Nowadays research and clinical studies of human reproductive endocrinology are generally carried out using human blood reproductive hormone assays. However the acquisition of human blood samples has some shortcomings. In search of new approaches, we paid attention to the fact that progesterone can be detected in cow's hair. Consequently we investigated whether or not steroid hormones are measurable in human hair. The results showed that the levels of steroid hormones in hair are not affected by shampoo and do not significantly vary between different segments of hair (i.e. top, middle and basal segments). The menstrual estradiol and progesterone rhythm of female hair is similar to that of female serum. The ratio of hair estradiol to serum estradiol in the female is 41.2% and that of hair progesterone to serum progesterone is 59.0%; the ratio of hair testosterone to serum testosterone in male is 116%. There are significant correlations between hair and serum steroid hormones of healthy human adult: γ (estradiol)=0.395 (n=20), p<0.05; γ (progesterone)=0.440 (n=22), p<0.025 and γ (testosterone)=0.395 (n=25), p<0.05.  相似文献

Summary Colonies of cells of epithelioid appearance were identified in monolayer cultures grown up to 50 days from normal human endometrial cell suspensions obtained by a method designed to insure a maximum harvest of glandular cells. Groups of these cells were separated from stromal cells by means of cloning cylinders. Studies comparing the ultrastructure of cells of this type to fresh endometrial tissue revealed a number of similarities. The morphological characteristics common to both types of samples included junctional complexes, perinuclear microfilaments and microvilli with glycocalyx. Other common features were prominent nucleoli, well developed Golgi, rough endoplasmic reticulum and membranebound electron-dense bodies in the cytoplasm. A stripping technique applie to the fetal bovine serum used in the nutrient medium made it possible to initiate cultures in a steroidfree environment and to maintain them in the presence of the specified concentration of estradiol and/or progesterone. Isolation of epithelial cells of endometrium in monolayer culture may provide a useful model system in which to study the specific effects of steroid hormones on cellular function and differentiation. Plasma steroid concentrations in two and three year-old male yellow perch maintained under two different photothermal regimes were investigated. Initially, all fish kept indoors were exposed to the same water temperature (22 °C) and photoperiod (15L:9D). By the end of August, following the first sampling, fish were exposed to different photothermal regimes. Groups A2 (2 year old) and A3 (3 year old) were maintained under photothermal conditions similar to those of southern Ohio. Groups B2 (2 year old) and B3 (3 year old) were exposed to a condensed light/temperature regime designed to accelerate maturation. Testosterone (T) was the major circulating androgen in all groups. In regime A fish, plasma concentrations of 11-ketotestosterone (11KT) and T were very low in August, increased in October and remained elevated until March. In regime B plasma androgens were high until February and then dropped abruptly in March. The elevated circulating levels of 11KT and T were associated with production of sperm. The highest sperm concentration in the groups A3 and B3 was observed in February and December, respectively. There were no major differences in profiles and levels of plasma steroids between two age categories within each photothermal regime. These data indicate that the compression of the photothermal cycle accelerated both the occurrence of the low postspawning levels of circulating steroids and the completion of milt production. Higher sperm concentration observed in B3 group earlier in the season compared to A3 group also support the notion that the condensed photothermal cycle accelerated gonadal maturation. It appears that modification of the environmental cues may be a useful tool for manipulation of reproductive processes in male yellow perch. The physicochemical properties of DPPC (1,2-dipalmitoyl-sn-glycero-3-phosphocholine) liposomes used for topical application are pharmaceutically important. Therefore the aim of our study was to establish rapid and efficient methods for the exact characterisation of the physicochemical properties of extruded DPPC liposomes containing low concentration (0.5%, w/w) of different, therapeutically interesting steroid hormones, named 17-beta-estradiol, cpa (cyproterone acetate) and finasteride. In a first step it could be shown that all drugs influenced the liposome size and changed the zeta potential compared to the placebo formulations. Our further analytical strategy was to use micro-calorimetry and ATR-FTIR (Fourier transformed infrared spectroscopy), two powerful and non-destructive methods to confirm the drug incorporation into the liposomes by proving interactions between the phospholipids and the steroids. Thereby it was even possible to localize the location of interaction. The characteristic phase transition temperatures of the phospholipid were decreased by the hormones which was detected by micro-DSC (differential scanning calorimetry). The results of the ATR-FTIR measurements indicated shifts of the specific lipid peaks, the C=O stretching bands and PO(2)(-) antisymmetric double stretching band, in the gel and liquid crystalline phase. A polar as well as a non-polar interaction could be proven. It could be shown that the investigated steroid hormones changed the physical properties of the phospholipid bilayers.  相似文献

To examine the effects of ovarian hormones on the behavior of female Macaca fascicularis and their male partners, daily 1-hr behavior tests were conducted while ovariectomized females were (1) untreated, (2) given estradiol benzoate (EB) (5 μg subcutaneously [s.c.]/day), (3) given estradiol benzoate together with increasing doses of progesterone (P) (5 mg, 10 mg, and 20 mg. s.c./day), and (4) given testosterone propionate (TP) (0.25 mg s.c./day) (six pairs, 540 tests). Weekly blood samples were analyzed by radioimmunoassay for plasma hormone levels (81 samples). Estrogen treatment produced plasma estradiol levels similar to those of intact females during the late follicular phase of the menstrual cycle. Additional progesterone at the lowest dose produced plasma progesterone levels similar to or somewhat higher than those during the midluteal phase, while higher doses produced supraphysiological levels. Androgen treatment resulted in plasma levels well above the physiological range. Hormone treatments produced highly significant effects on the sexual, social, and aggressive interactions of the pairs. As in rhesus monkeys, estrogen increased male and female sexual activity, and increasing doses of additional progesterone reversed these effects. Unlike in rhesus monkeys, testosterone propionate increased both female sexual motivation (invitations) and also male sexual activity and ejaculatory performance. The direction of the hormone-dependent changes in grooming and aggressive interactions confirmed earlier results with intact females and indicated that aggressive interactions and male grooming times were highest, and female grooming times were lowest, when copulatory activity was at its height.  相似文献

The catalytic requirements and the role of P450 3A9, a female-specific isoform of CYP3A from rat brain, in the metabolism of several steroid hormones were studied using recombinant P450 3A9 protein. The optimal steroid hormone hydroxylase activities of P450 3A9 required cholate but not cytochrome b5. P450 3A9 was active in the hydroxylation reactions of testosterone, androstenedione, progesterone and dehydroepiandrosterone (DHEA). No activity of P450 3A9 toward cortisol was detectable under our reconstitution conditions. Among all the steroid hormones examined, female-specific P450 3A9 seemed to catalyze most efficiently the metabolism of progesterone, one of the major female hormones, to form three mono-hydroxylated products, 6-, 16-, and 21-hydroxyprogesterone. Our data also showed that P450 3A9 can catalyze the formation of a dihydroxy product, 4-pregnen-6, 21-diol-3, 20-dione, from progesterone with a turnover number, 1.3 nmol/min/nmol P450. Based on the Vmax/Km values for P450 3A9 using either 21-hydroxprogesterone or 6-hydroxyprogesterone as a substrate, 4-pregnen-6, 21-diol-3, 20-dione may be formed either by 6-hydroxylation of 21-hydroxprogesterone or 21-hydroxylation of 6-hydroxyprogesterone. As a major isoform of CYP3A expressed in rat brain, the activities of P450 3A9 toward two major neurosteroids, progesterone and DHEA suggested a possible role for P450 3A9 in the metabolism of neurosteroids.  相似文献

Implicit motives for power and affiliation, salivary levels of testosterone, estradiol, and progesterone, and relationship status were measured in 18 normally cycling (NC) women, 18 women using oral contraceptives (OC), and 18 men at three assessments, corresponding to the menstrual, midcycle, and premenstrual phases of women's menstrual cycle. NC and OC women had elevated levels of affiliation motivation and decreased levels of power motivation at midcycle. Power motive changes were particularly pronounced in NC women across cycle phases. OC women and participants not engaged in an intimate relationship had significantly heightened levels of affiliation motivation, averaged across all cycle phases. Testosterone and power motivation, both averaged across all cycle phases, were positively correlated in men and in single women, but not in women engaged in an intimate relationship. Averaged levels of estradiol and power motivation were positively correlated in engaged women, but not in single women or men. Averaged levels of progesterone and affiliation motivation were negatively correlated in men, and there was evidence for a positive association between luteal affiliation motivation and periovulatory and luteal progesterone in NC women. This study therefore provides evidence that implicit motivational states fluctuate across the menstrual cycle, that the power motive is associated with testosterone and, in women, with estradiol, and that the affiliation motive and progesterone are associated in different ways in men and NC women.  相似文献

The growth regulators reviewed include auxins, gibberellins, cytokinins, ethephon, CCC, Alar, ABA, morphactin and TIBA. None of them is used routinely. The chlorophenoxy and naphthalene compounds are definitely harmful. Blood samples from 15 breeding male Murrah buffaloes were collected during the winter, summer and monsoon seasons. Seminal characteristics and sexual behaviour were also studied. Serum samples were analysed for testosterone, progesterone and estradiol-17beta levels by radioimmunoassay. The studies showed significantly lower values for testosterone during winter (0.53 +/- 0.06 ng/ml) than during summer (1.22 +/- 0.19 ng/ml) and monsoon (1.06 +/- 0.12 ng/ml). The progesterone level was lowest during monsoon (84 +/- 9 pg/ml), intermediate during winter (115 +/- 14 pg/ml) and highest during summer (224 +/- 24 pg/ml). The mean level of estradiol-17beta was almost double (9 +/- 0.7 pg/ml) during monsoon as compared to winter (5 +/- 0.1 pg/ml). The correlations between hormone levels, seminal characteristics and sexual behaviour were of low magnitude.  相似文献

Concentrations of estradiol-17β (E2), progesterone (Po), and luteinizing hormone (LH) in serum and estrone glucuronide (E1G) and pregnanediol glucuronide (PdG) in urine were determined by direct radioimmunoassay in five yearling and three adult female white-tailed deer (Odocoileus virginianus) from 14 October to 20 December 1985. Elevated levels of LH were recorded before the first estrus of the breeding season and immediately preceding or during behavioral estrus. Urinary E1G and PdG concentrations were poorly correlated with serum levels of E2 and Po, respectively. Serum Po levels prior to the first estrus of the breeding season indicate that pubertal yearlings experience silent ovulations and suggest that silent ovulations may be important in the transition from adolescence to cycling adult in white-tailed deer. © 1992 Wiley-Liss, Inc.  相似文献

Seven men, 21–30 years old, and six men, 72–90 years old, were given human chorionic gonadotropin (HCG), 3,000 units a day for four days. The concentration of 17β-estradiol (1), estrone and testosterone were measured in plasma samples drawn before and during the course of HCG administration. The administration of HCG resulted in higher levels of both 17β-estradiol and testosterone in the younger as compared to the older men although the percentage increases over baseline levels were similar in both groups. HCG administration resulted in similar, absolute and relative increases of estrone in both young and old men. The levels of 17β-estradiol were higher on day 3 as compared to day 5 in young men. The relative ability to respond to exogenous gonadotropins appears to be preserved despite ageing and loss of libido and potentia. The absolute response is, however, somewhat less in old men as compared to young.  相似文献

We have examined the incorporation and metabolism of [14C] stearic acid within the total lipids of HTC rat-hepatoma cells in suspension culture in presence and in absence of steroidal hormone stimulation. Both, glucocorticoids (dexamethasone, cortisol and corticosterone) and mineralocorticoids (deoxycorticosterone and aldosterone) as well as the estrogen -estradiol and the androgen testosterone enhanced the extent of 9 desaturation to oleic acid of the saturated precursors, whereas only the two mineralocorticoids affected the incorporation rate of the exogenous acid into total cellular lipids, thus promoting a little stimulation. Furthermore, all the hormones tested increased the radiolabelling of the total cellular phospholipids except deoxycorticosterone and testosterone, the former having no effect and the latter exerting a moderate inhibition. On the other hand, the incorporation of14C into neutral lipids was stimulated by testosterone, in contrast to the inhibition of this parameter observed exclusively with either the mineralocorticoids or the estrogen. Within the phospholipid subclasses, the radiolabelling of phosphatidylcholine was augmented by means of all the steroids tested save deoxycorticosterone and testosterone, whereas phosphatidylethanolamine exhibited a decrease only in the presence of testosterone. In a similar fashion, within the neutral lipids, the predominating triglyceride fraction was preferentially labelled—at the expense of other subclasses of lesser abundance—upon treatment with the steroids except aldosterone, which exerted no effect. Despite several studies on fish hormone therapy, finding new candidates may provide more reproductive efficiency in artificial propagation. Kisspeptins, being upstream of the hypothalamic–pituitary-gonadal axis, appear to play a key role in the reproduction process. In the present study, the effect of different variants of kisspeptide, including goldfish (Carassius auratus) kiss1 kisspeptin (Kiss1), human kisspeptin (Hkiss), and their combination (Kiss1 + H), on the reproductive indices of goldfish broodstock in comparison to Ovaprim (a typical synthetic Gnrh hormone) was investigated. Peptides (Kiss1 and Hkiss) were synthesized using a solid-phase synthesis approach. Kiss1 and Hkiss were injected at a dose of 100 μg kg−1 body weight, blood samples were taken 6 h after injection and sex hormones (E2, Dhp, and 11-Kt), gonadotropins (Lh and Fsh), cortisol and reproductive indices (fecundity, fertilization and hatching percentage) were measured. The results showed a significant increase of plasma sex hormones and gonadotropins in fish treated with kisspeptins. In addition, the cortisol and lipoprotein lipase in Kiss1, Hkiss and Kiss1 + H were remarkably increased compared to Ovaprim. In conclusion, kisspeptins could be a more suitable candidate than Ovaprim for accelerating and synchronizing oocyte maturation in the fisheries industry.  相似文献

Late wilt, a severe vascular disease of maize caused by the fungus Harpophora maydis, is characterized by rapid wilting of maize plants before tasseling and until shortly before maturity. The pathogen is currently controlled by resistant maize cultivars, but the disease is constantly spreading to new areas. The plant’s late phenological stage at which the disease appears suggests that plant hormones may be involved in the pathogenesis. This work revealed that plant growth hormones, auxin (Indole-3-acetic acid) and cytokinin (kinetin), suppress H. maydis in culture media and in a detached root assay. Kinetin, and even more auxin, caused significant suppression of fungus spore germination. Gibberellic acid did not alter colony growth rate but had a signal suppressive effect on the pathogens’ spore germination. In comparison, ethylene and jasmonic acid, plant senescing and defense response regulators, had minor effects on colony growth and spore germination rate. Their associate hormone, salicylic acid, had a moderate suppressive effect on spore germination and colony growth rate, and a strong influence when combined with auxin. Despite the anti-fungal auxin success in vitro, field experiments with dimethylamine salt of  2,4-dichlorophenoxyacetic acid (that mimics the influence of auxin) failed to suppress the late wilt. The lines of evidence presented here reveal the suppressive influence of the three growth hormones studied on fungal development and are important to encourage further and more in-depth examinations of this intriguing hormonal complex regulatory and its role in the maize-H. maydis interactions.  相似文献

Cyclodextrins with different ring size and ring substituents were tested for recombinant mink and porcine growth hormones aggregation suppression in the refolding process from Escherichia coli inclusion bodies. Methyl-β-cyclodextrin and 2-hydroxypropyl-β-cyclodextrin show a positive effect on the aggregation suppression of both proteins. The influence of different methyl-β-cyclodextrin and 2-hydroxypropyl-β-cyclodextrin concentrations on the renaturation yield of both growth hormones was investigated. Moreover, methyl-β-cyclodextrin and 2-hydroxypropyl-β-cyclodextrin suppress not only folding-related, but also temperature-related aggregates formation of both proteins. Circular dichroism experiments (monitoring of protein solution turbidity by registering high tension voltage) showed that the onset temperature of aggregation of both growth hormones increased with increasing 2-hydroxypropyl-β-cyclodextrin concentration. In conclusion, cyclodextrins have perspectives in biotechnology of veterinary growth hormones not only for protein production, but also for its storage.  相似文献

This study of the human growth hormone binding protein (GHBP) was undertaken using several samples of hGH, extractive or recombinant, from different origins. They were labelled in identical conditions and assayed by gel chromatography after incubation with three human sera having different levels of binding activity. For each serum the binding activities of the five recombinant hormones were very close and significantly higher (P less than 0.005) then the binding activities of the 2 extractive hormones. A radioactive peak which appeared in the zone of high molecular weights was more important with extractive than with recombinant hormones (P less than 0.01). This peak increased with the ageing of the tracer and appeared even when the tracer was incubated in the absence of serum. Thus, it is for its main part not related to another binding protein but, more likely, to a polymerization of the hormone. These data point out the importance of accurate technical conditions to have a reproducible assay for GHBP and to interpret the results in studies of growth disturbances.  相似文献

Summary The three segments (S1, S2, S3) of the proximal tubule of the rat kidney were investigated, with special reference to lysosomes, after castration, estradiol application, and at the end of pregnancy. Especially in S1 and S2 castration induces an increase of cellular autophagy. The nuclei become smaller; endoplasmic reticulum (ER), ribosomes, and Golgi apparatus are reduced; catabolism predominates. In S1 more giant lysosomes occur; the total number of lysosomes increases whereas acid phosphatase activity decreases at the same time. Sex differences which exist in untreated animals disappear. Substitution with estradiol causes an activation of the proximal tubule cells: Heterophagy predominates, and cellular autophagy is reduced. Nuclear size is unchanged; ER, ribosomes and Golgi apparatus show a clear increase. Giant lysosomes are absent in S1. On the whole lysosomes are larger, but less numerous than after castration. Acid phosphatase is highly active. All changes are most evident in S3. At the end of pregnancy the proximal tubule cells are stressed considerably: Pinocytotic activity increases, and large numbers of cell organelles and many lipid vacuoles can be observed. The basal lamina in S1 and S2 becomes thicker. Lysosomes enlarge and increase in number in all segments; giant lysosomes are absent in S1; acid phosphatase activity is extremely high. Twenty early pregnancies were diagnosed and monitored in vervet monkeys by ultrasonography. Non-gravid uteri became increasingly echogenic from cycle days 7 to 26. The first definite sign of pregnancy was a gestational cavity of 2 mm (+/- 0.80) at 33.0 (+/- 1.48) days menstrual age, which was also used to date all subsequent features. Earlier signs, such as an endometrial line swelling or endometrial 'pregnancy' ring, as reported for other non-human primate species, could not be reliably and consistently used to diagnose pregnancy in vervet monkeys. A rapid increase of the gestational cavity size from days 37 to 49 corresponded closely to a rapid increase in plasma progesterone concentration from day 39 to 49. The first yolk sac was recognizable at 38.0 days (+/- 3.10) and measured 3.3 mm (+/- 0.40) in diameter. A heart beat could be detected at 45.5 (+/- 1.73) days and the size of the first measurable embryo at 35 days was 2 mm. The dating of most features was within the range reported for other non-human primate species.  相似文献

This study aimed to investigate the effect of the menstrual cycle on strength, functioning of the GH/IGF-1 axis and collagen metabolism in physically active women. Twenty-four physically active and eumenorrheic women volunteered to participate in the study (body mass 60.3 ± 9.18 kg, age 21.8 ± 0.92 years). Blood samples were obtained between the 5th and 8th days (the follicular phase) and between the 19th and 22th days (the luteal phase) of the menstrual cycle to determine sex steroid concentrations (follicle-stimulating hormone (FSH), luteinizing hormone (LH), prolactin (PRL), oestradiol (E2) and progesterone (P)). Also insulin-like growth factor 1 (IGF-1) and collagen metabolism markers (synthesis (PICP) and breakdown (ICTP)) and maximum voluntary contraction (MVC) were measured. MVC was higher in the luteal phase 164.1 ± 34.77 [N m] (F(1.23) = 4.59; p = 0.043). The recorded collagen synthesis marker (PICP = 296.4 ± 35.61 [ng/ml]) was at the upper level of the reference range (30–300), with an insignificant decrease in the luteal phase (Z = 1.612; p = 0.107) and a significant increase in oestradiol concentration (Z = 4.286; p = 0.0001). The marker of collagen breakdown (ICTP = 4.16 ± 0.68 [μg/l]) was reduced by 6.8% in the same phase (Z = 1.764; p = 0.137). The variability of physical abilities (MVC) during the menstrual cycle showed that menstrual status should be taken into account in determination of the training loads. Increasing the load in the luteal phase seems to be favoured by a beneficial change in collagen metabolism (lower synthesis decrease, lower breakdown increase) observed in physically active women.  相似文献

The course of foveal dark adaptation was studied as a function of the intensity and duration of preexposure. Four intensities (11,300, 5,650, 1,130, and 565 mL.) and four durations (300, 150, 30, and 15 seconds) were used in all combinations of intensity and duration. The threshold-measuring instrument was a monocular Hecht-Shlaer adaptometer and the threshold measurements were recorded in log micromicrolamberts. There were two subjects and each went through the complete series of intensities and durations five times. The five logarithmic values obtained for each threshold were converted into a geometric mean and these means were the data used in the analysis of the results. The chief results were as follows:— 1. For each subject the final steady threshold value was in the region of 7.0 log µµL. 2. As the intensity, or duration, or both, were increased the initial foveal dark adaptation threshold rose, the slope of the curve decreased, and the time to reach a final steady threshold value increased. 3. For those values of preexposure intensity and time for which the product, I x t, is a constant it was found that for the two higher intensities and two longer durations and also for the two lower intensities and two shorter durations, the dark adaptation curves were the same. For other values of I x t = C the curves were generally not the same.  相似文献
