To study dietary pH effects on Lymantria dispar asiatica larvae and provide a theoretical basis for its control in different forests, phosphate buffers (PBs) of pH 6, 7, and 8 were used to prepare experimental diets. The diet prepared with pH 6 PB was named as DPB6, with pH 8 PB as DPB8, and with pH 7 PB as DPB7 (control). The dietary pH was 5.00 in DPB6, 6.05 in control, and 6.50 in DPB8. After feeding on the diets with different pH values for 84 hr, fourth-instar caterpillars were randomly collected. Growth and various physiological traits were determined and 16S recombinant DNA sequencing was performed using the intestinal microflora of surviving larvae. Results showed that the mortality was 30% in DPB6, and 10% in DPB8, while no mortality was observed in control. The partial least squares discriminant analyses suggested that diets prepared with PB of different pH resulted in different food intake, amount of produced feces, weight gain, digestive enzyme activities, and antioxidant enzyme activities in larvae. Interestingly, both the highest weight gain and the lowest total antioxidant capacities were seen in control larvae. Results also showed that the larval gut microbiota community structure was significantly affected by dietary pH. Moreover, linear discriminant analysis effect size suggested that the family Acetobacteraceae in control, genus Prevotella in DPB8, and genus Lactococcus, family Flavobacteriaceae, family Mitochondria, and family Burkholderiaceae in DPB6 contributed to the diversity of the larval gut microbial community.  相似文献

In the present study, diel pattern in gut microbial communities in insects were evaluated. Lymantria dispar asiatica fourth instar larvae (72 ± 2 hr after molting) at noon (LdD) and midnight (LdN) were used for a comparative analysis of the gut microbial community. Ten bacterial operational taxonomic units (OTUs) were shared between LdD and LdN samples. One bacterial OTU was specific to LdD. The dominant gut microbes were OTU72 in LdD and OTU75 in LdN. A linear discriminant analysis effect size cladogram suggested that ten bacterial OTUs maintain significant differences in relative abundances between LdD and LdN. These results agreed with the discrete ellipses between LdD and LdN in principal coordinates analysis plots. Additionally, using phylogenetic investigation of communities by reconstruction of unobserved states, the gut microbial community was assigned to 23 functional terms, among which 22 exhibited significant differences between LdD and LdN. To conclude, the present study documented a diel pattern in the gut microbial community of L. dispar asiatica larvae.  相似文献

The effect of interspecific competition between the solitary endoparasitoid Glyptapanteles porthetriae Muesebeck (Hymenoptera: Braconidae) and the gregarious Glyptapanteles liparidis Bouché (Hymenoptera: Braconidae), was investigated in larvae of Lymantria dispar L. (Lepidoptera: Lymantriidae). Host larvae were parasitized by both wasp species simultaneously in premolt to the 2nd or the 3rd host instar or in an additional approach with a 4‐day delay in parasitization by the second wasp species. Host acceptance experiments revealed that both wasp species do not discriminate between unparasitized host larvae and larvae parasitized previously by the same or the other species. In more than 90% female wasps parasitized the larva they encountered first. During the period of endoparasitic development, larvae of the competing parasitoid species never attacked the egg stage of the other species. When host larvae were parasitized simultaneously by both wasp species, the rate of successful development of both species depended on the age of the host larva at the time of its parasitization; G. liparidis emerged successfully from 44% of host larvae parasitized during the premolt to 2nd instar, G. porthetriae from 28%, and in 20% of the hosts both parasitoid species were able to develop in one gypsy moth larva. However, when host larvae were parasitized simultaneously during premolt to the 3rd instar, G. liparidis was successful in 90% of the hosts, compared to 8% from which only G. porthetriae emerged. In the experiments with delayed oviposition, generally the species that oviposited first succeeded in completing its larval development. Larvae of the species ovipositing with four days delay were frequently attacked and killed by larvae of the first parasitizing species or suffered reduced growth. As the secondary parasitoid species, G. porthetriae‐larvae were never able to complete their development, whereas G. liparidis developed successfully in at least 12,5% of the multiparasitized host larvae. Thus, multiparasitism of gypsy moth larvae by both Glyptapanteles species corresponds to the contest type; however, G. porthetriae is only able to develop successfully as the primary parasitoid of young host larvae.  相似文献

Lymantria dispar (L.) (Lepidoptera: Lymantriidae) larvae can be infected in the laboratory with a variety of entomopathogenic microsporidia. In many cases, however, L. dispar is only a semi‐permissive host for such infections. In this study, we analyzed changes in the melanization of hemolymph and hemocyte numbers in L. dispar larvae after inoculation with various entomopathogenic microsporidia. We compared the infections produced by microsporidia isolated from L. dispar and infections produced by isolates from other Lepidoptera to which L. dispar is only a semi‐permissive host. Microsporidiosis induced a significant activation of the prophenoloxidase system leading to melanization; activation was highest when the pathogen caused heavy infections of the fat body, which was the case with two microsporidia originally isolated from L. dispar. Infection of only the silk glands or light infection of the fat body by two Vairimorpha spp. from other lepidopteran hosts elicited a lower response. Very light infections caused by a microsporidium isolated from Malacosoma americanum were not accompanied by elevated hemolymph melanization activity. Heavy infections by Endoreticulatus spec. that remained restricted to the gut tissue likewise did not elicit melanization. One Vairimorpha spec. from L. dispar induced a significant increase in total hemocyte numbers; the other infections led to temporarily decreased numbers. Microscopic examinations showed that parts of infected tissue were encapsulated by hemocytes. We conclude that measured alterations in hemolymph melanization and hemocyte numbers were likely to be induced by the damaging effects of heavy infections. Observed defense responses did not prevent the progression of infections.  相似文献

We quantified horizontal transmission of three microsporidian pathogens, Endoreticulatus schubergi, Nosema lymantriae and Vairimorpha disparis that infect Lymantria dispar larvae in an experiment using caged, potted oak plants. Despite marked differences in the modes of spore release from infectious hosts, no significant differences in the transmission success to uninfected, susceptible test hosts were ascertained between the tested microsporidian species. The density of initially inoculated larvae and the exposure period, on the other hand, did influence the number of infected test larvae. Depending on the density of inoculated larvae (10%, 30% or 50%), between 0% and 26% of the test larvae became infected with one of the three tested microsporidian pathogens after an exposure period of 6 days. When the exposure period was 12 days, between 11% and 76% of the test larvae became infected.  相似文献

Infection of Lymantria dispar host larvae by the entomopathogenic microsporidium Vairimorpha sp. has a negative impact on the performance of the endoparasitic braconid Glyptapanteles liparidis. To investigate possible causes for this effect, we studied to what extent nutritional host suitability is altered by the microsporidium. Therefore, we analyzed carbohydrates and fatty acids in host larvae after Vairimorpha infection and/or parasitism by G. liparidis. Trehalose levels were significantly reduced in the hemolymph of infected hosts. After day five post infection, it was detected only in traces. Four to six days later, the glycogen resources were depleted in infected larvae. Parasitism by G. liparidis, on the other hand, led to increased hemolymph trehalose levels during the early endoparasitic phase but to a significant decrease at the end of its larval development. No effect of parasitism on the glycogen content was ascertained. Hemolymph levels of the fatty acids analyzed, such as palmitic, stearic, oleic, linoleic, and linolenic acid, were significantly reduced in microsporidia-infected L. dispar. Vairimorpha sp. develops as an intracellular parasite in the fat body of the host larva and synthesis of trehalose and fatty acids may be disturbed. Moreover, microsporidia may also harness metabolites or energy produced by host cells. We conclude that the microsporidia-induced decrease in hemolymph carbohydrates and fatty acids adversely affects growth and development of parasitoid larvae.  相似文献

Multiple‐choice assays were conducted in the laboratory to examine the effect of CO2‐induced changes in leaf quality on growth, nutritional indices and preferences of the gypsy moth (Lymantria dispar) larvae. The tested tree species, one‐year‐old aspen (Populus pseudo‐simonii Kitag.), two‐year‐old birch (Betula platyphylla) and three‐year‐old oak (Quercus mongolica Fisch.) were transplanted to open‐top chambers at ambient or elevated CO2 (650 ppm) concentrations in May 2005. The present study was conducted in 2006. Leaves from the upper and lower crowns of each tree species were sampled and analysed, respectively. On both measurement dates (24 July 2006 and 16 August 2006), there were significant CO2 effects on levels of condensed tannin, starch, nitrogen and C : N ratio. But only on the latter date, were the concentrations of total phenolics, soluble sugar, carbon and total non‐structural carbohydrates significantly affected by elevated CO2. Leaf dry weight content and specific leaf weight were almost completely unaffected by CO2 enrichment. Gypsy moth larvae exhibited a clear selectivity for tested leaf types (leaves from the upper and lower crowns of each tree species) even in their early instar stage, with the upper leaves of P. pseudo‐simonii being the most preferable and the lower leaves of Q. mongolica being the least preferred. The changes in leaf quality significantly reduced larval growth and altered partial indices of insect performance (e.g., relative growth rate and relative consumption rate). However, at least in this short‐term choice feeding assay (13 days), the CO2‐induced changes in leaf quality had no significant effects on food preference behaviour of the gypsy moth larvae, neither within the limited range of host plants nor within the leaves at different canopy heights of the same tree species.  相似文献

为了探明20-羟基蜕皮甾酮对昆虫蜕皮过程中体壁的表皮层、 皮细胞及其细胞器的具体影响过程, 本研究利用透射电镜技术研究了20-羟基蜕皮甾酮对舞毒蛾Lymantria dispar (Linnaeus)5龄幼虫体壁超微结构的变化。结果表明, 用高浓度20-羟基蜕皮甾酮溶液浸过的白桦叶片饲喂幼虫, 处理6 h, 摄入约400 μg 20-羟基蜕皮甾酮后, 幼虫停止取食; 处理12 h时表皮细胞顶膜上的微绒毛减少, 在皮细胞与旧表皮之间形成蜕皮间隙, 旧头壳从幼虫头部脱离; 处理24 h时蜕皮间隙继续增大, 旧表皮与皮细胞进一步分离, 新表皮质层开始形成; 处理36 h时皮细胞顶膜形成较短的微绒毛, 胞质区域出现数量较多的电子疏松泡, 新表皮由上表皮、 外表皮及8层左右内表皮片层组成; 处理48 h时顶膜与内表皮界限模糊, 内表皮继续合成至16层左右; 72 h时细胞内出现大面积电子疏松泡, 内表皮合成至20层左右。 处理96 h时, 与对照组相比, 皮细胞细胞器较少, 核仁周围出现小部分空白区域, 胞质区域内含物减少; 虫体发黑缩小, 即将死亡; 内表皮层数仍旧保持20层左右。对照组幼虫6-96 h虫体活跃, 正常取食, 外部观察及透射电镜结果均未显现蜕皮现象; 表皮层由上表皮、 外表皮及内表皮组成; 皮细胞顶膜微绒毛密度高; 表皮细胞分泌活动旺盛, 胞质区域细胞界限明显, 内含物丰富; 细胞器典型而且活跃; 内表皮片层随时间不断增加至50层左右。结果提示, 外源20-羟基蜕皮甾酮能够导致舞毒蛾5龄幼虫的致死性蜕皮。  相似文献

Abstract. . The independent and interactive effects of temperature and dietary nitrogen content on performance of the gypsy moth (Lymantria dispar L.) were examined. In long-term feeding trials, larvae were reared from egg hatch to pupation on low (1.5%) and high (3.7% dry weight) nitrogen diets, under three temperature regimes. Short-term feeding trials with fourth instars and the same treatments were conducted in order to calculate nutritional indices.
Higher temperatures did not influence larval survival and marginally increased final pupal weights, but strongly decreased long-term development rates. They also accelerated short-term growth and consumption rates, and tended to improve food processing efficiencies. High concentrations of dietary nitrogen increased survival rates and final pupal weights markedly, but decreased long-term development rates only marginally. A high content of dietary nitrogen also accelerated short-term development and growth rates, reduced consumption rates, and improved food digestibility. Insects responded to low nitrogen-content diets primarily by eating faster, rather than by altering efficiency of nitrogen use. In the short-term feeding trials, thermal regime and dietary nitrogen interacted to influence growth rates, overall food processing efficiencies and nitrogen consumption rates. No interactive effects were observed in long-term studies.
This research demonstrates that small changes in thermal regime and ecologically relevant variation in dietary nitrogen content can strongly affect gypsy moth performance. Moreover, various performance parameters are differentially sensitive to the direct and interactive effects of temperature and diet.  相似文献

Carvacrol is a terpene compound with various biological activities. However, few studies have specifically focused on its insecticidal activity and mechanism of carvacrol. The larvae of Lymantria dispar are seriously harmful herbivorous insect. This study measured the antifeedant, growth-inhibitory, and toxic effects of carvacrol on L. dispar larvae. To further clarify the insecticidal mechanism of carvacrol, the effects of carvacrol on detoxifying enzymes, antioxidative enzymes, digestive enzyme activities, and the mRNA expression of the above-mentioned enzyme genes were investigated. The results of the study showed that the median lethal concentration (LC50) and the sublethal concentration (LC20) of carvacrol were 1.120 mg/mL and 0.297 mg/mL, respectively, at 72 h. After LC20 treatment of L. dispar larvae for 72 h, food intake and weight gain were significantly lower compared with the control. Enzyme activity assays showed that carvacrol significantly inhibited the activities of carboxylesterase (CarE), glutathione S-transferase (GST), and acetylcholinesterase (AchE), and the inhibition rate of AchE activity was highest (66.51%). Carvacrol also activated the activities of superoxide dismutase (SOD) and catalase (CAT), while it inhibited the activities of lipase (LIP) and amylase (AMS), and first inhibited and then activated protease. In addition, qRT-PCR tests showed that carvacrol affected the mRNA expression levels of CarE, GST, AchE, SOD, CAT, LIP, AMS, and protease. This study helps to clarify the insecticidal mechanism of carvacrol on L. dispar larvae.  相似文献

ABSTRACT. Surgical removal of the brain or disconnection of the last abdominal ganglion from the ventral nerve cord prevented sex pheromone release in female Lymantria dispar (L.) (Lymantriidae), as assayed by the male wing-fanning response. The calling behaviour continued to occur in individuals whose terminal abdominal ganglion had been thus isolated, however, indicating that the neural mechanisms controlling calling function independently in the last abdominal ganglion.  相似文献

Lepidopterans are known to have different pheromone‐binding proteins with differential expression patterns that facilitate specific signal transduction of semiochemicals. Two PBPs of the Asian gypsy moth, Lymantria dispar, were reported to express in both females and males, but their physiological functions were unknown. Results showed that LdisPBP1 and LdisPBP2 were expressed in the sensilla trichodea of males and the s. trichodea and s. basiconica of females. When LdisPBP1 gene was targeted by RNA interference (RNAi) in males, the expression of LdisPBP1 and LdisPBP2 decreased by 69 and 76%, respectively, and when LdisPBP2 gene was targeted by RNAi, they decreased by 60 and 42%, respectively. In females, after treatment with LdisPBP1 dsRNA, LdisPBP1 and LdisPBP2 levels were reduced by 26 and 69%, respectively, and LdisPBP2 dsRNA reduced the relative expression of them by 4 and 62%, respectively. The expression of LdisPBP1 and LdisPBP2 was interdependent. Electroantennogram (EAG) recordings showed that LdisPBPs participate in the recognition of the sex pheromone in males, and the sex pheromone and plant volatiles in females. The function of LdisPBPs represents the sex‐specific roles.  相似文献

【目的】为了探究滞育激素经昆虫取食的生物效应, 需设计一种能够加速跨膜运输的融合蛋白, 克服经昆虫消化道的降解作用。【方法】本研究以分月扇舟蛾Clostera anastomosis滞育激素基因Cloan-DH和TAT穿膜肽编码序列为基础, 构建TAT-Cloan-DH融合基因; 然后以增强型绿色荧光蛋白(enhanced green fluorescent protein, EGFP)为标签, 构建在pET-22b载体中。在0.2 mmol/L IPTG诱导下, 在大肠杆菌Escherichia coli BL21 Rosetta(DE3)中表达出TAT-Cloan-DH-EGFP融合蛋白。 再分别将表达有TAT-Cloan-DH-EGFP和EGFP融合蛋白的大肠杆菌BL21菌体拌入人工饲料,经舞毒蛾Lymantria dispar幼虫持续取食0, 8, 24, 48, 72和90 h后, 随机挑选试虫进行组织固定并制作石蜡切片, 选取中肠所在体段切片进行组织荧光分析。【结果】37℃ 0.2 mmol/L IPTG诱导条件下, 表达的TAT-Cloan-DH-EGFP融合蛋白的分子量为61 kD, 主要以包涵体形式表达, 融合蛋白约占细胞总蛋白含量的18.1%; 随着试虫取食含TAT-Cloan-DH-EGFP融合蛋白饲料的增加, 虫体组织的绿色荧光强度逐渐增加, 取食72 h后, 组织荧光强度达到最高。【结论】实验结果说明融合蛋白TAT-Cloan-DH-EGFP可以通过消化道横向跨膜运输, 到达虫体其他组织, 且该过程与蛋白的摄入量成比例。  相似文献

Abstract. The effects of pheromone concentration and ambient temperature on male gypsy moth, Lymantria dispar (L.) (Lepidoptera), flight responses to pheromone were investigated in a wind tunnel. As the pheromone dose increased from 10 ng to 1000 ng, males flew at progressively slower airspeeds and ground speeds, and reduced their wingbeat frequencies. Furthermore, the moths steered significantly smaller course angles as the pheromone concentration increased, indicating that they were adopting a more upwind heading. The overall width of the flight tracks also decreased when males flew in more concentrated pheromone plumes. Estimation of plume dimensions using a male wing-fanning assay showed that as pheromone dosage increased, the resultant active spaces became wider, indicating that an inverse relationship existed between the dimensions of the time-averaged plume and the width of track reversals and that most turns were initiated within the plume. When males were flown at cool (20°C) and warm (26°C) ambient temperatures but to equivalent pheromone emission rates, they exhibited higher airspeeds and ground speeds at the higher temperature but steered larger course angles. Track widths, and length of track legs were, however, similar at the two temperatures. The mean turning frequency was nearly the same (c. 4 turns/s) across all the concentrations and temperatures tested even though the moths' thoracic temperature differed by 5°C when the ambient temperature was varied.  相似文献

1. This study investigated how phytochemical variation among clones of quaking aspen Populus tremuloides, growing in a common habitat, affects the growth and fecundity of a model herbivore. 2. Gypsy moth Lymantria dispar larvae were reared from egg hatch to pupation on 10 aspen clones in the field or on excised foliage in the laboratory. Foliage was collected from each clone, and concentrations of phenolic glycosides, condensed tannins, nitrogen, and water were determined. 3. Herbivore fitness parameters and aspen phytochemical concentrations varied significantly among clones. In both the field and laboratory, larvae reared on clones containing high concentrations of phenolic glycosides exhibited prolonged developmental times and reduced pupal weights and fecundity. Herbivore performance parameters were also related positively to foliar nitrogen concentrations in the laboratory. Food consumption, but neither growth nor reproductive parameters, were related positively to condensed tannin concentrations. 4. In this study, foliar concentrations of phenolic glycosides were implicated as a significant determinant of food quality for gypsy moths, consistent with results of previous laboratory experiments. Additionally, this study documents a case in which host plant variation at a local level influences the performance and possibly the distribution and abundance of an important herbivore.  相似文献

Two distinctly different patterns of gut enzyme activity were noted in relation to diapause in pharate first instar larvae of the gypsy moth, Lymantria dispar. Trypsin, chymotrypsin, elastase, aminopeptidase and esterase activities were low at the initiation of diapause and through the period of chilling needed to terminate diapause. At the completion of a 150 day chilling period, activity of each of these enzymes quickly increased when the pharate larvae were transferred to 25°C. By contrast, activity of alkaline phosphatase (ALP) increased rapidly at the onset of diapause, remained elevated throughout diapause, increased again during postdiapause, and then dropped at the time of hatching. In addition, zymogram patterns of ALP activity differed qualitatively in relation to diapause: several bands were detectable during the pre- and postdiapause periods, but only one band, a band of high mobility, was visible during diapause. The ALP isozyme present in diapausing pharate larvae had a pH optimum of 10.6. Diapause in the gypsy moth can be averted by application of an imidazole derivative, KK-42, and pharate larvae treated with KK-42 showed elevated protease and esterase activity, low ALP activity, and expressed ALP isozymes with low mobility. Thus the overall patterns of gut enzyme activity and the ALP zymogram in KK-42 treated individuals were similar to those observed in untreated individuals at the termination of diapause. 为探讨外源茉莉酸对青杨Populus cathayana的诱导抗性及其对舞毒蛾Lymantria dispar的影响, 室内对青杨扦插苗喷施不同浓度的茉莉酸(对照为0.17%丙酮), 分别在喷施后1, 5和10 d采集叶片分析其保护性酶活性的变化, 并接舞毒蛾幼虫于青杨苗木上观测其长发育情况。结果表明: 0.1 和0.001 mmol/L两种浓度的茉莉酸(JA)处理均使青杨叶过氧化物酶(POD)、 多酚氧化酶(PPO)、 苯丙氨酸解氨酶(PAL)、 胰凝乳蛋白酶抑制剂(CI)和胰蛋白酶抑制剂(TI)活性较对照增加(P<0.05)。取食茉莉酸诱导的青杨苗木后, 舞毒蛾幼虫的发育历期延长, 体重降低。0.1 mmol/L茉莉酸诱导的青杨苗木5 d后接虫, 使舞毒蛾幼虫的发育历期显著延长, 较对照长8 d; 接虫21 d后称重时, 取食茉莉酸诱导的青杨叶片的幼虫体重较对照组降低了50%~100%, 该结果说明外源茉莉酸诱导青杨产生了对舞毒蛾明显的抗虫性。  相似文献

Pheromone‐binding proteins (PBPs) are distributed widely on the antennae of insects, and they are believed to be involved in the process of chemical signal transduction, but their interaction with chemicals is largely unknown. Here, we present our findings on the key amino acid residues of PBPs in the gypsy moth, Lymantria dispar. Potential key residues were screened with the Calculate Mutation Energy program and molecular docking methods. Mutated proteins were obtained by mutating residues to alanine via site‐directed mutagenesis. Circular dichroism (CD) spectroscopy showed that the mutated proteins formed α‐helix, and the stability of protein structure was influenced due to mutations. Fluorescence binding assays were further conducted with the mutated proteins, sex pheromones and analogues. Results showed that to PBP 1, tyrosine at position 41 and phenylalanine at position 76 could be the key amino acid residues influencing the stability of structure; in addition, phenylalanine at 36 and lysine at position 94 could be key amino acid residues interacting with chemicals. To PBP 2, glycine at position 49, phenylalanine at position 76 and lysine at position 121 could be the key amino acid residues in the structural stability. These results shed light on the relationship between the specific amino acids and functions of PBPs in transmitting the chemical signals.  相似文献
