The effects of intercycle cover crops on Rotylenchulus reniformis population densities in pineapple were evaluated in one greenhouse and two field experiments. In the greenhouse, Crotalaria juncea, Brassica napus, and Tagetes erecta were planted for 3 months and then incorporated. These treatments were compared to weedy fallow with or without 1,3-dichloropropene (1,3-D) in three soils (Makawao fallow, Wahiawa fallow, and Wahiawa pineapple) naturally infested with R. reniformis. All cover crop incorporation suppressed R. reniformis numbers in cowpea more than did the weedy treatment in the Makawao (P < 0.05) but not in the Wahiawa soils. Crotalaria juncea treatment increased bacterivorous nematodes and nematode-trapping fungal population densities more than the other treatments in Makawao fallow and Wahiawa pineapple-planted soils. The field trials included the same plants as well as Sinapis alba. Treatments with Crotalaria juncea and 1,3-D maintained lower R. reniformis population densities on pineapple longer than other cover crops or weedy fallow treatments. Crotalaria juncea could have suppressed R. reniformis because it is a poor host and because it enhances nematode-trapping fungi when incorporated into soil. An ultrastructural study of dry and wet trichomes from the upperleaf surface of pineapple revealed that shield cells were normallydead and stalk cells were often living. In stalk cells of drytrichomes, polysaccharide material was present within vacuolesand a layered material was present between the plasma membraneand cell wall. In stalk cells of trichomes wet for 12 h vacuoleswere enlarged and appeared to contain little material. Layeredmaterial between the plasma membrane and cell wall was alsoabsent. In both wet and dry trichomes, numerous mitochondriawere present and plasmodesmata were observed connecting stalk,foot, and adjacent epidermal and mesophyll cells. It is suggestedthat the large numbers of mitochondria, which indicate highmetabolism, and the numerous plasmodesmata connecting the cells,provide indirect evidence for the function of the trichomesin uptake of dissolved nutrients. A procedure for the mass propagation of pineapple plants (Ananas comosus L. Merr) using a temporary immersion technique is described. This procedure involved three distinct phases in the automated
temporary immersion system: shooting, bud differentiation and elongation. To establish this protocol, we used in vitro shoots
obtained from established liquid culture as starting materials. Three culture methods (solid, liquid and temporary immersion)
were compared. Temporary immersion increased the multiplication rate and fresh and dry weight after 42 days. Conventional
micropropagation (liquid medium) and temporary immersion were compared in combination with paclobutrazol. Paclobutrazol promoted
the formation of compact bud clusters with limited leaf development. The highest multiplication rate (106) was found when
ex-plants were cultured in shooting medium (MS+2.1 mg/l BA+0.3 mg/l NAA) supplemented with 1 mg/l PB for 7 weeks. A 10-l temporary
immersion bioreactor was used to test two approaches during elongation stage: reduction of the shoot-formation period or decrease
of the initial number of explants. The highest number of competent and uniform plants (191.8 plant/l) was achieved when shoots
were cultured for 4 weeks in shooting medium supplemented with PB.

Multiple shoots were regenerated from leaf explants obtained from in vitro grown shoot cultures of pineapple. Each leaf was horizontally cut into three pieces (~ 0.5 cm, basal, middle and tip) and cultured onto MS basal medium supplemented with 2% sucrose and various growth regulators.The explant containing the basal part of the leaf gave rise to tiny protuberances which grew into shoots.The highest number of shoots were obtained on MS basal medium supplemented with 2,4-D (0.90 µM) and 2iP (0.98 pM).These shoots were subcultured ontowhite’s basal medium supplemented with 1% sucrose, NAA (0.54 µM) and IBA (1.97 µM). Plantlets produced in vitro were transferred to paper cups containing autoclaved soil or Soilrite, hardened in the greenhouse and established in soil.The protocol provides an easy propagation system for pineapple, an otherwise vegetatively propagated fruit crop. 相似文献

The effects of temperature, O2, and CO2 on titratable acid content and on CO2 exchange were measured in detached pineapple (Ananas comosus) leaves during the daily 15-hour light period. Comparative measurements were made in air and in CO2-free air. Increasing the leaf temperature from 20 to 35 C decreased the total CO2 uptake in air and slightly increased the total CO2 released into CO2-free air. Between 25 and 35 C, the activation energy for daily acid loss was near 12 kcal mol−1, but at lower temperatures the activation energy was much greater. 相似文献

Characterization of Phosphoenolpyruvate Carboxykinase from Pineapple Leaves Ananas comosus (L.) Merr
Phosphoenolpyruvate carboxykinase has been partially purified from pineapple (Ananas comosus [L.]) leaves. Specific activities obtained show it to be a major activity in this tissue. Above 15 C, the respective activation energies for decarboxylation and carboxylation are 13 and 12 kcal/mol. Below 15 C, there are discontinuities in Arrhenius plots with an associated large increase in activation energy. The adenine nucleotides are preferred to other nucleotides as substrates. The apparent Km values in the carboxylation direction are: ADP 0.13 mm, HCO(3) (-) 3.4 mm, and phosphoenolpyruvate 5 mm. In the decarboxylation direction, the apparent Km values are: ATP 0.02 mm, ADP 0.05 mm, and oxaloacetate 0.4 mm. The decarboxylation activity had an almost equal velocity with either ADP or ATP. The pH optima are between 6.8 and 7. Inhibition of the carboxylation reaction by ATP, pyruvate, and carbonic anhydrase was demonstrated. Decarboxylase specific activities are over twice carboxylation activities. The data support a model in which phosphoenolpyruvate carboxykinase is of physiological significance only during the light period and then only as a decarboxylase. 相似文献

以陕西杨凌地区主栽的番茄品种'金棚1号'为试验材料,通过固相微萃取和GC/MS联用技术,对番茄果实不同成熟阶段的香气成分及其组成变化进行了研究.结果表明,'金棚1号'番茄果实共检测到54种香气成分,主要成分为醛类、酸类、醇类、酮类、酯类、酚类等.在果实的不同发育阶段,香味组分及其含量差异较大.醛类物质在绿熟期相对含量较高,为45.87%,在半熟期、硬熟期、完熟期的相对含量分别为12.65%、16.62%、17.15%,其中C6醛在绿熟期占43.7%,完熟期占15.27%,为醛类物质的主要成分;酸类物质含量在4个发育时期中先上升后下降,在半熟期含量达到最高,为15.2%,在完熟期酸类物质含量下降,为6.93%;酮类物质在完熟期含量达到最大,为18.27%;在绿熟期检测到4种重要的番茄特征香气物质,半熟期检测到5种番茄特征香气物质,硬熟期和完熟期各检测到6种番茄特征香气物质.说明随着果实的成熟,特征香气物质种类增多. 相似文献

PAM (Pulse Amplitude Modulation) fluorometer techniques directly measure the light reactions of photosynthesis that are otherwise difficult to estimate in CAM (Crassulacean Acid metabolism) plants such as pineapple (Ananas comosus comosus cv. Phuket). PAM machines calculate photosynthesis as the Electron Transport Rate (ETR) through PSII (4 electrons per O2 produced) as mol m?2 s?1. P vs. E curves fitted the waiting-in-line function (an equation of the form $ {\hbox{ETR}} = \left( {{\hbox{ET}}{{\hbox{R}}_{{ \max }}} \times {\hbox{E}}/{{\hbox{E}}_{\rm{opt}}}} \right).{{\hbox{e}}^{{1} - {\rm{E}}/{\rm{Eopt}}}} $ ) allowing half-saturating and optimal irradiances (Eopt) to be estimated. Effective Quantum Yield (Ymax), Electron Transport Rate (ETRmax) and the Non-Photochemical Quenching parameter, NPQmax all vary on a diurnal cycle but the parameter qNmax does not show a systematic variation over a diurnal period. Phuket pineapple is a “sun plant” with Optimum Irradiance (Eopt) from 755 to 1,130 μmol m?2 s?1 (400–700 nm) PAR but photosynthetic capacity is very low in the late afternoon even though light conditions are favourable for rapid photosynthesis. Total CO2 fixed nocturnally as C4-dicarboxylic acids by leaves of the Phuket pineapple was only ≈0.14 gC m?2 d?1 (0.012 mol C m?2 d?1). Titratable acid of leaves was depleted about 3 pm (15:00) and shows a classical CAM diurnal cycle. The Phuket pineapple variety only stored enough CO2 as C4 acids to account for only about 2.5% of photosynthesis (Pg) estimated using the PAM machine (≈5.6 gC m?2 d?1). Phuket pineapples are classifiable as CAM-Cycling plants but nocturnal fixation of CO2 is so low compared to the more familiar Smooth Cayenne variety that it probably recycles only a small proportion of the respiratory CO2 produced in leaves at night and so even CAM-cycling is only of minor importance to the carbon economy of the plant. Unlike the Smooth Cayenne pineapple variety, which fixes large amounts of CO2 nocturnally, the Phuket pineapple is for practical purposes a C3 plant. 相似文献

Ethylene-releasing agents such as ethephon (2-chloroethylphosphonic acid) are used widely to induce flowering in pineapples
(Ananas comosus (L.) Merrill). However, ethephon treatment is less reliable in summer, particularly if plants are treated on abnormally hot
days. [14C]ethephon was used to follow uptake and translocation in leaf tissues. Up to 30% of the ethephon entered the leaf within
4 h, and up to 60% by 24 h. Uptake was dramatically modified by temperature, relative humidity, solution pH, and the surface
on which solution droplets were placed. Entry occurred across the leaf cuticle and probably also by way of stomatal pores,
and label was recovered at all depths within the leaf. 14C label entered more rapidly through the abaxial epidermis than through the adaxial epidermis. Low-volume spray applications
to whole plants resulted in rapidly drying droplets mainly on the adaxial, distal epidermis and were rather ineffective at
inducing flowering, possibly because little ethephon or ethylene reaches the shoot apex. High-volume sprays may facilitate
ethephon entry because solution accumulates in leaf axils and hence remains in prolonged contact with abaxial epidermis of
leaf bases close to the shoot apex. When poured into the center of the plant, 20% of a normal commercial ethephon dose induced
full flowering even under adverse temperatures. It is suggested that high-volume evening spraying and avoidance of hot days
may reduce the incidence of flowering failure.

Penicillium funiculosum Thom. was consistently isolated from pineapple-infected fruitlet (black spots). Polyphenol oxidase, peroxidase, and laccase activities were determined in extracts from contiguous and infected fruitlets. Healthy fruitlets showed a rather high level of polyphenol oxidase (optimum pH 7.0), and this activity was tremendously increased (×10) in contiguous infected fruitlets. Furthermore, infected fruitlets also exhibited laccase activity (optimum pH 4.0), while peroxidase was rather constant in both fruitlets. Browning reactions were attributed to qualitative and quantitative modifications of the enzymatic equipment (polyphenol oxidase and laccase) (p<0.0001). In infected fruiltets, sucrose and L-malic acid were present at significantly lower amounts than in healthy ones, likely owing to fungal metabolism (p<0.0001), whereas cell wall material was three times higher, which could be viewed as a defense mechanism to limit expansion of the mycelium. 为了分析不同肉色类型桃果实发育过程中类胡萝卜素成分的变化特点及其颜色差异形成的因素,选择‘半斤桃’(红肉)、‘图八德’(黄肉)、‘正姬’(白肉)桃品种为试材,采用紫外分光光度计和高效液相色谱法(HPLC)对桃果实类胡萝卜素及其组分含量进行定性定量分析.结果显示:(1)桃果实的主要类胡萝卜素成分为叶黄素、玉米黄素、β-隐黄质、α-胡萝卜素和β-胡萝卜素.(2)随着果实的发育,‘半斤桃’和‘正姬’的类胡萝卜素总含量逐渐降低,而‘图八德’则是先降低后上升,且明显高于前二者;3种类型桃果实的叶黄素均逐渐降低,果实成熟时接近0;玉米黄素呈先降后升的趋势,果实成熟时达到最大值;‘半斤桃’和‘正姬’的β-隐黄质、α-胡萝卜素和β-胡萝卜素呈逐渐下降的趋势,而‘图八德’的β-隐黄质含量先升后微降最后上升、α-胡萝卜素和p-胡萝卜素含量则逐渐增加,且均在果实成熟时达到最大值.(3)‘图八德’成熟果实中的类胡萝卜素总含量达到最大值,其中β-胡萝卜素含量最高,占5个成分之和的75.99%.研究表明,桃果实肉色形成与类胡萝卜素的含量与成分有着紧密的联系,尤其是其中的β-胡萝卜素含量. 相似文献

MADS-box转录因子在植物的发育过程特别是控制花器官的诱导与发育中起关键作用。利用同源克隆结合RACE技术, 从菠萝(Ananas comosus)花中分离出1个新的菠萝MADS-box基因, 命名为AcMADS1(GenBank登录号为KC257408)。AcMADS1基因的编码区为726 bp, 编码241个氨基酸, 蛋白质分子量为27.50 kDa, 等电点为9.26。序列比对和系统进化树分析表明, AcMADS1具有保守的MADS-box及半保守的K区, 属于AGL6亚家族MADS-box蛋白。生物信息学分析表明, AcMADS1是亲水碱性蛋白, 二级结构主要以α-螺旋、无规则卷曲和折叠延伸链为蛋白质骨架, 三级结构中蛋白核心结构符合转录因子与DNA结合的常见功能域MADS-box, 而且作为转录因子定位于细胞核中。组织特异性表达分析表明, AcMADS1基因在菠萝果肉以及花器官的雌蕊、花瓣和萼片中均有表达, 但在雄蕊以及营养器官的根、茎和叶中几乎不表达; 且在花器官早期发育过程中大量表达, 后期呈下降趋势。因此推测这个基因可能在菠萝花器官发育和开花诱导过程中起重要作用。 相似文献

利用SCo T分子标记技术对来自9个国家或地区的46份菠萝种质进行了遗传多样性分析,并对SCo T标记在菠萝研究中的效率做了探讨。结果表明,SCo T标记在菠萝种质中具有丰富的多态性,引物多态性条带百分比在75%~100%之间,平均为94.61%;引物的有效等位基因数(Ne)、Nei's基因多样性指数(H)、Shannon's信息指数(I)和多态性信息含量(PIC)平均值分别为1.45、0.27、0.41和0.28,表明SCo T标记具有较高的多态性检测效率。基于SCo T标记计算获得的遗传相似系数对菠萝种质做聚类分析,46份菠萝种质可被划分为5个类群,其中,第Ⅰ类群所包含的菠萝种质数量最多,占菠萝种质总数的84.78%。主成分分析获得了与聚类分析不尽一致的结论,但两者反映的种质亲缘关系基本一致。本研究结果将为我国菠萝种质的鉴定、保存和科学利用提供一定的理论依据。 相似文献

Present studies on the community characteristics of earthworms revealed the occurrence of 11 species of earthworms in the pineapple (Ananus comosus) and 14 species in the mixed fruit plantations of west Tripura (India). While 9 species of earthworms namely Drawida assamensis, Drawida papillifer papillifer, Drawida nepalensis, Kanchuria sp., Metaphire houlleti, Eutyphoeus gigas, Eutyphoeus scutarius, Eutyphoeus comillahnus and Pontoscolex corethrurus are of common occurrence to both the pineapple and the mixed fruit plantations, two and five earthworm species namely Kanchuria sumerianus, Eutyphoeus sp. and Metaphire posthuma, Perionyx excavatus, Lampito mauritii, Amynthus alexandri, Eutyphoeus gammiei are restricted to the pineapple and the mixed fruit plantations respectively. Earthworms were found mostly within 15 cm depth of soils having temperature 25–25.8 °C, moisture 18.8–22.4 %, water holding capacity 26–31.7 % and organic matter content 2.4–4.0 %. Mean earthworm densities (158 ind. m?2) was significantly higher (p < 0.01, t = 9.67) and biomass (36.67 g m?2) significantly lower (p < 0.01, t = ?5.98) in the pineapple plantation than the mixed fruit plantation (density 93 ind. m?2, biomass 56 g m?2). High density value of earthworms in pineapple plantation is linked with dominance of D. assamensis and high biomass value in mixed fruit plantation was due to the higher relative abundance of larger species like E. gigas, E. scutarius, E. comillahnus and E. gammiei. Compared to the mixed fruit plantation, significantly (p < 0.05) higher index of dominance, lower index of diversity, species richness index and species evenness were recorded in the pineapple plantation. 相似文献

An increase in exocarp peroxidase activity was observed in fruit at 5 to 30 days post pollination (DPP), and decreased at 40 and 50 DPP. Total peroxidase activity of the mesocarp was significantly lower than the exocarp in all developmental stages. Mesocarp peroxidase activity decreased consecutively from outer, to middle and, to inner tissue at every developmental stage. Total activity in the mesocarp peaked at 20 DPP. Native-PAGE of exocarp tissue showed at least two cathodic (basic) peroxidases and two anionic (acidic) peroxidases. The number of isozymes was greatest and bands most intense at 30 DPP. IEF-PAGE of the 5 to 50 DPP fruit exocarp showed at least 8 peroxidase isozymes (pI 4.6 to 9.6). Anion exchange chromatography showed only one peak of anionic peroxidase activity that was not evident until 15 DPP. This peak was greatest at 30 DPP and declined at 40 and 50 DPP. Cationic peroxidase isozymes appeared to be the predominant and most intense isoforms throughout fruit development. The changes in peroxidase activity corresponded to fruit formation and may be associated with susceptibility to fruit rot. 相似文献

This study investigated the diversity and risk status of the pests associated with pineapple fruit trade from Nigeria to the U.S.A. in a commodity-initiated Qualitative Pest Risk Assessment as required under the Sanitary and Phytosanitary agreement of member nations of the World Trade Organization. The study validated in the field as necessary, all fruit pests of pineapple, Ananas comosus in the southwest Nigeria that have potential to follow the risk pathway from the farm to the port of exit. Twenty four pests, comprising 18 species of insects, 5 fungi and 1 bacterium were identified. Among these, one insect — Zonocerus variegatus was identified as a quarantine pest of pineapple with potential to follow the pathway to the pest risk areas in the U.S.A. The likelihood of introduction based on climatic conditions matching the pest etiology and host biotics is medium. The climatic requirement of the pest is compared with those of the pest risk areas in the USA such as Georgia, Texas, Florida, California, and Alabama. The pest lays its eggs usually in soil but could be hidden in the leaf bract or scale on the pineapple fruit surface. The egg undergoes 6-9 months of diapause, depending on the environment. The infestive nymphs and adults are relatively large sized. The overall consequence of introduction is high but the risk of this pest following the pathway is rated medium. The mitigation in place in Nigeria such as pre-shipment inspection, cleaning and fumigation at the port of exit appears adequate but could still be improved to completely remove the likelihood of introduction. Gamma-irradiation which is likely to become available in Nigeria has the potential to remove totally the risk of introduction of this pest to the USA. 相似文献
