The degradation of an azo dye mixture by an aerobic bacterial consortium was studied in a rotating biological reactor. Laterite pebbles of particle size 850 microm to 1.44 mm were fixed on gramophone records using an epoxy resin on which the developed consortium was immobilized. Rate of degradation, BOD, biomass determination, enzymes involved, and fish bioassay were studied. The RBC has a high efficiency for dye degradation even at high dye concentrations (100 microg/mL) and high flow rate (36 L/h) at alkaline pH and salinity conditions normally encountered in the textile effluents. Bioassays (LD-50) using Thilapia fish in treated effluent showed that the percentage mortality was zero over a period of 96 h, whereas the mortality was 100% in untreated dye water within 26 h. Fish bioassay confirms that the effluent from RBC can be discharged safely to the environment.  相似文献

A phyto-tunnel was developed using a drilled PVC pipe. It was planted with Portulaca grandiflora and used for the treatment of a textile effluent and a dye mixture. COD, BOD, TOC, conductivity, turbidity, total suspended solids and total dissolved solids of the textile effluent, and dye mixture were decreased by 57, 45, 43, 52, 76, 77 and 24 % within 96 h, and 49, 62, 41, 63, 58, 71 and 33 %, within 60 h, respectively, after treatment. The effluent and dye mixture were decolorized up to 87 and 90 % within 96 and 60 h, respectively. Significant induction in activities of lignin peroxidase, tyrosinase and DCIP reductase was observed in root tissues of the plants. FTIR, HPLC and HPTLC of untreated and treated samples showed the formation of new metabolites and preferential dye removal. Phytotoxicity studies revealed the non-toxic nature of the metabolites.  相似文献

In this study, we attempted two investigational systems: one is treatment of sago industry effluent by aerobic bacterial consortium and the other is impact of treated and untreated effluent on seed germination. For the treatment system, the starch degrading bacteria were isolated from sago industry effluent and effluent contaminated soil. The genera, Alcaligenes, Bacillus and Corynebacterium were found efficient in starch degradation. The selected isolates were tested for their efficiency on the degradation of starch both in Mineral Salts Medium (MSM) and in sago industry effluent. About 85% of the starch was degraded in MSM by a bacterial consortium composed of Alcaligenes, Bacillus and Corynebacterium, whereas in effluent the degradation of starch was only 63%. The physico-chemical properties such as electrical conductivity, total solids, suspended solids, dissolved solids, BOD, COD, nitrogen and phosphate were found decreased in effluent after 72 h. The pH of the effluent was relatively increased from 3 to 6.7. The study of seed germination (maize and green gram) was carried out at 25, 50, 75 and 100% concentrations of treated and untreated effluent using soil sowing method. Shoot length, root length, fresh weight, dry weight and chlorophyll content showed an increase when treated effluent was tested whereas a decrease of growth was noticed in untreated effluent tested seedlings. The results revealed that effluent treated by aerobic microorganisms has no negative impact on the seed germination and can be effectively used for irrigation.  相似文献

Citrobacter sp. strain KCTC 18061P was found to be able to decolorize textile plant effluent containing different types of reactive dyes. Effects of physico-chemical parameters, such as aeration, nitrogen source, glucose and effluent concentrations on the color removal of real dye effluent by this strain were investigated. The observed changes in the visible spectra indicated color removal by the absorption of dye to cells during incubation with the strain. This strain showed higher decolorization ability under aerobic than static culture conditions. With 1% glucose, this strain removed 70% of effluent color within 5 days. Decolorization was not significantly dependent on the nitrogen sources tested. Chemical oxygen demand (COD) and biological oxygen demand (BOD) were decreased in proportion to incubation times, and their removal rates were about 35% and 50%, respectively, at 7 days of culture.  相似文献

Malachite green was detoxified into p-benzyl-N,N-dimethylaniline and N,N-dimethyl-aniline hydrochloride by Penicillium ochrochloron. Degradation metabolites were analyzed by TLC, HPLC, and FTIR and identified by GCMS analysis. Phytotoxicity testing revealed the nontoxic nature of these metabolites. The percentage decolorization of malachite green (50 mg/L) was 93% in czapek dox broth after 14 h with an optimum pH of 7 at 30°C. The induction in the activity of lignin peroxidase after degradation suggested that the degradation of malachite green was peroxidase-mediated. Fungal culture was also found to have detoxified the textile effluent. The values of TDS, TSS, COD, and BOD were reduced in the treated samples compared to the control effluent. The treated effluent was non-toxic to the plants of Triticum aestivum and Ervum lens Linn, and the amount of total chlorophyll was higher in plants with treated effluent when compared to control effluent.  相似文献

The combination of anaerobic and aerobic periods in the operation cycle of a Sequencing Batch Reactor (SBR) was chosen to study biological color removal from simulated textile effluents containing reactive, sulfonated, monoazo and diazo dyes, respectively, Remazol Brilliant Violet 5R and Remazol Black B. 90% color removal was obtained for the violet dye in a 24-h cycle with a Sludge Retention Time (SRT) of 15 days and an aerated reaction phase of 10 h. For the black dye only 75% color removal was achieved with the same operational conditions and no improvement was observed with the increase of the SRT to 20 days. For the violet dye a reduction of the color removal values from 90 to 75% was observed with the increase of the aerated reaction phase from 10 to 12 h. However, this increase did not promote the aerobic biodegradation of the produced aromatic amines. Abiotic tests were performed with sterilized SBR samples and no color removal was observed in cell-free supernatants. However color removal values of 30 and 12% were observed in the presence of sterilized cells and supernatants with violet and black dye, respectively and could be attributed to the presence of active reducing principles in the sterilized samples.  相似文献

The origin of polyploid wheat genomes has been the subject of numerous studies and is the key problem in wheat phylogeny. Different diploid species have been supposed to donate genomes to tetraploid and hexaploid wheat species. To shed light on phylogenetic relationships between the presumable A genome donors and hexaploid wheat species we have applied a new approach: the comparison of defensins from diploid Triticum species, Triticum boeoticum Boiss. and Triticum urartu Thum. ex Gandil., with previously characterized Triticum kiharae defensins [T.I. Odintsova et al., Biochimie 89 (2007) 605-612]. Defensins were isolated by acidic extraction of seeds followed by three-step chromatographic separation. Isolated defensins were identified by molecular masses using MALDI-TOF mass spectrometry and N-terminal sequencing. For the first time, we have shown that T. urartu defensins are more similar to those of the hexaploid wheat than T. boeoticum defensins, although variation among samples collected in different regions of the world was revealed. Our results clearly demonstrate that T. urartu of the Asian origin contributed the A genome to polyploid wheat species.  相似文献

Two new yeast strains (SPT1 and SPT2) were isolated and immobilized on glassy carbon electrodes to form microbial biosensors for estimation of biochemical oxygen demand (BOD). Ferricyanide was proven to be the most efficient mediator to shuttle electrons from the redox center of reduced microbial enzymes to the electrode in the presence of excess glucose/glutamic acid (GGA). With a 3-fold greater metabolic assimilation capability and greater responses to various effluent samples, SPT1 was selected for sensor-BOD measurements. BOD estimations for the GGA standard resulted in an extended linear range: 2-100 mg/l. Response reproducibility was +/-10% for a GGA standard containing 10 mg BOD/l. For analysis of pulp mill effluents, the BOD detection limit was 2 mg/l with a response time of 5 min.  相似文献

A microbial consortium capable of aerobic degradation of a mixture of azo dyes consisting of two isolated strains (RRL,TVM) and one known strain of Pseudomonas putida (MTCC 1194) was immobilized on laterite stones. The amount of bacterial biomass attached to the laterite stones was 8.64 g per 100 g of the stone on a dry weight basis. The packed bed reactor was filled with these stones and had a total capacity of 850 mL and a void volume of 210 mL. The feed consisted of an equal mixture of seven azo dyes both in water as well as in a simulated textile effluent, at a pH of 9.0 and a salinity of 900 mg/L. The dye concentrations of influent were 25, 50, and 100 microg/mL.The residence time was varied between 0.78 and 6.23 h. It was found that at the lowest residence time 23.55, 45.73, and 79.95 microg of dye was degraded per hour at an initial dye concentration of 25, 50, and 100 microg, respectively. The pH was reduced from 9.0 to 7.0. Simulated textile effluent containing 50 microg/mL dye was degraded by 61.7%. Analysis of degradation products by TLC and HPLC showed that the dye mixture was degraded to nontoxic smaller molecules. The bacteria-coated pebbles were stable, there was no washout even after 2 months, and the reactor was found to be suitable for the aerobic degradation of azo dyes.  相似文献

Osmotic priming of carrot seeds for 2 wk in polyethylene glycol (PEG, — 10 MPa) at 15 °C led to more rapid and synchronous germination at 20 °C compared to untreated seeds. These responses were enhanced by a 24 h pre-priming soak in water or a change of solution after the first 24 h of priming to remove leachate. The inclusion of 200 mg litre-1N-substituted phthalimide in the pre-priming soak and/or in the PEG further enhanced the results of priming. Leachate removal combined with phthalimide inclusion gave 79% and 86% germination from seeds of two carrot cultivars during the first day in 20°C water following priming. In contrast, cumulative germination of untreated seeds of the same cultivars was 18% and 61% respectively after 3 days in 20°C water. Seeds primed in PEG containing 200 mg phthalimide litre-1with the solution replaced after the first 24 h germinated earlier and more synchronously than untreated seeds over a range of germination temperatures (5, 10, 15, or 20°C), but the effects of priming were most marked at 5°C.  相似文献

Kraft mill effluents were biologically treated in an aerated lagoon (AL) (3.4 L) to evaluate the behavior of aromatic compounds by UV-vis spectroscopy. Under aerobic conditions, biological oxygen demand (BOD(5)) removal was between 84% and 95%, whereas soluble chemical oxygen demand (COD(s)) removal ranged between 40% and 60%. A slight increase was observed in the VIS(436)/COD(s) relationship (0.101-0.110), indicating that color reduction was less than the total organic matter reduction expressed by COD(s). Additionally, low values of UV(254)/UV(280) (1.23-1.12) indicate the presence of lignin-derived compounds in the aerobic effluent relationship. Ultrafiltration (UF) analysis shows that color was concentrated in the molecular weight fraction larger than 10,000 Da. The recalcitrant fraction was found to be resistant to further biodegradation, even under optimized microbiological conditions.  相似文献

Agriotes obscurus (L.) wireworms assembled in increasing numbers at rows of treated (Agrox DL Plus seed treatment) and untreated wheat, Triticum aestivum L., planted at increasing densities (0, 0.15, 0.30, and 0.60 seeds/cm). In treated wheat plots at all planting densities, no wireworm damage to seedlings was apparent, and total wireworms taken in core samples in wheat rows increased according to the asymptotic equation y = B0(1 -e(-Blx)), where B0 is the asymptote, B1 is the slope of the initial rise, and x is the seeding density. The number of dead wireworms in treated plots increased linearly and intercepted the asymptotic models (theoretical point at which 100% mortality of assembled population occurs) at 0.95 seeds/cm on 11 June and 1.14 seeds/cm on 18 June 1996. Untreated wheat at all densities planted had severe wireworm damage and significantly reduced stand. Populations that had assembled at the surviving untreated wheat were fewer than in the treated wheat plots, and although increasing with seeding density, did not follow the asymptotic model. The data suggest that A. obscurus populations can be assembled and killed in fallowed fields in large numbers at treated trap crops of wheat over a 19-d period when planted in rows spaced 1 m apart at a linear seeding density of 1.5 seeds/cm.  相似文献

This work shows the chemical characterization of a dye processing plant effluent that was contributing to the mutagenicity previously detected in the Cristais river, S?o Paulo, Brazil, that had an impact on the quality of the related drinking water. The mutagenic dyes Disperse Blue 373, Disperse Orange 37 and Disperse Violet 93, components of a Black Dye Commercial Product (BDCP) frequently used by the facility, were detected by thin layer chromatography (TLC). The blue and orange dyes were quantified by high performance liquid chromatography (HPLC/DAD) in a raw and treated effluent samples and their contribution to the mutagenicity was calculated based on the potency of each dye for the Salmonella YG1041. In the presence of S9 the Disperse Blue 373 accounted for 2.3% of the mutagenic activity of the raw and 71.5% of the treated effluent. In the absence of S9 the Disperse Blue 373 accounted for 1.3% of the mutagenic activity of the raw and 1.5% of the treated effluent. For the Disperse Orange 37, in the presence of S9, it contributed for 0.5% of the mutagenicity of the raw and 6% of the treated effluent. In the absence of S9; 11.5% and 4.4% of the raw and treated effluent mutagenicity, respectively. The contribution of the Disperse Violet 93 was not evaluated because this compound could not be quantified by HPLC/DAD. Mutagenic and/or carcinogenic aromatic amines were also preliminary detected using gas chromatograph/mass spectrometry in both raw and treated and are probably accounting for part of the observed mutagenicity. The effluent treatment applied by the industry does not seem to remove completely the mutagenic compounds. The Salmonella/microsome assay coupled with TLC analysis seems to be an important tool to monitor the efficiency of azo dye processing plant effluent treatments.  相似文献

A simple, efficient and cost-effective method for municipal wastewater treatment is examined in this paper. The municipal wastewater is treated using an upflow anaerobic sludge bed (UASB) reactor followed by flash aeration (FA) as the post-treatment, without implementing aerobic biological processes. The UASB reactor was operated without recycle, at hydraulic retention time (HRT) of 8 h and achieved consistent removal of BOD, COD and TSS of 60-70% for more than 12 months. The effect of FA on UASB effluent post-treatment was studied at different HRT (15, 30 and 60 min) and dissolved oxygen (DO) concentrations (low DO = 1-2 mg/L and high DO = 5-6 mg/L). The optimum conditions for BOD, COD and sulfide removal were 30-60 min HRT and high DO concentration inside the FA tank. The final effluent after clarification was characterized by BOD and COD values of 28-35 and 50-58 mg/L, respectively. Sulfides were removed by more than 80%, but the fecal coliform only by ~2 log. The UASB followed by FA is a simple and efficient process for municipal wastewater treatment, except for fecal coliform, enabling water and nutrients recycling to agriculture.  相似文献

A study was conducted to examine the potential of jute processing waste (JPW) for the treatment of wastewater contaminated with dye and other organics generated from various activities associated with jute cultivation and fibre production. Adsorption studies in batch mode have been conducted using dye solution as an adsorbate and JPW as an adsorbent. A comparative adsorption study was made with standard adsorbents such as powdered and granular activated carbon (PAC and GAC, respectively). A maximum removal of 81.7% was obtained with methylene blue dye using JPW as compared to 61% using PAC and 40% using GAC under similar conditions. The adsorption potential of JPW was observed to be dependent on various parameters such as type of dye, initial dye concentration, pH and dosage of adsorbent. The batch sorption data conformed well to the Langmuir and Freundlich isotherms. However, lower BOD (33.3%) and COD (13.8%) removal from retting effluent was observed using JPW as compared to 75.6% BOD removal and 71.1% COD removal obtained with GAC.  相似文献

The purpose is to develop a complete microbiological model system for the treatment of wastewater from textile mills in developing countries. Artificial wastewater was treated by microorganisms growing on wood shavings from Norway spruce during unsterile conditions. The microorganisms were inoculated from forest residues. Mixtures of the azo dyes Reactive Black 5 and Reactive Red 2 were degraded in batch as well as continuous experiments. Reactive Red 2 mixed with the anthraquinone dye Reactive Blue 4 was also treated in the continuous system. The system consisted of three reservoirs - the first two with an anaerobic environment and the third with an aerobic. The dye concentrations were 200 mg l−1 of each dye in the continuous system and the retention time was approximately 4 days and 20 h per reservoir. Samples from the process were analysed with spectrophotometer and LC/MS to monitor the degradation process. 86-90% of the colour was removed after a treatment of 4 days and 23 h in the continuous process. Two metabolites were found in the outlets of reactors one and two, but they were degraded to below the detection limit in the aerobic reactor.  相似文献

In the present study sequential anaerobic and aerobic treatment in two steps bioreactor was performed for removal of colour in the pulp and paper mill effluent. In anaerobic treatment, colour (70%), lignin (25%), COD (42%), AOX (15%) and phenol (39%) were reduced in 15 days. The anaerobically treated effluent was separately applied in bioreactor in presence of fungal strain, Paecilomyces sp., and bacterial strain, Microbrevis luteum. Data of study indicated reduction in colour (95%), AOX (67%), lignin (86%), COD (88%) and phenol (63%) by Paecilomyces sp. where as M. luteum showed removal in colour (76%), lignin (69%), COD (75%) AOX (82%) and phenol (93%) by day third when 7 days anaerobically treated effluent was further treated by aerobic microorganisms. Change in pH of the effluent, and increase in biomass of microorganisms substantiated results of the study, which was concomitant to the treatment method.  相似文献
