The aim of this study was to evaluate whether iron, like copper, could support Vitamin C mediated hydroxyl radical formation in bicarbonate-rich water. By using the hydroxyl radical indicator coumarin-3-carboxylic acid, we found that iron, in contrast to copper, was not capable to support Vitamin C induced hydroxyl radical formation. However, when 0.2?mg/l iron and 0.1?mg/l copper were both added to bicarbonate supplemented Milli-Q water, the Vitamin C induced formation of 7-hydroxycoumarin, as measured by HPLC analysis, was inhibited by 47.5%. The inhibition of hydroxyl radical formation by iron was also evident in the experiments performed on copper contaminated bicarbonate-rich household drinking water samples. In the presence of 0.2?mg/l of ferric iron the ascorbic acid induced hydroxyl radical formation was inhibited by 36.0–44.6%. This inhibition was even more significant, 47.0–59.2%, when 0.8?mg/l of ferric iron was present. None of the other redox-active metals, e.g. manganese, nickel or cobalt, could support ascorbic acid induced hydroxyl radical formation and did not have any impact on the ascorbic acid/copper-induced hydroxyl radical generation. Our results show, that iron cannot by itself produce hydroxyl radicals in bicarbonate rich water but can significantly reduce Vitamin C/copper-induced hydroxyl radical formation. These findings might partly explain the mechanism for the iron-induced protective effect on various copper related degenerative disorders that earlier has been observed in animal model systems.  相似文献

为研究维生素C多聚磷酸酯对小鼠抗热应激能力的影响 ,将 4 8只 3~ 4周龄、体重为 16~ 2 9g的健康雄性小鼠随机分为 4组 (对照组、Ⅰ、Ⅱ和Ⅲ组 ) ,在其饵料中分别添加 0、 5 0 0、 2 5 0 0和 5 0 0 0mg/kg的35 %维生素C多聚磷酸酯。喂食 4周后 ,每组取一半小鼠处死取其肝脏 ,另一半置于 (35± 1)℃条件下 ,2 4h后作同样处理。用硫代巴比妥酸分光光度法测肝脏脂质过氧化物 (LPO)的含量 ,用亚硝酸盐形成法测超氧化物歧化酶 (SOD)的活性 ,用分光光度法测过氧化氢酶 (CAT)和谷胱甘肽过氧化物酶 (GSH Px)的活性。经热应激与未经热应激相比 ,LPO :对照组和Ⅰ组显著升高 ,而Ⅱ和Ⅲ组无显著差异。SOD和GSH Px :对照组显著下降 ,其他 3组无显著差异 ;其中SOD :Ⅱ和Ⅲ组显著高于对照组和Ⅰ组 ,GSH Px :Ⅲ组显著高于其他 3组。CAT :对照组和Ⅰ组显著降低 ,而Ⅱ和Ⅲ组无显著差异 ;Ⅱ和Ⅲ组显著高于对照组和Ⅰ组。表明热应激促进了小鼠肝脏LPO的产生 ,抑制了抗化物酶的活性 ;而维生素C多聚磷酸酯对热应激造成的不利影响有缓解作用。  相似文献

目的探讨维生素A缺乏(vitamin A deficiency,VAD)对大鼠肺脏发育的影响及其可能的机制。方法建立VAD大鼠模型,取出生后2周正常及VAD幼鼠肺脏,HE、Weigert染色分别观察其肺脏组织形态变化和弹力纤维变化;Image Pro Plus5.0软件做肺脏形态学定量分析,蛋白质免疫印记检测大鼠肺组织MMP-9及TIMP-1蛋白含量变化。结果①VAD组大鼠肺上皮细胞堆积,出现明显的肺气肿,并发弥散的间质性肺炎,肺组织弹力纤维显著减少;②VAD组大鼠肺MMP-9的蛋白质表达水平明显高于对照组,TIMP-1水平变化不大,MMP-9与TIMP-1的比值升高。结论VAD导致大鼠肺脏正常发育受损,肺脏组织形态异常,可能通过上调MMP-9的表达诱发大鼠肺气肿。  相似文献

来自甲鱼养殖场的60只中华鳖(Pelodiscus sinensis)幼鳖驯养3周后,实验设5组:对照组、处理Ⅰ、Ⅱ、Ⅲ和Ⅳ组。各组设2个平行,依次在饵料中混合添加维生素C(Vc)和E(VE)为0和0、250和50、2500和50、250和250、2500和250mg/kg,喂食4周后,每组取半数幼鳖经酸应激处理24h。取幼鳖血液,用镜检法测定血细胞的吞噬率,透射比浊法测定血清溶菌活力、杀菌活力以及补体C3和C4含量。①经酸应激与未经酸应激处理相比:对照组血细胞吞噬率显著降低,而处理Ⅰ-Ⅳ组无显著变化;对照组和处理Ⅰ组血清溶菌活力和补体C3含量显著下降,而处理Ⅱ-Ⅳ组无显著变化;血清杀菌活力均有显著下降(对照组、处理Ⅰ和Ⅲ组极显著,处理Ⅱ组和Ⅳ组显著);对照组、处理Ⅰ和Ⅲ组血清补体C4显著下降,而处理Ⅱ和Ⅴ组无显著变化。②经酸应激处理,血细胞吞噬率、血清溶菌活力、杀菌活力和补体C3含量,处理Ⅰ~Ⅳ组的均显著高于对照组,处理Ⅳ组显著高于其他4组;血清杀菌活力,处理Ⅱ组又高于处理Ⅰ和Ⅲ组;血清补体C4,对照组显著低于处理Ⅰ-Ⅳ组,而处理Ⅰ-Ⅳ组间无显著相异。Vc和VE混合饲喂对酸应激后中华鳖血细胞吞噬率、血清溶菌活力、杀菌活力和补体C3含量有显著协同促进作用,对血清补体C4的合成无协同作用。说明Vc和VE混合饲喂能显著增强中华鳖抗酸应激能力,缓解或部分缓解酸应激造成的不利影响。  相似文献

Two experiments were conducted to evaluate the effects of vitamin C supplementation on performance, iron status and immune function of pigs during the 21-day post-weaning period. In experiment one, 48 crossbred pigs (Chester White ‐ Large White ‐ Yorkshire), weaned at 30 days of age and weighing 7.7 ± 0.9kg, were allotted to diets containing either 0 or 300 mg/kg vitamin C. In experiment two, 96 crossbred pigs (Chester White ‐ Large White ‐ Yorkshire), weaned at 20 ± 2 days and weighing 7.1 ± 0.5kg, were allotted to diets containing 0,75 or 300 mg/kg vitamin C. Six replicate pens were assigned to each treatment in experiment one while experiment two had eight replicates. All pens housed two barrows and two gilts. In both experiments, no improvement (P > 0.05) in growth rate, feed intake or feed conversion was observed as a result of vitamin C supplementation. Plasma iron concentration increased (P < 0.10) with increased vitamin C in the diet while free and total iron binding capacity were unaffected by treatment. There were no differences in the intradermal response to the mitogen phytohemaggutinin used as an indicator of cellular immunity (P > 0.05). In trial 2, the plasma levels of the immunoglobulin IgG showed a linear (P = 0.07) increase with increasing levels of vitamin C and the same trend was noted in trial 1. Antibody titers to bovine serum albumin also tended to increase in both trials but the increases were not statistically significant. In conclusion, the overall results of these experiments indicate that weanling pig performance is not improved as a result of vitamin C supplementation. Whether or not vitamin C plays a role in stimulating humoral immune function in pigs requires further study since the results of our experiments do not completely rule out the possibility that such a role exists.  相似文献

营养源对动物生长发育具有重要作用。本文采用静水实验浸泡法探究葡萄糖和维生素C对镇海林蛙(Rana zhenhaiensis)蝌蚪生长及其苹果酸脱氢酶、乳酸脱氢酶及淀粉酶活性的影响。在葡萄糖实验组共分为0.5、1.0和2.0 g/L三个浓度组,维生素C实验组共分为10.0、20.0和30.0 mg/L浓度组,另设1组加曝气水作为对照组。在葡萄糖实验组中,蝌蚪存活率在不同实验组间的差异不显著(P>0.05)。变态时间在0.5 g/L和1.0 g/L实验组最短,分别为(43.0±4.0)d和(43.0±3.4)d,2.0 g/L实验组最长,为(46.2±5.4)d,且实验组间差异显著(P <0.05)。变态时,0.5 g/L实验组的体重和体全长最大,且各实验组间的体重差异显著(P <0.01),但体全长差异不显著(P> 0.05)。增重率在0.5 g/L实验组最高,为(9.67±1.71)mg/d,对照组最低,为(7.54±1.22)mg/d,且各实验组之间差异显著(P<0.05)。维生素C实验组中,存活率在各实验组间差异不显著(P> 0.05)。所有实验...  相似文献

目的:探究磷酸肌酸钠配合大剂量维生素C对小儿肺炎支原体感染心肌损害患儿炎症因子的影响。方法:收集我院收治的肺炎支原体感染心肌损害患儿72例,根据治疗方法不同分为对照组和试验组,每组36例。对照组给予磷酸肌酸钠治疗,试验组给予磷酸肌酸钠联合维生素C治疗。观察并比较两组患儿治疗前后心肌酶、心功能、炎症因子水平、临床疗效及不良反应。结果:与治疗前比较,治疗后两组患儿血清心肌酶水平均降低,且试验组低于对照组,差异均具有统计学意义(P0.05)。与治疗前比较,治疗后两组患儿心排出量、每搏输出量均升高,且试验组高于对照组,差异均具有统计学意义(P0.05);与治疗前比较,治疗后两组患儿血清IL-18,IL-6及IL-17水平均降低,且试验组低于对照组,差异均具有统计学意义(P0.05);与治疗前比较,治疗后两组患儿血清TGF-β水平均升高,且试验组高于对照组,差异均具有统计学意义(P0.05)。试验组临床总有效率(91.67%)高于对照组(72.22%),差异具有统计学意义(P0.05)。结论:磷酸肌酸钠配合大剂量维生素C治疗小儿肺炎支原体感染心肌损害的临床疗效显著,能够改善心肌功能,抑制炎性反应,安全性较高。  相似文献

为探讨维生素E(VE)对中华鳖(Pelodiscus sinensis)幼鳖血清补体C3和C4的影响及补体在酸应激下的变化,在6组(对照组,实验Ⅰ、Ⅱ、Ⅲ、Ⅳ和Ⅴ组)幼鳖的饵料中依次添加0、50、250、500、1000和5000mg/kg的VE,喂食4周,每组取半数幼鳖经酸应激处理24h。取幼鳖血清,用透射比浊法测定血清补体C3和C4的含量。经和未经酸应激的实验Ⅱ、Ⅲ和Ⅳ组幼鳖血清补体C3的含量均明显高于对照组,实验Ⅰ和Ⅱ组C4含量也明显高于对照组;经酸应激的幼鳖与未经酸应激的比较,对照组和实验Ⅰ组C3和C4的含量显著下降,其余4组没有变化。分析说明,VE在一定剂量范围内能促进血清补体C3和C4的合成,酸应激能导致其下降;而高剂量的VE对酸应激导致的不利影响有拮抗作用。  相似文献

Guinea pigs were fed for five weeks with two diets with different levels of vitamin C, low (33 mg of Vit C/Kg diet) and high (13,200 mg of Vit C/Kg of diet). Catalase was inhibited with 3-amino-1,2,4-triazole (AT) in half of the animals from each dietary group. AT caused an almost complete depletion of liver catalase activity (90%) in both dietary groups. Vitamin C supplementation increased total glutathione peroxidase activity and tissue vitamin C level and decreased levels of protein carbonyls and malondialdehyde (MDA) in both treated and non-treated animals. This vitamin C supplementation did not change any of the other antioxidant defences studied. Our results show that dietary vitamin C supplementation increases global antioxidant capacity and decreases endogenous oxidative damage in the guinea pig liver under normal non-stressful conditions. This supports the protective value of dietary antioxidant supplementation.  相似文献

摘要 目的:探讨维生素C对万古霉素诱导肾损伤自噬水平的影响。方法:将20只雄性SD大鼠随机分为:对照组、万古霉素组、万古霉素+维生素C组和维生素C组。万古霉素组:连续每天腹腔注射400 mg/kg万古霉素;万古霉素+维生素C组:注射万古霉素之前30 min腹腔注射200 mg/kg维生素C;对照组和维生素C组分别单独注射同体积的生理盐水和200 mg/kg维生素C。连续给药7 d后,通过苏木精-伊红染色(HE)观察大鼠肾组织病理损伤;免疫组化和免疫荧光检测肾组织中LC3B和Beclin 1的表达情况,比较各组之间的表达差异。结果:相对于对照组,万古霉素诱导大鼠肾损伤模型组肾组织出现明显的病理改变,包括肾间质水肿,肾小管细胞质空泡性变化,细胞凋亡坏死等;同时观察到肾组织中LC3B光密度明显升高和Beclin 1的荧光强度显著增强。维生素C处理组,肾组织的病理损伤显著改善并且自噬相关蛋白LC3B和Beclin 1的表达显著降低。相对于对照组,维生素C单独处理组肾组织损伤和自噬相关蛋白的表达无明显变化。结论:维生素C可降低自噬相关蛋白LC3B和Beclin 1的表达,缓解万古霉素诱导的大鼠肾损伤。  相似文献

Vitamin B12 is destroyed by the addition of substantial amounts of vitamin C in the presence of copper. Effects of carnosine and anserine, natural water-soluble antioxidants, on the destruction of vitamin B12, were studied. Cortisol is known to induce lipoprotein lipase (LPL) activity in human adipose tissue in vitro and in vivo such as in Cushing's syndrome. The significance of the glucocorticoid receptor (GR) for this induction was evaluated in the present study. The synthetic steroid molecule RU 486, a potent glucocorticoid antagonist, was used as a tool to block the GR, in vitro and in vivo. In addition to LPL activity, glucose tolerance, blood pressure and plasma lipids, all variables influenced by Cortisol, were studied in order to evaluate the peripheral antiglucocorticoid activity of RU 486 in vivo, in man. Addition of both Cortisol and RU 486 to incubations of human adipose tissue pieces significantly inhibited the increase in LPL activity that could be induced by Cortisol alone (p<0.01). In a ten-fold molarity excess RU 486 totally abolished Cortisol action (p<0.01). With Cortisol and RU 486 in equimolar concentrations the RU 486 blockade was probably incomplete and LPL activity induced (p<0.05). The results imply that the stimulating effect of Cortisol on LPL activity in human adipose tissue is mediated via the GR. Administration of 400 mg RU 486 at 2200 hours on two consecutive days to healthy men caused a significant rise in serum Cortisol levels measured at 0800 hours (p<0.05). The mean LPL activity in the subcutaneous abdominal adipose tissue remained unchanged. The mean level of serum triglycerides decreased significantly (p<0.01) and there was a negative correlation between change in LPL activity and change in triglyceride levels (r=-0.73, p<0.05). Glucose tolerance and blood pressure were not affected. In conclusion, a total blockade of the GR with RU 486 can be achieved in human adipose tissue in vitro. The blockade abolishes the stimulating effect of Cortisol on LPL activity suggesting that the stimulation is GR dependent. In vivo, with the dose used, the negative pituitary feedback regulation probably compensates for the blockade, at least during the morning hours, thus obviating any peripheral effect of blockade of the GR.  相似文献

Microvoltammetric electrodes implanted in the caudate nucleus of the anesthetized rat have been used to monitor dopamine released following electrical stimulation of the medial forebrain bundle. These electrodes are fabricated from unmodified carbon fibers and have been used with normal pulse voltammetry. Dopamine appears in the vicinity of the electrode when the stimulation is initiated, and disappears almost immediately when the stimulation is terminated. The data suggest that the effective diffusion distance is less than 100 micron. Postmortem analysis using liquid chromatography with electrochemical detection shows that dopamine released in this manner is metabolized to 3,4-dihydroxyphenylacetic acid (DOPAC); however, neither substance is observed electrochemically in the extracellular fluid within seconds after the stimulation. In addition, inhibitors of neuronal uptake of dopamine, amphetamine (1.8 or 15 mg X kg-1) or benztropine (25 mg X kg-1), or of dopamine metabolism, pargyline (150 mg X kg-1) or tropolone (100 mg X kg-1), do not significantly affect the rate at which dopamine disappears from extracellular fluid, although they can affect the amount released. These results suggest that dopamine cannot freely diffuse in the extracellular fluid because an extraneuronal uptake mechanism exists that clears dopamine from extracellular fluid into an extraneuronal pool where metabolism to 3,4-dihydroxyphenylacetic acid occurs. Dopamine can be observed during electrical stimulation of the ascending fibers because neuronal and extraneuronal uptake systems are unable to remove dopamine on these short time scales.  相似文献

目的:研究大黄蛰虫胶囊联合维生素C钠治疗黄褐斑的临床疗效及对血清和血液流变学的影响。方法:选取2012年8月到2013年8月我院收治的黄褐斑患者140例,按照随机数字表法平均分成研究组和对照组,每组70例,研究组给予大黄蛰虫胶囊联合维生素C钠治疗,对照组给予大黄蛰虫胶囊治疗,两组均治疗1个月。3个月后比较两组临床疗效,并检测两组患者的血清睾酮(T)、雌二醇(E2)、孕酮(P)、过氧化脂质分解产物丙二醛(MDA)、超氧化物歧化酶(SOD)以及血液流变学水平的变化。结果:研究组总有效率85.7%(60/70)显著高于对照组的57.1%(40/70),两组比较差异具有统计学意义(P0.05);与治疗前和对照组相比,治疗后研究组SOD、E2显著升高,MDA、T显著降低,差异均具有统计学意义(P0.05);两组P水平均无明显变化;治疗后研究组血液流变学各指标较治疗前和对照组显著下降,差异均有统计学意义(均P0.05)。结论:大黄蛰虫胶囊联合维生素C钠治疗黄褐斑具有较好的临床疗效,可以调节患者的性激素水平,改善患者的血流变学。  相似文献

Senescence marker protein-30 (SMP30) is a gluconolactonase required for vitamin C (VC) synthesis. We examined effects of VC deficiency on the mouse skin using SMP30 knockout (KO) mice. SMP30 KO or wild type male mice were weaned around day 30 of age, and fed VC-deficient diet. They were given either VC water or control water. VC deficiency for 36 days did not affect skin hydroxyproline contents, while VC deficiency for 60 days decreased the hydroxyproline levels. Levels of some collagen mRNAs were different among the groups, but did not correlate with skin VC levels. The epidermis was morphologically abnormal in VC-deficient SMP30 KO mouse at 60 days after the weaning. Interestingly, the hair cycle was not synchronized among the groups. These data suggest low susceptibility of the mouse skin to VC deficiency and involvement of VC in the regulation of keratinocyte function and hair cycle in vivo.  相似文献

Monocyte recruitment and retention in the vasculature is influenced by oxidative stress and is involved in cardiovascular disease (CVD). Individuals with low plasma ascorbate are at elevated risk of CVD. It is unknown whether vitamin C supplementation affects monocyte adhesion to endothelial cells (ECs) in healthy non-smokers. In a randomised double-blind crossover study the effect of vitamin C supplementation (six weeks, 250 mg/day) was determined in subjects with normal (HIC) and below average (LOC) plasma vitamin C concentration at baseline (mean=67 microM, n=20, mean=32 microM, n=20, respectively). LOC subjects showed 30% greater monocyte adhesion to ECs. This was significantly reduced by 37% (P<0.02) following vitamin C supplementation to levels of HIC monocyte adhesion. No differences in plasma malondialdehyde concentrations were observed between groups or after supplementation. In conclusion, vitamin C supplementation normalises monocyte adhesion in subjects with low plasma vitamin C (LOC). This process may be related to a direct effect on monocytes, independent of lipid peroxidation.  相似文献

本文以湘研15号辣椒为材料,研究了脱落酸(ABA)与镉(Cd^2+)在辣椒米实不同发育时期对辣椒素和维生素C含量的影响,并分析了处理后辣椒素、维生素C含量变化与POD活性以及叶片光合特征之间的关系。结果表明:湘研15号辣椒植株经ABA与Cd^2+处理后果实中辣椒素含量峰值分别比对照提高了80.4%与61.4%,POD活性均显著升高,二者处理后辣椒果实中的辣椒素、POD变化趋势基本一致,处理后叶片光合速率均有下降趋势。ABA处理后果实中维生素C含量在转色期比对照降低12.8%~21.7%,叶片中叶绿素相对值显著增加;Cd^2+处理后果实中维生素C含量在转色期比对照减少18%~25%,红熟期减少13%-26%,叶片叶绿素相对值在红熟期之前比CK显著降低。  相似文献

为探讨维生素C (VC)和维生素E (VE)联用对应激和非应激中华鳖幼鳖的生长、肝脏VC 和VE 以及血清皮质醇含量的影响 ,作者使用了 5组饵料 ,VC 和VE 的添加量依次为 0和 0mg/kg (对照组 )、 2 5 0和 5 0mg/kg (实验Ⅰ组 )、2 5 0 0和 5 0mg/kg (实验Ⅱ组 ) ;2 5 0和 2 5 0mg/kg (实验Ⅲ组 ) ;2 5 0 0和 2 5 0mg/kg(实验Ⅳ组 )。中华鳖幼鳖的生长、肝脏VC 和VE以及血清皮质醇分别通过特定生长率、高压液相色谱法和放免法来测定。结果实验Ⅰ -Ⅳ组中华鳖的特定生长率明显高于不加VC 和VE的对照组 ,但实验Ⅰ -Ⅳ组间没有明显不同。非应激中华鳖肝脏VC 和VE的含量随饵料中VC 和VE 含量的增加而明显升高 ,并且实验Ⅱ -Ⅳ组肝脏VC 和VE都明显高于对照组和实验Ⅰ组。酸应激后 ,对照组和实验组中华鳖肝脏VC 和VE都有下降的趋势 ,但无显著差异 ;应激后实验Ⅱ -Ⅳ组肝脏VC 和VE均明显高于对照组 ,实验Ⅳ组明显高于其它 4组。血清皮质醇的含量在实验Ⅰ -Ⅳ组间没有明显不同 ,实验Ⅰ -Ⅲ组与对照组相比虽有降低的趋势但没有变化 ,实验Ⅳ组则明显低于对照组。酸应激后 ,对照组血清皮质醇明显升高 ,其他 4组虽有升高的趋势 ,但没有明显变化。应激后实验Ⅰ -Ⅳ组血清皮质醇的含量均明显低于对照组 ,实验Ⅰ -Ⅳ组间没有明显  相似文献
