假寄生和毒液注射实验表明啮小蜂Tetrastichussp .毒液能永久麻痹亚洲玉米螟蛹Ostriniafurnacalis,较高剂量的毒液对亚洲玉米螟蛹有不同程度的麻痹作用。麻痹的蛹在室温下较长时间内不腐烂和干化。低剂量毒液能延长亚洲玉米螟蛹期 1~ 3d ,导致畸形成虫。啮小蜂初产卵转移寄主实验证实毒液可以帮助啮小蜂卵在亚洲玉米螟蛹体内的发育 ,啮小蜂初产卵在转移寄主体内发育到羽化时间为 2 5d ,比它们在原寄主内的羽化时间增加 8d左右 ,初产蜂卵与 1个毒液储液囊当量的毒液同时注入转移寄主体内 ,初产卵的羽化时间明显缩短 ,死亡率降低  相似文献

1. Parasitoid wasps sting and inject venom into arthropod hosts, which alters host metabolism and development while keeping the host alive for several days, presumably to induce benefits for the parasitoid young. 2. This study investigates the consequences of host envenomation on development and fitness of wasp larvae in the ectoparasitoid Nasonia vitripennis, by comparing wasps reared on live unstung, previously stung, and cold‐killed hosts. Developmental arrest and suppression of host response to larvae are major venom effects that occur in both stung and cold‐killed hosts, but not in unstung hosts, whereas cold‐killed hosts lack venom effects that require a living host. Thus, cold‐killed hosts mimic some of the effects of venom, but not others. 3. Eggs placed on live unstung hosts have significantly higher mortality during development; however, successfully developing wasps from these hosts have similar lifetime fecundity to that of wasps from cold‐killed or stung hosts. Therefore, although venom is beneficial, it is not required for wasp survival. 4. While wasps developing on both cold‐killed and stung hosts have similar fitness levels, rearing multiple generations on cold‐killed hosts results in significant fitness reductions of wasps. 5. It is concluded that the largest benefits of venom are induction of host developmental arrest and suppression of host response to larva (e.g. immune responses), although more subtle benefits may accrue across generations or under stressful conditions.  相似文献

To determine differential gene expression profiles in the venom gland and sac (gland/sac) of a solitary hunting wasp species, Orancistrocerus drewseni Saussure (1857), a subtractive cDNA library was constructed by suppression subtractive hybridization. A total of 498 expressed sequence tags (EST) were clustered and assembled into 205 contigs (94 multiple sequences and 111 singletons). About 65% (134) of the contigs had matched BLASTx hits (E≤10?4). Among these, 115 contigs had similarity to proteins with assigned molecular function in the Gene Ontology database, and most of them (112 contigs, 83%) were homologous to genes from Hymenoptera, particularly to Apis mellifera (98 contigs). The contigs encoding hyaluronidase and phospholipase A2, known to be main components of wasp venoms, were found in high frequencies (27 and 4%, respectively, as judged by the number of ESTs) in the gene ontology category of catalytic activity. Full‐length open reading frames of hyaluronidase and phospholipase A2 were characterized and their abundance in the venom gland/sac was confirmed by quantitative real‐time PCR. Several contigs encoding enzymes, including zinc‐metallopeptidases that are likely involved in the processing and activation of venomous proteins or peptides, were also identified from the library. Discovery of venom gland/sac‐specific genes should promote further studies on biologically active components in the venom of O. drewseni. © 2009 Wiley Periodicals, Inc.  相似文献

The number of mature eggs carried by a female parasitoid at any given moment (egg load) is a fitness‐related parameter affecting reproductive potential and impacting upon host population dynamics. Microplitis rufiventris Kokujev (Hymenoptera: Braconidae) is a solitary koinobiont endoparasitoid wasp of several noctuid pests, including Spodoptera littoralis. The number of mature eggs carried by females at emergence is approximately 50. The rate of egg maturation is strongly affected both by feeding status and access to host larvae. In early adult life, egg maturation rates are lower for 6–72 h in fed wasps compared with food‐deprived wasps. When given access to hosts, honey‐fed wasps live for approximately 9 days with high lifetime fecundity (226 eggs). By contrast to early adult life, the total realized fecundity is positively affected by feeding status, where water‐fed and starved females have 140 and 107 eggs, respectively. Egg resorption is most pronounced in the later life of females. The results suggest, in addition to confirming the effect of honey‐feeding on total fecundity, that fecundity of starved wasps includes rapid egg maturation early in life, which potentially could improve the performance of the parasitoid as a biological control agent.  相似文献

Polybia paulista wasp venom possesses three major allergens: phospholipase A1, hyaluronidase and antigen-5. To the best of our knowledge, no hyaluronidase from the venom of Neotropical social wasps was structurally characterized up to this moment, mainly due to its reduced amount in the venom of the tropical wasp species (about 0.5% of crude venom). Four different glycoproteic forms of this enzyme were detected in the venom of the wasp Polybia paulista. In the present investigation, an innovative experimental approach was developed combining 2-D SDS-PAGE with in-gel protein digestion by different proteolytic enzymes, followed by mass spectrometry analysis under collision-induced dissociation CID) conditions for the complete assignment of the protein sequencing. Thus, the most abundant form of this enzyme in P. paulista venom, the hyaluronidase-III, was sequenced, revealing that the first 47 amino acid residues from the N-terminal region, common to other Hymenoptera venom hyaluronidases, are missing. The molecular modeling revealed that hyaluronidase-III has a single polypeptide chain, folded into a tertiary structure, presenting a central (β/α)5 core with alternation of β-strands and α-helices; the tertiary structure stabilized by a single disulfide bridge between the residues Cys189 and Cys201. The structural pattern reported for P. paulista venom hyaluronidase-III is compatible with the classification of the enzyme as member of the family 56 of glycosidase hydrolases. Moreover, its structural characterization will encourage the use of this protein as a model for future development of "component-resolved diagnosis".  相似文献

The venoms of the social wasps evolved to be used as defensive tools to protect the colonies of these insects against the attacks of predators. Previous studies estimated the presence of a dozen peptide components in the venoms of each species of these insects, which altogether comprise up to 70% of the weight of freeze-dried venoms. In the present study, an optimized experimental protocol is reported that utilizes liquid chromatography coupled to electrospray ionization mass spectrometry for the detection of peptides in the venom of the social wasp Polybia paulista; peptide profiles for both intra- and inter-colonial comparisons were obtained using this protocol. The results of our study revealed a surprisingly high level of intra- and inter-colonial variability for the same wasp species. We detected 78–108 different peptides in the venom of different colonies of P. paulista in the molar mass range from 400 to 3000 Da; among those, only 36 and 44 common peptides were observed in the inter- and intra-colony comparisons, respectively.  相似文献

The biological activity of venom from Pimpla turionellae L. (Hymenoptera: Ichneumonidae) was examined in vivo toward larvae and pupae of Galleriae mellonella L. (Lepidoptera: Pyralidae), and in vitro toward bacterial and fungal cultures, as well as cultured insect cells. Pupae of G. mellonella were far more susceptible to the venom than larvae. At low doses of venom [0.1 venom reservoir equivalents (VRE)], pupal abdominal mobility was inhibited within 30 min, and by 24 h, all pupae injected with venom concentrations >0.5 VRE were completely paralyzed. These same doses of venom resulted in an inhibition of adult emergence. Host larvae were far less sensitive to wasp venom as evidenced by all venom injected larvae remaining responsive to mechanical stimulation by 1 h post injection, even at concentrations equivalent to 1 venom reservoir. Eventually (>2 h at 25 degrees C), venom-injected larvae became immobile, then flaccid, and all died within 24 h post-injection. At lower concentrations of wasp venom, the onset of paralysis was delayed by comparison to that evoked by 1 VRE, and few host larvae were able to pupate. Development of host larvae to adult emergence was also reduced in a dose-dependent manner, with eclosion completely prevented at high concentrations (>0.5 VRE) of venom. Venom doses <0.5 VRE did not appear to induce paralysis or alter larval development. When venom was incubated with bacterial or fungal cultures, no antimicrobial activity was detected. However, wasp venom was found to be cytotoxic and cytolytic to cultured cells derived from the cabbage looper Trichoplusia ni Hubner (Lepidoptera: Noctuidae) and the yellow fever mosquito, Aedes aegypti (L.) (Diptera: Culcidae). Though both cell types displayed similar susceptibility in terms of LC50s, the lepidopteran cells responded much more rapidly with regard to the onset of morphological changes and the timing of cell death. The codling moth Cydia pomonella (Linnaeus) (Lepidoptera: Tortricidae) is a serious pest of pome fruit crops. A natural enemy of codling moth, the larval ectoparasitoid Mastrus ridibundus (Gravenhorst) (Hymenoptera: Ichneumonidae) has been imported into South America from the USA but little is known about the biology and ecology of the wasp, knowledge that is needed to design an efficient strategy of release and establishment. Experiments were carried out to assess important traits of the biology of the parasitoid in relation to its possible use as a biocontrol agent for codling moth. When M. ridibundus females were offered larvae ranging in weight from 37 to 78 mg, they oviposited more eggs on heavier hosts. In another study, the adult wasps were offered honey, diluted honey (10%) or pollen in paired choice tests and both males and females preferred honey over the other two foods. Females preferred 10% honey over pollen, while the males showed the opposite preference. Honey‐fed females lived longer than starved females. Adults died rapidly at 35°C, while they lived 20 days at 25°C and 12–17 days at 15°C. Female wasps had on average 25 ± 14 and 18 ± 11 progeny at 15 and 25°C, respectively, but they did not had progeny at 35°C. The development time (egg to adult emergence) was on average 44 ± 7 and 24 ± 2 days at 15 and 25°C respectively. Immature insects did not reach the adult stage at 35°C.  相似文献

Three different concentrations of the antibiotic tetracycline in honey were tested for their influence on the offspring production and longevity of the parasitoid wasp Encarsia formosa. Several earlier publications did not provide a conclusive answer on the effect that the Wolbachia have on these wasps. The results of our experiments show that at high tetracycline hydrochloride concentrations in honey (50mg/ml) the antibiotic is toxic to the females, all females died within three days after the antibiotic treatment. The concentration 5mg/ml was less toxic although the treated females also lived shorter and produced less offspring than the control females. At the lowest tested concentration of 1mg/ml there was no significant difference either in offspring production or in longevity between the control and the treated females. The antibiotic treatment at both 5 and 1mg/ml resulted in exclusively male progeny after the first two days of oviposition. These results are consistent with the theory that in species in which all individuals are infected the Wolbachia should not impose a large fitness cost.  相似文献

Parasitoid wasps are diverse and ecologically important insects that use venom to modify their host's metabolism for the benefit of the parasitoid's offspring. Thus, the effects of venom can be considered an ‘extended phenotype’ of the wasp. The model parasitoid wasp Nasonia vitripennis has approximately 100 venom proteins, 23 of which do not have sequence similarity to known proteins. Envenomation by N. vitripennis has previously been shown to induce developmental arrest, selective apoptosis and alterations in lipid metabolism in flesh fly hosts. However, the full effects of Nasonia venom are still largely unknown. In this study, we used high throughput RNA sequencing (RNA‐Seq) to characterize global changes in Sarcophaga bullata (Diptera) gene expression in response to envenomation by N. vitripennis. Surprisingly, we show that Nasonia venom targets a small subset of S. bullata loci, with ~2% genes being differentially expressed in response to envenomation. Strong upregulation of enhancer of split complex genes provides a potential molecular mechanism that could explain the observed neural cell death and developmental arrest in envenomated hosts. Significant increases in antimicrobial peptides and their corresponding regulatory genes provide evidence that venom could be selectively activating certain immune responses of the hosts. Further, we found differential expression of genes in several metabolic pathways, including glycolysis and gluconeogenesis that may be responsible for the decrease in pyruvate levels found in envenomated hosts. The targeting of Nasonia venom effects to a specific and limited set of genes provides insight into the interaction between the ectoparasitoid wasp and its host.  相似文献

Chemicals from the venom gland elicited alarm behaviour and attack in the Asian polistine wasp Polybioides raphigastra. When presented with crushed venom glands workers of this wasp respond with a mass stinging attack. Gas chromatography–mass spectrometry analyses show that the major volatiles in the venom gland are alkanes, monounsaturated alkenes and 2-alcohols. Several pyrazines, a spiroacetal and aromatics were also identified as trace compounds. The anatomy and morphology of the sting apparatus are reported, and we describe sting autotomy in this wasp. This is the first such report for the Ropalidiinae. The structures responsible for autotomy are likely to be large barbs present on the sting lancets, and a conspicuous tooth present on the medial side of the left lancet. Sting autotomy in P. raphigastra probably plays an important role in the localization of sites of attack by wasps defending the nest.  相似文献

Metaphycus flavus (Howard) and M. stanleyi Compere (Hymenoptera: Encyrtidae) are currently being screened for use as augmentative biological control agents of citrus-infesting soft scales (Homoptera: Coccidae). Two factors were investigated, host quality-dependent sex allocation and local mate competition, which likely influence these parasitoid's sex allocation strategies and are therefore of interest for their mass-rearing. The results of these studies suggested that, under the mass-rearing protocol that is envisioned for these parasitoids, offspring sex ratios in both M. flavus and M. stanleyi are dominated by host quality (= size) influences, but not by interactions with other females. These results indicated that host size strongly influences offspring sex ratios and brood sizes; larger hosts led to more female offspring and larger broods. In contrast, increasing the number of parental females did not lead to fewer female offspring as expected under local mate competition. Additionally, within-brood sex ratios did not vary with brood size; this result is inconsistent with expected sex ratios due to local mate competition. Other results also indicated that host quality was a dominant influence on M. flavus' and M. stanleyi's sex ratios. Larger hosts led to a larger size in the emerging wasps, and larger wasps had greater egg loads and lived longer than smaller wasps. However, wasp longevity, and the influence of wasp size on longevity were mediated by a wasp's diet. Metaphycus flavus females lived the longest when they had access to hosts, honey, and water, followed by honey and water, and shortest when they had access to water alone; M. stanleyi females lived longest with honey and water, followed by hosts, honey, and water, and shortest with water alone. Greater wasp size led to greater longevity in females only when they had access to food (honey, or hosts and honey). Finally, other results suggested that both M. flavus and M. stanleyi are facultatively gregarious. Wasp size did not decrease with brood size as expected under superparasitism. Overall, the results of these studies suggested that holding newly emerged females of both M. flavus and M. stanleyi for several days in the presence of an appropriate food source before field release could enhance a female's performance as an augmentative biological control agent. It increases their initial life expectancy following release, and maximizes the females' egg load (both Metaphycus species) and resources for replacing oviposited eggs (M. flavus only).  相似文献

In a study of the quality ofTrichogramma pretiosum Riley (Hymenoptera: Trichogrammidae), we compared female wasps emerging from natural hosts, parasitized in the laboratory or the field with those emerging from factitious hosts used for commercial mass production. Females from the natural hosts were larger, more fecund, and longer lived than those from the factitious hosts. Compared to small females, large female wasps are substantially more fecund when honey (carbohydrate) is available but marginally more fecund when honey is unavailable. The size of a femaleT. pretiosum depends on two factors: the size of the host egg from which it emerges even when the wasp was gregarious, and the number of conspecifics that emerge with it. The similarities in the size distribution of female wasps emerging from natural hosts, in conjunction with the mechanism by whichTrichogramma measure host size and allocate eggs accordingly, suggests the hypothesis that size related components of fitness in femaleT. pretiosum are under strong selection in the field.  相似文献

Dibrachys boarmiae was recently found as a parasitoid of the pistachio twig borer moth, Kermania pistaciella, an important pistachio pest in Iran. Several biological and behavioural aspects of the wasp were studied under controlled conditions using three different lepidopterous host species, K. pistaciella, Sitotroga cerealella and Galleria mellonella. Development time from egg to adult decreases from 54 days at 15°C to 12.6 days at 32.5°C. The thermal constant was estimated 270°D, and the lower development threshold was at 9.9°C. Longevity of females that had the opportunity to oviposit and access to food (honey) was on average 23 days, and 134 offspring were produced during 13 days of oviposition. Ninety percent of ovipositions occur during the first 10 days of the parasitoid's life, although no pre-oviposition period was found. If females were deprived of food, longevity as well as fecundity, were drastically reduced to 8 days and only 79 offspring respectively. Females were able to mate immediately after emergence and the sex ratio of the progeny was strongly female biased on all examined hosts, especially on the largest host G. mellonella (0.08 male). When females were kept with a male throughout their life, the total number of progeny decreased to 92 wasps on average but the sex ratio was unaffected. The parasitoid significantly preferred to attack the pre-pupal stage, and this influenced the size of clutches allocated to different host stages. The parasitoid laid larger clutches on bigger hosts.  相似文献

The endoparasitoid wasp Asobara japonica has highly poisonous venom: the host Drosophila larvae are killed by envenomation at a dose that is naturally injected by the female wasp at parasitism. This insecticidal venom is neutralized, however, because A. japonica introduces lateral oviduct components soon after venom injection at oviposition. Although the venom and lateral oviduct components of this parasitoid have been partially characterized, how the venom components favor successful development of wasp eggs and larvae in the host remains ambiguous. Here, we demonstrated that A. japonica venom did not affect host humoral immune responses, determined as expression of antimicrobial peptide (AMP) genes, but significantly diminished two cellular responses, spreading and phagocytosis, by host hemocytes. Moreover, venom components drastically elevated a serine protease‐like activity 4 h after its injection. The lateral oviduct components did not negate the detrimental effects of the venom on host cellular immunities, but significantly reduced the venom‐induced elevation of protease activity. Both active factors in venom and lateral oviduct components were roughly characterized as heat‐labile substances with a molecular mass of at least 10 kDa. Finally, venom of A. japonica, with a wide host range, was found to be much more toxic than that of Asobara rossica, which has a limited host range. These results reveal that A. japonica venom toxicity allows exploitation of a broader range of host insects because it is essential to overcome cellular immune responses of the host for successful parasitism.  相似文献

枸杞瘿螨姬小蜂Cirrospilus eniophyesiYang(新种)是枸杞瘿螨Aceria pallidaKeifer的重要天敌,以幼虫在虫瘿内取食枸杞瘿螨的成虫和若虫。在(25±1)℃下,该蜂完成1个世代需16~21d。成虫寿命及产卵量与补充营养有关,取食10%蜂蜜水的雌虫寿命为(6.94±0.67)d,雄虫为(5.14±0.49)d。不进行补充营养的雌雄成虫寿命分别为(1.96±0.27)d和(1.06±0.29)d;补充10%蜂蜜水和水可显著提高产卵量,每雌平均产卵量分别为(29.13±6.12)粒和(23.14±3.16)粒,产卵高峰期在羽化后的第2天。成虫羽化时间多集中在6:00~8:00am和4:00~6:00pm,羽化当天即可交尾。枸杞瘿螨姬小蜂自然种群消长与寄主虫口密度相关,在6月底7月初寄生率达到83.3%。  相似文献

为证明大胡蜂Vespa magnifica(Smith)蜂毒具有较大的药用开发价值,本研究采用超高效液相色谱-质谱和电泳技术对其多肽和蛋白质的分布进行分析,发现其蛋白质的相对分子质量主要分布在17~45kDa范围内。蜂毒多肽类物质的相对分子质量呈"单峰"式分布,61%在500~3000Da范围内,为大胡蜂蜂毒中多肽含量最为丰富的部分。通过牛津杯法对蜂毒的抑菌活性进行研究,且以HepG2人肝癌细胞及B16黑色素瘤细胞为研究对象,用MTT法检测蜂毒的细胞毒性活性,证明其具有良好的抑菌作用和细胞毒活性,其结果与已报道的其他蜂类既有相似性又存在具体差异,展示了大胡蜂蜂毒的分子多样性,为后续该毒素的物质基础研究及药用价值开发提供参考。  相似文献
