Effect of nitrogen supply on frost resistance, nitrogen metabolism and carbohydrate content in white clover (Trifolium repens) 总被引:1,自引:0,他引:1

Effects of mineral nitrogen (2, 4, 6 and 8 m M NH4 NO3 ) and nodulation with Rhizobium on frost hardiness in seedlings of white clover ( Trifolium repens ) have been studied. Seedlings of a population from Bodø (67°N lat.) were grown in Leonard jars under controlled conditions in a phytotron. For induction of frost hardening, plants were first exposed to 12 h photoperiod conditions for 2 weeks at 18°C, then for 2 weeks at 6°C and finally for 2 weeks at 0.5°C. Frost hardiness after treatments at 6 and 0.5°C was significantly enhanced by increasing nitrogen supply and was positively correlated with total nitrogen content of the stolons. Frost hardiness of nodulated plants correlated to the tissue nitrogen concentration. Content of soluble proteins in stolons decreased during hardening at 6°C but did not change during treatment at 0.5°C. There were minor changes in total amount of free amino acids during hardening. Both absolute and relative amounts of proline and arginine increased, and those of asparagine decreased during hardening. Absolute amounts of all free amino acids increased with increasing nitrogen supply, but the changes during hardening were similar in all treatments. There was a significant increase in the content of soluble carbohydrates during hardening. However, this increase was inversely related to nitrogen supply. 相似文献

Electron attachment rates and gas phase acidities for the canonical tautomers of the nucleobases and electron affinities for thymine, deprotonated thymine, and cytosine are reported The latter are from a new analysis of published photoelectron spectra. The values for deprotonated thymine are (all in eV) keto-N1-H, 3.327(5); enol-N3-H, 3.250(5), enol-C2OH, 3.120(5) enol-N1-H, 3.013(5), and enol-C4OH,3.123(5). The values for deprotonated cytosine, keto-N1-H, 3.184(5); trans-NH-H, 3.008(5); cis-NH-H, 3.039(5); and enol-N1-H, 2.750(5) and enol-O-H, 2.950(5). The gas phase acidities from these values are obtained from these values using experimental or theoretical calculations of bond dissociation energies. Kinetic and thermodynamic properties for thermal electron attachment to thymine are obtained from mass spectrometric data. We report an activation energy of 0.60 eV and electron affinity of thymine, 1.0(1) eV. 相似文献

Escherichia coli mutants which secreted thymidine, thymine, uracil, cytosine, and guanine into the culture medium were isolated. The isolation strategy was based on the combination of a sensitive screening method and a mutant-generating system. The screening method made use of a thyA mutant of E. coli. These cells, when spread on the agar surface with the 3-galactosidase indicator X-gal, will grow into bule colonies if a minute amount of thymidine is supplied to them from a nearby secretor colony. A chemostat was used as a mutant-generating system to select for E. coli mutants that were resistant to inhibitors of the pyrimidine biosynthetic pathway. Although many mutants were selected based on their secretion of thymidine, other kinds of nucleosides and nucleobases, such as cytosine, uracil, guanine, and thymine, were also present in larger quantities. This rational selection strategy should be applicable to other species of micro-organisms for the isolation of better producers of nucleosides. The production of nucleosides and nucleobases by fermentation could then become a possibility. 相似文献

The aim of this study was to analyse the influence of low-temperature wine fermentation on nitrogen consumption and nitrogen
regulation. Synthetic grape must was fermented at 25 and 13°C. Low-temperature decreased both the fermentation and the growth
rates. Yeast cells growing at low-temperature consumed less nitrogen than at 25°C. Specifically, cells at 13°C consumed less
ammonium and glutamine, and more tryptophan. Low-temperature seemed to relax the nitrogen catabolite repression (NCR) as deduced
from the gene expression of ammonium and amino acid permeases (MEP2 and GAP1) and the uptake of some amino acids subjected to NCR (i.e. arginine and glutamine). Low-temperature influences the quantity
and the quality of yeast nitrogen requirements. Nitrogen-deficient grape musts and low temperature are two of the main prevalent
causes of sluggish fermentations and, therefore, the effects of both growth conditions on yeast metabolism are of considerable

The contribution of carbon and nitrogen reserves to regrowth following shoot removal has been studied in the past. However, important gaps remain in understanding the effect of shoot cutting on nodule performance and its relevance during regrowth. In this study, isotopic labelling was conducted at root and canopy levels with both 15N2 and 13C‐depleted CO2 on exclusively nitrogen‐fixing alfalfa plants. As expected, our results indicate that the roots were the main sink organs before shoots were removed. Seven days after regrowth the carbon and nitrogen stored in the roots was invested in shoot biomass formation and partitioned to the nodules. The large depletion in nodule carbohydrate availability suggests that root‐derived carbon compounds were delivered towards nodules in order to sustain respiratory activity. In addition to the limited carbohydrate availability, the upregulation of nodule peroxidases showed that oxidative stress was also involved during poor nodule performance. Fourteen days after cutting, and as a consequence of the stimulated photosynthetic and N2‐fixing machinery, availability of Cnew and Nnew strongly diminished in the plants due to their replacement by C and N assimilated during the post‐labelling period. In summary, our study indicated that during the first week of regrowth, root‐derived C and N remobilization did not overcome C‐ and N‐limitation in nodules and leaves. However, 14 days after cutting, leaf and nodule performance were re‐established. 相似文献

为探寻增强苜蓿耐盐能力的调控途径,以甘农4号苜蓿品种为材料,采用NO供体硝普钠、NO清除剂c-PTIO及硝普钠类似物亚铁氰化钠处理苜蓿幼苗,研究盐胁迫下外源NO对苜蓿幼苗生长、光合特征、氮同化酶活性和氮代谢物含量的影响.结果表明: 外源NO能明显缓解盐胁迫对苜蓿幼苗生长及光合作用的抑制,单株干质量、叶绿素含量、净光合速率、蒸腾速率和可溶性蛋白含量增加;外源NO能增强硝酸还原酶、谷氨酰胺合成酶和谷氨酸合酶活性,抑制蛋白水解酶和谷氨酸脱氢酶活性, 降低叶片中游离氨基酸含量,提高硝态氮含量,加快铵的同化.NO供体SNP的类似物亚铁氰化钠对盐胁迫下苜蓿幼苗生长及氮代谢无调控作用;施用NO清除剂c-PTIO加剧了盐胁迫对苜蓿幼苗生长和氮代谢的抑制,添加外源NO能缓解c-PTIO的抑制效应.盐胁迫下,外源NO和内源NO均参与了苜蓿幼苗氮代谢的调控. 相似文献

近年来,植物所受到的诸如干旱、盐、高温、低氧、重金属胁迫和营养元素缺乏等环境胁迫越来越多,严重影响了植物的生长发育及作物的质量和产量。氮素是植物生长发育所需的必需营养元素,同时也是核酸、蛋白质和叶绿素的重要组成成分,其代谢过程与植物抵抗逆境的能力息息相关。氮代谢是指植物对氮素的吸收、同化和利用的全过程,是植物体内基础代谢途径之一。氮代谢主要从氮素吸收、同化及氨基酸代谢等方面参与植物的抗逆性,并通过调节离子吸收和转运、稳定细胞形态和蛋白质结构、维持激素平衡和细胞代谢水平、减少体内活性氧(reactive oxygen species,ROS)生成以及促进叶绿素合成等生理机制来影响植物抵抗非生物胁迫的能力。因此,提高植物在逆境下的氮代谢水平是减轻外界胁迫对其损伤的一种潜在途径。该文从氮素同化的基本途径出发,分别阐述了氮代谢在干旱胁迫、盐胁迫和高温胁迫等多个方面的逆境抵抗过程中的作用机理,为氮代谢参与植物抗逆性研究提供了有利参考。 相似文献

The enzyme guanine aminohydrolase (guanase) is inhibited by low levels of Pb2+. The inhibition is noncompetitive and the Ki is 3.0 X 10(-6) M. The only other heavy metals that are inhibitory at low concentrations are Ag+, which is 36% more, and Hg2+, which is about 50% less inhibitory than Pb2+. The inhibition of guanase by Pb2+ and Hg2+ is synergistic and the inhibition of the enzyme was readily reversed by EDTA. The relationship of these studies with guanase and to the etiology and treatment of saturnine gout, which appears in humans suffering from lead poisoning, is discussed. 相似文献

This study was carried out to determine the effect that seasonal changes have on the effect of localised cold stress on peripheral temperatures using the foot immersion method with a cold water bath. The subjects were six males and four females. The data were obtained in April, July, October and January. Skin temperature of the right index finger, the forehead, the arm, the cheek, the second toe and the instep were measured before, during and after the immersion of the feet in water at 15°C for 10 mins, as well as oxygen consumption before immersion of the feet.The average finger temperature was highest during foot immersion in the summer, next highest in the winter, then spring, and the lowest during foot immersion in the autumn. The finger temperatures during the pre-immersion period in the autumn tended to be lower than in other seasons. The finger temperatures during the pre-immersion period affected the temperature change of the finger during the immersion period. The rate of increase of the toe temperature and the foot temperature during post-immersion in the summer and the spring were greater than those in the autumn and winter. Oxygen consumption during the pre-immersion period in the autumn was significantly lower than in the other seasons (p<0.001 or 0.010). Cooling the feet caused no significant changes in the temperatures the cheek, forehead or forearm. The cheek temperature in the summer and autumn was cooler than corresponding temperatures taken in the winter and spring. 相似文献

Summary Ten alfalfa cultivars were used to study the effects of nodulation and variations in resistance to alfalfa sickness. The alfalfa
seedlings were planted in sick soil with three treatment,,i.e.: pasteurized sick soil inoculated with Rhizobium ‘Nitragin’ which served as the control, the inoculated non-pasteurized sick
soil and the non-inoculated non-pasteurized sick soil. None of the alfalfa cultivars were immune from the sickness. Cultivar
‘Anik’ fromMedicago falcata was among the most resistant cultivars. Three Phytophthora root rot resistant cultivars including ‘Agate’, ‘Apollo’ and ‘Ramsey’
were not resistant to the disease. Alfalfa inoculated with Rhizobium showed greatly improved seedling growth. Correlation
coefficients showed that those alfalfa cultivars more resistant to alfalfa sickness produced more dry weight. Dry weight increase
due to nodulation (82%) had more than compensated for the loss of dry weight due to alfalfa sickness (33%). The present study
suggested that the poor growth of alfalfa on sick soil was attributed to both the soil borne pathogens and the poor nodulation
of alfalfa, probably due to the absence of effective Rhizobium in sick soil. Highly significant differences were also obtained
among olfalfa cultivars for plant dry weight in the inoculation treatment. Selection for effective Rhizobium strains and for
alfalfa genotypes which are resistant to alfalfa sickness and are high in nitrogen fixation rates could improve alfalfa yield
