  • Genome size evolution and its relationship with pollen grain size has been investigated in sweet potato (Ipomoea batatas), an economically important crop which is closely related to diploid and tetraploid species, assessing the nuclear DNA content of 22 accessions from five Ipomoea species, ten sweet potato varieties and two outgroup taxa.
  • Nuclear DNA amounts were determined using flow cytometry. Pollen grains were studied using scanning and transmission electron microscopy.
  • 2C DNA content of hexaploid I. batatas ranged between 3.12–3.29 pg; the mean monoploid genome size being 0.539 pg (527 Mbp), similar to the related diploid accessions. In tetraploid species I. trifida and I. tabascana, 2C DNA content was, respectively, 2.07 and 2.03 pg. In the diploid species closely related to sweet potato e.g. I. ×leucantha, I. tiliacea, I. trifida and I. triloba, 2C DNA content was 1.01–1.12 pg. However, two diploid outgroup species, I. setosa and I. purpurea, were clearly different from the other diploid species, with 2C of 1.47–1.49 pg; they also have larger chromosomes. The I. batatas genome presents 60.0% AT bases.
  • DNA content and ploidy level were positively correlated within this complex. In I. batatas and the more closely related species I. trifida, the genome size and ploidy levels were correlated with pollen size. Our results allow us to propose alternative or complementary hypotheses to that currently proposed for the formation of hexaploid Ipomoea batatas.

Summary Tetraploid F1 hybrids between Ipomoea batatas, sweet potato (2n = 6x = ca. 90), and diploid (2n = 2x = 30) I. trifida (H. B. K.) Don. showed various degrees of fertility reduction. The present study aimed to clarify its causes by cytological analysis of meiotic chromosome behavior in the diploid and sweet potato parents and their tetraploid hybrids. The diploid parents showed exclusively 15 bivalents, and the sweet potato parents exhibited almost perfect chromosome pairing along with predominant multivalent formation. Their hybrids (2n = 4x= 57–63) formed 2.6–5.0 quadrivalents per cell, supporting the autotetraploid nature. The meiotic aberratios of the hybrids were characterized by the formation of univalents, micronuclei, and abnormal sporads (monad, dyad, triad, and polyad). The causes underlying these aberrations were attributed in part to the multivalent formation, and in part to a disturbance in the spindle function. Three hybrids showing serious meiotic aberrations were very low in fertility. The utilization of the sweet potato-diploid I. trifida hybrids for sweet potato improvement is described and, further, the role of interploidy hybridization in the study of the sweet potato evolution is discussed.  相似文献

  • 1 The sweet potato butterfly Acraea acerata is an indigenous species in Ethiopia that has become a major pest on the introduced sweet potato Ipomoea batatas. To assess the role of wild Ethiopian Ipomoea species as host plants, the presence of larvae on wild ipomoeas was studied, and female oviposition choice and larval performance were tested on five wild ipomoeas, as well as on sweet potato.
  • 2 In laboratory tests, oviposition and larval development were successful on two wild ipomoeas (Ipomoea tenuirostris and Ipomoea cairica) but no oviposition occurred on the remaining three species. Of the latter, larvae did not feed on Ipomoea hochstetteri and Ipomoea indica, and survival rates were extremely low on Ipomoea purpurea.
  • 3 Sweet potato was a better host plant than I. tenuirostris and I. cairica in terms of oviposition preference, larval survival and pupal size; pupae were larger, resulting in more fecund female butterflies.
  • 4 In the wild butterfly populations were abundant on I. tenuirostris but absent on I. cairica. Females also tended to prefer I. tenuirostris to I. cairica in oviposition choice experiments. However, no significant differences in performance were found between larvae raised on I. tenuirostris and I. cairica in the laboratory.
  • 5 Wild populations of A. acerata also existed on Ipomoea obscura, a plant not investigated in the present study.
  • 6 The abundance of A. acerata on wild ipomoeas is too low to likely affect butterfly population densities on sweet potato. However, wild populations may act as reservoirs subsequent to butterfly population bottlenecks on sweet potato.

Summary More than 28,000 pollinations were carried out between 5 Ipomoea batatas and 41 diploid I. trifida accessions of diverse origins to obtain 4x interspecific hybrids. From the resultant 730 seeds, 248 plants were finally obtained. Ploidy level determination of the progeny showed unexpected results: 52 individuals were hexaploid, 5 were pentaploid, 190 were tetraploid, as expected, and one was not determined. The existence of 5x and 6x progenies from 6x x 2x crosses not only confirmed the presence of 2n gametes but also their successful function in gene flow between ploidy levels and polyploidization within this genus. The progeny and their cultivated parents were planted in an observation field. The cultivated parents produced 0.49 kg/plant or less. Most 4x progenies did not produce storage roots or had very poor yields; nonetheless, and despite their cultivated parents' poor yields, 8 genotypes yielded between 0.81 and 1.50 kg/plant.A new scheme, using the 4x interspecific hybrids, is proposed for evaluating 2x and 4x wild accessions of the section Batatas to which the sweet potato belongs. Other possible uses of the 4x hybrids in breeding and genetics of the sweet potato are also discussed.  相似文献

Experiments were conducted to asymmetrically fuse protoplasts from sweet potato (Ipomoea batatas L. Lam.) and its wild relativesI. trifida Don. andI. lacunosa L. Protoplasts of sweet potato were treated with iodoacetamide, whereas those ofI. trifida Don. andI. lacunosa L. were irradiated with X-rays. The asymmetric protoplast fusion was carried out by the electrofusion method and by polyethylene glycol treatment. Electrically-fused protoplasts initiated cell division, and then formed calli earlier than the polyethylene glycol-fused protoplasts. Plant regeneration occurred only in electrofused calli, suggesting that polyethylene glycol had some toxic effect on plant regeneration ability. Analysis of peroxidase isozymes confirmed the interspecific hybrid characteristics of both the fusion-derived calli and regenerated plants.  相似文献

A protoplast-to-plant regeneration system has been established for sweet potato (Ipomoea batatas (L.) Lam.) and its wild relative, I. lacunosa L. Viable protoplasts, isolated from preplasmolyzed stems and petioles of in vitro-grown plants, were cultured on liquid MS (Murashige & Skoog 1962) medium that supported cell division and colony formation. Embryogenic calli of sweet potato were induced on agar-solidified MS medium supplemented with 3% (w/v) sucrose, 50 mg l-1 casamino acids, 0.2–0.5 mg l-1 2,4-d, 1.0 mg l-1 kinetin and 1.0 mg l-1 ABA. On average, 3 plants were regenerated from a single sweet potato callus subcultured on semi-solid MS medium containing 3% (w/v) sucrose, 800 mg l-1 glutamine, 2.0 mg l-1 BA or 1.0 mg l-1 kinetin and 1.0 mg l-1 GA3. Embryogenic calli of I. lacunosa L. were initiated on semi-solid MS medium containing 0.2–0.5 mg l-1 IAA and 1.0–2.0 mg l-1 BA. A whitefly-transmitted infectious agent, associated with geminate particles, induced distinct symptoms on several Ipomoea species, but not on I. batatas cv. Georgia Jet. The virus was transmitted by Bemisia argentifolii in a persistent manner and by grafting, but not mechanically. No transmission to species outside Ipomoea was obtained. Extracts from infected Ipomoea plants hybridised with a bean yellow mosaic virus riboprobe and a tomato yellow leaf curl virus riboprobe, although not so strongly as hybridisation of these riboprobes with extracts from plants infected with the homologous viruses. Based on host range, we consider this virus to be distinct from sweet potato leaf curl virus reported from the Far East, and propose it be named “Ipomoea crinkle leaf curl virus” (ICLCV).  相似文献

A full-length cDNA of Rubisco activase (IBrcaI) was cloned from sweet potato (Ipomoea batatas (L.) Lam) using Rapid-Amplification of cDNA Ends (RACE). IBrcaI contains a 1,347 bp open reading frame encoding a protein of 439 amino acids. The sequence alignment of multiple Rubisco activase genes from sweet potato and other plants showed high homology at two previously described ATP-binding sites. Western blot analysis indicated that there are two Rubisco activase proteins in sweet potato. Expression of IBrcaI was only detected in leaves. In the 14 h light and 10 h dark photoperiods, maximal and minimal IBrcaI mRNA expression levels were detected at 8:00 in the morning and at midnight, respectively.  相似文献

Investigations on interspecific crossability in 8Cucumis species (2n = 24) and chromosome pairing and pollen fertility of their hybrids from 15 combinations have been utilized for tracing the phylogenetic relationships among these taxa and factors responsible for their differentiation. A collective evaluation of data suggests that there are three broad groups of species, one of the spiny fruited interfertile species, whose hybrids show varying degree of chromosome associations and low to high pollen fertility; the second of species with non-spiny fruits, which are completely incompatible with the former but weakly compatible with the cultivated species,C. melo L. to produce partly developed seeds, and the third group ofC. metuliferus E. Mey. exSchrad. andC. melo and its different botanical varieties. Summary Attempts were made to obtain intergeneric hybrids between Diplotaxis siifolia, a wild species, and cultivars of Brassica (B. campestris, B. juncea, and B. napus). The crosses showed unilateral incompatibility. When the wild species was used as female parent, pollen germination and pollen tube growth were normal, but hybrid seeds aborted due to post-fertilization barriers. Reciprocal crosses (cultivars as female parent) showed strong pre-fertilization barriers; although pollen grains showed germination, pollen tubes failed to enter the stigma. Hybrids were realized in two of the crosses, D. siifolia x B. juncea and D. siifolia x B. napus, through ovary culture. The hybrids were multiplied in vitro by multiplication of axillary shoots, or somatic embryogenesis. Detailed studies were carried out on the hybrid D. siifolia x B. juncea. F1 hybrids had shrivelled anthers and were pollen sterile. Amphiploids of this hybrid showed 60% pollen fertility and produced seeds upon self-pollination as well as backcross pollination with the pollen of B. juncea.  相似文献

Sweet potato (Ipomoea batatas) is one of the most important crops in the world, and its production rate is mainly decreased by the sweet potato virus disease (SPVD) caused by the co-infection of sweet potato chlorotic stunt virus (SPCSV) and sweet potato feathery mottle virus. However, methods for improving SPVD resistance have not been established. Thus, this study aimed to enhance SPVD resistance by targeting one of its important pathogenesis-related factors (i.e., SPCSV-RNase3) by using the CRISPR-Cas13 technique. First, the RNA targeting activity of four CRISPR-Cas13 variants were compared using a transient expression system in Nicotiana benthamiana. LwaCas13a and RfxCas13d had more efficient RNA and RNA virus targeting activity than PspCas13b and LshCas13a. Driven by the pCmYLCV promoter for the expression of gRNAs, RfxCas13d exhibited higher RNA targeting activity than that driven by the pAtU6 promoter. Furthermore, the targeting of SPCSV-RNase3 using the LwaCas13a system inhibited its RNA silencing suppressor activity and recovered the RNA silencing activity in N. benthamiana leaf cells. Compared with the wild type, transgenic N. benthamiana plants carrying an RNase3-targeted LwaCas13a system exhibited enhanced resistance against turnip mosaic virus TuMV-GFP and cucumber mosaic virus CMV-RNase3 co-infection. Moreover, transgenic sweet potato plants carrying an RNase3-targeted RfxCas13d system exhibited substantially improved SPVD resistance. This method may contribute to the development of SPVD immune germplasm and the enhancement of sweet potato production in SPVD-prevalent regions.  相似文献

During 2010–2011, a severe leaf spot disease of sweet potato (Ipomoea batatas) was found in Haikou City, Hainan province of China. The disease is characterized with large, irregular, brown, necrotic lesions on the margin or in the centre of leaves. A species of Stemphylium was consistently recovered from pieces of symptomatic tissues on PDA. Based on morphological characteristics and molecular identification by rDNA‐ITS gene analysis, the fungal species was identified as Stemphylium solani Weber, and its pathogenicity was confirmed by Koch's postulates. This is the first report of leaf spot on sweet potato caused by Ssolani in China.  相似文献

Fusarium wilt, a disease caused by Fusarium oxysporum f.sp batatas (Fob) is an important disease in sweet potato production. Using endophytic bacteria for biological control of sweet potato diseases is one of the important ways. A Bacillus subtilis with antagonistic effect on Fusarium wilt of sweet potato was isolated from soil by confrontation culture. According to the biological characteristics, 16S rDNA sequence analysis, and physiological and biochemical analysis, the Bacillus subtilis HAAS01 was named. A pot experiment was conducted for the biological control experiment of strain HAAS01, and the endogenous hormone content, antioxidant enzyme activity, soluble protein content, and related gene expressions of sweet potato plants were detected. The results showed that the HAAS01 strain could promote the production of endogenous hormones and resist the infection of plant diseases together with defensive enzymes and upregulation of related gene expressions. In summary, Bacillus subtilis HAAS01 was effective in controlling Fusarium wilt of sweet potato and has potential for application and development.  相似文献

The biological containment of the potato (Solanum tuberosum) was assessed by establishing the crossability of this tuberous crop with the related wild non-tuberous species in The Netherlands, black nightshade (S. nigrum) and bittersweet (S. dulcamara). To circumvent crossability barriers, genotypes with different ploidy number were employed and crosses were performed under different environmental conditions. S. dulcamara was shown to be incongruent with potato at all ploidy levels, while S. nigrum displayed unilateral incompatibility. If S. nigrum was emasculated and used as female, fertilization by potato pollen resulted in berry set and seed development. Emasculation of S. nigrum was essential in this cross, because analysis of the fertilization process demonstrated that this species is highly self-compatible and potato pollen was outcompeted by pollen of S. nigrum. The hybrid seeds derived from this cross did not mature and appeared not to be viable. By application of the technique of embryo rescue of immature embryos, hybrid plants could be obtained. However, these hybrid plants proved to be sterile. These data demonstrate that gene flow by pollen dispersal from potato to its most common wild relatives in Western Europe is highly unlikely. The potato is thus a naturally contained species in this part of the world.  相似文献

The cDNA of the touch-induced genes (TCH) of the sweet potato [Ipomoea batatas (L.) Lam.] has been cloned and analyzed. IbTCH1, which exists as at least two-copy genes in the genome of the sweet potato, encodes for 148-amino acid polypeptides, and harbors four conversed Ca2+-binding motif EF-hands. IbTCH1 was shown to be expressed in the flower, leaf, thick pigmented root, and particularly in the white fibrous root, but expressed only weakly in the petiole. IbTCH1 is upregulated upon exposure to environmental stresses, dehydration, and jasmonic acid. Furthermore, IbTCH1 is developmentally regulated in the leaf and root. These results strongly indicate that the gene performs functions in both plant development and in defense/stress-signaling pathways.  相似文献

 True potato seed (TPS) may be an alternative method of potato production in developing countries. A breeding method for the sexual propagation of this vegetatively propagated crop should consider the development of parental lines and the type of cultivar to be released. Open-pollinated (OP) cultivars seem to be an inexpensive procedure to produce potato from true seed. However, OP progenies are the result of selfing and outcrossing in male-fertile tetraploid potatoes. The aim of the present research was to establish the effect of inbreeding and open pollination in TPS. Ten Andigena clones were used as parental material to derive hybrid (S0), inbred (S1 and S2), and open-pollinated (OP1 and OP2) generations. Significant differences among generations were found for pollen production, pollen viability (as determined by its stainability with aceto-carmine glycerol), number of flowers and berries plant-1, number of seeds berry-1, weight of 1000 seeds, and tuber yield plant-1. The parental populations were significantly different for most of the traits, but not for flower production and berry weight. The interaction of population ×generation was significant for pollen and seed production as well as for weight for 1000 seeds. All the traits evaluated except seed weight showed a strong inbreeding depression, while the OP progenies had intermediate values between the S0 and the S1. This demonstrates that open pollination in potatoes is not exclusively the product of selfing; it also results from outcrossing. Received: 10 November 1997 / Accepted: 3 March 1998  相似文献

Summary Intergeneric hybrids were produced between Diplotaxis siettiana and Brassica campestris through embryo rescue. The hybrids were completely pollen sterile and backcrosses with pollen of B. campestris did not yield any seeds. Induction of colchiploidy restored pollen fertility and backcross pollinations yielded viable seeds. Cytological details of the hybrid, amphidiploid and backcross progenies were studied. Both pollen-sterile and pollen-fertile plants have been obtained in backcross 2 progeny. This hybrid (D. siettiana x B. campestris) was used as a bridge cross to transfer the cytoplasm of D. Siettiana to two other incompatible cultivars of BrassicaB. juncea and B. napus. Pollinations of the amphidiploid (D. siettiana x B. campestris, 2n = 36) with pollen of B. juncea/B. napus readily produced seeds without embryo rescue. These hybrids were grown to flowering and their cytological details were studied. Seeds have been produced from backcross pollinations of both these hybrids with the pollen of the respective cultivars. The results clearly show the feasibility of producing alloplasmic lines in all the three oilseed brassicas.  相似文献

GAl5, GA3, GA5, GA19, GA20 and GA23 were identified by GC-MS in the acidic ethyl acetate-soluble fraction from the seeds of sweet potato (Ipomoea batatas Lam.). GA19 and GA23 were major GAs in the mature seeds, their contents being about 200 and 160 μg/kg fresh weight, respectively, while those of GA19 and GA23 in immature seeds were below 100 μg/kg fr. wt. The occurrence of glycosyl conjugates of GA3, GA5, GA8, GA17, GA19, GA20, GA23 and GA44 in the butanol fraction from mature seeds was shown by GC/MS analysis after enzymatic hydrolysis.

Besides the endogenous GAs in sweet potato, those in immature seeds of several other Convolvulaceae plants were investigated. The species of endogenous GAs were discussed in terms of chemotaxonomy.  相似文献

Cytogenetical studies were carried out on the successive generations of offsprings from the induced tetraploid hybrid (2n = 32) betweenRanunculus silerifolius (2n = 16) andR. chinensis (2n = 16). Aneuploids, 2n = 30 to 35, frequently occurred. In latter subsequent generations the deviation of aneuploids increased, but the proportion of euploids decreased, accompanied by the reduction of fertility of pollen grains and seed sets. F2 and F4 PMCs constantly exhibited meiotic abnormality, i.e. formation of quadrivalents and univalents.
