A survey of the sources of Clostridium botulinum type C toxin possibly utilized as food by aquatic birds in an epizootic area of avian botulism in northern Utah showed that living aquatic and terrestrial invertebrates normally found in close association with dead, decomposing birds commonly carried the toxin. Of 461 samples associated with 21 species of avian carcasses, 198 were toxin-positive. Invertebrate species not normally scavengers of vertebrate tissues were less commonly and less highly toxic, particularly when captured 30 cm or more from a carcass; six of 237 samples of such aquatic invertebrates low-level toxin. Of the species tested, blow fly larvae (Calliphoridae) were the most consistently and highly toxic, although others, particularly adult and larval stages of several species of beetles (Coleoptera), contained toxin at levels probably significant in the epizootiology of the disease. An estimated 0.05 to 0.25 g of the most toxic fly larvae or 15 g of the most toxic beetles tested carried a mediam lethal dose for an adult mallard duck. Examination of stomach contents of aquatic birds dead of botulism showed that some had consumed invertebrates.  相似文献

Colonization by blowflies (Diptera: Calliphoridae) of mouse carcasses exposed in open agricultural land near Durham (54 45'N) changed from early spring monopolization by Calliphora vicina R.-D. to a summer pattern of multiple species exploitation by this species together with Lucilia caesar L., L. illustris Mg., L. silvarum Mg., L. sericata Mg. and L. richardsi Collin. In a garden at the edge of Durham, mouse carcasses were dominated by C. vicina from spring to autumn. Difference in mouse colonization between the agricultural and garden sites seemed to reflect differences in the blowfly species present, as measured by baited trap catches at the sites. In sets of C. vicina reared from mice under conditions of competition for larval food, it was found that resulting females were significantly larger than males, size being measured as mean wing length. Blowfly production from three sheep carcasses exposed successively at the agricultural site was dominated by C. vomitoria L. and L. caesar, but also produced other Lucilia species in small numbers, including L. sericata. These L. sericata females from sheep that had died from causes other than myiasis included full-sized specimens, in contrast to those produced from mouse carcasses that were all undersized individuals. As L. sericata females trapped on sheep pastures are predominantly full-sized, this suggests that large carcasses may, in part, be a source of the L. sericata population that attacks sheep as a myiasis agent. The nature of large carcasses as possible sources of L. sericata in lowland Britain is discussed.  相似文献

Abstract.  Understanding the effects of low winter temperatures on mortality is essential in the development of a full understanding of the long-term population dynamics of any insect. The present study aims to examine the survival of pupae and larvae of the blow fly, Lucilia sericata , at overwintering temperatures. Groups of pupae and diapausing and nondiapausing third-stage larvae of L. sericata are maintained in cooled incubators at either 3 °C and 6 °C. Groups are removed from the incubators at 3–4-day intervals and transferred either to−8 °C or to 25 °C. After 1 h in the freezer, the larvae and pupae exposed to this cold-shock are also transferred to 25 °C. Larvae and pupae are then allowed to continue development and the number of adults emerging from each group is counted. The results demonstrate that survival decreases linearly with the period of exposure at both 3 °C and 6 °C. Mortality is higher at 3 °C than at 6 °C and, in groups that receive the cold shock, cold-shock reduces emergence by over 50%. However, there is no consistent tendency for diapausing larvae to survive prolonged cold or cold shock better than other life-cycle stages. The results suggest that the facultative development of an overwintering diapause stage in L. sericata does not appear to be an adaptation to enhance cold tolerance or resistance to cold shock. It is concluded that the survival of overwintering L. sericata is likely to be relatively less affected by low temperatures than it is by, for example, biotic factors, particularly given the buffered soil environment and short time-scales over which periods of cold act.  相似文献

Laboratory investigations were made into the effect of cultivar type, prior cold storage, fumigation temperature and methyl bromide concentration, on the efficacy of a fumigation treatment to control Liriomyza trifolii in chrysanthemum cuttings. The tests related to the standard quarantine treatment used in the UK to control Spodoptera littoralis on imported chrysanthemum cuttings: cold storage for 2 days at 1 – 2°C followed by methyl bromide fumigation at 15°C with a concentration time product (CTP) of 54 g h/m3. L. trifolii larvae, within detached leaves, and 1 – 2 and 2 – 3 day old pupae, were treated. Methyl bromide concentrations of 6·75 or 13·5 g/m3 were used to achieve a range of CTPs and thus obtain accurate dose-response lines and estimates of the LD99 and LD99·9 for each insect stage. Fumigation temperatures were 8, 11 or 15°C. Efficacy of the standard treatments differed between the three cultivars tested, but the LD99 for larvae remained below 54 g h/m3. Decreasing fumigation temperature to 11°C or less increased LD99 values for larvae and pupae and substantially increased variability. There is therefore little scope for using fumigation temperatures of less than 15°C for quarantine purposes. Omitting the cold storage treatment prior to fumigation did not significantly affect efficacy of fumigation. Reducing the methyl bromide concentration from 13·5 to 6·75 g/m3 did not significantly affect the LD99 for larvae but significantly reduced LD99s for pupae.  相似文献

Abstract. Duration of some pre-adult stages of Calliphora vicina Robineau-Desvoidy, C.alpina Zett., C.vomitoria Linn., Phormia terraenovae R.-D. and Lucilia sericata Meig. were determined at 3.5–26oC, in the context of use of these insects in forensic estimates of post-mortem intervals. Egg development in C. vicina occurs down to at least 3.5oC, larval growth to 4oC and puparial formation, development and perfect adult emergence to 5oC. Growth curves by successive weighing of individual larvae showed specific differences in rates of development of the above species at constant temperatures. Alternating temperatures for 12 h periods (such as 10oC and 20oC) to simulate diurnal changes, usually, but not at all temperature pairs tested, caused acceleration of growth relative to the corresponding constant temperature (15oC in the above case), in C.vomitoria, P.terraenovae and L.sericata , but usually caused retardation of larval growth in C.vicina. In this species, larvae as single individuals on beef liver in small tubes grew on average at the same rates as those (50–100 larvae) growing communally. All species showed variation in larval growth between individuals, more marked in C.vicina , and slower-growing larvae showed increased unexplained mortality. Because of this and to avoid complication by fast-growing larvae from later laid egg batches on a carcass, growth curves given are those of mean weights of the heaviest 25% (upper quartile) of groups of 40–90 feeding larvae. Variation in growth rates, and acceleration or retardation of growth in different species under alternating temperatures, thus affect suggested procedures in obtaining estimates of post-mortem intervals using these insects.  相似文献

Carcasses of mosquito larvae killed by Bacillus thuringiensis var. israelensis allow its complete growth cycle (germination, vegetative growth, and sporulation), thus becoming toxic themselves to scavenging larvae. In this study, we demonstrate that the bacterium is capable of inducing death of Aedes aegypti pupae and of recycling in the resulting carcasses. B. thuringiensis var. israelensis-killed pupae were obtained by treating 40-hr-old synchronized fourth instar larvae with a low dose of spores (8000/ml). The fraction of dead pupae was reduced by higher or lower spore concentrations as well as by treating younger or older larval populations (both fourth instar): Increased proportions of dead larvae were obtained at higher concentration or by earlier treatment, whereas lower concentrations or later treatment resulted in more living pupae. Multiplication of B. thuringiensis var. israelensis is shown to occur in the carcasses of dead pupae. The number of spores in each pupal carcass followed a similar kinetic as in larval carcasses, but the final yield was about 10-fold higher, apparently reflecting the difference in dry weight between the two mosquito developmental stages (426 micrograms vs 83 micrograms, respectively). The specific larvicidal activity in a homogenized dead pupa was similar to that of B. thuringiensis var. israelensis powder, LC50 of about 600 spores/ml.  相似文献

1. Freezing was lethal in the eggs, larvae, pupae and adults of the blowflies Calliphora vicina R-D. and Calliphora vomitoria (L.), but varying degrees of supercooling were found. 2. Cold resistance (as determined by their supercooling capacity) was greatest in eggs (to c. -25 degrees C), and moderate in other life stages (range -7 to -13 degrees C), which was evident from the distributions of their supercooling points. 3. Water contents of all life stages varied from 63% to 77% of fresh weight; pupae of C. vomitoria contain the smallest proportion of water. No relationship between the amount of body water and supercooling ability was detected. 4. Glucose was the major compound in all life stages of C. vicina, with a maximum concentration of 8.22 micrograms mg-1 fresh weight in the adults. In addition, five other potential cryoprotectants occurred in concentrations greater than 0.1% of fresh weight. No correlation between solute concentration and supercooling was observed. 5. Both C. vicina and C. vomitoria have sufficient cold hardiness to avoid lethal freezing in sheltered habitats in southern Britain, which may render diapause unnecessary in both species.  相似文献

The mortality of larvae and pupae of the blowfly Lucilia sericata (Meigen) were examined during winter and spring, for two or three years. In soil cores in the field, 70-95% of the larvae died overwinter. Larvae congregated in the top 10 cm of the soil core and did not move extensively throughout the column during the winter. Larvae and pupae at greater depth were less likely to pupariate and emerge successfully than larvae closer to the surface. Under semi-natural conditions, where pupae were placed outside in sawdust filled tubes, in the absence of the usual biotic mortality factors, the mortality of larvae was considerably lower and was also unaffected by low winter temperatures. Hence, low temperatures did not appear to be the primary cause of high overwintering mortality in the field which, it is suggested, is more likely to be the result of the action of biotic mortality factors, such as entomopathogenic nematodes and fungi.  相似文献

1. A simple artificial diet was devised for larvae of Lucilia sericata Meigen. 2. A basic diet of 20 g/l agar with 20% horse blood and added yeast sustained normal growth and development, as compared with a lamb meat control. 3. When yeast was added to the blood agar diet at 50 g/l, both peak and final larval weights were increased by 25-50% at higher larval densities (2-8 larvae/g diet) and the hatchling-adult development period was reduced by about 3 days. 4. No adult insects emerged on an agar-yeast diet without blood. Increasing the concentration of blood from 10% to 20% increased adult weight at emergence by 50%, though developmental period was not significantly affected. 5. Increasing larval density significantly reduced the weights of pupae and emergent adults (by up to 50%), both on the blood-agar-yeast diet and the lamb meat control. Percentage survival fell from 80-90% to below 10% on the blood-agar-yeast diet at the highest densities of 4 or 8 larvae/g diet, though survival on lamb was not significantly affected by density.  相似文献

Abstract. To examine the species composition of flies breeding in carrion in the field, the carcasses of mice and quail were exposed on sheep farms in the South West of England. Calliphora vicina was the dominant species of Diptera; 19,294 individuals emerged from 175 of the 241 infested carcasses recovered. Lucilia sericata emerged from only 39 of the infested carcasses, at a median of 10 adults per infested carcass. Other species of Lucilia present were L.ampullacea, L.caesar and L.illustris. The highest number of L.sericata emerged from carcasses placed in open pasture, the highest number of C.vicina emerged from carcasses in hedgerow, whereas the highest numbers of L.caesar, L.ampullacea and L.illustris emerged from carcasses in woodland. The duration of exposure of carcasses in the field was negatively related to the size of the adult L.sericata which emerged and, in woodland and hedgerow habitats, to the number of L.sericata which emerged. These data indicate that the larvae of L.sericata in carcasses experience significant levels of competition and that the intensity of this competition may be sufficient to reduce the numbers of L.sericata able to emerge successfully. The size distributions of female L. sericata which emerged from carcasses or which were caught as adults in the field showed only a small degree of overlap, suggesting that only a relatively small proportion of the wild L.sericata population emerge from carcass breeding sites. The results are discussed in relation to the development of new approaches to the control of blowfly strike of sheep.  相似文献

Injection of stationary phase culture-supernatants of Staphylococcus aureus and Pseudomonas aeruginosa into the hemolymph of silkworm larvae caused their death, whereas a culture-supernatant of a non-pathogenic strain of Escherichia coli did not. A culture-supernatant of a mutant of agr, a global virulence regulator of S. aureus that is required for exotoxin production, was much less toxic to silkworm larvae. A culture-supernatant of a disruption mutant of the S. aureus beta-toxin gene did not kill larvae, whereas one of a deletion mutant of alpha-toxin, gamma-toxin, or aureolysin killed larvae, indicating that the beta-toxin gene is required for staphylococcal supernatant-mediated killing of silkworm larvae. The 50% lethal doses (LD50) of staphylococcal alpha-toxin and beta-toxin, Pseudomonas exotoxin A and diphtheria toxin were 12 microg/g, 9 microg/g, 0.14 microg/g and 1.1 microg/g, respectively. As the purified toxins killed the larvae, silkworm larvae could be used as a model to study the actions of pathogenic bacterial toxins in animal bodies.  相似文献

We determined that the number of insect fragments, quantified using the standard flotation method, in flour milled from wheat infested with larvae, pupae, or preemergent adults of the lesser grain borer, Rhyzopertha dominica (F.), was proportional to infestation level. Wheat infested with a single preemergent adult contributed 28 and 10x as many fragments as wheat infested with a single larva or pupa, respectively. Using regression models that were developed from these data, we predicted that the maximum infestation level that would result in flour with fragment counts below the Food and Drug Administration defect action level (75 fragments/50 g of flour) was 0.95 and 1.5% (380-640 infested kernels/kg of wheat) for pupae and larvae, but it decreased to 0.05% (20 infested kernels/kg) when the grain was infested with preemergent adults. We also reexamined the accuracy and sensitivity of near-infrared spectroscopy (NIRS) for detecting insect fragments in flour by testing three different NIR spectrometers. NIRS-predicted numbers of insect fragments were correlated with the actual number of fragments. NIRS is less precise than the standard flotation method, but it is rapid, nondestructive, does not require extensive sample preparation, and could easily be automated for a more sophisticated sampling protocol for flour based on prescreening samples with NIRS followed up by use of the standard flotation method when necessary.  相似文献

Abstract Samples of dipteran larvae were collected from live sheep throughout Scotland, reared in the laboratory, and identified once adult flies emerged. Lucilia sericata was found in 77% of samples, and other species in 49%. The most common alternative species were L.caesar , which occurred in 31 % of samples, and Protophormia terraenovae , which occurred in 18%. Three other calliphorid species, Calliphora vomitoria, C.vicina and Lillustris , and the muscid Muscina pabulorum were also found. The proportion of samples containing alternative species was significantly lower in eastern Scotland than in western Scotland. Significantly higher proportions of samples containing alternative species were collected at altitudes of 200 m and above; from sheep of hill breeds; from rough grazing conditions and moorland; in the absence of trees; and in the presence of bracken.  相似文献

We investigated the neuroendocrine regulation of the development of seasonal morphs in a bivoltine race (Daizo) of the silkmoth, Bombyx mori, by decerebration, the transplantation of brain-suboesophageal ganglion (Br-SG) complexes and the injection of active neuropeptides. When brains were removed from fresh pupae destined to develop into summer morphs (SD pupae) by embryonic and larval exposures to short days at low temperature, the pupae developed into autumn or intermediate morphs. However, in pupae destined to develop into autumn morphs (LD pupae), the operation did not show an effect on seasonal morph development. Br-SG complexes were excised from fifth-instar LD and fifth-instar SD larvae 2 days after larval ecdysis and were transplanted into the abdomen of SD larvae of the same age. The Br-SG complexes of LD larvae, but not the Br-SG complexes of SD larvae, shifted the host's seasonal morph development toward the autumn morph. Furthermore, when treated with crude pupal SGs extract and diapause hormone (DH), fresh SD pupae developed into autumn or intermediate morphs, respectively. Possibly the development of seasonal morphs in the silkmoth, B. mori, is regulated by a novel function of DH. Alternatively, DH may act on the imaginal wing disks at an earlier stage than on the ovaries.  相似文献

For insight into the physiological indicators of diapause in Pieris melete, water and carbohydrate (glycogen and trehalose) levels were measured under both natural and laboratory conditions. The highest water content (3.71–3.79 mg/mg dry weight) was found in larvae and developing pupae, which was substantially higher than in diapausing pupae (2.59 mg/mg dry weight). Water content was almost stable during diapause, except for individuals approaching diapause termination (3.43–3.58 mg/mg dry weight). The total carbohydrate level was significantly higher in pre‐pupae (47.41 μg/mg) compared to larvae (22.80 μg/mg) and developing pupae (21.48 μg/mg). The highest level of trehalose was detected in winter diapausing pupae, and no trehalose was found in larvae or developing pupae. Levels of glycogen were highest in pre‐pupae and lowest in diapausing pupae. Levels of total carbohydrate decreased as diapause proceeded, and no significant changes were found in trehalose levels for diapausing pupae under natural conditions or treated for 60–90 days at 5°C. Pupae treated at 20°C for 60–90 days had significantly lower levels of trehalose than those treated for 30 days. Glycogen content was relatively stable at 5°C, but increased after treatment under natural conditions and 20°C for more than 60 days. These results suggest that the dynamics of water and carbohydrate levels are potential physiological diapause indicators, which show metabolic differences between trehalose and glycogen during diapause development.  相似文献

The habits of blowflies infesting slaughter-houses have been studied to determine the conditions under which control measures are most likely to be effective. Flies found in such numbers as to be considered pests of economic importance were: Calliphora erythrocephala (Meig.) and C. vomitoria (L.); Lucilia caesar (L.), L. illustris (Meig.) and L. sericata (Meig.); and Phormia terrae-novae (R.-D.). All were found breeding profusely on slaughter-house refuse.
The reactions of adult blowflies to weather and the working conditions at the slaughter-house are recorded; the three genera react in differing degrees to relatively minor changes in weather conditions, and redistribution of populations caused by these changes is so rapid as to make difficult the accurate assessment of degree of infestation. It was found that blowflies normally do not settle on the internal fabric of slaughter-house buildings but tend rather to congregate on nearby vegetation. The preferences were studied of adult flies for certain types of meat, for particular parts of carcasses, and for meat exposed to sunlight.
Conditions favouring rapid development of larvae were determined and the rates of breeding recorded for the three genera when in natural competition and when breeding independently. The larvae of P. terrae-novae were found to be predacious on other species and each other. Lucilia sericata was found to oviposit and breed successfully in vegetable matter.
Blowflies overwinter as fully grown larvae which pupate only a short while before emerging in the spring.
Various methods are discussed whereby the findings of this investigation into the life history and habits of blowflies can be utilized in the control of these pests.  相似文献

This study was carried out in the northeast region of Konya in Turkey, between April and December 2004, to determine the vertical distribution of Culicoides (Diptera: Ceratopogonidae) larvae and pupae. Larvae and pupae were collected using the saturated sugar flotation technique in mud samples taken from a range of depths of up to 20 cm. In total, 282 adult Culicoides were obtained. The number of larvae and pupae decreased as the depth increased; at 2.5 cm, 97 Culicoides larvae and 73 Culicoides pupae were found. Larvae and pupae of C. nubeculosus (Meigen) and C. puncticollis (Becker) were mostly found in organic matter-rich breeding sites, while larvae and pupae of C. gejgelensis Dzhafarov and C. festivipennis Kieffer were mostly found in organic matter-poor breeding sites. Culicoides larvae and pupae were obtained largely in July and August.  相似文献

1. The relative abundance of the blowflies Calliphora vicina (R.-D.) and Lucilia sericata (Meigen) in carrion was considered in relation to inter- and intraspecific larval competition and the distribution of adults between habitat types. 2. In mixed and pure laboratory cultures of L. sericata and C. vicina the mortality of both species increased and adult size declined as the initial larval number was increased. However, for L. sericata at initial numbers greater than ten larvae/g of liver, the effects of competition on adult size and mortality were greater in the mixed cultures than in the pure cultures. In contrast, for C. vicina at numbers greater than ten larvae/g of liver, the effects of competition on adult size and mortality were greater in the pure cultures than in the mixed cultures. 3. The laboratory data suggest therefore that for L. sericata the effects of interspecific competition with C. vicina on size and mortality were greater than the effects of intraspecific competition in a pure culture at the same initial number. Notably, however, the intensity of interspecific competition was not sufficiently asymmetric to allow C. vicina to exclude L. sericata even at the highest numbers examined. 4. In the field, higher numbers of adult L. sericata emerged from the carcasses of laboratory mice placed in open pasture than in woodland or hedgerow sites. In contrast, higher numbers of C. vicina emerged from carcasses placed in woodland and hedgerow sites. 5. Trapping showed that in the field adult L. sericata were relatively more abundant in open pasture than in woodland and hedgerow sites, while C. vicina were more abundant in woodland and hedgerow sites than in open pasture. 6. It is concluded that the low numbers of L. sericata that emerge from carrion relative to the numbers of C. vicina may, in part, be the result of asymmetric interspecific competition, but that the uneven distribution of adults of the two species among habitat types also plays a major role in shaping the blowfly community in carrion.  相似文献

Populations of the blowfly, Lucilia sericata (Meigen) (Diptera: Calliphoridae), have a considerable potential for rapid increase; the lifetime reproductive output of each adult female has been estimated to be between 130 and 172 eggs. Nevertheless, in the field, absolute population densities of this species are relatively low. To account for this difference, the levels of mortality affecting the eggs, feeding and wandering larvae and pupae of L. sericata were assessed in the field and laboratory. Percentage egg hatch was dependent on relative humidity with no egg eclosion at humidities below 50%; there was no significant effect of temperature on egg hatch. On infested sheep, the mean mortality of feeding larvae was 53%, but this ranged widely from 0% to 96%. There was no effect of atmospheric temperature or humidity on the mortality of feeding larvae in vivo. In the laboratory, only 10% of wandering larvae pupariated at 10 degrees C. At above 20 degrees C pupariation was consistently almost 100%. Percentage emergence increased from 0% at 10 degrees C to about 80% between 20 and 30 degrees C. The upper lethal temperature for pupae was approached at 35 degrees C. Analysis of the predation of pupae in the field revealed a weak, but significant curvilinear relationship between temperature and proportionate mortality and a median mortality of 0.49% per 24 h exposure (interquartile range = 5.2%). There was no evidence of density dependence in pupal predation. Overall, it is estimated that pre-adult mortality accounts for losses of approximately 97% of each generation, but this figure is subject to considerable variation depending on factors such as climate, time of year and host susceptibility.  相似文献

Serum samples taken from two infant botulism cases during hospitalization were titrated for botulinum toxin by both the intraperitoneal (ip) injection method and the score method in mice. By the ip method, in which death is the only parameter, such low levels of toxin as lower than 4 ip LD50/ml may not be titrated even though the surviving mice show abdominal palsy. By the score method based on the degree of abdominal palsy, such low levels of toxin as 1.1 and 0.8 ip LD50/ml were detected in specimens of one of the patient's serum. No antitoxin was demonstrated in either case of infant botulism by applying the score method. It is not known whether spontaneous recovery from infant botulism is due to the antitoxin production.  相似文献
