The crystal structure of Clostridium thermocellum endoglucanase CelA in complex with cellopentaose has been determined at 0.94 A resolution. The oligosaccharide occupies six D-glucosyl-binding subsites, three on either side of the scissile glycosidic linkage. The substrate and product of the reaction occupy different positions at the reducing end of the cleft, where an extended array of hydrogen-bonding interactions with water molecules fosters the departure of the leaving group. Severe torsional strain upon the bound substrate forces a distorted boat(2,5) B conformation for the glucosyl residue bound at subsite -1, which facilitates the formation of an oxocarbenium ion intermediate and might favor the breakage of the sugar ring concomitant with catalysis.  相似文献

The phytoplankton lysis rates in the different eutrophic regions of Lake Taihu were determined based on the activities of particle and dissolved esterase, as well as the decay rate of the latter, from August 2009 to March 2010. Two peaks were observed for the chlorophyll a concentration, one in September 2009 and another in January 2010. The highest phytoplankton lysis rates observed in October were associated with the decay of the summer bloom, whereas the minimum lysis rates observed in January were associated with the winter bloom. The highest cell lysis rates in Meiliang Bay, Lake centre, and Gonghu Bay were 0.67, 0.77, and 0.68 d–1, respectively, whereas the lowest lysis rates in these regions were 0.03, 0.01, and 0.05 d–1, respectively. Water temperature showed an apparent indirect effect on lysis rate. In addition, a significant inverse relationship was observed between lysis rates and nitrate concentration in the three lake regions, which suggests that phytoplankton cell lysis is associated with changes in nitrate concentration. The development of lithium–sulfur batteries necessitates a thorough understanding of the lithium‐deposition process. A novel full‐cell configuration comprising an Li2S cathode and a bare copper foil on the anode side is presented here. The absence of excess lithium allows for the realization of a truly lithium‐limited Li–S battery, which operates by reversible plating and stripping of lithium on the hostless‐anode substrate (copper foil). Its performance is closely tied to the efficiency of lithium deposition, generating valuable insights on the role and dynamic behavior of lithium anode. The Li2S full cell shows reasonable capacity retention with a Coulombic efficiency of 96% over 100 cycles, which is a tremendous improvement over that of a similar lithium‐plating‐based full cell with LiFePO4 cathodes. The exceptional robustness of the Li2S system is attributed to an intrinsic stabilization of the lithium‐deposition process, which is mediated by polysulfide intermediates that form protective Li2S and Li2S2 regions on the deposited lithium. Combined with the large improvements in energy density and safety by the elimination of a metallic lithium anode, the stability and electrochemical performance of the lithium‐plating‐based Li2S full cell establish it as an important trajectory for Li–S battery research, focusing on practical realization of reversible lithium anodes.  相似文献

Human exposure to N-nitrosodimethylamine (NDMA) from foods and beverages was modeled and upper-bound cancer risks were predicted for the United States and Canada. Approximately 0.5 (0–10.8) cancer incidents per million population from lifetime exposure to NDMA in drinking water were estimated. Lifetime exposure to NDMA from the major exogenous sources may result in 49.6 (range: 17.7–171.7) cancer incidents per million population, while meat products contribute the most (15.9/million) followed by milk products (10.9/million). Drinking water may contribute approximately 1% to the exogenous cancer risk and holds the 10th position among 10 exogenous sources. The sum of the cancer risks from the major exogenous sources (e.g., 49.6/million) is higher than the permissible limits (1–10/million) of several regulatory agencies. Thus, NDMA in exogenous sources can pose a significant source for cancer risk. Cancer risk from the exogenous sources was estimated to be much lower than that of the NDMA in the endogenous source (<1%).  相似文献

A new water mite species Sperchon (Hispidosperchon) serapae n. sp. is described. The material was collected from a slow-flowing stream during field study on the water mite fauna of the Lakes region in southwestern Turkey. An updated list of hitherto known species of Sperchon Kramer from Turkey, including nomenclatural changes and numerous new locality records to the species, is also given.  相似文献

This study has been conducted to estimate the occurrence of Cryptosporidium oocysts in water supplies in the Metropolitan area of Seoul, South Korea, for 10 years from 2000 to 2009. Water samples were collected quarterly at 6 intakes in the Han River and its largest stream and 6 conventional Water Treatment Plants (WTPs) serving drinking water for 10 million people of Seoul. Cryptosporidium oocysts were found in 22.5% of intake water samples and arithmetic mean was 0.65 oocysts/10 L (range 0-22 oocysts/10 L). Although the annual mean of oocyst number was as low as 0.04-1.90 oocysts/10 L, 3 peaks in 2004 and 2007 were observed and the pollution level was a little higher in winter. The lowest density was observed at Paldang intake and the pollution level increased at Kuui and Jayang intakes. At the end of the largest stream, oocysts were found in 70% of collected samples (mean 5.71 oocysts/10 L) and it seemed that its joining the Han River resulted in the increase at Kuui intake and downstream. Oocyst removal by physical process exceeded 2.0-2.3 log and then all finished water samples collected at 6 WTPs were negative for Cryptosporidium in each 100 L sample for 10 years. These results suggested that domestic wastewater from the urban region could be a source of Cryptosporidium pollution and separating sewage systems adjacent to the intakes could be meaningful for some intakes having weakness related to parasitological water quality.  相似文献

The ribosome inactivating protein PD-L4 from Phytolacca dioica is a N-beta-glycosidase, probably involved in plant defence. The crystal structures of wild type PD-L4 and of the S211A PD-L4 mutant with significantly decreased catalytic activity were determined at atomic resolution. To determine the structural determinants for the reduced activity of S211A PD-L4, both forms have also been co-crystallized with adenine, the major product of PD-L4 catalytic reaction. In the structure of the S211A mutant, the cavity formed by the lack of the Ser hydroxyl group is filled by a water molecule; the insertion of this non-isosteric group leads to small albeit concerted changes in the tightly packed active site of the enzyme. These changes have been correlated to the different activity of the mutant enzyme. This work highlights the importance of atomic resolution studies for the deep understanding of enzymatic properties.  相似文献

Cell lines can be useful experimental tools for studying marine fish, which are often difficult to routinely obtain and maintain in the laboratory. As few cell lines are available from coldwater marine fish, cultures were initiated from late gastrula embryos of haddock (Melanogrammus aeglefinus) in Leibovitz's L-15 with fetal bovine serum (FBS). From one culture, a cell line (HEW) emerged that has been grown for close to 100 population doublings, was heteroploid, and expressed telomerase activity, all of which suggest HEW is immortal. Growth occurred only if FBS was present and was optimal at 12 to 18°C. Usually most cells had an epithelial-like morphology, but under some conditions, cells drew up into round central bodies from which radiated cytoplasmic extensions with multiple branches. These neural-like cells appeared within a few hours of cultures being placed at 28°C or being switch to a simple salt solution (SSS). At 28°C, cells died within 24 h. In SSS, HEW cells survived as a monolayer for at least 7 days. The sensitivity of HEW cells to morphological change and their capacity to withstand starvation should make them useful for investigating cellular responses to environmental stresses.  相似文献

Water convolvulus, a vegetable, absorbed bisphenol A (BPA), an endocrine disruptor, from the medium. One week later, no BPA could be detected in the plant, indicating that BPA had been metabolized in the plant. BPA monoglucoside was detected as the BPA base at ca. 10% in the roots, some in the stems, but none in the leaves. 2H-NMR analyses of MeOH extracts and hydrolyzates of the plant treated with BPA-d16 showed the presence of metabolites (ca. 7% and 26%, respectively, as BPA equivalents) other than the glucoside. Over 50% of BPA might be polymerized and/or tightly bound in the plant residues.  相似文献

The central part of the tin ore deposit Ehrenfriedersdorf/Erzgebirge, which was exploited from the 13th century to 1990, was flooded from 1994 to 1996. Since that time mine waters have flown through the gallery “Tiefer Sauberger Stolln” to the creek Wilisch in the Elbe river catchment area. The water at the mine portal shows high concentrations of arsenic and heavy metals. The average arsenic concentration is about 0.5 mg/L. Approximately two thirds of arsenic are transported dissolved. Where the mine water ascends from deeper levels, arsenic concentrations of about 0.4 mg/L were found. Here arsenic occurs predominantly particular. The mining gallery “Tiefer Sauberger Stolln” provides the unique opportunity of subsurface sampling for the identification of the arsenic sources under different hydrological conditions (normal and high water level). The sources of dissolved arsenic in the gallery part between the raise and the portal were determined and analyzed. Between these two monitoring points, many inflows of infiltration water were detected. The concentration of As in the infiltration water reaches up to 1.8 mg/L, which varies depending on the location in the gallery and the hydrological situation. The first part of the gallery was straightened, heightened and partly concreted with modern mining technique. The arsenic concentrations can decrease owing to high precipitation rates and snow melt events. The last part of the gallery was preserved due to low coverage. Here the arsenic concentrations in the infiltration waters increase with the surface water inflow. At a normal water level, 1 kg arsenic per day leaves the raise and 2.1 kg the gallery portal, which means that 50 % of the arsenic load comes from the infiltration water. At a high water level, 2.5 kg arsenic per day are transported through the raise and 8.2 kg per day through the gallery portal, which means that about 70 % of the arsenic load comes from infiltration water. The area of Ehrenfriedersdorf is characterized by a superposition of anthropogenic soil pollution over the geogenic inventory. There is a close connection between ancient soil contaminations by high amounts of water‐soluble arsenic compounds, e.g. arsenic trioxide formed by roasting the ores during ancient tin smelting, and high concentrations of dissolved arsenic in the infiltration water. The contamination of surface water and river sediments by arsenic is originating from an anthropogenic pollution of soils by ancient tailings via infiltration of water rich in arsenic into the mine gallery.  相似文献

Abstract.  Little attention has been given to the effect of physiological limitations on the ability of introduced species to invade ecological communities. In the present study, the water balance of the invasive Argentine ant ( Linepithema humile ) is compared with five common ant species native to southern California. Total and critical water contents are measured, as well as rates of water loss and survival times. The results show that the body size of ant species has a strong effect on water balance, explaining between 99% of total and 90% of critical water content (i.e. the amount of water remaining when the ant becomes moribund). The measured survival times at 40 °C and approximately 0% relative humidity are shorter for L. humile than for the other five native ant species. The area-independent water-loss rate is higher for L. humile than for the other native ant species studied. This finding is corroborated by independent data using two different methods (i.e. gravimetric measurement and open-flow respirometry). Somewhat surprisingly, significant differences in the water-loss rates of different sizes of Solenopsis xyloni workers are found, even after adjusting for surface area. The area-independent water-loss rate is significantly lower for larger than for smaller workers. Consistent with circumstantial evidence from the field, the results suggest that the physiological constraints of invasive species, such as water-loss rates and critical water content, can limit their distribution and abundance not only at regional scales, but also at fine scales.  相似文献

Haliplus alberti sp. n. is described, based on three specimens collected in Pan de Guajaibon, San Juan de Sagüa, Pinar del Río, Cuba. Morphologically, the new species is similar to H. pseudofasciatus Wallis, 1933 and H. mutchleri Wallis, 1933, both from Florida (USA). The beetles were collected in a permanent lagoon with abundant aquatic vegetation, muddy bottom and macrophytes near the banks. Seven species of Haliplus are now known from Cuba.

Deer serve as reservoirs of tick-borne pathogens that impact on medical and veterinary health worldwide. In the Republic of Korea, the population of Korean water deer (KWD, Hydropotes inermis argyropus) has greatly increased from 1982 to 2011, in part, as a result of reforestation programs established following the Korean War when much of the land was barren of trees. Eighty seven Haemaphysalis flava, 228 Haemaphysalis longicornis, 8 Ixodes nipponensis, and 40 Ixodes persulcatus (21 larvae, 114 nymphs, and 228 adults) were collected from 27 out of 70 KWD. A total of 89/363 ticks (266 pools, 24.5% minimum infection rate) and 5 (1.4%) fed ticks were positive for Anaplasma phagocytophilum using nested PCR targeting the 16S rRNA and groEL genes, respectively. The 16S rRNA gene fragment sequences of 88/89 (98.9%) of positive samples for A. phagocytophilum corresponded to previously described gene sequences from KWD spleen tissues. The 16S rRNA gene fragment sequences of 20/363 (5.5%) of the ticks were positive for A. bovis and were identical to previously reported sequences. Using the ITS specific nested PCR, 11/363 (3.0%) of the ticks were positive for Bartonella spp. This is the first report of Anaplasma and Bartonella spp. detected in ticks collected from KWD, suggesting that ticks are vectors of Anaplasma and Bartonella spp. between reservoir hosts in natural surroundings.  相似文献
