SUMMARY 1. Unialgal cultures of three species common in the freshwater phytoplankton were used to test limitation of specific growth rate and final yield in defined media of low K+ concentration (range <0.3–6 μmol L−1 or mmol m−3).
2. Growth rate of the diatom Asterionella formosa was independent of K+ concentration above 0.7 μmol L−1. Final yield was dependent on initial concentration when accompanied by K+ depletion below this concentration, but not by lesser depletion with more residual K+. Analyses of particulate K in the biomass indicated a mean final cell content of 2.8 μmol K 10−8 cells, approximately 1.0% of the organic dry weight.
3. Less detailed work with the diatom Diatoma elongatum showed no dependence of growth rate or final yield upon the initial K+ concentration in the range 0.8–3.2 μmol L−1. The phytoflagellate Plagioselmis nannoplanctica suffered net mortality in the lowest concentration tested, 0.8 μmol L−1.
4. Comparison with the range of K+ concentration in natural fresh waters, including a depletion induced by an aquatic macrophyte, suggests that K+ is unlikely to limit growth of phytoplankton. Nevertheless, there can be correlation of K+ with lake trophy.  相似文献

Yeasts from high-altitude lakes: influence of UV radiation   总被引:1,自引:0,他引:1  
Mountain lakes located at a high elevation are typically exposed to high UV radiation (UVR). Little is known about the ecology and diversity of yeasts inhabiting these extreme environments. We studied yeast occurrence (with special emphasis on those producing carotenoid pigments) at five high-altitude (>1400 m a.s.l.) water bodies located in the Nahuel Huapi National Park (Bariloche, Argentina). Isolates were identified using a polyphasic approach. Production of photoprotective compounds (carotenoids and mycosporines) by yeast isolates, and UVB resistance of selected species were studied. All water samples contained viable yeast cells in variable numbers, generally ranging from 49 to 209 cells L−1. A total of 24 yeast species was found; at least four represented novel species. Carotenogenic yeasts prevailed in lakes with low water conductivity and higher transparency and chlorophyll a levels. Apparently, the ability to produce photoprotective compounds in yeasts was related to the transparency of mountain lake waters, and strains from more transparent waters developed increased UVB resistance. Our results indicate that UVR is an important environmental factor affecting the yeast community structure in aquatic habitats.  相似文献

This study reports for the first time the presence of Anabaenopsis arnoldi blooms in Saudi freshwaters. This species has been investigated with high cell densities (3.8 × 103–264 × 103 cells mL−1) during June–November 2007 in Tendaha Lake, one of the major freshwater sources in Saudi Arabia. High temperature and conductivity, and a high concentration of phosphate, and low nitrate concentrations may have contributed to the formation of these blooms. The blooms were found to produce microcystins (MCYSTs) at concentrations up to 364 μg g−1 dry weight as detected by an enzyme-linked immunosorbent assay. MCYSTs were also detected in the raw and treated water of the lake at concentrations (1.6–8.3 and 0.33–1.6 μg L−1, respectively) exceeding the World Health Organization guideline level of 1 μg L−1 for these toxins. HPLC analysis revealed that the extracts of A. arnoldi blooms contained MCYST-RR, -YR and two unidentified MCYSTs, but a pure culture of A. arnoldi isolated from Tendaha Lake during the present study produced MCYST-RR and –YR only. This is the first study to report MCYST production by A. arnoldi . Therefore, this cyanobacterium should be taken into consideration during monitoring of toxic cyanobacterial blooms in drinking and recreational water sources in the world, particularly arid and semi-arid countries including Saudi Arabia.  相似文献

Stoichiometric constraints within ecological interactions and their ecosystem consequences may depend on characteristics of the abiotic environment such as background nutrient levels. We assessed whether consumer identity, via differing body stoichiometry, could regulate periphyton stoichiometry across nutrient regimes in open systems. In 60 flow-through artificial streams, we factorially crossed dissolved inorganic nitrogen levels (elevated = 294  μ g L−1, ambient = 26  μ g L−1) with dissolved inorganic phosphorus levels (DIP: elevated = 15  μ g L−1, ambient = 3  μ g L−1) and consumer type [crayfish (body N : P = 18), snails (body N : P = 28) or a control]. At ambient DIP, periphyton in the crayfish treatment had a lower %P and a lower C : P than periphyton in the snail treatment suggesting that consumer identity, probably mediated by differing P-excretion, regulated periphyton P content. At high DIP, consumer identity no longer affected periphyton elemental composition. Therefore, the stoichiometry of consumer-driven nutrient recycling and consumer identity may be less important to ecosystem functioning in environments with elevated nutrient levels.  相似文献

1. Although large cladocerans are usually uncommon in large rivers, Daphnia lumholtzi (an exotic species in North America) is widespread and occasionally abundant. We collected zooplankton on the Illinois River (Illinois, U.S.A.) in 1995 and 1996 and found that the peak density of D. lumholtzi (25 L−1) typically exceeded that of all native species combined. Maximum density occurred during warm periods (up to 27 °C) when concentrations of inorganic suspended sediments were high (>50 mg L−1).
2. Using a life table experiment, we examined the effects of variation in suspended sediment (0 and 80 mg L−1) and food (104 and 105 Ankistrodesmus cells mL−1) on fitness of D. lumholtzi and the native Daphnia parvula. Daphnia lumholtzi had greater survivorship than D. parvula in most treatments and higher life-time fertility than D. parvula in all treatments. Both species achieved their fastest intrinsic rates of growth in treatments with high food, but their responses to suspended solids differed. The growth rate of D. lumholtzi in high food was slightly increased by higher turbidity, whereas that of D. parvula was depressed.
3. Results suggest that the ability of D. lumholtzi to tolerate suspended solids is an important factor contributing to its success in invading North American rivers.  相似文献

Aims:  To evaluate the effectiveness of organic acids and supercritical carbon dioxide (SC-CO2) treatments as well as their combined effect for the reduction of nonpathogenic Escherichia coli and three pathogenic bacteria in fresh pork.
Methods and Results:  The different treatment conditions were as follows: (i) treatment with acetic (1%, 2% or 3%) or lactic acid (1%, 2% or 3%) only, (ii) treatment with SC-CO2 at 12 MPa and 35°C for 30 min only and (iii) treatment with 3% acetic or lactic acid followed by treatment with SC-CO2. Within the same organic acid concentration, the lactic and acetic acid treatments had similar reductions. For the combined treatment of lactic acid and SC-CO2, micro-organism levels were maximally reduced, ranging from 2·10 to 2·60 log CFU cm−2 ( E. coli , 2·58 log CFU cm−2; Listeria monocytogenes , 2·60 log CFU cm−2; Salmonella typhimurium , 2·33 log CFU cm−2; E. coli O157:H7, 2·10 log CFU cm−2).
Conclusions:  The results of this study indicate that the combined treatments of SC-CO2 and organic acids were more effective at destroying foodborne pathogens than the treatments of SC-CO2 or organic acids alone.
Significance and Impact of the Study:  The combination treatment of SC-CO2 and organic acids may be useful in the meat industry to help increase microbial safety.  相似文献

Aims:  To study the evolution of rind microbial communities in Fontina PDO cheese.
Methods and Results:  Four batches were examined for their surface microflora during ripening, carried out in two different maturing caves, at Ollomont and Pré-Saint-Didier, Aosta Valley region, Northwest of Italy. Culture-dependent methodologies were combined with culture-independent analysis (PCR-DGGE). Yeasts were found to increase from 103 to 106 CFU cm−2 in 28 days, with consequent rise of surface pH, which allowed the growth of salt-tolerant bacteria, in particular coryneforms which reached 109 CFU cm−2 at the end of 3 months. Coagulase-negative cocci and lactic acid bacteria reached 107 CFU cm−2 in the same period. Debaryomyces hansenii and Candida sake were the species more constantly present throughout the whole maturing process. As early as after 1 day since manufacture, Lactococcus lactis subsp. lactis and Streptococcus thermophilus were detected on cheese rinds. Arthrobacter nicotianae , Brevibacterium casei and Corynebacterium glutamicum were found after 7–28 days .
Conclusions:  According to cluster analysis of DGGE profiles, the maturing environment seemed to influence the dynamics of microbial groups on Fontina surfaces.
Significance and Impact of the Study:  These results represent a first picture of micro-organisms colonizing Fontina PDO rinds. Further studies are in progress to better understand the origin of this surface microflora and to formulate surface starters.  相似文献

Aims:  To determine Legionella pollution prevalence, describe the amount of Legionellae with respect to temperature in Shanghai cooling tower water (CTWs) in various types of public sites.
Methods and Results:  Six urban districts were selected as the study fields, adopting multiple-phase sampling methods. Routine culture was used to identify Legionellae. Of the samples, 58·9% (189/321) were observed to be positive, 19·9% were isolated over 100 CFU ml−1. Legionella pneumophila serogroup 1 was the most frequently isolated species (155/189, 82·0%), followed by Leg. micdadei that was at the second place (44/189, 23·3%). The mean CFU ml−1 of Legionellae in CTWs reached its peak from July to September. Over all 15·4% of the samples exceeding 100 CFU ml−1 were observed in a hospital setting.
Conclusions:  The prevalence of Legionella pollution in CTWs, especially in CTWs of subway stations and hospitals, is worrying, and the positive rate and CFU ml−1 of Legionellae in CTWs have a close relationship with air temperature.
Significance and Impact of the Study:  The study demonstrates pollution prevalence rates in different types of sites and various seasons, and provides a proportion of different serogroups of Legionellae . It illuminates an urgent need for dealing with the potential risk of legionellosis in Shanghai, through improved control and prevention strategies.  相似文献

1. The effects of prolonged ultraviolet-B (UVB) radiation on freshwater communities were studied in indoor microcosms (600 L) with artificial light sources, simulating a clear, shallow, mesotrophic aquatic ecosystem. A range of six intensities (in duplicate) of UVB radiation, ranging from 0 (control) to 9.56 kJ m−2 day−1 at the water surface, was applied for 8 weeks. The UVB radiation levels, attenuation, shading and scattering were comparable to those in Dutch shallow freshwater systems. Physical, chemical and biological variables were monitored weekly.
2. The UVB treatment did not affect the abundance, species composition or biovolume of the phytoplankton or zooplankton communities, nor did it affect the periphyton or the macroinvertebrate community. A few species showed a significant response on some of the sampling dates, but there was no negative UVB effect at the community level. Overall, the ecosystems in the microcosms were not affected by the UVB treatment.
3. In a bio-assay, a laboratory clone of Daphnia pulex , not subjected to UVB radiation, was fed with seston from the microcosms. Daphnia pulex feeding on seston from the control microcosms grew faster, had better survival and better reproduction than D. pulex feeding on seston from the UVB treated microcosms. The phytoplankton–zooplankton interaction may have been influenced by the UVB treatment.
4. The dissolved oxygen content (DOC) concentrations in the microcosms were around 5 mg L−1. The DOC levels in Dutch systems rarely fall below 10 mg L−1. This might provide sufficient protection against the detrimental effects of increased UVB radiation.  相似文献

This was a preliminary investigation to define the conditions of colonization of a human skin equivalent (SE) model with cutaneous microorganisms. SEs of 24 mm diameter were constructed with a dermal matrix of fibrin containing fibroblasts and a stratified epidermis. Microbial colonization of the SEs was carried out in a dry environment, comparable to ' in vivo ' skin, using a blotting technique to remove inoculation fluid. The microbial communities were sampled by scrub washing and viable cells enumerated on selective growth medium. Staphylococcus epidermidis , Propionibacterium acnes and Malassezia furfur (human skin commensals) and Staphylococcus aureus (transient pathogen) were colonized at inoculum densities of 102–106 CFU SE−1 on the surface of replicate SEs. Growth of all species was supported for upto 72–120 h, with recovery densities of between 104–109 CFU SE−1. A novel, real-time growth monitoring method was also developed, using S. aureus containing a lux cassette. Light output increased from 20 to 95 h, and colonization increased from 102 to 108 CFU SE−1, as confirmed by conventional recovery. Thus, the SE model has potential to investigate interactions between resident and transient microbial communities with themselves and their habitat, and for testing treatments to control pathogen colonization of human skin.  相似文献

In this study, Saccharomyces cerevisiae was engineered for simultaneous saccharification and fermentation of cellulose by the overexpression of the endoglucanase D (EngD) from Clostridium cellulovorans and the β-glucosidase (Bgl1) from Saccharomycopsis fibuligera . To promote secretion of the two enzymes, the genes were fused to the secretion signal of the S. cerevisiae α mating factor gene. The recombinant developed yeast could produce ethanol through simultaneous production of sufficient extracellular endoglucanase and β-glucosidase. When direct ethanol fermentation from 20 g L−1β-glucan as a substrate was performed with our recombinant strains, the ethanol concentration reached 9.15 g L−1 after 50 h of fermentation. The conversion ratio of ethanol from β-glucan was 80.3% of the theoretical ethanol concentration produced from 20 g L−1β-glucan. In conclusion, we have demonstrated the construction of a yeast strain capable of conversion of a cellulosic substrate to ethanol, representing significant progress towards the realization of processing of cellulosic biomass in a consolidated bioprocessing configuration.  相似文献

The nutrient supply rates within the canopy of the economically important red algal species, Eucheuma serra J. Agardh were determined experimentally in a recirculating flow-chamber. A single individual was placed in the working section of the 2000 × 200 × 250 mm3 acrylic flow-chamber and subjected to unidirectional water velocities from 1.0 to 9.3 cm s−1. Rates of nutrient supply were determined using 9.7 mm diameter CaSO4 (gypsum) spheres that were attached to the thallus inside and outside of the canopy. The supply rates within the canopy were 56% less than outside of the canopy. Increases in internal and external water velocity asymptotically increased the nutrient supply rates regardless of location. A model was developed to examine how changes in ammonium and nitrate supply compared with the physiologically maximum uptake rates of these nutrients. The results suggest that when the ammonium concentration in the water was 20 µmol L−1 uptake rates were limited by the supply rate especially at velocities below 5 cm s−1, whereas in the case of 20 µmol L−1 of nitrate, the supply of nitrate was more than adequate to maximize nutrient uptake.  相似文献

We tested two predictions required to support the hypothesis that anthropogenic acidic episodes might explain the poor biological response of upland British streams otherwise recovering from acidification: (i) that invertebrate assemblages should differ between episodic and well-buffered streams and (ii) these effects should differentiate between sites with episodes caused by anthropogenic acidification as opposed to base-cation dilution or sea-salt deposition. Chronic and episodically acidic streams were widespread, and episodes reflected acid titration more than dilution. Nonmarine sulphate (16–18% vs. 5–9%), and nitrate (4–6% vs. 1–2%) contributed more to anion loading during episodes in Wales than Scotland, and Welsh streams also had a larger proportion of total stream sulphate from nonmarine sources (64–66% vs. 35–46%). Sea-salts were rarely a major cause of episodic ANC or pH reduction during the events sampled. By contrast, streams with episodes driven by strong anthropogenic acids had lower pH (5.0±0.6) and more dissolved aluminium (288±271 μg L−1) during events than where episodes were caused by dilution (pH 5.4±0.6; 116±110 μg Al L−1) or where streams remained circumneutral (pH 6.7±1.0; 50±45 μg Al L−1). Both biological predictions were supported: invertebrate assemblages differed among sites with different episode chemistry while several acid-sensitive species were absent only where episodes reflected anthropogenic acidification. We conclude that strong acid anions – dominantly nonmarine sulphate – still cause significant episodic acidification in acid-sensitive areas of Britain and may be a sufficient explanation for slow biological recovery in many locations.  相似文献

1. Sediment and nutrient loading in freshwater systems are leading causes of aquatic habitat degradation globally. We investigated the impacts of fine-sediment and nutrient additions on the growth and survival of western toad ( Bufo boreas ) tadpoles and emergent metamorphs in mesocosm and exclosure experiments.
2. Mesocosm tanks received weekly pulses of fine sediments to create initial concentrations of 0, 130 and 260 mg L−1 of suspended sediment and either bi-weekly additions of nutrients (N = 160 μg L−1, P = 10 μg L−1) or no additions in a factorial design. Within mesocosms, tadpole exclosures allowed for quantification of tadpole grazing pressure on periphyton biomass, chlorophyll- a and sediment deposition.
3. Tadpoles receiving sediment additions experienced slower growth rates and reduced survival to metamorphosis, although no effects of treatment were detected on size at metamorphosis or time to metamorphosis. Nutrient additions also lowered survival, but had no impact on other measured parameters of tadpole fitness. Dissections and gut content analysis revealed that tadpoles ingested sediment in large quantities altering the proportion of the organic content of ingested food.
4. Together these results suggest that although sediment was readily consumed by tadpoles, its presence in the larval environment had an overall negative effect on tadpole growth and survival, although not as severe as predicted.  相似文献

Many aspects of biodefense research require quantitative growth assessments of the test agent. This study evaluated the BioNanoPore (BNP™) technology to quantitate Bacillus anthracis and Yersinia pestis faster than traditional plate counting methods. The BNP™ technology enabled quantification of B. anthracis and Y. pestis in phosphate-buffered saline and naïve rabbit blood at 6 and 24 h, respectively. After 6 h of growth, counts for B. anthracis ranged from 6·19–6·45 log10 CFU ml−1 on BNP™, while counts after 24 h on tryptic soy agar (TSA) ranged from 6·51–6·58 log10 CFU ml−1. For Y. pestis , counts on BNP™ at 24 h ranged from 6·31–6·41 log10 CFU ml−1 on BNP™ and ranged from 6·44–6·89 log10 CFU ml−1 on TSA at 48 h. This study demonstrates that the BNP™ technology provides a more rapid detection of B. anthracis and Y. pestis , which could aid in the evaluation of potential medical countermeasures and treatments as well as other biological defense applications such as surface sampling or decontamination efficacy.  相似文献

Yeast communities associated with sugarcane leaves, stems and rhizosphere during different phases of plant development were studied near Campos, in Rio de Janeiro, Brazil. Atmospheric temperature, soil granulometry and pH, and sugar cane juice degree Brix and pH were determined. Yeast communities associated with sugarcane were obtained after cellular extraction by shaking, blending and shaking plus sonication, and cultured on Yeast Nitrogen Base Agar plus glucose (0.5%) and Yeast Extract-Malt Extract Agar. No significant differences in yeast counts were found among the cellular extraction treatments and culture media. 230 yeast cultures were identified according to standard methods, and distinct yeast communities were found for each substrate studied. The prevalent species isolated from sugarcane were Cryptococcus laurentii, Cryptococcus albidus, Rhodotorula mucilaginosa and Debaryomyces hansenii.  相似文献

1. A sediment core from the shallow, hypertrophic Lake Søbygaard (mean depth ∼1 m; [TP] 310 μg P L−1) was analysed for subfossil remains to reconstruct chironomid community changes in relation to the succession and disappearance of aquatic macrophytes. 2. Species composition in the 1.10 m core indicates a succession from a 'naturally' eutrophic state to a hypertrophic state during recent centuries. Radiometric dating (210Pb) of the uppermost 20 cm of the sediment core (∼1932–93) indicates that sediment accumulation rate had doubled in recent decades. 3. Changes in chironomid assemblages were in close agreement with changes in both diatoms and macrophyte remains in the same core. Distinct changes in chironomid communities reflect the eutrophication process and macrophyte succession through Chara , Ceratophyllum and Potamogeton dominance to the present state, with complete loss of submerged vegetation and dominance by phytoplankton. 4. The co-occurrence and relationship between aquatic macrophyte diversity and recent subfossil chironomid assemblages were assessed from an additional 25 Danish lakes. There was good agreement between the macrophyte and chironomid-based lake groupings. Overall, a significant difference ( P <0.001) was found in chironomid assemblages among lakes in different macrophyte classes. In a pair-wise comparison, the poorly buffered mesotrophic lakes and the alkaline eutrophic lakes had significantly different chironomid assemblages. 5. Chironomid taxa commonly reported to be associated with macrophytes ( Cricotopus , Endochironomus and Glyptotendipes ) were shown also to be indicators of highly productive lakes lacking abundant submerged vegetation.  相似文献

1. Taxonomic composition and abundance of heterotrophic flagellates (HF) were studied in 55 lakes with different trophy in northern Germany using a live-counting technique.
2. Mean abundances and biomasses of HF ranged from 169 cells L−1 and 22 μg L−1 in mesotrophic lakes to 2439 cells mL–1 and 475 μg L−1 in hypertrophic lakes, respectively. Highest values were generally observed in spring, but mesotrophic lakes showed maximum values in early summer.
3. The taxonomic composition of HF was not significantly influenced by lake trophy and season. The major fraction of HF consisted of chrysomonads and Protista incertae sedis; other important groups were choanoflagellates and bicosoecids. The size distribution of HF changed with lake trophy and season, with a higher proportion of large HF (> 10 μm) in hypertrophic lakes and in spring, respectively.
4. Correlation analyses revealed a strong negative impact of cladocerans on total HF biomass and especially on large HF. Ciliates and large bacteria (> 10 μm) were strongly positively correlated with HF biomass; small bacteria (< 2 μm) showed a weak positive correlation.
5. Analyses at the level of species and genera revealed distinct distribution patterns of some taxa. Paraphysomonas , Aulacomonas and Quadricilia as large-bodied HF showed highest abundance in hypertrophic lakes and in spring. Attached taxa (e.g. Monosiga , Salpingoeca amphoridium ) were highly abundant in late summer and autumn, whereas Spumella and Kathablepharis occurred frequently in most samples.  相似文献

The ratio of dissolved fixed inorganic nitrogen to soluble inorganic phosphate (N:P) in the ocean interior is relatively constant, averaging ~16 : 1 by atoms. In contrast, the ratio of these two elements spans more than six orders of magnitude in lakes and other aquatic environments. To understand the factors influencing N:P ratios in aquatic environments, we analyzed 111 observational datasets derived from 35 water bodies, ranging from small lakes to ocean basins. Our results reveal that N:P ratios are highly correlated with the concentration of dissolved O2 below ~100 µmol L−1. At higher concentrations of O2, N:P ratios are highly variable and not correlated with O2; however, the coefficient of variation in N:P ratios is strongly related to the size of the water body. Hence, classical Redfield ratios observed in the ocean are anomalous; this specific elemental stoichiometry emerges not only as a consequence of the elemental ratio of the sinking flux of organic matter, but also as a result of the size of the basins and their ventilation. We propose that the link between N:P ratios, basin size and oxygen levels, along with the previously determined relationship between sedimentary δ15N and oxygen, can be used to infer historical N:P ratios for any water body.  相似文献

The growing resistance against antibiotics demands the search for alternative treatment strategies. Photodynamic therapy is a promising candidate. The natural intermediate of chlorophyll biosynthesis, protochlorophyllide, was produced, purified and tested as a novel photosensitizer for the inactivation of five model organisms including Staphylococcus aureus, Listeria monocytogenes and Yersinia pseudotuberculosis , all responsible for serious clinical infections. When microorganisms were exposed to white light from a tungsten filament lamp (0.1 mW cm−2), Gram-positive S. aureus, L. monocytogenes and Bacillus subtilis were photochemically inactivated at concentrations of 0.5 mg L−1 protochlorophyllide. Transmission electron microscopy revealed a disordered septum formation during cell division and the partial loss of the cytoplasmic cell contents. Gram-negative Y. pseudotuberculosis and Escherichia coli were found to be insensitive to protochlorophyllide treatment due to the permeability barrier of the outer membrane. However, the two bacteria were rendered susceptible to eradication by protochlorophyllide (10 mg L−1) upon addition of polymyxin B nonapeptide at 50 and 20 mg L−1, respectively. The release of DNA and a detrimental rearrangement of the cytoplasm were observed.  相似文献
