit and to mount an efficient defensive response to localized UV‐B radiation.  相似文献

The chlorophyte Ulva is perceived as a simple and uniform algal form, with little functional differentiation within a thallus. We compared morphology, pigmentation, photosynthesis, growth, reproduction, and UV‐B sensitivity between different thallus regions of Ulva pertusa Kjellman. Thallus thickness and cell size were significantly greater, whereas cell number was less in the basal region than in other regions. Photosynthetic pigment contents were lowest in the basal region and increased toward the marginal region. Photosynthetic capacity and photosynthetic efficiency normalized to fresh weight, area, volume, and cell number showed a progressive increase from the basal to marginal parts; however, on a chl basis those values were equal regardless of thallus part. Values of light saturation point were not statistically different between regions. Growth rates increased from marginal to basal and to middle parts of the thallus, whereas sporulation was highest in marginal (100%) followed by middle (30%) and basal parts (0%). Daily observation over 9 days showed that 56% of the basal cells divided once and did not produce spores, whereas every marginal cell went through its first division and 89% of the primary daughter cells also divided, resulting in 100% sporulation. A 7‐day treatment with PAR and PAR + UV‐A caused a significant decrease in the effective quantum yield of all thallus regions, followed by a recovery toward the initial values, whereas PAR + UV‐A + UV‐B irradiation led to greater photoinhibition and less recovery. Marked differences in the UV‐B sensitivity were observed, with marginal parts being more sensitive and basal parts most resistant.  相似文献

The bloom‐forming cyanobacterium Microcystis aeruginosa Kütz 854 was cultured with 1.05 W·m?2 UV‐B for 3 h every day, and its growth, pigments, and photosynthesis were investigated. The specific growth rates represented by chl a concentration and OD750 were inhibited 8% and 9% by UV‐B exposure, respectively. Six days of UV‐B treatment significantly reduced cellular contents of phycocyanin and allophycocyanin by 32% and 62%, respectively, and markedly increased the carotenoid content by 27%, but had little effect on the chl a content. The initial values of optimal photosynthetic efficiency for UV‐B treated samples were, respectively, 52%, 87%, and 93% of controls on days 4, 7, and 10 of growth. The light‐saturated photosynthetic rates at day 6 were significantly lower than controls grown without UV‐B. The probability of electron transfer beyond QA decreased during UV‐B exposure, and this indicated that the acceptor side of PSII was one of main damage sites. The adaptation of M. aeruginosa 854 to UV‐B radiation could be observed from light‐saturated photosynthetic rates on day 13 and diurnal changes of chl fluorescence during the late growth phase. When both exposed to higher UV‐B, samples cultured under 1.05 W·m?2 UV‐B for 9 days recovered faster than controls. It is suggested that M. aeruginosa 854 had at least three adaptive strategies to cope with the enhanced UV‐B: increasing the synthesis of carotenoids to counteract reactive oxidants caused by UV‐B exposure, degrading phycocyanin and allophycocyanin to avoid further damage to DNA and reaction centers, and enhancing the repair of UV‐B induced damage to the photosynthetic apparatus.  相似文献

The effects of elevated UV‐B (280–315 nm) radiation on the long‐term decomposition of Quercus robur leaf litter were assessed at an outdoor facility in the UK by exposing saplings to elevated UV‐B radiation (corresponding to a 30% increase above the ambient level of erythemally weighted UV‐B, equivalent to that resulting from a c. 18% reduction in ozone column) under arrays of cellulose diacetate‐filtered fluorescent UV‐B lamps that also produced UV‐A radiation (315–400 nm). Saplings were also exposed to elevated UV‐A radiation alone under arrays of polyester‐filtered fluorescent lamps and to ambient solar radiation under arrays of nonenergized lamps. After 8 months of irradiation, abscised leaves were placed into litter bags and allowed to decompose in the litter layer of a mixed deciduous woodland for 4.08 years. The dry weight loss of leaf litter from saplings irradiated with elevated UV‐B and UV‐A radiation during growth was 17% greater than that of leaf litter irradiated with elevated UV‐A radiation alone. Annual fractional weight loss of litter (k), and the estimated time taken for 95% of material to decay (3/k) were respectively increased and decreased by 27% for leaf litter exposed during growth to elevated UV‐B and UV‐A radiation, relative to that exposed to UV‐A alone. The present data corroborate those from a previous study indicating that UV‐B radiation applied during growth accelerates the subsequent decomposition of Q. robur leaf litter in soil, but indicate that this effect persists for over four years after abscission.  相似文献

Plants face various abiotic and biotic environmental factors and therefore need to adjust their phenotypic traits on several levels. UV‐B radiation is believed to impact herbivorous insects via host plant changes. Plant responses to abiotic challenges (UV‐B radiation) and their interaction with two aphid species were explored in a multifactor approach. Broccoli plants [Brassica oleracea L. convar. botrytis (L.), Brassicaceae] were grown in two differently covered greenhouses, transmitting either 80% (high UV‐B) or 4% (low UV‐B) of ambient UV‐B. Three‐week‐old plants were infested with either specialist cabbage aphids [Brevicoryne brassicae (L.), Sternorrhyncha, Aphididae] or generalist green peach aphids [Myzus persicae (Sulzer), Sternorrhyncha, Aphididae]. Plants grown under high‐UV‐B intensities were smaller and had higher flavonoid concentrations. Furthermore, these plants had reduced cuticular wax coverage, whereas amino acid concentrations of the phloem sap were little influenced by different UV‐B intensities. Cabbage aphids reproduced less on plants grown under high UV‐B than on plants grown under low UV‐B, whereas reproduction of green peach aphids in both plant light sources was equally poor. These results are likely related to the different specialisation‐dependent sensitivities of the two species. The aphids also affected plant chemistry. High numbers of cabbage aphid progeny on low‐UV‐B plants led to decreased indolyl glucosinolate concentrations. The induced change in these glucosinolates may depend on an infestation threshold. UV‐B radiation considerably impacts plant traits and subsequently affects specialist phloem‐feeding aphids, whereas aphid growth forces broccoli to generate specific defence responses.  相似文献

Cuttings of Populus kangdingensis and Populus cathayana originating from altitudes of 3500 and 1500 m in southwestern China, respectively, were grown for one growing season in the field under ambient or ambient plus supplemental ultraviolet‐B (UV‐B) radiation with two levels of nutrients. In both species, enhanced UV‐B radiation significantly increased UV‐B absorbing compounds and guaiacol peroxidase (GPX) activity, while no significant effects were observed in photosynthetic pigments and proline content. On the other hand, cuttings grown with high‐nutrient availability had larger leaf area, higher total biomass and GPX activity as well as higher water use efficiency (WUE) (as measured by stable carbon isotope composition, δ13C) when compared with low‐nutrient conditions, while UV‐B absorbing compounds and ascorbic acid (AsA) content significantly decreased. Differences in responses to enhanced UV‐B radiation and nutrient availability were observed between the two species. Nutrient‐induced increases in chlorophyll a, chlorophyll b and total chlorophyll as well as in carotenoids were greater in P. kangdingensis than in P. cathayana. In P. cathayana, enhanced UV‐B radiation significantly decreased leaf area and total biomass, while it significantly increased WUE and ascorbate peroxidase (APX). In contrast, such changes were not observed in P. kangdingensis. In addition, the effects of enhanced UV‐B radiation on leaf area, total biomass and UV‐B absorbing compounds were closely related to the nutrient status. Our results indicated that P. kangdingensis, which originates from the altitude of 3500 m and is apparently adapted to low‐nutrient and high‐UV‐B habitats, exhibits better tolerance to enhanced UV‐B radiation and greater growth under low‐nutrient availability than does P. cathayana originating from the altitude of 1500 m.  相似文献

UV‐absorbing films and nets are frequently used as covering materials for netted greenhouses and film tunnels in protected cultivation systems. This study explored the effects of such materials on the dispersal behaviour of western flower thrips (WFT), Frankliniella occidentalis, in flight cages under greenhouse conditions with additional artificial UV‐A light sources. The study involved release–recapture experiments in choice and no‐choice layouts. Different trapping methods were compared (blue sticky cards, plants and transparent cards) for recapture of thrips. In choice experiments, insects were released from a black box compartment between two tunnels covered with either UV‐transmitting or UV‐absorbing materials. A significantly higher proportion of (82–98%) WFT was recaptured in UV‐transmitting tunnels compared with UV‐absorbing tunnels. In no‐choice experiments, WFT were found to infiltrate the tunnels at different rates depending on the trap type used and experimental layout. In small‐scale dispersal experiments using blue sticky cards and plants as traps, infiltration was not significantly different between UV‐absorbing and UV‐transmitting tunnels, whereas when using transparent cards, WFT penetrated further into the UV‐transmitting plastic film tunnels. In larger‐scale dispersal experiments, plants or blue sticky cards were arranged in concentric circles around a source plant at the release point. Dispersal was found to differ depending on the method of release, but WFT tended to exhibit reduced dispersal from source plants under UV‐deficient conditions. In conclusion, our data support the hypothesis that manipulation of spectral light properties using UV‐absorbing cladding materials for protected crop stands interferes with the orientation and host finding of WFT, resulting in reduced dispersal into and within plant stands in UV‐deficient environments.  相似文献

Iron deficiency has been considered one of the main limiting factors of phytoplankton productivity in some aquatic systems including oceans and lakes. Concomitantly, solar ultraviolet‐B radiation has been shown to have both deleterious and positive impacts on phytoplankton productivity. However, how iron‐deficient cyanobacteria respond to UV‐B radiation has been largely overlooked in aquatic systems. In this study, physiological responses of four cyanobacterial strains (Microcystis and Synechococcus), which are widely distributed in freshwater or marine systems, were investigated under different UV‐B irradiances and iron conditions. The growth, photosynthetic pigment composition, photosynthetic activity, and nonphotochemical quenching of the different cyanobacterial strains were drastically altered by enhanced UV‐B radiation under iron‐deficient conditions, but were less affected under iron‐replete conditions. Intracellular reactive oxygen species (ROS) and iron content increased and decreased, respectively, with increased UV‐B radiation under iron‐deficient conditions for both Microcystis aeruginosa FACHB 912 and Synechococcus sp. WH8102. On the contrary, intracellular ROS and iron content of these two strains remained constant and increased, respectively, with increased UV‐B radiation under iron‐replete conditions. These results indicate that iron‐deficient cyanobacteria are more susceptible to enhanced UV‐B radiation. Therefore, UV‐B radiation probably plays an important role in influencing primary productivity in iron‐deficient aquatic systems, suggesting that its effects on the phytoplankton productivity may be underestimated in iron‐deficient regions around the world.  相似文献

UV RESISTANCE LOCUS8 (UVR8) is a photoreceptor for ultraviolet‐B (UV‐B) light that initiates photomorphogenic responses in plants. UV‐B photoreception causes rapid dissociation of dimeric UVR8 into monomers that interact with CONSTITUTIVELY PHOTOMORPHOGENIC1 (COP1) to initiate signal transduction. Experiments with purified UVR8 show that the dimer is maintained by salt‐bridge interactions between specific charged amino acids across the dimer interface. However, little is known about the importance of these charged amino acids in determining dimer/monomer status and UVR8 function in plants. Here we evaluate the use of different methods to examine dimer/monomer status of UVR8 and show that mutations of several salt‐bridge amino acids affect dimer/monomer status, interaction with COP1 and photoreceptor function of UVR8 in vivo. In particular, the salt‐bridges formed between arginine 286 and aspartates 96 and 107 are key to dimer formation. Mutation of arginine 286 to alanine impairs dimer formation, interaction with COP1 and function in vivo, whereas mutation to lysine gives a weakened dimer that is functional in vivo, indicating the importance of the positive charge of the arginine/lysine residue for dimer formation. Notably, a UVR8 mutant in which aspartates 96 and 107 are conservatively mutated to asparagine is strongly impaired in dimer formation but mediates UV‐B responses in vivo with a similar dose–response relationship to wild‐type. The UV‐B responsiveness of this mutant does not correlate with dimer formation and monomerisation, indicating that monomeric UVR8 has the potential for UV‐B photoreception, initiating signal transduction and responses in plants.  相似文献

UV‐B is a high‐energy component of the solar radiation perceived by the plant and induces a number of modifications in plant growth and development, including changes in flowering time. However, the molecular mechanisms underlying these changes are largely unknown. In the present work, we demonstrate that Arabidopsis plants grown under white light supplemented with UV‐B show a delay in flowering time, and this developmental reprogramming is mediated by the UVR8 photoreceptor. Using a combination of gene expression analyses and UV‐B irradiation of different flowering mutants, we gained insight into the pathways involved in the observed flowering time delay in UV‐B‐exposed Arabidopsis plants. We provide evidence that UV‐B light downregulates the expression of MSI1 and CLF, two of the components of the polycomb repressive complex 2, which in consequence drives a decrease in H3K27me3 histone methylation of MIR156 and FLC genes. Modification in the expression of several flowering time genes as a consequence of the decrease in the polycomb repressive complex 2 activity was also determined. UV‐B exposure of flowering mutants supports the involvement of this complex in the observed delay in flowering time, mostly through the age pathway.  相似文献

To better understand the interactions between PAR and UV‐B radiation in microalgae, the marine chlorophyte alga Dunaliella tertiolecta was subjected to a UV‐B flux of 4.1 W·m ? 2 (unweighted) with varying PAR fluxes. Rate constants for damage and repair processes during UV‐B exposure increased with PAR flux. However, recovery after UV‐B exposure increased with PAR up to 300 μmol quanta·m ? 2·s ? 1 1 Received 17 September 2002. Accepted 19 February 2003. , beyond which photoinhibition of PSII electron transport was found to decrease recovery rates. In the absence of PAR during the post UV‐B exposure period, no recovery was seen, indicating that perhaps the lack of light available for photosynthesis depresses repair either directly or indirectly by affecting ATP synthesis. Possible mechanisms for the observed interactions between PAR and UV‐B exposure are discussed.  相似文献

Although planar‐structured perovskite solar cells (PSCs) have power conversion efficiencies exceeding 24%, the poor photostability, especially with ultraviolet irradiance (UV) severely limits commercial application. The most commonly‐used TiO2 electron selective layer has a strong photocatalytic effect on perovskite/TiO2 interface when TiO2 is excited by UV light. Here a UV‐inert ZnTiO3 is reported as the electron selective layer in planar PSCs. ZnTiO3 is a perovskite‐structured semiconductor with excellent chemical stability and poor photocatalysis. Solar cells are fabricated with a structure of indium doped tin oxide (ITO)/ZnTiO3/Cs0.05FA0.81MA0.14PbI2.55Br0.45/Sprio‐MeOTAD/Au. The champion device exhibits a stabilized power conversion efficiency of 19.8% with improved photostability. The device holds 90% of its initial efficiency after 100 h of UV soaking (365 nm, 8 mW cm?2), compared with 55% for TiO2‐based devices. This work provides a new class of electron selective materials with excellent UV stability in perovskite solar cell applications.  相似文献

Ultraviolet‐B (UV‐B) stress activates MAP kinases (MAPKs) MPK3 and MPK6 in Arabidopsis. MAPK activity must be tightly controlled in order to ensure an appropriate cellular outcome. MAPK phosphatases (MKPs) effectively control MAPKs by dephosphorylation of phosphothreonine and phosphotyrosine in their activation loops. Arabidopsis MKP1 is an important regulator of MPK3 and MPK6, and mkp1 knockout mutants are hypersensitive to UV‐B stress, which is associated with reduced inactivation of MPK3 and MPK6. Here, we demonstrate that MPK3 and MPK6 are hyperactivated in response to UV‐B in plants that are deficient in photorepair, suggesting that UV‐damaged DNA is a trigger of MAPK signaling. This is not due to a block in replication, as, in contrast to atr, the mkp1 mutant is not hypersensitive to the replication‐inhibiting drug hydroxyurea, hydroxyurea does not activate MPK3 and MPK6, and atr is not impaired in MPK3 and MPK6 activation in response to UV‐B. We further show that mkp1 leaves and roots are UV‐B hypersensitive, whereas atr is mainly affected at the root level. Tolerance to UV‐B stress has been previously associated with stem cell removal and CYCB1;1 accumulation. Although UV‐B‐induced stem cell death and CYCB1;1 expression are not altered in mkp1 roots, CYCB1;1 expression is reduced in mkp1 leaves. We conclude that the MKP1 and ATR pathways operate in parallel, with primary roles for ATR in roots and MKP1 in leaves.  相似文献

Epidermally located ultraviolet (UV)‐absorbing phenolic compounds, flavonoids and hydroxycinnamic acid esters (HCAs), can shield the underlying tissues in plants against harmful UV‐radiation. The relative importance of the two different classes of phenolic compounds for UV‐screening was a matter of recent debate. Using a non‐invasive method based on chlorophyll fluorescence measurements to estimate epidermal UV transmittance, the relationship between epidermal UV shielding and the content of the two different groups of secondary phenolic compounds in the epidermal layers and the underlying photosynthetic mesophyll of developing rye primary leaves grown under supplementary UV‐B radiation was investigated. From the fourth to the tenth day after sowing, epidermally located flavonoids increased in an age‐ and irradiation‐dependent manner, whereas mesophyll flavonoids and epidermal HCAs, mainly ferulic acid and p‐coumaric acid esters, were constitutively present and did not vary in their contents over the observed time period. There was an excellent correlation between epidermal UV‐A and UV‐B absorbances as assessed by chlorophyll fluorescence measurements and contents of epidermal flavonoids. However, HCAs showed an additional contribution to UV‐B shielding. In contrast, mesophyll flavonoids did not seem to play a respective role. When absorbances of the abaxial and adaxial epidermal layers were compared, it became apparent that in fully expanded primary leaves epidermal tissues from both sides were equally effective in absorption of UV‐radiation. However, the earlier and more UV‐exposed abaxial epidermis of young unrolling leaves showed a significantly higher absorption. It is shown that in early stages of development the epidermal HCAs are the dominant UV‐B protective compounds of the primary leaf. This function is increasingly replaced by the epidermal flavonoids during leaf development and acclimation. The application of chlorophyll fluorescence measurements has been proven to be a useful tool for estimating relative contents of these compounds in epidermal tissue.  相似文献
